Enhanced microbial remediation of uranium tailings through red soil utilization

Seepage of uranium tailings has become a focus of attention in the uranium mining and metallurgy industry, and in-situ microbial remediation is considered an effective way to treat uranium pollution. However, this method has the drawbacks of easy biomass loss and unstable remediation effect. To overcome these issues, spare red soil around the uranium mine was used to enhance the efficiency and stability of bioremediation. Furthermore, the bioremediation mechanism was revealed by employing XRD, FTIR, XPS, and 16S rRNA. The results showed that red soil, as a barrier material, had the adsorption potential of 8.21∼148.00 mg U/kg soil, but the adsorption is accompanied by the release of certain acidic and oxidative substances. During the dynamic microbial remediation, red soil was used as a cover material to neutralize acidity, provide a higher reduction potential (<-200 mV), and increase the retention rate of microbial agent (19.06 mL/d) compared to the remediation group without red soil. In the presence of red soil, the anaerobic system could maintain the uranium concentration in the solution below 0.3 mg/L for more than 70 days. Moreover, the generation of new clay minerals driven by microorganisms was more conducive to the stability of uranium tailings. Through alcohol and amino acid metabolism of microorganisms, a reducing environment with reduced valence states of multiple elements (such as S2-, Fe2+, and U4+) was formed. At the same time, the relative abundance of functional microbial communities in uranium tailings improved in presence of red soil and Desulfovirobo, Desulfocapsa, Desulfosporosinus, and other active microbial communities reconstructed the anaerobic environment. The study provides a new two-in-one solution for treatment of uranium tailings and resource utilization of red soil through in-situ microbial remediation.

Examining the potential impacts of a coastal renourishment project on the presence and abundance of Escherichia coli.

Erosion poses a significant threat to oceanic beaches worldwide. To combat this threat, management agencies often utilize renourishment, which supplements eroded beaches with offsite sand. This process can alter the physical characteristics of the beach and can influence the presence and abundance of microbial communities. In this study, we examined how an oceanic beach renourishment project may have impacted the presence and abundance of Escherichia coli (E. coli), a common bacteria species, and sand grain size, a sediment characteristic that can influence bacterial persistence. Using an observational field approach, we quantified the presence and abundance of E. coli in sand (from sub-tidal, intertidal, and dune zones on the beach) and water samples at study sites in both renourished and non-renourished sections of Folly Beach, South Carolina, USA in 2014 and 2015. In addition, we also measured how renourishment may have impacted sand grain size by quantifying the relative frequency of grain sizes (from sub-tidal, intertidal, and dune zones on the beach) at both renourished and non-renourished sites. Using this approach, we found that E. coli was present in sand samples in all zones of the beach and at each of our study sites in both years of sampling but never in water samples. Additionally, we found that in comparison to non-renourished sections, renourished sites had significantly higher abundances of E. coli and coarser sand grains in the intertidal zone, which is where renourished sand is typically placed. However, these differences were only present in 2014 and were not detected when we resampled the study sites in 2015. Collectively, our findings show that E. coli can be commonly found in this sandy beach microbial community. In addition, our results suggest that renourishment has the potential to alter both the physical structure of the beach and the microbial community but that these impacts may be short-lived.

