Determination of bactericidal activity against 3HC-2-Tre-labelled Mycobacterium abscessus (Mycobacteroides abscessus) by automated fluorescence microscopy

The minimum bactericidal concentration (MBC) of antibiotics is an important parameter for the potency of a drug in eradicating a bacterium as well as an important measure of the potential of a drug candidate in research and development. We have established a fluorescence-based microscopy method for the determination of MBCs against the non-tuberculous mycobacterium Mycobacterium abscessus (Mycobacteroides abscessus) to simplify and accelerate the performance of MBC determination compared to counting colony forming units on agar. Bacteria are labelled with the trehalose-coupled dye 3HC-2-Tre and analysed in a 96-well plate. The results of the new method are consistent with MBC determination by plating on agar. The method was used to evaluate the bactericidality of the antibiotics rifabutin, moxifloxacin, amikacin, clarithromycin and bedaquiline. Bactericidal effects against M. abscessus were observed, which are consistent with literature data.

Inhibitory potential of natural antimicrobial compounds against histamine-forming lactic acid bacteria

This work evaluated the potential of natural antimicrobial compounds to prevent histamine formation by inhibiting the microbial growth of Lentilactobacillus parabuchneri and Limosilactobacillus reuteri, bacteria selected for their ability to produce histamine in cheese. Of the evaluated compounds, eugenol, carvacrol, thymol and cinnamaldehyde were the most effective in inhibiting histamine-forming lactic acid bacteria and, therefore, in reducing histamine generation, even at concentrations below the minimum bactericidal concentration (MBC). Carvacrol significantly impacted both microbial viability and the metabolism of bacteria with the inhibition of decarboxylase activity. In the case of L. parabuchneri, the use of 0.7 mg/mL carvacrol (MBC/2) resulted in an 88% reduction in histamine content, and for L. reuteri a 96% reduction was observed by using 0.65 mg/(MBC/2). Therefore, using essential oil components during food processing and preservation would be a good strategy to prevent histamine accumulating in cheese and to, consequently, mitigate its adverse effects.

Bioactive compound from marine seagrass Streptomyces argenteolus TMA13: combatting fish pathogens with time-kill kinetics and live-dead cell imaging.

Actinobacteria, pervasive in aquatic and terrestrial environments, exhibit a filamentous morphology, possess DNA with a specific G + C content and production of numerous secondary metabolites. This study, focused on actinobacteria isolated from marine seagrass, investigating their antibacterial activity against fish pathogens. Among 28 isolates, Streptomyces argenteolus TMA13 displayed the maximum zone of inhibition against fish pathogens-Aeromonas hydrophila (10mm), Aeromonas caviae (22mm), Edwardsiella tarda (17mm), Vibrio harveyi (22mm) and Vibrio anguillarum (12mm) using the agar plug method. Optimization of this potent strain involved with various factors, including pH, temperature, carbon source and salt condition to enhance both yield production and antibacterial efficacy. In anti-biofilm assay shows the maximum percentage of inhibition while increasing concentration of TMA13 extract. Minimal Inhibitory Concentration (MIC) and Minimal Bactericidal Concentration (MBC) assays with TMA13 crude extract demonstrated potent activity against fish pathogens at remarkably low concentrations. Time-kill kinetics assay showcased growth curve variations over different time intervals for all fish pathogens treated with a 100µg/ml concentration of crude extract, indicating a decline in cells viability and progression into the death phase. Additionally, fluorescence microscopic visualization of bacterial cells exposed to the extracts emitting green and red fluorescence, enabling live-dead cell differentiation was also studied. Further characterization of the crude extract through GC-MS and FT-IR analyses performed and identified secondary metabolites with functional groups exhibiting significant antibacterial activity. This study elucidates the capacity of Streptomyces argenteolus TMA13 to enhance the production of antibiotic compounds effective against fish pathogens.

Ratanjot (Alkanna tinctoria L.) Root Extract, Rich in Antioxidants, Exhibits Strong Antimicrobial Activity against Foodborne Pathogens and Is a Potential Food Preservative.

