Comparison of Enzyme Activity in Order to Describe the Metabolic Profile of Dairy Cows during Early Lactation.

Enzymatic diagnostics have practical applications in diseases of the liver, heart, pancreas, muscles, blood, and neoplastic diseases. This study aimed to compare enzyme activity to describe dairy cows’ metabolism during early lactation. Based on their general health symptoms, the cows were assigned to one of three groups: acidotic, healthy and ketotic. Samples of milk, blood and rumen fluid were collected at 12 ± 5 days postpartum. Ketotic cows were characterized by the highest malondialdehyde (MDA, 76.098 nM/mL), glutathione reductase (GluRed, 109.852 U/L), superoxide dismutase (SOD, 294.22 U/L) and gamma-glutamyltranspeptidase (GGTP, 71.175 U/L) activity. In comparing ketotic and acidotic cows, MDA, GluRed, SOD and GGTP activity were higher by a factor of almost: 1.85, 1.89, 0.79 and 2.50, respectively. Acidotic cows were characterized by the highest aspartate aminotransferase activity (AspAT, 125.914 U/L). In comparing acidotic and ketotic cows, AspAT activity was higher by a factor of almost 1.90. The use of enzymatic markers could limit the frequency of sampling for laboratory analyses and may result in a faster diagnosis of metabolic disorders. AspAT activity in blood serum seems to be a good indicator of acidosis; GGTP may participate in the pathogenesis of ketosis.

Alterations in the Milk Metabolome of Dairy Cows Supplemented with Different Levels of Calcium Propionate in Early Lactation

This study aimed to investigate the effects of dietary supplementation with different levels of calcium propionate on the lactation performance, blood energy metabolite parameters, and milk metabolites of dairy cows in early lactation. Thirty-two multiparous Holstein cows were randomly divided into 4 groups, which were orally drenched with 0, 200, 350, and 500 g/d calcium propionate per cow supplemented to a basal diet for 5 weeks from calving. The milk and blood of the dairy cows were sampled and measured every week. The milk samples from the last week were used for the metabolomic analysis via liquid chromatography–mass spectrometry (LC-MS). The results showed that the calcium propionate supplementation quadratically increased the dry matter intake, energy-corrected milk yield, and 4% fat-corrected milk yield; linearly reduced the milk protein and milk lactose concentrations; and quadratically decreased the somatic cell count in the milk. With the increase in calcium propionate, the serum glucose content showed a linear increase, while the serum insulin content showed a quadratic increase. The diets supplemented with calcium propionate quadratically decreased the β-hydroxybutyric acid and linearly decreased the non-esterified fatty acid content in the serum. The metabolomic analysis revealed that eighteen different metabolites were identified in the milk samples of the dairy cows supplemented with calcium propionate at 350 g/d, which decreased the abundance of genistein and uridine 5-monophosphate and increased the abundance of adenosine, uracil, protoporphyrin IX, and sphingomyelin (d 18:1/18:0) compared with the control group. The milk metabolic analysis indicated that the calcium propionate effectively improved the milk synthesis and alleviated the mobilization of adipose tissue and bone calcium. In summary, the calcium propionate could improve the lactation performance and energy status and promote the milk metabolic profile of dairy cows in early lactation. Calcium propionate (350 g/d) is a well-recommended supplement for dairy cows for alleviating negative energy balance and hypocalcemia in early lactation.

Early metabolic profiling in the periparturient period reduces the occurrence of postparturient metabolic diseases in cows.

Objective:To study the impact of early metabolic profiling and intervention measures in the periparturient period on the occurrence of postparturient metabolic diseases in cows.Materials and Methods:Using a cohort of dairy cows from two selected areas of Bangladesh, we routinely tested the serum calcium level and ketone bodies in the urine at periparturient periods. In addition, milk samples were tested for the presence of mastitis at different stages of lactation. Animals showing reduced serum calcium levels, high ketone bodies in the urine or the presence of clinical and subclinical mastitis received appropriate therapeutic intervention immediately after detection. After the intervention, the number of animals that got sick with diseases or conditions like milk fever, ketosis, mastitis, and dystocia, which are caused by metabolic problems, was recorded.Results:In the periparturient period, most of the animals had lower serum calcium levels (8.13 ± 1.2 mg/dl), which were significantly increased by the following intervention (10.05 ± 1.4 mg/dl). On the other hand, there was a higher number of ketosis-affected animals (33.9%, 20/59) during the periparturient period, which decreased (18.6%, 11/59) during the postparturient period. Similarly, the number of mastitis-affected cows was also decreased in postparturient cows (30.9%, 13/42) than in periparturient cows (59.5%, 25/42), following improved hygienic measures. After early intervention, the number of study animals with metabolic diseases decreased from 51.7% before intervention to 15.3% after intervention.Conclusions:Early metabolic profiling significantly reduced the occurrence of metabolic diseases in cows. Therefore, we recommend regular metabolic profiling of dairy cows and receiving early intervention measures to reduce the occurrence of metabolic diseases on the farm.

