Autonomous and extrinsic regulation of thymopoiesis inhuman immune system (HIS) mice

Human Immune System (HIS) mice represent a novel biotechnology platform to dissect human haematopoiesis and immune responses. However, the limited human T-cell development that is observed in HIS mice restricts its utility for these applications. Here, we address whether reduced thymopoiesis in HIS mice reflects an autonomous defect in T-cell precursors and/or a defect in the murine thymic niche. Human thymocyte precursors seed the mouse thymus and their reciprocal interactions with murine thymic epithelial cells (TECs) led to both T-cell and TEC maturation. The human thymocyte subsets observed in HIS mice demonstrated survival, proliferative and phenotypic characteristics of their normal human counterparts, suggesting that the intrinsic developmental program of human thymocytes unfolds normally in this xenograft setting. We observed that exogenous administration of human IL-15/IL-15Rα agonistic complexes induced the survival, proliferation and absolute numbers of immature human thymocyte subsets, without any obvious effect on cell-surface phenotype or TCR Vβ usage amongst the newly selected mature single-positive (SP) thymocytes. Finally, when IL-15 was administered early after stem cell transplantation, we noted accelerated thymopoiesis resulting in the more rapid appearance of peripheral naïve T cells. Our results highlight the functional capacity of murine thymic stroma cells in promoting human thymopoiesis in HIS mice but suggest that the "cross-talk" between murine thymic stroma and human haematopoietic precursors may be suboptimal. As IL-15 immunotherapy promotes early thymopoiesis, this novel approach could be used to reduce the period of T-cell immunodeficiency in the post-transplant clinical setting.

Thymic epithelial cells: the multi-tasking framework of the T cell “cradle”

The thymus provides the anatomical "cradle" that fosters developing thymocytes. Thymic epithelial cells (TECs) are specialized cellular components that may be viewed as a multifunctional "frame" to nurture distinct stages of thymopoiesis. A symbiotic relationship between TECs and thymocytes exists because reciprocal interactions are required to achieve complete maturation of both cell types. Here, we propose that crucial instructive signals delivered by developing thymocytes negatively regulate functional attributes of immature TECs (including the expression of Delta-like 4 (DLL4) and interleukin-7 (IL-7)) that are required during early stages of thymopoiesis, while promoting the diversification of more mature TEC subsets. Thus, the division of labour among TECs may be coordinated directly by local cellular feedback mechanisms operating within distinct thymic niches.

Early defects in human T-cell development severely affect distribution and maturation of thymic stromal cells: possible implications for the pathophysiology of Omenn syndrome

AbstractThymocytes and thymic epithelial cell (TEC) cross-talk is crucial to preserve thymic architecture and function, including maturation of TECs and dendritic cells, and induction of mechanisms of central tolerance. We have analyzed thymic maturation and organization in 9 infants with various genetic defects leading to complete or partial block in T-cell development. Profound abnormalities of TEC differentiation (with lack of AIRE expression) and severe reduction of thymic dendritic cells were identified in patients with T-negative severe combined immunodeficiency, reticular dysgenesis, and Omenn syndrome. The latter also showed virtual absence of thymic Foxp3+ T cells. In contrast, an IL2RG-R222C hypomorphic mutation permissive for T-cell development allowed for TEC maturation, AIRE expression, and Foxp3+ T cells. Our data provide evidence that severe defects of thymopoiesis impinge on TEC homeostasis and may affect deletional and nondeletional mechanisms of central tolerance, thus favoring immune dysreactive manifestations, as in Omenn syndrome.

A roadmap for thymic epithelial cell development

Thymic epithelial cells (TEC) are essential components of the thymus that guide and control the development and TCR repertoire selection of T cells. Previously, TEC have been considered as postmitotic cells that, once generated during ontogeny, were maintained in their mature state. Recently, it has become clear that TEC can be generated from common or committed medullary and cortical TEC progenitor cells in ontogeny, that stages of immature and mature TEC are phenotypically separable, and that TEC undergo a rapid turnover in a matter of a few weeks. All of these findings strongly suggest that in the adult thymus mature TEC are constantly regenerated from a pool of stem or progenitor cells, a view that renders the thymus structure potentially much more dynamic than previously thought. However, the identity of "thymus stem cells" is elusive, and developmental stages of TEC development are only beginning to be elucidated.

An Epithelial Progenitor Pool Regulates Thymus Growth

Thymic epithelium provides an essential cellular substrate for T cell development and selection. Gradual age-associated thymic atrophy leads to a reduction in functional thymic tissue and a decline in de novo T cell generation. Development of strategies tailored toward regeneration of thymic tissue provides an important possibility to improve immune function in elderly individuals and increase the capacity for immune recovery in patients having undergone bone marrow transfer following immunoablative therapies. In this study we show that restriction of the size of the functional thymic epithelial progenitor pool affects the number of mature thymic epithelial cells. Using an embryo fusion chimera-based approach, we demonstrate a reduction in the total number of both embryonic and adult thymic epithelium, which relates to the initial size of the progenitor cell pool. The inability of thymic epithelial progenitor cells to undergo sufficient compensatory proliferation to rescue the deficit in progenitor numbers suggests that in addition to extrinsic regulation of thymus growth by provision of growth factors, intrinsic factors such as a proliferative restriction of thymic epithelial progenitors and availability of progenitor cell niches may limit thymic epithelial recovery. Collectively, our data demonstrate an important level of regulation of thymic growth and recovery at the thymic epithelial progenitor level, providing an important consideration for developing methods targeted toward inducing thymic regeneration.

