Metabolic coordination of liver and kidney in mercapturic acid biosynthesis in vivo.

When S-carbamido(14C)methyl glutathione, a model compound of glutathione S-conjugate, was administered i.v. to mice, radioactivity accumulated in the kidney within 1 to 2 min and then decreased gradually during the following 10 to 15 min with concomitant increase in hepatic radioactivity. Most hepatic radioactivity was accounted for by S-carbamidomethyl cysteine and its N-acetyl derivative, a mercapturic acid. The i.v. administration of S-carbamido(14C)methyl cysteine resulted in rapid and predominant accumulation of radioactivity in the liver. In both cases, the radioactive urinary metabolites were fully accounted for by N-acetyl-S-carbamidomethyl cysteine. N-Acetyl-S-carbamido(14C)methyl cysteine administered to mice were accumulated preferentially in the kidney and was excreted into urine very rapidly. These results suggest the following series of events: glutathione S-conjugate accumulated mainly in the kidney and is hydrolyzed into its component amino acids, presumably by gamma-glutamyl transferase and some peptidase(s) on the renal brush border membranes. The cysteine S-conjugate which is formed in the tubular lumen is reabsorbed and transferred to the liver, acetylated to form N-acetylcysteine S-conjugate, and excreted in the urine. Thus, renal hydrolysis of glutathione S-conjugates seems to be coordinated with acetylation in liver and with mercapturic acid biosynthesis in vivo.

GLUTATHIONE S-CONJUGATE FORMATION AND METABOLISM IN HepG2 CELLS: A CELL MODEL OF MERCAPTURIC ACID BIOSYNTHESIS

Mercapturic acid biosynthesis is mediated by a series of at least four enzymatic steps and three cell membrane transport events, and is believed to require the interorgan shuttling of the various metabolic intermediates. To identify a single cell type that can carry out all of these m etabolic and transport steps, the present study exam ined whether HepG2 cells, a human hepatoma-derived cell line, can convert an electrophilic chemical (1-chloro-2,4-dinitrobenzene, CDNB) to its corresponding mercapturic acid (S -dinitrophenyl- N -acetylcysteine, DNP-NAC). The results demonstrate that HepG2 cells are able to convert CDNB to DNP-NAC in a dose- and time-dependent fashion. Intracellular conjugation with glutathione occurred rapidly, and the resulting glutathione S -conjugate was promptly transported into the culture medium, where it was sequentially degraded to the cysteinylglycine and cysteine S -conjugates. The cysteine conjugate was then presumably reabsorbed, and N -acetylated intracellularly to form the mercapturic acid. The mercapturic acid was found to accumulate slowly in the culture medium, such that after 4 h of incubation, 4-10% of the CDNB dose was recovered as the mercapturic acid. These data provide the first demonstration that a single cell type can carry out all of the transport and enzymatic steps required for mercapturic acid biosynthesis. HepG2 cells may provide a useful model system for studying this important detoxification pathway.

Purification and characterization of cysteine-S-conjugate N-acetyltransferase from pig kidney.

Microsomal cysteine-S-conjugate N-acetyltransferase catalyses the N-acetylation of various S-substituted cysteines in liver and kidney. We describe here the purification and more detailed characterization of this enzyme catalysing the final reaction of mercapturic acid biosynthesis, and thus playing a crucial role in the detoxicating metabolism of many xenobiotics. The solubilization of cysteine-S-conjugate N-acetyltransferase by deoxy-BIGCHAP [N,N'-bis-(3-D-gluconamidopropyl)deoxycholamide] was the prerequisite for partial purification by means of anion-exchange chromatography. The molecular mass of the enzyme was determined by gel filtration. A polyclonal antiserum was raised against the excised protein band from SDS/PAGE and purified antibodies were used for the complete purification of native cysteine-S-conjugate N-acetyltransferase by immuno-affinity chromatography. A dimeric form of the enzyme was sometimes detected on SDS/PAGE, depending on the degree of purification. For further characterization of cysteine-S-conjugate N-acetyltransferase, the stability of catalytic activity, the pH optimum and K(m) values were determined. The inhibitory effects of various agents were tested, revealing a substantial, yet not complete, loss of cysteine-S-conjugate N-acetyltransferase activity after treatment with cysteine proteinase inhibitors or probenecid under various conditions.

