Foliar Water Uptake Supports Water Potential Recovery but Does Not Affect Xylem Sap Composition in Two Salt-Secreting Mangroves.

Mangroves are highly salt-tolerant species, which live in saline intertidal environments, but rely on alternative, less saline water to maintain hydraulic integrity and plant productivity. Foliar water uptake (FWU) is thought to assist in hydration of mangroves, particularly during periods of acute water deficit. We investigated the dynamics of FWU in Avicennia marina and Aegiceras corniculatum by submerging and spraying excised branches and measuring leaf water potential (Ψ) at different time intervals. Daily changes in xylem sap composition (ionic concentrations, pH and surface tension) were monitored during 2 days characterised by the presence of morning dew and difference in tides. In both species, FWU occurred over relatively short times, with leaf Ψ recovering from -4.5 MPa to about -1.5 MPa in 120-150 min. At predawn, Ψ was higher (-1.5 MPa) than sea water Ψ, indicating that leaves had been partially rehydrated by absorbed dew. Tides did not affect Ψ, but high tides increased the overall ionic content of xylem sap. The results indicated mangroves are extremely efficient in absorbing non-saline water via the leaves and restoring the water balance to Ψ higher than seawater. Changes in xylem sap composition, which were strongly influenced by tides, were not affected by observed FWU.

Should we delay leaf water potential measurements after excision? Dehydration or equilibration?

BackgroundAccurate leaf water potential (Ψw) determination is crucial in studying plant responses to water deficit. After excision, water potential decreases, even under low evaporative demand conditions, which has been recently attributed to the equilibration of pre-excision Ψw gradients across the leaf. We assessed the influence of potential re-equilibration on water potential determination by monitoring leaf Ψw and relative water content decline after excision using different storage methods.ResultsEven though leaf Ψw declined during storage under low evaporative demand conditions, this was strongly reduced when covering the leaf with a hydrophobic layer (vaseline) and explained by changes in relative water content. However, residual water loss was variable between species, possibly related to morpho-physiological leaf traits. Provided water loss was minimized during storage, pre-excision leaf transpiration rate did not affect to the magnitude of leaf Ψw decline after excision, confirming that transpiration-driven Ψw gradients have no effect on leaf Ψw determination.ConclusionsDisequilibrium in water potentials across a transpiring leaf upon excision is dissipated very quickly, well within the elapsed time between excision and pressurization, therefore, not resulting in overestimation of leaf Ψw measured immediately after excision. When leaf storage is required, the effectiveness of a storage under low evaporative demand varied among species. Covering with a hydrophobic layer is an acceptable alternative.

Dynamic soil hydraulic resistance regulates stomata.

The onset of stomatal closure reduces transpiration during drought. In seed plants, drought causes declines in plant water status which increases leaf endogenous abscisic acid (ABA) levels required for stomatal closure. There are multiple possible points of increased belowground resistance in the soil-plant atmospheric continuum that could decrease leaf water potential enough to trigger ABA production and the subsequent decreases in transpiration. We investigate the dynamic patterns of leaf ABA levels, plant hydraulic conductance and the point of failure in the soil-plant conductance in the highly embolism-resistant species Callitris tuberculata using continuous dendrometer measurements of leaf water potential during drought. We show that decreases in transpiration and ABA biosynthesis begin before any permanent decreases in predawn water potential, collapse in soil-plant hydraulic pathway and xylem embolism spread. We find that a dynamic but recoverable increases in hydraulic resistance in the soil in close proximity to the roots is the most likely driver of declines in midday leaf water potential needed for ABA biosynthesis and the onset of decreases in transpiration.

