TRPM4 Conductances in Thalamic Reticular Nucleus Neurons Generate Persistent Firing during Slow Oscillations.

During sleep, neurons in the thalamic reticular nucleus (TRN) participate in distinct types of oscillatory activity. While the reciprocal synaptic circuits between TRN and sensory relay nuclei are known to underlie the generation of sleep spindles, the mechanisms regulating slow (<1 Hz) forms of thalamic oscillations are not well understood. Under in vitro conditions, TRN neurons can generate slow oscillations in a cell-intrinsic manner, with postsynaptic Group 1 metabotropic glutamate receptor activation triggering long-lasting plateau potentials thought to be mediated by both T-type Ca2+ currents and Ca2+-activated nonselective cation currents (ICAN). However, the identity of ICAN and the possible contribution of thalamic circuits to slow rhythmic activity remain unclear. Using thalamic slices derived from adult mice of either sex, we recorded slow forms of rhythmic activity in TRN neurons, which were driven by fast glutamatergic thalamoreticular inputs but did not require postsynaptic Group 1 metabotropic glutamate receptor activation. For a significant fraction of TRN neurons, synaptic inputs or brief depolarizing current steps led to long-lasting plateau potentials and persistent firing (PF), and in turn, resulted in sustained synaptic inhibition in postsynaptic relay neurons of the ventrobasal thalamus (VB). Pharmacological approachesindicated that plateau potentials were triggered by Ca2+ influx through T-type Ca2+ channels and mediated by Ca2+- and voltage-dependent transient receptor potential melastatin 4 (TRPM4) channels. Together, our results suggest that thalamic circuits can generate slow oscillatory activity, mediated by an interplay of TRN-VB synaptic circuits that generate rhythmicity and TRN cell-intrinsic mechanisms that control PF and oscillation frequency.SIGNIFICANCE STATEMENT Slow forms of thalamocortical rhythmic activity are thought to be essential for memory consolidation during sleep and the efficient removal of potentially toxic metabolites. In vivo, thalamic slow oscillations are regulated by strong bidirectional synaptic pathways linking neocortex and thalamus. Therefore, in vitro studies in the isolated thalamus offer important insights about the ability of individual neurons and local circuits to generate different forms of rhythmic activity. We found that circuits formed by GABAergic neurons in the thalamic reticular nucleus and glutamatergic relay neurons in the ventrobasal thalamus generated slow oscillatory activity, which was accompanied by persistent firing in thalamic reticular nucleus neurons. Our results identify both cell-intrinsic and synaptic mechanisms that mediate slow forms of rhythmic activity in thalamic circuits.

Intrinsic membrane properties of spinal dorsal horn neurones modulate nociceptive information processingin vivo

The dorsal horn of the spinal cord is the first central relay where nociceptive inputs are processed. Based on the expression and modulation of intrinsic electrophysiological properties in in vitro slice preparations, dorsal horn neurones (DHNs) display different discharge patterns (tonic, plateau or rhythmic), which shape the neurone's response to sensory inputs. However, it is unclear whether intrinsic properties play any role in sensory processing in vivo. Using in vivo patch clamp recordings in the adult rat, we here examine whether these intrinsic properties are present, and to what extent they determine the DHN response to natural stimulation. We focused primarily on wide dynamic range neurones in deep laminae. These cells displayed a multicomponent peripheral receptive field, comprising an excitatory firing zone, a low-probability firing fringe, and adjacent inhibitory zones. Deep DHNs presented similar intrinsic properties to those observed in vitro, including plateau potentials. These plateaus, underlying high frequency accelerating discharges and after-discharges, were triggered by mechanical stimulation of the excitatory receptive field. Persistent activities induced by activation of plateau potentials were interrupted by stimulation of peripheral inhibitory zones. Moreover, we show that plateau activation is necessary for the expression of windup in response to repetitive, nociceptive stimulation. Finally, using the spinal nerve ligation model of neuropathy, we demonstrate a significant increase in the proportion of plateau neurones in deep dorsal laminae. Our data, therefore, establish that intrinsic amplification properties are expressed within intact spinal circuits and suggest their involvement in neuropathy-induced hyperexcitability of deep DHNs.