Oxidation and acetylation as determinants of 2-bromocystein-S-ylhydroquinone-mediated nephrotoxicity.

2-Bromodiglutathion-S-ylhydroquinone is a more potent nephrotoxicant than 2-bromomonoglutathion-S-ylhydroquinones. In the present study we examined the activity of enzymes involved in mercapturic acid biosynthesis toward both the glutathione conjugates and their cysteine and N-acetylcysteine metabolites and compared the results to the relative nephrotoxicity of these conjugates. Although differences were observed in the kinetics of the gamma-glutamyl transpeptidase (gamma-GT)-mediated hydrolysis and transpeptidation of the glutathione conjugates, the concentration of this enzyme within the kidney probably precludes it from contributing to their differential toxicity. In contrast, the rate at which the cysteine and corresponding mercapturate conjugates underwent N-deacetylation/N-acetylation cycling correlated with previously reported differences in toxicity. The relative rates of these two reactions are important because electrochemical data suggest that 2-bromodicystein-S-ylhydroquinone is more readily oxidized to the reactive quinone than its corresponding mercapturic acid. In addition, 2-bromodi(N-acetylcystein-S-yl)hydroquinone, which is the most potent of the mercapturic acid conjugates, exhibited the highest N-deacetylation/N-acetylation ratio. In contrast, 2-bromo-3-(N-acetylcystein-S-yl)hydroquinone, which is essentially not toxic in vivo, was not a substrate for the renal cysteine conjugate N-deacetylase. The data suggest that the rate-determining step for the in vivo toxicity of these conjugates is probably the N-acetylation reaction and the availability of the corresponding acetyl-CoA cofactor.

Glutathione conjugation and conversion to mercapturic acids can occur as an intrahepatic process.

By catalyzing the reaction of electrophilic compounds with the sulfhydryl group of glutathione, the glutathione S-transferases play physiologically important roles in the detoxication of potential alkylating agents. The glutathione S-conjugates thus formed are transported out of cells for further metabolism by gamma-glutamyltransferase and dipeptidases, ectoproteins that catalyze the sequential removal of the glutamyl and glycyl moieties, respectively. These ectoproteins are not found in all cells, but are localized predominantly to the apical surface of epithelial tissues. The resulting cysteine S-conjugates can be reabsorbed by specific cell types, and acetylated on the amino group of the cysteinyl residue by intracellular N-acetyl-transferases, to form the corresponding mercapturic acids (N-acetylcysteine S-conjugates). Mercapturic acids are then released into the circulation and delivered to the kidney for excretion in urine, or they may undergo further metabolism. Mercapturic acid biosynthesis is generally considered to be an interorgan process, with the liver serving as the major site of glutathione conjugation, and the kidney as the primary site for conversion of glutathione conjugates to cysteine conjugates. Cysteine conjugates formed in the kidney appear to be transported back to the liver for acetylation. This interorgan model of mercapturic acid synthesis is based largely on the interorgan distribution of the enzymes involved in their formation, and in particular of the enzyme gamma-glutamyltransferase. Rats have relatively low hepatic and high renal activities of gamma-glutamyltransferase, the only protein known to initiate the breakdown of glutathione S-conjugates. The low gamma-glutamyltransferase activity in rat liver limits the hepatic degradation of glutathione S-conjugates, particularly after large doses of xenobiotic. In contrast, hepatic gamma-glutamyltransferase is significantly higher in species such as rabbit, guinea pig, and dog, and as a consequence, nearly all of the glutathione and glutathione S-conjugates released by liver cells of these species is degraded within the liver. Recent studies demonstrate that glutathione S-conjugates synthesized within hepatocytes are secreted preferentially across the canalicular membrane into bile, and are broken down within biliary spaces to form cysteine S-conjugates. The latter are then reabsorbed by the liver, N-acetylated to form mercapturic acids, and reexcreted into bile, completing an intrahepatic pathway for mercapturic acid biosynthesis. The contribution of this intrahepatic pathway to overall mercapturate formation is dependent on dose of the electrophile, route of exposure, and the physicochemical properties of the glutathione S-conjugate formed, as well as the tissue distribution and activity of gamma-glutamyltransferase.(ABSTRACT TRUNCATED AT 400 WORDS)

