On the influence of graphene oxide and hydroxyapatite modification on alginate-based hydrogel matrix: thermal, physicochemical, and biological considerations

Sodium alginate (SA) hydrogels with an addition of graphene oxide (GO) and hydroxyapatite (HAp) crosslinked by calcium chloride solution were investigated as potential materials for osteochondral tissue regeneration. The influence of various ratios of the nanoadditives in the natural derived polymer matrix on the thermal, physiochemical and biological properties was studied. Two thermal characterization methods (DSC and TGA) were employed to examine the thermal properties of the materials and provide information regarding the different types of water within the hydrogel structure. These parameters are crucial for the assessing and understanding of the adsorption/desorption processes in hydrogels and also impact their biocompatibility. The effect of GO and HAp addition on thermal characteristics of alginate hydrogel is reported, as well as the nanoadditives polymer chains interaction, as evidenced by FTIR results. The compression test confirmed that the nanoadditives, uniformly dispersed in the polymer matrix, improved the mechanical properties of the hydrogels, but only up to a certain content of additives. The composite hydrogels exhibited a very low friction coefficient. Both GO and HAp also enhanced chemical stability of alginate hydrogels under in vitro conditions. Biological assays demonstrated that most of the tested hydrogel extracts were not cytotoxic to hUC-MSCs, but they can affect the proliferation rate of the cells. Developed materials may present an intriguing alternative for osteochondral tissue regeneration.

Laser-printed paper ELISA and hydroxyapatite immobilization for colorimetric congenital anomalies screening in saliva

Alpha-fetoprotein (AFP) is a fetal protein that can indicate congenital anomalies such as Down syndrome and spinal canal blockage when detected at abnormal levels in pregnant women. Current AFP detection methods rely on invasive blood or serum samples, which require sophisticated equipment. From the many solutions proposed, colorimetric paper-based assays excel in point-of-care settings. The concept of paper-based ELISA (p-ELISA) enhances traditional methods, aligning with the ASSURED criteria for diagnostics in resource-limited regions. Despite success in microfluidic paper-based assay devices, laser printing remains underexplored for p-ELISA. Additionally, modifying the paper surface provides an additional layer of sensitivity enhancement. In this study, we developed a novel laser-printed paper-based ELISA (LP-pELISA) for rapid, sensitive, and noninvasive detection of AFP in saliva samples. The LP-pELISA platform was fabricated by printing hydrophobic barriers on filter paper using a laser printer, followed by depositing hydroxyapatite (HAp) as an immobilization material for the antibodies. The colorimetric detection was achieved using AuNPs functionalized with anti-AFP antibodies and silver nitrate enhancement. The LP-pELISA exhibited a linear response for AFP detection in both buffer and saliva samples over a range of 1.0-800ngmL-1, with a limit of detection (LOD) reaching 1.0ngmL-1. The assay also demonstrated good selectivity, repeatability, reproducibility, and stability. The LP-pELISA was further validated by testing spiked human saliva samples, showing its potential for point-of-care diagnosis of congenital disabilities. The LP-pELISA is a noninvasive platform showcasing simplicity, cost-effectiveness, and user-friendliness, utilizing laser printing, hydroxyapatite modification, and saliva samples to efficiently detect AFP. Beyond its application for AFP, this method's versatility extends to other biomarkers, positioning it as a catalyst for the evolution of paper-based biosensors. The LP-pELISA holds promise as a transformative tool for point-of-care diagnostics, fostering advancements in healthcare with its innovative technology.

Chitin scaffolds derived from the marine demosponge Aplysina fistularis stimulate the differentiation of dental pulp stem cells.

