Theoretical Issues of Creation of Model Municipal Libraries

Creation of model municipal libraries of new generation is carried out within the framework of the National project “Culture” developed in pursuance of the Edict of The President of the Russian Federation № 204 of May 7, 2018. The aim of the project is to find new models of library development that combine cultural, educational, scientific and educational functions. Modernization of municipal libraries is carried out on the basis of principles of modernization of public libraries, recommended by the Model Standard for Public Library Services. The authors consider the library as a system object and on the basis of internal and external relations of the object determine the place, role and functions of library in the communication process. The study is based on the concept of library as a four-element system. The article describes each of its elements in the process of their evolution in the modern information environment. Based on the spatial approach, the authors identify the components and levels of the library space, determine directions of its development and substantiate the principles of its construction.The authors conclude that development of the model of library operation in the territorial information space should reflect the specific features of the formation of its own information space as a documentary-communication institution.

PTO-QuickStep: A Fast and Efficient Method for Cloning Random Mutagenesis Libraries.

QuickStep is a cloning method that allows seamless point integration of a DNA sequence at any position within a target plasmid using only Q5 High-Fidelity DNA Polymerase and DpnI endonuclease. This efficient and cost-effective method consists of two steps: two parallel asymmetric PCRs, followed by a megaprimer-based whole-plasmid amplification. To further simplify the workflow, enhance the efficiency, and increase the uptake of QuickStep, we replaced the asymmetric PCRs with a conventional PCR that uses phosphorothioate (PTO) oligos to generate megaprimers with 3′ overhangs. The ease and speed of PTO-QuickStep were demonstrated through (1) right-first-time cloning of a 1.8 kb gene fragment into a pET vector and (2) creating a random mutagenesis library for directed evolution. Unlike most ligation-free random mutagenesis library creation methods (e.g., megaprimer PCR of whole plasmid [MEGAWHOP]), PTO-QuickStep does not require the gene of interest to be precloned into an expression vector to prepare a random mutagenesis library. Therefore, PTO-QuickStep is a simple, reliable, and robust technique, adding to the ever-expanding molecular toolbox of synthetic biology and expediting protein engineering via directed evolution.

Implementation of tactical maneuvers with maneuver libraries

This study creates and combines the general maneuver libraries for fixed-wing aircraft to implement tactical maneuvers. First, the generalized maneuver libraries are established by analyzing the characteristics of tactical maneuvers required in battlefields. The 7th order polynomial is applied to both the creation of the maneuver libraries and the generation of the trajectories or flight paths for modal inputs. To track the desired trajectory, we design the Attitude Command Attitude Hold (ACAH) system and the Rate Command Rate Hold (RCRH) system using Model Following Controller (MFC). Moreover, the Line-of-Sight (LOS) guidance law is designed. In particular, the CONDUIT® is employed to optimize the gains so that the control systems meet the aircraft Handling Qualities (HQ) criteria. Finally, flight simulations are performed for the longitudinal loop, immelmann-turn, and climb-slalom-descent maneuvers to verify that tactical aggressive maneuvers are realizable via the combination of maneuver libraries. This study can contribute to the development of flight techniques for aircraft tactical maneuvers and to the revision of air force operational manuals.

To the 90th Birth Anniversary of E.A. Fenelonov, the Librarian, Leader, Scientist

The article is devoted to the 90th birth anniversary of E.A. Fenelonov, the prominent figure of library construction, scientist-librarian and the Honored worker of culture of the RSFSR. The authors present his biographical data, data on the most important stages of his life and activity: study at the Moscow State Library Institute, postgraduate study, defence of the dissertation thesis “Organization of library service for rural population”, leadership of the newly organized library of the Lipetsk region; work as the Head of Department (then Division) of libraries of the Ministry of Culture of RSFSR, the Head of the Main information-computing centre for culture and arts of the Ministry of Culture of RSFSR, the Deputy Director for scientific and methodical work of the V.I. Lenin State Library of the USSR (nowadays — the Russian State Library, RSL), the research scientist in the Research Department of the RSL. The paper describes the general characteristic features of his administrative activity and scientific creativity. The authors emphasize his role in the centralization of the library network, creation of depository libraries, introduction of open access and computers in the librarianship, participation in the development of the “Regulations on Librarianship in the USSR” (1984), the laws “On Librarianship”, “On the Legal Deposit Copy of Documents”, etc. The paper considers the main works of E.A. Fenelonov, dedicated to the management of librarianship, comprehensive planning of library construction, economic efficiency of libraries, organization of the library network, library zoning, activities of the centralized library systems, methodical work of libraries. The authors reveal the personality of E.A. Fenelonov as organizer, manager, scientist and person.