GC/MS and LC/MS Based Serum Metabolomic Analysis of Dairy Cows With Ovarian Inactivity.

Metabolic disorders may lead to the inactive ovaries of dairy cows during early lactation. However, the detailed metabolic profile of dairy cows with inactive ovaries around 55 days postpartum has not been clearly elucidated. The objective of this study was to investigate the metabolic difference in cows with inactive ovaries and estrus from the perspective of serum metabolites. According to clinical manifestations, B-ultrasound scan, rectal examination, 15 cows were assigned to the estrus group (E; follicular diameter 15–20 mm) and 15 to the inactive ovary group (IO; follicular diameter <8 mm and increased <2 mm within 5 days over two examinations). The blood was collected from the tail vein of the cow to separate serum 55–60 days postpartum, and then milked and fasted in the morning. Serum samples were analyzed using gas chromatography time-of-flight mass spectrometry technology (GC-TOF-MS) and ultra-high-pressure liquid chromatography-quadrupole-time-of-flight mass spectrometry (UHPLC-QTOF-MS). Differences in serum metabolites were identified using multivariate statistical analysis and univariate analysis. Thirty differentially abundant metabolites were identified between the two groups. In cows with inactive ovaries compared with cows in estrus, 20 serum metabolites were significantly higher (beta-cryptoxanthin (p = 0.0012), 9-cis-retinal (p = 0.0030), oxamic acid (p = 0.0321), etc.) while 10 metabolites were significantly lower (monostearin (p = 0.0001), 3-hydroxypropionic acid (p = 0.0005), D-talose (p = 0.0018), etc.). Pathway analysis indicated that the serum differential metabolites of multiparous cows in estrus obtained by the two metabolomics techniques were mainly involved in β-alanine metabolism and steroid biosynthesis metabolism, while other involved metabolic pathways were related to metabolism of glyoxylate; dicarboxylate metabolism; fructose, mannose, glutathione, glycerolipid, glycine, serine, threonine, propanoate, retinol, and pyrimidine metabolism. This indicates that the abnormalities in glucose metabolism, lipid metabolism, amino acid metabolism, and glutathione metabolism of postpartum dairy cows obstructed follicular development.

Effects of dietary N-carbamylglutamate supplementation on milk production performance, nutrient digestibility and blood metabolomics of lactating Holstein cows under heat stress

Heat stress is a serious threat to dairy cow health worldwide. N-carbamylglutamate (NCG), a novel feed additive, has been applied in dairy cow industry. However, the effects and mechanisms of NCG on dairy cows under heat stress are still unknown. A total of 48 Holstein dairy cows with similar days in milk (154 ± 13.6 d), parity (1-3), and body condition score were randomly divided into 4 groups of 12 animals each. Under heat stress, the cows were fed a TMR diet supplemented with 0 (control), 15, 20, 25 g of NCG/d per cow for 60 days. Milk production performance was recorded and serum parameters were examined. Meanwhile, metabolomics study of plasma based on liquid chromatography-mass spectrometry was also applied to investigate metabolites and pathways in response to NCG supplementation. Milk yield was increased but milk urea nitrogen was reduced by the NCG treatment. Addition of 15 g of NCG/d increased the milk protein percentage compared with the control. No effect of NCG was observed on dry matter intake, lactose percentage, milk fat percentage and somatic cell count. Serum glucose concentrations in cows fed with 15, 20, 25 g of NCG/d were 5.34 ± 0.312, 5.78 ± 0.473 and 5.54 ± 0.711 mmol/L, which were greater than those in the control (4.87 ± 0.367 mmol/L, P < 0.05). Meanwhile, the concentrations of blood urea nitrogen and blood ammonia were decreased with the addition of NCG. Results for antioxidant capacity and immune function showed beneficial effects of NCG, such as the increases in total antioxidant capacity, superoxide dismutase, glutathione peroxidase, IgG, IgM and IgA. Clear separations of plasma metabolic profiles between control and NCG groups were observed in the score plots. Sixteen different metabolites involved in metabolisms of amino acids, ketone bodies, butanoate and energy, as well as gut microbiome-derived metabolism were regulated by NCG supplementation. In conclusion, supplementation with NCG has the potential to enhance production performance, antioxidant capacity and immune function, and improved the metabolic profile of dairy cows under heat stress.

