Assessing plasma activated water irrigation effects on tomato seedlings

Introduction: The study investigates the potential of Plasma Activated Water (PAW) as an innovative irrigation medium to enhance growth and defense responses in tomato seedlings. It explores PAW’s utility in both healthy seedlings and those inoculated with Tomato mottle mosaic virus (ToMMV).Methods: PAW, produced through a dielectric barrier volume discharge, serves as a chemical-free alternative to traditional fertilizers. Tomato seedlings were irrigated with PAW or control solutions. The study employs biometric measurements to assess growth and biochemical analysis to evaluate antioxidant levels and pigments. Gene expression analysis was conducted to evaluate the plant response, while the distribution of macro and micronutrients was assessed through micro X-ray fluorescence.Results and discussion: Results indicate that PAW-irrigated seedlings exhibit significant growth enhancement compared to those receiving conventional fertilization. Increased levels of antioxidant molecules and pigments suggest improved photosynthetic activity and stress tolerance. Gene expression analysis shows up-regulation of defense genes in PAW-treated plants post-viral infection. The up-regulation of defense genes and the restoration of mineral nutrient distribution in PAW-treated, virus-infected plants highlight PAW’s role in enhancing plant resilience against pathogens and mitigating nutrient deficiencies. These findings emphasize PAW’s potential as a sustainable agricultural solution, promoting plant growth, enhancing defense mechanisms, and reducing biotic stress due to virus infections.

Identification of the ALMT gene family in the potato (Solanum tuberosum L.) and analysis of the function of StALMT6/10 in response to aluminum toxicity.

Aluminum (Al)-activated malate transporters (ALMTs) play an important role in the response to Al toxicity, maintenance of ion homeostasis balance, mineral nutrient distribution, and fruit quality development in plants. However, the function of the StALMT gene family in potato remains unknown. In this study, 14 StALMT genes were identified from the potato genome, unevenly distributed on seven different chromosomes. Collinearity and synteny analyses of ALMT genes showed that potatoes had 6 and 22 orthologous gene pairs with Arabidopsis and tomatoes, respectively, and more than one syntenic gene pair was identified for some StALMT gene family members. Real-time quantitative polymerase chain reaction (qPCR) results showed differential expression levels of StALMT gene family members in different tissues of the potato. Interestingly, StALMT1, StALMT6, StALMT8, StALMT10, and StALMT12 had higher expression in the root of the potato cultivar Qingshu No. 9. After being subjected to Al3+ stress for 24 h, the expression of StALMT6 and StALMT10 in roots was evidently increased, displaying their decisive role in Al3+ toxicity. In addition, overexpression of StALMT6 and StALMT10 in Arabidopsis enhanced its tolerance to Al toxicity. These results indicate that StALMT6 and StALMT10 impart Al3+ resistance in the potato, establishing the foundation for further studies of the biological functions of these genes.

Asymmetric distribution of mineral nutrients aggravates uneven fruit pigmentation driven by sunlight exposure in litchi.

Sunlight boosts anthocyanin synthesis/accumulation in sunny pericarp of litchi fruit, directly leading to uneven pigmentation. Distribution discrepancy of mineral element aggravates uneven coloration by modulating synthesis/accumulation of anthocyanin and sugar. Uneven coloration, characterized by red pericarp on sunny side and green pericarp on shady side, impacts fruit quality of 'Feizixiao' (cv.) litchi. The mechanisms of this phenomenon were explored by investigating the distribution of chlorophyll, flavonoids, sugars, and mineral elements in both types of pericarp. Transcriptome analysis in pericarp was conducted as well. Sunny pericarp contained higher anthocyanins in an order of magnitude and higher fructose, glucose, co-pigments (flavanols, flavonols, ferulic acid), and mineral elements like Ca, Mg and Mn, along with lower N, P, K, S, Cu, Zn and B (P < 0.01), compared to shady pericarp. Sunlight regulated the expression of genes involved in synthesis/accumulation of flavonoids and sugars and genes functioning in nutrient uptake and transport, leading to asymmetric distribution of these substances. Anthocyanins conferred red color on sunny pericarp, sugars, Ca and Mg promoted synthesis/accumulation of anthocyanins, and co-pigments enhanced color display of anthocyanins. The insufficiencies of anthocyanins, sugars and co-pigments, and inhibition effect of excess K, S, N and P on synthesis/accumulation of anthocyanins and sugars, jointly contributed to green color of shady pericarp. These findings highlight the role of asymmetric distribution of substances, mineral elements in particular, on uneven pigmentation in litchi, and provide insights into coloration improvement via precise fertilization.