Natural and sustainable plant-based antioxidants and antimicrobials are highly desirable for improving food quality and safety. The present investigation assessed the antimicrobial and antioxidant properties of active components from Alkanna tinctoria L. (herb) roots, also known as Ratanjot root. Two methods were used to extract active components: microwave-assisted hot water (MAHW) and ethanolic extraction. MAHW extract yielded 6.29%, while the ethanol extract yielded 18.27%, suggesting superior Ratanjot root extract powder (RRP) solubility in ethanol over water. The ethanol extract showed significantly higher antioxidant activity than the MAHW extract. Gas Chromatography-Mass Spectrometry analysis revealed three major phenolic compounds: butanoic acid, 3-hydroxy-3-methyl-; arnebin 7, and diisooctyl pthalate. The color attributes (L*, a*, b*, H°ab, C*ab) for the ethanolic and MAHW extracts revealed significant differences (p < 0.05) in all the above parameters for both types of extracts, except for yellowness (b*) and chroma (C*ab) values. The ethanol extract exhibited antimicrobial activity against 14 foodborne bacteria, with a significantly higher inhibitory effect against Gram-positive bacteria (Listeria monocytogenes and Staphylococcus aureus) than the Gram-negative bacteria (Salmonella enterica serovar Typhimurium and Escherichia coli). The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were both 25 mg/mL for the Gram-negative bacteria, while the MIC and MBC concentrations varied for Gram-positive bacteria (0.049-0.098 mg/mL and 0.098-0.195 mg/mL) and the antimicrobial effect was bactericidal. The antimicrobial activities of RRP extract remained stable under broad temperature (37-100 °C) and pH (2-6) conditions, as well as during refrigerated storage for 30 days. Application of RRP at 1% (10 mg/g) and 2.5% (25 mg/g) levels in a cooked chicken meatball model system prevented lipid oxidation and improved sensory attributes and retarded microbial growth during refrigerated (4 °C) storage for 20 days. Furthermore, the RRP extract was non-toxic when tested with sheep erythrocytes and did not inhibit the growth of probiotics, Lacticaseibacillus casei, and Lactiplantibacillus plantarum. In conclusion, the study suggests that RRP possesses excellent antimicrobial and antioxidant activities, thus making it suitable for food preservation.

Production and characterization of the lipopeptide with anti-adhesion for oral biofilm on the surface of titanium for dental implants.

Titanium implants are subject to bacterial adhesion and peri-implantitis induction, and biosurfactants bring a new alternative to the fight against infections. This work aimed to produce and characterize the biosurfactant from Bacillus subtilis ATCC 19,659, its anti-adhesion and antimicrobial activity, and cell viability. Anti-adhesion studies were carried out against Streptococcus sanguinis, Staphylococcus aureus, Fusobacterium nucleatum, Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis, and Proteus mirabilis as the minimum inhibitory concentration and the minimum bactericidal concentration. Cell viability was measured against osteoblast and fibroblast cells. The biosurfactant was classified as lipopeptide, with critical micelle concentration at 40µg mL- 1, and made the titanium surface less hydrophobic. The anti-adhesion effect was observed for Staphylococcus aureus and Streptococcus sanguinis with 54% growth inhibition and presented a minimum inhibitory concentration of 15.7µg mL- 1 for Streptococcus sanguinis and Aggregatibacter actinomycetemcomitans. The lipopeptide had no cytotoxic effect and demonstrated high potential application against bacterial biofilms.

Antibacterial Size Effect of ZnO Nanoparticles and Their Role as Additives in Emulsion Waterborne Paint.

Nosocomial infections, a prevalent issue in intensive care units due to antibiotic overuse, could potentially be addressed by metal oxide nanoparticles (NPs). However, there is still no comprehensive understanding of the impact of NPs' size on their antibacterial efficacy. Therefore, this study provides a novel investigation into the impact of ZnO NPs' size on bacterial growth kinetics. NPs were synthesized using a sol-gel process with monoethanolamine (MEA) and water. X-ray diffraction (XRD), transmission electron microscopy (TEM), and Raman spectroscopy confirmed their crystallization and size variations. ZnO NPs of 22, 35, and 66 nm were tested against the most common nosocomial bacteria: Escherichia coli, Pseudomonas aeruginosa (Gram-negative), and Staphylococcus aureus (Gram-positive). Evaluation of minimum inhibitory and bactericidal concentrations (MIC and MBC) revealed superior antibacterial activity in small NPs. Bacterial growth kinetics were monitored using optical absorbance, showing a reduced specific growth rate, a prolonged latency period, and an increased inhibition percentage with small NPs, indicating a slowdown in bacterial growth. Pseudomonas aeruginosa showed the lowest sensitivity to ZnO NPs, attributed to its resistance to environmental stress. Moreover, the antibacterial efficacy of paint containing 1 wt% of 22 nm ZnO NPs was evaluated, and showed activity against E. coli and S. aureus.