The biochemical and metabolic profiles of dairy cows with mycotoxins-contaminated diets.

BackgroundPrevious studies on the effects of mycotoxins have solely focused on their biochemical profiles or products in dairy ruminants. Changes in metabolism that occur after exposure to mycotoxins, as well as biochemical changes, have not been explored.MethodsWe measured the biochemical and metabolic changes in dairy cows after exposure to mycotoxins using biochemical analyses and nuclear magnetic resonance. Twenty-four dairy cows were randomly assigned to three different treatment groups. Control cows received diets with 2 kg uncontaminated cottonseed. Cows in the 50% replacement group received the same diet as the control group, but with 1 kg of uncontaminated cottonseed and 1 kg of cottonseed contaminated with mycotoxins. Cows in the 100% replacement group received the same diet as the control, but with 2 kg contaminated cottonseed.ResultsThe results showed that serum γ-glutamyl transpeptidase and total antioxidant capacities were significantly affected by cottonseed contaminated with mycotoxins. There were also significant differences in isovalerate and NH3-N levels, and significant differences in the eight plasma metabolites among the three groups. These metabolites are mainly involved in amino acid metabolism pathways. Therefore, the results suggest that amino acid metabolism pathways may be affected by mycotoxins exposure.

Relation between Liver Lipid Content and Plasma Biochemical Indicators in Dairy Cows

Background: The incidence of metabolic disorders in dairy cows has increased as a result of intensive production practices and genetic selection for milk yield. Among such disorders, liver lipidosis is very frequently observed and has significant negative effects on cow health and milk yield. Severe energy supply deficit causes high fat mobilization, resulting in rapid body condition loss and liver lipid infiltration, which cannot always be detected by known blood biochemical indicators. The main objective of the study was to determine possible associations of blood biochemical parameters with liver lipid infiltration severity in high-yield multiparous Holstein cows along prepartum and postpartum periods.Materials, Methods &amp; Results: Fifty-four high-yield Holstein cows from a herd managed in a free-stall system in the northwest region of the state of Rio Grande do Sul, Brazil were evaluated. Cows were divided in two groups according to their physiological status. Group 1 included 19 prepartum cows, with an average milk yield of 44.7 L/d in the previous lactation, and 2.8 average parity, and Group 2 included 35 postpartum cows, with an average milk yield of 46.8 L/d in the previous lactation, and 2.5 average parity. The farm produced 10,000 kg milk per year, containing 3.5% of fat, 3.2% of protein and 4.7% of lactose. Total cholesterol (CH), plasma triglycerides (TGp), beta-hydroxybutyrate (BHB), albumin (ALB), non-esterified free fatty acids (NEFA) levels, and aspartate aminotransferase (AST) and creatine kinase (CK) activities were analyzed in the plasma and liver triglycerides (TGl) levels were determined in liver tissue samples.The SPSS statistical software was used for the statistical analyses. The obtained data were analyzed for normality by the Shapiro-Wilk test and for homoscedasticity by Bartlett test. Data were submitted to analysis of variance, and means were compared by the t-test (PROC TTEST). Pearson correlation test (PROC CORR) was used to compare the results between prepartum and postpartum cows, and between those with and without lipidosis.An overall incidence of 20% severe hepatic lipidosis was determined both in prepartum and postpartum cows. Positive significant correlations were obtained between days in milk (DIM) and AST (r= 0.43; P &lt; 0.01), CH and TGp (r= 0.44; P &lt; 0.01), TGl and AST (r=0.32; P &lt; 0.05), NEFA and AST (r= 0.45; P &lt; 0.01) and CK and AST (r= 0.447; P &lt; 0.01). Negative significant correlations were detected between TGp and DIM (r= -0.45; P &lt; 0.01), and between TGl and TGp (r= -0.30; P &lt; 0.05).Discussion: Prepartum cows showed moderate fat infiltration in the liver, while 25.7% of postpartum cows presented severe lipidosis. AST values increased as lactation progressed, suggesting a greater tendency of liver damage, in early postpartum. The obtained CK values were within the reference range, except for the cows with hepatic lipidosis, indicating that the measured AST activity was originated from the liver and not from the muscle tissues. TGp, NEFA and AST values indicate the degree of fat mobilization and, possibly, liver infiltration, while TGl indicates fat infiltration in the liver. Although negative energy balance (NEB) may trigger lipolysis and some degree of hepatic lipidosis, plasma BHB values did not significantly increase. The obtained results show that the evaluated plasma parameters may be useful to establish guidelines for the identification of changes in the metabolic profile of dairy cows during different production stages, as well as measures for the prevention and control of metabolic disorders.

