Comparative Assessment of the Anti-Helicobacter pylori Activity and Gastroprotective Effects of Three Herbal Formulas for Functional Dyspepsia In Vitro

Helicobacter pylori has been implicated in various gastrointestinal disorders, including functional dyspepsia. This study aimed to compare the anti-H. pylori activity and gastroprotective effects of three typical herbal formulas used for gastrointestinal disorders in Korea: Shihosogan-tang (ST), Yijung-tang (YT), and Pyeongwi-san (PS). Firstly, we assessed the total phenolic and flavonoid contents, as well as the antioxidative capacity. Additionally, we evaluated the antibacterial effect on H. pylori using an ammonia assay, minimum inhibitory concentration (MIC) test, and the disk agar diffusion method. Furthermore, we examined alterations in the gene expression of tight junction proteins, pro-inflammatory cytokines, and cellular vacuolation using an AGS cell model infected with H. pylori. While ST exhibited a higher total phenolic content, superior free radical scavenging, and inhibition of H. pylori compared to YT and PS, YT more evidently inhibited gastric cellular morphological changes such as vacuolation. All formulations significantly ameliorated changes in inflammatory and gastric inflammation-related genes and cellular morphological alterations induced by H. pylori infection. Overall, the present in vitro study suggests that all three herbal formulas possess potential for ameliorating gastrointestinal disorders, with ST relatively excelling in inhibiting H. pylori infection and inflammation, while YT potentially shows greater efficacy in directly protecting the gastric mucosa.

Synthesis of Green Brucite [NixMg1−x(OH)2] by Incorporation of Nickel Ions in the Periclase Phase (MgO) Applied as Pigments

Magnesium oxide is typically white and can be colorized with transition metal insertion by doping. We present the preparation of a green-colored hydroxide by the exchange of Mg2+ on the crystalline lattice with Ni2+ in MgO, using three nickel salts. MgOst was prepared by the colloidal starch suspension method, using cassava starch. The oxides and hydroxides, before and after, were characterized by X-ray diffraction (XRD), and show that a phase change occurs: a transition from periclase (MgO) to brucite (Mg(OH)2) due to the incorporation of nickel ions from different salts (acetate, chloride, and nitrate), resulting in the solid solution [NixMg1−x(OH)2]. The FTIR spectrum corroborates the crystallographic structure identified through XRD patterns, confirming the formation of a crystal structure resembling brucite. The new samples present a green color, indicative of the incorporation of the Ni2+ ions. The antimicrobial activity of products resulting from the doping of magnesium oxide with nickel and the precursor MgOst was assessed through the minimum inhibitory concentration (MIC) test. The evaluation included three bacterial strains: Staphylococcus aureus (ATCC 25923), Escherichia coli (ATCC 25922), Salmonella gallinarum (ATCC 9184), and a yeast strain, Candida albicans (ATCC 10231). The obtained results were promising; the tested samples exhibited antimicrobial activity, with a MIC ranging from 0.312 to 0.625 μg.μL−1. The nickel compound, derived from the precursor chloride salt, demonstrated superior MIC activity. Notably, all tested samples displayed bactericidal activity against the S. aureus strain and exhibited a broad spectrum of inhibition, encompassing both Gram-positive and Gram-negative strains. Only the nickel compounds derived from precursors with acetate and nitrate anions demonstrated antimicrobial activity against C. albicans, exhibiting a fungistatic behavior. Based on the conducted studies, [NixMg1−x(OH)2] has emerged as a promising antimicrobial agent, suitable for applications requiring the delay or inhibition of bacterial growth.

Quorum-quenching enzyme Est816 assisted antibiotics against periodontitis induced by Aggregatibacter actinomycetemcomitans in rats.