The use of stem cells for tissue regeneration is a prominent trend in regenerative medicine and tissue engineering. In particular, dental pulp stem cells (DPSCs) have garnered considerable attention. When exposed to specific conditions, DPSCs have the ability to differentiate into osteoblasts and odontoblasts. Scaffolds are critical for cell differentiation because they replicate the 3D microenvironment of the niche and enhance cell adhesion, migration, and differentiation. The purpose of this study is to present the biological responses of human DPSCs to a purified 3D chitin scaffold derived from the marine demosponge Aplysina fistularis and modified with hydroxyapatite (HAp). Responses examined included proliferation, adhesion, and differentiation. The control culture consisted of the human osteoblast cell line, hFOB 1.19. Electron microscopy was used to examine the ultrastructure of the cells (transmission electron microscopy) and the surface of the scaffold (scanning electron microscopy). Cell adhesion to the scaffolds was determined by neutral red and crystal violet staining methods. An alkaline phosphatase (ALP) assay was used for assessing osteoblast/odontoblast differentiation. We evaluated the expression of osteogenic marker genes by performing ddPCR for ALP, RUNX2, and SPP1 mRNA expression levels. The results show that the chitin biomaterial provides a favorable environment for DPSC and hFOB 1.19 cell adhesion and supports both cell proliferation and differentiation. The chitin scaffold, especially with HAp modification, isolated from A. fistularis can make a significant contribution to tissue engineering and regenerative medicine.

Recycling of phosphate tailings for an efficient hydroxyapatite-based adsorbent to immobilize heavy metal cations.

Hydroxyapatite (HAP) is a promising adsorbent for immobilizing heavy metals in soil and water. However, the preparation and modification of HAP from pure chemicals increases its cost and limits its large-scale practical application. In this study, a hydroxyapatite-based adsorbent (HAPPT) was prepared from phosphate tailing produced in the phosphorus industry to sequester Pb, Cd and Zn from solution. The results showed that HAPPT was composed of HAP and MgO, with a surface area of 27.74 m2/g. The kinetics studies showed that most Pb and Cd were removed from the initial solution in 4h and the adsorption of Zn increased with increasing contact time. Metals presented higher adsorption capacities at 35°C than that at 25°C. The adsorption isotherms showed that HAPPT presented high adsorption capacities for Pb, Cd and Zn in mono-metal solutions. The adsorption capacity of Cd at pH 6 was higher than that at pH 3, but the adsorption for Pb and Zn was similar at both pHs. HAPPT has selectivity for Pb in Pb-Cd-Zn multi-metals solution, and competitive adsorption reduced the adsorption quantity by 53%, 93% and 79% for Pb, Cd and Zn, respectively. The combined results of TEM-EDS, XRD and XPS showed that Pb was immobilized by forming phosphates due to the dissolution of HAP, whereas Cd and Zn were immobilized by forming hydroxide precipitates resulting from the function of MgO in HAPPT. The results of this study provided an efficient adsorbent for the removal of heavy metals in solution and provided a new perspective on the recycling of phosphate tailings in the phosphorus industry.

Synthesis of iron-substituted hydroxyapatite nanomaterials by co-precipitation method for defluoridation

The consumption of too much fluoride ions through drinking water can seriously harm human health. Thus fluoride ions need to be removed by the novel and efficient nanomaterials materials synthesized via eco-friendly method. The pure and iron-doped hydroxyapatites were synthesized using a simple co-precipitation technique for the removal of fluoride from water. The synthesized materials were characterized by advanced technical tools. The point of zero charge of the materials was determined by the salt addition method. Crystallite size and degree of crystallinity were observed to decrease with the substitution of calcium. However, the surface area and pore volume were found to have enhanced with modification of iron in the apatite. Batch adsorption experimental data were well fitted to pseudo-second order and Langmuir models, which implied that the sorption process is chemisorption through a monolayer on a homogenous surface. The maximum sorption capacities of HA and Fe-HA were found to be 40.46 and 83.86 mg g−1, respectively. The thermodynamic data revealed that the adsorption process is endothermic and spontaneous. The regeneration and reuse analysis insured that the materials have good potential for reuse. The adsorption mechanism was inferred as chemisorption through electrostatic interaction and ion exchange. The modification of hydroxyapatite using iron considered as a competent sorbent for the removal of fluoride ions.

