Quantum theory of a potential biological magnetic field sensor: Radical pair mechanism in flavin adenine dinucleotide biradicals

Recent studies in vitro and in vivo suggest that flavin adenine dinucleotide (FAD) on its own might be able to act as a biological magnetic field sensor. Motivated by these observations, in this study, we develop a detailed quantum theoretical model for the radical pair mechanism (RPM) for the flavin adenine biradical within the FAD molecule. We use the results of existing molecular dynamics simulations to determine the time-varying distance between the radicals on FAD, which we then feed into a quantum master equation treatment of the RPM. In contrast to previous semi-classical models, which are limited to the low-field and high-field cases, our quantum model can predict the full magnetic field dependence of the transient absorption signal. Our model's predictions are consistent with experiments at physiological pH values.

Tetraphenylethene-Based Molecular Cage with Coenzyme FAD: Conformationally Isomeric Complexation toward Photocatalysis-Assisted Photodynamic Therapy.

Flavin adenine dinucleotide (FAD), serving as a light-absorbing coenzyme factor, can undergo conformationally isomeric complexation within different enzymes to form various enzyme-coenzyme complexes, which exhibit photocatalytic functions that play a crucial role in physiological processes. Constructing an artificial photofunctional system using FAD or its derivatives can not only develop biocompatible photocatalytic systems with excellent activities but also further enhance our understanding of the role of FAD in biological systems. Here, we demonstrate a supramolecular approach for constructing an artificial enzyme-coenzyme-type host-guest complex with photoinduced catalytic function in water. First, we have designed and synthesized a water-soluble tetraphenylethene (TPE)-based octacationic molecular cage (1) with a large and flexible cavity, which can adaptively encapsulate with two FAD molecules with "U-shaped" conformation (uFAD) to form a 1:2 host-guest complex (1⊃uFAD2) in water. Second, based on the conformationally isomeric complexation of FAD within 1, the 1⊃uFAD2 complex facilitates electron and energy transfers to molecular oxygen upon the white-light illumination, efficiently producing reactive oxygen species (ROS) such as superoxide radical (O2•-) and singlet oxygen (1O2). To our knowledge, the 1⊃uFAD2 complex acts as a photocatalyst to achieve the highest turnover frequency (TOF) of 35.6 min-1 for the photocatalytic oxidation reaction of NADH via a photoinduced superoxide radical catalysis mechanism in an aqueous medium. At last, combining the cytotoxic effects of ROS and the disruption of the intracellular redox balance involving NADH, 1⊃uFAD2 as a supramolecular photosensitizer displays an excellent oxygen-independent photocatalysis-assisted photodynamic therapy in hypoxic tumors.

Functional and structural impact of 10 ACADM missense mutations on human medium chain acyl-Coa dehydrogenase

Medium chain acyl-CoA dehydrogenase (MCAD) deficiency (MCADD) is associated with ACADM gene mutations, leading to an impaired function and/or structure of MCAD. Importantly, after import into the mitochondria, MCAD must incorporate a molecule of flavin adenine dinucleotide (FAD) per subunit and assemble into tetramers. However, the effect of MCAD amino acid substitutions on FAD incorporation has not been investigated. Herein, the commonest MCAD variant (p.K304E) and 11 additional rare variants (p.Y48C, p.R55G, p.A88P, p.Y133C, p.A140T, p.D143V, p.G224R, p.L238F, p.V264I, p.Y372N, and p.G377V) were functionally and structurally characterized. Half of the studied variants presented a FAD content <65 % compared to the wild-type. Most of them were recovered as tetramers, except the p.Y372N (mainly as dimers). No correlation was found between the levels of tetramers and FAD content. However, a correlation between FAD content and the cofactor's affinity, proteolytic stability, thermostability, and thermal inactivation was established. We showed that the studied amino acid changes in MCAD may alter the substrate chain-length dependence and the interaction with electron-transferring-flavoprotein (ETF) necessary for a proper functioning electron transfer thus adding additional layers of complexity to the pathological effect of ACADM missense mutations. Although the majority of the variant MCADs presented an impaired capacity to retain FAD during their synthesis, some of them were structurally rescued by cofactor supplementation, suggesting that in the mitochondrial environment the levels and activity of those variants may be dependent of FAD's availability thus contributing for the heterogeneity of the MCADD phenotype found in patients presenting the same genotype.