Identification and characterization of Xenopus laevis homologs of mammalian TRAF6 and its binding protein TIFA

Tumor necrosis factor receptor (TNFR)-associated factor 6 (TRAF6) transduces signals from members of the TNFR superfamily and the Toll/IL-1R family, leading to activation of transcription factors such as NFκB and AP-1. Genetic disruption of the TRAF6 gene in mice results in various developmental abnormalities during embryogenesis, including osteopetrosis, failure of neural tube closure, defective formation of skin appendices, absence of lymph nodes, and absence of mature thymic epithelial cells. To clarify the effect of TRAF6 in development, we previously identified a TRAF-interacting protein with a forkhead-associated domain (TIFA), which binds and activates TRAF6 upon extracellular stimulation. To understand the physiological roles of TRAF6 and TIFA in early development, we studied these genes in Xenopus laevis. Here, we describe identification of X. laevis homologs of mammalian TRAF6 (XTRAF6) and TIFA (XTIFA). As was the case for the mammalian homologs, overexpression of XTRAF6 or XTIFA activated NFκB, whereas XTIFA carrying a mutation that abolishes XTRAF6 binding failed to activate NFκB, suggesting that XTIFA activates NFκB by binding to XTRAF6. XTIFA and XTRAF6 mRNAs were expressed at similar levels in zygotes from the neurula stage and then increased. Whole-mount in situ hybridization revealed that XTRAF6 mRNA was expressed in the head region and neural tube during the neurula stage, and the expression expanded to the pharyngeal apparatus during the tailbud stage. This localization is consistent with the defective neural tube closure and abnormal thymus organogenesis observed in TRAF6-deficient mice. Our results suggest possible cooperation between XTRAF6 and XTIFA during embryogenesis.

Mutations affecting thymus organogenesis in Medaka, Oryzias latipes

The thymus is an organ for T lymphocyte maturation and is indispensable for the establishment of a highly developed immune system in vertebrates. In order to genetically dissect thymus organogenesis, we carried out a large-scale mutagenesis screening for Medaka mutations affecting recombination activating gene 1 ( rag1) expression in the developing thymus. We identified 24 mutations, defining at least 13 genes, which led to a marked reduction of rag1 expression in the thymus. As thymus development depends on pharyngeal arches, we classified those mutations into three classes according to the defects in the pharyngeal arches. Class 1 mutants had no or slight morphological abnormalities in the pharyngeal arches, implying that the mutations may include defects in such thymus-specific events as lymphocyte development and thymic epithelial cell maturation. Class 2 mutants had abnormally shaped pharyngeal arches. Class 3 mutants showed severely attenuated pharyngeal arch development. In Class 2 and Class 3 mutants, the defects in thymus development may be due to abnormal pharyngeal arch development. Those mutations are expected to be useful for identifying the molecular mechanisms underlying thymus organogenesis.

Characterization of a monoclonal antibody, RTE-21, that binds to keratohyalin granule-associated proteins in epithelial cells of human skin and thymus

The role of skin and thymic epithelium in the promotion of T-cell activation and maturation is currently an area of great interest. In thymus. epithelium is located in the cortex, medulla, and in medullary epithelial swirls called Hassall's bodies. During studies of antigens shared by skin and thymic epithelial cells, we produced a murine monoclonal antibody (RTE-21) raised against the rat thymic epithelial cells line, IT26R21, that identified an antigen present in terminally differentiated epithelium of normal human skin and thymus. In indirect immunofluorescence studies, antibody RTE-21 identified cytoplasmic granules located in the stratum granulosum of normal human skin. Hassall's bodies of human thymus, and in a subset of cells of the IT26R21 rat thymic epithelial cell line. Moreover, the granular reactivity pattern of antibody RTE-21 in the stratum granulosum could be extracted by 1 M potassium phosphate (1 M KPO 4). A 1 M KPO 4 extract of epidermis containing keratohyalin granule proteins was dialyzed against distilled water, solubilized in reduced sample buffer and subjected to polyacrylamide gel electrophoresis and Western immunoblot analysis. In this assay, antibody RTE-21 recognized proteins of 70, 36, 34, and 30 kDa. Using antibody RTE-21 and indirect immunofluorescence, we demonstrated that keratohyalin granules in human thumus were localized exclusively to Hassall's bodies. These data support the notion that human thymic Hassall's bodies result from terminal differentiation of medullary thymic epithelium. Thus, antibody RTE-21 should be an important probe for the study of skin and thymic epithelial cell maturation in vitro and in vivo.

IN VITRO AND IN VIVO DEFINITION OF A NEW VARIANT OF SEVERE COMBINED IMMUNODEFICIENCY DISEASE (SCID)

SCID Is a heterogeneous disease caused by at least two known primary defects; specific enzyme deficiency or a defect in thymic epithelial cell maturation. A 12 nonth old male presented with a 7 month history of recurrent fevers, pyogenic infections, and documented Pneumocystis carinii pneumonia. Examination revealed hyperplastic lymphoid tissue. Laboratory data included normal red cell enzymes, normal lymphocyte numbers, profound hypogammaglobulinemia (except for an IgM of 22-47 mg%), no antibody response, but normal numbers of B-cells. 9-15% of his peripheral blood lymphocytes formed E-rosettes; both his PHA and MLR responses were markedly reduced (10% of normal). He did not respond to several skin test antigens or DNCB, and he failed to reject an allogeneic skin graft. Lymph node biopsy demonstrated normal follicle formation, poor germinal center formation, and depletion of cells in the paracortex. Thymus biopsy revealed a large thymus with predominantly E-rosetting cells, PAS-positive epithelial cells, but no Hassall's bodies. Unlike other SCID patients, bone marrow precursor cells were responsive normally to human thymus conditioned medium, suggesting the initial stage(s) of T-cell maturation were achieved in this patient. On the basis of the thymus biopsy and bone marrow precursor cell response, we believe this case of SCID represents a failure of terminal differentiation of intrathymic T-lymphocytes.
