FRESH-WATER GREEN ALGAE (CHLOROPHYTA) AS A NATURAL PIGMENT FOR MOJOSARI DUCKS

An experiment in a completely randomize design was undertaken to study the use of fresh-watergreen algae as a yolk coloring agent in Mojosari ducks during a laying period on productiveperformance and egg quality from 36 to 44 wk of age. A total of 80 thirty-six wk–old laying ducks weredivided into four dietary treatments and each of four replicates with 5 birds. Diets were formulated witha commercial concentrate, rice bran and yellow corn (2:4:4) according to a commercial standard diet asa control, and three other dietary treatments with 2, 4 or 8% of green algae were included. Fresh watergreen algae had a significant effect on the feed uptake, egg production, and feed conversion ratio (FCR)(p 0.05) except eggyolk color. The yolk color increased within 7 days after feeding with the test diets. The present studyindicated that fresh-water green algae could be used as a natural coloring agent in laying ducks and at8% of green algae showed the highest score of (Roche Yellow Color-15).

Review on the Performances of Cihateup Duck (Anas platyrhynchos Javanica) as Genetic Resource of Local Poultry in Indonesia

Cihateup duck is one of the local Indonesian poultry genetic resources originated from West Java. These ducks are raised in Tasikmalaya and its surrounding area. Cihateup duck raised as layer or meat type duck is potential to provide business opportunities for people in West Java. For future development of Cihateup duck, some important potential characteristics of Cihateup ducks are required. This paper describes the phenotype performance, rearing management of Cihateup ducks and its genetic relationship with other ducks. Cihateup ducks have longer neck, wing, femur and tibia compared with other Indonesian ducks. Egg production (200 egg/head/year) is lower than Alabio and Mojosari ducks. The quality of meat (taste and flavour) is less preferred by consumers because of its odor. Genetic relationship between Cihateup duck with local ducks in Java is closer than Alabio ducks from South Kalimantan. Key words: Cihateup duck, performance, genetic resource

Efficacy of application of vaccine AI H5N1 clade 2.1.3 on Mojosari ducks challenge against AI H5N1 clade 2.3.2 in laboratory conditions

Influenza virus HPAI H5N1 clade 2.3.2 outbreaks since September 2012 caused high mortality in ducks. Vaccination is one of strategies recommended by government. However, AI H5N1 clade 2.3.2 vaccine not yet available during this research, while AI H5N1 clade 2.1.3 vaccines available in markets. Therefore it was important to do study on efficay of HPAI H5N1 clade 2.1.3. vaccines on duck at laboratory scale. Three groups of Mojosari duck were used in this study, they were 1 group vaccinated with A Vaccine, 1 group vaccinated with B Vaccine, and 1 group as control (not vaccinated). Vacination groups consisted of 9 DOD and control group was consisted of 6 DOD. Vaccination was conducted when the duck at three weeks old of age using single dose recommended by producer. At three weeks later (ducks at 6 weeks old of age) all Groups of ducks were challenged with virus HPAI H5N1 clade 2.3.2 at dose 106 EID50/ml by drops intranasaly. Result showed that Group 1 (vaccinated with A Vaccine) produced 67% protection (3 out of 9 ducks died), Group 2 (vaccinated with B Vaccine) produced 100% protection (non out of 9 ducks died), and Group 3 (control, not vaccinated) produce 0% protection (all of 9 ducks died). This study give an alternative of choise to use AI H5N1 Clade 2.1.3 vaccine with high protection when AI H5N1 Clade 2.3.2 vaccine not available in markets to controll high mortality in ducks caused by HPAI H5N1 clade 2.3.2 outbreaks. UJI POTENSI PROBIOTIK Lactobacillus plantarium SECARA IN-VITRO

