Advantages and prospects of cell derived decellularized extracellular matrix as tissue engineering scaffolds

To review the application of cell derived decellularized extracellular matrix (CDM) in tissue engineering. The literature related to the application of CDM in tissue engineering was extensively reviewed and analyzed. CDM is a mixture of cells and their secretory products obtained by culturing cells in vitro for a period of time, and then the mixture is treated by decellularization. Compared with tissue derived decellularized extracellular matrix (TDM), CDM can screen and utilize pathogen-free autologous cells, effectively avoiding the possible shortcomings of TDM, such as immune response and limited sources. In addition, by selecting the cell source, controlling the culture conditions, and selecting the template scaffold, the composition, structure, and mechanical properties of the scaffold can be controlled to obtain the desired scaffold. CDM retains the components and microstructure of extracellular matrix and has excellent biological functions, so it has become the focus of tissue engineering scaffolds. CDM is superior in the field of tissue engineering because of its outstanding adjustability, safety, and high bioactivity. With the continuous progress of technology, CDM stents suitable for clinical use are expected to continue to emerge.

PBF-LB fabrication of microgrooves for induction of osteogenic differentiation of human mesenchymal stem cells

Stem cell differentiation has important implications for biomedical device design and tissue engineering. Recently, inherent material properties, including surface chemistry, stiffness, and topography, have been found to influence stem cell fate. Among these, surface topography is a key regulator of stem cells in contact with materials. The most important aspect of ideal bone tissue engineering is to control the organization of the bone extracellular matrix with fully differentiated osteoblasts. Here, we found that laser powder bed fusion (PBF-LB)-fabricated grooved surface inspired by the microstructure of bone, which induced human mesenchymal stem cell (hMSC) differentiation into the osteogenic lineage without any differentiation supplements. The periodic grooved structure was fabricated by PBF-LB which induced cell elongation facilitated by cytoskeletal tension along the grooves. This resulted in the upregulation of osteogenesis via Runx2 expression. The aligned hMSCs successfully differentiated into osteoblasts and further organized the bone mimetic-oriented extracellular matrix microstructure. Our results indicate that metal additive manufacturing technology has a great advantage in controlling stem cell fate into the osteogenic lineage, and in the construction of bone-mimetic microstructural organization. Our findings on material-induced stem cell differentiation under standard cell culture conditions open new avenues for the development of medical devices that realize the desired tissue regeneration mediated by regulated stem cell functions.

ECM-Engineered Electrospun Fibers with an Immune Cascade Effect for Inhibiting Tissue Fibrosis.

Tissue regeneration/fibrosis after injury is intricately regulated by the immune cascade reaction and extracellular matrix (ECM). Dysregulated cascade signal could jeopardize tissue homeostasis leading to fibrosis. Bioactive scaffolds mimicking natural ECM microstructure and chemistry could regulate the cascade reaction to achieve tissue regeneration. The current study constructed an ECM-engineered micro/nanofibrous scaffold using self-assembled nanofibrous collagen and decorin (DCN)-loaded microfibers to regulate the immune cascade reaction. The ECM-engineered scaffold promoted anti-inflammatory and pro-regenerative effects, M2 polarization of macrophages, by nanofibrous collagen. The ECM-engineered scaffold could release DCN to inhibit inflammation-associated fibrous angiogenesis. Yet, to prevent excessive M2 activity leading to tissue fibrosis, controlled release of DCN was expected to elicit M1 activity and achieve M1/M2 balance in the repair process. Regulated cascade reaction guided favorable crosstalk between macrophages, endothelial cells and fibroblasts by proximity. Additionally, decorin could also antagonize TGF-β1 via TGF-β/Smad3 pathway to suppress fibrotic activity of fibroblasts. Hence, ECM-engineered scaffolds could exert effective regulation of the immune cascade reaction by microstructure and DCN release and achieve the balance between tissue fibrosis and regeneration. STATEMENT OF SIGNIFICANCE: : With the incidence of up to 74.6%, failed back surgery syndrome (FBSS) has been a lingering issue in spine surgery, which poses a heavy socio-economic burden to society. Epidural fibrosis is believed to be responsible for the onset of FBSS. Current biomaterial-based strategies treating epidural fibrosis mainly rely on physical barriers and unidirectional suppression of inflammation. Regulation of the immune cascade reaction for inhibiting fibrosis has not been widely studied. Based on the simultaneous regulation of M1/M2 polarization and intercellular crosstalk, the ECM-engineered micro/nanofibrous scaffolds constructed in the current study could exert an immune cascade effect to coordinate tissue regeneration and inhibit fibrosis. This finding makes a significant contribution in the development of a treatment for epidural fibrosis and FBSS.