Toward the replacement of conventional fertilizer with polyhalite in eastern China to improve peanut growth and soil quality

Abstract Background Polyhalite fertilizer application is an effective way to alleviate a shortage of potassium. This study explored the effects of polyhalite fertilizer application as a total or partial replacement for conventional potassium fertilizer to improve peanut growth and soil quality. Results The index of peanut yield and its economic benefits, the content and distribution of mineral nutrients in different organs, soil chemical properties, and rhizosphere microbial diversity in response to the treatments were examined. The results show that the M4P6T treatment (60% polyhalite fertilizer replacing potassium chloride as the base fertilizer, and 40% potassium chloride fertilizer applied as a topdressing) increased profit by 7.2% without affecting the yield. The M4P6T treatment significantly improved the accumulation and distribution of potassium, calcium and magnesium in the kernels compared with the M10B treatment (no polyhalite fertilizer; potassium chloride fertilizer only as the base fertilizer). Soil treated with polyhalite fertilizer had higher alpha-diversity values and greater relative abundance of microbes at the phylum and genus levels. Conclusions Partial substitution of polyhalite for potassium chloride improved soil quality and peanut growth more than did single applications of polyhalite and potassium chloride. Kinetics of Macro and Micronutrients during Germination of Habanero Pepper Seeds in Response to Imbibition

The aim of this study was to evaluate the concentration and distribution of a number of macro- and micronutrients in response to imbibition in habanero pepper seeds extracted from fruits with different stages of maturity and postharvest storage times, as well as in their seedlings. Fruits were harvested unripe, half-ripe, and ripe, and were subjected to storage for 0 (control), 7, and 14 days postharvest prior to seed extraction. An X-ray microfluorescence analysis was carried out on seeds pre- and post-imbibition, as well as on seedlings at 10 and 14 days after sowing. K, Ca, Fe, P, Mg, and Mn were detected by elemental analysis. The results indicated that the elements had a higher concentration and distribution in seeds extracted from half-ripe fruits and ripe unstored fruits, as well as in seeds extracted from fruits stored for 7 and 14 days postharvest. K and Ca were the elements with the highest distributions and concentrations in seeds and seedlings pre- and post-imbibition compared to the other elements. At all maturity stages, postharvest storage increased the concentration and distribution of mineral nutrients in seeds and seedlings before and after imbibition. Storage translocated mineral elements to the radicle before germination, increased meristem growth in emerged seedlings, decreased electrical conductivity, and increased germination. Fourteen days of postharvest storage increased the distribution of macro- and micronutrients in immature seeds, decreasing electrical conductivity, potentiating germination, and improving mineral element distribution in seedlings.

Exogenous 5-aminolevulinic acid alleviates low-temperature damage by modulating the xanthophyll cycle and nutrient uptake in tomato seedlings

5-Aminolevulinic acid (ALA), an antioxidant existing in plants, has been widely reported to participate in the process of coping with cold stress of plants. In this study, exogenous ALA promoted the growth of tomato plants and alleviated the appearance of purple tomato leaves under low-temperature stress. At the same time, exogenous ALA improved antioxidant enzyme activities, SlSOD gene expression, Fv/Fm, and proline contents and reduced H2O2 contents, SlRBOH gene expression, relative electrical conductivity, and malondialdehyde contents to alleviate the damage caused by low temperature to tomato seedlings. Compared with low-temperature stress, spraying exogenous ALA before low-temperature stress could restore the indicators of photochemical quenching, actual photochemical efficiency, electron transport rate, and nonphotochemical quenching to normal. Exogenous ALA could increase the total contents of the xanthophyll cycle pool, the positive de-epoxidation rate of the xanthophyll cycle and improved the expression levels of key genes in the xanthophyll cycle under low-temperature stress. In addition, we found that exogenous ALA significantly enhanced the absorption of mineral nutrients, promoted the transfer and distribution of mineral nutrients to the leaves, and improved the expression levels of mineral nutrient absorption-related genes, which were all conducive to the improved adaptation of tomato seedlings under low-temperature stress. In summary, the application of exogenous ALA can increase tomato seedlings’ tolerance to low-temperature stress by improving the xanthophyll cycle and the ability of the absorption of mineral nutrients in tomato seedlings.

Characterization of Stem Nodes Associated with Carbon Partitioning in Maize in Response to Nitrogen Availability.

