In situ magnesium-doped calcium phosphate microtube-reinforced magnesium-doped hydroxyapatite composites with tunable mechanical properties and biodegradability

Hydroxyapatite, commonly employed in bone defect repair, demonstrates limitations in load-bearing applications due to its poor mechanical property and biodegradability. This research introduces an innovative bone substitute material, namely, in situ magnesium-doped calcium phosphate microtube (Mg-CaPMT)-reinforced magnesium-doped hydroxyapatite composites (Mg-CaPMT/Mg-HA) inspired by microtube material with outstanding mechanical property and capillary tube in a plant. The in situ Mg-CaPMT, with a ~7 μm diameter and 400-500nm wall thickness, was formed by pore-forming carbon fiber (CF) and the coating on it. These microtubes are uniformly distributed within Mg-HA matrix. The composites can meet load-bearing bone requirements, demonstrating compressive strength of 167.0±20.7MPa and fracture toughness of 2.04±0.13MPa·m1/2. Notably, it exhibits exceptional permeability and biodegradability, attributed to capillary action by the Mg-CaPMT structure. The adjustable mechanical and biological properties of Mg-CaPMT/Mg-HA, based on microtube dimensions, content, and composition, offer a versatile solution for repairing various bone defects in load-bearing areas.

Enhancing Bone Implants: Magnesium-Doped Hydroxyapatite for Stronger, Bioactive, and Biocompatible Applications.

Hydroxyapatite (HAp) with the chemical formula Ca10(PO4)6(OH)2 is an inorganic material that exhibits morphology and composition similar to those of human bone tissues, making it highly desirable for bone regeneration applications. As one of the most biocompatible materials currently in use, HAp has undergone numerous attempts to enhance its mechanical strength. This research focuses on investigating the influence of magnesium (Mg) incorporation on the structural and mechanical properties of synthesized magnesium-doped hydroxyapatite (MgHAp) samples. Apart from its biocompatibility, Mg possesses a density and elasticity comparable to those of human bone. Therefore, incorporating Mg into HAp can be pivotal for improving bone formation. Previous studies have not extensively explored the structural changes induced by Mg substitution in HAp, which motivated us to revisit this issue. Hydrothermal synthesis technique was used to synthesize MgHAp samples with varying molar concentrations (x = 0, 0.5, 1.0, and 1.5). Theoretical simulation of HAp and MgHAp for obtaining 3D structures has been done, and theoretical X-ray diffraction (XRD) data have been compared with the experimental XRD data. Rietveld analysis revealed the alteration and deviation of lattice parameters with an increase in the Mg content, which ultimately affect the structure as well the mechanical properties of prepared samples. The findings revealed an increase in compressive stress and fracture toughness as the Mg concentration in the composition increased. Furthermore, using a finite-element analysis technique and modeling of the mechanical testing data, the von Mises stress distribution and Young's modulus values were calculated, demonstrating the similarity of the prepared samples to human cortical bone. Biocompatibility assessments using NIH-3T3 fibroblast cells confirmed the biocompatible and bioactive nature of the synthesized samples. MgHAp exhibits great potential for biomedical applications in the dental, orthopedic, and tissue engineering research fields.

Magnesium-enhanced porous hydroxyapatite ceramics: Morphometric parameters, physical properties and bioactivity