Product Data Management for Sustainability: An Interoperable Approach for Sharing Product Data in a BIM Environment

Several research and development projects in the construction industry show the need of coherent product libraries to ease information sharing among involved stakeholders. In these projects, designers are often the main actors addressed in the creation and fruition of such product libraries. Besides designers, the presented research addresses the needs of manufacturers, clients and users. To achieve this coherence of product libraries, there are issues such as standardisation of product data templates, numerosity and heterogeneity of products categories, interoperability in sharing data and scarce thrust of manufacturers in developing extensive libraries. Considering the European directives and regulations, manufacturers are required to provide detailed and structured information about their products, e.g. in the form of CE marks and/or Declaration of Performance (DOP). Hence, developing data structures that are already compliant with CE marks and DOPs information requirements improves manufacturers’ capability of promoting their products. Moreover, this makes clients and users aware of provided information, giving them the possibility of comparing performances of different products and consequently choosing only those products that fit better their needs. The aim of the here described research activity is twofold: defining a common data structure for product data and using existing technologies to share these data. The presented research proposes a conceptual model for providing such structure for product data to facilitate Product Data Management (PDM). This structure relies on standardised Product Data Templates (PDT) and Product Data Sheets (PDS) for enhancing the sharing process within to a Building Information Modelling (BIM) environment. Moreover, an eXtensible Markup Language (XML) structure has been developed for transferring product data allowing a machine-readable exchange of information. The result of the described research is the definition of information requirements for developing an online product library that permits a user-friendly consultation by stakeholders and an automated extraction of data by software.

Mass Spectrometry Data Repository Enhances Novel Metabolite Discoveries with Advances in Computational Metabolomics.

Mass spectrometry raw data repositories, including Metabolomics Workbench and MetaboLights, have contributed to increased transparency in metabolomics studies and the discovery of novel insights in biology by reanalysis with updated computational metabolomics tools. Herein, we reanalyzed the previously published lipidomics data from nine algal species, resulting in the annotation of 1437 lipids achieving a 40% increase in annotation compared to the previous results. Specifically, diacylglyceryl-carboxyhydroxy-methylcholine (DGCC) in Pavlova lutheri and Pleurochrysis carterae, glucuronosyldiacylglycerol (GlcADG) in Euglena gracilis, and P. carterae, phosphatidylmethanol (PMeOH) in E. gracilis, and several oxidized phospholipids (oxidized phosphatidylcholine, OxPC; phosphatidylethanolamine, OxPE; phosphatidylglycerol, OxPG; phosphatidylinositol, OxPI) in Chlorella variabilis were newly characterized with the enriched lipid spectral databases. Moreover, we integrated the data from untargeted and targeted analyses from data independent tandem mass spectrometry (DIA-MS/MS) acquisition, specifically the sequential window acquisition of all theoretical fragment-ion MS/MS (SWATH-MS/MS) spectra, to increase the lipidomic annotation coverage. After the creation of a global library of precursor and diagnostic ions of lipids by the MS-DIAL untargeted analysis, the co-eluted DIA-MS/MS spectra were resolved in MRMPROBS targeted analysis by tracing the specific product ions involved in acyl chain compositions. Our results indicated that the metabolite quantifications based on DIA-MS/MS chromatograms were somewhat inferior to the MS1-centric quantifications, while the annotation coverage outperformed those of the untargeted analysis of the data dependent and DIA-MS/MS data. Consequently, integrated analyses of untargeted and targeted approaches are necessary to extract the maximum amount of metabolome information, and our results showcase the value of data repositories for the discovery of novel insights in lipid biology.