Stem node has been found to be a hub for controlling mineral nutrient distribution in gramineous plants. However, the characteristics of stem nodes associated with whole-plant carbon partitioning in maize (Zea mays L.) and their responses to nitrogen (N) availability remains elusive. Maize plants were grown in greenhouse under low to high N supply. Plant growth, sugar accumulation, and sugar transporters in nodes and leaves, as well as the anatomical structure of nodes, were investigated at vegetative phase. When compared to N-sufficient plants, low-N availability stunted growth and resulted in 49–64% less sugars in leaves, which was attributed to low photosynthesis or the accelerated carbon export, as evidenced by more 13C detected further below leaf tips. Invariably higher sugar concentrations were found in the stem nodes, rather than in the leaves across N treatments, indicating a crucial role of nodes in facilitating whole-plant carbon partitioning. More and smaller vascular bundles and phloem were observed in stem nodes of N-deficient plants, while higher sugar levels were found in the bottom nodes than in the upper ones. Low-N availability upregulated the gene expressions of sugar transporters, which putatively function in nodes such as ZmSWEETs and ZmSUTs at the bottom stem, but suppressed them in the upper ones, showing a developmental impact on node function. Further, greater activity of sugar transporters in the bottom nodes was associated with less sugars in leaves. Overall, these results highlighted that stem nodes may play an important role in facilitating long-distance sugar transport in maize.

Arbuscular mycorrhizal fungi species improve the fatty acids profile and nutrients status of soybean cultivars grown under drought stress.

To investigate the effects of arbuscular mycorrhizal fungi (AMF) species on the absorption and distribution of mineral nutrients in soybean cultivars under drought stress, an experiment was carried out through a factorial method in the form of randomized complete blocks with six replicates in 2020. Experimental factors include: drought stress at three irrigation levels (well-watered [WW], medium stress [MS] and severe stress [SS], i.e., 100%, 70% and 40% FC), soybean cultivars at two levels (Sepideh and Williams), and mycorrhizae application at four levels (nonapplication, Funneliformis mosseae, Glomus hoi, Rhizophagus intraradices). The results indicated that drought stress increased the concentration of grain potassium (K) and sodium (Na), leaf K, stem and leaf Na, and decreased the concentrations of grain phosphorus (P), copper (Cu), and zinc (Zn), leaf P, stem and leaf Cu, and manganese (Mn). In addition, there was no significant difference in terms of concentrations of grain K. The highest reduction was observed under SS conditions (40% FC). G. hoi colonization increased the concentrations of grain P and Zn, leaf K and Cu and stem Mn under SS conditions (40% FC). In addition, compared to Sepideh cv., Williams cv. showed higher stress resistance. Seed oil content decreased in the plants exposed to drought stress. Severe drought treatments have a deleterious effect on seed fatty acid composition, resulting in enhanced linoleic, oleic and linolenic acids. AMF colonization is a useful tool for improving the plant nutrient uptake, fatty acid profile, efficiency of resource utilization and stabilizing yield, hence reducing the production risks of crops grown under drought stress conditions. It was concluded that AMF colonization should be employed to help alleviate the adverse effects of drought stress. AMF colonization is an effective biotechnological strategy that can alter nutrient uptake and fatty acid composition and enhance oil quality in soybean cultivars under drought conditions.

From plant biology research to technology transfer and knowledge extension: improving food quality and mitigating environmental impacts

Academic scientists face an unpredictable path from plant biology research to real-life application. Fundamental studies of γ-aminobutyrate and carotenoid metabolism, control of Botrytis infection, and the uptake and distribution of mineral nutrients illustrate that most academic research in plant biology could lead to innovative solutions for food, agriculture, and the environment. The time to application depends on various factors such as the fundamental nature of the scientific questions, the development of enabling technologies, the research priorities of funding agencies, the existence of competitive research, the willingness of researchers to become engaged in commercial activities, and ultimately the insight and creativity of the researchers. Applied research is likely to be adopted more rapidly by industry than basic research, so academic scientists engaged in basic research are less likely to participate in science commercialization. It is argued that the merit of Discovery Grant applications to the Natural Sciences and Engineering Research Council (NSERC) of Canada should not be evaluated for their potential impact on policy and (or) technology. Matching industry funds in Canada rarely support the search for knowledge. Therefore, NSERC Discovery Grants should fund basic research in its entirety.

Early sensing of phosphate deprivation triggers the formation of extra root cap cell layers via SOMBRERO through a process antagonized by auxin signaling.