Organic soil amendment effects on soil hydrology in an almond orchard evaluated using time-lapse electrical resistivity tomography

Adding organic amendments to soils in orchards has been suggested as a climate-smart agricultural practice that can increase resilience to extremes such as drought. The benefits of adding almond hulls and shells to soil include releasing potassium into the soil, improving water infiltration, reducing soil water evaporation, and enhancing water-holding capacity. In this study, soil infiltration and root water uptake (RWU) patterns of almond trees in amended and control soil treatments were investigated. An almond orchard was mulched with a mixture of almond hulls and shells used as surface-applied organic matter amendments. The combined use of time-lapse electrical resistivity tomography (ERT), stem water potential (SWP) and leaf water potential (LWP) measurements were used to evaluate RWU, soil infiltration patterns, and tree water status at different times during the study. The results of the ERT showed that the RWU patterns for the almond trees are distinct between the amended and the control treatments after the irrigation was applied. Compared to the control treatment, the amended treatment allowed the soil to store more water. Regardless of treatment, the majority of RWU patterns were observed in the top 0.5–1 m root zone depth. Almond trees began to recover from stress, as indicated by the SWP and LWP values, four hours after the start of irrigation. In addition, this was demonstrated by the ERT measurements, which revealed the RWU activity four hours after the irrigation had been applied. This research reveals that time-lapse ERT surveys combined with soil and tree water status data can infer patterns of RWU in almond trees grown with organic soil amendments and under controlled conditions. Furthermore, it was concluded that using almond hulls and shells as organic matter amendments can help almond growers improve infiltration, making almond production more resilient to climate change-related extremes such as droughts.

Rootstocks affect the vulnerability to embolism and pit membrane thickness in Citrus scions.

Embolism resistance of xylem tissue varies among species and is an important trait related to drought resistance, with anatomical attributes like pit membrane thickness playing an important role in avoiding embolism spread. Grafted Citrus trees are commonly grown in orchards, with the rootstock being able to affect the drought resistance of the whole plant. Here, we evaluated how rootstocks affect the vulnerability to embolism resistance of the scion using several rootstock/scion combinations. Scions of 'Tahiti' acid lime, 'Hamlin', 'Pera' and 'Valencia' oranges grafted on a 'Rangpur' lime rootstock exhibit similar vulnerability to embolism. In field-grown trees, measurements of leaf water potential did not suggest significant embolism formation during the dry season, while stomata of Citrus trees presented an isohydric response to declining water availability. When 'Valencia' orange scions were grafted on 'Rangpur' lime, 'IAC 1710' citrandarin, 'Sunki Tropical' mandarin or 'Swingle' citrumelo rootstocks, variation in intervessel pit membrane thickness of the scion was found. The 'Rangpur' lime rootstock, which is known for its drought resistance, induced thicker pit membranes in the scion, resulting in higher embolism resistance than the other rootstocks. Similarly, the rootstock 'IAC 1710' citrandarin generated increased embolism resistance of the scion, which is highly relevant for citriculture.

Foliar Pectins and Physiology of Diploid and Autotetraploid Mango Genotypes under Water Stress.

The cultivation of mango in Mediterranean-type climates is challenged by the depletion of freshwater. Polyploids are alternative genotypes with potential greater water use efficiency, but field evaluations of the anatomy and physiology of conspecific adult polyploid trees under water stress remain poorly explored. We combined field anatomical evaluations with measurements of leaf water potential (Ψl) and stomatal conductance (Gs) comparing one diploid and one autotetraploid tree per treatment with and without irrigation during dry summers (when fruits develop). Autotetraploid leaves displayed lower Ψl and Gs in both treatments, but the lack of irrigation only affected Gs. Foliar cells of the adaxial epidermis and the spongy mesophyll contained linear pectin epitopes, whereas branched pectins were localized in the abaxial epidermis, the chloroplast membrane, and the sieve tube elements of the phloem. Cell and fruit organ size was larger in autotetraploid than in diploid mango trees, but the sugar content in the fruits was similar between both cytotypes. Specific cell wall hygroscopic pectins correlate with more stable Ψl of autotetraploid leaves under soil water shortage, keeping lower Gs compared with diploids. These preliminary results point to diploids as more susceptible to water deficits than tetraploids.