<p>Probiotic is a product containing non-pathogenic microbes live, which was given to animals or humans to fix the rate of growth, efficiency, and increase conversion ration of animal or human health by affecting positively the balance of the gut microbes and microbial pathogen control in the digestive tract. In order to potentially lactic acid bacteria as probiotic candidates must pass the test selection among others such as probiotic test resistance to acid pH, the bile salts, against pathogenic bacteria. This experimental research methods for descriptive use that aim to provide information about the probiotic potential of Lactobacillus plantarum was isolated from the small intestine Mojosari duck (<em>Anasplathyrinchos</em>) with the tests in vitro i.e. the resistance of <em>Lactobacillus plantarum</em> on the condition of the acidicpH (2, 3, and 4), bile salt concentration of 0.3% (b/v), inhibition of pathogenic bacteria (<em>Escherichia coli, Staphylococcus aureus</em> and <em>Salmonella typhi</em>) The results showed that on pH 2 the number of cells of bacteria that lives was 4.3. 10<sup>7</sup> CFU/ml, pH 3 was 3.8. 10<sup>9</sup> CFU/ml and pH 4 was 2.7. 10<sup>10</sup> CFU/ml, the resistance of <em>Lactobacillus plantarum</em> on bile salt showed the number of cells of bacteria that lives was 1.2. 10<sup>9</sup> CFU/ml.inhibition of the pathogenic bacteria showed that<em>Lactobacillus plantarum</em>could inhibited strong on Escherichia coli and Staphylococcus aureus with diameter of zone was 12.7 mm and 13.3 mm but not<em>Salmonellatyphi</em> with diameter of zone was 9.3 mm.</p>

GENETIC VARIATION IN THREE BREEDS OF INDONESIAN LOCAL DUCKS BASED ON BLOOD AND EGG WHITE PROTEIN POLYMORPHISM

A study of genetic variation of four blood protein loci and three egg white loci in three breeds ofIndonesian local ducks (Magelang duck = MAD, Tegal duck = TED and Mojosari duck= MOD) wasinvestigated by using polyacrilamide gel electrophoresis (PAGE). The result showed that four bloodprotein loci (Pre-albumin, Albumin, Transferrin and Post-transferrin) and three egg white loci(Ovalbumin, Ovoglobulin and Conalbumin) were found to be polymorphic in three breeds of localducks. There was no difference on blood and egg white protein polyimorphism. The average value ofheterozygosity on three breeds of local ducks had a genetic potential to determine the controlled matingsystem and selection to obtain pure line. The closest phylogenetic relationship among the three breeds oflocal ducks occurred between MAD and TED.

Polymorphism of D-Loop Mitochondrial DNA Region and Phylogenetic in Five Indonesian Native Duck Population

2 Abstract: This research aimed to determine mitochondrial DNA (mtDNA) D-loop region polymorphism and phylogenetic in five Indonesian native ducks population namely Magelang duck, Tegal duck, Mojosari duck, Bali duck and Alabio duck. The significance of this research was applicable to conservation and refinement strategy as well as the improvement of genetic quality by utilizing the available native duck plasma nutfah. It further concerned with determining the genealogy of family, channel, breed or maternal inheritance that bred individuals in native duck population in Indonesia and discovering phylogenetic relations with Mallards duck (Anas platyrhynchos) and the other Anas ducks. Fragment of 718-bp was amplified by PCR and determined the nucleotide sequence of 701-bp. The sequence was then analyzed using Single Nucleotide Polymorphism (SNP) and compared to the standard nucleotide sequence from Anas Platyrhynchos complete genome (HM010684.1) in GenBank Accession. It obtained nucleotide percentage equation of 93.59±8.23%. Phylogenetic investigation used sequencing products and was analyzed using MEGA5 software. Indonesian native ducks have a relatively close genetic relationship with Anas Platyrhynchos and Anas zonorhyncha shown by the genetic distance varying from 0.000-0.786 compared to the other Anas ducks in the world (0.073-1.037) or to Cairina moschata (2.972-5.776). Highly distant genetic variation was found in Magelang duck compared to the other native ducks ranged from 0.000-0.950 to 0.000-0.312. The research concluded that polymorphism of mtDNA D-loop region was found in Indonesian native ducks and had relatively similar maternal inheritance with Anas platyrhyncos dan Anas zonorhyncha.

