Effects of Azadirachta indica Leaf Aqueous Extract supplemented Fish Diets on Innate Immune Response and Disease Resistance of Tilapia (Oreochromis niloticus)

Pseudomonas fluorescens is an important bacterial pathogen causing economic losses in aquaculture worldwide. The adverse effects of antibiotics raise public concerns on using them in aquaculture. The objective of this study was to examine the effectiveness of aqueous extract of Azadirachta indica leaves on nonspecific immunity and disease resistance in tilapia (Oreochromis niloticus). Fish diets supplemented with aqueous extract of A. indica at the concentrations of 0% (Diet 1), 1% (Diet 2), 1.5% (Diet 3) and 2% (Diet 4) were investigated for their effects on nonspecific immune response and disease resistance in tilapia. The fish given diet 3 and diet 4 for 3 weeks had increased levels of leukocyte phagocytic activity though higher activity was observed in diet 4 group. Diet 3 and diet 4 also enhanced plasma lysozyme activity in tilapia. The similar levels of lysozyme activity were found in the fish receiving both diets for 3 weeks. Only diet 4 had stimulatory effect on respiratory burst activity in tilapia. This effect was observed 3 weeks after feeding the fish with diet 4. Throughout the experiment, no significant differences (P > 0.05) in phagocytic, lysozyme and respiratory burst activities were found between group fed with diet 1 (control) and diet 2. Following 3 weeks after feeding, fish were infected with Pseudomonas fluorescens at median lethal dose (LD50). No mortality was found in the diet 4 group while 50%, 48% and 28% of cumulative mortality were found in the fish receiving diet 1, 2 and 3 respectively. These results indicate that the aqueous extract of A. indica may elevate the nonspecific immune response and enhance disease resistance in tilapia.

Lactic Acid Bacteria-Fermented Diet Containing Bacterial Extracellular Vesicles Inhibited Pathogenic Bacteria in Striped Beakfish (Oplegnathus fasciatus)

In recent years, probiotics have received considerable attention for improving the health of aquaculture organisms, such as fish and shrimp, by stimulating immune activity and increasing growth rates. Oplegnathus fasciatus is a common and economically important cultured fish species in Asia. In this study, we aimed to investigate the potential of lactic acid bacteria (LAB; Limosilactobacillus reuteri)-fermented feed to promote growth and enhance immune function in O. fasciatus. The feed contained the highest proportion of LAB after L. reuteri fermentation for 3 days in anaerobic conditions. Oplegnathus fasciatus was fed LAB-fermented feed for 30 days. The administration of LAB-fermented feed (live bacteria > 109 CFU/g) significantly increased the growth rate (weight gain = 174.8%; FCR = 4.23) and intestinal probiotic levels of O. fasciatus. After LAB-fermented feeding, the immunity index was evaluated by superoxide anion production, the phagocytic activity of leukocytes, and bactericidal and lysozyme activities in the serum of O. fasciatus. We found that LAB-fermented feed treatment potentially elevated the proportions of intestinal Bifidobacterium, Blautia, and Dorea species and reduced pathogenic bacterial growth (Acinetobacter, Escherichia_Shigella, and Megasphaera) in O. fasciatus. This study demonstrated that LAB-fermented feed containing extracellular vesicles improves growth performance and the inhibition of pathogenic Acinetobacter baumannii.

Purpose: to determine the state of local and general nonspecific resistance to the body in the dynamics of surgical treatment of patients with odontogenic maxillary sinusitis in the presence of galvanic pathology in the oral cavity and to clarify the causes of postoperative inflammatory complications. Methods: 56 examined subjects with chronic odontogenic maxillary sinusitis were divided into two observation groups, which were subjected to immunological and general clinical examination methods. Results. In 30 patients with chronic odontogenic maxillary sinusitis with oral galvanism (compensated and decompensated forms), no decrease in local and general nonspecific resistance was observed. In 26 patients with oral galvanism (atypical and typical forms), a decrease in immunity was observed. Conclusions. Based on the examination of 56 patients with chronic odontogenic maxillary sinusitis in the presence of galvanic pathology in the oral cavity, it was found that in the case of galvanosis, there is a decrease in local and general nonspecific resistance of the body, which, in turn, in all patients leads to the development of inflammatory complications in the periosteal soft tissues and in the bone wound of the jaw, i.e., osteomyelitis (34.6%). From stable to stable: how does stress influence the first line of immune defense in sport horses