Diencephalic Locomotor Region in the Lamprey—Afferents and Efferent Control

In vertebrates, locomotion can be initiated by stimulation of the diencephalic locomotor region (DLR). Little is known of the different forebrain regions that provide input to the neurons in DLR. In the lamprey, it had been shown previously that DLR provides monosynaptic input to reticulospinal neurons, which in turn elicit rhythmic ventral root activity at the spinal level. To show that actual locomotor movements are produced from DLR, we use a semi-intact preparation in which the brain stem is exposed and the head fixed, while the body is left to generate actual swimming movements. DLR stimulation induced symmetric locomotor movements with an undulatory wave transmitted along the body. To explore if DLR is under tonic GABAergic input under resting conditions, as in mammals, GABAergic antagonists and agonists were locally administered into DLR. Injections of GABA agonists inhibited locomotion, whereas GABA antagonists facilitated the induction of locomotion. These findings suggest that GABAergic projections provide tonic inhibition that once turned off can release locomotion. Double-labeling experiments were carried out to identify GABAergic projections to the DLR. Populations of GABAergic projection neurons to DLR originated in the caudoventral portion of the medial pallium, the lateral and dorsal pallium, and the striatal area. These different GABAergic projection neurons, which also project to other brain stem motor centers, may represent the basal ganglia output to DLR. Moreover, electrical stimulation of striatum induced long-lasting plateau potentials in reticulospinal cells and associated locomotor episodes dependent on DLR being intact, suggesting that striatum may act via the basal ganglia output identified here.

Dendritic Calcium Plateau Potentials Modulate Input–Output Properties of Juxtaglomerular Cells in the Rat Olfactory Bulb

Understanding the intrinsic membrane properties of juxtaglomerular (JG) cells is a necessary step toward understanding the neural basis of olfactory signal processing within the glomeruli. We used patch-clamp recordings and two-photon Ca(2+) imaging in rat olfactory bulb slices to analyze a long-lasting plateau potential generated in JG cells and characterize its functional input-output roles in the glomerular network. The plateau potentials were initially generated by dendritic calcium channels. Bath application of Ni(2+) (250 microM to 1 mM) totally blocked the plateau potential. A local puff of Ni(2+) on JG cell dendrites, but not on the soma, blocked the plateau potentials, indicating the critical contribution of dendritic Ca(2+) channels. Imaging studies with two-photon microscopy showed that a dendritic Ca(2+) increase was always correlated with a dendritic but not a somatic plateau potential. The dendritic Ca(2+) conductance contributed to boosting the initial excitatory postsynaptic potentials (EPSPs) to produce the plateau potential that shunted and reduced the amplitudes of the following EPSPs. This enables the JG cells to act as low-pass filters to convert high-frequency inputs to low-frequency outputs. The low frequency (2.6 +/- 0.8 Hz) of rhythmic plateau potentials appeared to be determined by the intrinsic membrane properties of the JG cell. These properties of the plateau potential may enable JG cells to serve as pacemaker neurons in the synchronization and oscillation of the glomerular network.

Spontaneous voltage oscillations in striatal projection neurons in a rat corticostriatal slice.

In a rat corticostriatal slice, brief, suprathreshold, repetitive cortical stimulation evoked long-lasting plateau potentials in neostriatal neurons. Plateau potentials were often followed by spontaneous voltage transitions between two preferred membrane potentials. While the induction of plateau potentials was disrupted by non-NMDA and NMDA glutamate receptor antagonists, the maintenance of spontaneous voltage transitions was only blocked by NMDA receptor and L-type Ca2+ channel antagonists. The frequency and duration of depolarized events, resembling up-states described in vivo, were increased by NMDA and L-type Ca2+ channel agonists as well as by GABAA receptor and K+ channel antagonists. NMDA created a region of negative slope conductance and a positive slope crossing indicative of membrane bistability in the current-voltage relationship. NMDA-induced bistability was partially blocked by L-type Ca2+ channel antagonists. Although evoked by synaptic stimulation, plateau potentials and voltage oscillations could not be evoked by somatic current injection--suggesting a dendritic origin. These data show that NMDA and L-type Ca2+ conductances of spiny neurons are capable of rendering them bistable. This may help to support prolonged depolarizations and voltage oscillations under certain conditions.

Calcium-dependent plateau potentials in rostral ambiguus neurons in the newborn mouse brain stem in vitro.