Biocompatible MXene (Ti3C2Tx) Immobilized with Flavin Adenine Dinucleotide as an Electrochemical Transducer for Hydrogen Peroxide Detection in Ovarian Cancer Cell Lines.

Flavin adenine dinucleotide (FAD) is a coenzyme and acts as a redox cofactor in metabolic process. Owing to such problems as poor electron transfer properties, unfavorable adsorption, and lack of stability on rigid electrodes, the bio-electrochemical applications of FAD have been limited. Herein, a novel fabrication method was developed for the immobilization process using 2D MXene (Ti3C2Tx), which enhanced the redox property of FAD and improved the electro-catalytic reduction of hydrogen peroxide (H2O2) in neutral medium. The FAD-immobilized Ti3C2Tx electrode (FAD/Ti3C2Tx) was studied by UV-Visible and Raman spectroscopies, which confirmed the successful adsorption of FAD on the Ti3C2Tx surface. The surface morphology and the elemental composition of Ti3C2Tx were investigated by high resolution transmission electron microscopy and the energy dispersive X-ray analysis. The redox property of the FAD/Ti3C2Tx modified glassy carbon electrode (FAD/Ti3C2Tx/GCE) was highly dependent on pH and exhibited a stable redox peak at −0.455 V in neutral medium. Higher amounts of FAD molecules were loaded onto the 2D MXene (Ti3C2Tx)-modified electrode, which was two times higher than the values in the reported work, and the surface coverage (ᴦFAD) was 0.8 × 10−10 mol/cm2. The FAD/Ti3C2Tx modified sensor showed the electrocatalytic reduction of H2O2 at −0.47 V, which was 130 mV lower than the bare electrode. The FAD/Ti3C2Tx/GCE sensor showed a linear detection of H2O2 from 5 nM to 2 µM. The optimization of FAD deposition, amount of Ti3C2Tx loading, effect of pH and the interference study with common biochemicals such as glucose, lactose, dopamine (DA), potassium chloride (KCl), ascorbic acid (AA), amino acids, uric acid (UA), oxalic acid (OA), sodium chloride (NaCl) and acetaminophen (PA) have been carried out. The FAD/Ti3C2Tx/GCE showed high selectivity and reproducibility. Finally, the FAD/Ti3C2Tx modified electrode was successfully applied to detect H2O2 in ovarian cancer cell lines.

Humic acid/halloysite nanotube/flavin adenine dinucleotide nanocomposite based selective electrochemical biosensor for hydrogen peroxide

Abstract Humic acid (HA) is a natural source of organic matter obtained from the soil. It is a stable and water-soluble compound with heterogeneous organic components which helps the plants to absorb water and other nutrients from the soil. HA contains quinoid redox active groups for electron transfer reactions. In this work, we have used HA as a dispersing agent to prepare Halloysite nanotube (HNT) dispersion. HNT is a one-dimensional, layered aluminosilicate nanotube which contains hydroxyl groups on the surface. HNT has several unique properties such as high surface area, mesoporous material, presence of polar charges and low-cost. As-prepared HA/HNT dispersion was characterized using Ultraviolet–Visible spectroscopy (UV–Vis), X-Ray Diffraction (XRD), and High-resolution transmission electron microscopy (HR-TEM). Furthermore, HA/HNT dispersion was used to modify a glassy carbon electrode (GCE) for successful immobilization of flavin adenine dinucleotide (FAD) from the deoxygenated phosphate buffered saline (PBS, pH 7.4) solution containing FAD molecules. Electrochemical properties of FAD immobilized on GCE/HA/HNT (GCE/HA/HNT/FAD) were studied by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The GCE/HA/HNT/FAD biosensor showed a redox peak centered at (Eo′) −0.45 V in PBS (pH 7.4) at a scan rate of 50 mV/s. This new FAD (electron transfer mediator) based biosensor showed good electrocatalytic activity towards the reduction of hydrogen peroxide (H2O2) at −0.48 V. The calibration graph was obtained from 1 to 250 μM of H2O2 and the limit of detection (LOD) was found to be 0.49 μM. The interference study was also performed in the presence of uric acid (UA), dopamine (DA), ascorbic Acid (AA) and oxalic acid (OA) which confirmed that these molecules did not interfere in the electrochemical measurements. Moreover, the GCE/HA/HNT/FAD biosensor exhibited high reproducibility and repeatability during electrochemical analysis. The real applicability of the biosensor was also demonstrated by detecting H2O2 in an antiseptic solution.