The role of the root cap in the plant response to phosphate deprivation has been scarcely investigated. Here we describe early structural, physiological and molecular changes prior to the determinate growth program of the primary roots under low Pi and unveil a critical function of the transcription factor SOMBRERO in low Pi sensing. Mineral nutrient distribution in the soil is uneven and roots efficiently adapt to improve uptake and assimilation of sparingly available resources. Phosphate (Pi) accumulates in the upper layers and thus short and branched root systems proliferate to better exploit organic and inorganic Pi patches. Here we report an early adaptive response of the Arabidopsis primary root that precedes the entrance of the meristem into the determinate developmental program that is a hallmark of the low Pi sensing mechanism. In wild-type seedlings transferred to low Pi medium, the quiescent center domain in primary root tips increases as an early response, as revealed by WOX5:GFP expression and this correlates with a thicker root tip with extra root cap cell layers. The halted primary root growth in WT seedlings could be reversed upon transfer to medium supplemented with 250 µM Pi. Mutant and gene expression analysis indicates that auxin signaling negatively affects the cellular re-specification at the root tip and enabled identification of the transcription factor SOMBRERO as a critical element that orchestrates both the formation of extra root cap layers and primary root growth under Pi scarcity. Moreover, we provide evidence that low Pi-induced root thickening or the loss-of-function of SOMBRERO is associated with expression of phosphate transporters at the root tip. Our data uncover a developmental window where the root tip senses deprivation of a critical macronutrient to improve adaptation and surveillance.

Potential Networks of Nitrogen-Phosphorus-Potassium Channels and Transporters in Arabidopsis Roots at a Single Cell Resolution.

Nitrogen (N), phosphorus (P), and potassium (K) are three major macronutrients essential for plant life. These nutrients are acquired and transported by several large families of transporters expressed in plant roots. However, it remains largely unknown how these transporters are distributed in different cell-types that work together to transfer the nutrients from the soil to different layers of root cells and eventually reach vasculature for massive flow. Using the single cell transcriptomics data from Arabidopsis roots, we profiled the transcriptional patterns of putative nutrient transporters in different root cell-types. Such analyses identified a number of uncharacterized NPK transporters expressed in the root epidermis to mediate NPK uptake and distribution to the adjacent cells. Some transport genes showed cortex- and endodermis-specific expression to direct the nutrient flow toward the vasculature. For long-distance transport, a variety of transporters were shown to express and potentially function in the xylem and phloem. In the context of subcellular distribution of mineral nutrients, the NPK transporters at subcellular compartments were often found to show ubiquitous expression patterns, which suggests function in house-keeping processes. Overall, these single cell transcriptomic analyses provide working models of nutrient transport from the epidermis across the cortex to the vasculature, which can be further tested experimentally in the future.

A transporter for delivering zinc to the developing tiller bud and panicle in rice.

Tiller number is one of the most important agronomic traits that determine rice (Oryza sativa) yield. Active growth of tiller bud (TB) requires high amount of mineral nutrients; however, the mechanism underlying the distribution of mineral nutrients to TB with low transpiration is unknown. Here, we found that the distribution of Zn to TB is mediated by OsZIP4, one of the ZIP (ZRT, IRT-like protein) family members. The expression of OsZIP4 was highly detected in TB and nodes, and was induced by Zn deficiency. Immunostaining analysis revealed that OsZIP4 was mainly expressed in phloem of diffuse vascular bundles in the nodes and the axillary meristem. The mutation of OsZIP4 did not affect the total Zn uptake, but altered Zn distribution; less Zn was delivered to TB and new leaf, but more Zn was retained in the basal stems at the vegetative growth stage. Bioimaging analysis showed that the mutant aberrantly accumulated Zn in enlarged and transit vascular bundles of the basal node, whereas in wild-type high accumulation of Zn was observed in the meristem part. At the reproductive stage, mutation of OsZIP4 resulted in delayed panicle development, which is associated with decreased Zn distribution to the panicles. Collectively, OsZIP4 is involved in transporting Zn to the phloem of diffuse vascular bundles in the nodes for subsequent distribution to TBs and other developing tissues. It also plays a role in transporting Zn to meristem cells in the TBs.