Calcium-dependent plateau potentials in rostral ambiguus neurons in the newborn mouse brain stem in vitro. J. Neurophysiol. 78: 2483-2492, 1997. The nucleus ambiguus contains vagal and glossopharyngeal motoneurons and preganglionic neurons involved in respiration, swallowing, vocalization, and control of heart beat. Here we show that the rostral compact formation's ambiguus neurons, which control the esophageal phase of swallowing, display calcium-dependent plateau potentials in response to tetanic orthodromic stimulation or current injection. Whole cell recordings were made from visualized neurons in the rostral nucleus ambiguus using a slice preparation from the newborn mouse. Biocytin-labeling revealed dendritic trees with pronounced rostrocaudal orientations confined to the nucleus ambiguus, a morphological profile matching that of vagal motoneurons projecting to the esophagus. Single-stimulus orthodromic activation, using an electrode placed in the dorsomedial slice near the nucleus tractus solitarius, evoked single excitatory postsynaptic potentials (EPSPs) or short trains of EPSPs (500 ms to 1 s). However, tetanic stimulation (5 pulses, 10 Hz) induced voltage-dependent afterdepolarizations or long-lasting plateau potentials (>1 min) with a constant firing pattern. Depolarizing or hyperpolarizing current pulses elicited voltage-dependent afterdepolarizations or plateau potentials lasting a few seconds to several minutes. Constant spike activity accompanied the long-lasting plateau potentials, which ended spontaneously or could be terminated by weak hyperpolarizing current pulses. Current-induced afterdepolarizations and plateau potentials were dependent on extracellular and intracellular Ca2+, as they were blocked completely by extracellular Co2+, Cd2+, or intracellular bis-(o-aminophenoxy)-N,N,N',N'-tetraacetic acid (BAPTA). Orthodromically induced afterdepolarizations and plateau potentials were blocked by intracellular BAPTA. Afterdepolarizations and plateau potentials were completely blocked by substitution of extracellular Na+ with choline. Afterdepolarizations persisted in tetrodotoxin. We conclude that rostral ambiguus neurons have a Ca2+-activated inward current carried by Na+. Synaptic activation of this conductance may generate prolonged spike activity in these neurons during the esophageal phase of swallowing.

Cholinergic-Dependent Plateau Potential in Hippocampal CA1 Pyramidal Neurons

Cholinergic stimulation of the hippocampal formation results in excitation and/or seizure. We report here, using whole-cell patch-clamp techniques in the hippocampal slice (34-35 degrees C), a cholinergic-dependent slow afterdepolarization (sADP) and long-lasting plateau potential (PP). In the presence of 20 microM carbachol, action potential firing evoked by weak intracellular current injection elicited an sADP that lasted several seconds. Increased spike firing evoked by stronger depolarizing stimuli resulted in long-duration PPs maintained close to -20 mV. Removal of either Na+ or Ca2+ from the external media, intracellular Ca2+ ([Ca2+]i) chelation with 10 mM bis(2-aminophenoxy)ethane-N,N,N',N'-tetra-acetic acid, or the addition of 100 microM Cd2+ to the perfusate abolished both the sADP and PP. The sADP was depressed and the PP was abolished by either 10 microM nimodipine or 1 microM omega-conotoxin, whereas 1.2 microM tetrodotoxin was ineffective. The involvement of a Na+/Ca2+ exchanger was minimal because both the sADP and PP persisted after equimolar substitution of 50 mM Li+ for Na+ in the external media or reduction of the bath temperature to 25 degrees C. Finally in the absence of carbachol the sADP and PP could not be evoked when K+ channels were suppressed, suggesting that depression of K+ conductances alone was not sufficient to unmask the conductance. Based on these data, we propose that a Ca2+-activated nonselective cation conductance was directly enhanced by muscarinic stimulation. The sADP, therefore, represents activation of this conductance by residual [Ca2+]i, whereas the PP represents a novel regenerative event involving the interplay between high-voltage-activated Ca2+ channels and the Ca2+-activated nonselective cation conductance. This latter mechanism may contribute significantly to ictal depolarizations observed during cholinergic-induced seizures.

Morphological and electrophysiological aspects of dorsal median paired neurons generating plateau action potentials in the terminal abdominal ganglion of the insect central nervous system

Among the three clusters of dorsal unpaired median neurons of the Periplaneta americana terminal abdominal ganglion, another type of neuron has been characterized by anterograde cobalt stainings and microelectrode technique. These neurons are bilaterally distributed in the ganglion. Their axons ipsilaterally exit the ganglion via the anterior proctodeal nerves, to innervate the proctodeum. They are characterized by a long-duration overshooting action potentials and a low firing frequency. Most often the depolarizing phase is composed of two peaks: a fast spike followed by a slow phase. Tetrodotoxin suppressed the fast peak and blocked the spontaneous activity suggesting that sodium channels are involved in the depolarizing phase as well as in the initiation of the action potential. Calcium channel blockers induced a disappearing of the slow depolarizing phase indicating the participation of calcium ions and a reduction of the afterhyperpolarization reflecting the participation of calcium-activated potassium channels. Furthermore, cadmium, as lanthanum or barium, induced a long-lasting plateau potential, which would be due to a persistent sodium conductance. Tetraethylammonium increased the duration of the action potential indicating that potassium channels are implicated in the falling phase. The results demonstrate that these neurons are different from other cells, especially dorsal unpaired median neurons, of the central nervous system of the cockroach.