Fluorescence Quenching-Based Evaluation of Glucose Oxidase Composite with Conducting Polymer, Polypyrrole

Glucose oxidase (GOx) composites with conducting polymers (e.g., polypyrrole (Ppy)) are excellent nanobiomaterials suitable for the design of bioelectronic devices such as biosensors and biofuel cells. Here, we address the spectroscopic properties of GOx, flavin adenine dinucleotide (FAD), and composites of these compounds with polypyrrole (Ppy). The exploration of native GOx and FAD solutions confirmed that about 5% of FAD dissociated from GOx during the period of solution preparation, and this fraction remained constant for 1 month. It has been found that the Ppy, which formed composites with FAD and GOx, facilitated the removal of FAD molecules from GOx and twice reduced the fluorescence decay rate. Differences in the FAD and Ppy average fluorescence relaxation times showed that the FAD composite with Ppy and Ppy effectively quenched the FAD fluorescence and FAD could not freely unfold. A Novel Approach to Simulate a Charge Transfer in DNA Repair by an Anacystis nidulans Photolyase.

An Anacystis nidulans photolyase enzyme containing two chromophore cofactors was simulated for a photoreaction DNA repairing process via molecular dynamics (MD) method. A novel approach has been introduced for the electron transfer between the FAD (flavin adenine dinucleotide; flavin) molecule and CPD (cyclobutane pyrimidine dimer). This approach involves four simulation stages with different charges for the FAD and CPD fragments and a role of a charged state of the active cofactor was qualified during the MD modeling. Observations show that flavin has actively participated in a charge transfer process, thereby involving the conformational changes of the DNA and CPD substrate fragment. The DNA conformation behavior has shown to correlate with the electron transfer from flavin to CPD. This is manifested on the similarities of the DNA repairing process by excision repair of the UV photoproducts.

MD studies on conformational behavior of a DNA photolyase enzyme

In this work, molecular dynamics (MD) simulations were performed on a DNA photolyase protein with two cofactors, FAD (flavin adenine dinucleotide) and MTHF (methenyltetrahydrofolate), inside the enzyme pocket. A DNA photolyase is a highly efficient light-driven enzyme that repairs the UV-induced cyclobutane-pyrimidine dimer in damaged DNA. We were aimed to compare the conformational changes of the FAD cofactor and other constituent fragments of the molecular system under consideration. The conformational behavior of the FAD molecule is very important for understanding the functional and structural properties of the DNA repair protein photolyase. The photoactive FAD is an essential cofactor both for specificial binding to damaged DNA and for catalysis. The second chromophore (MTHF or 8-HDF) is not necessary for catalysis and has no effect on specific enzyme—substrate binding. The obtained results were discussed to gain insight into the light-driven mechanism of DNA repair by a DNA photolyase enzyme—based on the enzyme structure, the FAD mobility, and conformation shape.

Cover Picture: On the Importance of Anion–π Interactions in the Mechanism of Sulfide:Quinone Oxidoreductase (Chem. Asian J. 11/2013)

The anion–π interaction is a well-recognized noncovalent force that is established between electron-deficient rings and anions. In the Communication by Antonio Frontera and co-workers on page 2708 ff., the relevance of this noncovalent interaction has been put on the scene by highlighting its critical role in the mechanism of sulfide:quinone oxidoreductase. In the first step of the enzymatic mechanism, an anionic intermediate is formed and is stabilized by an anion–π interaction that involves a cysteine residue and a flavin adenine dinucleotide (FAD) molecule. In the Cover picture, a dragon spits fireballs that represent the sulfur atoms of the two cysteine residues of the active site (disulfide bridge), of which one interacts with the FAD molecule placed on a branch.