Quorum-quenching enzyme Est816 hydrolyzes the lactone rings of N-acyl homoserine lactones, effectively blocking the biofilm formation and development of Gram-negative bacteria. However, its applications in the oral field is limited. This study aimed to evaluate the efficacy of enzyme Est816 in combination with antibiotics against periodontitis induced by Aggregatibacter actinomycetemcomitans in vitro and in vivo. The antimicrobial efficacy of enzyme Est816 in combination with minocycline, metronidazole, and amoxicillin was determined using the minimum inhibitory concentration test. The anti-biofilm effect of enzyme Est816 was assessed using scanning electron microscopy, live/dead bacterial staining, crystal violet staining, and real-time quantitative PCR. Biocompatibility of enzyme Est816 was assessed in human gingival fibroblasts (HGF) by staining. A rat model of periodontitis was established to evaluate the effect of enzyme Est816 combined with minocycline using micro-computed tomography and histological staining. Compared to minocycline, metronidazole, and amoxicillin treatment alone, simultaneous treatment with enzyme Est816 increased the sensitivity of biofilm bacteria to antibiotics. Enzyme Est816 with minocycline exhibited the highest rate of biofilm clearance and high biocompatibility. Moreover, the combination of enzyme Est816 with antibiotics improved the antibiofilm effects of the antibiotics synergistically, reducing the expression of the virulence factor leukotoxin gene (ltxA) and fimbria-associated gene (rcpA). Likewise, the combination of enzyme Est816 with minocycline exhibited a remarkable inhibitory effect on bone resorption and inflammation damage in a rat model of periodontitis. The combination of enzyme Est816 with antibiotics represents a prospective anti-biofilm strategy with the potential to treat periodontitis.

Inhibitory effects of alkyl‐galactosides on the coaggregation of Fusobacterium nucleatum and oral bacteria

AbstractPurposeCoaggregation and coadhesion are important mechanisms for oral bacteria to colonize the oral cavity and exert pathogenic effects. Fusobacterium nucleatum has been reported to coaggregate with various oral bacteria. Some of the coaggregation are inhibited by D‐galactose. D‐galactose, however, will be metabolized by oral bacteria when applied to the oral cavity, resulting in the acid generation that causes dental caries. In the present study, we developed a novel material, alkyl‐galactosides, that inhibits the coaggregation of F. nucleatum and is not metabolized by oral bacteria.MethodsAcid production from alkyl‐galactoside by Streptococcus mutans was determined by pH lowering during liquid culture. Antibacterial activity was measured using the minimum inhibitory concentration test, and coaggregation inhibitory effects were evaluated based on the relative coaggregation ratio between F. nucleatum and several oral bacteria using absorbance measurements.ResultsD‐galactose was rapidly metabolized by S. mutans, resulting in a decreased pH. Alkyl‐galactoside, however, was not metabolized by the bacterium at 1 mM. Minimum inhibitory concentration of alkyl‐galactoside against S. mutans is 1–4 mM. Alkyl‐galactoside inhibited coaggregation between F. nucleatum and several oral bacteria almost to the same extent as D‐galactose.ConclusionsAlkyl‐galactoside inhibits coaggregation between F. nucleatum and oral bacteria and is not assimilated by oral bacteria suggesting to be a potent novel material for prevention of dental plaque formation.

Green synthesis of ZnO/catechin nanocomposite: Comprehensive characterization, optical study, computational analysis, biological applications and molecular docking

Herein, a green and cost-efficient approach was employed to develop the ZnO/Catechin nanocomposite. Margosa leaf extract was utilized as a reducing and capping agent in the fabrication of ZnONPs. Catechin extracted from green tea was employed to modify the ZnONPs, and various conjugation strategies between ZnONPs and extracted catechin were explored. Comprehensive characterization techniques including SEM, elemental mapping, EDX, UV–vis spectroscopy, FT-IR, and XRD were employed to validate the successful formation of the ZnO/Catechin nanocomposite, revealing an average crystal size of 49.37 nm. In vitro antimicrobial assessment was conducted on green-synthesized ZnONPs (A), extracted catechin (B), and the nanocomposite (C) against both gram-positive bacteria (Bacillus cereus, Staphylococcus aureus) and gram-negative bacteria (Escherichia coli, Pseudomonas aeruginosa). Minimum inhibitory concentration (MIC) testing highlighted the robust inhibitory effects of the nanocomposite (C) at notably lower concentrations (10–60 μg/mL), surpassing the efficacy of precursor compounds (A and B), which necessitated higher concentrations (20–600 μg/mL) for moderate effectiveness. Components (A-C) exhibited enhanced antibacterial efficacy, yielding inhibitory zones ranging from 9 ± 0.01 to 13 ± 0.22 mm against the tested pathogens, outperforming Azithromycin as a positive control. Exploration of Frontier molecular orbitals (FMOs) unveiled smaller band gap energies (Eg) of 1.86, 0.49, and 2.14 eV for the prepared nanocomposites (C1-3) in contrast to precursor compounds (A = 4.04 eV, B = 4.03 eV).