The application of porous bioactive ceramics as bone substitutes requires their effective osseointegration, which relies on specific physical and biological properties. In this study, we developed porous magnesium-doped hydroxyapatite (MgHA) ceramics with four different magnesium concentrations (0.25, 2, 5 and 10 mol%). The ceramics were prepared through the polymeric replication sponge method and subjected to physical and biological characterizations. Upon sintering, the porous 10 mol% MgHA ceramics exhibited increased densification, which resulted in the highest compressive strength of 2.17 MPa at the lowest porosity of 31 %.The porous samples were analysed via microcomputed tomography. The scaffolds with 0.25 and 10 mol% Mg doping concentrations were selected to elucidate the influence of Mg on morphometric parameters and biological properties at two distinct levels. The high level of Mg doping led to considerable improvements in relative bone volume, connectivity density, trabecular number and trabecular thickness. Cell attachment and proliferation tests using Vero cell lines were conducted on both samples to investigate the correlations between their morphometric parameters and bioactivities. Substantially more cells attached to and proliferated on the surface of the 10 mol% MgHA ceramics compared with those on the ceramics containing 0.25 mol% MgHA. In summary, our study underscores the effectiveness of Mg incorporation in improving the physical and biological properties of porous HA ceramics, which makes them promising candidates for bone substitute applications.

New Physico-Chemical Analysis of Magnesium-Doped Hydroxyapatite in Dextran Matrix Nanocomposites.

The new magnesium-doped hydroxyapatite in dextran matrix (10MgHApD) nanocomposites were synthesized using coprecipitation technique. A spherical morphology was observed by scanning electron microscopy (SEM). The X-ray diffraction (XRD) characterization results show hydroxyapatite hexagonal phase formation. The element map scanning during the EDS analysis revealed homogenous distribution of constituent elements of calcium, phosphor, oxygen and magnesium. The presence of dextran in the sample was revealed by Fourier transform infrared (FTIR) spectroscopy. The antimicrobial activity of the 10MgHAPD nanocomposites was assessed by in vitro assays using Staphylococcus aureus ATCC 25923, Pseudomonas aeruginosa ATCC 27853, Streptococcus mutans ATCC 25175, Porphyromonas gingivalis ATCC 33277 and Candida albicans ATCC 10231 microbial strains. The results of the antimicrobial assays highlighted that the 10MgHApD nanocomposites presented excellent antimicrobial activity against all the tested microorganisms and for all the tested time intervals. Furthermore, the biocompatibility assays determined that the 10MgHApD nanocomposites did not exhibit any toxicity towards Human gingival fibroblast (HGF-1) cells.

Complex Evaluation of Nanocomposite-Based Hydroxyapatite for Biomedical Applications.

A magnesium-doped hydroxyapatite in chitosan matrix (MgHApC) sample was developed as a potential platform for numerous applications in the pharmaceutical, medical, and food industries. Magnesium-doped hydroxyapatite suspensions in the chitosan matrix were obtained by the coprecipitation technique. The surface shape and morphological features were determined by scanning electron microscopy (SEM). The hydrodynamic diameter of the suspended particles was determined by Dynamic light scattering (DLS) measurements. The stability of MgHApC suspensions was evaluated by ultrasonic measurements. The hydrodynamic diameter of the MgHApC particles in suspension was 29.5 nm. The diameter of MgHApC particles calculated from SEM was 12.5 ± 2 nm. Following the SEM observations, it was seen that the MgHApC particles have a spherical shape. The Fourier-transform infrared spectroscopy (FTIR) studies conducted on MgHApC proved the presence of chitosan and hydroxyapatite in the studied specimens. In vitro antimicrobial assays were performed on Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923, Pseudomonas aeruginosa ATCC 27853, and Candida albicans ATCC 10231 microbial strains. The antimicrobial experiments showed that MgHApC exhibited very good antimicrobial properties against all the tested microorganisms. More than that, the results of the in vitro studies revealed that the antimicrobial properties of the samples depend on the incubation time. The evaluation of the sample's cytotoxicity was performed using the human colon cancer (HCT-8) cell line. Our results suggested the great potential of MgHApC to be used in future applications in the field of biomedical applications (e.g., dentistry, orthopedics, etc.).