Em Portugal, durante a I República (1910-1926), os políticos consideravam essencial a formação escolar das gerações mais novas, uma vez que seriam estas a dar continuidade ao espírito revolucionário. Nesta época surgiram novos escritores, jornais e coleções infantis e, ainda que a maioria dos livros fosse de caráter educativo, verifica-se um novo espírito na escrita para crianças. Era necessário substituir o dever de obediência pelo sentimento cívico e pela ideia de responsabilidade de cada cidadão no progresso do país. Daí a introdução de novos temas nos livros para crianças como a História e/ou a Geografia do país, de que são testemunhos as obras dos grandes autores da época como Ana de Castro Osório, Virgínia de Castro e Almeida, Aquilino Ribeiro, António Sérgio, Fernanda de Castro e Jaime Cortesão. Na ilustração destaque para alguns dos mais exímios artistas portugueses da época, como as irmãs Mamia Roque Gameiro e Raquel Roque Gameiro, Alonso, Leal da Câmara e Milly Possoz, Sarah Afonso, entre outros. A preocupação com a alfabetização, a ampliação da rede escolar e a criação de bibliotecas, fez com que, a pouco e pouco, a leitura passasse a tomar o seu devido lugar.

Accurate Measurement of Characteristic Response for Unexploded Ordnance With Transient Electromagnetic System

Characteristic response parameterized in time-dependent target intrinsic polarizabilities has been widely used to create feature libraries for unexploded ordnance (UXO) discrimination. This paper discusses the accurate measurement of characteristic response for UXOs with transient electromagnetic (TEM) system. First, a specially designed TEM system, which utilizes a modified solenoid and a pair of rectangular coils as transmitting coils to ensure the uniformity of primary field up to 96% throughout the target, is described. Then, the characteristic responses of 12 UXOs are measured from directions vertical or parallel to the axis at distances from 40 to 120 cm. The measured characteristic responses show a power law decay function at an early time and an exponential decay function at a later time. The characteristic responses parallel to the target axis are larger, and the decay is considerably slower than those vertical to the axis, leading to one to two orders of magnitude difference at a later time. Results also show that the characteristic response measured from the direction vertical to the axis of the target is more precise than that from the direction parallel to the axis at the same distance. The portion of the target that is close to the system accounts for a large proportion of the response. A target can be approximated as a single dipole only when it is located no less than twice its length from the system. Finally, the characteristic responses of 12 targets are parameterized with the Pasion–Oldenburg expression for feature library creation.

Electronic Library of the RSL: Development Stages and Features of Formation of Digital Collections

The article for the first time covers the history of the Electronic Library (EL) of the Russian State Library (RSL) from the beginning of its formation in the mid-1990s to the present. The author considers the stages of formation and development of the EL of the RSL, the changes that have occurred depending on various factors: changes in legislation in the field of intellectual activity, the emergence of new conceptual approaches and technological platforms. The article identifies two basic approaches to the creation of electronic library: formation of large electronic collections by type of publications or presented information and the creation of electronic version of the collections of documents stored in the RSL holdings. The author gives the key decisions made in relation to the EL: the resources that have passed into the public domain are freely available on the website of the RSL; all documents (not fragments) digitized on the basis of the RSL holdings shall be included in the EL of the RSL; the quality of electronic materials is strictly regulated, etc. The article presents the history of creation and features of large digital collections and separate collections, as well as participation in Russian and international projects and programs. The author considers the development of various electronic services, search capabilities, and features of access to different collections depending on the content. The article highlights the role of the “Electronic Library of Dissertations”, which became the basis for enriching the EL of the RSL with relevant scientific knowledge, for working out new technical and technological solutions, and for providing unique access opportunities within the current legislation. The author notes that the EL creators are focused on the organization of access to modern scientific and educational content within the framework of legislative initiatives, conceptual and special technical solutions, as well as on providing free access to materials of great cultural and historical value for Russia and the world community

Enhancement of cellulase production in Trichoderma reesei RUT-C30 by comparative genomic screening