In situ aerobic methane oxidation rates in a stratified lake

AbstractMicrobial aerobic methane oxidation is an important sink for aquatic methane worldwide. Despite its importance to global methane fluxes, few aerobic methane oxidation rates have been obtained in freshwater or marine environments without imposing changes to the microbial community through use of ex situ methods. A novel in situ incubation method for continuous time‐series measurements was used in Jordan Lake, North Carolina, during 2020–2021, to determine reaction kinetics for aerobic methane oxidation rates across a wide range of naturally varying methane (55–1833 nM) and dissolved oxygen (DO; 28–366 μM) concentrations and temperatures (17–30°C). Methane oxidation began immediately at the start of each of 21 incubations and methane oxidation rates were 1st order with respect to methane. The data density allowed for accurate calculation of 1st‐order rate constants, k, that ranged from 0.018 to 0.462 h−1 (R2 &gt; 0.967). Addition of ammonium (20–45 μM) to natural concentrations ranging from 0.057 to 2.4 μM did not change aerobic methane oxidation rate kinetics, suggesting that the natural population of aerobic methane oxidizers in this eutrophic lake was not nitrogen limited. Values of k inversely correlated most strongly with initial DO concentrations (R2 = 0.82) rather than temperature. Values for k increased with Julian day throughout our sampling period, suggesting seasonal influences on methane oxidation via responses to geochemical changes or shifts in microbial community abundance and composition. These experiments demonstrate a high variability in the enzymatic capacity for 1st‐order methane oxidation rates in this eutrophic lake that is tightly and inversely coupled to oxygen concentrations. Measurements of in situ aerobic methane oxidation rate constants allow for the direct quantification and modeling of the microbial community's capacity for methane oxidation over a wide range of natural methane concentrations.

Heavy metals in a typical industrial area-groundwater system: Spatial distribution, microbial response and ecological risk

The environmental burden due to industrial activities has been quite observable in the last few years, with heavy metals (HMs) like lead, cadmium, and arsenic inducing serious perturbations to the microbial ecosystem of groundwater. Studies carried out in North China, a region known for interconnection of industrial and groundwater systems, sought to explore the natural mechanisms of adaptation of microbes to groundwater contamination. The results showed that heavy metals permeate from surface increased the diversity and abundance of microbial communities in groundwater, producing an average decrease of 40.84% and 34.62% in the relative abundance of Bacteroidetes and Proteobacteria in groundwater, respectively. Meanwhile, the key environmental factors driving the evolution of microbial communities shift from groundwater nutrients to heavy metals, which explained 50.80% of the change in the microbial community composition. Microbial indicators are more sensitive to HMs pollution and could accurately identify industrial area where HMs permeation occurred and other extraneous pollutants. The phylum Bacteroidetes could act as appropriate indicators for the identification. Significant genera that were identified, being Mesorhizobium, Clostridium, Bacillus and Mucilaginibacter, were found to play important roles in the microbial network in terms of the potential to assist in groundwater clean-up. Notably, pollution from heavy metals has diminished the effectiveness and resilience of microbial communities in groundwater, thereby heightening the susceptibility of these normally stable microbial ecosystems. These findings offer new perspectives on how to monitor and detect groundwater pollution, and provide scientific guidance for developing suitable remediation methods for groundwater contaminated with heavy metals. Future research is essential explore the application of metal-tolerant or resistant bacteria in bioremediation strategies to rehabilitate groundwater systems contaminated by HMs.

Prairie restoration promotes the abundance and diversity of mutualistic arbuscular mycorrhizal fungi.

Predicting how biological communities assemble in restored ecosystems can assist in conservation efforts, but most research has focused on plants, with relatively little attention paid to soil microbial organisms that plants interact with. Arbuscular mycorrhizal (AM) fungi are an ecologically significant functional group of soil microbes that form mutualistic symbioses with plants and could therefore respond positively to plant community restoration. To evaluate the effects of plant community restoration on AM fungi, we compared AM fungal abundance, species richness, and community composition of five annually cultivated, conventionally managed agricultural fields with paired adjacent retired agricultural fields that had undergone prairie restoration 5-9 years prior to sampling. We hypothesized that restoration stimulates AM fungal abundance and species richness, particularly for disturbance-sensitive taxa, and that gains of new taxa would not displace AM fungal species present prior to restoration due to legacy effects. AM fungal abundance was quantified by measuring soil spore density and root colonization. AM fungal species richness and community composition were determined in soils and plant roots using DNA high-throughput sequencing. Soil spore density was 2.3 times higher in restored prairies compared to agricultural fields, but AM fungal root colonization did not differ between land use types. AM fungal species richness was 2.7 and 1.4 times higher in restored prairies versus agricultural fields for soil and roots, respectively. The abundance of Glomeraceae, a disturbance-tolerant family, decreased by 25% from agricultural to restored prairie soils but did not differ in plant roots. The abundance of Claroideoglomeraceae and Diversisporaceae, both disturbance-sensitive families, was 4.6 and 3.2 times higher in restored prairie versus agricultural soils, respectively. Species turnover was higher than expected relative to a null model, indicating that AM fungal species were gained by replacement. Our findings demonstrate that restoration can promote a relatively rapid increase in the abundance and diversity of soil microbial communities that had been degraded by decades of intensive land use, and community compositional change can be predicted by the disturbance tolerance of soil microbial taxonomic and functional groups.