Chemical characterization and antibacterial activities of Brazilian propolis extracts from Apis mellifera bees and stingless bees (Meliponini).

The aim of this study was to evaluate the physicochemical composition and antibacterial activity of Brazilian propolis extracts from different types, concentrations, and extraction solvents and from different regions in Brazil. A total of 21 samples were analyzed, comprising 14 samples from Apis mellifera (12 green, 1 brown, and 1 red) and 7 samples from stingless bees (3 mandaçaia, 2 jataí, 1 hebora, and 1 tubuna). The analyses performed were dry extract, total phenolic content (TPC) and antioxidant activity (DPPH and ABTS). The antibacterial activity was performed by Determination of Minimal Inhibitory Concentration (MIC) and Minimal Bactericidal Concentration (MBC). The results showed that very low levels of phenolic compounds and antioxidant activity decreased the antimicrobial activity of the propolis extracts from tubuna and jataí. However, there was no correlation between the increase in propolis concentration in the extract, and the increase in antimicrobial activity. The highest TPC and antioxidant activity was obtained for green propolis extract made with 70% raw propolis that presented similar antibacterial activity to the samples formulated with 30% or less raw propolis. The aqueous propolis extract showed lower antimicrobial activity compared to the alcoholic extracts, indicating that ethanol is a better solvent for extracting the active compounds from propolis. It was observed that the MIC (0.06 to 0.2 mg/mL) and MBC (0.2 to 0.5 mg/mL) values for Gram-negative bacteria were higher compared to Gram-positive bacteria (MIC 0.001-0.2 mg/mL, and the MBC 0.02-0.5 mg/mL). The propolis extracts that exhibited the highest antimicrobial activities were from stingless bees hebora from the Distrito Federal (DF) and mandaçaia from Santa Catarina, showing comparable efficacy to samples 5, 6, and 7, which were the green propolis from the DF. Hence, these products can be considered an excellent source of bioactive compounds with the potential for utilization in both the pharmaceutical and food industries.

Effects of Hexagonal Boron Nitride and Mesoporous Silica Nanoparticles on the Morphology, Mechanical Properties and Antimicrobial Activity of Dental Composites

AbstractHexagonal boron nitride (HBN), an artificial material with unique properties, is used in many industries. This article focuses on the extent to which hexagonal boron nitride and silica nanoparticles (MSN) affect the physicochemical and mechanical properties and antimicrobial activity of prepared dental composites. In this study, HBN, and MSN were used as additives in dental composites. 5% and 10% by weight of HBN are added to the structure of the composite materials. FTIR analysis were performed to determine the components of the produced boron nitride powders, hexagonal boron nitride-containing composites, and filling material applications. The structural and microstructural properties of dental composites have been extensively characterized using X-ray diffractometry (XRD). Surface morphology and distributions of nano boron nitride were determined by scanning electron microscopy (SEM)-EDS. In addition, the solubility of dental composites in water and their stability in water and chemical solution (Fenton) were determined by three repetitive experiments. Finally, the antimicrobial activity of dental composites was detected by using Minimum Inhibitory Concentration (MIC) measurement, as well as Minimum Fungicidal Concentration (MFC) method against yeast strain Saccharomyces cerevisiae, and Minimum Bactericidal Concentration (MBC) method against bacteria strains, Staphylococcus aureus and Escherichia coli. Since the HMP series have better antimicrobial activity than the HP series, they are more suitable for preventing dental caries and for long-term use of dental composites. In addition, when HMP and HP series added to the composite are compared, HMP-containing dental composites have better physicochemical and mechanical properties and therefore have a high potential for commercialization.

Biocompatibility, Antimicrobial and Wound Healing Activities of Lavandula dentata Leaves Extract