Pengaruh Kombinasi Sagu Kukus (Metroxylon Spp) dan Tepung Keong Mas (Pomacea Spp) sebagai Pengganti Jagung Kuning terhadap Penampilan Itik Jantan Alabio, Mojosari dan Hasil Persilangannya (The Effect of Steaming Sago (Metroxylon Spp) and Golden Snail Meal

<p>The aim of the experiment was to find out the effects of steaming sago (Metroxylon Spp) and golden snail meal (Pomacea Spp) combination for yellow corn substitution, on the performance of male Alabio, Mojosari, and their cross<br />(MA). One hundred and ninety two young male ducks from the three breeds of 7 day old were assigned in 48 units of pens (4 ducks/pen). The experimental design was Completely Randomized Design with factorial (3x4) the first factor<br />was duck breeds (a) consisted of three duck breeds, Alabio, Mojosari, and Raja ducks, and the second factor was the type of ration of: R0 (control/100% basal ration), R1 (basal ration + 13% steaming sago and 2% golden snail flour), R2<br />(basal ration + 26% steaming sago and 4% golden snail flour) and R3 (basal ration + 39% steaming sago and 6% golden snail flour). All treatments were repeated four times. The observed variables were the performance of ducks<br />(body weight, body weight gain, ration consumption, and feed conversion ratio). The results indicated that breed had significant effects (P<0.05) on body weight, body weight gain, and ration conversion. Meanwhile, types of feed had<br />significant effects (P<0.05) on feed consumption, body weight gain, and final body weight. It was concluded that the combination of steaming sago and golden snail meal up to 45% of the ration could replace the need for yellow corn as<br />source of energy without affecting the performance of male ducks of 1–8 weeks old. Cross male ducks were better than male Alabio and Mojosari ducks because they were more efficient in converting ration in to meat which resulting in<br />higher body weight gain.</p><p>(Key words: Steaming sago, Golden snail flour, Male ducks, Performance)<br /><br /></p>

Effect of genotypes and aflatoxin levels in the diets on early laying characteristics of local ducks

Layer ducks are known for their sensitivity to aflatoxin contamination in their diets, and this has been causing problem in egg productivity of local ducks due to feeding system practiced by traditional farmers. This study was aimed at exploring the level of sensitivity of Mojosari and Tegal ducks to different aflatoxin levels in their diets, especially on characteristics during early laying period. Three hundreds and nine female Mojosari ducks and 260 female Tegal ducks were used in this study. Two levels of aflatoxin in the diets, 0 and 150 ppb, were given to 4 week-old ducklings for 4 weeks. Observations were taken on age at first laying, body weight at first laying, and egg qualities as early indications of the egg production potentials. The results showed that different levels of aflatoxin in the diets only affected live weight at first laying, and not age at first laying. The two genotypes differed significantly in egg weight, yolk weight, and shell weight. It is concluded that aflatoxin contamination in the diets during early growth did not really affect characteristics of early laying of Mojosari and Tegal ducks. Key Words: Local Ducks, Aflatoxin

Effect of steamed sago and golden snail flour as substitution of yellow corn on performance of male duck of Alabio, Mojosari and MA

This study was aimed at finding out the effects of combining steaming sago ( Metroxylon Spp) and golden snail flour ( Pomacea Spp) as the source of energy in duck ration, substituting yellow corn, on the performance of male Alabio, Mojosari, and their cross (MA). One hundred and ninety two young male ducks from the three breeds of 7 days old were assigned in 48 units of cage (4 ducks/cage). Research method applied was Completely Randomized Design with factorial pattern (3x4). The first factor was duck breeds (a) consisted of tree duck breeds: Alabio duck, Mojosari duck and Raja duck and the second factor was types of ration: R0 (control/100% basal ration), R1 (basal ration + 13% steaming sago and 2% golden snail flour), R2 (basal ration + 26% steaming sago and 4% golden snail flour) and R3 (basal ration + 39% steaming sago and 6% golden snail flour). All treatments were repeated four times. The observed variables were performance of ducks: body weight, body weight gain, ration consumption and ration conversion. Results indicated that breed had significant effect (P < 0.05) on body weight, body weight gain and ration conversion. Meanwhile, ration had significant effect (P < 0.05) on ration consumption, body weight gain, and final body weight. It is concluded that the combination of steaming sago and golden snail flour up to 45% of the ration could replace the need for yellow corn as the source of energy in the ration without affecting the performance of male ducks of 1 – 8 weeks old. Cross male ducks were better than that male Alabio ducks and Mojosari because they were more efficient in converting ration into meat which resulted in higher body weight gain. Heterosis of the crossbred between Tegal and Mojosari ducks under sub-optimal condition

