Simultaneous separation and determination of seven biphenyl cyclooctene lignans in Schisandra chinensis and its preparations by micellar electrokinetic chromatography with dual organic solvent system.

Gomisin is a natural dibenzo cyclooctene lignan, which is mainly derived from the family Magnoliaceae. It has anti-inflammatory, antioxidant, anti-tumor, anti-aging, and hypoglycemic effects. Gomisins play important roles as medicines, nutraceuticals, food additives, and cosmetics. The objective of this study is to establish a micellar electrokinetic chromatography (MEKC) method for simultaneous separation and determination of seven biphenyl cyclooctene lignans (Gomisin D, E, G, H, J, N, and O) in Schisandra chinensis and its preparations. The method was optimized by studying the effects of the main parameters on the separation. The method has been validated and successfully applied to the determination of seven Gomisins in S.chinensis and its preparations. In the separation system, the running buffer was composed of 20 mM Na2HPO4, 8.0 mM sodium dodecyl sulfate (SDS), 11% (v/v) methanol, and 6.0% (v/v) ethanol. A diode array detector was used with a detection wavelength of 230 nm, a separation voltage of 17 kV, and an operating temperature of 25°C. Under this condition, the seven analytes were separated at baseline within 20 min, and a good linear relationship was obtained with correlation coefficient ranging from 0.9919 to 0.9992. The limit of detection (LOD, S/N = 3) and the limit of quantification (LOQ, S/N = 10) ranged from 0.8 to 0.9μg/mL and from 2.6 to 3.0μg/mL, respectively. The recovery rate was between 99.1% and 102.5%. The experimental results indicated that this method is suitable for the separation and determination of seven Schisandra biphenyl cyclooctene lignan compounds in real samples. At the same time, it provides an effective reference for the quality control of S.chinensis and its preparations.

A novel cation-selective exhaustive injection and sweeping micellar electrokinetic chromatography method for the simultaneous determination of second-generation triazoles in human plasma

In this study, we introduce the first capillary electrophoresis method that combines cation-selective exhaustive injection and sweeping micellar electrokinetic chromatography (CSEI-sweeping-MEKC) for the simultaneous and sensitive quantification of voriconazole (VRC), posaconazole (PSC), and isavuconazole (ISV) levels in human plasma. The sensitivity was remarkably improved, with a 5374-fold enhancement compared to the traditional MEKC method. We systematically optimized key parameters affecting sensitivity, resolution, and efficiency. The separation buffer included 100 mM NaH2PO4, 13 % acetonitrile, and 13 % tetrahydrofuran. This was followed by a high-conductivity buffer zone (1 psi, 50 s) of 100 mM NaH2PO4, an extended electrokinetic sample injection (10 kV, 600 s), and analyte sweeping with 100 mM sodium dodecyl sulfate micelles at −20 kV. Under these optimized conditions, the method allowed for an analytical run in 10 min and was validated for linearity (r ≥ 0.9997), precision (%RSD ≤ 8.8 %), and accuracy (%RE ≤ 2.20 %), delivering satisfactory results. The limit of detection was 2.55 ng/mL for ISV, 0.89 ng/mL for PSC, and 1.04 ng/mL for VRC. This method was successfully applied to analyze plasma samples from patients undergoing triazole antifungal treatment, demonstrating its feasibility for clinical testing.

Design of experiments for micellar electrokinetic chromatography method development for the monitoring of water-soluble vitamins in cell culture medium.