A cervical cancer tissue-derived decellularized extracellular matrix scaffold for cervical cancer tissue reconstruction in vitro

The prepare decellularized extracellular matrix (ECM) scaffold materials derived from human cervical carcinoma tissues for 3D culture of cervical carcinoma cells. Fresh human cervical carcinoma tissues were treated with sodium lauryl ether sulfate (SLES) solution to prepare decellularized ECM scaffolds. The scaffolds were examined for ECM microstructure and residual contents of key ECM components (collagen, glycosaminoglycan, and elastin) and genetic materials by pathological staining and biochemical content analysis. In vitro 3D culture models were established by injecting cultured cervical cancer cells into the prepared ECM scaffolds. The cells in the recellularized scaffolds were compared with those in a conventional 2D culture system for cell behaviors including migration, proliferation and epithelial-mesenchymal transition (EMT) wsing HE staining, immunohistochemical staining and molecular biological technology analysis. Resistance to 5-fluorouracil (5-Fu) of the cells in the two culture systems was tested by analyzing the cell apoptosis rates via flow cytometry. SLES treatment effectively removed cells and genetic materials from human cervical carcinoma tissues but well preserved the microenvironment structure and biological activity of ECM. Compared with the 2D culture system, the 3D culture models significantly promoted proliferation, migration, EMT and 5-Fu resistance of human cervical cancer cells. The decellularized ECM scaffolds prepared using human cervical carcinoma tissues provide the basis for construction of in vitro 3D culture models for human cervical cancer cells.

Hydrogel-based microenvironment engineering of haematopoietic stem cells.

Haematopoietic Stem cells (HSCs) have the potential for self-renewal and multilineage differentiation, and their behaviours are finely tuned by the microenvironment. HSC transplantation (HSCT) is widely used in the treatment of haematologic malignancies while limited by the quantity of available HSCs. With the development of tissue engineering, hydrogels have been deployed to mimic the HSC microenvironment in vitro. Engineered hydrogels influence HSC behaviour by regulating mechanical strength, extracellular matrix microstructure, cellular ligands and cytokines, cell-cell interaction, and oxygen concentration, which ultimately facilitate the acquisition of sufficient HSCs. Here, we review recent advances in the application of hydrogel-based microenvironment engineering of HSCs, and provide future perspectives on challenges in basic research and clinical practice.

ECM roles and biomechanics in cardiac tissue decellularization.

Natural biomaterials hold enormous potential for tissue regeneration. The rapid advance of several tissue-engineered biomaterials, such as natural and synthetic polymer-based scaffolds, has led to widespread application of these materials in the clinic and in research. However, biomaterials can have limited repair capacity; obstacles result from immunogenicity, difficulties in mimicking native microenvironments, and maintaining the mechanical and biochemical (i.e., biomechanical) properties of native organs/tissues. The emergence of decellularized extracellular matrix (ECM)-derived biomaterials provides an attractive solution to overcome these hurdles since decellularized ECM provides a nonimmune environment with native three-dimensional structures and bioactive components. More importantly, decellularized ECM can be generated from the tissue of interest, such as the heart, and keep its native macro- and microstructure and tissue-specific composition. These decellularized cardiac matrices/scaffolds can then be reseeded using cardiac cells, and the resulting recellularized construct is considered an ideal choice for regenerating functional organs/tissues. Nonetheless, the decellularization process must be optimized and depends on tissue type, age, and functional goal. Although most decellularization protocols significantly reduce immunogenicity and deliver a matrix that maintains the tissue macrostructure, suboptimal decellularization can change ECM composition and microstructure, which affects the biomechanical properties of the tissue and consequently changes cell-matrix interactions and organ function. Herein, we review methods of decellularization, with particular emphasis on cardiac tissue, and how they can affect the biomechanics of the tissue, which in turn determines success of reseeding and in vivo viability. Moreover, we review recent developments in decellularized ECM-derived cardiac biomaterials and discuss future perspectives.

