Investigation of the structure of silk film regenerated with lithium thiocyanate solution

AbstractThe Silk fibers of the mulberry (Bombyx Mori) were dissolved in a 70% lithium thiocyanate solution. Dissolved silk was regenerated by casting the films from the solution after dialyzing. The films were investigated by infrared (IR) spectroscopy and x‐ray diffraction analysis. It was found that the freshly prepared film was amorphous. The transformation to the β‐form could be brought about by heating, solvent induced crystallization, ultraviolet (UV) radiation, and prolonged storage. The mechanism of this transformation is discussed.

Formation of Cross‐links in Silk by Dyeing with a Difluorochloropyrimidyl Dye

The dyeing of silk using a monoazo difluorochloropyrimidyl reactive dye has been studied with respect to exhaustion, fixation ratio, solubility of the dyed fibres in lithium thiocyanate solution, work recovery and tensile strain recovery. The results indicate the formation of cross‐links between the reactive group by reaction of the two fluorine atoms in the dye with functional groups in the silk fibroin, probably mainly tyrosine OH groups.

Anion solvation I. Solvation of the thiocyanate anion in aprotic solvents by near infrared spectroscopy

Solutions of tetrabutylammonium and lithium thiocyanates have been examined in the infrared 2000–5500 cm−1 region at room temperature. New bands have been observed which can be assigned to simultaneous transitions between the v (CN) vibrations of the anion and the v (CH) vibrations of the solvent CH3 group. These transitions which are independent of the state of association (free ion, ionpairs, dimers, triple cation) provide evidence of anion solvation by aprotic solvents.

Anion solvation II. Solvation of thiocyanate and halide anions in mixtures of protic and aprotic solvents

Infrared and Raman spectra of tetraalkylammonium and lithium thiocyanates and bromides in protic-aprotic solvent mixtures have been recorded in the 2100–2300 cm−1 region at room temperature. It is shown that the anion is hydrogen bonded to the protic solvent, whether free or linked to a cation. Models are given for the solvated ion-pair taking into account both anion and cation solvation.

NMR studies of chitin and chitin derivatives

AbstractThe 13C NMR spectra of N‐acetyl‐D‐glucosamine (1) and chitobiose (2), monomer and dimer of chitin, were assigned and used to assign the 13C NMR spectra of chitin in lithium thiocyanate and in LiCl/dimethylacetamide solutions. The dissolution of chitin in formic acid was studied. IR, 13C and 1H NMR spectra show that dissolution occurs by a reaction of formic acid with chitin with the formation of formyl chitin as a statistical derivative of chitin. 13C NMR spectra of diformyl‐ and diacetylchitin were also assigned to characterize these regular polymers.

Dielectric and ultrasonic relaxation of lithium thiocyanate and lithium perchlorate in dimethoxymethane

Ultrasonic absorption at radio frequencies, complex dielectric permittivities at microwave frequencies, electrical conductances, and infrared spectra of solutions of LiSCN and LiClO/sub 4/ in dimethoxymethane are reported. The conductance data, analyzed in the concentration range 7 X 10/sup 14/ to 10/sup -2/ M, indicate that the major species are neutral pairs of ions although a significant amount of triple ions are present. Band analysis of the ir spectra of solutions with concentrations equal and above 0.05 M reveal the presence of three major species, ion-pairs quadrupoles and chains, the latter with Li/sup +/ bound also to the S-end of the SCN ion. The dielectric spectra indicate that there are three dipolar species present, the solvent and two solute species, in accord with information from the ir spectra. Alternate analysis of the dielectric spectra in terms of a Cole-Davidson distribution does not seem to be consistent with the ir findings. Consequently a possible theoretical interpretation of the Cole-Davidson spectrum (based on our earlier proposed mechanism of collisional diffusion and rotation of ion pairs) is considered as unlikely to be relevant. The same applies to recent improvements on our proposal based on a three-dimensional diffusion model. With the aid of the structuralmore » information above, the ultrasonic spectra at c approx. 0.1 M for both LiSCN and LiClO/sub 4/ are interpreted as due to an ion-pair-quadrupole equilibrium involving cation desolvation. At higher concentrations a new relaxation process centered at approx. 10 MHz appears.« less

Vibrational study of ionic association in aprotic solvents—II. Structure and force constants of the lithium thiocyanate dimer in ether solutions

Lithium thiocyanate is dimerized in ethers (Et 2 O, Bu 2 O, dimethoxy and diethoxymethane). The dimer is centrosymmetric. An important decrease in the force constant F CN and an increase in F CS are observed with respect to SCN − LiNCS. Hence, the only possible structure involves two lithium bridges between the two nitrogen atoms. A simple electrostatic model well accounts for the mechanical coupling between the two SCN groups through the lithium bridges.

The molecular mass of solubilized fibroin prepared from silk was analyzed by SDS-PAGE. It was found that the fibroin molecule was degraded during reeling, degumming (removal of sericin), and dissolution of silk threads. A protocol for the preparation of solubilized fibroin conserving its native molecular size is offered, that is, (1) to use only fresh cocoons (not dry cocoons or reeled silk threads), (2) to degum by heating in 8 M aqueous urea, and (3) to dissolve by saturated (∼9 M) aqueous lithium thiocyanate at room temperature.