Biopharmaceutical production takes place in complex processes which should be thoroughly understood. Therefore, the iConsensus project focuses on developing a monitoring platform integrating several process analytical technology tools for integrated, automated monitoring of the biopharmaceutical process. Water-soluble vitamin monitoring using (microchip) capillary electrophoresis (CE) is part of this platform. This work comprises the development of conventional CE methods as the first part towards integrated vitamin monitoring. The vitamins were divided based on their physical-chemical properties to develop two robust methods. Previously, a method for the analysis of cationic vitamins (pyridoxine, pyridoxal, pyridoxamine, thiamine and nicotinamide) in cell culture medium was developed. This work focused on the development of a micellar electrokinetic chromatography method for anionic and neutral vitamins (riboflavin, d-calcium pantothenate, biotin, folic acid, cyanocobalamin and ascorbic acid). By employing multivariate design of experiments, the background electrolyte (BGE) could be optimised within one experiment testing only 11 BGEs. The optimised BGE conditions were 200mM borate with 77mM sodium dodecyl sulphate at a pH of 8.6. Using this BGE, all above-mentioned cationic, anionic and neutral vitamins could be separated in clean samples. In cell culture medium, most anionic and neutral vitamins could be separated. Combining the two methods allows for analysis of cationic, anionic and neutral vitamins in cell culture medium samples. The next step towards integrated vitamin monitoring includes transfer to microchip CE. Due to the lack of fast and reliable methods for vitamin monitoring, the developed capillary methods could be valuable as stand-alone at-line process analytical technology solutions as well.

Method Development for Determination of Doripenem in Human Plasma via Capillary Electrophoresis Coupled with Field-Enhanced Sample Stacking and Sweeping.

In this study, we established a novel capillary electrophoresis method for monitoring the concentration of doripenem in human plasma. As a time-dependent antibiotic, doripenem maximizes its antibacterial effects and minimizes the potential for antibiotic resistance through careful therapeutic drug monitoring. Two online preconcentration techniques, field-enhanced sample stacking (FESS) and sweeping, were coupled to enhance the detection sensitivity. Briefly, an uncoated fused silica capillary (40 cm × 50 μm i.d) was rinsed with a high conductivity buffer (HCB) composed of 150 mM phosphate buffer (NaH2PO4, pH 2.5) and 20% methanol. A large sample plug prepared in a low-conductivity phosphate buffer (50 mM NaH2PO4, pH 2.5) was then hydrodynamically injected (5 psi, 80 s) into the capillary. Under an applied voltage of -30 kV, the analyte was accumulated at the FESS boundary and swept by the negatively charged micelles toward the UV detector. Plasma samples were pretreated by solid-phase extraction (SPE) to eliminate endogenous interferences. The validation results demonstrated a high coefficient of determination (r2 > 0.9995) for the regression curve with impressive precision and accuracy: relative standard deviation (RSD) <5.86% and relative error <4.63%. The limit of detection (LOD, S/N = 3) for doripenem was determined to be 0.4 μg/mL. Compared to the conventional micellar electrokinetic chromatography method, our developed method achieved a sensitivity enhancement of up to 488-fold for doripenem. Furthermore, the newly developed method successfully quantified doripenem concentrations in plasma samples obtained from patients accepting doripenem regimens, proving its application potential in the clinical realm.

On-line preconcentration of fluorescein derivatives of essential amino acids using sweeping in a nonionic micellar system

A rapid and effective method of nonionic micellar electrokinetic chromatography with laser-induced fluorescence detection and sweeping as an online preconcentration method was developed for the simultaneous determination of free essential amino acids in supplement formulations. The analytes were derivatized using fluorescein isothiocyanate isomer I dissolved in dimethyl sulfoxide. Optimal separation conditions were achieved using an uncoated fused silica capillary (40.0 cm effective length, 50.2 cm total length, 50 µm internal diameter), 20 mM Na2B4O7, (pH 9.3), 50 mM Brij-35, and 15% (v v-1) methanol as running buffer. Complete separation of all analytes was achieved within 15 min. The optimized method exhibited good linearity (r2 > 0.997), acceptable precision (1.3 – 12.4%), and accuracy (88 – 112%) in concentration range 20 – 1000 ng mL−1. The limit of detection was 6 ng mL−1. The method successfully determined the quantity of free essential amino acids in a dietary supplement containing spirulina.