Potential-dependent single molecule blinking dynamics for flavin adenine dinucleotide covalently immobilized in zero-mode waveguide array of working electrodes

Single molecules exhibit a set of behaviors that are characteristic and distinct from larger ensembles. Blinking is one such behavior that involves episodic transitions between luminescent and dark states. In addition to the common blinking mechanisms, flavin adenine dinucleotide (FAD), a cofactor in many common redox enzymes, exhibits blinking by cycling between a highly fluorescent oxidized state and a dark reduced state. In contrast to its behavior in flavoenzymes, where the transitions are coupled to chemical redox events, here we study single FAD molecules that are chemically immobilized to the Au region of a zero-mode waveguide (ZMW) array through a pyrroloquinoline quinone (PQQ) linker. In this structure, the Au functions both to confine the optical field in the ZMW and as the working electrode in a potentiostatically controlled 3-elecrode system, thus allowing potential-dependent blinking to be studied in single FAD molecules. The subset of ZMW nanopores housing a single molecule were identified statistically, and these were subjected to detailed study. Using equilibrium potential, E(eq), values determined from macroscopic planar Au electrodes, single molecule blinking behavior was characterized at potentials E < E(eq), E - E(eq), and E > E(eq). The probability of observing a reduced (oxidized) state is observed to increase (decrease) as the potential is scanned cathodic of E(eq). This is understood to reflect the potential-dependent probability of electron transfer for single FAD molecules. Furthermore, the observed transition rate reaches a maximum near E(eq) and decreases to either anodic or cathodic values, as expected, since the rate is dependent on having significant probabilities for both redox states, a condition that is obtained only near E(eq).

Extremophile Enzymatic DNA Repair

Flavin adenine dinucleotide (FAD) - dependent DNA photolyase repairs UV-damaged DNA by a photo-induced electron transfer reaction, the rate constant of which is exponentially dependent on temperature. However, how this process is adapted to function in extremophiles, inhabiting thermally extreme environments, is currently unknown. To study the temperature dependence of biological electron transfer under extreme conditions, we have produced recombinant DNA photolyases in E.coli from two extremophiles: Sulfolobus solfataricus, a hyperthermophilic archaeon (rSsPL) and Colwellia psychrerythraea, a psychrophilic bacterium (rCpPL). To the best of our knowledge, rCpPL is the first cold-adapted DNA photolyase to be isolated.Both photolyases contain a noncovalently bound FAD molecule and their absorption spectra display unique features of the protein-bound FAD, suggestive of temperature adaptation. Structural modeling of both photolyases revealed unique features related to their respective thermal environments. The repair activity of rSsPL, in vitro, was comparable to E.coli DNA photolyase (EcPL) at ambient temperature, whereas the repair activity of rCpPL appears to be much slower than its mesophilic and thermophilic counterparts. A comparative study of the repair activities of all three photolyases (EcPL, rCpPL and rSsPL) as a function of various physical and chemical parameters such as, temperature, ionic strength, viscosity, and pH is presented. Denaturation studies on rSsPL reveal its extreme stability as compared to rCpPL, which is extremely sensitive to its thermal and chemical environments.

Time-resolved emission of flavin adenine dinucleotide in water and water–methanol mixtures

Time-resolved fluorescence decay of flavin adenine dinucleotide (FAD) was studied at room temperature in water and water-methanol mixtures by a fluorescence upconversion technique. The observations were focused on the most initial decay phase (200 ps), before the residual fluorescence assumes a single exponential decay, typical for an extended conformation of the fluorophore. Within the first few picoseconds, where most of the electron transfer coupled quenching takes place, the emission decay curves could be fitted by a stretched exponent, compatible with the inhomogeneous distance dependent electron transfer model. This implies that the population of the excited FAD molecules exhibits a large number of non-identical states, each with its own separation between the donor (adenine) and acceptor (isoalloxazine) moieties, having its own rate of electron transfer. To evaluate the distribution of the separation between the donor-acceptor pair, we carried out molecular dynamics simulations of closed conformation of the FAD in water and water-methanol mixtures, sampling the structure at 10 fs intervals. The analysis of the dynamics reveals that within the 4 ps time frame, where most of the nonexponential fluorescence relaxation takes place, the relative motion of the donor-acceptor pair is consistent with a one-dimensional Brownian motion, where the diffusion coefficient and the shape of the confining potential well are solvent dependent. The presence of methanol enhances the diffusion constant and widens the width of the potential well. On the basis of these parameters, the relaxation dynamics was accurately reconstructed as an electron transfer reaction in an inhomogeneous system where the reactants are diffusing within the time frame of the observation.

