Oxidative Stress and Serum S100B Levels in Adolescents with First-Episode Drug-Naive Unipolar Depression.

Unipolar depression is common among adolescents and has high recurrence rates. Studies conducted with adults show that oxidative stress plays a role in etiology of depression but studies with adolescent patients are limited. In addition, baseline S100B level in adult patients with depression is considered as a marker of response to treatment. The purpose of this study was to measure the levels of serum S100B, Malondialdehyde (MDA), total oxidant status (TOS), and total antioxidant status (TAS), which have not been previously investigated in adolescent patients with first-episode, drug-naive unipolar depression, and to investigate the relationship of these parameters with disease severity and patient-specific variables. This study was conducted with 37 adolescents diagnosed with unipolar depression and 37 healthy peers. Participants were asked to fill out the Beck Depression Scale, Screen for Child Anxiety Related Disorders, and suicide probability questionnaires. After this procedure, 5 cc blood was collected from the adolescents and serum S100B, MDA, TOS, and OSI levels measured. Serum S100B, MDA, TOS, and OSI levels were higher and TAS level was lower in patients than their healthy peers. There was no relationship between the patients' severity of depression or suicide probability and these parameters. The serum S100B, MDA, TOS, and OSI levels of female patients were higher than their healthy peers, but the TAS level was not different. Male patients had higher TOS and OSI levels and lower TAS levels than their healthy peers. The results show that increased serum S100B, MDA, TOS and OSI levels may contribute to etiology of depression regardless of gender. The gender-specific increase in S100B and MDA levels, which were significantly increased in female adolescent patients but not in males, should be supported by further follow-up studies.

Relationship between behcet's disease and oxidant and antioxidant markers

Behcet’s disease is known as a chronic and inflammatory vasculitis with multiple system and organ involvement. In this study, we aimed to evaluate the relationship between BD and oxidative stress by evaluating the total antioxidant status (TAS), total oxidant status (TOS) and oxidative stress index (OSI) in healthy control group and patients with Behcet’s disease (BD). Materials and Methods: Study groups were composed of BD patients (n = 57) and healthy controls (n = 54). Serum total TAS, TOS, (OSI) and serum lipid levels were measured by photometric method. All measurements were made on Beckman Coulter AU5800 Series (Japan). OSI was determined by the TOS / TAS ratio. Serum TAS levels in BD were lower than the healthy controls (p = 0.039). Serum levels of TOS, OSI and lipids did not differ significantly in BD patients compared to healthy controls. Also, the sensitivity of TAS for BD was high (0.825). Low TAS levels can play an important role in the etiopathology of BD. Increasing serum TAS levels may be beneficial for the clinical process. More studies are needed in this regard.

Evaluation of thiol-disulfide homeostasis in pseudoexfoliation glaucoma and primary open-angle glaucoma.

This study aimed to compare the serum thiol-disulfide homeostasis, total antioxidant status (TAS), and total oxidant status (TOS) in patients with pseudoexfoliation glaucoma (PEG) patients, primary open-angle glaucoma (POAG) patients, and healthy individuals (control). Ninety subjects were included in this study. Three groups were separated as PEG, POAG, and control. All groups were chosen to be similar in terms of age and gender. Blood samples were obtained following an overnight fasting state and were collected on the ice at 4°C. The serum samples were separated from the cells by centrifugation at 3000 rpm for 15 min and were stored at -80°C. Serum samples analyzed for TAS and TOS, native thiol, total thiol, disulfide, and native thiol/disulfide ratio. TAS and TOS levels of PEG patients were 1.2892 ± 0.0905 mmol/L; 5.0191 ± 2.7722 μmol/L, respectively. TAS and TOS levels of POAG patients were 1.2741 ± 0.1252 mmol/L; 4.1674 ± 1.7723 μmol/L, respectively. TAS and TOS levels of the control group were 2.3414 ± 0.1409 mmol/L; 4.0931 ± 0.1107 μmol/L, respectively. The TAS level was significantly lower in PEG and POAG groups compared to control. TOS level showed no significant differ ¬ ence between PEG, POAG, and control groups (P > 0.05). The mean serum total thiol and native thiol levels were significantly lower in patients with PEG compared to POAG and control group; there was no significant difference between the POAG and control group (P > 0.05). The mean serum disulfide level was significantly lower in patients with PEG compared to POAG (P = 0.018). Low levels of TAS were observed in patients with glaucoma, which was likely a response to the increased oxidative stress observed in these patients. While total thiol and native thiol levels were higher in the PEG group, the disulfide level was higher in the POAG group. TAS and TOS levels showed no significant difference between POAG and PEG groups.