Green versatile micellar electrokinetic chromatographic method for determination of six antimicrobial and anti-inflammatory drugs in combined dosage forms

AbstractA fast reliable micellar electrokinetic methodology was investigated for the concurrent quantitation of six antimicrobial and anti-inflammatory drugs, namely, ciprofloxacin, dexamethasone, metronidazole, ornidazole, spiramycin and tinidazole. The method has the merits of rapidity, precision, and sensitivity. The separation was carried out in less than 7 min by applying a basic background electrolyte consisting of 25 mM disodium tetraborate buffer, pH 9 containing 50 mM SDS at 25 kV using photodiode array detector at 230 and 315 nm. The internal standard used during analysis was cromolyn sodium and validation was carried out following ICH guidelines. The proposed method showed linear response over the range from 0.5 to 10.0 μg mL−1 reaching limits of detection and limits of quantitation in the ranges of 0.09–0.2 μg mL−1 and 0. 27–0.6 respectively. The method's greenness was estimated using the GAPI tool where excellent greenness was concluded. Co-formulated or single-ingredient commercial preparations were investigated and the results were statistically evaluated.

Analytical Quality by Design-Compliant Development of a Cyclodextrin-Modified Micellar ElectroKinetic Chromatography Method for the Determination of Trimecaine and Its Impurities.

In 2022, the International Council for Harmonisation released draft guidelines Q2(R2) and Q14, intending to specify the development and validation activities that should be carried out during the lifespan of an analytical technique addressed to assess the quality of medicinal products. In the present study, these recommendations were implemented in Capillary Electrophoresis method development for the quality control of a drug product containing trimecaine, by applying Analytical Quality by Design. According to the Analytical Target Profile, the procedure should be able to simultaneously quantify trimecaine and its four impurities, with specified analytical performances. The selected operative mode was Micellar ElectroKinetic Chromatography employing sodium dodecyl sulfate micelles supplemented with dimethyl-β-cyclodextrin, in a phosphate-borate buffer. The Knowledge Space was investigated through a screening matrix encompassing the composition of the background electrolyte and the instrumental settings. The Critical Method Attributes were identified as analysis time, efficiency, and critical resolution values. Response Surface Methodology and Monte Carlo Simulations allowed the definition of the Method Operable Design Region: 21-26 mM phosphate-borate buffer pH 9.50-9.77; 65.0 mM sodium dodecyl sulfate; 0.25-1.29% v/v n-butanol; 21-26 mM dimethyl-β-cyclodextrin; temperature, 22 °C; voltage, 23-29 kV. The method was validated and applied to ampoules drug products.

Multicomponent antihypertensive pharmaceuticals determination with related impurity analysis using MEKC: Whiteness and greenness assessment

In the present study, a white and green micellar electrokinetic chromatography method (MEKC) coupled with a photo diode array detector (DAD) has been established for challenging multi-component analysis of two antihypertensive combinations therapy. The first one is the supramolecular complex LCZ696 of valsartan (VAL) and sacubitril (SAC) with sacubitril related impurity (BMP) and the second combination is nebivolol (NEB) with thiazide like diuretics: hydrochlorothiazide (HCZ) and indapamide (IND). Separation was done using fused silica capillary (40 cm × 50 μm id) along with 10 mM phosphate buffer adjusted to pH 10 in presence of 25 mM sodium dodecyl sulphate (SDS). Linearity was assessed in the concentration ranges of 10–300, 10–300, and 5–50 μg. mL−1 for VAL, SAC, and BMP (Mix I) respectively, while for Mix II 20–200 μg. mL−1 for HCZ, IND and NEB with r > 0.9991. LOD were 3.12,3.11 and1.53 μg. mL−1for VAL, SAC, and BMP respectively. In addition, LOD for Mix II were 5.92,4.44 and 5.30 μg. mL−1for HCZ, IND and NEB respectively. The applicability of the suggested method was proved by investigating the five antihypertensive drugs at various concentrations levels in different pharmaceutical preparations with sacubitril related impurity analysis,BMP,at minute level in presence of parent drug, SAC, reaching 1.6%. Moreover, the proposed method did not rely only on the greenness evaluation tool using the most widely used Analytical Eco-Scale and the novel AGREE tool but also was merged with whiteness appraisal using RGB 12 algorithm.