Exploring the relationship between leaf water potential, defoliation, and grape berry physical properties of Merlot (Vitis vinifera L.) grapevine

The aim of this study was to examine the impact of leaf water potential and defoliation treatments on the physical properties of grape berries. The research was conducted over two consecutive years (2019-2020) using ‘Merlot’/41B graft combination grapevines grown in the Chateau Kalpak vineyards located in Tekirdağ, Şarköy. The experiment involved four distinct water stress levels (S0, S1, S2, and S3), which were determined based on leaf water potential measurements. These stress levels were subjected to different irrigation levels. Additionally, defoliation treatments were applied, including Control (C), Full Window (FW), Right Window (RW), and Left Window (LW). The results showed that the effects of water stress and defoliation treatments on berry physical properties were statistically insignificant. However, in the second year of the study, the FW treatment was observed to have led to changes in the desired direction for grapevines. This was likely due to the cumulative decrease in water reserves caused by reduced precipitation over multiple years, making the effects of FW treatment more prominent. Moreover, the study found that both current and past vegetation period conditions influence vine production year, leaf water potentials (Ψleaf), and stress levels. Finally, the data revealed that berry weight and % dry weight increased with higher stress levels.

Plants lacking OST1 show conditional stomatal closure and wildtype-like growth sensitivity at high VPD.

Climate change-associated rise in VPD (atmospheric vapor pressure deficit) results in increased plant transpiration and reduced stomatal conductance, photosynthesis, biomass, and yield. High VPD-induced stomatal closure of Arabidopsis is an active process regulated via the kinase SnRK2.6 (OPEN STOMATA 1, OST1). Here, we performed gas exchange, leaf water potential and rosette growth measurements to study, whether (1) high VPD-induced stomatal closure is detected in plants carrying loss-of-function mutations in OST1 (ost1-3) when they are grown at reduced soil water content or measured at increased air temperature; (2) ost1-3 plants expressing OST1 construct with no ABA-activation domain, but intact ABA-independent activation, show stronger stomatal VPD response compared with ost1-3 plants; and (3) rosette area and biomass of ost1-3 are more affected by growth at high VPD compared with Col-0. The stomata of well-watered ost1-3 plants were insensitive to high VPD regardless of air temperature, but in deficit-irrigated ost1-3, leaf water potential decreased the most and stomata closed at high VPD. Differences between VPD-induced stomatal closures of ost1-3 plants and ost1-3 plants expressing OST1 with no ABA-activation domain point at gradual VPD-induced ABA-independent activation of OST1. High VPD conditions led to similar reductions in rosette area and specific leaf area of well-watered Col-0 and ost1-3 plants. Rosette dry mass was unaffected by high VPD. Our results show that OST1 loss-of-function plants display conditional stomatal closure and no extra sensitivity of rosette area growth compared with Col-0 wildtype under high VPD conditions.

Best Procedures for Leaf and Stem Water Potential Measurements in Grapevine: Cultivar and Water Status Matter.

The pressure chamber is the most used tool for plant water status monitoring. However, species/cultivar and seasonal effects on protocols for reliable water potential determination have not been properly tested. In four grapevine cultivars and two times of the season (early season, Es; late season, Ls, under moderate drought), we assessed the maximum sample storage time before leaf water potential (Ψleaf) measurements and the minimum equilibration time for stem water potential (Ψstem) determination, taking 24 h leaf cover as control. In 'Pinot gris', Ψleaf already decreased after 1 h leaf storage in both campaigns, dropping by 0.4/0.5 MPa after 3 h, while in 'Refosk', it decreased by 0.1 MPa after 1 and 2 h in Es and Ls, respectively. In 'Merlot' and 'Merlot Kanthus', even 3 h storage did not affect Ψleaf. In Es, the minimum Ψstem equilibration was 1 h for 'Refošk' and 10 min for 'Pinot gris' and 'Merlot'. In Ls, 'Merlot Kanthus' required more than 2 h equilibration, while 1 h to 10 min was sufficient for the other cultivars. The observed cultivar and seasonal differences indicate that the proposed tests should be routinely performed prior to experiments to define ad hoc procedures for water status determination.

Daytime stomatal regulation in mature temperate trees prioritizes stem rehydration at night.