Electroactive extracellular Matrix/Polypyrrole composite films and their microenvironmental effects on osteogenic differentiation of BMSCs

Bone marrow mesenchymal stem cells (BMSCs) could be regulated through their microenvironment, which includes biological cues of extracellular matrix (ECM) and other endogenous physical cues. In this work, cell-derived ECM was used as a framework to prepare extracellular matrix/polypyrrole composite films by chemical polymerization of polypyrrole (Ppy). The composite film retained the microstructure of ECM, while the microscopic surface exhibited dual-exposure of ECM and Ppy. The content of Ppy had a decisive influence on the protein exposure area and electrochemical performance to generate different biological or electrical microenvironment. It was worth noting that the ECM/Ppy composite films with appropriate Ppy content had unique biological microenvironments that could promote the osteogenic differentiation of BMSCs. In addition, the electrochemical performance of ECM/Ppy composite films had been significantly improved compared with ECM, which meant that they were electroactive. When BMSCs were simultaneously affected by biological microenvironment and electrical microenvironment, this new composite electroactive film showed the best ability to promote osteogenic differentiation. Correspondingly, the microenvironment provided by ECM/Ppy composite films significantly up-regulated the expression of genes related to osteogenic differentiation of ALP, Col-I, and RunX-2. Furthermore, the electrical microenvironment had been shown to regulate the osteogenic differentiation of BMSCs through the influence on the intracellular calcium ion (Ca2+) concentration and related signal pathways. This new composite electroactive biomaterial has unique biochemical properties and good electrochemical properties to construct biological and electrical microenvironment, having great potential in research and application of bone tissue regeneration and regenerative medicine.

Three-dimensional cancer cell migration directed by dual mechanochemical guidance.

Directed cell migration guided by external cues plays a central role in many physiological and pathophysiological processes. The microenvironment of cells often simultaneously contains various cues and the motility response of cells to multiplexed guidance is poorly understood. Here we combine experiments and mathematical models to study the three-dimensional migration of breast cancer cells in the presence of both contact guidance and a chemoattractant gradient. We find that the chemotaxis of cells is complicated by the presence of contact guidance as the microstructure of extracellular matrix (ECM) vary spatially. In the presence of dual guidance, the impact of ECM alignment is determined externally by the coherence of ECM fibers and internally by cell mechanosensing Rho/Rock pathways. When contact guidance is parallel to the chemical gradient, coherent ECM fibers significantly increase the efficiency of chemotaxis. When contact guidance is perpendicular to the chemical gradient, cells exploit the ECM disorder to locate paths for chemotaxis. Our results underscore the importance of fully characterizing the cancer cell microenvironment in order to better understand invasion and metastasis.