Simultaneous determination of triclosan，triclocarban and p-chloro-m-xylenol in disinfectant，personal care products and oiltment by nonaqueous capillary electrophoresis

Triclosan （TCS）， triclocarban （TCC）， and p-chloro-m-xylenol （PCMX） are some of the most widely used antibiotics because of their broad-spectrum and highly-efficient bactericidal effects. In the context of disinfection， the National Standard GB 38598-2020 stipulates that the contents of the effective ingredients present in a disinfectant must be specified， wherein their range must fall within 90%-110% of the specified central value. To ensure a suitable product quality， analysis by high performance liquid chromatography （HPLC） is recommended by both the GB/T 27947-2020 and GB/T 34856-2017. However， the results analyzed according to the National Standard method often exceed the declared contents， thereby indicating the necessity to establish a new method based on a completely different principle （e. g.， capillary electrophoresis）， especially since it was not possible to analyze TCS， TCC， and PCMX in a single injection using the National Standard method. Moreover， using this method， large amounts of methanol were consumed， which could be potentially harmful to both operators and the environment. In terms of their water solubilities， this decreases in the order of PCMX>TCS>TCC， wherein TCC is insoluble in water. As such， the use of nonaqueous capillary electrophoresis （NACE） based on running buffer solutions prepared in pure organic solvents （e. g.， methanol or acetonitrile） is necessary. In this paper， a new NACE approach combined with an ultraviolet detection method was developed for the simultaneous analysis of TCS， TCC， and PCMX in disinfectants， personal care products， and ointments. For this purpose， an uncoated fused silica capillary （20 cm×50 μm， total length=30.2 cm） was used as the separation column with a separation buffer composed of 14 mmol/L sodium borate， 2 g/L polyethylene glycol （PEG） 20000， and 0.5 mmol/L dodecyltrimethylammonium bromide （DTAB） in methanol. Following optimization of the separation parameters， the complete and simultaneous separation of TCS， TCC， and PCMX was achieved when the sample solution was prepared using 5 g/L PEG 20000 in methanol-acetonitrile （50∶50， v/v）. It was possible to directly inject the sample into the analysis system after a simple dilution with the sample medium， and no interference was observed in any of the sample electropherograms when a separation voltage and detection wavelength of -12 kV and 214 nm were employed， respectively. Furthermore， TCS， TCC， and PCMX showed good linear relationships with their corrected peak areas within a mass concentration range of 1-100 mg/L， and the correlation coefficients （r） were all greater than 0.99. Moreover， the limits of detection （LODs， S/N=3） and limits of quantification （LOQs， S/N=10） were determined to be 0.2 and 1 mg/L， respectively. The spiked recoveries ranged from 94.5% to 104.4% with relative standard deviations of ≤4.8% in all cases. Subsequently， the established method was used to analyze 31 commercial samples， including hand sanitizer， disinfectant， baby powder， and antibacterial cream. A comparative analysis of HPLC， the developed NACE method， and our previously reported micellar electrokinetic chromatographic （MEKC） method was also carried out for the quantitative determination of TCS， TCC， and PCMX. Although no statistically significant differences were observed among the three methods， the results determined for 16 out of the 31 samples did not match the claimed contents. These results therefore indicate the necessity to further control the compositions of disinfectant products. Our results indicate that the newly established NACE method can be an important alternative to HPLC for routine laboratory analyses， especially considering that it minimizes waste generation， requires only a simple sample pretreatment process， and exhibits a good selectivity to the target compounds. It is therefore hoped that the NACE method will be incorporated into the National Standard method in the near future.

Tailoring two white chromatographic platforms for simultaneous estimation of ritonavir-boosted nirmatrelvir in their novel pills: degradation, validation, and environmental impact studies.