Alteration of bursting properties in interneurons during locust flight.

1. The contribution of bursting properties to the generation of the flight motor pattern was examined for two identified interneurons (interneurons 566 and 567) in the flight system of the locust Locusta migratoria by means of intracellular recording and stimulation techniques. These interneurons are important elements in transmitting proprioceptive information from the hindwing tegula to wing elevator motoneurons. 2. Offset currents injected into these neurons revealed that bursts are triggered in the intact flying animal by synaptic input from tegula afferents (n = 10). These bursts lead to an amplification of proprioceptive input that is crucial for the generation of the intact flight motor pattern. In the absence of afferent input the activity of these neurons remained subthreshold for triggering a burst. This explains why these neurons exhibit only weak rhythmic oscillations in deafferented animals. 3. The property of interneuron 566 to burst was conditional, always being expressed during flight (n = 14) and occurring only occasionally in the quiescent animal. In the absence of flight, stimulation of tegula afferents never evoked bursts in interneuron 566 (n = 7) and depolarizing current pulses evoked weak bursts in only three of nine preparations. In 2 of 14 animals, bursting property of interneuron 566 was enhanced just after the termination of flight. 4. Variability in the bursting property was also found for interneuron 567. In the quiescent animal, tegula-evoked compound excitatory postsynaptic potentials were not sufficient to trigger bursts (n = 3) but depolarizing current pulses evoked always weak rhythmic bursting activity (n = 4). This bursting property was also variable and in one animal we found long-lasting plateau potentials that could be evoked by current injection after flight was elicited several times. 5. The data presented demonstrate that the capacity to burst is conditional in the interneurons 566 and 567. Bursting properties are always induced during flight and function to amplify proprioceptive pathways that are important for the generation of the intact flight motor pattern.

Gastric mill activity in the lobster. III. Effects of proctolin on the isolated central pattern generator.

1. The response of the isolated gastric central pattern generator (CPG) to bath application of proctolin is characterized and compared with the previously analyzed behavioral response. 2. Proctolin had an excitatory effect on the ongoing spontaneous rhythm of "combined" preparations, in which the stomatogastric ganglion (STG) is connected to the esophageal and commissural ganglia by the stomatogastric nerve (STN). The effect started between 20 s and 5 min and was characterized by strongly increased burst durations as well as increased spike rates in all units except the two lateral posterior gastric (LPG) motoneurons. The effect was strongest in the dorsal gastric (DG) and lateral gastric (LG) motoneurons and was accompanied by a phase change of the DG burst. DG continued spiking throughout large parts of the burst of LG and of the gastric mill (GM) motoneurons, which are antagonists of DG. 3. The threshold concentration was approximately 10(-10) M, and the effects were dose dependent and reversible. 4. LG and DG were identified as target cells for the action of proctolin. In LG regenerative plateau properties were induced, as revealed by its long-lasting plateau potentials, sensitivity for triggering inputs, and the occurrence of oscillatory prepotentials. An induction of endogenous bursting in DG was concluded from preparations, in which DG was cycling alone or bursting with a much shorter period duration than other gastric neurons. Hyperpolarization of DG, which normally has no or weak driving power within the gastric network, demonstrated that under the influence of proctolin, firing of DG can accelerate the gastric rhythm from a 27- to a 9-s period duration. 5. Proctolin does not only have a modulatory influence on an ongoing rhythm, but it also can trigger gastric activity. This function was first concluded from proctolin-treated STGs, which, unlike normal preparations, continue bursting if inputs via the STN are blocked. Finally, triggering was demonstrated directly, since isolated STGs that were not oscillating started a gastric rhythm after 20-30 min of perfusion with proctolin. 6. The proctolin-induced changes of the CPG activity in isolated preparations are in agreement with the effect on gastric mill chewing in the intact animal, in which, depending on the dose, different modes of chewing could be elicited.