Amino acid transport and glutathione homeostasis: What is the mechanism for cysteine uptake from bile?

Transport of L-cysteine and a cysteine S-conjugate, S-(1,2-dichlorovinyl)-L-cysteine (DCVC) was investigated in rat liver canalicular plasma membrane (cLPM) vesicles. Cysteine uptake into an osmotically active intravesicular space was temperature sensitive and further enhanced by an inwardly directed Na+ gradient. Na+ -dependent and -independent L-cysteine uptake exhibited saturation kinetics with apparent Km of 53 ± 0.7 and 1300 ± 300 μM and Vmax of 95 ± 21 and 1600 ± 200 pmol ± mg protein−1; 10 sec−1 for the Na+ dependent components, and an apparent Km of 207 ± 48 μM and a Vmax of 355 ± 71 pmol ± mg protein−1 ± 10 sec−1 for the Na+ -independent component. Na+-dependent uptake was inhibited by L-alanine, glycine, L-phenylalanine and L-leucine, whereas Na+-independent uptake was inhibited by L-phenylalanine, L-leucine and 2-amino-2-norbornanecarboxylic acid. Both Na+ -dependent and -independent L-cysteine transport processes were inhibited by several cysteine S-conjugates, with DCVC having the strongest effect. Inhibition of [35S]-cysteine uptake by DCVC was noncompetitive with a K, of 1.2 ± 0.1 mM. On the other hand, uptake of [35 S IDCVC by the rat cLPM vesicles was not stimulated by a Na+-gradient, but was inhibited by several other amino acids, including L-cysteine. Further investigation of [35SlDCVC uptake in rat cLPM vesicles indicated a saturable Na+ -independent process with an apparent K, of 155 f 42 μM, and a Vmax of 393 f 53 pmol mg protein −1 5 sec −1. Time-dependent nonspecific binding of DCVC to rat cLPM vesicles was diminished by addition of aminooxyacetic acid, an inhibitor of cysteine conjugate β-lyase activity. These findings demonstrate that cysteine and DCVC uptake into cLPM vesicles occurs by both saturable and nonsaturable transport mechanisms. These transport pathways may function in vivo for uptake and recycling of biliary L-cysteine for glutathione turnover and some cysteine S-conjugates for intrahepatic mercapturic acid biosynthesis.

Polarity of hepatic glutathione and glutathione S-conjugate efflux, and intraorgan mercapturic acid formation in the skate