Trees remain sufficiently hydrated during drought by closing stomata and reducing canopy conductance (Gc ) in response to variations in atmospheric water demand and soil water availability. Thresholds that control the reduction of Gc are proposed to optimize hydraulic safety against carbon assimilation efficiency. However, the link between Gc and the ability of stem tissues to rehydrate at night remains unclear. We investigated whether species-specific Gc responses aim to prevent branch embolisms, or enable night-time stem rehydration, which is critical for turgor-dependent growth. For this, we used a unique combination of concurrent dendrometer, sap flow and leaf water potential measurements and collected branch-vulnerability curves of six common European tree species. Species-specific Gc reduction was weakly related to the water potentials at which 50% of branch xylem conductivity is lost (P50 ). Instead, we found a stronger relationship with stem rehydration. Species with a stronger Gc control were less effective at refilling stem-water storage as the soil dries, which appeared related to their xylem architecture. Our findings highlight the importance of stem rehydration for water-use regulation in mature trees, which likely relates to the maintenance of adequate stem turgor. We thus conclude that stem rehydration must complement the widely accepted safety-efficiency stomatal control paradigm.

Shrub Invasion Overrides the Effect of Imposed Drought on the Photosynthetic Capacity and Physiological Responses of Mediterranean Cork Oak Trees.

Mediterranean ecosystems face threats from both climate change and shrub invasion. As shrub cover increases, competition for water intensifies, exacerbating the negative effects of drought on ecosystem functioning. However, research into the combined effects of drought and shrub invasion on tree carbon assimilation has been limited. We used a Mediterranean cork oak (Quercus suber) woodland to investigate the effects of drought and shrub invasion by gum rockrose (Cistus ladanifer) on cork oak carbon assimilation and photosynthetic capacity. We established a factorial experiment of imposed drought (ambient and rain exclusion) and shrub invasion (invaded and non-invaded) and measured leaf water potential, stomatal conductance and photosynthesis as well as photosynthetic capacity in cork oak and gum rockrose over one year. We observed distinct detrimental effects of gum rockrose shrub invasion on the physiological responses of cork oak trees throughout the study period. Despite the imposed drought, the impact of shrub invasion was more pronounced, resulting in significant photosynthetic capacity reduction of 57% during summer. Stomatal and non-stomatal limitations were observed under moderate drought in both species. Our findings provide significant knowledge on the impact of gum rockrose invasion on the functioning of cork oak and can be used to improve the representation of photosynthesis in terrestrial biosphere models.

Accurate estimates of land surface energy fluxes and irrigation requirements from UAV-based thermal and multispectral sensors

The two-source energy balance model estimates canopy transpiration (Tr) and soil evaporation (E) traditionally from satellite partitions of remotely sensed land surface temperature (LST) and the Priestley-Taylor equation (TSEB-PT) at seasonal time with limited accuracy. The high spatial–temporal resolution spectral data collected by unmanned aerial vehicles (UAVs) provide valuable opportunity to estimate Tr and E precisely, improve the understanding of the seasonal and the diurnal cycle of evapotranspiration (ET), and timely detect agricultural drought. The UAV data vary in spatial resolution and the uncertainty imposed on the TSEB-PT outcome has thus far not being considered. To address these challenges and prospects, a new energy flux modelling framework based on TSEB-PT for high spatial resolution thermal and multispectral UAV data is proposed in this paper. Diurnal variations of LST in agricultural fields were recorded with a thermal infrared camera installed on an UAV during drought in 2018 and 2019. Observing potato as a test crop, LST, plant biophysical parameters derived from corresponding UAV multispectral data, and meteorological forcing variables were employed as input variables to TSEB-PT. All analyses were conducted at different pixelation of the UAV data to quantify the effect of spatial resolution on the performance. The 1 m spatial resolution produced the highest correlation between Tr modelled by TSEB-PT and measured by sap flow sensors (R2 = 0.80), which was comparable to the 0.06, 0.1, 0.5 and 2 m pixel sizes (R2 = 0.76–0.78) and markedly higher than the lower resolutions of 2 to 24 m (R2 = 0.30–0.72). Modelled Tr was highly and significantly correlated with measured leaf water potential (R2 = 0.81) and stomatal conductance (R2 = 0.74). The computed irrigation requirements (IRs) reflected the field irrigation treatments, ET and conventional irrigation practices in the area with high accuracy. It was also found that using a net primary production model with explicit representation of temperature influences made it possible to distinguish effects of drought vis-a-vis heat stress on crop productivity and water use efficiency. The results showed excellent model performance for retrieving Tr and ET dynamics under drought stress and proved that the proposed remote sensing based TSEB-PT framework at UAV scale is a promising tool for the investigation of plant drought stress and water demand; this is particularly relevant for local and regional irrigations scheduling.