A Fluorescence Perspective on the Differential Interaction of Riboflavin and Flavin Adenine Dinucleotide with Cucurbit[7]uril

The interaction of the macrocyclic host, cucurbit[7]uril (CB7), with riboflavin (RF) and its derivative, flavin adenine dinucleotide (FAD), has been investigated using absorption and steady-state and time-resolved fluorescence measurements. Interestingly, in the presence of CB7, the fluorescence intensity of RF is quenched, whereas the fluorescence intensity of FAD is enhanced. It is proposed that the fluorescence quenching of RF results from the tautomerization of its isoalloxazine moiety from the lactam to the lactim forms, upon binding to CB7. Such a tautomerization can be brought about since the two lactim forms have higher dipole moments than the lactam form of RF, and thus experience much stronger dipole-dipole interactions (and hence greater binding affinities) with CB7 in the former cases than in the latter. This tautomerization in the presence of CB7 leads to a significant reduction in the observed radiative decay rate and hence a reduction in the fluorescence intensity of RF. Binding of CB7 with RF is confirmed by an increase in the rotational correlation time of RF in the presence of CB7. Geometry optimization studies indicate the formation of an exclusion complex between CB7 and RF, possibly stabilized by H-bonding interactions, as also suggested by the characteristic red shift in the absorption spectra of the CB7-RF system. In the case of FAD, both the isoalloxazine ring and the adenine moiety can interact with the CB7 host. In aqueous solutions, a good fraction of FAD molecules exists in a "closed" conformation with the adenine and isoalloxazine rings stacked together, thus leading to very efficient fluorescence quenching due to the ultrafast intramolecular electron transfer from adenine to the isoalloxazine moiety. Binding of the adenine and/or the isoalloxazine moiety of FAD with CB7 inhibits the stacking interaction and changes the "closed" conformation to the "open" conformation, wherein the adenine and isoalloxazine moieties are widely separated, thus prohibiting the electron transfer process. This reduces the inherent fluorescence quenching of FAD molecule and results in the observed fluorescence enhancement. As observed for RF, the interaction of CB7 with the isoalloxazine ring of FAD should cause fluorescence quenching due to the lactam to lactim tautomerization process. However, in the interplay between the above two opposing effects, the fluorescence enhancement due to the modulation in the conformational dynamics of FAD by the CB7 host predominates. The conformational change is in fact supported by the observation of a long lifetime component in the fluorescence decay of FAD in the presence of CB7. Moreover, at acidic pH, when FAD is already present mainly in the "open" form, the conformational dynamics no longer plays any major role and the fluorescence of FAD is quenched by CB7, as expected, due to the tautomerization at the isoalloxazine moiety.

Inhibiting Intramolecular Electron Transfer in Flavin Adenine Dinucleotide by Host−Guest Interaction: A Fluorescence Study

Modulation in the photophysical properties and intramolecular electron transfer behavior of the flavin adenine dinucleotide (FAD) molecule has been investigated in the presence of the macrocyclic hosts, alpha-, beta- and gamma-cyclodextrins (CDs), using absorption and steady-state and time-resolved fluorescence measurements. The results demonstrate that only the beta-CD host has a suitable cavity dimension to form a weak inclusion complex with FAD by encapsulating the adenine moiety, which is the preferred binding site in the large FAD molecule. Interestingly, in spite of the weak binding interaction, a significant enhancement in the fluorescence intensity of FAD is observed on complexation with beta-CD, and this has been attributed mainly to the modulation in the conformational dynamics of FAD in the presence of beta-CD. In aqueous solutions, a good fraction of FAD molecules exist in a "closed" conformation with the adenine and isoalloxazine rings stacked on each other, thus leading to very efficient fluorescence quenching due to the ultrafast intramolecular electron transfer from adenine to the isoalloxazine moiety. Complex formation with beta-CD inhibits this intramolecular electron transfer by changing the "closed" conformation of FAD to the "open" form, wherein the adenine and isoalloxazine moieties are widely separated, thus prohibiting the fluorescence quenching process. Further evidence for the conformational changes has been obtained by the observation of a long lifetime component in the fluorescence decay of FAD in the presence of beta-CD, which corresponds to the decay of the unquenched "open" form of FAD. Fluorescence up-conversion studies also indicate the absence of any ultrafast component in the fluorescence decay arising from the complexed FAD, thus supporting the formation of the "open" form in the presence of beta-CD, with no intramolecular electron transfer.