As the COVID-19 pandemic is not yet over, Pfizer has launched the novel pill Paxlovid® (Nirmatrelvir (NMV) co-packaged with ritonavir (RIT)) as an effective medication for hospitalized and non-hospitalized patients. Making pharmaceutical analysis greener and more sustainable has lately become the main direction of the research community. In this context, two fast, green, and stability-indicating chromatographic methods were designed for the neat quantitative determination of NMV and RIT in their bulk and dosage forms. Method I is deemed the first electro-driven attempt for the assay of Paxlovid®. Herein, the optimized conditions of the Micellar Electrokinetic Chromatographic (MEKC) method were 50 mM borate buffer at pH 9.2 with 25 mM sodium lauryl sulfate (SDS) being used as the background electrolyte (BGE) on a deactivated fused silica capillary (50 cm effective length × 50 μm id). Method II was an isocratic reversed-phase HPLC separation method using Zorbax-Eclipse C18 (4.6 × 250 mm, 5 μm particle size) column and 50 mM ammonium acetate buffer at pH 5 and acetonitrile as mobile phase constituents at a flow rate of 1 mL min-1. For the sake of simplicity and increasing sensitivity, a single wavelength of 210 nm was used for the two methods to assay both drugs. Linear correlations between peak areas and concentration were observed in the ranges of 10-200 μg mL-1 for NMV and 5-100 μg mL-1 RIT in both methods. The impact of versatile stress conditions such as hydrolysis, oxidation, and photolysis on the stability of NMV and RIT was studied. Fortunately, both methodologies were able to separate both drugs from their degradants. Thus, the stability indicating power of the methods was proved. The derived methods were statistically validated in agreement with the ICH guidelines. Furthermore, the environmental friendliness and sustainability of these methods were investigated and compared with the cited methods using the holistic multicriteria evaluation tools namely Hexagon, AGREE, and RGB12 metrics. Conclusively, the proposed methods offered reliable, feasible, economic, white, and stability-indicating alternatives to the cited chromatographic methods.

Screening of prolyl hydroxylase 2 inhibitors based on quantitative strategy of peptides

Prolyl hydroxylase 2 (PHD2) is a key oxygen receptor regulating oxygen homeostasis in human body, and it is one of the important targets for drug research and development of hypoxia related diseases. In PHD2 enzymatic reaction, the structure of substrate (HIF-1α556–574) and product (hydroxylated HIF-1α) peptide only differ from one oxygen atom (MW>2000), which makes it a great challenge to separate them accurately and efficiently. In this work, the direct separation and detection of HIF-1α and hydroxylated HIF-1α has been firstly reported based on micellar electrokinetic chromatography (MEKC). Under optimized conditions, the intraday RSD of peak area and apparent electrophoretic mobility of hydroxylated HIF-1α were 1.87% and 0.81% respectively, and the interday RSD were 2.01% and 1.03% respectively. The LOD and LOQ of the MEKC method were 10 µM and 50 µM respectively, and the recoveries was 98.42–105.38%. Subsequently, the feasibility and accuracy of MEKC method to screen PHD2 inhibitors were confirmed by using roxadustat, and the IC50 (10.36 µM) and inhibitor type (competitive) were consistent with literature. Finally, the method was used to screen the PHD2 inhibitory activity of five traditional Chinese medicines (TCMs). The present work not only overcomes the difficulties of direct quantitative detection of hydroxylated HIF-1α, but also provides technical support for exploring and discovering new drug leads for hypoxia-related diseases from complex matrix such as TCMs.

Decision-making tree for improving selectivity and sensitivity of sweeping-MEKC determination of hydrophobic analytes: Application to five cardiovascular drugs with greenness assessment

Efficient separation of five cardiovascular drugs has been achieved by a new micellar electrokinetic chromatographic method based on using a general strategy that can be applied for the separation of different hydrophobic analytes, while keeping the highest possible sweeping efficiency and hence the highest possible sensitivity. The studied drugs include valsartan, atorvastatin, ezetimibe, amlodipine, and simvastatin. The separation is accomplished in less than 12 min by applying a basic background electrolyte composed of 10 mM disodium tetraborate buffer of pH 10.5 containing 25 mMSDS and 11% n-propanol at 30 kv. Cromolynsodium is used as an internal standard. Variables that may influence the electrophoretic mobility of the five compounds are investigated. The calibration plots are linear within ranges of 0.5–20.0 μg/mL, 0.5–10.0 μg/mL and 10.0–100.0 μg/mL for valsartan, ezetimibe and simvastatin (at 200 nm) and 1.0–30.0 μg/mL for amlodipine, atorvastatin and simvastatin (at 240 nm). Correlation coefficients are ≥ 0.9997 and detection limits are ranged between 0.12 and 2.05 μg/mL. Thedeveloped procedure is successfully utilized to estimate the studied compounds in different pharmaceutical preparations. The obtained data are statistically analyzed and evaluated. The greenness profile of the investigated approach has been assessed by a recent assessment tool to ensure the principles of green chemistry.