Crossbreeding between two different strains or breeds is often done in a livestock production system to take advantage of the heterosis (hybrid vigour) which may resulted from the crossing. The level of heterosis largely depend on the genetic distance betweeen the two parental groups, and is also affected by the environmental condition in which the heterosis would be more significant under less optimal condition. The aim was to evaluate the heterosis level of the crosses between Tegal and Mojosari ducks under stressful environment of low nutritional content (14% protein) in the diet. The experiment was conducted in the animal facilities of the Indonesian Research Institute for Animal production, using 40 Tegal (TT) ducks, 44 Mojosari ducks (MM), 46 crossbreds between Tegal drakes and Mojosari ducks (TM), and 42 crossbreds between Mojosari drakes and Tegal ducks (MT). Results showed that heterosis was not significant for the average age at first laying (AFL) and weight of first egg (WFE), in which AFL of TT (171.8 d) was not significantly different from that of MT (163.4 d) and both were shorter than the AFL of MM (182.3 d) or that of TM (182.8 d). Heterosis also did not exist on egg production to 68 weeks old (EP-68), in which the EP-68 of TT was 141.3 eggs and that of MT was 122.6 eggs, and both were lower than EP-68 of MM (156.8 eggs) or of TM (154.0 eggs). For egg qualities, heterosis also did not show any significant level and difference between genotypes only existed for the Haugh Unit values. It seems that even under stressful environment heterosis did not exist among crosses between Tegal and Mojosari ducks, and therefore it can be concluded that the crosses between these two breeds will not produce any beneficial effects in a production system under any condition. Estimation of genetic parameters for body weight of Alabio and Mojosari ducks at starter period

A selection program is one of many important tools in livestock breeding in improving the quality of breeding stock. The choice of an effective selection method requires some information on the value of genetic parameters for some economically important traits, such as heritabilities and genetic correlation coefiicients. This experiment used 25 drakes and 100 ducks of each Alabio and Mojosari ducks, mated at random 1 drake to 4 ducks. A number of ducklings were obtained from each mating in each population, and their body weights were observed from hatching to 8 weeks old. Results showed that the heritability estimation for body weight to 8 weeks old were generally low either in Alabio or in Mojosari, ranging between 0.061 to 0.227. The highest heritability estimation was obtained for 6-week body weight 0.151 for Alabio and 0.227 for Mojosari ducks. The estimates of genetic correlation among body weights varied widely but generally high. It is concluded that 6-week body weight can be considered as a selection criterion depending on the selection objective in the local Indonesian ducks. Moulting patterns of Alabio and Mojosari ducks and their relation on blood lipids (tryglycerides), egg production and egg quality