Stressful factors, including physical exercise, can influence the body’s defense mechanisms and can be involved in the etiology and evolution of systemic diseases. The aim of this study was to investigate the influence of various stressors to which sport horses are commonly subjected on the innate immune system. For this, four experimental protocols were designed including different animals, namely: (a) resting horses subjected to everyday medium level exercise (n = 18, average age = 7 years, range: 6 to 11 years), (b) medium course transportation, where the distance of one-way trips represented 270 km (n = 8, average age = 7 years, range: 6 to 9 years), (c) a long course, with a 561 km one-way distance (n = 5, average age = 8.2, range: 7 to 11 years) and (d) pre- and post-competition investigations (n = 9, average age = 7 years, range: 6 to 11). Blood was sampled on heparine (50 UI/ml) before and after the workout or transportation. The phagocytic activity of leukocytes was measured by a carbon particle inclusion test, along with total immunoglobulin/opsonin levels (Ig) (24% Zn sulphate precipitation test) and circulating immune complexes (4.2% polyethylene glycol precipitation test). The results were statistically interpreted by use of Micrososft Excel program (Student’s t test). The various types of stress differently influenced the equine athletes. While a moderate daily exercise did not substantially influence the humoral and cellular effectors of innate immunity, transportation exerted stronger beneficial effects based on the distance to the competition site. As a fast acting antimicrobial mechanism, the phagocytosis underwent the most pronounced changes, positively influenced mainly by the short course transportation. While the transportation stress enhanced phagocytosis, the competition decreased the potential of the cells to react, without significantly influencing their numbers. The experiment suggested that management measures, such as age-tailored exercise and maternal care and diet could help in alleviating stress from stable to stable, during the competition season, thus improving animal welfare and allowing a better health forecast.

Morphometric and immuno-biochemical markers of placenta in patients with uterine eclampsia

Purpose: to study the segmentation of organometric and immuno-biochemical digital indicators in ewes with ESO at the end of gestation.Materials and methods. The experiment involved three flocks of Tsygai, Volgograd and Stavropol breeds, each with 700 suyag sheep. Placental tissue samples were taken from clinically healthy and sick pregnant ewes, which were placed in 10% neutral formalin. Using a Cytadel 2000 histoprocessor (Shendon), the obtained samples were clarified in chloroform and enclosed in a Histomix paraffin medium (Biovitrum). Histosections were prepared on a rotary microtome (MICROM HM340E). Using the AxioScope.A1 (ZEISS) microscope, histopreparations were photographed and processed using the ZENpro 2012 (ZEISS) program. According to Schmidt and Thannhauser, the RNA content was determined using a two-wave spectrophotometer in UV. The activity of the G-6-Phase was studied by Swanson. In vacuum tubes Vacuette ™ (Austria), blood samples were taken in the morning before feeding, from the jugular vein. Results. In sheep with eclampsia, a decrease in the total mass of the placenta, its cotiledons, as well as an increase in the length of the umbilical cord, in which stroma sclerosis is noted in the vascular wall, was observed. Small calcification foci were found in the tissues of the afterbirth, and small terminal villi are located around the stem villi near the vascular membrane, tightly adjacent to them. The proportion of syncytiotrophoblast of crypts of carbuncles and villi of the chorion of cotyledons of mature syncytium decreases in comparison with physiologically occurring severity. The content of RNA in its subcellular fractions is mainly concentrated in the microsomal fraction, 18% - in mitochondria and 7% - in the nuclear fraction. Albumins in pregnant ewes with eclampsia were reduced by 1.35 times. BASC and phagocytic activity of leukocytes in queens with eclampsia of pregnant sheep is reduced 15 days before lambing, which poses a threat to the reproductive health of the mother and the viability of newborn lambs.