Evaluating plasma oxidative stress markers in prelingual profound sensorineural hearing loss

Background and Aim: The etiopathogenesis of sensorineural hearing loss (SNHL) is an ess­ential contributing factor to its morbidity, which cannot be explained entirely so far. The current study aimed to determine the oxidative stress (OS) status by comparing the total antioxidant status (TAS), total oxidant status (TOS) and oxidative stress index (OSI) values in patients with pediatric prelingual profound SNHL. We also evaluated the correlation between OS parameters and audiological test results.
 Methods: The study included 25 participants (9 females; age range: 6‒34 months) diagnosed with pediatric prelingual profound SNHL and 25 healthy subjects (10 females; age range: 9‒28 months). Their TAS, TOS, and OSI levels were measured in the plasma of both groups. We evaluated the correlation between OS para­meters and audiological test results in the pati­ent group.
 Results: We found significantly higher serum TOS levels and OSI values in the patient group (mean ± SD of TOS: 16.08 ± 1.88 μmol H2O2 eq/L, p < 0.001; mean ± SD of OSI: 1.71 ± 0.48 arbitrary units, p < 0.001), compared to the
 controls. Moreover, we found lower serum TAS levels in the patient group (mean ± SD of TAS: 0.99 ± 0.20 mmol Trolox eq/L), compared to the controls. There was a strong correlation between OS parameters and audiological test results of the patient group.
 Conclusion: We detected significantly higher TOS, OSI, and lower TAS levels in pediatric patients with SNHL, compared to the healthy subjects. The obtained data indicated that pediatric SNHL is under OS influence.

Serum Oxidative Stress Levels in Patients with Nasal Septal Deviation.

Comparison of the total oxidant status (TOS), total antioxidant status (TAS) and paraoxonase (PON1) serum levels in patients with nasal septum deviation (NSD) and healthy subjects. The TAS, TOS, and PON1 serum levels of 47 patients with NSD (mean age 35.3) and 50 healthy subjects (mean age 37.8) were compared in this study. We found significantly higher TAS levels in the control group (p<0.001). The mean TAS value was 1.196 mmoL/L in the control group, whereas it was 1.046 mmoL/L in the NSD group. On the other hand, TOS was significantly higher in the NSD group (p<0.001). We found that the mean TOS value was 6.600 mmoL/L in the control group, and 20.194 mmoL/L in the NSD group. The NSD and control groups had similar PON1 levels (p=0.446). The mean PON1 value was 279.64 U/L in the control group, and 324.21 U/L in the NSD group. We detected higher TOS and lower TAS levels in patients with NSD compared to healthy subjects. These results indicate that patients with NSD are exposed to oxidative stress.