Morphological and fractal features of cancer cells anchored on composite layers based on magnesium-doped hydroxyapatite loaded in chitosan matrix

In the present study, we report the development and characterization of composite layers (by spin coating) based on magnesium-doped hydroxyapatite in a chitosan matrix, containing human osteosarcoma MG63 cells anchored. Studies regarding the biocompatibility of the composite layers were performed with the aid of a MTT (3–4,5-Dimethylthiazol 2,5-diphenyltetrazolium bromide) assay. The data determined that the composite layers did not inhibit the growth and adhesion of MG63 cells to their surfaces exhibiting good biocompatibility properties. Furthermore, the attachment and development of MG63 cells on the surface of MgHApCh composite layers were investigated using atomic force microscopy (AFM). AFM topographical maps emphasized that the HApCh and 8MgHApCh composite layers surface promoted the attachment and proliferation of MG63 cells on their surface. Meanwhile, in the case of 30MgHApCh layers incubated for 48 h, a slight modification of the morphological features of the MG63 cells. In addition, the effects of the composite layers against Candida albicans ATCC 10231 were also evaluated. The data results from the in vitro antifungal assay depicted that the composite layers successfully inhibited the growth of the fungal cells onto their surface. Morphological and fractal analyses unveil cancer cell surfaces on Mg-containing composite layers with intricate 3D patterns, driven by high-frequency components. Their remarkable complexity and roughness arises from a strong multifractal nature, supporting more effective vertical growth compared to Si and HApCh surfaces. The cell viability reduced of uncoated Si surface is highlighted by its less intense 3D pattern growth. Our results show that the uncoated Si surface promotes lower viability of MG63 cancer cells, with less rough and complex 3D spatial patterns.

Synthesis and Characterization of Mg-Hydroxyapatite and Its β-Cyclodextrin Composite as Enhanced Bio-Carrier of 5-Fluorouracil Drug; Equilibrium and Release Kinetics.

An advanced form of magnesium-doped hydroxyapatite (Mg·HAP) was integrated in composite with β-cyclodextrin producing a safe biocomposite (β-CD/HAP) as an enhanced delivery structure of traditional 5-fluorouracil (5-FU) chemotherapy during the treatment stages of colorectal cancer cells. The qualifications of β-CD/HAP as a carrier for 5-FU were followed based on the loading, release, and cytotoxicity as compared to Mg·HAP. β-CD/HAP composite exhibits notably higher 5-FU encapsulation capacity (272.3 mg/g) than Mg·HAP phase (164.9 mg/g). The 5-FU encapsulation processes into β-CD/HAP display the isotherm behavior of the Freundlich model (R2 = 0.99) and kinetic assumptions of pseudo-first order kinetic (R2 > 0.95). The steric studies reflect a strong increment in the quantities of the free sites after the β-CD integration steps (Nm = 61.2 mg/g) as compared to pure Mg·HAP (Nm = 42.4 mg/g). Also, the capacity of each site was enhanced to be loaded by 5 of 5-FU molecules (n = 4.45) in a vertical orientation. The 5-FU encapsulation energy into β-CD/HAP (<40 kJ/mol) reflects physical encapsulation reactions involving van der Waals forces and hydrogen bonding. The 5-FU release profiles of β-CD/HAP exhibit slow and controlled properties for about 80 h either in gastric fluid (pH 1.2) or in intestinal fluid (pH 7.4). The release kinetics and diffusion exponent (>0.45) signify non-Fickian transport and complex erosion/diffusion release mechanism. The free β-CD/HAP particles display a considerable cytotoxic effect on the HCT-116 cancer cells (33.62% cell viability) and its 5-FU-loaded product shows a strong cytotoxic effect (2.91% cell viability).