Modification of hydroxyapatite (HA) powder by carboxymethyl chitosan (CMCS) for 3D printing bioceramic bone scaffolds

The poor mechanical properties of 3D printed HA bone scaffold is always a challenge in tissue engineering, to address this issue, carboxymethyl chitosan (CMCS) was proposed to modify HA bone scaffolds by a physical blending method in this research. A series of HA and HA/CMCS composite ceramic scaffolds were printed by using piezoelectric inkjet 3D printing technology, and their properties were investigated in terms of forming quality, structural morphology, mechanical properties, degradability, cytotoxicity, and cell adhesion growth. The results of forming quality and structural morphology show that with the increase of CMCS content, the forming quality of the samples deteriorated, the pore size and porosity increased. However, when the content of CMCS reached 5 wt%, obvious cracks appeared on the surface of the sample, and the forming quality was relatively poor. The mechanical testing results indicated the toughness of composites could be enhanced by incorporating CMCS into HA, which was attributed to the higher strength connections of the CMCS polymer network between HA particles and the stronger interaction between HA and CMCS molecules. FTIR spectra further revealed the strong hydrogen bonding interaction between CMCS and HA. Moreover, the degradation rate and mineralization ability of the sample increased with the content of CMCS, but the compressive strength during degradation increased with the CMCS content, indicating that incorporating CMCS into HA cannot only improve the mechanical property and biological activity of the scaffold but also makes up the defect of slow degradation of pure HA scaffold. Finally, the cytotoxicity, cell adhesion, and cell proliferation tests show that HA and HA/CMCS composite samples had good cytocompatibility, HA/CMCS sample with 3 wt% CMCS possessed the best bioactivity. In summary, HA/CMCS composite powder with 3 wt% CMCS content is the optimal matrix material for 3D printing bone scaffolds.

First-Principles Computational Study of the Modification Mechanism of Graphene/Graphene Oxide on Hydroxyapatite

Due to its unique crystal structure and nano-properties, hydroxyapatite (HA) has become an important inorganic material with broad development prospects in electrical materials, for fire resistance and insulation, and in bone repair. However, its application is limited to some extent because of its low strength, brittleness and other shortcomings. Graphene (G) and its derivative graphene oxide (GO) are well known for their excellent mechanical properties, and are widely used to modify HA by domestic and foreign scholars, who expect to achieve better reinforcement and toughening effects. However, the enhancement mechanism has not been made clear. Accordingly, in this study, G and GO were selected to modify HA using the first-principles calculation method to explore the theory of interfacial bonding of composites and explain the microscopic mechanism of interfacial bonding. First-principles calculation is a powerful tool used to solve experimental and theoretical problems and predict the structure and properties of new materials with precise control at the atomic level. Therefore, the bonding behaviors of hydroxyapatite (100), (110) and (111) crystal planes with G or GO were comprehensively and systematically studied using first-principles calculation; this included analyses of the density of states and differential charge density, and calculations of interfacial adhesion work and elastic moduli. Compared to HA (100) and (111) crystal planes, HA (110) had the best bonding performance with G and with GO, as revealed by the calculation results. The composite material systems of HA (110)/G and HA (110)/GO had the smallest density of states at the Fermi level, the largest charge transfers of Ca atoms, the largest interfacial adhesion work and the most outstanding elastic moduli. These results provide a theoretical basis for the modification of HA to a certain extent, and are beneficial to the expansion of the scope of its application.

Plasma-Sprayed Hydroxyapatite Coatings and Their Biological Properties

Air plasma spraying (APS) is a common method of producing hydroxyapatite (HA) coatings for alloprosthetic implants. Modification of HA spraying potentially may diminish the risk of inflammation and local infection during bone implantation. Titanium implants were HA coated with different deposition process parameters. HA powder was deposited using APS with axial powder injection at three different distances (100, 120, and 140 mm). The surface morphology of each implant was examined and the direct contact cytotoxicity of each HA coating was evaluated according to norm ISO 10993 5:2009. The response of monocytes to HA was assessed via the activation of transcription nuclear factor. All coatings had a lamellar structure. HA sprayed at a distance of 120 mm showed the highest roughness and little phase change. The analysis of the results of the conducted research showed that plasma-spraying distance during the HA coating process had a negligible impact on biocompatibility. The results obtained for a distance of 120 mm showed a slight increase in the biological properties tested. Moreover, HA coatings sprayed at different distances were not cytotoxic and did not stimulate the NF-kB. Bare titanium was less susceptible to colonization by Staphylococcus aureus than HA-coated surfaces. HA constitutes a potentially good, low-cost, non-cytotoxic material for joint prostheses.