Epiphytic and endophytic bacteria on Camellia oleifera phyllosphere: exploring region and cultivar effect

The epiphytic and endophytic bacteria play an important role in the healthy growth of plants. Both plant species and growth environmental influence the bacterial population diversity, yet it is inconclusive whether it is the former or the latter that has a greater impact. To explore the communities of the epiphytic and endophytic microbes in Camellia oleifera, this study assessed three representative C. oleifera cultivars from three areas in Hunan, China by Illumina high-throughput sequencing. The results showed that the diversity and species richness of endophytic microbial community in leaves were significantly higher than those of microbial community in the epiphytic. The diversity and species richness of epiphytic and endophytic microbes are complex when the same cultivar was grown in different areas. The C. oleifera cultivars grown in Youxian had the highest diversity of epiphytic microbial community, but the lowest abundance, while the cultivars grown in Changsha had the highest diversity and species richness of endophytic microbes in leaves. It was concluded that the dominant phylum mainly included Proteobacteria, Actinobacteriota and Firmicutes through the analysis of the epiphytic and endophytic microbial communities of C. oleifera. The species and relative abundances of epiphytic and endophytic microbial community were extremely different at the genus level. The analysis of NMDS map and PERMANOVA shows that the species richness and diversity of microbial communities in epiphytes are greatly influenced by region. However, the community structure of endophytic microorganisms in leaves is influenced by region and cultivated varieties, but the influence of cultivars is more significant. Molecular ecological network analysis showed that the symbiotic interaction of epiphytic microbial community was more complex.

Linking phosphorus fertility to soil microbial diversity and network complexity in citrus orchards: Implications for sustainable agriculture

Continuous input of chemical fertilizers causes soil acidification, land degradation, and water eutrophication, but its impact on soil microbial communities and co-occurrence patterns of microboes is unclear. In this study, we evaluated the impact of chemical fertilizer on bacterial community diversity and ecological network at 75 citrus sites in a watershed where chemical fertilizer is continuously applied. In addition, 25 natural forest sites adjacent to citrus sites were used as references to compare the potential effects of land conversion from natural forests to citrus orchards on soil bacterial diversity and microbial network. The results showed that the cultivation of citrus results in soil phosphorus (P) accumulation, with significantly higher available and mineral-bound phosphate contents than those found in soils of natural forests. Citrus soil average soil available P was 73.2 mg/kg, which was 20 times higher than that in forest soils. As the P content accumulates, soil bacterial Shannon index linearly decreases from 7.06 to 5.93 and is significantly lower than that of adjacent natural forest soils (7.13). Low P fertility soils have higher microbial network complexity and stability, containing more microbial communities and tighter relationships between microbes compared to higher P fertility soils. Soil P content and soil pH regulates soil microbial network complexity and stability. In addition, soil bacterial community structure in soils of natural forests is significantly different from that in citrus soils, and the bacterial community structure in high P soils is different from that in medium and low P soils. Soil P fertility reduces the relative abundance of dominant communities including Proteobacteria, Bacteroidetes, and Gemmatimonadetes, and also changes the relative abundance of some functional microbial communities, such as some phosphorus cycling microorganisms, including Acetobacteraceae and Beijerinckiaceae. In conclusion, soil phosphate accumulation severely alters soil bacterial community diversity, complexity, stability, and functionality. This could have negative impacts on soil functional stability and the sustainability of citrus orchards.