Mechanisms of hepatic glutathione and glutathione S-conjugate efflux were investigated in isolated hepatocytes and perfused liver of the little skate ( Raja erinacea). Glutathione was released by isolated skate hepatocytes at a rate of 0.12 ± 0.03 nmol · hr −1 · (mg protein) −1. In the perfused liver, glutathione concentrations in bile were high (∼0.7mM) compared to hepatic tissue levels (0.61 ± 0.11 μmol · g −1). During the first hour of perfusion, the biliary glutathione excretion rate was 3 nmol · hr −1 · (g liver) −1, whereas glutathione accumulated in the recirculating perfusate at a rate of only 1.5nmol · hr −1 · (g liver) −1. Release of glutathione by isolated hepatocytes and perfused liver was not affected by the addition of acivicin, an inhibitor of γ-glutamyltransferase (EC 2.3.2.2), to cell suspension medium or liver perfusate. 1-Chloro-2,4-dinitrobenzene (CDNB) was taken up by isolated hepatocytes, conjugated to glutathione, and released as 5-(2,4-dinitrophenyl) (DNP)-glutathione. After infusion of 0.5 μmol CDNB in perfused liver, S-DNP-glutathione was concentrated in bile (0.5 mM) and was associated with choleresis. S-DNP-Conjugates of cysteinylglycine, cysteine and N-acetylcysteine, were also found in bile, suggesting intrahepatic breakdown of S-DNP-glutathione and subsequent acetylation of the resulting cysteine conjugate to form the mercapturic acid, S-DNP- N-acetylcysteine. This mercapturic acid accounted for 31% of the total S-DNP-conjugates collected in bile. In contrast, neither S-DNP-glutathione nor other S-DNP-conjugates were detected in the perfusate (< 0.5 μM). These findings demonstrate that biliary excretion is the predominant route for efflux of glutathione and a glutathione S-conjugate from skate liver. The results also identify an intrahepatic pathway for mercapturic acid biosynthesis facilitated by biliary glutathione S-conjugate excretion.

Intrahepatic conversion of a glutathione conjugate to its mercapturic acid. Metabolism of 1-chloro-2,4-dinitrobenzene in isolated perfused rat and guinea pig livers.

Because of the low hepatic activity of gamma-glutamyl-transferase in the rat, the liver is generally considered to play only a minor role in the degradation of glutathione conjugates, a limiting step in mercapturic acid formation. Recent findings indicate, however, that the liver has a prominent role in glutathione catabolism, particularly in species other than rat. To examine the contributions of liver to mercapturic acid biosynthesis, mercapturate formation was compared in isolated perfused livers from rats and guinea pigs dosed with either 0.3 or 3.0 mumol of 1-chloro-2,4-dinitrobenzene (CDNB). Chemically synthesized glutathione conjugate, mercapturic acid, and intermediary metabolites of CDNB were used as standards in the high performance liquid chromatography analysis of bile and perfusate samples. Biliary excretion accounted for almost all of the recovered metabolites. A marked species difference was observed in the pattern of CDNB metabolism. Rat livers dosed with 0.3 mumol of CDNB excreted 55% of total biliary metabolites as the glutathione conjugate and 8.2% as the mercapturic acid, whereas guinea pig livers excreted only 4.8% as the glutathione conjugate and 47% as the mercapturate. Mercapturic formation was also dose-dependent, with a larger fraction formed at the 0.3- versus the 3.0-mumol dose (8.2 versus 3.7% in the rat; 47 versus 19% in the guinea pig). Hepatic conversion of the glutathione conjugate to the mercapturic acid was markedly inhibited in both species after retrograde intrabiliary infusion of acivicin, an inhibitor of gamma-glutamyltransferase activity. These findings provide direct evidence for intrahepatic biosynthesis of mercapturic acids. Thus, glutathione conjugates synthesized within hepatocytes are secreted into bile and broken down to cysteine conjugates; the latter are then presumably reabsorbed by the liver, N-acetylated to form the mercapturic acid and re-excreted into bile.

Localization and capacity of the last step of mercapturic acid biosynthesis and the reabsorption and acetylation of cysteine S-conjugates in the rat kidney