The uptake performance and microscopic mechanism of inorganic-organic phosphorus hybrid amorphous hydroxyapatite for multiple heavy metal ions

Our previous studies have shown that the modification of hydroxyapatite with organic phosphonates can greatly improve the adsorption effect of hydroxyapatite on heavy metal ion pollutants. Hydroxyethylydene-1,1-diphosphonate (HEDP) is a widely used multi-purpose chemical agent with the advantage of lower-cost, but the preparation and application of hydroxyapatite composites doped by HEDP have not been reported. Therefore, in the present work, calcium nitrate and diammonium hydrogen phosphate were used as the calcium source and inorganic phosphorus source, respectively. 5%, 10%, 20% and 25% HEDP was added as an organophosphine source in the process of preparing hydroxyapatite （HAP）to obtain inorganic-organic phosphorus hybrid HAP composite materials with different HEDP doping amounts, respectively. XRD characterization showed that when HEDP doping was 20% of total phosphorus, the crystallinity of the resulting composite material was greatly reduced and became amorphous HAP, and the maximum adsorption capacity of the obtained amorphous HAP was 1893.7 mg/g, 239.3 mg/g, 285.4 mg/g, and 339.7 mg/g for Pb2+, Cu2+, Zn2+ and Cd2+, respectively, which was 8.47, 4.25, 3.29, 3.41 times that of the commercial HAP, respectively. Furthermore, Hirshfeld surface analysis specially including fingerprint plot was applied to investigate the interaction between HEDP and HAP, and reduced density gradient (RDG) method was carried out to explore the interaction mechanism of HEDP doping amorphous HAP composite and heavy metal ions. Because of the low price of HEDP, the simple and low-consumption preparation method and the excellent removal effect to various heavy metal pollutants, the inorganic-organic phosphorus hybrid amorphous HEDP-HAP composite material will have broad application prospects in the field of heavy metal pollution control.

Effects of Incorporation of Marine Derived Hydroxyapatite on the Microhardness, Surface Roughness, and Fluoride Release of Two Glass-Ionomer Cements

Background: The aim of this study was to evaluate the effects of incorporation of hydroxyapatite (HA) derived from cuttlefish bone on the microhardness, surface roughness (SR), and fluoride release (FR) of conventional cure, and resin-modified glass-ionomer cement. Methods: There were four groups for each tested material; experimental glass-ionomer were made by addition and of 2, 5, and 10 wt % HA respectively to conventional glass-ionomers Fuji II LC and Fuji IX GP Extra. One group was prepared without the addition of HA particles. For SR and microhardness measurements sectional Teflon molds (5 mm in diameter and 2 mm deep) were used to prepare 10 samples per group (n = 80). The samples were stored in distilled water at 37 °C for 7 days prior to testing. The SR was measured using a contact type profilometer and the microhardness was determined using a Vickers micro-hardness tester at a load of 980 g for 15 s. For FR measurements, there were six samples per group (n = 48), prepared in Teflon molds (8 mm in diameter and 2 mm deep). The FR was measured with an ionoselective electrode in triplicates after 24 h, 7 days, and 45 days. Statistical analysis was performed using one-way ANOVA with Tukey post-hoc test. Results and Conclusion: Microhardness values obtained for Fuji II modified with 10 wt % HA were significantly higher compared to the other two groups tested. Comparison of materials with respect to SR showed significant difference between them (p < 0.0001) with Fuji II and Fuji IX modified with HA having higher SR values. Regarding FR, Fuji IX showed statistically significant higher results than Fuji II, independently of HA modification, and groups modified with 2 and 5 wt % HA showed significantly increased fluoride release in all three time points.