A new experimental rat model of pancreatitis using Ecballium elaterium

BackgroundWe originally aimed to determine the beneficial effects of Ecballium Elaterium (EE) on acute pancreatitis; however, we observed negative effects of EE on the pancreas. Thus, we used EE in rats to generate a new model of pancreatitis, which we compared with other established pancreatitis models. MethodsA total of 32 Wistar albino rats were used. Rats were divided into 4 groups, each of which contained 8 rats. Group 1 (Control), Group 2 (L-Arginine (LA), Group 3 (LA + EE), Group 4 (EE): Twenty-four hours after that serum parameters were analyzed in the collected blood. Blood samples were transported on mice to the Biochemistry Laboratory following cardiac puncture. The levels of amylase, interleukin (IL)-6, interleukin (IL) 1-β (IL-1β), malondialdhyde (MDA), tumor necrosis factor (TNF)-α, total antioxidant status (TAS), and total oxidant status (TOS) were analyzed. Histopathological analysis: The pancreas and lung tissue samples obtained from the rats. Edema, inflammation, vacuolization, and necrosis of the pancreas were assessed using a scoring system ranging from 0 to 4. Edema, hemorrhage and inflammation of the lung tissue were evaluated using a scoring system ranging from 0 to 3. ResultsHistopathological analysis revealed that edema, inflammation, necrosis, and hemorrhage were significantly higher in the LA + EE group than in the control group. Moreover, necrosis was higher in the rats that received LA and EE compared to the rats that received only LA or EE. Increases in inflammatory mediator levels, including IL-6, IL-1β, TNF-α, MDA, and TOS, were observed in all groups as compared to the control group. Moreover, lower TAS levels were detected in all groups but the control group. The increase in IL-1β and TNF-α levels and the decrease in TAS were statistically significant in all groups (P < 0.05). ConclusionsEE may be used to create a successful acute pancreatitis (AP) model, resulting in edema, necrosis, hemorrhage, and inflammation of the pancreas. The major advantage of this model is that it does not require laparotomy, and can be implemented with only an intraperitoneal injection (IP). Moreover, EE may be combined with other agents, such as LA, to create severe pancreatitis. Further molecular studies are warranted to determine the underlying effects of EE on the pancreas.

Effect of Montelukast Monotherapy on Oxidative Stress Parameters and DNA Damage in Children with Asthma

Background: There is ample knowledge reported in the literature about the role of oxidative stress in asthma pathogenesis. It is also known that the interaction of reactive oxygen species with DNA may result in DNA strand breaks. The aim of this study was to investigate if montelukast monotherapy affects oxidative stress and DNA damage parameters in a population of pediatric asthma patients. Methods: Group I consisted of 31 newly diagnosed asthmatic patients not taking any medication, and group II consisted of 32 patients who had been treated with montelukast for at least 6 months. Forty healthy control subjects were also enrolled in the study. Plasma total oxidant status (TOS) and total antioxidant status (TAS) were measured to assess oxidative stress. DNA damage was assessed by means of alkaline comet assay. Results: The patients in both group I and group II had statistically significant higher plasma TOS (13.1 ± 4 and 11.1 ± 4.1 μmol H₂O₂ equivalent/liter, respectively) and low TAS levels (1.4 ± 0.5 and 1.5 ± 0.5 mmol Trolox equivalent/liter, respectively) compared with the control group (TOS: 6.3 ± 3.5 μmol H₂O₂ equivalent/liter and TAS: 2.7 ± 0.6 mmol Trolox equivalent/liter; p < 0.05). DNA damage was 18.2 ± 1.0 arbitrary units (a.u.) in group I, 16.7 ± 8.2 a.u. in group II and 13.7 ± 3.4 a.u. in the control group. There were statistically significant differences only between group I and the control group (p < 0.05). Conclusions: According to the findings, montelukast therapy makes only minimal but not statistically significant improvement in all TOS, TAS and DNA damage parameters.

Reduced SIRT1 expression correlates with enhanced oxidative stress in compensated and decompensated heart failure

Sirtuin-1 (SIRT1) is a longevity factor in mammals initiating the cell survival mechanisms, and preventing ischemic injury in heart. In the etiopathogenesis of heart failure (HF), impairment in cardiomyocyte survival is a notable factor. Oxidative stress comprises a critical impact on cardiomyocyte lifespan in HF. The aim of the present study was to investigate SIRT1 expression in patients with compensated (cHF) and decompensated HF (dHF), and its correlation with oxidative stress. SIRT1 expression in peripheral leukocytes was examined using quantitative RT-PCR in 163 HF patients and 84 controls. Serum total oxidant status (TOS) and total antioxidant status (TAS) were measured via colorimetric assays, and oxidative stress index (OSI) was calculated. Lipid parameters were also determined by routine laboratory methods. SIRT1 mRNA expression was significantly downregulated in HF with more robust decrease in dHF (p=0.002, control vs cHF; p<0.001, control vs dHF). Markedly increased oxidative stress defined as elevated TOS, OSI and low TAS levels were detected in HF patients comparing with the controls (TAS; p=0.010, control vs cHF, p=0.045 control vs dHF, TOS; p=0.004 control vs cHF; p<0.001 control vs dHF, OSI; p<0.001 for both comparisons, respectively). With SIRT1 expression levels, TAS, TOS, OSI, and high density lipoprotein levels in cHF and dHF were determined correlated. SIRT1 expression were significantly reduced in both HF subtypes, particularly in dHF. SIRT1 expression was correlated with the oxidant levels and antioxidant capacity. Data suggest that SIRT1 may have a significant contribution in regulation of oxidant/antioxidant balance in HF etiology and compensation status.