Effect of Polyethylene Microplastics on the Microbial Community of Saline Soils

Mulching to conserve moisture has become an important agronomic practice in saline soil cultivation, and the effects of the dual stress of salinity and microplastics on soil microbes are receiving increasing attention. In order to investigate the effect of polyethylene microplastics on the microbial community of salinized soils, this study investigated the effects of different types (chloride and sulphate) and concentrations (weak, medium, and strong) of polyethylene (PE) microplastics (1% and 4% of the dry weight mass of the soil sample) on the soil microbial community by simulating microplastic contamination in salinized soil environments indoors. The results showed that:PE microplastics reduced the diversity and abundance of microbial communities in salinized soils and were more strongly affected by sulphate saline soil treatments. The relative abundance of each group of bacteria was more strongly changed in the sulphate saline soil treatment than in the chloride saline soil treatment. At the phylum level, the relative abundance of Proteobacteria was positively correlated with the abundance of fugitive PE microplastics, whereas the relative abundances of Bacteroidota, Actinobacteriota, and Acidobacteria were negatively correlated with the abundance of fugitive PE microplastics. At the family level, the relative abundances of Flavobacteriaceae, Alcanivoracaceae, Halomonadaceae, and Sphingomonasceae increased with increasing abundance of PE microplastics. The KEGG metabolic pathway prediction showed that the relative abundance of microbial metabolism and genetic information functions were reduced by the presence of PE microplastics, and the inhibition of metabolic functions was stronger in sulphate saline soils than in chloride saline soils, whereas the inhibition of genetic information functions was weaker than that in chloride saline soils. The secondary metabolic pathways of amino acid metabolism, carbohydrate metabolism, and energy metabolism were inhibited. It was hypothesized that the reduction in metabolic functions may have been caused by the reduced relative abundance of the above-mentioned secondary metabolic pathways. This study may provide a theoretical basis for the study of the effects of microplastics and salinization on the soil environment under the dual pollution conditions.

Effects of reduced snowpack due to climate warming on abiotic and biotic soil properties in alpine and boreal forest systems

Reduction in snow cover, depth, onset, and duration of seasonal snow in mid-latitude regions due to climate warming has multiple global and local scale ecosystem impacts. These effects include modulations of the hydrological cycles and increases in land surface solar radiation absorption due to decreased albedo. Changes in snow cover characteristics also affect underlying soils. Snow has an insulating effect on soils by decoupling air and soil temperatures, thus seasonal snow cover reduction leads to overall lower soil temperatures and an increase in freeze-thaw cycles. This is especially prominent during the fall and spring thaw seasons when the snow cover is not as extensive. This in turn has downstream impacts on soil physical, chemical, and biological properties. Among these impacts are soil moisture reduction, temperature, frost regimes, soil pH shifts, and alteration in nutrient flux dynamics during winter, snowmelt period and the following summer growing season. These changes in soil physicochemical properties due to snowpack reduction can then impact the biological soil properties via increased plant root mortality, reduced abundance and diversity of soil arthropods, and shifts in composition, abundance and activity of soil microbial communities. All these soil biotic factors can in turn alter the dynamics of soil nutrient fluxes and future greenhouse gas emissions. Here, we integrate data on the effects of snow cover reduction on abiotic and biotic soil properties, with focus on temperate alpine and forest ecosystems and with an outlook on future impacts.