Effect of Silica Addition on Mechanical Properties of Eggshell-Derived Hydroxyapatite

Eggshell is a solid waste that is available in abundance but is being left unused. Eggshell containing calcium in a high amount. Calcium can be used as a precursor for hydroxyapatite (HAp). Modification of HAp with SiO2 is expected to improve its low mechanical properties for biomedical applications. In this study, HAp is synthesized from the eggshell. Then, it was modified by adding SiO2 utilizing the coprecipitation method with concentrations of 10%, 20%, 30%, and 40%, respectively. The HAp and HAp/SiO2 were characterized using; X-ray diffraction and Fourier transform infrared spectroscopy. The analysis HAp and HAp/SiO2 were density, compressive strength, and hardness. The best mechanical properties of HAp/SiO2 were characterized using SEM-EDS. The HAp were prepared successfully with a ratio of Ca/P was 1.673, close to the theoretical 1.67. The addition of SiO2 caused a decrease in crystallite size and density but increased compressive strength and hardness. The best mechanical properties of HAp/SiO2 were obtained with SiO2 of 30% and 40% with similar values.

Hydroxyapatite-modified micro/nanostructured titania surfaces with different crystalline phases for osteoblast regulation

Surface structures and physicochemical properties critically influence osseointegration of titanium (Ti) implants. Previous studies have shown that the surface with both micro- and nanoscale roughness may provide multiple features comparable to cell dimensions and thus efficiently regulate cell-material interaction. However, less attention has been made to further optimize the physicochemical properties (e.g., crystalline phase) and to further improve the bioactivity of micro/nanostructured surfaces. Herein, micro/nanostructured titania surfaces with different crystalline phases (amorphous, anatase and anatase/rutile) were prepared and hydroxyapatite (HA) nanorods were deposited onto the as-prepared surfaces by a spin-assisted layer-by-layer assembly method without greatly altering the initial multi-scale morphology and wettability. The effects of crystalline phase, chemical composition and wettability on osteoblast response were investigated. It is noted that all the micro/nanostructured surfaces with/without HA modification presented superamphiphilic. The activities of MC3T3-E1 cells suggested that the proliferation trend on the micro/nanostructured surfaces was greatly influenced by different crystalline phases, and the highest proliferation rate was obtained on the anatase/rutile surface, followed by the anatase; but the cell differentiation and extracellular matrix mineralization were almost the same among them. After ultrathin HA modification on the micro/nanostructured surfaces with different crystalline phases, it exhibited similar proliferation trend as the original surfaces; however, the cell differentiation and extracellular matrix mineralization were significantly improved. The results indicate that the introduction of ultrathin HA to the micro/nanostructured surfaces with optimized crystalline phase benefits cell proliferation, differentiation and maturation, which suggests a favorable biomimetic microenvironment and provides the potential for enhanced implant osseointegration in vivo.

Synthesis and characterization of injectable self-healing hydrogels based on oxidized alginate-hybrid-hydroxyapatite nanoparticles and carboxymethyl chitosan

Injectable hydrogels are of great interest in tissue engineering, and those incorporating hydroxyapatite (HA) are especially acclaimed in the application of bone repair. Synthetic micro-HA were generally used for this purpose and in some cases, surface modification of HA was further applied to improve the interfacial compatibility of rigid inorganic HA with soft organic matrix. In this study, the injectable hydrogels based on oxidized alginate hybrid HA nanoparticles and carboxymethyl chitosan were achieved via Schiff base reaction. Physicochemical characterization confirmed that oxidized HA/Alg hybrids (OHAH) were successfully prepared. Rheological measurements verified the formation of hydrogels based on the dynamic imine bonding, and the gelation time showed a negative correlation to the concentration and oxidation time of OHAH, while the storage moduli exhibited a positive correlation. The self-healing property of these hydrogels was validated by the splicing experiments and rheological experiments. The lyophilized hydrogels showed porous structures with numerous HA nanoparticles distributed on the surface of pore wall. MTT assays and live/dead staining of cell experiments confirmed the cytocompatibility of these hydrogels. The injectable hydrogels with self-healing and tunable gelling properties were ingeniously prepared with functionalized alginate-mediated HA hybrid nanoparticles, and these hydrogels are promising for applications in bone tissue engineering.