Biomimetic Glycopolypeptide Hydrogels with Tunable Adhesion and Microporous Structure for Fast Hemostasis and Highly Efficient Wound Healing

AbstractDespite clinical applications of the first‐generation tissue adhesives and hemostats, the correlation among microstructure and hemostasis of hydrogels with wound healing is less understood and it is elusive to design high‐performance hydrogels to meet worldwide growing demands in wound closure, hemostasis, and healing. Inspired by the microstructure of extracellular matrix and mussel‐mimetic chemistry, two kinds of coordinated and covalent glycopolypeptide hydrogels are fabricated, which present tunable tissue adhesion strength (14.6–83.9 kPa) and microporous structure (8–18 µm), and lower hemolysis <1.5%. Remarkably, the microporous size mainly controls the hemostasis, and those hydrogels with larger pores of 16–18 µm achieve the fastest hemostasis of ≈14 s and the lowest blood loss of ≈6% than fibrin glue and others. Moreover, both biocompatibility and hemostasis affect wound healing performance, as assessed by hemolysis, cytotoxicity, subcutaneous implantation, and hemostasis and healing assays. Importantly, the glycopolypeptide hydrogel‐treated rat‐skin defect model achieves full wound closure and regenerates thick dermis and epidermis with some hair follicles on day 14. Consequently, this work not only establishes a versatile method for constructing glycopolypeptide hydrogels with tunable adhesion and microporous structure, fast hemostasis, and superior healing functions, but also discloses a useful rationale for designing high‐performance hemostatic and healing hydrogels.

Multiple Particle Tracking Detects Changes in Brain Extracellular Matrix and Predicts Neurodevelopmental Age.

Brain extracellular matrix (ECM) structure mediates many aspects of neural development and function. Probing structural changes in brain ECM could thus provide insights into mechanisms of neurodevelopment, the loss of neural function in response to injury, and the detrimental effects of pathological aging and neurological disease. We demonstrate the ability to probe changes in brain ECM microstructure using multiple particle tracking (MPT). We performed MPT of colloidally stable polystyrene nanoparticles in organotypic rat brain slices collected from rats aged 14-70 days old. Our analysis revealed an inverse relationship between nanoparticle diffusive ability in the brain extracellular space and age. Additionally, the distribution of effective ECM pore sizes in the cortex shifted to smaller pores throughout development. We used the raw data and features extracted from nanoparticle trajectories to train a boosted decision tree capable of predicting chronological age with high accuracy. Collectively, this work demonstrates the utility of combining MPT with machine learning for measuring changes in brain ECM structure and predicting associated complex features such as chronological age. This will enable further understanding of the roles brain ECM play in development and aging and the specific mechanisms through which injuries cause aberrant neuronal function. Additionally, this approach has the potential to develop machine learning models capable of detecting the presence of injury or indicating the extent of injury based on changes in the brain microenvironment microstructure.

Mechanical response of composite fiber networks subjected to local contractile deformation

The extracellular matrix (ECM), forming the cell microenvironment, is a random fiber network whose heterogeneous microstructure serves a vital role in cellular functions such as migration, proliferation, and differentiation. Although prior computational studies used a network of randomly crosslinked fibers for investigating the ECM mechanical response due to the cell contraction, we hereby investigate the cell-induced deformation by considering the interplay between the collagen fibers and soft proteoglycan matrix domain. For this purpose, we represent the complex ECM microstructure by a double network model in which a random network of relatively stiff fibers (representing collagens) is embedded in a homogeneous network (representing the matrix domain). We demonstrate that the interaction between the network of stiff fibers and the network of matrix fibers plays a significant role in the propagation of cell-induced displacements. In particular, as the stiffness of the matrix domain increases, the long-range displacement propagation becomes suppressed. Furthermore, the degree of inhomogeneity in the displacement propagation is controlled by the relative stiffness of fiber and matrix networks. The influence of matrix stiffness becomes prominent for composite networks with random networks having a low average network connectivity and/or composed of fibers with low bending rigidity.

Scaffolds made from natural or synthetic matrix have many shortcomings in providing ideal extracellular matrix microstructure for cell proliferation and differentiation. Deselularized native scaffold can maintain the extracellular matrix of native organs, as an ideal microstructure scaffold for cell proliferation and differentiation. This technique answers the challenge of high need for liver transplants due to the limitations of liver donors and the need as liver models for drug testing.for drug testing. Liver is a complex organ, so ideal microstructure is needed in liver organoid reconstruction techniques to produce good synthetic and metabolic functions of the liver.