Antibacterial Activity of Ethanol, Ethyl Acetate, and n-Hexane Extracts of Bidara Laut Lignum (Strychnos lucida R.Br) against Staphylococcus aureus and Escherichia coli

Strychnos lucida is empirically believed to have efficacy in treating various diseases such as fever, malaria, toothache, and stomachache and to treat wounds. The secondary metabolite of Strychnos lucida has various polarity. The purpose of this study was to compare the inhibitory activity of Strychnos lucida extracts against E. coli and S. aureus bacteria. The test was carried out by measuring the Diameter of the Inhibitory Region (DIR) using the disc diffusion method on a concentration of 90%, 75%, 60%, 45%, 30%, and 15%. The Minimum Inhibitory Concentration (MIC) was carried out by agar dilution method on a concentration of 15%, 12.5%, 10%, 7.5%, 5%, and 2.5%. The positive control was 1% ampicillin, and the negative control was 10% DMSO. The ethyl acetate extract showed the highest DIR of 14.30 mm against S. aureus and E. coli on a concentration of 45%, ethanol extract was 8,38 mm against S. aureus and 8.71 mm against E. coli, and n-hexane had no inhibitory activity on a concentration 45%. The result of the MIC test for ethanol and ethyl acetate extract for both bacteria was found at a concentration of 15%, while n-hexane extract against Staphylococcus aureus was found at a concentration of 90% and a concentration of 75% against Escherichia coli.

Cefiderocol – An effective antimicrobial for MDR infections but a challenge for routine antimicrobial susceptibility testing

PurposeCefiderocol is a novel cephalosporin–siderophore conjugate antibiotic that holds promise to thwart infections caused by multi-drug-resistant gram-negative bacilli. Its antibacterial activity against normally susceptible species is not affected by most β-lactamases, including metallo-β-lactamases. Due to the siderophore-mediated entry into the cell, the activity of cefiderocol is less affected by porin loss or active efflux resistance than many other β-lactam antibiotics. The aim of this study was to assess in vitro susceptibility to the cefiderocol of carbapenemase-producing gram-negative bacilli from clinical samples of hospitalized patients. Materials and methodsWe analyzed 102 clinical strains of carbapenemase-producing Enterobacterales and non-fermentives from hospital centers in Łódź, Poland. Antimicrobial susceptibility to cefiderocol was tested by the minimum inhibitory concentration test strips and disc diffusion methods. ResultsThe obtained results turned out to be ambiguous, and the area of technical uncertainty made their interpretation very difficult. ConclusionsThe cost of therapy with this antibiotic, and difficulties in interpreting the drug susceptibility are the limitations to the use of cefiderocol. Intensive work should be carried out to finally standardize an easily accessible and reliable method for the determination of susceptibility to cefiderocol.

Synthesis, characterization, and investigation of antibacterial activity of Novel CMC/CuO NPs/CQDs bionanocomposite coating

In recent decades, healthcare-associated infections (HAIs) have become a common problem in healthcare facilities such as hospitals. As a result, researchers are currently developing nanocomposite coatings that are strengthened with antibacterial nanoparticles. In this research, a novel antibacterial bionanocomposite coating based on carboxymethyl cellulose polymer/copper oxide nanoparticles/carbon quantum dots was coated on medical grade 316 stainless steel by sol-gel dip-coating method. The effect of the concentration of nanocomposite components was investigated at four different levels to determine the best ratio with the most antibacterial activity. Structural characteristics of nanocomposite and coating were investigated using different analysis methods. The coating analysis showed that reinforcements are uniformly distributed in the polymer matrix. Antibacterial test of disc diffusion was performed by the Kirby-Bauer method and minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) antibacterial test. The results showed that bionanocomposite was effective in the MIC assays against Staphylococcus aureus and Escherichia coli with MIC values of 25 mg/ml and >50 mg/ml, respectively. The inhibition zones for E. coli and S. aureus were 17 and 32 mm, respectively, at 10 μg/disc of gentamicin. SEM images displayed significant and evident alterations in the structure of bacterial morphology, indicating cellular damage.