The ecosystem wilting point defines drought response and recovery of a Quercus-Carya forest.

Soil and atmospheric droughts increasingly threaten plant survival and productivity around the world. Yet, conceptual gaps constrain our ability to predict ecosystem-scale drought impacts under climate change. Here, we introduce the ecosystem wilting point (ΨEWP ), a property that integrates the drought response of an ecosystem's plant community across the soil-plant-atmosphere continuum. Specifically, ΨEWP defines a threshold below which the capacity of the root system to extract soil water and the ability of the leaves to maintain stomatal function are strongly diminished. We combined ecosystem flux and leaf water potential measurements to derive the ΨEWP of a Quercus-Carya forest from an "ecosystem pressure-volume (PV) curve," which is analogous to the tissue-level technique. When community predawn leaf water potential (Ψpd ) was above ΨEWP (=-2.0MPa), the forest was highly responsive to environmental dynamics. When Ψpd fell below ΨEWP , the forest became insensitive to environmental variation and was a net source of carbon dioxide for nearly 2 months. Thus, ΨEWP is a threshold defining marked shifts in ecosystem functional state. Though there was rainfall-induced recovery of ecosystem gas exchange following soaking rains, a legacy of structural and physiological damage inhibited canopy photosynthetic capacity. Although over 16 growing seasons, only 10% of Ψpd observations fell below ΨEWP , the forest is commonly only 2-4 weeks of intense drought away from reaching ΨEWP , and thus highly reliant on frequent rainfall to replenish the soil water supply. We propose, based on a bottom-up analysis of root density profiles and soil moisture characteristic curves, that soil water acquisition capacity is the major determinant of ΨEWP , and species in an ecosystem require compatible leaf-level traits such as turgor loss point so that leaf wilting is coordinated with the inability to extract further water from the soil.

Plant hydraulic resistance controls transpiration of soybean in rotational paddy fields under humid climates

Efficient irrigation and drainage management are highly required for increasing crop productivity in paddy rice and upland crop rotation. However, conventional management does not sufficiently consider the water status of the plants and soil in the root zone. The aim of this study was to evaluate whether the hydraulic resistance of soil (Rs) or plant (Rp) principally controlled transpiration in rotational paddy fields (RPFs) located in humid regions. To achieve this, we conducted field measurements of soil water conditions, evapotranspiration rate, and leaf water potential in RPFs cropped with soybean after the flowering stage and calculated Rs and Rp based on the theory of root water uptake. After the flowering stage, the soil was sometimes saturated owing to intermittent precipitation, and thus Rs was maintained at a low value. By contrast, Rp gradually increased over time and ranged between 5.1 × 108 and 10.3 × 108 s, which was one to three orders of magnitude higher than Rs. The ratio of the actual to the potential transpiration rate decreased throughout the investigation period and hardly reached 1.0, even though the soil was sufficiently wet. These results indicate that Rp, which probably increases with continuous soil saturation, controls crop transpiration in RPFs under humid climates. Our results suggest that drainage systems are essential in RPFs to avoid a change in Rp and improve crop productivity.