Light-scattering and infrared-spectrophotometric studies of chitin and chitin derivatives

A sample of chitin isolated from the shell of the crab Scylla serrata had, when in lithium thiocyanate solution, an average, weight-average molecular weight ( 1) of 1.036 x 10 6 daltons, an intrinsic dissymmetry ( 2) of 1.93, and a Z-average radius of gyration ( 3) of 64.14 nm. Carboxymethylchitin and glycol chitin, in 0.5 M sodium chloride, had, respectively, ( 1) 1.896 and 1.819 x 10 6 daltons, ( 2) 3.25 and 4.31. and ( 3) 143.49 and 251.57 nm. They had similar degrees of polymerization, they underwent dissociation as the concentration of sodium chloride was increased to 2.5 M, and the molecular packing of the chains was essentially side-by-side. Chitin in 5.55 M lithium thiocyanate and carboxymethylchitin in 2.5 M sodium chloride had similar degrees of polymerization. It is concluded that a small but significient number of the amino groups in the chitin molecule are not acetylated.

Studies on silk fibroin of the silkworm Bombyx mori

A procedure has been developed to obtain native fibroin in a pure state from the reservoir part of the silk gland. The purified protein has a sedimentation coefficient of 10 S as determined on sucrose density gradients and the amino acid composition is similar to that reported for fibroin from the cocoons. The effects of various solvents has been studied; lithium thiocyanate was found to be the solvent of choice. By in vivo labeling of fibroin with [ 3H]glycine and [ 14C]alanine it was demonstrated that fibroin synthesized in the posterior part of the gland and that stored in the reservoir part are identical.

Electron Microscopic Observations of Adeno-Associated Satellite Virus Type 1 Capsids and Subunits

Adeno-associated satellite virus type 1 was disrupted by lyophilization and subsequent rehydration with sodium dodecyl sulfate (SDS), or by treatment with lithium thiocyanate (LiSCN).The virus subunits, measured on disrupted capsids or individually, have a mean diameter of 4.1–4.6 nm, depending on negative staining conditions. Their relation to the capsid geometry is discussed.

Anion exchange of metal complexes—VI

Abstract The distribution of tracer 63Ni between the strongly basic anion exchange resin, Dowex 1 × 8, and solutions of sodium, potassium, ammonium, and lithium thiocyanate have been determined. No effects attributable to polynucleation were observed with concentrations of tracer up to 2·92 × 10−6 M. Since counting 63Ni, a soft β-emitter, is best accomplished with liquid scintillation techniques, a preliminary investigation was necessary to establish and characterize the best liquid scintillation mixture. The distributions were determined for the 0·05–5·0 M concentration range of the thiocyanate solution. Above 2·60 M, increasing sorbability was in the order NH4+ > K+ > Na+ > Li+, with a maximum at ∼ 0·3 M for Na+ and Li+, and 1·0 M for K+ and NH4+. Corrected distributions were determined by employing resin invasion data; from the corrected distributions, overall complexity parameters relating to the neutral complex were evaluated by curve fitting. Their values were found to be: k +2 ∗ =2·36×10 −3 ; k +1 ∗ =4·68×10 −1 ; k 0 ∗ =1 ( by definition ); k −1 ∗ =2·64; k −2 ∗ =1·89 . Resin sorption effects ∼ 85-fold stabilization of the neutral species over the aqueous phase.

Polysaccharides in germination. Xyloglucans ( amyloids') from the cotyledons of white mustard.

Two xyloglucan fractions have been isolated from the cotyledons of resting white-mustard seeds, the first by extraction with hot EDTA, and the second by subsequent extraction with alkali or lithium thiocyanate. Although both appear to have the ;amyloid' type of structure in which chains of (1-->4)-linked beta-d-glucopyranose residues carry d-xylose-rich side chains through position 6, these side chains are rather different in structure in the two polysaccharide fractions, and the second or ;insoluble' xyloglucan has fewer of them. The side chains in both polysaccharides are also different from those in other seed amyloids, especially in having xylose linked through positions 3 and 4 (instead of through position 2 as usual) and in containing fucose residues. Both polysaccharides show the characteristic blue ;amyloid' colour with iodine in the presence of sodium sulphate, and it is suggested that this arises by the interaction of iodine molecules and possibly iodide ions within the interstices between aggregated xyloglucan chains. ;Soluble' xyloglucan is metabolized during germination and is presumed to have a reserve function. ;Insoluble' xyloglucan is metabolized less completely over the period studied but its lack of turnover during cell-wall differentiation indicates that it also is a reserve. These and other beta-(1-->4)-linked reserve polysaccharides of seeds might also have a structural function which is of particular value for the survival of the dormant seed.

Reactions of the dirhodium cation Rh2 4+, with triphenylphosphine and alkali metal carboxylates, dithiocarbamates and other anions

Rhodium(I) carboxylates of stoicheiometry Rh(OCOR)(PPh3)3, (where R = alkyl, aryl, or substituted alkyl), have been obtained by the interaction of the dirhodium(II) cation, Rh24+, with a stoicheiometric amount of triphenyl-phosphine and an excess of the lithium salt of the appropriate carboxylic acid. The chemical properties of these carboxylates are described, and include reactions with hydrogen, oxygen, carbon monoxide and aldehydes. In benzene solution the RhI carboxylates catalyse the homogeneous hydrogenation of alkenes and alkynes; the rates are compared with those using chlorotris(triphenylphosphine)rhodium(I). The interaction of Rh24+ with triphenylphosphine in presence of sodium diethyldithiocarbamate, mercaptobenzthiazole, pyridine-2-thiol, toluene-3-thiol, diphenylphosphorodithioic acid, diethoxyphosphorodithioic acid and lithium thiocyanate have been studied, and also the interaction of chlorotris(triphenylphosphine)rhodium(I) with tin(II) chloride.