We investigated the capacity and the localization of N-acetylation of the mercapturic acid precursor S-benzyl-L-cysteine (BC), as well as the tubular reabsorption of this compound in the rat kidney in vivo et situ by renal clearance and continuous microinfusion and microperfusion experiments. In renal clearance experiments. 450 mumol BC was infused intravenously for 180 min. During the time of BC infusion and the following 180 min, the two kidneys excreted 400 mumol or 90% of the infused BC dose as the mercapturate N-acetyl-S-benzyl-L-cysteine (AcBC). Comparison of the amounts of BC and AcBC entering the left kidney via the renal artery with those leaving it via the renal vein and the ureter showed that 0.13 +/- 0.04 mumol BC/min (mean +/- SEM) was extracted and 0.24 +/- 0.08 mumol AcBC/min was formed by one kidney. The intrarenal acetylation can account for the formation of 38% of the mercapturate excreted in the final urine. In additional experiments, 50 pmol/min [14C]BC was microinfused into single superficial tubules at three different sites. During microinfusion into early proximal tubules, the final urine contained 16.3 +/- 1.8% of the microinfused radioactivity as AcBC, but no BC. When [14C]BC was microinfused into late proximal tubules, 13.0 +/- 2.3% of the infused label was recovered as BC, 28.1 +/- 2.3% as AcBC. During microinfusion into early distal tubules, the final urine contained no AcBC, but 90.3 +/- 2.1% of the infused [14C]BC was recovered.(ABSTRACT TRUNCATED AT 250 WORDS)

Is the Toxicity of Cysteine Conjugates Formed During Mercapturic Acid Biosynthesis Relevant to the Toxicity of Covalently Bound Drug Residues?

In this brief review, we have focused on the relevance of the data on cysteine conjugate toxicity to the potential hazard of bound drug residues. A resonable scenario, based on assumptions as well as literature data, has been presented for the release of cysteine conjugates of drug residues from protein. Furthermore, we have presented evidence that should this occur, the conjugate would be bioavailable. Finally, the mechanisms which could lead to cysteine conjugate-induced toxicity have been discussed. The question which must be answered is, how realistic is the treat of toxicity to the consumer from cysteine-bound drug residues in food products? Based on the data presented here, the danger is minimal, though it cannot be excluded. This is particularly true of the potential for renal complications. However, an important caveat which must not be overlooked is the marked species differences in cysteine conjugate toxicity. Though S-(1,2LD50-dichlorovinyl)-L-cysteine (DCVC) is a renal toxin in rodent models (LD50 = 66-83 mg/kg) [88], a single dose of 4-5 mg/kg causes fatal aplastic anemia in calves [44,59]. Though such a response has never been reported for any other cysteine conjugate, these data must be reckoned with if attempts are made to place acceptable limits on the amount of residues allowable in food products.

In vivo metabolism of leukotriene C4 in germ-free and conventional rats. Fecal excretion of N-acetylleukotriene E4.

[5,6,8,9,11,12,14,15-3H8]Leukotriene C4 was subcutaneously injected into rats. Substantial amounts of the administered radioactivity were excreted in feces of germ-free and conventional animals during a 72-h period (78 and 64%, respectively). Analyses of fecal extracts by high performance liquid chromatography showed eight radioactive components for each type of animal. One metabolite amounted to 4.6% of the injected radioactivity in germ-free and 0.6% in conventional rats. Its chemical structure, 5-hydoxy-6-S-(2-acetamido-3-thiopropionyl)-7,9-trans-11,14-c is-eicosatetraenoi c acid (N-acetylleukotriene E4) was determined by ultraviolet spectroscopy, fast atom bombardment mass spectrometry, chemical and enzymatic transformations, and confirmed by chemical synthesis. Another metabolite (2.7% of the administered radioactivity in germ-free and 0.5% in conventional rats) was characterized as the 11-trans isomer of the former metabolite. The pathway of formation of these compounds appears to be analogous to the pathway of mercapturic acid biosynthesis.