Background: The human skin is a natural barrier that protects against external stress, but its physiological structure can be compromised when it gets damaged. In the field of medical science, wounds are a significant issue that requires immediate attention. Lavandula species are used in the food, perfume and cosmetics industries due to their antimicrobial, anti-inflammatory and wound-healing properties. Methods: The present study was done to assess the biocompatibility, antimicrobial and wound-healing potential of L. dentata leave methanolic extract (LE) that is cultivated in the Taif region of Saudi Arabia. The biocompatibility of LE was analyzed against the normal human skin fibroblast (HSF) cell line using SRB assay. A quarter of LE IC50 was used for wound healing assay. Finally, Escherichia Coli, Staphylococcus aureus, Pseudomonas aeruginosa and Streptococcus pyogenes were used to determine LE antibacterial potential through the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) measurement. Result: LE appears safe on HSF cell line with IC50 = 97 mg/mL. LE decreased wound width, wound area and the mean cell migratory rate of wound border cells during scratch closure, while increased the wound closure% in a time-dependent manner compared to negative control. LE shows antimicrobial potential against all four strains at MIC and MBC greater than 3.0 mg/mL, except for Streptococcus pyogenes MIC= 3.0 mg/mL. In conclusion, LE offers therapeutic benefits against wound healing with an antimicrobial effect, however, higher concentrations of LE should be used to assess its effect.

Conjugation of Lysozyme and Epigallocatechin Gallate for Improving Antibacterial and Antioxidant Properties.

One of the main interests in the food industry is the preservation of food from spoilage by microorganisms or lipid oxidation. A novel alternative is the development of additives of natural origin with dual activity. In the present study, a chemically modified lysozyme (Lys) with epigallocatechin gallate (EGCG) was developed to obtain a conjugate (Lys-EGCG) with antibacterial/antioxidant activity to improve its properties and increase its application potential. The modification reaction was carried out using a free radical grafting method for the Lys modification reaction, using ascorbic acid and hydrogen peroxide as radical initiators in an aqueous medium. The synthesis of Lys-EGCG conjugate was confirmed by spectroscopic (FT-IR, 1H-RMN, and XPS) and calorimetry differential scanning (DSC) analyses. The EGCG binding to the Lys biomolecule was quantified by the Folin-Ciocalteu method; the antibacterial activity was evaluated by minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MCB) against Staphylococcus aureus and Pseudomonas fluorescens; the antioxidant activity was evaluated by ABTS, DPPH, and FRAP. The spectroscopic results showed that the Lys-EGCG conjugate was successfully obtained, and the DSC analysis revealed a 20°C increase (P < 0.05) in the denaturation temperature of Lys due to EGCG modification. The EGCG concentration in Lys-EGCG was 97.97 ± 4.7µmol of EGCG/g of sample. The antibacterial and antioxidant activity of the Lys-EGCG conjugate was higher (P < 0.05) than pure EGCG and Lys. The chemical modification of Lys with EGCG allows for the bioconjugate with a dual function (antibacterial/antioxidant), broadening the range of Lys and EGCG applications to different areas such as food, cosmetic, and pharmaceutical industries.

Uncovering the Antibacterial Potential of a Peptide-Rich Extract of Edible Bird's Nest against Staphylococcus aureus.

The diverse pharmacological properties of edible bird's nest (EBN) have been elucidated in recent years; however, investigations into its antibacterial effects are still limited. In the present study, we explored the antibacterial activity of a peptide-rich extract of EBN against Staphylococcus aureus, a notorious pathogen. The EBN extract (EEE) was prepared by soaking EBN in 80% ethanol for 2 days at 60°C. Biochemical analyses showed that peptides at the molecular weight range of 1.7-10 kDa were the major biochemical compounds in the EEE. The extract exhibited strong inhibition against S. aureus at a minimum inhibitory concentration (MIC) of 125 μg/ml and a minimum bactericidal concentration (MBC) of 250 μg/ml. This activity could be attributed to the impact of the extract on cell membrane integrity and potential, biofilm formation, and reactive oxidative species (ROS) production. Notably, the expression of biofilm- and ROS-associated genes, including intercellular adhesion A (icaA), icaB, icaC, icaD, and superoxide dismutase A (sodA), were deregulated in S. aureus upon the extract treatment. Our findings indicate a noteworthy pharmacological activity of EBN that could have potential application in the control of S. aureus.

Antibacterial, Antibiofilm, and Antioxidant Activities of Aqueous Crude Gymnema inodorum Leaf Extract against Vancomycin-Resistant Enterococcus faecium.