Effect of yeast products supplementation during transition period on metabolic profile and milk production in dairy cows.

The aim of the current study was to assess the effect that supplementation with yeast culture plus enzymatically hydrolyzed yeast (YC-EHY) during the transition period and lactation had on the performance, somatic cell count (SCC), and metabolic profile of dairy cows. Thirty multiparous Holstein dairy cows were divided into two groups. The treatments were 0 supplementation (control) and supplementation with 28g/cow/day of YC-EHY. The supplementation began 35 ± 5days before the expected calving date. The cows were kept in their respective treatments for 50days after the calving date. Body condition score (BCS), body weight, milk composition, SCC, and milk yield were assessed on a 2-weekly basis. Plasma samples were collected on days - 21st, - 7th, 0, 3rd, 7th, and weekly thereafter until 42days postpartum and analyzed for albumin, β-hydroxybutyrate (BHBA), urea, and non-esterified fatty acids (NEFA). There was an effect of treatment on milk yield in the supplemented animals in comparison to the control group (27.88 ± 0.98 vs 24.58 ± 0.99kg/days, P = 0.03). There was no effect of treatment (P > 0.05) on variables like 3.5% fat-corrected milk (FCM) and energy-corrected milk (ECM), milk component (%), milk composition yield (kg/day), and SCC. There was an interaction between group × days on ECM (P = 0.04) and protein (P = 0.008). The supplementation had no effect on the analyzed metabolites and on body weight and BCS. The supplementation with YC-EHY during the transition period and lactation improved milk yield without altering the metabolic profile.

Milk Metabolomics Data Reveal the Energy Balance of Individual Dairy Cows in Early Lactation

In early lactation, dairy cows typically have a negative energy balance which has been related to metabolic disorders, compromised health and fertility, and reduced productive lifespan. Assessment of the energy balance, however, is not easy on the farm. Our aims were to investigate the milk metabolic profiles of dairy cows in early lactation, and to obtain models to estimate energy balance from milk metabolomics data and milk production traits. Milk samples were collected in week 2 and 7 after calving from 31 dairy cows. For each cow, the energy balance was calculated from energy intake, milk production traits and body weight. A total of 52 milk metabolites were detected using LC-QQQ-MS. Data from different lactation weeks was analysed by partial least squares analysis, the top 15 most relevant variables from the metabolomics data related to energy balance were used to develop reduced linear models to estimate energy balance by forward selection regression. Milk fat yield, glycine, choline and carnitine were important variables to estimate energy balance (adjusted R2: 0.53 to 0.87, depending on the model). The relationship of these milk metabolites with energy balance is proposed to be related to their roles in cell renewal.

Plasma amino acids and metabolic profiling of dairy cows in response to a bolus duodenal infusion of leucine.