Moulting is a biological condition that can happen in poultry. It is resulted from the complex interaction which involves the function of thyroxine hormone. Moulting can reduce or even stop the egg production. A study was conducted to observe the moulting patterns of local ducks (Alabio and Mojosari) and to determine the relation of moulting with blood lipids (triglycerides), egg production and quality. Each breed consisted of ten female dukcs were observed for moulting pattern, blood triglycerides, egg production and quality. Fourty ducks were used for simulation of egg production. Data from moulting patterns,egg production and quality were analyzed using t-based on Least Square means with Statistical Analysis System. The relation of breeds and moulting patterns with triglycerides were analyzed using Analysis of Variance (ANOVA) for a completely randomized design in a factorial arrangement of 2x2. The main factor was kind of breeds, while the subfactor was the period of moulting, before and during moulting. There were not interaction in every variables between both factors. The average moulting periode of Alabio was significantly (P<0.05) shorter than that of Mojosari (69 vs 76 days). There were 40% of Alabio ducks moulting for 61-70 days, while 40% of Mojosari ducks moulting for 71-80 days. Egg production of Alabio ducks before and after moulting were higher than those Mojosari ducks. The triglycerides content of Alabio and Mojosari ducks was decreased during moulting, in Alabio ducks they were 32.02 and 27.64 μg/ml before and during moulting, while in Mojosari ducks they were 32.83 and 29.32 μg/ml respectively. Egg weight, albumin weight, yolk weight, and haugh unit of the two breeds increased after moulting, while yolk colour decreased. The average yolk colour of Alabio ducks before and after moulting were 6.90 and 5.11, while in Mojosari ducks they were 7.90 and 4.60 respectively. Expression of recessive homozygote gene (c/c) on the quality of first eggs in Mojosari duck

Recessive homozygote gene ( c/ c) in poultry is usually expressed in white plumage, and thought to affect growth and egg production. This effect is still under discussion because can be positive or negative. In order to study further the expression of the recessive gene on Mojosari duck this study was aimed to investigate the quality of first eggs from ducks with brown and white plumage. The result showed that weight of first eggs of ducks with dominant gene ( C/c ), was 52.91 g higher than that of duck with homozygote recessive gene ( c/c ) which 51.43 g. For other variable, there was no significant different between ducks with dominant gene ( C/c ) and with recessive gene ( c/c ), i.e. weight of egg yolk (14.99 vs. 14.94 g), weight of egg white (31.34 vs. 29.94 g) weight of wet shell (6.62 vs. 6.56 g) and thickness (0.36 vs. 0.34). However there was significant different between the two group for score of Haugh Unit (89.67 vs. 101.12) and egg yolk color (7.30 vs. 5.35). It is obvious that the expression of the recessive homozygote gene ( c/c ) did not give any significant difference to the quality of first egg, except for the color which need to the confirmed with more and longer observations. Predicted of production curve and persistensy by wood non-linier model on white Mojosari duck

Until recent, the biology data from white Mojosari duck have been never reported. Different from their sister, the brown Mojosari duck was establish for laying duck. Equation model of non-linier Wood was commonly used to predict the milk production curve. This method is possible for egg production of laying duck. The purpose of the research is understanding of production curve fitted and persistency. The data were collected from 39 birds of White Mojosari duck in Balitnak Ciawi. Four types of analysis data set, those are partial data (16 weeks), full data (52 week), collective and individual data of egg production. The equation of mathematic non-linier Wood is Y=AtBexp-kt by DUD method. The result show that egg production was 236 for 52 weeks record and twenty five percent were reached up than 290 eggs. Production trait will decrease on 4-5th month and after that slow up for increasing, but never came as high as the first and second month production. Estimation of A parameter is 5.5- 6.9 eggs per week, meanwhile the constantan of B is 1.0-1.1 and k is 0.007-0.02 respectively for production data. By percent, showed that A (83-84%), B (0.02-0.06) and k (0.006-001), that all for partial and full data. Genetic correlation between parameter A and B is negative, which is 0.94–0.098 (production data) and 0.24-0.76 respectively. Positive correlation was resulted between B and k parameters. But for A and k parameters were both are positive and negative result. Estimation of persistency was 3.8 weeks for individual data and 3.7 weeks for collective data by partial (16 weeks) data set. And than the fulldata (52 weeks) are 4.0 and 4.1 respectively for collective and individual data set. Key words: White Mojosari duck, non-linear, production curve

The estimation of genetic distance and discriminant variables on breed of duck (Alabio, Bali, Khaki Campbell, Mojosari and Pegagan) by morphological analysis