Landform affects the distribution of mineral nutrients in the tropical peats: a case study in a peatland of Siak, Indonesia

ABSTRACT Most peatlands in Indonesia border mineral upland, yet research reporting the effects of the presence of mineral upland on the distribution of mineral nutrients are limited. We collected multiple samples in surface layers from a peatland located between the Siak River and a mineral upland in Riau Province, Indonesia and measured electrical conductivity, pH, and exchangeable Ca2+, Mg2+, K+, and Na+. Transects were investigated for ash content and chemical composition of its profiles. Although electrical conductivity (183 ± 79 μS cm–1) and pH values (3.7 ± 0.3) were similar at surface layers, exchangeable Ca2+ (especially near the mineral upland) and Mg2+ were high at 0–15 cm deep (8.3 ± 5.1 cmolc kg–1 and 2.7 ± 1.1 cmolc kg–1, respectively) and tended to decline in the lower layers. Exchangeable K+ and Na+ did not show a clear trend, probably because of weaker bonding with peat material. High ash content (>10%) was found more frequently alongside the mineral upland than by the riverbank. In the profiles near the mineral upland, total Ca, Mg, Al, and Fe levels in the surface layer were higher (4.5 ± 0.3 g kg–1, 0.9 ± 0.0 g kg–1, 3.0 ± 0.5 g kg–1, and 1.6 ± 0.5 g kg–1, respectively), than those near the riverbank, which were 1.7 ± 0.7 g kg–1, 0.5 ± 0.2 g kg–1, 1.4 ± 0.3 g kg–1, and 0.6 ± 0.3 g kg–1, respectively. Based on the distribution of ash, exchangeable Ca2+ and Mg2+, and total Ca, Mg, Al, and Fe content, the present study suggests that the presence of the mineral upland nutritionally enriches the peat with Ca, Mg, and Fe. The influence of landform should be considered for a comprehensive understanding of nutrients sources in peatlands.

Vascular Cambium-Localized AtSPDT Mediates Xylem-to-Phloem Transfer of Phosphorus for Its Preferential Distribution in Arabidopsis

During plant growth and development mineral elements are preferentially delivered to different organs and tissues to meet the differential demand. It has been shown that the preferential distribution of mineral nutrients in gramineous plants is mediated by node-based transporters, but the mechanisms of preferential distribution in dicots are poorly understood. Here, we report a distinct mechanism for the preferential distribution of phosphorus (P) in Arabidopsis plants, revealed by detailed functional analysis of AtSPDT/AtSULTR3;4 (SULTR-like P Distribution Transporter), a homolog of rice OsSPDT. Like OsSPDT, AtSPDT is localized at the plasma membrane and showed proton-dependent transport activity for P. Interestingly, we found that AtSPDT is mainly expressed in the rosette basal region and leaf petiole, and its expression is up-regulated by P deficiency. Tissue-specific analysis showed that AtSPDT is mainly located in the vascular cambium of different organs, as well as in the parenchyma tissues of both xylem and phloem regions. Knockout of AtSPDT inhibited the growth of new leaves under low P due to decreased P distribution to those organs. The seed yields of the wild-type and atspdt mutant plants are similar, but the seeds of mutant plants contain – less P. These results indicate that AtSPDT localized in the vascular cambium is involved in preferential distribution of P to the developing tissues, through xylem-to-phloem transfer mainly at the rosette basal region and leaf petiole.

Common mycorrhizal networks influence the distribution of mineral nutrients between an invasive plant, Solidago canadensis, and a native plant, Kummerowa striata.

Invasive species often reduce ecosystem services and lead to a serious threat to native biodiversity. Roots of invasive plants are often linked to roots of native plants by common mycorrhizal networks (CMNs) of arbuscular mycorrhizal (AM) fungi, but whether and how CMNs mediate interactions between invasive and native plant species remains largely uninvestigated. We conducted two microcosm experiments, one in which we amended the soil with mineral N and another in which we amended the soil with mineral P. In each experiment, we grew a pair of test plants consisting of Kummerowia striata (native to our research site) and Solidago canadensis (an invasive species). CMNs were established between the plants, and these were either left intact or severed. Intact CMNs increased growth and nutrient acquisition by S. canadensis while they decreased nutrient acquisition by K. striata in comparison with severed CMNs. 15N and P analyses indicated that compared to severed CMNs, intact CMNs preferentially transferred mineral nutrients to S. canadensis. CMNs produced by different species of AM fungi had slightly different effects on the interaction between these two plant species. These results highlight the role of CMNs in the understanding of interactions between the invasive species S. canadensis and its native neighbor.

Arbuscular common mycorrhizal networks mediate intra- and interspecific interactions of two prairie grasses.