Light grazing reduces the abundance of carbon cycling functional genes by decreasing oligotrophs microbes in desert steppe

Understanding the functional genes of microorganisms involved in carbon cycling processes can link microbial community structure to potential ecological functions. However, the effects of grazing on the relationship between carbon cycling functional genes and soil environment need to be further investigated as a global land use activity. A replicated sheep grazing platform was established in Northern China in 2004 with four different stocking rates: no grazing (0 sheep ha−1 month−1), light grazing (0.15 sheep ha−1 month−1), moderate grazing (0.30 sheep ha−1 month−1), and heavy grazing (0.45 sheep ha−1 month−1). To examine the effects of biotic and abiotic factors on microbial species and functional gene abundance, we collected soil samples after 17 years of grazing analyzed for the abundance of microbial communities, the abundance of carbon function genes, plant carbon input and soil environmental factors. We found that light grazing reduced carbon cycling functional genes compared to no grazing (fencing). With increasing grazing intensity, the absolute abundance of functional genes increased. Changes in microbial species abundance were mainly responsible for the changes in the structure of carbon cycling functional genes. Our results emphasize that elucidating how grazing affects the carbon cycling functional genes through the effects of plant carbon inputs and the environment, and the interactions between them, will provide important information for understanding carbon cycling processes in grassland ecosystems.

Effects of molasses-based microbial dust suppressant on soil dust and microbial community

This study aims at the practical needs of soil dust control and molasses resource utilization. By adding molasses to the soil and combining it with the components of microbial dust suppressants, it explores its impact on the surface soil dust suppression performance and microbial communities. The results show that after spraying treatment, the soil surface hardness is positively correlated with the molasses concentration, the surface hardness is the highest (41.14 HA) when the molasses concentration is 10%; the anti-wind erosion effect shows a trend of first increasing and then decreasing with the molasses concentration, the anti-rain erosion effect needs to be improved; microbial community analysis show that molasses significantly affected the relative abundance of soil microbial community, and appropriate addition (1%) could optimize the microbial community structure and enhance the functions of carbohydrate and energy metabolism pathways, and the abundance of pathogenic species increased when the molasses concentration was higher (above 5%). Therefore, considering the dust suppression effect and environmental safety, a dust suppressant with a molasses concentration of 1% is more suitable for dust suppression. However, the synergistic pathway and mechanism of dust suppression through the interaction between molasses and MICP technology need to be further explored.

A Comparative Analysis of Microbial Communities in the Rhizosphere Soil and Plant Roots of Healthy and Diseased Yuanyang Nanqi (Panax vietnamensis) with Root Rot

Microbial communities are not only an important indicator of soil status but also a determinant of plant nutrition and health levels. Loss of microbial community ecosystem control can directly lead to microbial disease occurrence. During the process of Yuanyang Nanqi wild imitation planting, root rot diseases frequently occur, seriously affecting their yield and quality. Via amplicon sequencing, this study mainly compared the microbial community composition between the rhizosphere soil and roots of healthy and diseased Yuanyang Nanqi with root rot. The α-diversity showed that the microbial community diversity and abundance in the roots of diseased Yuanyang Nanqi were much lower than those of those in healthy specimens, while no significant difference was found in the rhizosphere soil. The β-diversity showed that the bacterial community in the Gejiu region and the fungal community in the Honghe region were significantly different from those in other regions. The species relative abundance map showed that there was no obvious difference in microbial community composition between the rhizosphere soil and roots of healthy and diseased Nanqi, but in diseased specimens with root rot, the proportions of Pseudomonas and Fusarium increased. Based on a functional prediction analysis of FUNGuild, the results showed that the Nanqi roots were mainly pathological saprophytic type and that their rhizosphere soil was mainly saprophytic type. The microorganisms in the roots of Yuanyang Nanqi tubers with root rot were also isolated and identified through the use of the culture method. The possible pathogenic strains were tested via anti-inoculation, and Fusarium oxysporum was identified as one of the main pathogenic fungi of Nanqi root rot, which was consistent with the amplicon sequencing results. These results will help us understand the change trend of microbial communities in healthy and diseased plants and analyze the pathogens involved, the pathogenesis, and the beneficial microorganisms, which would provide a theoretical basis for effective biological control.