Orientation of carbon fiber in magnesium-doped hydroxyapatite and its effect on mechanical and tribological properties of carbon fiber reinforced composites

Carbon fiber (CF) reinforced magnesium-doped hydroxyapatite (Mg-HA) composites have been shown possess excellent mechanical performance and tailored biological properties. In order to further explore the effect of the orientation for short CF reinforced Mg-HA composites for mechanical and tribological properties. Micro-sized CF was oriented in Mg-HA by a self-developed mold and highly oriented CF reinforced magnesium doped hydroxyapatite (OCF/Mg-HA) composites were prepared by spark plasma sintering (SPS). The morphology, structure, wettability, mechanical properties and tribological properties of the composites were investigated. The results show that the average contact angle of oriented CF/Mg-HA composites was 67.65° and had good hydrophilicity. Compressive strength of OCF/Mg-HA in the direction parallel to the fiber was 129.8 MPa, which met the requirements of mechanical properties of human cortical bone. In addition, the wear resistance of OCF/Mg-HA ceramics was significantly improved by the addition of CF. The average friction coefficient of OCF/Mg-HA ceramics in the direction parallel to the fiber was 0.157. OCF/Mg-HA ceramics exhibited excellent tribological properties and mechanical properties which were close to that of human load-bearing bone due to the oriented arrangement of CF. This laid a foundation for the clinical application of OCF/Mg-HA ceramics in the repair of bone defects at load-bearing sites.

Vapor-Induced Pore-Forming Atmospheric-Plasma-Sprayed Zinc-, Strontium-, and Magnesium-Doped Hydroxyapatite Coatings on Titanium Implants Enhance New Bone Formation—An In Vivo and In Vitro Investigation

Titanium implants are regarded as a promising treatment modality for replacing missing teeth. Osteointegration and antibacterial properties are both desirable characteristics for titanium dental implants. The aim of this study was to create zinc (Zn)-, strontium (Sr)-, and magnesium (Mg)-multidoped hydroxyapatite (HAp) porous coatings, including HAp, Zn-doped HAp, and Zn-Sr-Mg-doped HAp, on titanium discs and implants using the vapor-induced pore-forming atmospheric plasma spraying (VIPF-APS) technique. The mRNA and protein levels of osteogenesis-associated genes such as collagen type I alpha 1 chain (COL1A1), decorin (DCN), osteoprotegerin (TNFRSF11B), and osteopontin (SPP1) were examined in human embryonic palatal mesenchymal cells. The antibacterial effects against periodontal bacteria, including Porphyromonas gingivalis and Prevotella nigrescens, were investigated. In addition, a rat animal model was used to evaluate new bone formation via histologic examination and micro-computed tomography (CT). The ZnSrMg-HAp group was the most effective at inducing mRNA and protein expression of TNFRSF11B and SPP1 after 7 days of incubation, and TNFRSF11B and DCN after 11 days of incubation. In addition, both the ZnSrMg-HAp and Zn-HAp groups were effective against P. gingivalis and P. nigrescens. Furthermore, according to both in vitro studies and histologic findings, the ZnSrMg-HAp group exhibited the most prominent osteogenesis and concentrated bone growth along implant threads. A porous ZnSrMg-HAp coating using VIPF-APS could serve as a novel technique for coating titanium implant surfaces and preventing further bacterial infection.

Synthesis and characterization of Mg-hydroxyapatite and its cellulose hybridized structure as enhanced bio-carrier of oxaliplatin drug; equilibrium and release kinetics.

An advanced form of magnesium-doped hydroxyapatite (Mg HAP) was synthesized and hybridized with cellulose fibers, producing a safe biocomposite (CF/Mg HAP) as an enhanced delivery structure of traditional oxaliplatin (OXPN) chemotherapy drug during the treatment stages of colorectal cancer. The qualifications of CF/Mg HAP as a carrier for OXPN were followed based on loading, release, and cytotoxicity as compared to Mg HAP. The CF/Mg HAP composite exhibits a notably higher OXPN encapsulation capacity (256.2 mg g-1) than the Mg HAP phase (148.9 mg g-1). The OXPN encapsulation process into CF/Mg HAP displays the isotherm behavior of the Langmuir model (R2 = 0.99) and the kinetic assumptions of pseudo-first-order kinetics (R2 > 0.95). The steric studies reflect a strong increment in the quantities of the free sites after the cellulose hybridization steps (Nm = 178.58 mg g-1) as compared to pure Mg HAP (Nm = 69.39 mg g-1). Also, the capacity of each site was enhanced to be loaded by 2 OXPN molecules (n = 1.43) in a vertical orientation. The OXPN encapsulation energy into CF/Mg HAP (<40 kJ mol-1) reflects physical encapsulation reactions involving van der Waals forces and hydrogen bonding. The OXPN release profiles of CF/Mg HAP exhibit slow and controlled properties for about 100 h, either at pH 5.5 or pH 7.4. The release kinetics and diffusion exponent (>0.45) signify non-Fickian transport and a complex erosion/diffusion release mechanism. The free CF/Mg HAP particles display a considerable cytotoxic effect on the HCT-116 cancer cells (21.82% cell viability), and their OXPN-loaded product shows a strong cytotoxic effect (1.85% cell viability).