Sorption of bismuth(iii) and its chloride complexes with 2-aminopyrimidine cation on hydroxyapatite of various textures

Specific features of the interaction between bismuth(iii) and hydroxyapatite (HAP) of various morphologies during its sorption and co-crystallization binding were revealed. The obtained sorption isotherms nearly coincide with each other regardless of the HAP type used and cannot be described in terms of the Langmuir or Freundlich models. Bismuth can form the intrinsic phase of bismuth phosphate due to the chemical or topochemical reaction with HAP when the sorption method is used. In the case of co-crystallization binding of bismuth ions, the morphological modification of HAP occurs. The bismuth complexes with aminopyrimidine in neutral and weakly acidic solutions are readily hydrolyzed to form a precipitate, and no binding with HAP occurs.

A review on adsorptive removal of dyes from wastewater by hydroxyapatite nanocomposites.

Dye removal from wastewater is of prominence due to its hostile effects on human health and the environment. The complex structure of the dye molecule is responsible for its difficulty in removal. Adsorption is found to be a promising technique to eliminate dye wastes due to its high removal capacity at low concentration. Among different adsorbents used, hydroxyapatite is a biocompatible adsorbent that is relatively efficient in both anionic and cationic dye removal. Recently, modification of hydroxyapatite by doping with other materials to increase its removal efficiency has gained much attention. This review summarizes compilation of recent literature on the removal of anionic and cationic dye by different hydroxyapatite nanocomposites, comparison of adsorption capacities of different hydroxyapatite nanocomposites, the possible adsorption mechanism of removal of dyes, the general isotherm, and kinetic and thermodynamic studies explaining the type of adsorption and the characteristics, advantages, and limitations of adsorbents.

Nano-Structure Designing Promotion Osseointegration of Hydroxyapatite Coated Ti-6Al-4V Alloy Implants in Diabetic Model.

Mammalian diabetes mellitus which contains altered microenvironment always impairs diverse cellular processes such as osteogenesis, angiogenesis and tissue regeneration via different mechanisms. For researches in materials science, modifying the ability of osteogenesis and angiogenesis in dental implants shows its significant importance. Nano-structure designing is considered as a facile strategy to improve the surface bioactivity of the implants. In this study, the nanorod-structured hydroxyapatite (HA) coatings on Ti-6Al-4V implants were facilely designed by the combination of plasma-spraying and hydrothermal treatment via varying reaction media. Intriguingly, hydrothermal treatment eliminated the glassy phase and impurity phases of HA coatings, and nanorod-structured surface was successfully constructed under hydrothermal treatment in Na₃PO₄ solution. Additionally, the HA coatings with nanorod-structured surface effectively promoted the adhesion and proliferation and further enhanced osteogenic differentiation of DM-rBMSCs in vitro. Moreover, the osseointegration of Ti-6Al-4V implants with nanorod-structured HA coating was also enhanced in diabetes mellitus rabbit model in vivo. Therefore, the nano-structured surface modification of HA coating on Ti-6Al-4V implants could target pathological bone loss via strengthening osteogenesis and angiogenesis and further potentially used as a therapeutic coating to promote diabetic osteointegration.

Synergized poly(lactic acid)–hydroxyapatite composites: Biocompatibility study

ABSTRACT In vitro biocompatibility of impact modified composites produced from poly(lactic acid) (PLA) and hydroxyapatite (HA) is reported in this study. Surface modification was previously used to facilitate the dispersion of HA in PLA, whereas impact property of the PLA‐HA composites was deliberately enhanced as it was necessary. Herein, osteoblast cell culture assay was used to assess the possible effects of HA surface modification and impact modification on the cell behavior in physiological media. Furthermore, antimicrobial properties of the HA were assessed. Evidence of HA modification was confirmed through elemental and spectroscopic analysis. Incorporation of HA offered better cell attachment and proliferation to the PLA matrix, with significant increase in the cell viability (%). Also, modification of HA did not present obvious cytotoxicity to the PLA‐HA composite. Conversely, incorporation of impact modifier slowed down the rate of cell proliferation on the composite surface but facilitates increased wettability. © 2018 Wiley Periodicals, Inc. J. Appl. Polym.