High stochasticity in rare bacterial community assembly in rice-wheat rotation soils at a regional scale

A growing body of evidence highlights the important role of both abundant and rare microorganisms in agroecosystem for their respective ecological functioning. However, the ecological processes that govern the assembly of abundant and rare taxa microbial communities in rice-wheat rotation soils at a regional scale have not yet been elucidated. Here, we conducted a comprehensive investigation of bacterial diversity, community structure, and assembly in samples from 238 sites under rice-wheat rotation in eastern China. Soil pH was the most important edaphic factor shaping bacterial diversity and community structure. The Shannon diversity of both abundant and rare bacterial taxa had a parabolic relationship with soil pH, with diversity reaching its peak at pH values of 7.50 and 7.31, respectively. Abundant bacteria displayed greater sensitivity to soil pH than rare taxa. Stochastic processes, especially dispersal limitation, predominantly governed assembly for both abundant and rare bacteria, explaining 80% of observed variation. Remarkably, stochastic processes contributed >90% of variation in the assembly of rare taxa communities, surpassing its importance in abundant taxa community assembly. Moreover, the assembly of specific bacterial phyla was predominantly governed by stochastic processes in the rice-wheat rotation soils. Overall, our findings provide the first data that although soil pH can make an initial assessment of the variation of bacterial communities, the high level of stochasticity in rare taxa community assembly would make them highly unpredictable during agricultural managements in rice-wheat rotation soils. These findings contribute new perspectives to understanding the main processes underpinning the observed bacterial communities, with implications for soil sampling, biodiversity conservation and ecosystem functionality in rice-wheat rotation soils.

Analysis of Microbial Diversity in Rhizosphere Soil of Panax notoginseng under Different Water and Microbial Fertilizer Conditions

Panax notoginseng is a highly regarded medicinal plant that has obstacles associated with continuous cropping. Understanding soil microorganisms is crucial, as they play a major role in this regard. However, soil microorganisms are affected by multiple factors; therefore, we need to conduct more in-depth research. This study investigated the combined effects of irrigation and microbial fertilizer treatments (J1F1, J1F2, J2F1, J2F2, J3F1, J3F2, and CK) on the diversity of bacterial and fungal microbial communities in the rhizosphere of Panax notoginseng. The bacterial 16S rRNA genes and fungal internal transcribed spacer (ITS) sequences were sequenced using Illumina HiSeq. The results showed that, without microbial fertilizer (CK), the microbial community abundance and diversity were significantly lower than in the other treatments; moreover, among the microbial fertilizer treatments, the microbial abundance in F1 was higher than that in F2. Under the same microbial fertilizer application, the incidence rate of Panax notoginseng root rot was J2 &gt; J1 &gt; J3, and the yield of Panax notoginseng was J3 &gt; J2 &gt; J1. Under the same irrigation conditions, the incidence rate of Panax notoginseng root rot was F1 &gt; F2, and the yield of Panax notoginseng was F2 &gt; F1. This study provides important guidance for Panax notoginseng plant microbiota and sustainable agriculture.

Effects of Microtopography on Soil Microbial Community Structure and Abundance in Permafrost Peatlands.

Soil microorganisms play crucial roles in the stability of the global carbon pool, particularly in permafrost peatlands that are highly sensitive to climate change. Microtopography is a unique characteristic of peatland ecosystems, but how microtopography affects the microbial community structures and their functions in the soil is only partially known. We characterized the bacterial and fungal community compositions by amplicon sequencing and their abundances via quantitative PCR at different soil depths in three microtopographical positions (hummocks, flats, and hollows) in permafrost peatland of the Greater Xing'an Mountains in China. The results showed that the soil of hummocks displayed a higher microbial diversity compared to hollows. Microtopography exerted a strong influence on bacterial community structure, while both microtopography and soil depth greatly impacted the fungal community structure with variable effects on fungal functional guilds. Soil water content, dissolved organic carbon, total phosphorus, and total nitrogen levels of the soil mostly affected the bacterial and fungal communities. Microtopography generated variations in the soil water content, which was the main driver of the spatial distribution of microbial abundances. This information stressed that the hummock-flat-hollow microtopography of permafrost peatlands creates heterogeneity in soil physicochemical properties and hydrological conditions, thereby influencing soil microbial communities at a microhabitat scale. Our results imply that changes to the water table induced by climate warming inducing permafrost degradation will impact the composition of soil microbes in peatlands and their related biogeochemical functions, eventually providing feedback loops into the global climate system.