Leucine (Leu), one of the three branch chain amino acids, acts as a signaling molecule in the regulation of overall amino acid (AA) and protein metabolism. Leucine is also considered to be a potent stimulus for the secretion of insulin from pancreatice β-cells. Our objective was to study the effects of a duodenal bolus infusion of Leu on insulin and glucagon secretion, on plasma AA concentrations, and to do a metabolomic profiling of dairy cows as compared to infusions with either glucose or saline. Six duodenum-fistulated Holstein cows were studied in a replicated 3 × 3 Latin square design with 3 periods of 7 days, in which the treatments were applied at the end of each period. The treatments were duodenal bolus infusions of Leu (DIL; 0.15 g/kg body weight), glucose (DIG; at Leu equimolar dosage) or saline (SAL). On the day of infusion, the treatments were duodenally infused after 5 h of fasting. Blood samples were collected at -15, 0, 10, 20, 30, 40, 50, 60, 75, 90, 120, 180, 210, 240 and 300 min relative to the start of infusion. Blood plasma was assayed for concentrations of insulin, glucagon, glucose and AA. The metabolome was also characterized in selected plasma samples (i.e. from 0, 50, and 120 min relative to the infusion). Body weight, feed intake, milk yield and milk composition were recorded throughout the experiment. The Leu infusion resulted in significant increases of Leu in plasma reaching 20 and 15-fold greater values than that in DIG and SAL, respectively. The elevation of plasma Leu concentrations after the infusion led to a significant decrease (P<0.05) in the plasma concentrations of isoleucine, valine, glycine, and alanine. In addition, the mean concentrations of lysine, methionine, phenylalanine, proline, serine, taurine, threonine, and asparagine across all time-points in plasma of DIL cows were reduced (P<0.05) compared with the other groups. In contrast to the working hypothesis about an insulinotropic effect of Leu, the circulating concentrations of insulin were not affected by Leu. In DIG, insulin and glucose concentrations peaked at 30–40 and 40–50 min after the infusion, respectively. Insulin concentrations were greater (P<0.05) from 30–40 min in DIG than DIL and SAL, and glucose was elevated in DIG over DIL and SAL from 30–75 min and 40–50 min, respectively. Multivariate metabolomics data analysis (principal component analysis and partial least squares discriminant analysis) revealed a clear separation when the DIL cows were compared with the DIG and SAL cows at 50 and 120 min after the infusion. By using this analysis, several metabolites, mainly acylcarnitines, methionine sulfoxide and components from the kynurenine pathway were identified as the most relevant for separating the treatment groups. These results suggest that Leu regulates the plasma concentrations of branched-chain AA, and other AA, apparently by stimulating their influx into the cells from the circulation. A single-dose duodenal infusion of Leu did not elicit an apparent insulin response, but affected multiple intermediary metabolic pathways including AA and energy metabolism by mechanisms yet to be elucidated.

Assessment of the main plasma parameters included in a metabolic profile of dairy cow based on Fourier Transform mid-infrared spectroscopy: preliminary results.

BackgroundAlthough a metabolic profile represents a valid tool utilized in dairy herds to determine abnormalities in blood chemistry related to an increased risk of production diseases, there are no studies on application of Fourier Transform mid-infrared (FT-MIR) spectroscopy. This study assesses the potential application of FT-MIR to analyze the main blood biochemical parameters included in the metabolic profile of dairy cows. Infrared transmission spectra were acquired for 35 plasma samples (two replicates on each sample) of Italian Friesian dairy cows (14 primiparous and 21 pluriparous), all without clinical events, and at different stages of lactation, although mainly in the transition phase. Each sample was also analyzed independently using accepted reference clinical chemical methods and these results were used as calibrating values to perform predictive models by PLS method using cross validation.ResultsMeasured blood parameters concentrations were all within the reference ranges reported for healthy dairy cows. The number of extracted factors with the PLS procedure for each prediction model ranged between 3 and 7. The coefficient of determination (R2) of the prediction models ranged between 0.1 to values close to 1. R2 values greater than 0.9 were observed for the prediction models of total cholesterol, total protein, globulin, and albumin; values between 0.75 and 0.9 were observed for urea, NEFA, and total bilirubin, while values of R2 lower than 0.6 were observed for all minerals and for enzyme activity. The range error ratio (RER) and prediction to deviation (RPD) ranged from 5.1 to 43.8 and from 1 to 13.8 for RER and RPD, respectively. Values of RPD greater than 5 were observed for total cholesterol, total protein, albumin, and globulin. RPD ranged between 2 and 5 for the prediction models of urea, NEFA, and total bilirubin, while RPD and RER were low for minerals and enzyme activities.ConclusionsAlthough the results of this study require further validation, the use of FT-MIR spectroscopy was possible and provides fairly accurate measurement of various parameters of great importance in the evaluation of the metabolic and inflammatory status in dairy cows.

Nuclear magnetic resonance-based serum metabolic profiling of dairy cows with footrot.