Paraoxonase and arylesterase activity and total oxidative/anti-oxidative status in patients with idiopathic Parkinson’s disease

This study investigated serum paraoxonase (PON1) and arylesterase activity along with determination of oxidative status via measurement of total oxidant status (TOS), total anti-oxidant status (TAS) and oxidative stress index (OSI) in patients with Parkinson’s disease (PD) and compared results with data from healthy controls. A total of 82 subjects, including 42 patients with idiopathic PD, newly diagnosed and untreated (24 men, 18 women, aged 47–66years) and 40 healthy controls were enrolled in this study. We aimed to evaluate the oxidative status of PD patients via measurement of serum TOS and TAS and estimation of OSI using new automated methods. PON1 and arylesterase activities were measured spectrophotometrically. Serum total cholesterol, high density lipoprotein cholesterol, low density lipoprotein (LDL) cholesterol and triglyceride levels were measured using routine methods. TAS levels of PD patients were significantly lower than that of controls (p<0.05). TOS levels of PD patients were higher than those of controls (p<0.05). PON1 and arylesterase activities of PD were lower than those of controls (p<0.05). Serum levels of total and LDL cholesterol were significantly reduced in PD patients. In conclusion, the presence of high TOS and OSI levels together with low levels of TAS in PD patients supports the important role of oxidative stress in the pathophysiology of PD. Since oxidative stress is involved in neurodegeneration, selecting anti-oxidants, metal chelators or other compounds boosting endogenous enzymatic and non-enzymatic defense mechanisms seems to be an obvious choice as treatment for PD.

Caffeic acid phenethyl ester (CAPE) protects against acute urogenital injury following pneumoperitoneum in the rat

Purpose: Our aim is to determine the biochemical and histologic changes induced in the kidneys, testis and prostate on possible ischemia and reperfusion (I/R) injury caused by pneumoperitoneum (PNP) in a rat model and to evaluate the ability of protective effects of caffeic acid phenethyl ester (CAPE). Methods: Twenty-eight adult male Wistar albino rats were randomly divided to one of three treatment groups, with seven animals in each group. Sham, laparoscopy (L), and laparoscopy plus CAPE (L + C) group were subjected to 60 min of PNP with 15 mmHg one hour before the desufflation period. Total oxidant status (TOS) and total antioxidant status (TAS) levels were determined in kidney, testis, and prostate. Kidney and testis tissues were removed to obtain a histologic score. Also, Johnsen scoring system was used for testicular tissue analysis. Results: L group had significantly higher TOS and lower TAS levels on kidney and testis compared to the other groups. In prostate biochemical analysis, there was not any difference between groups. No difference was found between groups according to kidney and testis tissues’ histologic evaluation. In evaluation of Johnsen scoring, L group showed significant lower score compared to the other two groups. Conclusions: Increased intraabdominal pressure (IAP) had an oxidative effect on kidney and testis but not on prostate in rats. Moreover, it could affect the testicular Johnsen score. All these adverse effects of IAP on both kidney and testis could be prevented by CAPE administration.

Oxidative stress parameters and ceruloplasmin levels in patients with severe preeclampsia.

Oxidative stress is believed to have a role in the development of preeclampsia (PE). It is known that an increased ceruloplasmin (CP) level is also associated with PE. The aim of this study was to investigate the relationship between oxidative stress parameters and CP levels in patients with severe PE. Sixty patients with severe PE and 60 healthy pregnant women were recruited to the study. All study subjects were divided into 2 groups; group 1(n=60) consisted of patients with severe PE, and group 2 (n=60) consisted of healthy pregnant subjects. Blood samples were obtained to measure CP, total antioxidant status and total oxidant status from all subjects. Oxidative stress index was calculated. Compared to group 2; group 1 had significantly higher CP, total oxidant status, oxidative stress index and lower total antioxidant status levels (p<0.001, p<0.001, p=0.001, p=0.008, respectively). Serum CP levels were significantly correlated with oxidative stress index levels (r=0.385, p=0.002). The present study demonstrated that both oxidative stress and CP levels increased in patients with PE, and increased CP levels seem to be a consequence of oxidative stress.