Effects of Different Conditioners on Soil Microbial Community and Labile Organic Carbon Fractions under the Combined Application of Swine Manure and Straw in Black Soil

The return of straw and manure to agricultural fields can impact soil organic carbon (SOC) and biological properties. However, there is a lack of research on how using swine manure, maize straw, and various conditioners together affects soil bacterial and fungal populations. This study aimed to investigate six treatments, namely, only maize straw (S00), maize straw combined with swine manure (S0Z), maize straw combined with biochar and swine manure (SCZ), maize straw combined with boron slag and swine manure (SBZ), maize straw combined with biological agent and swine manure (SJZ), and maize straw combined with bio-organic fertilizer and swine manure (SFZ). The results showed that after the two-year return, all treatments increased the SOC content in 2023, which was 12.55–26.89% higher than S00, and the SCZ treatment significantly increased the soil organic carbon (SOC), dissolved organic carbon (DOC), easily oxidizable carbon (EOC), particulate organic carbon (POC), and microbial biomass carbon (MBC) content by 26.89%, 25.44%, 56.88%, 16.08%, and 43.54%, compared to S00. A redundancy analysis (RDA) showed that the continuous application of manure, maize straw, and conditioners has a positive impact on the diversity and abundance of soil microbial communities, enhancing the accumulation of soil carbon. Furthermore, our research revealed that soil fungi exhibited higher sensitivity in soil carbon composition following the addition of manure, straw, and conditioners to agricultural fields than bacteria. In conclusion, the addition of different conditioners to the fields is beneficial to biodiversity conservation from the perspective of achieving soil carbon storage and soil protection. Our findings suggested that the combination of maize straw, biochar, and swine manure has been proven to be the most effective treatment for increasing labile organic carbon fractions and enhancing the microbial community.

Added Biochars Promoted Nitrogen and Phosphorus Removal from Ecological Ditches at Low Temperature

The global issue of ecological ditches being poor in removing nitrogen and phosphorus under cold winter temperatures has been identified. This study introduced three types of biochar (reed, rice, and corn) into ecological ditch sediments via two application methods: rhizosphere and mixed addition. The purpose was to explore how these methods affect the removal of nitrogen and phosphorus, as well as their influences on microbial communities in sediments. The results indicated that the addition of biochar to ecological ditches significantly enhanced the removal of nitrogen and phosphorus. Among the three types of biochar, the mixed addition of corn biochar yielded the greatest results, achieving removal rates of 77.1% for total nitrogen (TN), 93.3% for NH4+-N, and 90.3% for total phosphorus (TP). The growth of Vallisneria natans was greatly improved by the mixed addition method, resulting in an average increase of 154%. This improvement was superior to the rhizosphere addition group, which led to a growth increase of 125%. In comparison, the control group (CK) showed a decrease of 4.8% in growth. Different methods of biochar addition resulted in changes in the physicochemical properties and stoichiometry of the plants. Microbial analyses showed that the addition of biochar reduced the diversity and abundance of the substrate microbial community.