A computational study of amoeboid motility in 3D: the role of extracellular matrix geometry, cell deformability, and cell-matrix adhesion.

Amoeboid cells often migrate using pseudopods, which are membrane protrusions that grow, bifurcate, and retract dynamically, resulting in a net cell displacement. Many cells within the human body, such as immune cells, epithelial cells, and even metastatic cancer cells, can migrate using the amoeboid phenotype. Amoeboid motility is a complex and multiscale process, where cell deformation, biochemistry, and cytosolic and extracellular fluid motions are coupled. Furthermore, the extracellular matrix (ECM) provides a confined, complex, and heterogeneous environment for the cells to navigate through. Amoeboid cells can migrate without significantly remodeling the ECM using weak or no adhesion, instead utilizing their deformability and the microstructure of the ECM to gain enough traction. While a large volume of work exists on cell motility on 2D substrates, amoeboid motility is 3D in nature. Despite recent progress in modeling cellular motility in 3D, there is a lack of systematic evaluations of the role of ECM microstructure, cell deformability, and adhesion on 3D motility. To fill this knowledge gap, here we present a multiscale, multiphysics modeling study of amoeboid motility through 3D-idealized ECM. The model is a coupled fluid‒structure and coarse-grain biochemistry interaction model that accounts for large deformation of cells, pseudopod dynamics, cytoplasmic and extracellular fluid motion, stochastic dynamics of cell-ECM adhesion, and microstructural (pore-scale) geometric details of the ECM. The key finding of the study is that cell deformation and matrix porosity strongly influence amoeboid motility, while weak adhesion and microscale structural details of the ECM have secondary but subtle effects.

Decellularizing the Porcine Optic Nerve Head: Toward a Model to Study the Mechanobiology of Glaucoma.

PurposeStudying the extracellular matrix (ECM) remodeling of the lamina cribrosa in vivo can be extremely challenging and costly. There exist very few options for studying optic nerve head (ONH) mechanobiology in vitro that are able to reproduce the complex anatomic and biomechanical environment of the ONH. Herein, we have developed a decellularization procedure that will enable more anatomically relevant and cost-efficient future studies of ECM remodeling of the ONH.MethodsPorcine posterior poles were decellularized using a detergent and enzyme-based decellularization protocol. DNA quantification and histology were used to investigate the effectiveness of the protocol. We subsequently investigated the ability of a polyethylene glycol (PEG)-based hydrogel to restore the ONH's ability to hold pressure following decellularization. Anterior-posterior displacement of the decellularized and PEG treated ONH in a pressure bioreactor was used to evaluate the biomechanical response of the ONH.ResultsDNA quantification and histology confirmed decellularization using Triton X-100 at low concentration for 48 hours successfully reduced the cellular content of the tissue by 94.9% compared with native tissue while preserving the ECM microstructure and basal lamina of the matrix. Infiltrating the decellularized tissues with PEG 6000 and PEG 10,000 hydrogel restored their ability to hold pressure, producing displacements similar to those measured for the non-decellularized control samples.ConclusionsOur decellularized ONH model is capable of producing scaffolds that are cell-free and maintain the native ECM microstructure.Translational RelevanceThis model represents a platform to study the mechanobiology in the ONH and potentially for glaucoma drug testing.

Role of the Extracellular Matrix in Loss of Muscle Force With Age and Unloading Using Magnetic Resonance Imaging, Biochemical Analysis, and Computational Models.

