Linoleate Isomerase Complex Contributes to Metabolism and Remission of DSS-Induced Colitis in Mice of Lactobacillus plantarum ZS2058.

A linoleate isomerase complex including myosin-cross-reactive antigen, short-chain dehydrogenase/oxidoreductase, and acetoacetate decarboxylase has been confirmed as the pivotal factor for conjugated linoleic acid (CLA) production in Lactobacillus plantarum. However, its role in the metabolism and health-associated benefits of Lactobacillus remain unclear. In the current study, the mild type, knockout, and complemented mutants of the linoleate isomerase complex of L. plantarum ZS2058 were used to investigate those putative effects. The metabonomic results showed that a linoleate isomerase complex could significantly influence the glycol-metabolism, lipid metabolism, and antioxidant compounds. Especially, with the stress of linoleic acid, linoleate isomerase complex knockout mutants induced the increase of several antioxidant compounds, such as glutamic acid, glycine, l-cysteine, glycerol, and l-sorbosone. Moreover, the linoleate isomerase complex played a pivotal role in ameliorating DSS-induced colitis. The knockout mutants showed effects similar to those in the DSS group, whereas complementation of the corresponding gene in the knockout mutants could restore the anti-inflammatory activity, wherein the integrity of a mucus layer was repaired, the level of pro-inflammatory cytokines decreased, and the amount of anti-inflammatory cytokines increased significantly. All the results indicated that the linoleate isomerase complex plays a key role in CLA production and metabolism as well as the health-associated benefits of L. plantarum ZS2058. These results are conducive to promote clinical trials and product development of probiotics for colitis.

Synergistic Effect of Eugenol and Probiotic Lactobacillus Plantarum Zs2058 Against Salmonella Infection in C57bl/6 Mice.

Previously, we showed the preventive effects of Lactobacillus plantarum ZS2058 (ZS2058) on Salmonella infection in murine models. In this work, we found that eugenol has a selective antibacterial effect, which inhibited Salmonella more than probiotics ZS2058 in vitro. It suggested a synergistic effect of them beyond their individual anti-Salmonella activity. We verified the conjecture in murine models. The results showed that the combination of ZS2058 and eugenol (CLPZE) significantly increased (p = 0.026) the survival rate of Salmonella-infected mice from 60% to 80% and the effect of CLPZE on preventing Salmonella-infection was 2-fold that of ZS2058 alone and 6-fold that of eugenol alone. CLPZE had a synergistic effect on inhibiting ST growth (the coefficient drug interaction ((CDI) = 0.829), reducing its invasiveness (CDI = 0.373) and downregulating virulence genes’ expression in vitro. CLPZE helped the host form a healthier gut ecosystem. CLPZE also elicited a stronger and earlier immune response to systemic infection. In conclusion, these obtained results suggest that ZS2058 and eugenol have a synergistic effect on preventing Salmonella infection and open new perspectives in the strategies of controlling the prevalence of Salmonella by combination of probiotics and functional food components.

Antiproliferation Activity and Mechanism of c9, t11, c15-CLNA and t9, t11, c15-CLNA from Lactobacillus plantarum ZS2058 on Colon Cancer Cells.

Conjugated linolenic acid (CLNA) is a type of ω-3 fatty acid which has been proven to have a series of benefits. However, there is no study about the function of Lactobacillus-derived CLNA isomer. Lactobacillus plantarum ZS2058 has been proven to manifest comprehensive functions and can produce CLNA. To investigate the specific functions of CLNA produced by this probiotic bacterium, two different conjugated α-linolenic acid (CLNA) isomers were successfully isolated. These isoforms, CLNA1 (c9, t11, c15-CLNA, purity 97.48%) and CLNA2 (c9, t11, t15-CLNA, purity 99.00%), both showed the ability to inhibit the growth of three types of colon cancer cells in a time- and concentration-dependent manner. In addition, the expression of MDA in Caco-2 cells was increased by CLNA1 or CLNA2, which indicated that lipid peroxidation was related to the antiproliferation activity of CLNAs. An examination of the key protein of pyroptosis showed that CLNA1 induced the cleavage of caspase-1 and gasdermin-D, while CLNA2 induced the cleavage of caspase-4, 5 and gasdermin-D. The addition of relative inhibitors could alleviate the pyroptosis by CLNAs. CLNA1 and CLNA2 showed no effect on caspase-3, 7, 9 and PARP-1, which were key proteins associated with apoptosis. No sub-diploid apoptotic peak appeared in the result of PI single staining test. In conclusion, CLNA1 activated caspase-1 and induced Caco-2 cell pyroptosis, whereas CLNA2 induced pyroptosis through the caspase-4/5-mediated pathway. The inhibition of Caco-2 cells by the two isomers was not related to apoptosis. This is the first study on the function of Lactobacillus-derived CLNA isomer. The inhibition pathway of Lactobacillus-derived CLNA isomer on colon cancer cells were proved.

C9, t11, c15-CLNA and t9, t11, c15-CLNA from Lactobacillus plantarum ZS2058 Ameliorate Dextran Sodium Sulfate-Induced Colitis in Mice.