A challenging case of carbapenem resistant Klebsiella pneumoniae-related pyogenic liver abscess with capsular polysaccharide hyperproduction: a case report

BackgroundCarbapenem-resistant Klebsiella pneumoniae (CRKP) infections are a major public health problem, necessitating the administration of polymyxin E (colistin) as a last-line antibiotic. Meanwhile, the mortality rate associated with colistin-resistant K. pneumoniae infections is seriously increasing. On the other hand, importance of administration of carbapenems in promoting colistin resistance in K. pneumoniae is unknown.Case presentationWe report a case of K. pneumoniae-related pyogenic liver abscess in which susceptible K. pneumoniae transformed into carbapenem- and colistin-resistant K. pneumoniae during treatment with imipenem. The case of pyogenic liver abscess was a 50-year-old man with diabetes and liver transplant who was admitted to Abu Ali Sina Hospital in Shiraz. The K. pneumoniae isolate responsible for community-acquired pyogenic liver abscess was isolated and identified. The K. pneumoniae isolate was sensitive to all tested antibiotics except ampicillin in the antimicrobial susceptibility test and was identified as a non-K1/K2 classical K. pneumoniae (cKp) strain. Multilocus sequence typing (MLST) identified the isolate as sequence type 54 (ST54). Based on the patient’s request, he was discharged to continue treatment at another center. After two months, he was readmitted due to fever and progressive constitutional symptoms. During treatment with imipenem, the strain acquired blaOXA−48 and showed resistance to carbapenems and was identified as a multidrug resistant (MDR) strain. The minimum inhibitory concentration (MIC) test for colistin was performed by broth microdilution method and the strain was sensitive to colistin (MIC < 2 µg/mL). Meanwhile, on blood agar, the colonies had a sticky consistency and adhered to the culture medium (sticky mucoviscous colonies). Quantitative real-time PCR and biofilm formation assay revealed that the CRKP strain increased capsule wzi gene expression and produced slime in response to imipenem. Finally, K. pneumoniae-related pyogenic liver abscess with resistance to a wide range of antibiotics, including the last-line antibiotics colistin and tigecycline, led to sepsis and death.ConclusionsBased on this information, can we have a theoretical hypothesis that imipenem is a promoter of resistance to carbapenems and colistin in K. pneumoniae? This needs more attention.

Effectiveness of Graptophyllum pictum L. Griff Extract as a Complementary Therapy for Peri-Implantitis: An In Vitro and In Silico Study

Background: Dental peri-implantitis is an inflammation that occurs in the peri-implant soft tissue and is accompanied by bone resorption, decreased osseointegration, and pockets. Graptophyllum pictum L. Griff extract may act as complementary therapy for peri-implantitis. Objectives: To investigate the effectiveness of G. pictum L. Griff extract as a complementary therapy for peri-implantitis. Materials and Methods: A phytochemical test was performed to determine the active compounds using each compound's reagents. Antioxidant assay to examine the antioxidant activity compared with vitamin C. Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) tests were used to investigate the antibacterial activity of Aggregatibacter actinomycetemcomitans (Aa), Porphyromonas gingivalis (Pg), Fusobacterium nucleatum (Fn), and Provotella intermedia (Pi) as peri-implantitis-related bacteria. Molecular docking is an in-silico test to visualize the interaction of G. pictum L. Griff molecules with targeted proteins using several software programs and databases. This study was analyzed by parametric and non-parametric variance analysis and continued with a post-hoc test. Results: G. pictum L. Griff extract contained flavonoids, alkaloids, saponins, tannins, steroids, and terpenoids. The antibacterial effect of G. pictum L. Griff can inhibit the growth of dental peri-implantitis-related bacteria such as Aa, Pg, Pi, and Fn with a minimum 25% extract concentration. In silico, the interaction of ethylcholest and proteins shows possible pharmalogical effects such as anti-inflammatory, antibacterial, and antioxidant activity. Conclusion: G. pictum L. Griff extract possesses good antibacterial, antioxidant, and anti-inflammatory activity that may be effective for dental peri-implantitis complementary therapy as documented in silico and in vitro.

Comparison of IncK-blaCMY-2 Plasmids in Extended-Spectrum Cephalosporin-Resistant Escherichia coli Isolated from Poultry and Humans in Denmark, Finland, and Germany.