The focus of this review is the application of advanced MRI to study the effect of aging and disuse related remodeling of the extracellular matrix (ECM) on force transmission in the human musculoskeletal system. Structural MRI includes (i) ultra-low echo times (UTE) maps to visualize and quantify the connective tissue, (ii) diffusion tensor imaging (DTI) modeling to estimate changes in muscle and ECM microstructure, and (iii) magnetization transfer contrast imaging to quantify the macromolecular fraction in muscle. Functional MRI includes dynamic acquisitions during contraction cycles enabling computation of the strain tensor to monitor muscle deformation. Further, shear strain extracted from the strain tensor may be a potential surrogate marker of lateral transmission of force. Biochemical and histological analysis of muscle biopsy samples can provide “gold-standard” validation of some of the MR findings. The review summarizes biochemical studies of ECM adaptations with age and with disuse. A brief summary of animal models is included as they provide experimental confirmation of longitudinal and lateral force transmission pathways. Computational muscle models enable exploration of force generation and force pathways and elucidate the link between structural adaptations and functional consequences. MR image findings integrated in a computational model can explain and predict subject specific functional changes to structural adaptations. Future work includes development and validation of MRI biomarkers using biochemical analysis of muscle tissue as a reference standard and potential translation of the imaging markers to the clinic to noninvasively monitor musculoskeletal disease conditions and changes consequent to rehabilitative interventions.

Microstructural densification and alignment by aspiration-ejection influence cancer cell interactions with three-dimensional collagen networks.

Extracellular matrix microstructure and mechanics are crucial to breast cancer progression and invasion into surrounding tissues. The peritumor collagen network is often dense and aligned, features which in vitro models lack. Aspiration of collagen hydrogels led to densification and alignment of microstructure surrounding embedded cancer cells. Two metastasis-derived breast cancer cell lines, MDA-MB-231 and MCF-7, were cultured in initially 4 mg/ml collagen gels for 3 days after aspiration, as well as in unaspirated control hydrogels. Videomicroscopy during aspiration, and at 0, 1, and 3 days after aspiration, epifluorescence microscopy of phalloidin-stained F-actin cytoskeleton, histological sections, and soluble metabolic byproducts from constructs were collected to characterize effects on the embedded cell morphology, the collagen network microstructure, and proliferation. Breast cancer cells remained viable after aspiration-ejection, proliferating slightly less than in unaspirated gels. Furthermore, MDA-MB-231 cells appear to partially relax the collagen network and lose alignment 3 days after aspiration. Aspiration-ejection generated aligned, compact collagen network microstructure with immediate cell co-orientation and higher cell number density apparently through purely physical means, though cell-collagen contact guidance and network remodeling influence cell organization and collagen network microstructure during subsequent culture. This study establishes a platform to determine the effects of collagen density and alignment on cancer cell behavior, with translational potential for anticancer drug screening in a biomimetic three-dimensional matrix microenvironment, or implantation in preclinical models.

Biaxial Basal Tone and Passive Testing of the Murine Reproductive System Using a Pressure Myograph.

The female reproductive organs, specifically the vagina and cervix, are composed of various cellular components and a unique extracellular matrix (ECM). Smooth muscle cells exhibit a contractile function within the vaginal and cervical walls. Depending on the biochemical environment and the mechanical distension of the organ walls, the smooth muscle cells alter the contractile conditions. The contribution of the smooth muscle cells under baseline physiological conditions is classified as a basal tone. More specifically, a basal tone is the baseline partial constriction of smooth muscle cells in the absence of hormonal and neural stimulation. Furthermore, the ECM provides structural support for the organ walls and functions as a reservoir for biochemical cues. These biochemical cues are vital to various organ functions, such as inciting growth and maintaining homeostasis. The ECM of each organ is composed primarily of collagen fibers (mostly collagen types I, III, and V), elastic fibers, and glycosaminoglycans/proteoglycans. The composition and organization of the ECM dictate the mechanical properties of each organ. A change in ECM composition may lead to the development of reproductive pathologies, such as pelvic organ prolapse or premature cervical remodeling. Furthermore, changes in ECM microstructure and stiffness may alter smooth muscle cell activity and phenotype, thus resulting in the loss of the contractile force. In this work, the reported protocols are used to assess the basal tone and passive mechanical properties of the nonpregnant murine vagina and cervix at 4-6 months of age in estrus. The organs were mounted in a commercially available pressure myograph and both pressure-diameter and force-length tests were performed. Sample data and data analysis techniques for the mechanical characterization of the reproductive organs are included. Such information may be useful for constructing mathematical models and rationally designing therapeutic interventions for women's health pathologies.