Purpose. The study of the productive traits of chickens when fed sunflower polysaccharide extract (hereinafter referred to as SPE), which can activate protein and carbohydrate metabolism, stimulate the body's natural defense factors. Materials and Мethods. The studies were carried out in a certified laboratory using classical and modern zootechnical, biochemical and other methods. Laboratory hematological studies were carried out on automatic biochemical analyzers URIT-800Vet and URIT-3020 (China) in the complex analytical laboratory of VRIMMP (Volgograd, Russia). The obtained results were processed using the software, calculation of the mean (M), standard errors of the mean (±SEM) and determination of the Student-Fisher test for the significance of the difference. Results. In I-III experimental groups, an increase in the yield of hatching eggs was recorded, respectively, by 88, 239 and 149 pcs. relative to the control, the intensity of egg production of hens – by 0.65; 1.35 and 0.77%, feed conversion per unit of egg mass – by 0.03; 0.07 and 0.05 kg. There has been a steady trend towards an increase in the mass of eggs in the experimental groups by 0.27 (0.43%); 0.81 (1.29%) and 0.63 g (1.01%), the acid number of the yolk decreased by 8.01 relative to the control; 9.42 (P≤0.05) and 10.29% (P≤0.05), which suggests the presence of antioxidant properties of PPE. The use of the studied additive had a positive effect on the morphological composition of the blood of laying hens of the experimental groups: the content of erythrocytes increased by 15.41 (P≤0.05), 17.47 (P≤0.05) and 17.81% (P≤0, 05), hematocrit – by 5.57 (P≤0.05); 10.22 (P≤0.01) and 11.15% (P≤0.01), hemoglobin – by 6.25 (P≤0.05), 8.67 (P≤0.05) and 9.19 % (P≤0.05) against the background of the control. In laying hens of the experimental groups, an increase in AST activity by 4.86 was recorded; 15.89 and 10.79 u/l, with a decrease in ALT activity by 0.70; 1.37 and 1.25 u/l. A significant increase in glucose in the blood serum of laying hens of the experimental groups was recorded in comparison with the control group by 15.26 (P≤0.05); 23.66 (P≤0.01) and 25.51% (P≤0.01), lactic acid level and glycogen content respectively by 15.26 (P≤0.05); 25.06 (P≤0.01); 25.51% (P≤0.01) and 2.19 (P≤0.05); 3.77 (P≤0.01) and 3.86% (P≤0.01). The bactericidal activity of blood serum of chickens of I-III experimental groups exceeded the control group by 5.02 (P≤0.05); 5.51 (P≤0.05) and 5.70% (P≤0.05 ), lysozyme activity – by 12.31 (P≤0.05); 15.89 (P≤0.01) and 16.36% (P≤0.01), phagocytic activity of leukocytes – by 3.06 (P≤0.05); 4.63 (P≤0.01) and 4.79% (P≤0.01), respectively. Conclusion. The results obtained confirm the activation of metabolic processes in the body of chickens of all experimental groups, under the influence of PPE, however, the studied additive at a dosage of 5.0% in the diet structure had the greatest efficiency in the production of hatching eggs.

The effect of feeding different doses of herbal supplements from Hungarian sainfoin on the growth and metabolism in the body of calves

There have been studied the influence of feeding plant raw material obtained from Hungarian sainfoin having an immunostimulating effect on the growth rate of dairy calves, as well as on morphological and biochemical blood parameters and natural resistance of the animal organism. Calves of the I and II experimental groups in addition to the main diet were fed a phyto supplement made of Hungarian sainfoin mixed with concentrates in doses of 150 and 300 g per head per day during 92 days. The use of herbal supplements in feeding calves ensured an increase in live weight in the animals of experimental groups by 11.0 kg (6.55 %) and by 11.7 kg (6.96 %) (p <0.05) over the period of scientific and economic experiment, compared with analogues of the control group. From the analysis of blood parameters, it should be noted that protein indicators were higher by 1.36 % (I experimental) and by 4.74 % (p < 0.01) (II experimental); there was an increase in hemoglobin at the end of the experiment - by 5.88 % (I experimental) and by 12.15 % (p ≤ 0.01) (II experienced). When determining the indicators of natural resistance, it was found that in calves of experimental groups I and II, the phagocytic activity of leukocytes was higher by 4.16 and 17.29 % (p ≤ 0.01), respectively, in comparison with control animals. Analysis of the data obtained showed that the most optimal dose for use in the diets of heifers is 300 g per head per day, fed to calves of the II experimental group. This is confirmed by the improvement of biochemical, morphological and immunological parameters of animal blood, as well as by higher values of their live weight gain.

The relationship between the character of the course of the disease, the level of tetanus and diphtheria antibodies, and indicators of general immunity in adults with infectious mononucleosis