Serum prolidase activity and oxidative status in patients with diabetic neuropathy

We found no data in the literature related to oxidative stress index (OSI), total oxidative status (TOS) and prolidase activity in patients with diabetic neuropathy (DN). In this study, we aimed to evaluate the oxidative status of DN patients via measurement of TOS and serum total antioxidant status (TAS) and estimation of OSI using new automated methods. Thirty-eight healthy participants, 40 diabetic patients without neuropathy, and 39 patients with DN were included. Electrophysiological and neurological examinations were performed. The activity of prolidase and levels of TOS and TAS were determined in the serum of patients. The level of TAS was lower, while the levels of TOS and OSI, and activity of prolidase were higher in both DN and diabetic control groups compared with the healthy subjects (p < 0.05). Prolidase activity was found to be higher in the DN group than in the diabetic control group (p = 0.001). In conclusion, the presence of high TOS and OSI levels together with low levels of TAS in diabetic patients with or without neuropathy may support a role of oxidative stress in the pathogenesis of diabetes mellitus. In addition, increased serum prolidase activity in DN may be interpreted as evidence of increased collagen turnover.

Antioxidant and Oxidant Levels of Pepper (Capsicum annuum cv. ‘Charlee’) Infected with Pepper Mild Mottle Virus

Total antioxidant status (TAS), total oxidant status (TOS), as well as total phenol (TP), free phenol (FP), conjugated phenol (CP), and the vitamin C content of pepper plants (Capsicum annuum cv. ‘Charlee’) infected with Pepper mild mottle virus (PMMoV) were determined via a novel automated antioxidant system using test kits. PMMoV was first identified in Turkey in 1994 in commercial pepper (Capsicum annuum cv. ‘Charlee’) fields and caused an outbreak in southern Turkey by 2006, especially in greenhouses. Infected plants were confirmed by ELISA using commercial kits for the virus. The infected plants had stunted growth; and leaves and fruits became mottled, puckered, malformed, deformed, and marked by off-colored sunken areas. Fruits expressing virus symptoms were utilized for biochemical analyses. The percent ratio of TOS to TAS was calculated as the oxidative stress index (OSI). The TAS level and the vitamin C content were found to be significantly lower in infected peppers as compared to healthy peppers. However, TOS, OSI, TP, FP, and CP levels were significantly higher in infected pepper fruits than in healthy ones. In this study, it has been demonstrated that low levels of TAS and high levels of TOS, as well as high OSI status, reflects the quality of peppers. Using this method, the quality of fruits and the condition of plants can be determined in advance of stress development.

Homocysteine oxidative stress and relation to bone mineral density in post-menopausal osteoporosis

Mildly elevated homocysteine (Hcy) and oxidative stress are novel and potentially modifiable risk factors for post-menopausal osteoporosis. We hypothesized that imbalance of oxidant/ antioxidant status and increased Hcy concentration stimulates osteoporotic activity, leading to increased collagen I breakdown in post-menopausal women. Patients were divided into 2 groups (NOP and OP). Group NOP had normal bone mineral density (BMD) and group OP low BMD. Thirty-four (69%) were in group OP and 15 (31%) in group NOP. Serum Total Antioxidant Status (TAS) and Total Peroxide (TPx) levels were determined with new automated methods. The study included measurement of Deoxypyridinoline (DPD). In OP patients plasma t-Hcy, urine deoxypyridinoline and plasma TPx were significantly higher than those in NOP controls. In addition, OP patients also had lower TAS levels than controls, which represent the oxidative imbalance. Pearson's correlation analysis revealed a negative correlation between t-Hcy and TAS (p<0.038). A significant negative correlation was also found between TAS level and BMD values for the spine in OP patients (p<0.035). In contrast, a positive correlation between t-Hcy and TPx in OP patients was demonstrated significantly, r=0.52, p<0.029. We show that the OP group had reduced TAS, whereas the elevated TPx was different from that in the NOP group. Slightly elevated homocysteinemia may contribute to increasing TPx and reducing TAS in the OP group. However, our results suggest a weak but negative relationship between TAS and BMD. Further investigations are needed to examine the relationship of oxidative stress as an endogenous bioactive agent to bone loss in post-menopausal women. Since oxidative stress is the imbalance between total oxidants and antioxidants in the body, any single oxidant/ antioxidant parameter may not reflect overall oxidative stress. Further studies are needed to understand the underlying mechanisms of these findings.