Assessment of drought stress responsive genes expression profiles and proline accumulation in a diverse set of grapevine rootstocks

In order to study plant responses to drought stress, it is important to have markers for such responses. This issue has been a challenge for decades and numerous traits have been used for this purpose. In the current study, the influence of drought stress on proline biosynthesis, leaf water potential, and also gene expression levels of 10 genes probably involved in drought stress response regulation pathways were investigated. Also, all experiments were performed in both root and leaf tissues of studied rootstocks to investigate the response of plant tissues which is more suitable for the assessment of possible markers. As a result of the study, the highest percentage of proline increase in leaf and root tissues was mostly observed in susceptible rootstocks. The leaf water potential (-MPa) measurements showed the most decrease in leaf water potential values in highly tolerant rootstocks, so the maximum differences between control and drought stress treated plants were determined in SO4, 140Ru, 1045, and 44-53M rootstocks, respectively. Gene expression levels of HAT5, RAP2-4, WRKY23, GRP, GDH, PRP2, GAS2, PD2, STPK, and genes were described in 18 different grapevine rootstocks and it was concluded that the root samples indicating more significant gene expression changes, could be more informative than leaf samples.

Ethylene constrains stomatal reopening in Fraxinus chinensis post moderate drought.

Clarifying the mechanisms underlying the recovery of gas exchange following drought is the key to providing insights into plant drought adaptation and habitat distribution. However, the mechanisms are still largely unknown. Targeting processes known to inhibit gas exchange during drought recovery, we measured leaf water potential, the leaf hydraulic conductance, stomatal reopening, abscisic acid (ABA) and the ethylene emission rate (EER) following moderate drought stress in seedlings of the globally pervasive woody tree Fraxinus chinensis. We found strong evidence that the slow stomatal reopening after rehydration is regulated by a slow decrease in EER, rather than changes in leaf hydraulics or foliar ABA levels. This was supported by evidence of rapid gas exchange recovery in plants after treatment with the ethylene antagonist 1-methylcyclopropene. These findings provide evidence to rigorously support ethylene as a key factor constraining stomatal reopening from moderate drought directly, thereby potentially opening new windows for understanding species drought adaptation.

Variety-specific response of bulk stomatal conductance of grapevine canopies to changes in net radiation, atmospheric demand, and drought stress.

In wine growing regions around the world, climate change has the potential to affect vine transpiration and overall vineyard water use due to related changes in daily atmospheric conditions and soil water deficits. Grapevines control their transpiration in response to such changes by regulating conductance of water through the soil-plant-atmosphere continuum. The response of bulk stomatal conductance, the vine canopy equivalent of stomatal conductance, to such changes were studied on Cabernet-Sauvignon, Merlot, Tempranillo, Ugni blanc, and Semillon vines in a non-irrigated vineyard in Bordeaux France. Whole-vine sap flow, temperature and humidity in the vine canopy, and net radiation absorbed by the vine canopy were measured on 15-minute intervals from early July through mid-September 2020, together with periodic measurements of leaf area, canopy porosity, and predawn leaf water potential. From these data, bulk stomatal conductance was calculated on 15-minute intervals, and multiple linear regression analysis was performed to identify key variables and their relative effect on conductance. For the regression analysis, attention was focused on addressing non-linearity and collinearity in the explanatory variables and developing a model that was readily interpretable.Variability of vapour pressure deficit in the vine canopy over the day and predawn water potential over the season explained much of the variability in bulk stomatal conductance overall, with relative differences between varieties appearing to be driven in large part by differences in conductance response to predawn water potential between the varieties. Transpiration simulations based on the regression equations found similar differences between varieties in terms of daily and seasonal transpiration. These simulations also compared well with those from an accepted vineyard water balance model, although there appeared to be differences between the two approaches in the rate at which conductance, and hence transpiration is reduced as a function of decreasing soil water content (i.e., increasing water deficit stress). By better characterizing the response of bulk stomatal conductance, the dynamics of vine transpiration can be better parameterized in vineyard water use modeling of current and future climate scenarios.