In situ synthesis of magnesium-doped hydroxyapatite aerogel for highly efficient U(VI) separation with ultra high adsorption capacity and excellent recyclability

Mg-doped HAP aerogel (MHAPA) was firstly in situ prepared via freeze-drying-calcination technology to capture U(VI). The U(VI) removal capacity by MHAPA even arrived 2685.6 mg g−1, which was about 2 times over purchased HAP, illustrating that the incorporation of Mg ions could greatly enhance the U(VI) removal capacity. Compared with HAP, MHAPA also showed better anti-ion interference ability and dynamic removal performances. In comparison with other HAP-based adsorbents, MHAPA possessed good recyclability and its desorption rate was up to 93.4% in the first cycle. The excellent U(VI) removal performances of MHAPA might be owing to its low crystallinity and grain size, fast ion exchange rate and partial ionization under acidic conditions, which would accelerate the process of electrostatic attraction, ion-exchange, and complexation to immobilize U(VI). To sum up, the prepared MHAPA was expected to be an environmentally friendly, recyclable and effective adsorbent to immobilize U(VI) in actual wastewater.

Hydroxyapatite nanoparticles-cell interaction: New approaches to disclose the fate of membrane-bound and internalised nanoparticles

Hydroxyapatite nanoparticles are popular tools in bone regeneration, but they have also been used for gene delivery and as anticancer drugs. Understanding their mechanism of action, particularly for the latter application, is crucial to predict their toxicity. To this end, we aimed to elucidate the importance of nanoparticle membrane interactions in the cytotoxicity of MG-63 cells using two different types of nanoparticles. In addition, conventional techniques for studying nanoparticle internalisation were evaluated and compared with newer and less exploited approaches. Hydroxyapatite and magnesium-doped hydroxyapatite nanoparticles were used as suspensions or compacted as specular discs. Comparison between cells seeded on the discs and those supplemented with the nanoparticles allowed direct interaction of the cell membrane with the material to be ruled out as the main mechanism of toxicity. In addition, standard techniques such as flow cytometry were inconclusive when used to assess nanoparticles toxicity. Interestingly, the use of intracellular calcium fluorescent probes revealed the presence of a high number of calcium-rich vesicles after nanoparticle supplementation in cell culture. These structures could not be detected by transmission electron microscopy due to their liquid content. However, by using cryo-soft X-ray imaging, which was used to visualise the cellular ultrastructure without further treatment other than vitrification and to quantify the linear absorption coefficient of each organelle, it was possible to identify them as multivesicular bodies, potentially acting as calcium stores. In the study, an advanced state of degradation of the hydroxyapatite and magnesium-doped hydroxyapatite nanoparticles within MG-63 cells was observed. Overall, we demonstrate that the combination of fluorescent calcium probes together with cryo-SXT is an excellent approach to investigate intracellular calcium, especially when found in its soluble form.

Biological and Physico-Chemical Properties of Composite Layers Based on Magnesium-Doped Hydroxyapatite in Chitosan Matrix.