ABSTRACT. EBV infection is one of the most common infections in the world. About 90 % of the world's population is infected with the Epstein-Barr virus. The Epstein-Barr virus is capable of affecting immunocompetent cells, causing lymphadenopathy, hepato- and splenomegaly, being stored forever in the body's lymphocytes, and causing an immunodeficient state. The aim of the study. The purpose of the study was to determine the relationships between the nature of the course of the disease for infectious mononucleosis (IM) in adults, the level of blood antibodies before tetanus and diphtheria, and general immunoreactivity. Materials and methods. In the first series of studies, blood antibodies was studied before diphtheria and tetanus in 79 adults. The control group consisted of 41 people. In the second series of studies, the immune status of 45 adults who became ill with IM was studied: 19 people who did not have a protective level of blood antibodies to diphtheria and tetanus (1st group) at the same time and 26 people (2nd group) who had a protective level of blood antibodies to these infections . In this work, the level of blood antibodies to diphtheria and tetanus toxin was studied by the ELISA method; population and subpopulation composition of blood lymphocytes using monoclonal antibodies: anti-CD3, CD4, CD8, CD22, CD16 (eBioscience, Beckman Coulter, R&D System). The proliferative activity of blood lymphocytes in the reaction of lymphocyte blast transformation was studied and the level of IgA, IgM, IgG in blood serum and the phagocytic activity of leukocytes. The results of the research were processed by the method of variation and correlation statistics using the "Statistica 10.0 for Windows" program. Results. It was established that 34.2% and 54,5 % of adults who developed MI in a moderate form did not have a protective level of blood antibodies for tetanus and diphtheria at the same time. A protective level of blood antibodies simultaneously before diphtheria and tetanus was observed in 55.2% of those who fell ill with IM in a moderate form and in 45,4 % of those who fell ill in a severe form. In persons who had a mild form of IM, the blood antibodies did not drop below the protective level before diphtheria and tetanus.The study of the immune status proved that in adults who do not have a protective level of blood antibodies against diphtheria and tetanus, the level of leukocytes, lymphocytes, CD3+, CD4+ - cells and the blast-transforming ability of T cells, the level of IgA, IgM, IgG, and the phagocytic ability of blood leukocytes are probably lower than in adults who have had an IM and have the protective blood antibodies level before these infections. Conclusions. The lack of a protective level of blood antibodies against diphtheria and tetanus in adults who have had IM is associated with a general low immunoreactivity of the body. A low level of blood antibodies before diphtheria and tetanus in persons who have had IM is closely related to the course of the disease. The lowest level of blood antibodies before these infections was observed in patients who fell ill with a severe form of IM, in patients with a mild form of the disease, significant changes in the level of blood antibodies before diphtheria and tetanus were not observed, as well as in the immune status.

Dynamics of changes in immunological parameters in adults with COVID-19

Aim. To evaluate the immune response in patients with COVID-19 and to track the dynamics of changes in immunological parameters 2-3 weeks after the onset of the disease. Materials and methods. The study included the results of a survey of 55 people aged 19 to 55 years with a new coronavirus infection (NCVI, COVID-19) in 2020. 27 of them were re-examined. The first sample (n=55) was taken on average 4.3 ± 0.49 days from the onset of the disease, the second (n=27) – 12.7 ± 1.12 days. The control group consisted of practically healthy individuals (n=41). The following indicators were determined in all patients: the presence of SARSCoV-2 RNA, the parameters of the general blood test, the main subpopulations of lymphocytes, the level of total immunoglobulins (IgM, IgG, IgA), the number of CEC, the absorption and bactericidal activity of leukocytes, the level of specific IgM and IgG to SARS-CoV-2. Results. At the onset of the disease, a statistically significant decrease in the level of lymphocytes (p<0.001) and their subpopulations (TNK cells (p<0.001), T-helper lymphocytes (<0.001), cytotoxic T-lymphocytes (p<0.001), NK cells (<0.001) and T-lymphocytes (<0.001)). At the same time, the level of activated T-lymphocytes was statistically significantly higher (p<0.001) than the same indicator in the control group. When analyzing immunological parameters after 12.7±1.12 days, an increase in the total number of lymphocytes (p<0.001), T-lymphocytes (p=0.003), T-helper cells (p=0.015) was found; an increased content of activated T-lymphocytes (p<0.001) and a low level of TNK cells (p<0.001). The indicators of the phagocytic link in patients with COVID-19 in the first days of the disease were within the reference range (similar indicators in the control group). Repeated examination of patients (sample 2) showed an increase in the phagocytic activity of leukocytes and monocytes (p<0.003 and p<0.09, respectively). At the beginning of the disease, already on the 4th-5th day of the disease, specific IgM and IgG antibodies to SARS-CoV-2 were detected in the blood serum of patients, by the second week of the disease their level was significantly higher than in the first study (p<0.009). Conclusion. At the onset of the disease, insufficiency of the cellular link of immunity was noted, which is probably as sociated with a violation of immunoregulatory mechanisms. At the same time, an increase in the level of activated T-lymphocytes indicates an overactivation of immune reactions, i.e. it can be said that there is an imbalance in the immune response in COVID-19.