Determination of four naphthalenediols in cosmetic samples by sweeping-micellar electrokinetic chromatography and a comparison with HPLC

Abstract A sweeping micellar electrokinetic chromatography (sweeping-MEKC) method was developed for the determination of 1,7-naphthalenediol, 2,3-naphthalenediol, 1,5-naphthalenediol and 2,7-naphthalenediol in cosmetics. Several parameters affecting sweeping-MEKC method were studied systematically and the separation conditions were optimized as 20 mM NaH2PO4–110 mM SDS and 40% (v/v) MeOH (pH 2.4), with −22 kV applied voltage and UV detection at 230 nm. The sample matrix is 60 mmol L−1 NaH2PO4 and sample introduction was performed at 3 psi for 6 s. Separation of the four naphthalenediols was completed in less than 17 min. Limit of detection (LOD) and limit of quantitation (LOQ) are 0.0045∼0.0094 μg mL−1 and 0.015∼0.031 μg mL−1. Linear relationship (r 2 &gt; 0.999) is satisfactory at the range of 0.1–10 μg mL−1. The developed method has been successfully applied to the determination of the four naphthalenediols in real cosmetic samples, with recoveries in foundation, sun cream and lotion in the range of 92.3%∼106.8% and relative standard deviation (RSD) less than 4.15%. A HPLC method described in the National Standards of the People’s Republic of China was carried out for the comparison with the proposed method. The results showed that the proposed sweeping-MEKC method has the advantages of fast, low cost with comparative sensitivity.

Micellar electrokinetic chromatography method for the analysis of synthetic and phytocannabinoids

Synthetic cannabinoids (SCs) are the largest group of illicit compounds currently monitored in Europe. Reliable analytical methods for the differentiation and quantification of SCs and phytocannabinoids (PCs) from plant-based products are needed to reduce possible public health risks. The objective of this research was to develop a new method for the detection of four SCs (JWH-018, JWH-073, JWH-200, JWH-250) and two PCs (THC, CBD) from plant materials by using micellar electrokinetic chromatography with UV-absorbance detection. A novel method with a water-based background electrolyte containing cholate micelles was developed and optimized using two sets of the Box-Behnken experimental design. The method enables the quantification of JWH-018, JWH-073, JWH-200, JWH-250, and detection of THC and CBD. The selectivity studies revealed that several plants that are prevalently used as carrier matrixes for SCs could be analyzed without an added sample purification procedure. A basic method validation was performed. The detection and quantification limits were 3.0 and 5.0 for SCs, and 3.7 and 6.2 mg/L for PCs. For all analytes, the method intra-day precision was under 8%, the inter-day precision under 13%, and recoveries up to 100%.

A novel reverse migration micellar electrokinetic chromatography method for in-capillary screening and quantifying of antioxidant components in Sanyetangzhiqing using 2,2'-Azinobis-(3-ethylbenzthiazoline-6-sulphonate) as oxidation-free radical.

A novel method of reverse migration micellar electrokinetic chromatography (RM-MEKC) using 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate) as oxidation-free radical was developed to screen the major antioxidants from Sanyetangzhiqing (SYTZQ), which is a new patent drug for diabetes. For simultaneous detection and separation of 2,2'-bis (3-ethylbenzothiazoline-6-sulfonic acid) ammonium salt (ABTS+ ) and chemical components of SYTZQ, the key factors were optimized and the method was validated under the optimal conditions. All the relative standard deviations of recovery, precision, and stability were below 6.6%. Based on these, the RM-MEKC method has been successfully used to screen the main antioxidants from SYTZQ tablets. Five compounds, including rosmarinic acid, salvianolic acid B, lithospermic acid, hyperoside, and isoquercetin, were separated and identified as the major antioxidants. There was a linear relationship with correlation coefficient of 0.923 between the total amount of major antioxidants and total antioxidative activity of SYTZQ. Consequently, these five ingredients could be selected as combinatorial markers for quality control of SYTZQ, and the established method was concluded to be a simple, reliable, and powerful tool to screen and quantify active compounds for quality evaluation of SYTZQ.