In the present study, we report the development and characterization of composite layers (by spin coating) based on magnesium-doped hydroxyapatite in a chitosan matrix, (Ca10−xMgx(PO4)6(OH)2; xMg = 0, 0.08 and 0.3; HApCh, 8MgHApCh and 30MgHApCh). The MgHApCh composite layers were investigated using scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), and X-ray photoelectron spectroscopy (XPS) techniques. The in vitro biological evaluation included the assessment of their cytotoxicity on MG63 osteoblast-like cells and antifungal activity against Candida albicans ATCC 10231 fungal cell lines. The results of the physico-chemical characterization highlighted the obtaining of uniform and homogeneous composite layers. In addition, the biological assays demonstrated that the increase in the magnesium concentration in the samples enhanced the antifungal effect but also decreased their cytocompatibility. However, for certain optimal magnesium ion concentrations, the composite layers presented both excellent biocompatibility and antifungal properties, suggesting their promising potential for biomedical applications in both implantology and dentistry.

Impact of Gamma Irradiation on the Properties of Magnesium-Doped Hydroxyapatite in Chitosan Matrix.

This is the first report regarding the effect of gamma irradiation on chitosan-coated magnesium-doped hydroxyapatite (xMg = 0.1; 10 MgHApCh) layers prepared by the spin-coating process. The stability of the resulting 10 MgHApCh gel suspension used to obtain the layers has been shown by ultrasound measurements. The presence of magnesium and the effect of the irradiation process on the studied samples were shown by X-ray photoelectron spectroscopy (XPS). The XPS results obtained for irradiated 10 MgHApCh layers suggested that the magnesium and calcium contained in the surface layer are from tricalcium phosphate (TCP; Ca3(PO4)2) and hydroxyapatite (HAp). The XPS analysis has also highlighted that the amount of TCP in the surface layer increased with the irradiation dose. The energy-dispersive X-ray spectroscopy (EDX) evaluation showed that the calcium decreases with the increase in the irradiation dose. In addition, a decrease in crystallinity and crystallite size was highlighted after irradiation. By atomic force microscopy (AFM) we have obtained images suggesting a good homogeneity of the surface of the non-irradiated and irradiated layers. The AFM results were also sustained by the scanning electron microscopy (SEM) images obtained for the studied samples. The effect of gamma-ray doses on the Fourier transform infrared spectroscopy (ATR-FTIR) spectra of 10 MgHApCh composite layers was also evaluated. The in vitro antifungal assays proved that 10 MgHApCh composite layers presented a strong antifungal effect, correlated with the irradiation dose and incubation time. The study of the stability of the 10 MgHApCh gel allowed us to achieve uniform and homogeneous layers that could be used in different biomedical applications.

Multi-ionic interaction with magnesium doped hydroxyapatite-zeolite nanocomposite porous polyacrylonitrile polymer bead in aqueous solution and spiked groundwater

Removal of multi-ionic contaminants from water resources has been a major challenge faced during the treatment of water for drinking and industrial applications. In the present study, varying composition of magnesium doped hydroxyapatite (Mg-HAp) and zeolite nanocomposite embedded porous polymeric beads were synthesized using solvent displacement method and its sorption efficiency towards multi-ion contaminant (such as Ag, Al, As, Ba, Be, Cd, Co, Cr, Cu, Mn, Ni, Pb, Se, Tl, Th, U, V and Zn) was investigated in aqueous solution and spiked groundwater. The prepared beads were characterized using suitable techniques like high-resolution transmission electron microscopy (HRTEM), scanning electron microscopy (SEM), and Brunauer-Emmett-Teller (BET) equation. The surface area and pore radius of the beads varied from 6.996 to 66.469 m2/g and 1.698–3.960 nm respectively according to the composition of the bead. The control bead without nanocomposite showed maximum surface area. Multi-ion adsorptions onto beads were confirmed using an inductively coupled plasma-optical emission spectrophotometer (ICP-OES) and X-ray photoelectron spectrophotometer (XPS). The sorption efficiency was high at pH 5 owing to its anionic surface charge leading to an increase in affinity towards the cations. For validating field application, selected high performance beads were tested in multi-ion spiked groundwater. The results indicated that the Mg-HAp nanocomposite bead dominate all the other bead compositions with more than 90% removal efficiency for most of the multi-ion contaminants. The feasible adsorption mechanism has been discussed. This adsorption study revealed that the Mg-HAp nanocomposite bead is a promising material that is cost-effective, non-toxic, biodegradable, eco-friendly and highly efficient towards the removal of multi-ionic contaminants from groundwater.