Phospholipid Bilayers Supported on Thiolate‐Covered Nanostructured Gold: In Situ Raman Spectroscopy and Electrochemistry of Redox Species

Thiol-covered nanostructured gold has been tested as a platform for the preparation of high-area phospholipid bilayer systems suitable for optical and electrochemical sensing. In situ and ex situ Raman spectroscopy and electrochemical measurements are made to study methylene blue (MB) and flavin-adenine dinucleotide (FAD) incorporation into dimyristoylphosphatidylcholine (DMPC) bilayers prepared by vesicle fusion on dithiothreitol (DTT)-covered nanostructured gold. Results show that lipophilic positively charged MB molecules are incorporated in the bilayer reaching the DTT-gold interface. On the other hand, the negatively charged FAD molecules are immobilized at the outer part of the phospholipid bilayer and cannot be electrochemically detected. Our results demonstrate that DTT-covered nanostructured gold provides a suitable high-area platform for phospholipid membranes that are able to separate and sense different kinds of molecules and biomolecules.

Involvement of PEG-carboxylate dehydrogenase and glutathione S-transferase in PEG metabolism by Sphingopyxis macrogoltabida strain 103

Sphingopyxis terrae and the Sphingopyxis macrogoltabida strains 103 and 203 are able to degrade polyethylene glycol (PEG). They possess the peg operon, which is responsible for the conversion of PEG to PEG-carboxylate-coenzyme A (CoA). The upstream (3.0 kb) and downstream (6.5 kb) regions of the operon in strain 103 were cloned and sequenced. The structure was well conserved between S. macrogoltabida strain 203 and S. terrae, except that two sets of transposases are absent in strain 203. The downstream region contains the genes for PEG-carboxylate dehydrogenase (PCDH), glutathione S-transferase (GST), tautomerase, and a hypothetical protein. The genes for pcdh and gst were transcribed constitutively and monocistronically, indicating that their transcription is independent of the operon regulation. PCDH and GST were expressed in Escherichia coli and characterized biochemically. PCDH is a homotetramer of 64-kDa subunits and contains one molecule of flavin adenine dinucleotide per subunit. The enzyme dehydrogenates PEG-carboxylate to yield glyoxylate, suggesting that the enzyme is the third enzyme involved in PEG degradation. GST is a homodimer of 28-kDa subunits. GST activity was noncompetitively inhibited by acyl-CoA and PEG-carboxylate-CoA, suggesting the interaction of GST with them. The proposed role for GST is to buffer the toxicity of PEG-carboxylate-CoA.

Cores and pH-dependent Dynamics of Ferredoxin-NADP+ Reductase Revealed by Hydrogen/Deuterium Exchange

NMR-detected hydrogen/deuterium (H/D) exchange of amide protons is a powerful way for investigating the residue-based conformational stability and dynamics of proteins in solution. Maize ferredoxin-NADP(+) reductase (FNR) is a relatively large protein with 314 amino acid residues, consisting of flavin adenine dinucleotide (FAD) and nicotinamide adenine dinucleotide phosphate (NADP(+))-binding domains. To address the structural stability and dynamics of FNR, H/D exchange of amide protons was performed using heteronuclear NMR at pD(r) values 8.0 and 6.0, physiologically relevant conditions mimicking inside of chloroplasts. At both pD(r) values, the exchange rate varied widely depending on the residues. The profiles of protected residues revealed that the highly protected regions matched well with the hydrophobic cores suggested from the crystal structure, and that the NADP(+)-binding domain can be divided into two subdomains. The global stability of FNR obtained by H/D exchange with NMR was higher than that by chemical denaturation, indicating that H/D exchange is especially useful for analyzing the residue-based conformational stability of large proteins, for which global unfolding is mostly irreversible. Interestingly, more dynamic conformation of the C-terminal subdomain of the NADP(+)-binding domain at pD(r) 8.0, the daytime pH in chloroplasts, than at pD(r) 6.0 is likely to be involved in the increased binding of NADP(+) for elevating the activity of FNR. In light of photosynthesis, the present study provides the first structure-based relationship of dynamics with function for the FNR-type family in solution.