Body resistance of calves during the imprinting period of growth and development

The results of the conducted research allow us to assert the different level of resistance of the body of calves at the end of the imprinting period, depending on the functional state of the body at birth. In calves born with signs of hypoxia, during the imprinting period, the number of leukocytes in the blood was 1.54 times higher than in functionally active animals. The number of neutrophils in the blood of the calves of the second group was 2.39 times more (p>0.001), and lymphocytes were 1.49 times less (p<0.001). The phagocytic activity of leukocytes in the blood of functionally active calves in the imprinting period of life was significantly higher than in calves born with signs of hypoxia. The phagocytic number of leukocytes in the blood of the calves of the first group was 1.40 times higher, and the phagocytic index was 15.70% more than that of the animals of the control group. At the end of the imprinting period, the resistance index of calves of the control group was 3.47 times higher than this index of calves of the experimental group. The percentage of active leukocytes (AL) reached 38.35 ± 2.17% in the calves of the first group and 13.63 ± 1.01% in the calves of the experimental group. The number of active lymphocytes in the blood of calves of the control group was found to be 1.83 times higher, and microbes were neutralized 2.18 times more than the white blood cells of the calves of the experimental group. The research results indicate the negative impact of hypoxia on the growth and development of the fetus. In newborn calves, adaptation to new living conditions was significantly less than in functionally active calves. The decrease in the resistance of the body of calves born with signs of hypoxia is most pronounced in the imprinting period of growth and development of animals.

Analgesic and anti-inflammatory potential of ethanolic extract from Serjania erecta leaves

Ethnopharmacological relevanceThe infusion of Serjania erecta Radlk (Sapindaceae) (popular name “cipó-cinco-folhas”) leaves is used in popular medicine to treat back pain. The anti-inflammatory, anti-hyperalgesic and anti-nociceptive properties of the ethanolic extract from S. erecta leaves (EESE) has not been yet completely clarified. Aim of the studyThe present study investigated the anti-hyperalgesic, anti-nociceptive and anti-inflammatory properties of EESE in experimental models in mice. Material and methodsEESE was fractionated by chromatographic techniques and the compound was identified by nuclear magnetic resonance (NMR), infrared (IR) spectrum, ultraviolet (UV) methods. Mice received a single dose of EESE by oral route (30, 100, and 300 mg/kg, p.o.) and were submitted to nociception induced by formalin, pleurisy induced by carrageenan and peritonitis induced by zymosan models. Mice also received EESE (30 and 100 mg/kg, p.o.) for 22 days in Complete Freund Adjuvant (CFA) model and another group received EESE for 7 days (30 and 100 mg/kg, p.o.) in pleurisy induced by Bacillus Calmette-Guerin (BCG). The cytotoxicity (MTT), phagocytic and chemotactic inhibitory activities of EESE were performed in in vitro assays. ResultsThe fractionation of EESE led to the identification of kaempferol-3-O-α-L-rhamnopyranoside. The oral administration of all doses of EESE decreased the nociceptive response induced by formalin. EESE significantly inhibited leukocyte migration in carrageenan-induced pleurisy and zymosan peritonitis models. The daily administration of EESE during for 7 days inhibited the leukocyte migration and the mycobacteria growth of pleural material obtained from animals which received BCG. EESE significantly reduced edema, cold allodynia and mechanical hyperalgesia responses induced by CFA. EESE did not induce cytotoxicity, and also decreased the leukocyte phagocytic activity, as well as, neutrophil chemotaxis. ConclusionsEESE showed analgesic and anti-inflammatory properties in acute and persistent experimental models in mice. EESE also reduced in vitro leukocyte chemotaxis and phagocytic activity without inducing cytotoxicity. The continuous oral treatment with EESE was effective against hyperalgesia and inflammation and these results could explain the popular use of S. erecta as an analgesic natural agent.

Characteristics of the Immune and Neuroendocrine Status of Children Exposed to Non-Ionizing Radiation in the School Environment

Introduction: The modern educational environment involves the use of various electronic devices generating physical hazards. In addition to the widespread use of these devices, other physical factors of the school environment also affect schoolchildren’s health. However, the impact of the combined exposure to physical factors of various nature on the regulatory systems of the body has been studied insufficiently so far. However, the influence of a complex of physical factors of various nature on the regulatory systems of the body has not been sufficiently studied. Objective: To evaluate the features of the immune and neuroendocrine status of schoolchildren exposed to heterogeneous physical factors of a non-ionizing nature at school and to analyze the relationship between the studied physical factors and changes in the parameters of regulatory systems. Materials and methods: The exposed group included 144 students of a gymnasium from the city of Perm and the unexposed (reference) group consisted of 114 schoolchildren from the town of Kungur, Perm Region. We measured physical factors of the school environment, including noise, artificial illumination, electromagnetic radiation, and air ion concentrations affecting elementary, secondary, and high school students from both groups and compared the results with respective permissible levels. The indicators of immune (CD-phenotyping, phagocytic activity of leukocytes, concentrations of immunoglobulins and cytokines) and neuroendocrine (TSH, free T4, and cortisol) systems were established and compared. Mathematical models were calculated and analyzed in order to find the links between the exposure to physical factors under study and the parameters of the immune and neuroendocrine systems. Results: The indoor air study showed differences in the number and charge of air ions between the gymnasium with its numerous e-learning tools and the school of comparison. The study of the immune and neuroendocrine systems in the exposed group revealed age-related changes in the expression of lymphocyte subpopulations, moderate changes in humoral and phagocytic parameters, an imbalance in the expression of pro- and anti-inflammatory cytokines and hormones. The construction of logistic regression models allowed us to establish the link between the parameters of electromagnetic radiation in the school environment and changes in the immune status of students (CD19+ and CD3+CD8+ lymphocytes). Conclusion: The study demonstrated a number of changes in the regulatory systems of schoolchildren associated with the impact of physical factors of the school environment.