Study of total antioxidant status and glutathione peroxidase activity in Tunisian vitiligo patients

Background:Vitiligo affects one to two percent of the word population. Its pathogenesis has not been clarified yet. Multiple mechanisms such as autoimmune, neuronal, endocrine and oxidative stress resulting from unbalanced antioxidant defense system have been proposed.Aims:Our purpose was to study the total antioxidant status and glutathione peroxidase activity in Tunisian vitiligo patients with or without diabetes or dysthyroidism.Materials and Methods:We studied 60 vitiligo patients and 62 healthy controls. The sex ratio male/female in vitiligo patients was (27/33 = 0.81). Patients with vitiligo were divided into three groups, according to the association with diabetes or dysthyroidism. The total antioxidant status (TAS), glutathione peroxidase activity (GPX activity) was evaluated by adaptable methods using Kits.Results and Conclusion:The generalized vitiligo was the most frequent type (35 patients versus 25 of focal ones). All patients having vitiligo showed low levels of TAS: 0.85 ± 0.7 and low GPX activity: 45 ± 0.6, as compared to the control group: 1.40 ± 0.12 mmol/L; 49 ± 1.8 U/L, (p < 0.01), for TAS and GPX, respectively. The association of low TAS and GPX activities was more pronounced in diabetic vitiligo patients than in dysthyroid vitiligo patients. This study demonstrated that antioxidant processes depletion (low TAS and low GPX activity) is clearly involved with vitiligo in Tunisian patients, regardless of the association of the disease with diabetes or dysthyroidism.

Maternal–neonatal serum paraoxonase 1 activity in relation to the mode of delivery

Aim: To investigate the effect of the mode of labour and delivery on the total antioxidant status (TAS) and the paraoxonase 1 (PON 1) serum activity in mothers and their newborns. Subjects and methods: One hundred six women with normal pregnancy were divided into 4 groups: group A ( n = 28) with normal labour and vaginal delivery (VG), group B ( n = 25) with scheduled caesarean section (CS), group C ( n = 26) with “emergency” CS and group D ( n = 27) with prolonged labour + VG. Blood was obtained from the mothers at the beginning of the labour process and immediately after delivery (pre- and post-delivery) as well as from the umbilical cord (CB). PON 1 activity and blood chemistry were determined using the Bayer Advia 1650 Clinical Chemistry System, whereas TAS levels were measured spectrophotometrically at 450 min in microtiter plates. Results: TAS levels were similar pre-delivery and low in CB in all the groups. In contrast, TAS levels were remarkably reduced in group C and in group D post-delivery whereas they were nearly unchanged in group B and just lowered in group A, at the same time of study. PON 1 activity was practically unaltered in group A and group B pre- vs. post-delivery. Interestingly, the enzyme activity was remarkably decreased in group C (222 ± 16 vs. 153 ± 14 U/min/mL) and group D (216 ± 16 vs. 135 ± 15 U/min/mL, p < 0.001) as compared with those of the other groups at the same time of study. Additionally, PON 1 activity was higher in the newborns of group A and group B than those in group C and group D. TAS and HDL positively correlated with PON 1 activity. Conclusions: The low TAS levels and the decreased PON 1 activity, which were found in groups C and D post-delivery, may be due to the increased production of free radicals, during long-lasting labour + VG and obstructive labour + CS. PON 1 activity was low in CB irrespectively of the mode of delivery, probably due to the low lipid levels in the serum of the umbilical cord. Neonates born with normal delivery or scheduled CS are benefited with a higher antiatherogenic enzyme activity perinatally.