Arbuscular mycorrhizal fungi form extensive common mycorrhizal networks (CMNs) that may interconnect neighboring root systems of the same or different plant species, thereby potentially influencing the distribution of limiting mineral nutrients among plants. We examined how CMNs affected intra- and interspecific interactions within and between populations of Andropogon gerardii, a highly mycorrhiza dependent, dominant prairie grass and Elymus canadensis, a moderately dependent, subordinate prairie species. We grew A. gerardii and E. canadensis alone and intermixed in microcosms, with individual root systems isolated, but either interconnected by CMNs or with CMNs severed weekly. CMNs, which provided access to a large soil volume, improved survival of both A. gerardii and E. canadensis, but intensified intraspecific competition for A. gerardii. When mixed with E. canadensis, A. gerardii overyielded aboveground biomass in the presence of intact CMNs but not when CMNs were severed, suggesting that A. gerardii with intact CMNs most benefitted from weaker interspecific than intraspecific interactions across CMNs. CMNs improved manganese uptake by both species, with the largest plants receiving the most manganese. Enhanced growth in consequence of improved mineral nutrition led to large E. canadensis in intact CMNs experiencing water-stress, as indicated by 13C isotope abundance. Our findings suggest that in prairie plant communities, CMNs may influence mineral nutrient distribution, water relations, within-species size hierarchies, and between-species interactions.

‘Chemlali’ olive trees in an irrigated grove: effects of tree age on virgin olive oil quality and mineral nutrient distribution in roots, leaves and fruits

Introduction. The effects of tree age on the quality of olive oil are not very clear. Data on the mineral element content in different plant organs of cv. Chemlali are also very scarce, and the correlation between tree age and mineral content is still unclear. Materials and methods. A two-year study was conducted on irrigated ‘Chemlali’ olive trees of two different ages (12 years old vs. 24 years old) in order to verify possible effects on either the nutritional balance in different plant organs (leaves, roots, fruits) or the virgin olive oil (VOO) quality. Results and discussion. The 24-year-old trees showed a higher mineral concentration with respect to 12-year-old trees in roots and fruits, whereas the opposite was reported for the leaves. Total phenols and induction time values were significantly lower in VOO from 24-year-old trees. Palmitic, palmitoleic, stearic and linoleic acid concentrations were higher in VOO of 24-year-old trees. Conclusion. These results can be useful in order to discriminate the characteristics of VOO obtained from trees of different ages and the mineral content data can be used for the optimization of the fertilization schedules with respect to the olive tree age for more sustainable management of olive groves.

Comparison of the distribution of mineral nutrients and active substances in dehydration- and salinity-stressed Aloe vera L.

ABSTRACTChanges in growth and distribution of inorganic mineral elements and organic active substances in seedlings of A. vera were examined under treatments of −0.44 and −0.88 MPa polyethylene glycol 6000 (PEG) and corresponding isosmotic sodium chloride (NaCl). All stress treatments inhibited plant growth, with NaCl severely inhibiting the growth of the seedlings than PEG. However, leaf biomass did not change significantly. The nitrogen concentration under PEG was maintained higher than that under NaCl. Salinity caused more severe ionic imbalance than dehydration. However, when the concentration of NaCl was increased, ion homeostasis was maintained. Stressors induced an increase in aloin concentration in leaves, especially in young leaves. Polysaccharide concentrations in leaves did not change under dehydration. However, the polysaccharide concentration decreased in the middle and base leaves under NaCl. A. vera exhibited high adaptability to suitable osmotic stress, which increases its main active ingredients in leaves.

Natural genetic variation of Arabidopsis thaliana root morphological response to magnesium supply

Plants dynamically cope with the variability of mineral nutrient distribution in soil by constantly modulating nutrient uptake and shaping root-system architecture. The changes in root morphology in response to major essential elements are largely documented, but little is known about how the root system responds to magnesium (Mg) availability. Thirty-six natural accessions of the model species Arabidopsis thaliana were subjected to an in vitro screen for identifying variation in root system architecture in response to Mg availability. Response of root morphology was observed on 2-dimensional agar plates. Low Mg supply repressed the elongation of the lateral roots more than of the primary root. However, some accessions exhibited higher number and length of lateral roots than the reference Columbia-0. Across all accessions, the root morphological traits did not correlate with tissue Mg concentrations. Interestingly, shoot calcium and root phosphorus concentrations were positively correlated with the number and length of lateral roots, whereas root iron concentration was negatively correlated with the primary root length. The diversity of root phenotypes identified in this report is a useful resource to study the genetic component determining root morphology in response to Mg availability.