Using δ<sup>13</sup>C and hydroscapes for discriminating cultivar specific drought responses

Measurement of carbon isotope discrimination in berry juice at maturity (δ13C) provides an integrated assessment of vine water status and water use efficiency (WUE) during the period of berry ripening, and when collected over multiple seasons, can provide an indication of drought stress responses. Berry juice δ13C measurements were carried out on 48 different varieties planted in a common garden experiment in Bordeaux, France from 2014 through 2020 and found important differences across this large panel of varieties. Cluster analysis showed that δ13C values are likely affected by the differing phenology of each variety, resulting in berry ripening of different varieties taking place under different conditions of soil water availability within the same year. Accounting for these phenological differences, the cluster analysis created a classification of varieties that corresponds well to our current empirical understanding of their relative drought tolerance. In addition, using measurements of predawn and midday leaf water potential measurements collected over four seasons on a subset of six varieties, a hydroscape approach was used to develop a list of metrics indicative of the sensitivity of stomatal regulation to water stress (i.e., an/isohydric behaviour). Key hydroscape metrics were also found to be well correlated with some δ13C metrics. A variety’s water potential regulation as characterized by a minimum critical leaf water potential as determined from hydroscapes was strongly correlated to δ13C values under well-watered conditions, suggesting that the latter may be a useful indicator of drought stress response.

Foliar Water Uptake Capacity in Six Mangrove Species

Foliar water uptake (FWU) is a mechanism that enables plants to acquire water from the atmosphere through their leaves. As mangroves live in a saline sediment water environment, the mechanism of FWU might be of vital importance to acquire freshwater and grow. The goal of this study was to assess the FWU capacity of six different mangrove species belonging to four genera using a series of submersion experiments in which the leaf mass increase was measured and expressed per unit leaf area. The foliar water uptake capacity differed between species with the highest and lowest average water uptake in Avicennia marina (Forssk.) Vierh. (1.52 ± 0.48 mg H2O cm−2) and Bruguiera gymnorhiza (L.) Lam. (0.13 ± 0.06 mg H2O cm−2), respectively. Salt-excreting species showed a higher FWU capacity than non-excreting species. Moreover, A. marina, a salt-excreting species, showed a distinct leaf anatomical trait, i.e., trichomes, which were not observed in the other species and might be involved in the water absorption process. The storage of leaves in moist Ziplock bags prior to measurement caused leaf water uptake to already occur during transport to the field station, which proportionately increased the leaf water potential (A. marina: −0.31 ± 0.13 MPa and B. gymnorhiza: −2.70 ± 0.27 MPa). This increase should be considered when performing best practice leaf water potential measurements but did not affect the quantification of FWU capacity because of the water potential gradient between a leaf and the surrounding water during submersion. Our results highlight the differences that exist in FWU capacity between species residing in the same area and growing under the same environmental conditions. This comparative study therefore enhances our understanding of mangrove species’ functioning.

Leaf water potential measurements using the pressure chamber: Synthetic testing of assumptions towards best practices for precision and accuracy.

Leaf water potential (ψ leaf), typically measured using the pressure chamber, is the most important metric of plant water status, providing high theoretical value and information content for multiple applications in quantifying critical physiological processes including drought responses. Pressure chamber measurements of ψ leaf (ψ leafPC) are most typical, yet, the practical complexity of the technique and of the underlying theory has led to ambiguous understanding of the conditions to optimize measurements. Consequently, specific techniques and precautions diversified across the global research community, raising questions of reliability and repeatability. Here, we surveyed specific methods of ψ leafPC from multiple laboratories, and synthesized experiments testing common assumptions and practices in ψ leafPC for diverse species: (i) the need for equilibration of previously transpiring leaves; (ii) leaf storage before measurement; (iii) the equilibration of ψ leaf for leaves on bagged branches of a range of dehydration; (iv) the equilibration of ψ leaf across the lamina for bagged leaves, and the accuracy of measuring leaves with artificially ‘elongated petioles’; (v) the need in ψ leaf measurements for bagging leaves and high humidity within the chamber; (vi) the need to avoid liquid water on leaf surfaces; (vii) the use of ‘pulse’ pressurization versus gradual pressurization; and (viii) variation among experimenters in ψ leafPC determination. Based on our findings we provide a best practice protocol to maximise accuracy, and provide recommendations for ongoing species‐specific tests of important assumptions in future studies.