Escherichia coli carrying IncK-blaCMY-2 plasmids mediating resistance to extended-spectrum cephalosporins (ESC) has been frequently described in food-producing animals and in humans. This study aimed to characterize IncK-blaCMY-2-positive ESC-resistant E. coli isolates from poultry production systems in Denmark, Finland, and Germany, as well as from Danish human blood infections, and further compare their plasmids. Whole-genome sequencing (Illumina) of all isolates (n = 46) confirmed the presence of the blaCMY-2 gene. Minimum inhibitory concentration (MIC) testing revealed a resistant phenotype to cefotaxime as well as resistance to ≥3 antibiotic classes. Conjugative transfer of the blaCMY-2 gene confirmed the resistance being on mobile plasmids. Pangenome analysis showed only one-third of the genes being in the core with the remainder being in the large accessory gene pool. Single nucleotide polymorphism (SNP) analysis on sequence type (ST) 429 and 1286 isolates showed between 0-60 and 13-90 SNP differences, respectively, indicating vertical transmission of closely related clones in the poultry production, including among Danish, Finnish, and German ST429 isolates. A comparison of 22 ST429 isolates from this study with 80 ST429 isolates in Enterobase revealed the widespread geographical occurrence of related isolates associated with poultry production. Long-read sequencing of a representative subset of isolates (n = 28) allowed further characterization and comparison of the IncK-blaCMY-2 plasmids with publicly available plasmid sequences. This analysis revealed the presence of highly similar plasmids in ESC-resistant E. coli from Denmark, Finland, and Germany pointing to the existence of common sources. Moreover, the analysis presented evidence of global plasmid transmission and evolution. Lastly, our results indicate that IncK-blaCMY-2 plasmids and their carriers had been circulating in the Danish production chain with an associated risk of spreading to humans, as exemplified by the similarity of the clinical ST429 isolate to poultry isolates. Its persistence may be driven by co-selection since most IncK-blaCMY-2 plasmids harbor resistance factors to drugs used in veterinary medicine.

Characterization of Antimicrobial Activities of Bifidobacterium lactis BB-12 and Their Inhibitory Effect Against Some Foodborne Pathogens.

Bifidobacterium animalis subsp. lactis BB-12, a probiotic, has shown potential to promote health benefits and control pathogens. This study aimed to investigate the effectiveness of BB-12 and its cell-free supernatant (CFS) in inhibiting the growth of Listeria monocytogenes and Salmonella enterica serovar Typhimurium. To assess the antimicrobial activity of BB-12, agar well diffusion, disk diffusion, and minimum inhibitory concentration (MIC) tests were conducted. The bicinchoninic acid (BCA) assay was performed to measure the protein concentration in CFS. The study's results indicated that the BB-12 strain inhibited the pathogens' growth. The disk diffusion test using BB-12 showed inhibitory results ranging from 11 to 14 mm for both bacteria. The agar well diffusion test reported the zone of inhibition ranging from 11.6 to 16 mm for both bacteria. The MIC test was conducted as a confirmatory test, which demonstrated the highest inhibitory zone using 2 McFarland (6 × 108 CFU/mL) concentrations of probiotics on L. monocytogenes (44.98%) and S. Typhimurium (66.41%). The disk diffusion test revealed that the probiotic CFS had a significant inhibitory impact on S. Typhimurium with a 16.6 mm zone of inhibition. The BCA test findings indicated that the 24- and 48-h CFSs exhibited inhibitory properties against infections. Notably, the 24-h CFS, including a protein level of 78.47 μg/mL, demonstrated a more pronounced inhibitory impact on both pathogens. The findings highlight that utilizing the BB-12 strain and its CFS can serve as a viable approach to battle infections, enhancing food safety and public health.

Occurrence, virulence, and resistance genes in Salmonella enterica isolated from an integrated poultry company in Jordan