Noninvasive ultrasound imaging for assessment of intact microstructure of extracellular matrix in tissue engineering.

Development of artificial tissues or organs is one of the actual tasks in regenerative medicine that requires observation and evaluation of intact volume microstructure of tissue engineering products at all stages of their formation, from native donor tissues and decellularized scaffolds to recipient cell migration in the matrix. Unfortunately in practice, methods of vital noninvasive imaging of volume microstructure in matrixes are absent. In this work, we propose a new approach based on high-frequency acoustic microscopy for noninvasive evaluation and visualization of volume microstructure in tissue engineering products. The results present the ultrasound characterization of native rat diaphragms and lungs and their decellularized scaffolds. Verification of the method for visualization of tissue formation in the matrix volume was described in the model samples of diaphragm scaffolds with stepwise collagenization. Results demonstrate acoustic microscopic sensitivity to cell content concentration, variation inlocal density, and orientation of protein fibers in the volume, micron air inclusions, and other inhomogeneities of matrixes.

Benzalkonium bromide is effective in removing Bacillus cereus biofilm on stainless steel when combined with cleaning-in-place

Bacillus cereus is a common foodborne pathogen in dairy environment, which is hard to remove for its strong biofilm forming ability. In our study, Clean-in-place (CIP) regime and benzalkonium bromide (BK) were used in combination to enhance biofilm removal effectiveness. The CIP regime (1.5% NaOH at 65 °C for 10 min-water rinse-1% HNO3 at 65 °C for 10 min-water rinse) often used in dairy industry only achieved a 4.69 log reduction/cm2. A step of 5000 μg/mL BK following this CIP regime could effectively remove the biofilm cells on stainless steel, and biofilm cells were removed to below detection limit. The efficacy of BK in inhibiting Bacillus cereus was investigated. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of BK against Bacillus cereus were 6.25 μg/mL and 25 μg/mL. The antimicrobial activity of BK took effect by inducing intracellular proteins and electrolytes leakage. Moreover, the microstructure of extracellular matrix was broken under confocal microscopy (CLSM) and the amounts of proteins, polysaccharides and DNA in extracellular matrix reduced by 44.8%, 69.2% and 60.9% respectively after treatment of 5000 μg/mL BK. By detecting the zeta potential of biofilm extracellular matrix and contact angle assay, we found BK could bind to extracellular matrix with electrostatic interaction and increasing the hydrophilicity of biofilm, making biofilm removal easier. So, BK combined with CIP regime could be used as an effective strategy to remove Bacillus cereus biofilm.

A novel three-dimensional culture system maintaining the physiological extracellular matrix of fibrotic model livers accelerates progression of hepatocellular carcinoma cells

Liver fibrosis is characterized by the progressive accumulation of extracellular matrix (ECM) and is a strong predictor of hepatocellular carcinoma (HCC) development and progression. However, the effect of ECM in fibrotic livers on HCC cells is poorly understood. The aims of this study were to create a new culture system that retained the natural ECM of fibrotic model livers and to establish whether natural ECM regulated the characteristics of HCC cells. Using an organ decellularization technique, we created a new culture system that preserved the tissue-specific ECM of fibrotic model livers from CCl4-treated rats. The content of ECM in fibrotic model liver scaffolds was increased and the ECM microstructure was distorted. Quantitative polymerase chain reaction and immunofluorescence assays of HCC cells cultured in fibrotic model liver scaffolds for 7 days showed an epithelial-mesenchymal transition phenotype. Moreover, the ECM of fibrotic model livers promoted proliferation and chemoresistance of HCC cells. These results showed a novel effect of natural ECM in fibrotic model livers on the malignant behaviour of HCC cells. This new culture system will be useful for both understanding the cell biology of fibrotic livers and developing novel anti-cancer drugs.