Vancomycin-resistant Enterococcus faecium (VREF) causes nosocomial infections with high mortality and morbidity rates. This study aimed to evaluate the antibacterial and antibiofilm activities of aqueous crude Gymnema inodorum leaf extract (GIE) against the VREF ATCC 700221 strain. The antimicrobial activity of GIE against VREF was performed using disk diffusion and broth microdilution. The antibiofilm activities were evaluated using the crystal violet staining assay. The antioxidant potential was evaluated. Preliminary screening of the antimicrobial activity of 50 and 100 µg/disk of GIE against VREF revealed inhibition zones of 8.33 ± 0.58 mm and 8.67 ± 0.29 mm, respectively. Additionally, the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values against VREF were 125 and ≥ 250 mg/mL, respectively. SEM analysis showed that treatment with GIE caused morphological changes, including incomplete cell division, damaged cell walls, and cell content leakage, suggesting a disruption of bacterial cells. GIE also inhibited and eradicated biofilms formed by VREF. The extract exhibited antioxidant activities in the DPPH and ABTS assays. While GIE shows potential as an antibacterial and antibiofilm agent, further studies are necessary to fully understand the underlying mechanisms and optimize its use for therapeutic applications.

Comparison of quantity, quality and antibacterial activity of essential oil Mentha longifolia (L.) L. under different traditional and modern extraction methods.

Extraction is the first and most important step in obtaining the effective ingredients of medicinal plants. Mentha longifolia (L.) L. is of considerable economic importance as a natural raw material for the food and pharmaceutical industries. Since the effect of different extraction methods (traditional and modern methods) on the quantity, quality and antimicrobial activity of the essential oil of this plant has not been done simultaneously; the present study was designed for the first time with the aim of identifying the best extraction method in terms of these features. For this purpose, extracting the essential oil of M. longifolia with the methods of hydrodistillation with Clevenger device (HDC), steam distillation with Kaiser device (SDK), simultaneous distillation with a solvent (SDE), hydrodistillation with microwave device (HDM), pretreatment of ultrasonic waves and Clevenger (U+HDC) and supercritical fluid (SF) were performed. Chemical compounds were identified by gas chromatography coupled with mass spectrometer (GC-MS). Antimicrobial activity of essential oils against various clinical microbial strains was evaluated by agar diffusion method and determination of the minimum inhibitory concentration and minimum bactericidal concentration (MIC and MBC). The results showed that the highest and lowest yields of M. longifolia leaf essential oil belonged to HDC (1.6083%) and HDM (0.3416%). The highest number of compounds belonged to SDK essential oil and was equal to 72 compounds (with a relative percentage of 87.13%) and the lowest number of compounds was related to the SF essential oil sample (7 compounds with a relative percentage of 100%). Piperitenone (25.2-41.38%), piperitenone oxide (22.02-0%), pulegone (10.81-0%) and 1,8-cineole (5-35.0%) are the dominant and main components of M. longifolia essential oil were subjected to different extraction methods. Antimicrobial activity results showed that the lowest MIC value belonged to essential oils extracted by HDM, SDK, SDE and U+HDC methods with a value of 1000 μg/mL was observed against Gram-negative bacteria Shigella dysenteriae, which was 5 times weaker than rifampin and 7 times weaker than gentamicin. Therefore, it can be concluded that in terms of efficiency of the HDC method, in terms of the percentage of compounds of the HDM method, and in terms of microbial activity, the SDK, HDM and U+HDC methods performed better.

The Effect of Salinity Stress on the Antibacterial Activity of SpirulinaPlatensis Algae

Background: The production of secondary metabolites in different strains of microalgae varies and is likely dependent on environmental conditions. Consequently, the production of bioactive substances as secondary metabolites occurs in microalgae to aid their survival in adverse environmental conditions such as salinity stress. Objectives: The purpose of this study is to investigate the antibacterial activity of the methanolic extract of S. platensis algae cultivated under different salinity stresses against Yersinia rukeri, Escherichia coli, Salmonella sp., and Vibrio cholerae. Methods: The broth microdilution method was used to evaluate the growth inhibitory activity of the extracts against Yersinia ruckeri, Salmonella sp., Escherichia coli, and Vibrio cholerae bacteria. The minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) were determined by this method, and the diameter of the inhibition zone was assessed through the well diffusion method. Results: In the treatment with algae extract grown under 3.5 ppt salinity stress, the concentrations of 50 and 25 mg/mL extract showed the largest inhibition zone diameter against Salmonella sp. compared to other bacteria. Under 7 ppt salinity stress, all studied concentrations of algal extract exhibited a higher inhibition zone diameter against V. cholerae compared to other bacteria. The results of comparing different concentrations of algae extract between the two salinity stresses of 3.5 and 7 ppt for each bacterium showed a significant difference (P &lt; 0.05). As salinity increased, the diameter of the inhibition zone also increased for all bacteria. Conclusions: Our results showed an increase in the antibacterial activity of the methanolic extract of Spirulina platensis with higher salinity stress levels. Therefore, cultivating Spirulina algae in salt water can be a cost-effective and suitable method to produce more secondary metabolites for use in the pharmaceutical industry.