BackgroundCellulolytic enzymes produced by the filamentous fungus Trichoderma reesei are commonly used in biomass conversion. The high cost of cellulase is still a significant challenge to commercial biofuel production. Improving cellulase production in T. reesei for application in the cellulosic biorefinery setting is an urgent priority.ResultsTrichoderma reesei hyper-cellulolytic mutant SS-II derived from the T. reesei NG14 strain exhibited faster growth rate and more efficient lignocellulosic biomass degradation than those of RUT-C30, another hyper-cellulolytic strain derived from NG14. To identify any genetic changes that occurred in SS-II, we sequenced its genome using Illumina MiSeq. In total, 184 single nucleotide polymorphisms and 40 insertions and deletions were identified. SS-II sequencing revealed 107 novel mutations and a full-length wild-type carbon catabolite repressor 1 gene (cre1). To combine the mutations of RUT-C30 and SS-II, the sequence of one confirmed beneficial mutation in RUT-C30, cre196, was introduced in SS-II to replace full-length cre1, forming the mutant SS-II-cre196. The total cellulase production of SS-II-cre196 was decreased owing to the limited growth of SS-II-cre196. In contrast, 57 genes mutated only in SS-II were selected and knocked out in RUT-C30. Of these, 31 were involved in T. reesei growth or cellulase production. Cellulase activity was significantly increased in five deletion strains compared with that in two starter strains, RUT-C30 and SS-II. Cellulase production of T. reesei Δ108642 and Δ56839 was significantly increased by 83.7% and 70.1%, respectively, compared with that of RUT-C30. The amount of glucose released from pretreated corn stover hydrolyzed by the crude enzyme from Δ108642 increased by 11.9%.ConclusionsThe positive attribute confirmed in one cellulase hyper-producing strain does not always work efficiently in another cellulase hyper-producing strain, owing to the differences in genetic background. Genome re-sequencing revealed novel mutations that might affect cellulase production and other pathways indirectly related to cellulase formation. Our strategy of combining the mutations of two strains successfully identified a number of interesting phenotypes associated with cellulase production. These findings will contribute to the creation of a gene library that can be used to investigate the involvement of various genes in the regulation of cellulase production.

Multicomponent Reaction Toolbox for Peptide Macrocyclization and Stapling

In the past decade, multicomponent reactions have experienced a renaissance as powerful peptide macrocyclization tools enabling the rapid creation of skeletal complexity and diversity with low synthetic cost. This review provides both a historical and modern overview of the development of the peptide multicomponent macrocyclization as a strategy capable to compete with the classic peptide cyclization methods in terms of chemical efficiency and synthetic scope. We prove that the utilization of multicomponent reactions for cyclizing peptides by either their termini or side chains provides a key advantage over those more established methods; that is, the possibility to explore the cyclic peptide chemotype space not only at the amino acid sequence but also at the ring-forming moiety. Owing to its multicomponent nature, this type of peptide cyclization process is well-suited to generate diversity at both the endo- and exo-cyclic fragments formed during the ring-closing step, which stands as a distinctive and useful characteristic for the creation and screening of cyclic peptide libraries. Examples of the novel multicomponent peptide stapling approach and heterocycle ring-forming macrocyclizations are included, along with multicomponent methods incorporating macrocyclization handles and the one-pot syntheses of macromulticyclic peptide cages. Interesting applications of this strategy in the field of drug discovery and chemical biology are provided.

Isolation of novel quorum-sensing active bacteria from microbial mats in Shark Bay Australia.

Quorum sensing is a potent system of genetic control allowing phenotypes to be coordinated across localized communities. In this study, quorum sensing systems in Shark Bay microbial mats were delineated using a targeted approach analyzing whole mat extractions as well as the creation of an isolate library. A library of 165 isolates from different mat types were screened using the AHL biosensor E. coli MT102. Based on sequence identity 30 unique isolates belonging to Proteobacteria, Actinobacteria and Firmicutes were found to activate the AHL biosensor, suggesting AHLs or analogous compounds were potentially present. Several of the isolates have not been shown previously to produce signal molecules, particularly the members of the Actinobacteria and Firmicutes phyla including Virgibacillus, Halobacillius, Microbacterium and Brevibacterium. These active isolates were further screened using thin-layer chromatography (TLC) providing putative identities of AHL molecules present within the mat communities. Nine isolates were capable of producing several spots of varying sizes after TLC separation, suggesting the presence of multiple signalling molecules. This study is the first to delineate AHL-based signalling in the microbial mats of Shark Bay, and suggests quorum sensing may play a role in the ecosphysiological coordination of complex phenotypes across microbial mat communities.