66] Cysteine S-conjugate N-acetyltransferase

Publisher Summary Cysteine S-conjugate N-acetyltransferase from rat kidney microsomes catalyzes the final reaction in mercapturic acid biosynthesis, the acetylation of cysteine thioethers. This enzyme is not active with substrates of the cytosolic amine N-acetyltransferase. The transfer of a radiolabeled acetyl group from CoASAc to a cysteine conjugate is used as the basis for measurement of the enzyme. After acidification, the mercapturic acid is extracted into cyclo-hexanone for determination of radioactivity. Solubilization and purification of cysteine S-conjugate N-acetyltransferase is carried out with kidneys from male Sprague-Dawley rats of 175–200 g. Kidneys are stored at –70° prior to use. All purification steps are conducted at 4°. The steps of the purification procedure include preparation of microsomes, solubilization, and hydroxylapatite. Although the enzyme obtained by this procedure is not purified to high specific activity, it is soluble and looses only 30% of its activity after storage at 4° for 3 months.

Topological aspects of microsomal N-acetyltransferase, an enzyme responsible for the acetylation of cysteine S-conjugates of xenobiotics.

Acetylation of cysteine S-conjugates of xenobiotics by microsomal N-acetyltransferase is the final step of detoxicative metabolism leading to mercapturic acid biosynthesis. To elucidate the subcellular site of N-acetylation and the effective mechanism by which the final metabolites are eliminated from the organisms, topological aspects and catalytic properties of microsomal N-acetyltransferase and mercapturic acid biosynthesis in vivo were investigated. Intravenous administration of radioactive S-benzyl-L-cysteine, a model compound of cysteine S-conjugates, resulted in rapid acetylation of the conjugate in liver and kidney to a similar extent. The acetylation was followed by a rapid excretion of the metabolite, a mercapturic acid, into the urine; about 60% of the injected dose appeared in urine within 60 min of administration. Limited proteolysis of microsomal vesicles obtained from liver and kidney by chymotrypsin or trypsin inactivated the transferase by 49-62% and 62-73%, respectively. Proteolytic inactivation of the transferase was not significantly affected by the presence of 0.04% sodium deoxycholate by which the vesicles became permeable to macromolecules due to its detergent action. To determine the sidedness of the active site of N-acetyltransferase on the microsomal membranes, two S-acetyldextran polymer derivatives (Mr 500 000) of cysteine and N-acetylcysteine which represent an nonpermeant substrate and product for this enzyme, respectively, were examined for their effects on the vesicle-associated enzyme activity. Both derivatives inhibited the transferase activity in a dose-dependent fashion; maximum inhibition of the enzyme activity was 40% by the former and 60% by the latter. Sulfobromophthalein strongly inhibited the enzyme activity and this inhibition was completely reversed by adding an equimolar amount of hepatic glutathione S-transferases (ligandins). In contrast to the strong inhibition by sulfobromophthalein itself, its glutathione S-conjugate did not inhibit the enzyme activity. These results indicate that the active site and the protease-sensitive domain(s) of the microsomal N-acetyltransferase are localized on the outer surface (cytoplasmic side) of endoplasmic reticulum and that the ligandin(s) might protect membranous N-acetyltransferase from inhibition by organic anions by binding them and catalyzing the conjugation with glutathione.

Studies on the effect of phenobarbital on γ-glutamyl transferase activity in various housefly strains

The effect of pretreatment with phenobarbital (PB) on γ-glutamyl transferase (GGT) activity in an insecticide-susceptible housefly strain (CSMA) and three insecticide-resistant housefly strains (Fc, Rutgers, and Hirokawa) was investigated. Pretreatment with PB had little if any effect on GGT activity in the susceptible and resistant housefly strains, indicating that not all the enzymes involved in mercapturic acid biosynthesis in the housefly are inducible.