Comparable Antibacterial Effects of Silver and Iron Oxide Nanoparticles on Urinary Calculi Obtained After Percutaneous Nephrolithotomy

Background: In recent years, science and industry have focused on preparing nanoparticles (NPs). Due to the resistance of bacteria to antibiotics, there is an overwhelming need to find effective antimicrobials with fewer side effects. Objectives: The aim of this study was to synthesize silver and iron oxide NPs and investigate their antimicrobial effects on bacteria isolated from urinary stones. Methods: Cases of urolithiasis from 45 patients with staghorn stones, extracted through percutaneous nephrolithotomy (PCNL), were included in the study. Urinary stone cultures were performed, and the isolation and identification of bacteria were done using standard microbiological techniques. Silver and iron oxide NPs were synthesized, and the minimum bactericidal concentration (MBC) and minimum inhibitory concentration (MIC) were evaluated for the isolates. Results: Seventeen (60.7%) out of 28 positive cultured cases of the isolated bacteria were gram-negative, and 11 (39.3%) were gram-positive. The antibacterial effects of silver and iron oxide NPs revealed that the maximum inhibitory zone for Ag NPs at 1000 ppm was 23 mm, and for Fe3O4 NPs, it was 18.5 mm. The MIC was 180 µg/mL for Ag and 250 µg/mL for Fe3O4. The MBC was 228 µg/mL for Ag and 300 µg/mL for Fe3O4. Conclusions: Nanoparticles exhibited antibacterial effects on the bacteria studied in a concentration-dependent manner. Ag NPs showed a more pronounced bactericidal effect than Fe3O4 NPs. As a result, these two NPs demonstrated effective antibacterial activity against both gram-positive and gram-negative bacteria.

Comparable Antibacterial Effects of Silver and Iron Oxide Nanoparticles on Urinary Calculi Obtained After Percutaneous Nephrolithotomy

Background: In recent years, science and industry have focused on preparing nanoparticles (NPs). Due to the resistance of bacteria to antibiotics, there is an overwhelming need to find effective antimicrobials with fewer side effects. Objectives: The aim of this study was to synthesize silver and iron oxide NPs and investigate their antimicrobial effects on bacteria isolated from urinary stones. Methods: Cases of urolithiasis from 45 patients with staghorn stones, extracted through percutaneous nephrolithotomy (PCNL), were included in the study. Urinary stone cultures were performed, and the isolation and identification of bacteria were done using standard microbiological techniques. Silver and iron oxide NPs were synthesized, and the minimum bactericidal concentration (MBC) and minimum inhibitory concentration (MIC) were evaluated for the isolates. Results: Seventeen (60.7%) out of 28 positive cultured cases of the isolated bacteria were gram-negative, and 11 (39.3%) were gram-positive. Evaluation of the Antibacterial, Anti-Inflammatory, And Bone-Promoting Capacity of UiO-66 Loaded with Thymol or Carvacrol.

Oral infectious diseases have a significant impact on the health of oral and maxillofacial regions, as well as the overall well-being of individuals. Carvacrol and thymol, two isomers known for their effective antibacterial and anti-inflammatory properties, have gained considerable attention in the treatment of oral infectious diseases. However, their application as topical drugs for oral use is limited due to their poor physical and chemical stability. UiO-66, a metal-organic framework based on zirconium ion (Zr4+), exhibits high drug loading capability. Carvacrol and thymol were efficiently loaded onto UiO-66 with loading rates of 79.60 ± 0.71% and 79.65 ± 0.76%, respectively. The release rates of carvacrol and thymol were 77.82 ± 0.87% and 76.51 ± 0.58%, respectively, after a period of 72 h. Moreover, Car@UiO-66 and Thy@UiO-66 demonstrated excellent antibacterial properties against Candida albicans, Escherichia coli, and Staphylococcus aureus with minimum bactericidal concentrations (MBC) of 0.313 mg/mL, 0.313 mg/mL, and 1.25 mg/mL, respectively. Furthermore, based on the results of the CCK8 cytotoxicity assay, even at concentrations as high as 1.25 mg/mL, Car@UiO-66 and Thy@UiO-66 exhibited excellent biocompatibility with a relative cell survival rate above 50%. These findings suggest that Car@UiO-66 and Thy@UiO-66 possess favorable biocompatibility properties without significant toxicity towards periodontal membrane cells. Additionally, in vivo studies confirmed the efficacy of Car@UiO-66and Thy@UiO-66 in reducing inflammation, promoting bone formation through inhibition of TNF-a and IL6 expression, enhancement of IL10 expression, and acceleration of bone defect healing. Therefore, the unique combination of antibacterial, anti-inflammatory, and osteogenic properties make Car@UiO-66 and Thy@Ui O-66 promising candidates for the treatment of oral infectious diseases and repairing bone defects.

