Male germ cell transplantation: an experimental approach with a clinical perspective

Germ cell transplantation was developed in strains of mice. The infusion of germ cell preparations into the seminiferous tubules of infertile hosts led to repopulation of the testis with donor germ cells and restored fertility. Meanwhile, this technique has become a powerful tool to study the expansion of the testicular stem cell population and the kinetics of spermatogonial proliferation and re-induction of spermatogenesis. Further approaches, such as the transfer of rat/hamster/rabbit germ cells into mouse testes or cryopreservation or culture of spermatogonia, have widened the spectrum of applications associated with germ cell transplantation. Since the devastating effects of chemo- or radiotherapy on spermatogonia are the cause of infertility in oncological patients, germ cell transplantation might become an alternative approach for gonadal protection in prepubertal and postpubertal male tumour patients. This review focusses on recent developments with respect to germ cell transplantation and highlights the problems and perspectives of future clinical applications.

Male germ cell transplantation: present achievements and future prospects.

Germ cells are unique, since their surviving descendants can undergo meiosis and differentiate into gametes, which transmit genetic material from one generation to another. We now know that male germ cells, whether they be primordial germ cells in gonadal ridges, gonocytes, or stem spermatogonia, are transplantable. The donor cells can be transferred by direct microinjection into the seminiferous tubules, rete testis or efferent ducts, depending on the recipient species. Following transplantation, the donor cells undergo spermatogenesis in the host's seminiferous tubules in rats and mice, and have even sired offspring in mice. Interspecific germ cell transfer is possible if the recipient's immune system is defective; nude or SCID mice can even produce rat spermatozoa. However, the major obstacle restricting widespread use of this new technology is its extremely low success rate. This article discusses some ideas for improving the success rate of the transfer technique, and considers several potential applications.

Male germ cell transplantation in rats: apparent synchronization of spermatogenesis between host and donor seminiferous epithelia

Primordial germ cells (PGC) and gonocytes from male Sprague-Dawley rat fetuses and neonates were transplanted via the rete testis into the lumen of the seminiferous tubules of recipient adult Long Evans rats. The donor germ cells apparently differentiated into mini-tubules or irregular segments of seminiferous epithelium within the lumen of the host seminiferous tubules, and exhibited qualitatively normal spermatogenesis in 10 out of 16 recipients. The stage of spermatogenesis of the intraluminal epithelium was synchronized closely with that of the adjacent seminiferous tubule epithelium, suggesting that the spermatogenic cycle is regulated locally by the intraluminal microenvironment. Male germ cell transplantation provides an interesting new tool for investigating the control of spermatogenesis.

Spermatogenesis following male germ-cell transplantation.

In the adult male, a population of diploid stem-cell spermatogonia continuously undergoes self-renewal and produces progeny cells, which initiate the complex process of cellular differentiation that results in mature spermatozoa. We report here that stem cells isolated from testes of donor male mice will repopulate sterile testes when injected into seminiferous tubules. Donor cell spermatogenesis in recipient testes showed normal morpholigical characteristics and produced mature spermatozoa. This methodology, besides opening new avenues of basic research into spermatogenesis and stem-cell self-renewal, may prove useful as a tool for biomedical science and biotechnology.

Nuclear transplantation of male primordial germ cells in the mouse

We examined the developmental ability of enucleated eggs receiving embryonic nuclei and male primordial germ cells (PGCs) in the mouse. Reconstituted eggs developed into the blastocyst stage only when an earlier 2-cell nucleus was transplanted (36%) but very rarely if the donor nucleus was derived from a later 2-cell, 8-cell, or inner cell mass of a blastocyst (0-3%). 54-100%, 11-67%, 6-43% and 6-20% of enucleated eggs receiving male PGCs developed to 2-cell, 4-cell, 8-cell and blastocyst stage, respectively, in culture. The overall success rate when taking into account the total number of attempts at introducing germ cells was actually 0-6%. Live fetuses were not obtained after transfer of reconstituted eggs to recipients, although implantation sites were observed. The developmental ability of reconstituted eggs in relation to embryonic genome activation and genomic imprinting is discussed.