Physicochemical and Biological Evaluation of Chitosan-Coated Magnesium-Doped Hydroxyapatite Composite Layers Obtained by Vacuum Deposition

In the present work, the effectiveness of vacuum deposition technique for obtaining composite thin films based on chitosan-coated magnesium-doped hydroxyapatite Ca10−xMgx(PO4)6 (OH)2 with xMg = 0.025 (MgHApCh) was proved for the first time. The prepared samples were exposed to three doses (0, 3, and 6 Gy) of gamma irradiation. The MgHApCh composite thin films nonirradiated and irradiated were evaluated by scanning electron microscopy (SEM), atomic force microscopy (AFM), and X-ray photoelectron spectroscopy (XPS) studies. The biological evaluation of the samples was also presented. All the results obtained from this study showed that the vacuum deposition method allowed for obtaining uniform and homogeneous layers. Fine cracks were observed on the MgHApCh composite thin films’ surface after exposure to a 6 Gy irradiation dose. Additionally, after gamma irradiation, a decrease in Ca, P, and Mg content was noticed. The MgHApCh composite thin films with doses of 0 and 3 Gy of gamma irradiation showed a cellular viability similar to that of the control. Samples with 6 Gy doses of gamma irradiation did not cause significantly higher fibroblast cell death than the control (p &gt; 0.05). On the other hand, the homogeneous distribution of pores that appeared on the surface of coatings after 6 Gy doses of gamma irradiation did not prevent the adhesion of fibroblast cells and their spread on the coatings. In conclusion, we could say that the thin films could be suitable both for use in bone implants and for other orthopedic and dentistry applications.

Osteogenic potential evaluation of biotin combined with magnesium-doped hydroxyapatite sustained-release film

Biotin is one of the water-soluble B-complex group of vitamins. Recent studies have found that the relative protein expression of BMP2, BSP and OPG in MC3T3-E1 cells is prominent after 14 days of co-culture with biotin film, especially for BMP2. It is also found that the rapid degradation of biotin film in vivo limits its application value. In this work, magnesium-doped hydroxyapatite (MgHA) film can form a porous network structure as a biological sustained-release film. Therefore, the multilayer (MgHA|biotin|MgHA|biotin) film was prepared by pulsed laser assisted electron beam deposition technique. The morphology, structure and properties of biotin film and multilayer film were analyzed and characterized. Also, the osteogenic effect of biotin film and multilayer film was evaluated after implantation into the femoral bone marrow cavity of SD rats. The results of micro-CT scan and 3D reconstruction showed that there were a large number of trabecular bones around the multilayer film, which was superior to biotin film in osteogenesis. Hematoxylin-eosin staining showed cancellous bone structure and intact bone marrow structure around the multilayer film, and the newly formed bone became lamellar. Masson-trichromatic staining revealed abundant osteoid and braided bone formation around the multilayer film. In conclusion, MgHA sustained release film can realize the continuous release of bioactive drugs, which provides a new route to accelerate the repair of bone defects.

Potential Load-Bearing Bone Substitution Material: Carbon-Fiber-Reinforced Magnesium-Doped Hydroxyapatite Composites with Excellent Mechanical Performance and Tailored Biological Properties.