A study on morphological body conformation of Alabio, Bali, Khaki Campbell, Mojosari and Pegagan ducks was carried out to determine the genetic distance and discriminant variables. This research was held in Research Institute for Animal Production, Ciawi, Bogor using 65 Alabio ducks, 40 Bali ducks, 36 Khaki Campbell ducks, 60 Mojosari ducks and 30 Pegagan ducks. Seven different body parts were measured, they were the length of femur, tibia, tarsometatarsus, the circumference of tarsometatarsus, the length of third digits, wing and maxilla. General Linear Models and simple discriminant analysis were used in this observation (SAS package program). Male and female Pegagan ducks had morphological size bigger than Alabio, Bali, Khaki Campbell and Mojosari ducks. Khaki Campbell ducks were mixed with Bali ducks (47.22%) and Pegagan ducks from isolated location in South Sumatera were lightly mixed with Alabio and Bali. Mahalanobis genetic distance showed that Bali and Khaki Campbell ducks, also, Alabio and Mojosari ducks had similarity, with genetic distance of 1.420 and 1.548, respectively. Results from canonical analysis showed that the most discriminant variables were obtained from the length of femur, tibia and third digits. Key words: Duck, genetic distance, morphological

Reciprocal crosses between Alabio and Mojosari ducks : early egg production

alternative to increasing productivity and production efficiency. Crossbreeding has been used widely for increasing productivity, and in this experiment Alabio and Mojosari ducks were crossed reciprocally in order to evaluate their egg production and egg quality, when compared to their parental breeds. Four genotypes AA, AM, MA and MM were compared and each consisted of 50 layer ducks. Each animal was kept in individual cage, and individual egg production and egg quality were recorded. They were given layer feed containing 20% crude protein and 3000 kcal/kg metabolisable energy. Results showed that the crossbred ducks (AM and MA) laid eggs earlier than their parental breeds (AA and MM), and MA laid earlier than AM. The weight of first eggs of the crossbreeds did not differ significantly, AM was the same as MM and MA the same as AA. For the body weight at first lay, MA and AM weighed in between their parental breeds, AA was the heaviest and then followed by MA, AM and MM. For the 3-month egg production, MA ducks laid significantly more egg than the other 3 genotypes, and this is related to the age of first lay being the earliest. In terms of egg quality such as egg weight, yolk color, weight and thickness of shell, weight of the white and HU value, the crossbred ducks showed quality in between AA and MM. The AM ducks tended to be closer to MM and MA closer to AA, and this shows a strong influence of maternal effects. The average level of heterosis for weight of first egg is 2.41% and for 3-month egg production is 2.1%, but for MA cross the heterosis level is 11.69% for egg production. The overall results showed that crossbreeds between Alabio and Mojosari have the potential in improving production traits in the effort to increase productivity and production efficiency. Reciprocal crossing between Tegal and Mojosari ducks : I . Early gowth and early egg production

In !ndonesia, duck farming plays an important role in meeting the demand for eggs and as an alternative source of income for small farmers . However, the production efficiency of duck farming is still very low because of small ownership and low quality of breeding stock . One way of improving the quality of breeding stock is by using crossbreds from various breeds of local ducks to exploit heterosis which may arise from the crossings . In order to test the performance of Tegal and Mojosari crossbreds, 250 ducks each of Mojosari and Tegal breeds were used and crossed to produce TT (Tegal x Tegal), TM (Tegal x Mojosari), MT (Mojosari x Tegal), and MM (Mojosari x Mojosari) . In this report, observations were taken only during the early gowth and the first egg laying. Results show that crossbreds between Mojosari did and Tegal not show any superiority to the parental breeds on young drakes at early gowth, while on young female ducks, the crossbred TM even showed a smaller body weight gain up to 8 weeks, although still similar to MM. The crossbred MT approached the performance of TT which is higher than TM or MM. This indicates a strong maternal effect, which was also confirmed by the findings on the age at first laying . In this early stage of gowth, there is no significant heterosis among the crossbreds. This confirms that these duck breeds can not be expected as meat producers . Observation of heterosis on components of egg production will describe more of the production potentials of these breeds. Keywords : Duck, crossbreeding