Footrot is a debilitating and contagious disease in dairy cows, caused by the Gram-negative anaerobe Dichelobacter nodosus. 1H-NMR (nuclear magnetic resonance)-based metabolomics has been previously used to understand the pathology and etiology of several diseases. The objective of this study was to characterize serum from dairy cows with footrot (n=10) using 1H-NMR-based metabolomics and chemometric analyses. 1H-NMR spectroscopy with multivariate pattern recognition (principal component analysis and orthogonal partial least-squares discriminant analysis) was performed to identify biomarkers in cows with footrot (F) and healthy controls (C). 1H-NMR analysis facilitated the identification of 21 metabolites. Among these metabolites, 4 metabolites were higher and 17 metabolites were lower in the F group than in the C group. The serum levels of 5 metabolites were significantly different (P<0.05) between the two groups. The results revealed that cows with footrot have altered carbohydrate, amino acid, lipid and energy metabolic pathways. Metabolomic approaches are a clinically useful diagnostic tool for understanding the biochemical alterations and mechanisms of several diseases.

Identifying alterations in metabolic profiles of dairy cows over the past two decades in Japan using principal component analysis

The aim of this study was to identify the metabolic changes that have occurred in Holstein-Friesian cows in Japan over the past 2 decades, based on long-term metabolic profiles. From 1987 to 2004, metabolic profile tests were conducted in 1,700 dairy herds containing ~50,000 cows. The cows were divided into 5 lactation stage groups: early (calving to 49 d in milk, DIM), peak (50–109 DIM), mid (110–209 DIM), and late (210 to dry-off) lactation and the dry period. Principal component analysis was applied to the metabolic profiles at each lactation stage separately to investigate changes in metabolic profiles across the multiyear periods 1987–1992, 1993–1998, and 1999–2004. We determined that cows have probably experienced increasing negative energy balance, energy deficiency, and liver dysfunction during the past 2 decades in Japan.

(1)H-Nuclear Magnetic Resonance-Based Plasma Metabolic Profiling of Dairy Cows with Fatty Liver.

Fatty liver is a common metabolic disorder of dairy cows during the transition period. Historically, the diagnosis of fatty liver has involved liver biopsy, biochemical or histological examination of liver specimens, and ultrasonographic imaging of the liver. However, more convenient and noninvasive methods would be beneficial for the diagnosis of fatty liver in dairy cows. The plasma metabolic profiles of dairy cows with fatty liver and normal (control) cows were investigated to identify new biomarkers using 1H nuclear magnetic resonance. Compared with the control group, the primary differences in the fatty liver group included increases in β-hydroxybutyric acid, acetone, glycine, valine, trimethylamine-N-oxide, citrulline, and isobutyrate, and decreases in alanine, asparagine, glucose, γ-aminobutyric acid glycerol, and creatinine. This analysis revealed a global profile of endogenous metabolites, which may present potential biomarkers for the diagnosis of fatty liver in dairy cows.

Neste estudo foi avaliado o perfil metabólico de vacas leiteiras no período de transição durante o verão e o inverno. Foram utilizados 31 animais pluríparos mestiços girolando em cada estação, totalizando 62 vacas. No verão permaneciam em pasto com suplementação de silagem de milho e concentrado. No inverno, a exigência nutricional era suprida apenas com silagem de milho e concentrado. Foram feitas um total de 11 coletas de sangue de cada animal segundo o seguinte protocolo: quatro coletas pré-parto espaçadas semanalmente, no dia do parto e com 2, 5, 10, 15, 21 e 30 dias pós-parto. Foram avaliadas as concentrações de cálcio, fósforo e magnésio. Todos os analitos variaram em função do estatus fisiológico. As concentrações médias de cálcio e magnésio foram maiores no verão do que no inverno. Em ambas as estações, as menores concentrações médias de cálcio ocorreram próximas ao parto, sendo que 75% dos animais no inverno e 35,48% dos animais no verão estavam hipocalcêmicos. Apenas no décimo dia as concentrações de cálcio voltaram aos níveis do pré-parto, demonstrando que esse tempo é necessário para a adaptação da nova condição de lactante. As concentrações de magnésio foram menores no pós-parto do que no pré-parto nas duas estações, demonstrando a necessidade desse mineral para a produção de leite. As concentrações médias de magnésio permaneceram sempre dentro dos valores de referência, porém 19% dos animais aos dois dias pós-parto no inverno e 7% dos animais aos 10 dias pós-parto no verão tinham concentrações inferiores a esses limites. As concentrações de fósforo sofreram grande variação ao longo do peri-parto, porém sempre em níveis elevados, sendo que 49,8% dos animais no inverno e 37,3% no verão tinham níveis de fósforo acima dos valores de referência