Determination of the Monoclonal Antibody Tocilizumab by a Validated Micellar Electrokinetic Chromatography Method

Tocilizumab is a monoclonal antibody used in the treatment of several inflammatory and autoimmune diseases as well as cancers. Tocilizumab improves clinical outcomes and reduce mortality rates in patients with COVID-19 disease. A novel, simple and reliable method was developed to determine tocilizumab using micellar electrokinetic chromatography (MEKC). Separation of tocilizumab and the internal standard, methotrexate, was achieved with a background electrolyte consisting of phosphoric acid buffer and sodium dodecyl sulfate (SDS) with UV detection at 195 nm. The method was linear in the concentration range from 10 to 250 µg/mL with correlation coefficient greater than 0.995. The method was successfully applied to the analysis of human and rat plasma samples with good recoveries. Sample preparation involved protein precipitation followed by dilution of the supernatant. The intra‐ and inter-day precisions were less than 5%, the accuracy varied from − 2.71 to 3.84%. The proposed method has acceptable analytical performance and could be applied in future clinical and pharmacokinetic studies including anticancer therapy.

Comparative Greenness Metric Estimates for Content Uniformity Testing of Anti-Cov-2, GS-5734 in Commercial Vials: Validated Micellar Electrokinetic Chromatographic Assay.

BackgroundThe antiviral drug GS-5734 remdesivir is a new phosphoramidate prodrug developed initially as a treatment for Ebola virus which then proved to have antiviral properties against other viruses. After clinical trials, it was the first antiviral to be approved by the U.S. Food and Drug Administration in 2020 to treat severe coronavirus (COVID-19) cases. The widespread current pandemic gave an urge to its fast production and marketing. Thus, new analytical methods must be available for its analysis in a fast and easy manner with low cost to be applicable in all laboratories.ObjectiveIn the current study, a green and economic micellar electrokinetic chromatographic (MEKC) method is proposed for remdesivir analysis.MethodsA fused-silica capillary (58.5 cm × 50 μm id, 50 cm effective length) with 20 mM borate buffer (pH 9) and 25 mM sodium dodecyl sulfate was used under a positive potential of 30 kV at 25°C with detection at 245 nm.ResultsRemdesivir analysis was achieved in approximately 5 min. The method proved to be linear in range of 1–50 μg/mL with correlation coefficient, r > 0.999.ConclusionThe MEKC method proposed was applied to the analysis of remdesivir in its commercial vials. The method was validated per International Conference on Harmonization guidelines.HighlightsGreen chemistry has been the focus of the analytical community in the past few years. This method is considered green due to its low energy and solvent consumption without sacrificing the method’s sensitivity or selectivity. The method’s green profile has been assessed by different greenness assessment scales to ensure the method is eco-friendly and can be used in the pharmaceutical industry.

Simultaneous determination of ten nucleosides and bases in Ganoderma by micellar electrokinetic chromatography

Ganoderma (lingzhi) is a famous herbal medicine and edible supplement in oriental countries for a long history. In this study, a simple micellar electrokinetic chromatography (MEKC) method was established for the analysis of nucleosides and bases, the major bioactive components in Ganoderma for the first time. By optimizing the borate concentration, the sodium dodecyl sulfate (SDS) concentration and the pH value of running buffer, 10 nucleosides and bases achieved an ideal separation. In real sample analysis, the developed method was successfully used to determine the 10 target analytes in 23 batches of Ganoderma samples from different regions. Results indicated that contents of 10 investigated analytes in each sample showed obvious variation. The principal components analysis (PCA) and hierarchical cluster analysis (HCA) analysis classified the samples into three groups, and the HCA tree visualized the relationships which was mainly contributed by geographical partition. The results indicated geographical origin to be an important factor that affect the accumulation of nucleosides and bases in Ganoderma. In summary, this study provides a simple and practical strategy for quality assessment and cultivation reference of Ganoderma.