Salmonella is considered one of the most common foodborne pathogens worldwide. The annual number of hospitalizations and deaths related to zoonotic salmonellosis, which is transmitted from animals to humans and infects poultry and meat, is expected to be significant. Hence, the primary aims of this research were to isolate and characterize Salmonella species obtained from an integrated poultry company and identify some virulence, and antimicrobial resistance, with a specific concern about colistin resistance genes. A total of 635 samples collected from various sources in an integrated company in Jordan were screened for Salmonella species accompanying their virulence and antimicrobial resistance genes. Samples were collected from parent stock house drag swabs, broiler farms, premix, cecum at the slaughterhouse level, pre-chilling and post-chilling stages, and the final product. Salmonella species were detected in 3% (6\\200) of investigated parent stock house drag swabs, 13.8% (11/80) from cloacal swabs from broiler farms, 16.9% (11/65) from boiler farms premix, 24.4% (11/45) from the cecum at slaughterhouse level, 16.4% (9/55) from the pre-chilling stage, 37.8% (17/45) from the post-chilling stage and 53.3% (24/45) from the final product stage. No isolates were detected in feed mills (0/20), parents' premix (0/40), or hatcheries (0/40). Salmonella isolates were resistant to ciprofloxacin (91.0%), nalidixic acid (86.5%), doxycycline (83.1%), tetracycline (83.1%), sulphamethoxazole-trimethoprim (79.8%) and ampicillin (76.4%). Serotyping shows that S. Infantis was the predominant serovar, with 56.2%. Based on the minimum inhibitory concentration (MIC) test, 39.3% (35/89) of the isolates were resistant to colistin; however, no mcr genes were detected. Among antimicrobial-resistant genes, blaTEM was the most prevalent (88.8%). Furthermore, the spvC, ompA, and ompF virulence genes showed the highest percentages (97.8%, 97.8%, and 96.6%, respectively). In conclusion, Salmonella isolates were found at various stages in the integrated company. S. Infantis was the most prevalent serotype. No mcr genes were detected. Cross-contamination between poultry production stages highlights the importance of good hygiene practices. Furthermore, the presence of virulence genes and the patterns of antimicrobial resistance present significant challenges for public health.

Green Synthesis, Characterization, and Evaluation of the Antimicrobial Properties and Compressive Strength of Hydroxyapatite Nanoparticle-Incorporated Glass Ionomer Cement.

Background Glass ionomer cement (GIC) plays a vital role in dental restorative procedures, serving purposes such as filling, luting, and adhesion. However, its inadequate mechanical properties pose challenges, especially in areas experiencing significant stress. To overcome this limitation, nanohydroxyapatite (nHA), known for its bioactive phosphate content, is added to the GIC at specific concentrations to improve its properties. Aim We aim to evaluate the antimicrobial property and compressive strength of green-mediated nHA-incorporated GIC. Material and methods Green synthesis of hydroxyapatite nanoparticles was prepared using Moringa oleifera extract in a solvent form and eggshell waste served as the calcium source. These nHApowders were then integrated into the GICat varying concentrations (3%, 5%, and 10%) designated as Group I, Group II, and Group III, respectively, while Group IV (control) consisted of conventional GIC. Specimens were fabricated and subjected to chemical structure analysis through Fourier transform infrared spectroscopy (FTIR), energy dispersive X-ray (EDX), and scanning electron microscopy (SEM). The antimicrobial activity and compressive strength of all groups were investigated. The antimicrobial activity against Streptococcus mutans and Lactobacillus was evaluated through the minimum inhibitory concentration (MIC) test, while compressive strength was evaluated by measuring the maximum force endured by the specimen before fracturing. Data analysis utilized IBM SPSS Statistics software, employing repeated measures ANOVA to determine mean MIC values and compressive strength, with Tukey's posthoc test for pairwise comparisons. Results The results of the study showed that the antimicrobial efficacy of nHA GIC improved with increasing weight percent (% wt) of the additive, exhibiting significantly enhanced activity against Streptococcus mutans and Lactobacillus compared to the control group (Group IV) with statistical significance (p < 0.05). Moreover, the compressive strength exhibited notable enhancements in the modified groups, including Group I (172.55 ± 0.76), Group II (178.16 ± 0.760), and Group III (182.45 ± 0.950), when compared to the control (162.46 ± 1.606), with statistically significant differences (p < 0.05). Conclusion The study demonstrates that the incorporation of green-mediated nHA-containing GIC results in superior antimicrobial efficacy and compressive strength compared to the control group (Group IV). In particular, the highest concentration of nHA-modified GIC (10%) exhibited the most favorable antimicrobial properties along with increased strength. Therefore, utilizing green-mediated nHA in the GIC shows promise as an effective restorative material. Future investigations should delve into the molecular chemistry and bonding mechanisms to further explore its potential.