1H NMR-based Plasma Metabolic Profiling of Dairy Cows with Type I and Type II Ketosis

This study identified differences in plasma metabolites among three groups of dairy cows: type I ketotic (K1), type II ketotic (K2), and healthy control cows (C). 50 cows with two or three parities were selected at 7–28 days postpartum. Cows were classified as type I ketotic (K1, 20 cows), type II ketotic (K2, 20 cows), or healthy control cows (C, 10 cows). Plasma metabolomic profiles were analyzed by 1H-nuclear magnetic resonance technology (1H NMR). The data were processed by principal component analysis and orthogonal partial least-squares discriminant analysis (OPLS-DA). Results-The results revealed that OPLS-DA was more effective at distinguishing amongst the three groups. Additionally, there were seven different metabolites between K2 and C, 19 different metabolites between K1 and C, and 24 different metabolites between K1 and K2. Therefore, the combination of 1H-NMR and multivariate statistical analyses can effectively distinguish the differential metabolites among the K1, K2, and C groups, thereby providing important information on the pathogenesis, early diagnosis, and prevention of type I and type II ketosis in dairy cows.

Plasma metabolic profiling of dairy cows affected with clinical ketosis using LC/MS technology

Background: Ketosis in dairy cattle is an important metabolic disorder. Currently, the plasma metabolic profile of ketosis as determined using liquid chromatography–mass spectrometry (LC/MS) has not been reported.Objective: To investigate plasma metabolic profiles from cows with clinical ketosis in comparison to control cows.Animals and methods: Twenty Holstein dairy cows were divided into two groups based on clinical signs and plasma β-hydroxybutyric acid and glucose concentrations 7–21 days postpartum: clinical ketosis and control cows. Plasma metabolic profiles were analyzed using LC/MS. Data were processed using principal component analysis and orthogonal partial least-squares discriminant analysis.Results: Compared to control cows, the levels of valine, glycine, glycocholic, tetradecenoic acid, and palmitoleic acid increased significantly in clinical ketosis. On the other hand, the levels of arginine, aminobutyric acid, leucine/isoleucine, tryptophan, creatinine, lysine, norcotinine, and undecanoic acid decreased markedly.Conclusion: Our results showed that the metabolic changes in cows with clinical ketosis involve complex metabolic networks and signal transduction. These results are important for future studies elucidating the pathogenesis, diagnosis, and prevention of clinical ketosis in dairy cows.

Characterization of the serum metabolic profile of dairy cows with milk fever using 1H-NMR spectroscopy

Background: Milk fever (MF) is a common calcium metabolism disorder in perinatal cows. Currently, information regarding the detailed metabolism in cows suffering from MF is scant.Objective: The purpose was to study the metabolic profiling of serum samples from cows with MF in comparison to control cows, and thereby exploring other underlying pathological mechanisms of this disease.Animals and methods: In the current study, we compared the serum metabolomic profile of dairy cows with MF (n = 8) to that of healthy dairy cows (n = 24) using a 500-MHz digital 1H-nuclear magnetic resonance (1H-NMR) spectrometer. Based on their clinical presentation and serum calcium concentration, cows were assigned either to the control group (no MF symptoms and serum calcium concentration >2.5 mmol/L) or to the MF group (MF symptoms and serum calcium concentration <1.4 mmol/L). For statistical analysis, a one-way analysis of variance was performed.Results: We identified differences regarding nine metabolites between the two groups, among which glucose, alanine, glycerol, phosphocreatine, and gamma-aminobutyrate decreased, and β-hydroxybutyrate, acetone, pyruvate, and lysine increased in cows with MF. Most of these were carbohydrates and amino acids involved in various energy metabolism pathways.Conclusion: The different metabolites in cows with MF reflected the pathological features of negative energy balance and fat mobilization, suggesting that MF is associated with altered energy metabolism.Clinical importance: The 1H-NMR spectroscopy can be used to understand the pathogenesis of MF and identify biomarkers of the disease.