Effect of cisplatin on oral ulcer-induced nociception in rats

ObjectiveThe aim of this study is to investigate effects of cisplatin preadministration on oral ulcerative mucositis-induced nociception by using an experimental model of rats. DesignAfter two rounds of cisplatin administration, oral ulcers developed with topical acetic acid treatment in rats. Spontaneous mouth rubbing behavior was observed as spontaneous nociceptive behavior in a plastic cage. Head-withdrawal behavior was observed as mechanical allodynia by using von Frey test in the oral mucosa of conscious rats. Bacterial invasion and inflammatory cell infiltration into oral ulcerative region and systemic leukocyte phagocytic activity were assessed. ResultsFollowing cisplatin preadministration, oral ulcerative mucositis-induced spontaneous nociceptive behavior was not observed in the model. The preadministration enhanced leukocyte phagocytic activity, leading to reduce bacterial invasion and inflammatory cell infiltration in the oral ulcerative region. In contrast, oral ulcerative mucositis-induced mechanical allodynia was induced. The exaggerated mechanical allodynia in the oral ulcerative region was largely inhibited by topical treatment with the antioxidative drug, ɑ-lipoic acid, or the blocker of N-formyl peptide receptor 1, N-t-butoxycarbonyl-methionyl-leucyl-phenylalanine. ConclusionsThese results suggest that cisplatin preadministration suppresses spontaneous nociception in oral ulcerative region, due to antiinflammatory effects by enhancement of leukocyte phagocytic activity, but exaggerates mechanical allodynia due to oxidative stress with N-formyl peptide receptor 1 activation. The suppression of spontaneous nociception is one of the advantages of cisplatin treatment for head and neck cancer patients although the exaggerated allodynia is a serious symptom.

Oil-producing enterprise

Arterial hypertension is an urgent health problem worldwide causing an increase in temporary and permanent disability, invalidity and mortality due to cardiovascular diseases. Researchers recognize the multifactorial nature of arterial hypertension, but environmental factors are of particular potential importance. The working conditions at oil production enterprises are characterized by a more pronounced influence of these factors which may predispose for early development of disadaptation disorders, functional changes in immune and humoral regulation, and, finally, for increased risk of cardiovascular diseases in people engaged in oil production. The aim of the present work was to study the features of immunity and humoral risk factors of arterial hypertension in hypertensive employees at an oil-producing enterprise. To this purpose, a comparative analysis of lymphocyte subpopulations (CD3+CD4+, CD3+CD25+), markers of apoptosis (CD3+CD95+, TNFR, p53, Bax, Annexin V-FITC+7AAD), phagocytic activity of leukocytes (absolute phagocytosis index), and the levels of vascular humoral factors (nitric oxide and homocysteine) was performed in employees of an oil production enterprise exposed to adverse production factors. The observational group consisted of employees with established episodes of increased blood pressure. A comparison group consisted of individuals without clinical manifestations of cardiovascular disease. As a result of the clinical and laboratory examination of employees at the oil-producing enterprise with arterial hypertension, some functional changes in immune regulation were revealed. This group was characterized by a significantly (p 0.05) decreased CD3+CD4+, and CD3+CD25+ lymphocyte contents, along with increased levels of regulatory CD127 lymphocytes against the comparison group (p 0.05). The workers at an oil production enterprise with arterial hypertension are characterized by decreased (p 0.05) phagocytic activity of peripheral blood leukocytes using the criteria of absolute phagocytosis. We found some signs of inhibited lymphocyte apoptosis (p 0.05), i.e., a decrease in CD95+, TNFR, and p53 over the background values, as well as increased Bax levels over the comparison group (p 0.05). However, the content of TNFR and p53 significantly (p 0.05) exceeded the reference level, regardless of previous arterial hypertension episodes. Development of the high blood pressure episodes among the employees at oi-producing plant showed a significant association (p 0.05) with elevated levels of homocysteine and nitric oxide concentrations which are known to induce endothelial dysfunction, atherogenesis and, hence, a persistent increase in blood pressure.