A potential load-bearing bone substitution and repair material, that is, carbon fiber (CF)-reinforced magnesium-doped hydroxyapatite (CF/Mg-HAs) composites with excellent mechanical performance and tailored biological properties, was constructed via the hydrothermal method and spark plasma sintering. A high-resolution transmission electron microscopy (TEM) was employed to characterize the nanostructure of magnesium-doped hydroxyapatite (Mg-HA). TEM images showed that the doping of Mg-induced distortions and dislocations in the hydroxyapatite lattice, resulting in decreased crystallinity and enhanced dissolution. Compressive strengths of 10% magnesium-doped hydroxyapatite (1Mg-HAs) and CF-reinforced 1Mg-HAs (CF/1Mg-HAs) were within the range of that of cortical bone. Compared with 1Mg-HAs, the fracture toughness of CF/1Mg-HAs increased by approximately 38%. The bioactivity, biocompatibility, and osteogenic induction properties of Mg-HAs and CF/Mg-HAs composites were evaluated in vitro using simulated body fluid (SBF) immersion, cell culture, osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs), and expression of genes associated with osteogenesis. When Mg-HAs were immersed in SBF, Mg2+ continued to release for up to 21 days. Mg-HAs demonstrated a satisfactory ability to induce apatite formation in comparison with HAs. The cell proliferation and morphology on CF/1Mg-HAs were similar to those of 1Mg-HAs, suggesting that adding CF had no adverse effect on cellular activity. The expression levels of osteogenesis-related genes [osteocalcin (OPN), osteopontin (OCN), and runt-related transcription factor 2 (Runx2)] on 1Mg-HAs were significantly higher at days 3 and 7 than those on HAs and 0.5Mg-HAs groups. This finding suggests that a certain amount of Mg doping had beneficial influences in the different stages of osteogenic differentiation and could induce osteogenic differentiation of BMSCs. The new bone volume to total volume ratio of implanted 1Mg-HAs (30.9% ± 4.1%) and CF/1Mg-HAs (25.4% ± 5.4%) was remarkably higher than that of HAs (21.6% ± 3.9%). 1Mg-HAs and CF/1Mg-HAs tailored an ideal effect of new bone information and implant osseointegration. The excellent mechanical performance and tailored biological properties of CF/Mg-HAs were attributed to nano Mg-doped HA, CF reinforcing, refined microstructure, and controlled composition.

Synthesis of luminescent Mg-incorporated hydroxyapatite by reflux condensation method: Photoluminescence, in-vitro drug release and kinetic studies

The role of biomaterials in the field of medicine has immense applications because they are engineered to be in close proximity to the biological organ or tissue and thereby healing or replacing the damaged tissues or organs. These biomaterials are of natural and synthetic made of different components to interact with the biological system. They can be tailored in such a way either for therapeutic or diagnostic purposes. The focus of the present study is to prepare nanosized magnesium doped hydroxyapatite (Mg-HAp) using magnesium nitrate as the source of magnesium (Mg). The reflux condensation method was used to synthesize pure HAp and Mg-HAp samples that were subjected to microwave irradiation. XRD studies verify that the integration of Mg into HAp did not bring about any phase changes in HAp. Further, the changes in the lattice parameters of Mg-HAp show that Mg is well incorporated into the HAp lattice. FTIR and Raman spectroscopic investigations show the absence of the hydroxyl group, which adds to the confirmation of Mg substitution in the HAp lattice. Crystallinity and size of the Mg-HAp decreased for microwave irradiated samples compared to as-synthesized samples because of the uniform heating of the sample. The morphology of Mg-HAp samples further indicates that particle size reduces with increasing Mg concentration which was proven by SEM results. Photoluminescence spectra of microwave irradiated Mg-HAp samples exhibit increased intensity for increased Mg concentration because of the blocking of recombination of charge carriers. The self-luminescent property of Mg-HAp particles, which is because of their surface plasmon, can be used in imaging and drug delivery applications.