Creation and characterisation of probiotic libraries for use in poultry and pigs

Probiotics are defined as live micro-organisms which provide the host with a growth or health advantage. In recent year’s with the emergence of antimicrobial resistance, probiotics, and in particular Lactobacillus, have become increasingly popular as an alternative control strategies for bacterial pathogens. Recent studies have demonstrated that Lactobacillus is not only able to increase the growth rate of animals, an important consideration for commercial farming entities, but is also able to inhibit the colonisation of animals with pathogenic bacteria species such as Salmonella, Campylobacter and Escherichia. This study aimed to isolate and characterise lactobacilli isolates from commercially reared chickens and pigs. Particular attention was paid to the probiotic potential of the isolates. Our approach combined molecular techniques with in vitroscreening to rapidly identify this potential. To date 80 and 105 isolates have been collected from pigs and chickens, respectively. All isolates have been confirmed to belong to the Lactobacillus genus by PCR, speciated by 16S sequencing and determined to be clonally unique using RAPD PCR. Isolates were subsequently tested for their ability to tolerate low pH, bile, aerobic and anaerobic conditions before assessing their AMR profile and determine their ability to inhibit a panel of clinical and type strains of pathogenic bacteria. Isolates identified as potential probiotics are currently undergoing whole genome sequenced as per EFSA guidelines. This study has demonstrated that lactobacilli isolates with suitable probiotic properties can be isolated from commercial poultry and pigs. Furthermore, these isolates may prove useful as control strategies for zoonotic bacterial pathogens.

A Mini-ISY100 Transposon Delivery System Effective in γ Proteobacteria.

Transposons are invaluable biological tools for the genetic manipulation of microorganisms. ISY100 from Synechocystis sp. PCC6803 is a member of the Tc1/mariner/IS630 superfamily, and is characterized by high transposition efficiency and a strong preference for TA target sequences. In this paper, we describe the design and application of a mini-ISY100 suicide vector for the in vivo creation of stable random transposon insertion libraries. The system was successfully applied in seven species belonging to four different orders of γ proteobacteria. In all cases, delivery using conjugation consistently showed the highest transposition efficiency compared to chemical transformation or electroporation. We determined the frequency of transposon insertions in all the species and proved the utility of the system by identifying genes involved in colony coloration in Shewanella oneidensis. The ease and the efficiency of the protocol developed here allow the creation of complete knock-out libraries in an extensive range of host microorganisms in less than a week with no requirement for preparatory modification.

Stereodivergent Synthesis through Catalytic Asymmetric Reversed Hydroboration.

The control of chemo-, regio-, diastereo-, and enantioselectivity is a central theme in organic synthesis. The capability to obtain the full set of stereoisomers of a molecule would significantly enhance the efficiency for the synthesis of natural product analogues and creation of chiral compound libraries for drug discovery. Despite the tremendous progress achieved in the field of asymmetric synthesis in the past decades, the precise control of both relative and absolute configurations in catalyst-controlled reactions that create multiple stereocenters remains a significant synthetic challenge. We report here the development of a catalyst-controlled hydroboration with hitherto unattainable selectivity. The Rh-catalyzed hydroboration of α, β-unsaturated carbonyl compounds with pinacolborane proceeds with high levels of regio-, diastereo-, and enantioselectivities to provide a hydroboration product with two vicinal stereocenters. Through the appropriate choice of substrate geometry ( E or Z) and ligand enantiomer ( S or R), all the possible diastereoisomers are readily accessible. The boron-containing products underwent many stereospecific transformations, thus providing a strategy for collective stereodivergent synthesis of diverse valuable chiral building blocks.

Methodology for Developing an Educational and Research Video Library in Minimally Invasive Surgery

Explore the methods used and costs necessary for the creation and maintenance of a surgical video library with an emphasis on its applications in surgical education and scholarship. A methodology paper highlighting how to develop and utilize a surgical video library for trainee operative preparation, development of research projects, and surgeon credentialing. The study was conducted at the University of Pittsburgh Medical Center, a tertiary care medical center. Not applicable. The video library includes all recorded robotic operations performed by the Division of Surgical Oncology at the University of Pittsburgh from 2010 to 2018. It includes 929 videos of which 110 selected videos are uploaded for trainee review online to prepare for upcoming operations. These procedures are broken into steps to create intraoperative time metrics for trainee integration. Fellows operated from console in 85% of robotic cases and all 30 fellows could obtain robotic privileges based on case logs. To date 102 short scholarly videos have been created: 7 for manuscripts, 13 as video submissions, 27 in book chapters, and 55 for presentations. Three papers have been published using video review to determine clinical outcomes with four more under evaluation. The cost of the program is <$10,000 annually. Video libraries can be efficiently created utilizing intraoperative recorders for minimally invasive surgery. Breaking surgeries into distinct steps can aid in deliberate integration and answer clinical questions. Overall video libraries are cost effective tools for trainee education, research, and ultimately surgeon credentialing.