Renal transtubular transport of mercapturic acid in vivo

When S- benzyl-N- acetyl- l-[ U- 14 C] cysteine , a mercapturic acid, was administered to rats intravenously, the plasma level of radioactivity decreased very rapidly with a concomitant increase in the renal level of radioactivity. The renal radioactivity reached its maximum within 2 min and then decreased rapidly with concomitant appearance of the radioactive mercapturic acid in the urine. Bilateral ligation of the ureters resulted in only a slight decrease in the rate of disappearance of mercapturic acid from the plasma, while bilateral nephrectomy caused a marked retardation of its clearance from the plasma. Intravenous administration of probenecid, a well known inhibitor of a renal transbular transport system for organic acids, caused a significant retardation of mercapturate clearance from the plasma in both of the control and ureter-ligated animals. The renal accumulation of this mercapturic acid as well as its excretion into urine was inhibited by probenecid. All these data suggested that a mercapturic acid in the plasma was preferentially taken up by renal tubule cells from the basolateral side of plasma membranes via the probenecid-sensitive transtubular transport system and then excreted rapidly into the lumenal space. This transtubular transport of a mercapturic acid seems to constitute an important process in the hepato-renal cooperation in the mercapturic acid biosynthesis in vivo.

The effect of phenobarbital on mercapturic acid biosynthesis in the house fly Musca domestica

The metabolism of S-(2,4- dinitrophenyl) glutathione was investigated in the housefly by two routes of application. Various intermediates of S-(2,4- dinitrophenyl) glutathione were isolated and identified by TLC, indicating that the mercapturic acid biosynthesis pathway is operative in the housefly. Induction of mercapturic acid biosynthesis by phenobarbital resulted in a 1.75 fold increase in mercapturic acid formation.

The Role of Glutathione and Glutathione S-Transferases in the Metabolism of Chemical Carcinogens and Other Electrophilic Agents

Publisher Summary This chapter discusses the role of glutathione (GSH) and glutathione s-transferases in metabolism of chemical carcinogens and other electrophilic agents. GSH is a tripeptide (I) that is present in nearly all living cells and is the most abundant sulfhydryl compound present in animal tissues, mainly in the cytosol. The chapter illustrates the wide range of electrophilic agents, including several known mutagens and carcinogens, which conjugate with GSH, a process usually catalyzed by the GSH S-transferases. This conjugation is probably a protective mechanism and is the initial stage in mercapturic acid biosynthesis for the elimination of foreign compounds from the body. GSH S-transferases provide protection not only by catalyzing the conjugation of a potential toxicant with GSH, but also by preferentially binding, even covalently, that toxicant. The reactive electrophiles that conjugate with GSH also bind to DNA, RNA, and protein and identification of GSH conjugates provide information on the nature of these biologically active intermediates or even their immediate precursors. Thus, the knowledge of the way in which mutagens and carcinogens are metabolized is essential to a better understanding of their mode of action and of the processes for their detoxication.

The metabolism of benzyl isothiocyanate and its cysteine conjugate.

1. The corresponding cysteine conjugate was formed when the GSH (reduced glutathione) or cysteinylglycine conjugates of benzyl isothiocyanate were incubated with rat liver or kidney homogenates. When the cysteine conjugate of benzyl isothiocyanate was similarly incubated in the presence of acetyl-CoA, the corresponding N-acetylcysteine conjugate (mercapturic acid) was formed. 2. The non-enzymic reaction of GSH with benzyl isothiocyanate was rapid and was catalysed by rat liver cytosol. 3. The mercapturic acid was excreted in the urine of rats dosed with benzyl isothiocyanate or its GSH, cysteinyl-glycine or cysteine conjugate, and was isolated as the dicyclohexylamine salt. 4. An oral dose of the cysteine conjugate of [14C]benzyl isothiocyanate was rapidly absorbed and excreted by rats and dogs. After 3 days, rats had excreted a mean of 92.4 and 5.6% of the dose in the urine and faeces respectively, and dogs had excreted a mean of 86.3 and 13.2% respectively. 5. After an oral dose of the cystein conjugate of [C]benzyl isothiocyanate, the major 14C-labelled metabolite in rat urine was the corresponding mercapturic acid (62% of the dose), whereas in dog urine it was hippuric acid (40% of the dose). 5. Mercapturic acid biosynthesis may be an important route of metabolism of certain isothiocyanates in some mammalian species.