Short communication: Comparison of hemato-immunological parameters in rainbow trout (Oncorhynchus mykiss): Palomino vs naturally pigmented varieties

Aim of the study: To evaluate hemato-immunological parameters in the Palomino trout compared to naturally pigmented rainbow trout. Area of study: Fars province (SW Iran). Material and methods: Sixty fish of both varieties (30 Palomino and 30 naturally pigmented) were sampled from the same pond. After blood sampling, hematological parameters, total serum protein and immunoglobulin (Ig), phagocytic activity, respiratory burst activity, serum lysozyme, and bactericidal activity were measured. Main results: Number of white blood cells, total protein, total immunoglobulins, lysozyme, and bactericidal activities in serum of Palomino trout were significantly higher than those in naturally pigmented rainbow trout. However, no significant changes were observed in no. of red blood cells, hemoglobin, hematocrit, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, differential leukocyte count, phagocytic, and respiratory burst activity. Research highlights: The immune system performance of Palomino trout was similar to naturally pigmented fish. However, in some cases, the immune response of Palomino fish may be even more robust than that of naturally pigmented fish. This finding helps us to favor Palomino trout in breeding programs and genetic modification.

Effect of different dietary regimens at dry-off on performance, metabolism, and immune system in dairy cows

Concentrate withdrawal and feed restriction are commonly used to reduce milk production and to facilitate dry-off, but may impair immune function in dairy cows. We investigated the effect of feed rations providing different amounts of nutrients in combination with feed restriction on performance, endocrine, and metabolic responses, as well as on leukocyte function before and after abrupt dry-off. Forty-three cows were studied from d 12 before until d 6 after dry-off (56 d before scheduled calving). Cows were fed experimental concentrates rich in crude protein (nitrogenic, n = 14), glucogenic precursors (glucogenic, n = 14), or lipids (lipogenic, n = 15). On d 3 before dry-off, total feed allowance was restricted to 50% in half of the animals of each dietary group, whereas feed allowance remained unchanged in the other animals. Performance parameters (milk yield, milk composition, and dry matter intake) were recorded, and daily blood and milk samples were taken and analyzed for various metabolic and endocrine parameters. Additionally, activity and mRNA abundance of several genes in leukocytes were measured at selected time points before and after feed restriction and dry-off, respectively. Feed restriction immediately resulted in a negative energy balance and decreased milk production. Concomitantly, concentrations of nonesterified fatty acids increased, whereas insulin, insulin-like growth factor-1, and glucagon decreased. After dry-off, energy balance turned positive and plasma nonesterified fatty acids decreased. Plasma glucose, insulin, and insulin-like growth factor-1 concentrations increased in all groups after dry-off. Glucose, insulin, and glucagon concentrations in plasma were higher in nonrestricted compared with restricted animals after dry-off. The experimental concentrate types marginally affected the investigated metabolic and endocrine factors, with the exception of elevated milk and plasma urea concentrations in cows fed the nitrogenic concentrate. Chemotactic and phagocytic activity of leukocytes were not affected by diets, feed restriction, or dry-off. Likewise, blood leukocyte mRNA abundance encoding for tumor necrosis factor α (TNF), heat shock protein family A (HSP70), and the glucose transporters (GLUT) 1 and 3 remained unchanged throughout the study period. Overall, the short-term negative energy balance induced by feed restriction was temporarily accompanied by metabolic adaptations, but did not alter the studied factors related to the immune system. Metabolic and endocrine adaptations supporting milk synthesis were continued during the first days after dry-off despite cessation of milking. Thus, the abrupt dry-off resulted in a short-term increase of glucose and triglyceride concentrations, with a delayed endocrine response to re-establish nutrient homeostasis in blood.

USE OF PREPARATION OF RECOMBINANT INTERFERONS FOR CORRECTION OF OVULATORY FUNCTION IN OVARIES OF DAIRY COWS

The article presents the material on studying the efficacy of the preparation of recombinant interferons containing granulocyte-macrophage colony-stimulating factor (GM-CSF) for the prevention of the post-estral uterine bleeding in dairy cows, accompanied by a retardation in ovulation of the dominant follicle. It has been found that a single administration of the drug containing GM-CSF in its composition to cows at a dose of 5.0 ml during insemination, contributes to the ovulatory functional recovery in 86.7% of animals, and reduces the period from calving to fertilization by 27.7 – 37.8 days. The introduction of the recombinant interferon preparation to cows provides activation of the cellular link in the immune defense, normalization of the blood hormonal-cytokine profile, recovery of the ovulatory function in the genital gonads. This is manifested by an increase in the serum lysozyme activity – by 29.5%, leukocyte phagocytic activity – by 12.4%, phagocytic index – by 38.8%, phagocytic number – by 58.8%, progesterone – by 59.9%, estradiol – by 60.0%, testosterone – by 59.3%, DHEA-S – by 56.8%, IL-4 – by 42.5%, IL-10 – by 53.4%, with a decrease in the content of IL-2 – by 40.4% and IL-1β – by 37.2%.