HBIM FOR ARCHAEOLOGICAL SITES: FROM SFM BASED SURVEY TO ALGORITHMIC MODELING

Abstract. The implementation of new technologies often leads research towards experimentation with new methodological approaches. The BIM process applied in cultural heritage is increasingly consolidating its practices, expanding its potential even in the field of archaeological research. Integrating archaeological data into BIM models has clear benefits but, due to the complexity of the involved elements, archaeological case studies can fit effectively into the BIM process only with the creation of semantic-aware libraries implementing dedicated objects. Cultural heritage can advantage of BIM-based knowledge management only integrating approaches and tools with different characteristics from those commonly used in BIM workflows. In the phase of the geometric modeling process, following that of the survey process and preparatory to the subsequent information modeling process, when the detected geometries are not easily traceable to libraries of pre-compiled objects, an alternative to the so-called "Scan to BIM" local modeling can be represented by algorithmic modeling. The paper, focusing on the geometric modelling phase of the HBIM process, presents first results and prospective tasks of an ongoing research project on the last architectural phase of the colonnade of a multilayered temple in the Antas valley in Sardinia (Italy), affected by imposing works of anastylosis and restoration.

Facile synthesis of a diverse library of mono-3-substituted β-cyclodextrin analogues

ABSTRACTSubstituted cyclodextrins (CDs) have many applications, but synthetic challenges have limited the derivatives that can routinely be accessed. In particular, although there is considerable interest in selective derivatization at the 2- and 3-hydroxyls on the secondary face, since bulky guest molecules are most likely to project through this larger aperture, syntheses of such derivatives have required arduous procedures that afford poor yields, limiting their accessibility and utility. We address this challenge via synthetic strategies that allow facile creation of diverse libraries of water-soluble mono-3-substituted-β-cyclodextrins, via the commercially available mono-3-amino-β-cyclodextrin. The power of these strategies is confirmed through synthesis of twenty water-soluble cyclodextrin analogues with amide, thioureas or urea linkers, using one-pot reactions producing ≥55% yields and purifications that do not require chromatography. This work opens new possibilities for the design of selective host molecules for use in supramolecular chemistry, chemical separations, pharmaceutical formulations, and calibration of molecular simulations.

A new strategy for the in vitro selection of stapled peptide inhibitors by mRNA display.

Hydrocarbon stapled peptides are promising therapeutics for inhibition of intracellular protein-protein interactions. Here we develop a new high-throughput strategy for hydrocarbon stapled peptide discovery based on mRNA display of peptides containing α-methyl cysteine and cyclized with m-dibromoxylene. We focus on development of a peptide binder to the HPV16 E2 protein.

Quantitative Metabolomics Using Isotope Residue Outlier Analysis (IROA®) with Internal Standards.

Various research strategies involving biomarker discovery and mechanistic studies in system biology depend on reproducible and reliable quantification of all metabolites from tissue(s) of interest. Contemporary analytical methods rely on mass spectrometry-based targeted and/or untargeted metabolomics platforms. The robustness of these analyses depends on the cleanliness of the samples, accuracy of the database, resolution of the instrument, and, the most variable of the list, the personal preferences of the researcher and the instrument operator. In this chapter, we introduce a simple method to prepare murine liver samples and carry it through the Isotope Ratio Outlier Analysis (IROA®) pipeline. This pipeline encompasses sample preparation, LC-MS-based peak acquisition, proprietary software-based library creation, normalization, and quantification of metabolites. IROA® offers a unique platform to create and normalize a local library and account for run-to-run variability over years of acquisition using the internal standards (IROA®-IS) and long-term reference standards (IROA®-LTRS).