Phytochemical screening and investigation on the antibacterial activity of fermented noni (Morinda citrifolia) fruit juice extracts on selected microorganisms in Makurdi Metropolis

Plant products are generally useful for medicinal purposes, one of which is the noni plant. Noni or pace (Morinda citrifolia L.) fruit juice was extracted using aqueous and methanol solvents, then fermented and screened for the presence or absence of bioactive constituents (phytochemicals) using the modified standard procedures. The major phytochemical constituents revealed were alkaloids, anthraquinones, cardiac glycosides, and flavonoids, phenolic compounds, saponins, tannins, reducing sugar, saponnins, carbohydrate and terpenoids. Bacterial cell viability and MIC values were also determined by turbidity test. The lowest concentration of the extract with clear suspension was considered the MIC values. The lowest concentration of the extract in the post-incubation suspensions which did not harbour any bacterial growth upon spotting on MHA after overnight incubation at 37° C was considered the MBC values. The test was performed in triplicates alongside antibiotics ciprofloxacin (5μg) as a positive control at a concentration of 0.1mg/ml. Antibacterial activity was perfumed against three bacteria (Salmonella typhi, Escherichia coli, and Staphylococcus aureus) by agar-well diffusion method. Activity was observed for both methanolic and aqueous extracts of Morinda citrifolia L. fruit juice against the organisms tested. The extracts had both bacteriostatic and bactericidal activity against the test microorganisms. The Minimum Inhibitory Concentration (MIC) test revealed that the bacterial isolates were inhibited between concentrations of 50mg/ml and 25mg/ml for the methanolic extract, and between 100mg/ml and 50mg/ml for the aqueous extract of the fermented fruit juice. The extract was bactericidal from the range of 100mg/ml to 200mg/ml for both extracts, though with the exception of S. aureus which was bactericidal at 50mg/ml for methanolic extract.

Determination of minimum inhibitory concentrations using machine-learning-assisted agar dilution.

This research uses modern artificial intelligence and machine-learning approaches to standardize and automate the interpretation of agar dilution minimum inhibitory concentration testing. Artificial intelligence is currently of significant topical interest to researchers and clinicians. In our manuscript, we demonstrate a use-case in the microbiology laboratory and present validation data for the model's performance against manual interpretation.

Clinical Features and Treatment Outcomes of Carbapenem-Resistant Pseudomonas aeruginosa Keratitis

Evaluation of the microbiological diagnostic profile of multidrug-resistant Pseudomonas aeruginosa keratitis and potential management with rose bengal-photodynamic antimicrobial therapy (RB-PDAT) is important. To document the disease progression of carbapenemase-resistant P aeruginosa keratitis after an artificial tear contamination outbreak. This retrospective observation case series included 9 patients 40 years or older who presented at Bascom Palmer Eye Institute and had positive test results for multidrug-resistant P aeruginosa keratitis between January 1, 2022, and October 31, 2023. Evaluation of type III secretion phenotype, carbapenemase-resistance genes blaGES and blaVIM susceptibility to antibiotics, and in vitro and in vivo outcomes of RB-PDAT against multidrug-resistant P aeruginosa keratitis. Among the 9 patients included in the analysis (5 women and 4 men; mean [SD] age, 73.4 [14.0] years), all samples tested positive for exoU and carbapenemase-resistant blaVIM and blaGES genes. Additionally, isolates were resistant to carbapenems as indicated by minimum inhibitory concentration testing. In vitro efficacy of RB-PDAT indicated its potential application for treating recalcitrant cases. These cases highlight the rapid progression and challenging management of multidrug-resistant P aeruginosa. Two patients were treated with RB-PDAT as an adjuvant to antibiotic therapy and had improved visual outcomes. This case series highlights the concerning progression in resistance and virulence of P aeruginosa and emphasizes the need to explore alternative therapies like RB-PDAT that have broad coverage and no known antibiotic resistance. The findings support further investigation into the potential effects of RB-PDAT for other multidrug-resistant microbes.

Identification of vanA gene on Vancomycin-Resistant Staphylococcus aureus from Diabetic Ulcer Isolate at Lampung Province