The state of functional activity of neutrophils in patients with various variants of carbohydrate metabolism disorders who have had SARS-COV-2S infection

According to the Federal State Statistics Service of the Russian Federation, there has been a significant increase in the prevalence of prediabetes and type 2 diabetes mellitus (DM). Type 2 diabetes is characterized by frequent disability due to the progression of complications of diabetes and high mortality. Domestic and foreign authors have shown that neutrophils play an important role in the development of diabetic micro- and macroangiopathies. It has been shown that in DM, changes in the functional activity of neutrophils are observed, manifested in a decrease in chemotactic activity, changes in phagocytic activity, leukotriene production, and secretion of lysosomal enzymes. According to the results of previous studies, ambiguous data were obtained on the effect of high hyperglycemia in DM on the phagocytic activity of neutrophils. Thus, foreign authors have shown that in DM, when the target levels of glycemic control are not reached, the phagocytic activity of neutrophils is inhibited. Other researchers have found that with DM, activation of the phagocytic function of neutrophils is detected, but at the same time, the reaction of neutrophils to additional stimulation with latex (according to the results of the HCT test), a decrease in the expression of adhesion molecules on their surface and the ability to form extracellular traps occurs. Neutrophils, being the main cells of innate immunity, play an important role in the antiviral response. Patients with impaired carbohydrate metabolism are at high risk of severe course and unfavorable outcome of SARS-CoV-2 infection. The course and outcome of an infectious disease depends on how the effector functions of neutrophils are realized. The aim of the study was to study the features of the functional activity of neutrophils in patients with various variants of carbohydrate metabolism disorders who had SARS-CoV-2 infection. The study included 20 patients with impaired carbohydrate metabolism who had suffered SARS-CoV-2 infection, the control group included 15 clinically healthy volunteers. In all groups, the phagocytic activity of latex particles with a diameter of 1.7 microns was evaluated by neutrophils (phagocytosis activity, phagocytosis intensity, phagocytic number); spontaneous and induced NBT activity of neutrophils by morphological method. A significant increase in the absorption capacity of peripheral blood neutrophils, an increase in the phagocytic activity of leukocytes in the group of patients with impaired carbohydrate metabolism was revealed. A simultaneous increase in the indicators of spontaneous and induced HCT tests was found in the group of patients with impaired glucose tolerance and in the group of patients with type 2 diabetes mellitus. The observed disorders in the phagocytic link are an important factor in reducing the overall immunoreactivity of patients, which determines the long rehabilitation period. Identifying the features of innate immunity in patients with various variants of carbohydrate metabolism disorders who have suffered from SARS-CoV-2 infection will further help develop an individual strategy for the prevention and treatment of these diseases.

The effect of the drug “Amiksyn” on the immune status of carp

The productivity of pond fish, in particular carp, largely depends on the intensity of use of aquatic bioresources. The aim of the study was to investigate the effect of “Amiksyn” on the immunological parameters of carp fish, namely: the number of T- and B-lymphocytes, immunoglobulins, the level of circulating immune complexes, lysozyme, phagocytic activity and bactericidal activity of serum. 10 fish were selected for hematological and biochemical studies. The research was conducted in the aquarium of the Department of Aquatic Bioresources of Stepan Gzhytskyi NUVMB Lviv. Fish were kept in the pool for 21 days to adapt to the new conditions, and at the beginning of the series of experiments were placed in aquariums with a volume of 200 liters. Aeration and mechanical filtration of water were provided in the tanks. The temperature during the experiments fluctuated slightly and was 18 ± 1.5 °C. The main hydrochemical parameters corresponded to fish farming standards. In order to adjust the immune response, and thus prevent possible manifestations of fish damage by pathogens, we studied the possibility of using the immunostimulant “Amiksyn”, which was used for 5 – 10 – 15 – 20 days and then dosing 5 – 10 – 15 mg/kg body weight Pisces. The use of “Amiksyn” at a dose of 10 mg/kg of fish weight four times during the fifteen-day period with an interval of 5 days provided a decrease in the number of T-lymphocytes by 9.1 % and B-lymphocytes by 23.0 %; caused the activation of non-specific resistance indicators, namely lysozyme activity increased by 28.7 %, phagocytic activity of leukocytes increased by 14.0 % and bactericidal activity of serum increased by 19.0 %. Therefore, to adjust the immune system and stimulate the metabolic processes of fish in order to prevent the negative effects of various pathogens, the most optimal dose was 10 mg/kg of fish weight within 15 days of use.