Recalcification stabilizes cadmium but magnifies phosphorus limitation in wastewater-irrigated calcareous soil

Long-term wastewater irrigation leads to the loss of calcium carbonate (CaCO3) in the tillage layer of calcareous land, which irreversibly damages the soil's ability to retain cadmium (Cd). In this study, we selected calcareous agricultural soil irrigated with wastewater for over 50 years to examine the recalcification effects of sugar beet factory lime (SBFL) at doses of 0%, 2.5%, 5%, and 10%. We found that SBFL promoted Cd transformation in the soil from active exchangeable species to more stable carbonate-bonded and residual species, which the X-ray diffraction patterns also confirmed results that CdSO4 reduced while CdS and CaCdCO3 increased. Correspondingly, the soil bioavailable Cd concentration was significantly reduced by 65.6–84.7%. The Cd concentrations in maize roots and shoots were significantly reduced by 11.7–50.6% and 13.0−70.0%, respectively, thereby promoting maize growth. Nevertheless, SBFL also increased the proportion of plant-unavailable phosphorus (P) in Ca8–P and Ca10–P by 4.3–13.0% and 10.7−25.9%, respectively, reducing the plant-available P (Olsen P) content by 5.2–22.1%. Consequently, soil P-acquiring associated enzyme (alkaline phosphatase) activity and microbial (Proteobacteria, Bacteroidota, and Actinobacteria) community abundance significantly increased. Our findings showed that adding SBFL to wastewater-irrigated calcareous soil stabilized Cd, but exacerbated P limitation. Therefore, it is necessary to alleviate P limitations in the practice of recalcifying degraded calcareous land.

Zhibai dihuang pill (ZBDH) exhibits therapeutic effects on idiopathic central sexual precocity in rats by modulating the gut microflora

To reveal the role of gut microbiota (GM) in the occurrence and development of idiopathic central precocious puberty (ICPP) using 16S rDNA sequencing and bioinformatics analysis. The Danazol-induced ICPP model was successfully constructed in this study. ZBDH and GnRHa treatments could effectively inhibit ICPP in rats, as manifested by the delayed vaginal opening time, reduced weight, decreased uterine organ coefficient, and decreased uterine wall thickness and corpus luteum number, as well as remarkably reduced serum hormone (LH, FSH, and E2) levels. According to 16S rDNA sequencing analysis results, there was no significant difference in the GM community diversity across different groups; however, the composition of the microbial community and the abundance of the dominant microbial community were dramatically different among groups. ZBDH and GnRHa treatments could effectively reduce the abundance of Muribaculateae and Lactobacillus and promote Prevotella abundance. ZBDH and GnRHa were effective in treating Danazol-induced ICPP model rats. The therapeutic effects of ZBDH and GnRHa could be related to the changes in GM in rats.

Fermentation properties and functional stability of dough starter Jiaozi and Laomian after frozen storage.

This study aims to investigate the effects of frozen storage on the stability of traditional dough starters in China. The microbial community structure and abundance of related metabolic genes in different fermented sourdough prepared by Jiaozi (JZ) and Laomian (LM) starters before and after frozen storage at -20°C for half a year were analyzed using the shotgun metagenomic sequencing method, and differences in characteristics of texture in steamed bread were also compared by formal methods. The fermentation ability (FA) and metabolic activities of yeast in the JZH sourdough (started by JZ which was stored at -20°C for half a year) were better than those of LMH sourdough (started by LM which was stored at -20°C for half a year). The dominant genera of Acetobacter were found to be increased in the JZH0 sourdough (started by JZH and fermented for 0 h) and those of Lactobacillus were found to be decreased. Lactobacillus (98.72%), Pediococcus (0.37%), Saccharomyces (0.27%), and Acetobacter (0.01%), were dominant in sourdough LMH0 (started by LMH and fermented for 0 h). The abundances of "oxidative phosphorylation-related enzymes" and the "biosynthesis of glutamate"-related enzymes and genes related to "biosynthesis of glutamate" and "unsaturated fatty acid" were higher in JZH0 than in the JZ0 sourdough (started by JZ without being frozen and fermented for 0 h). The good FA of yeast, the acid production capacity of bacteria in the sourdough, and the quality of the JZH steamed bread (made by the JZH starter) indicated the better freezing tolerance of the microorganisms in JZ than in LM. The conclusion of this study suggests the better application potential of the JZ as the fermentation starter in actual production.
