Objective To investigate the influence and mechanism of proliferation inhibition and apoptosis induced by polypyrimidine tract-binding protein-associated splicing factor (PSF) silence in colon cancer cells.Methods Small interfering RNA (siRNA) that targeted PSF gene (siRNA-PSF) was constructed and transfected into human colon cancer DLD-1 cells,for 48 h to observe the effect of PSF silence on proliferation and apoptosis of DLD-1 cells.Furthermore,real-time fluorescent quantitative polymerase chain reaction (FQ-PCR) and Western blotting were used to detect the change of microtubule-associated protein 1 light chain-3B (LC3B) expression in DLD-1 cells with PSF silence by siRNA-PSF transfection.Results Compared with control cells and cells with siRNA-vector transfection,transfection with siRNA-PSF significantly reduced PSF expression in DLD-1 cells (P < 0.05).At the same time,proliferation of DLD-1 cells was inhibited (luminescence value:25.05 ± 3.94 vs.132.88 ± 8.73 and 127.61 ± 8.02,P<0.05),whereas the apoptosis of DLD-1 cells was obviously increased [apoptosis rate:(54.6 ±5.5) % vs.(8.5 ± 1.3) % and (10.1 ± 2.0) %,P < 0.05].FQ-PCR and Western blotting still revealed that the expression of LC3B was significantly decreased in DLD-1 cells with PSF silence by siRNA-PSF transfection.Conclusion Silence of PSF inhibits proliferation and induces apoptosis of colon cancer cells by prohibiting LC3B expression.