Determination of five saponins in Xuesaitong Dropping Pills by micellar electrokinetic chromatography and evaluation method of between-batch consistency

The present study determined five saponins in Xuesaitong Dropping Pills(XDP) by micellar electrokinetic chromatography(MEKC), and evaluated between-batch consistency by MEKC fingerprints and similarity analysis. A background buffer was composed of 20 mmol·L~(-1) sodium tetraborate-20 mmol·L~(-1) boric acid solution(pH 8.5), 55 mmol·L~(-1) sodium dodecyl sulfate(SDS), 23 mmol·L~(-1) β-cyclodextrin, and 13% isopropyl alcohol. All separations were performed at 25 ℃,20 kV and the detection wavelength was set at 203 nm. The separation channel was a fused silica capillary with a dimension of 75 μm I.D. and a total length of 50.2 cm(effective length of 40.0 cm). The contents of notoginsenoside R_1, and ginsenosides Rg_1, Re, Rb_1, Rd were determined with their quality control ranges set. The fingerprints of XDP were established and the between-batch consistency was evaluated by similarity analysis. The contents of five saponins from the 19 batches of XDP were stable in the fixed ranges. Statistical analysis was carried out on the results of multiple batches of samples, and the specific quality control ranges were recommended as follows: notoginsenoside R_1 21.92-34.16 mg·g~(-1), ginsenosides Rg_1 83.54-131.78 mg·g~(-1), ginsenosides Re 13.58-19.82 mg·g~(-1), ginsenosides Rb_1 89.40-129.90 mg·g~(-1), and ginsenosides Rd 22.34-35.67 mg·g~(-1). Eleven characteristic peaks were identified in the fingerprints. Five peaks, notoginsenoside R_1 and ginsenosides Rg_1, Re, Rb_1, Rd, were identified with reference standards. The similarities of the 19 batches of samples were all above 0.988, indicating good between-batch consistency. This method is green and simple, and can be used for the quantitative determination and quality evaluation of XDP. It can also provide references for the quality control of other Chinese medicinal dripping pills.

The critical evaluation of the effects of imidazolium-based ionic liquids on the separation efficiency of selected biogenic amines and their metabolites during MEKC analysis

Ionic liquids (ILs) such as imidazole can be used to prevent the sorption of analytes onto the quartz walls of the capillary. Coating the capillary wall with a cation layer increases its surface stability, consequently improving the repeatability of separation process. Currently, examining the effects of dynamic coatings on the capillary wall is an emerging trend in capillary electrophoresis (CE) research. This study uses micellar electrokinetic chromatography (MEKC) to evaluate how ILs in the background electrolyte (BGE) affect the separation efficiency of biogenic amines (BAs). Specifically, this research focuses on 12 ILs built from cations containing an imidazole ring with different alkyl substituents and anions, as well as one IL containing a pyridinium cation with tetrafluoroborate anion. All analyzed ILs, which were added to the BGE in concentrations ranging from 1 to 20 mM, were tested for their ability to improve the electrophoretic separation of selected BAs, namely: homovanillic acid (HVA), vanililmandelic acid (VMA), dihydroxyphenylglicol (DHPG), 3-metoxy-4-hydroxyphenyl glicol (MHPG), normetanephrine (NM), metanephrine (M), and dihydroxyphenylacetic acid (DOPAC). The results showed that the most effective ILs added to the BGE were those with a chloride anion (1-hexyl-3-methylimidazolium chloride [HMIM+Cl−] and 1-ethyl-3-methylimidazolium chloride [EMIM+Cl−]) and those with a tetrafluoroborate anion (1-hexyl-3-methylimidazolium tetrafluoroborate [HMIM + BF4−]). Improved separation efficiency was also obtained for the BGE containing 1-hexyl-3-methylimidazolium hexafluorophosphate [HMIM + PF6−]. On the other hand, ILs with trifluoromethanesulfonate [OTf−] or bis(trifluoromethylsulfonyl)imide [NTf2-] anions, even at low concentrations in the BGE, disturbed the flow of current through the capillary and worsened the separation process. Overall, this study provides a critical evaluation of the impact of different types and concentrations of ILs on the performance of the MEKC method during the analysis of selected BAs.