Staphylococcus aureus is a type of bacteria that causes an increasing number of infections in hospitals, particularly in diabetic ulcers. Over the last few decades, there has been an alarming increase in the prevalence of pathogen strains of Staphylococcus aureus resistant to antibiotics such as Vancomycin. This study aimed to identify the presence of the vanA gene, which is responsible for the mode of Vancomycin resistance in Staphylococcus aureus Lampung isolate. Ulcer swab was collected from 32 patients with ulcer complications from surgical Installation, Internal Medicine, Home Diabetes Wound Clinic and Diabetes Wound Center in Bandar Lampung. Among the total ulcer swab, 12 samples of S. aureus were identified and subjected to the Minimum Inhibitory Concentration test to identify Vancomycin Resistant Staphylococcus aureus and evaluated the vanA gene by Polymerase Chain Reaction. To detect the presence of the vanA gene, a Polymerase Chain Reaction was performed on Staphylococcus aureus, Vancomycin-Resistant Staphylococcus aureus, and Vancomycin-intermediate Staphylococcus aureus using a specific primer arrangement. There were two samples of Vancomycin- Resistant Staphylococcus aureus and one sample of Vancomycin-intermediate Staphylococcus aureus from the Minimum Inhibitory Concentration test but only one sample tested positive for the vanA gene on Polymerase Chain Reaction. There is Staphylococcus aureus resistance to Vancomycin in Lampung isolate and the vanA gene was detected in some resistant isolates. The arrangement of the vanA gene in the Lampung isolate is different from that of the vanA gene in other places isolates.

Evaluating the Antimicrobial Efficacy of Apple Cider Vinegar on Bacteria and Fungi Isolated from Vaginitis

Vaginitis is a prevalent medical illness that affects a substantial proportion of women, mostly attributed to the excessive proliferation of bacteria and fungi. The objective of this study was to assess the antimicrobial effectiveness of Apple Cider Vinegar against bacteria and fungi that were obtained from cases of vaginitis. 50 vaginal swabs were collected from vaginitis patients at private clinics in Kirkuk City, Iraq (10 pregnant women and 40 non-pregnant women). The research included the isolation and identification of bacterial and fungal isolates. All isolates performed the Minimum Inhibitory Concentration and disc diffusion techniques tests. Different concentrations of apple cider vinegar (20%, 30%, 40%, 50% and 100%) were tested against the isolates and Standard antibiotic drugs were used as positive controls. The results of 50 patients were examined, showing that 22 (44%) had bacterial vaginitis and 39 (78%) had fungal vaginitis. Vaginitis was more prevalent in pregnant women compared with non-pregnant. Staphylococcus aureus recorded 14 (24.9%) out of the 54 bacterial isolates, followed by Echerichia coli, Group B- Streptococcus, Klebsiella pneumoniae, Staphylococcus epidermidis and Pseudomonas aeruginosa respectively. whereas Candida albicans recorded 30 (61.2%) out of the 49 fungal isolates, followed by Candida tropicalis and Candida parapsilosis, respectively. The findings showed a range of sensitivity to apple cider vinegar among the isolated samples. The efficacy of apple cider vinegar was found to be greater against fungal isolates compared to bacterial isolates, with a higher effectiveness observed against gram-negative bacteria in comparison to gram-positive bacteria. The highest MIC values were found for Gram-positive bacteria, which recorded 125 (µg/mL), followed by Gram-negative bacteria, 62.5 μg/ml. While the lowest MIC values for the isolated fungi were (31.25 μg/ml). A comparison to traditional antimicrobial drugs has highlighted ACV's potential as an alternative treatment for vaginitis. According to these findings, ACV may be an effective treatment choice for vaginitis.

Safety Assessment of Levilactobacillus brevis KU15006: A Comprehensive Analysis of its Phenotypic and Genotypic Properties.

Levilactobacillus brevis KU15006, isolated from kimchi, exhibits pathogen-antagonistic and anti-diabetic activities; however, the safety of this strain has not been assessed. In the present study, L. brevis KU15006 was evaluated to elucidate its safety as a probiotic strain using phenotypic and genotypic analyses. Its safety was assessed using a minimum inhibitory concentration test comprising nine antibiotics, 26 antibiotic resistance genes, a single conjugative element, virulence gene analysis, hemolysis, cell cytotoxicity, mucin degradation, and toxic metabolite production. L. brevis KU15006 exhibited equal or lower minimum inhibitory concentration for the nine antibiotics than the cut-off value established by the European Food Safety Authority. It did not harbor antibiotic resistance and virulence genes. L. brevis KU15006 lacked β-hemolysis, mucin degradation, cytotoxicity against Caco-2 cells, gelatin liquefaction, bile salt deconjugation, and toxic metabolite production abilities. Based on the results, L. brevis KU15006, which has antagonistic and anti-diabetic effects, could be marketed as a probiotic in the future.