Design of Thermosensitive Niosomes by Eutectic Mixture of Natural Fatty Acids.

In the current study, a smart release system responsive to temperature was developed to improve the efficiency of tetracycline (TC) in antibacterial therapy. The nanovesicles designed consist of a non-ionic surfactant, SPAN60, cholesterol and a phase change material (PCM) as a thermoresponsive gating material. Niosomes were prepared using an increasing amount of PCM and characterized in terms of size, zeta potential, colloidal stability and thermoresponsive properties. The vesicles that developed were homogenous in size, had good biocompatibility and stability for up to 3 months and demonstrated thermoresponsive behavior. A low drug leakage was observed at 37 °C, while a rapid release occurred at 42 °C, due to the faster diffusion rate of the drug trough the melted PCM. This controllable drug release capacity allows us to avoid premature drug release, minimizing unwanted and toxic effects and ensuring a long retention time in the nanodevice so that it reaches the infected sites. In addition, TC-loaded niosomes were screened to investigate their antibacterial activity against various Gram-positive and Gram-negative bacteria by minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) assays. An interesting temperature-dependent antibacterial activity was observed against some bacterial strains: the niosomes activity against S. epidermis, for example, was improved by the temperature increase, as suggested by a reduction in MIC values from 112.81 to 14.10 μM observed at 37 and 42 °C, respectively. Taken together, the thermoresponsive platform developed allows us to use lower antibiotic amounts while ensuring therapeutic efficacy and, so, will advance the development of a novel antibacterial agent in clinical practice.

Kinetic study of in vitro release of curcumin from chitosan biopolymer and the evaluation of biological efficacy

Sustained release of curcumin from the polymeric carrier system chitosan, a natural biopolymer material derived from chitin originated from natural shrimp shell waste, was studied. Six kinetic models, zero order, first order, Korsmeyer–Peppas (KP), Peppas – Sahlin (PS), Higuchi, and Hixson–Crowell, were applied to study the drug release kinetics. The release mechanism of the drug from the curcumin-chitosan composite was evaluated by changing the pH, ionic strength of the release media, and drug concentration. KP and PS models were selected among the studied models to investigate the drug release mechanism from the chitosan biopolymer based on the R2 values (R2 > 0.99). The model constants m in the PS model and n in the KP model stand for the case II relaxation and Fickian diffusion contribution, respectively. The n being < 0.43 in the KP model suggests that the Fickian diffusion governs the drug release. Furthermore, there is a noticeable difference between the values obtained for model parameters m and n in the PS and KP models, indicating that Case II relaxation and Fickian diffusion play crucial roles in the curcumin release mechanism from chitosan. Polymer relaxation has been proven to play a predominant role in releasing curcumin from the composite at lower ionic strengths and higher pH values. Anti-inflammatory activity was tested using the egg-albumin denaturation assay, and the diphenyl-2-picrylhydrazyl assay was carried out to determine the antioxidant activity of the composite. The composite material showed IC50 values of 0.29 mg/ mL and 1.08 mg/ mL for anti-inflammatory and anti-oxidant activities, respectively. The drug composite has shown antibacterial activity against Pseudomonas aeruginosa, Klebsiella pneumoniae, and Staphylococcus aureus, which are highly effective against S.aureus. The resulting inhibition zones for S.aureus were 13.34 ± 0.34 mm, 16.34 ± 0.60 mm, and 13.34 ± 0.73 mm for 5, 10, and 20 mg/ml concentrations, respectively. The drug composite’s minimum inhibitory concentration/ minimum bactericidal concentration ratio for S.aureus, K. pneumoniae, and P.aeruginosa was greater than 4, suggesting that they cause bacteriostatic effects.