Purification, Characterization, and Crystallization of the Components of the Nitrobenzene and 2-Nitrotoluene Dioxygenase Enzyme Systems

The protein components of the 2-nitrotoluene (2NT) and nitrobenzene dioxygenase enzyme systems from Acidovorax sp. strain JS42 and Comamonas sp. strain JS765, respectively, were purified and characterized. These enzymes catalyze the initial step in the degradation of 2-nitrotoluene and nitrobenzene. The identical shared reductase and ferredoxin components were monomers of 35 and 11.5 kDa, respectively. The reductase component contained 1.86 g-atoms iron, 2.01 g-atoms sulfur, and one molecule of flavin adenine dinucleotide per monomer. Spectral properties of the reductase indicated the presence of a plant-type [2Fe-2S] center and a flavin. The reductase catalyzed the reduction of cytochrome c, ferricyanide, and 2,6-dichlorophenol indophenol. The ferredoxin contained 2.20 g-atoms iron and 1.99 g-atoms sulfur per monomer and had spectral properties indicative of a Rieske [2Fe-2S] center. The ferredoxin component could be effectively replaced by the ferredoxin from the Pseudomonas sp. strain NCIB 9816-4 naphthalene dioxygenase system but not by that from the Burkholderia sp. strain LB400 biphenyl or Pseudomonas putida F1 toluene dioxygenase system. The oxygenases from the 2-nitrotoluene and nitrobenzene dioxygenase systems each had spectral properties indicating the presence of a Rieske [2Fe-2S] center, and the subunit composition of each oxygenase was an alpha(3)beta(3) hexamer. The apparent K(m) of 2-nitrotoluene dioxygenase for 2NT was 20 muM, and that for naphthalene was 121 muM. The specificity constants were 7.0 muM(-1) min(-1) for 2NT and 1.2 muM(-1) min(-1) for naphthalene, indicating that the enzyme is more efficient with 2NT as a substrate. Diffraction-quality crystals of the two oxygenases were obtained.

Amperometric cholesterol biosensor based on in situ reconstituted cholesterol oxidase on an immobilized monolayer of flavin adenine dinucleotide cofactor

A new amperometric biosensor for determining cholesterol based on deflavination of the enzyme cholesterol oxidase (ChOx) and subsequent reconstitution of the apo-protein with a complexed flavin adenine dinucleotide (FAD) monolayer is described. The charge transfer mediator pyrroquinoline quinone (PQQ) was covalently bound to a cystamine self-assembled monolayer (SAM) on an Au electrode. Boronic acid (BA) was then bound to PQQ using the carbodiimide procedure, and the BA ligand was complexed to the FAD molecules on which the apo-ChOx was subsequently reconstituted. The effective release of the FAD from the enzyme and the successful reconstitution were verified using molecular fluorescence and cyclic voltammetry. The optimal orientation of FAD toward the PQQ mediator and the distances between FAD and PQQ and between PQQ and electrode enhance the charge transfer, very high sensitivity (about 2500 nA mM −1 cm −2) being obtained for cholesterol determination. The biosensor is selective toward electroactive interferents (ascorbic acid and uric acid) and was tested in reference serum samples, demonstrating excellent accuracy (relative errors below 3% in all cases). The biosensor activity can be successfully regenerated in a simple process by successive reconstitution with batches of recently prepared apo-ChOx on the same immobilized Au/SAM–PQQ–BA–FAD monolayer (it was tested five times); the lifetime of the biosensor is about 45–60 days.