A ouabain-sensitive hyperpolarization in rat striatal neurones in vitro

Intracellular recordings were made from rat striatal neurones in vitro. In the presence of intracellular caesium and extracellular tetraethylammonium chloride (TEA) (5 mM) and barium (3 mM), long-lasting plateau potentials developed followed by a prominent voltage independent hyperpolarization which lasted several seconds. A similar afterhyperpolarization was observed when calcium was replaced by barium. The afterhyperpolarization was reduced in a potassium free medium and reversibly abolished in a Na +-free solution or by cooling the slice to 21–24°C. It was also irreversibly blocked by ouabain (50 μM). This hyperpolarization may therefore result from the activation of a Na +,K +-ATPase electrogenic pump.

An inward calcium current underlying regenerative calcium potentials in rat striatal neurons in vitro enhanced by BAY K 8644

The single electrode voltage clamp technique was used to characterize the currents underlying the calcium potentials in rat caudate neurons in vitro. In current clamp experiments, long depolarizing current pulses evoked repetitive firing of fast somatic action potentials. These were abolished by tetrodotoxin (1 μM) and replaced by slow graded depolarizing potentials. These were preceded by a transient hyperpolarizing notch. Addition of 4-aminopyridine (100 μM) abolished the hyperpolarizing notch, enhanced the slow graded depolarizing response and induced the appearance of a slow all-or-nothing action potential. Both the slow graded response and the all-or-nothing action potential were abolished by cobalt (2 mM), suggesting the involvement of voltage-dependent calcium conductances. When the neurons were loaded intracellularly with caesium the action potential duration increased. Substitution of the extracellular calcium by barium (1–3 mM) or external addition of tetraethylammonium (5 mM) further prolonged spike duration and induced the appearance of long-lasting plateau potentials. These were insensitive to tetrodotoxin and were reversibly blocked by the calcium antagonists cobalt (2 mM), manganese (2 mM) or cadmium (500 μM). The calcium potentials were enhanced by the calcium ‘agonist’ BAY K 8644 (1–5 μM). In voltage clamp experiments when intracellular caesium was used to reduce outward currents and tetrodotoxin to block fast regenerative sodium currents, depolarizing voltage steps from a holding potential of −50, −40 mV activated an inward current. This current peaked in 50–80 ms and inactivated in two phases: an initial one at 150–200 ms followed by a second one after several hundred ms. The current was still inward at the end of the command step and often was followed by an inward tail current. The current/voltage curve was N-shaped with a region of negative slope conductance between −25 and −10 mV. The inward current was larger in the presence of barium (1–3 mM) and tetraethylammonium (5 mM). It was abolished by the calcium antagonists cobalt (2 mM) and cadmium (500 μM) and enhanced by the calcium ‘agonist’ BAY K 8644 (5 μM). In conclusion, like many other central neurons, striatal neurons bear calcium conductances that underlie the calcium potentials observed in current clamp experiments.

Long-term depression of parallel fibre synapses following stimulation of climbing fibres

The results from several investigations suggest that climbing fibres heterosynaptically depress parallel fibre responses in Purkinje cells. In the present investigation the mechanism behind the depression has been studied by extracellular recording of responses in single Purkinje cells, evoked by electrical stimulation of parallel fibres and climbing fibres. The results show that a short time of conjunctive stimulation of climbing fibres and parallel fibres results in a long-lasting depression of the parallel fibre responses and that this depression can be prevented if the Purkinje cells are inhibited during the conjunctive stimulation. Since inhibition has been shown to shorten or abolish the long-lasting plateau potentials which are evoked by climbing fibre impulses this finding supports the assumption that the climbing fibre evoked plateau potentials mediate the heterosynaptic depression of parallel fibre responses.

Calcium depletion produces Na+-dependent, sustained depolarizations of Retzius cell membranes in the leech CNS

1. Ca++ free/EGTA Ringer produces an abrupt change in the Retzius cell (RZ) membrane conductance resulting in a sustained depolarization: a plateau potential. 2. The long-lasting plateau potential is a consequence of Ca++ depletion and not a specific effect of EGTA. 3. The amplitude of this increased conductance plateau depends upon the external concentration of Na+. 4. Mn++ and La+++ completely block Ca++ depletion plateaus while Sr++ and Mg++ decrease plateau duration. Co++ has effects which are intermediate between these two cation groups. 5. We suggest that RZ membranes have a repolarizing potassium conductance which depends primarily upon Ca++ conductance and not membrane potential.