To investigate the specific functions of conjugated fatty acids (CFAs) produced by the probiotic bacterium, α-linolenic acid was isomerized by Lactobacillus plantarum ZS2058, and two different conjugated linolenic acid (CLNA) isomers were successfully isolated: c9, t11, c15-CLNA (CLNA1) and t9, t11, c15-CLNA (CLNA2). The effects and mechanism of CLNA crude extract and individual isomers on colitis were explored. CLNA significantly inhibited weight loss, the disease activity index, and colon shortening. Additionally, CLNA alleviated histological damage, protected colonic mucus layer integrity, and significantly upregulated the concentration of tight junction proteins (ZO-1, occludin, E-cadherin 1, and claudin-3). CLNA significantly attenuated the level of proinflammatory cytokines (TNF-α, IL-1β, and IL-6) while upregulating the expression of the colonic anti-inflammatory cytokine IL-10 and nuclear receptor peroxisome-activated receptor-γ. Moreover, CLNA increased the activity of oxidative stress-related enzymes (SOD, GSH, and CAT), and the myeloperoxidase activity was significantly decreased by CLNA. Meanwhile, the concentrations of CLNA in the liver and conjugated linoleic acid in the colonic content were significantly increased because of the treatment of CLNA. Furthermore, CLNA could rebalance the intestinal microbial composition of colitis mice, including increasing the α-diversity. CLNA1 and CLNA2 increased the abundance of Ruminococcus and Prevotella, respectively.

Genetic determinates for conjugated linolenic acid production in Lactobacillus plantarum ZS2058.

To investigate the genetic determinates for conjugated linolenic acid (CLNA) production in Lactobacillus plantarum ZS2058, a high CLNA producer. After culturing with α-linolenic acid (ALA) in the medium, the fatty acid compositions of supernatant fluid and cell pellets were analysed via GC-MS. cis9,trans11,cis15-CLNA was identified to be the predominant isomer. And during CLNA production, 10-hydroxy-cis12-cis15-octadecenoic acid (10-HOEA) and 10-oxo-cis12-cis15-octadecenoic acid (10-OXOA) were accumulated. The E. coli recombinants harbouring genes encoding myosin-cross-reactive antigen (MCRA), short-chain dehydrogenase/oxidoreductase (DH) and acetoacetate decarboxylase (DC), respectively, were analysed for their roles in CLNA production. The results indicated that MCRA converted ALA to 10-HOEA, following converted to 10-OXOA by DH. While with the combination of three recombinants, ALA could be transformed into CLNA plus 10-HOEA and 10-OXOA. When the three genes were deleted, none of the L. plantarum ZS2058 knockout mutants could produce any CLNA, after complementation, and all the complementary mutants recovered the CLNA-production ability at similar levels as the wild strain. Lactobacillus plantarum ZS2058 produced CLNA from ALA with 10-HOEA and 10-OXOA as intermediates. The triple-component isomerase of MCRA, DH and DC was the unique genetic determinant for CLNA generation. The current results firstly provided conclusive evidence that the triple-component isomerase complex was shared by both CLA and CLNA production in lactobacilli.

Lactobacillus plantarum ZS2058 and Lactobacillus rhamnosus GG Use Different Mechanisms to Prevent Salmonella Infection in vivo

Pathogen-induced infectious diseases pose great threats to public health. Accordingly, many studies have investigated effective strategies targeting pathogenic infections. We previously reported the preventive effects of Lactobacillus plantarum ZS2058 (ZS2058) and L. rhamnosus GG (LGG) against Salmonella spp. in a murine model. Here, we compared the mechanisms underlying the preventive effects of these Lactobacillus strains in vivo. Notably, reduced C-reactive protein levels were observed with both ZS2058 and LGG, which suggests abrogated anti-infection and inflammatory responses. ZS2058 more efficiently reduced the pathogenicity of Salmonella by increasing the level of propionic acid in feces and production of mucin 2 in the mouse colon and activity through the interleukin (IL)-23/IL-22 and IL-23/IL-17 pathways. Meanwhile, LGG more strongly alleviated gut inflammation, as indicated by changes in the levels of tissue necrosis factor (TNF)-α, IL-10 and myeloperoxidase (MPO) in infected mice. Moreover, both ZS2058 and LGG restored the levels of interferon (INF)-γ, a cytokine suppressed by Salmonella, albeit through different pathways. Our results demonstrate that ZS2058 and LGG prevent Salmonella infection via different mechanisms.

Optimization of the quenching and extraction procedures for a metabolomic analysis of Lactobacillus plantarum

This study evaluated the quenching and extraction procedures using different solvents at different temperatures for Lactobacillus plantarum ZS2058. Eight quenching methods that used readily available organic solvents (methanol and ethanol) or cryoprotectants (glycerol and dimethyl sulfoxide) were evaluated quantitatively, and their effects on cell membrane integrity and metabolic inactivation were assessed by propidium iodide and energy charge assays, respectively. The combination of 20% methanol and 0.9% sodium chloride (−4 °C), with the lowest propidium iodide labelling rate of 3.06 ± 0.16% and the highest energy charge value of 0.849 ± 0.003, outperformed most of its counterparts and was adopted for further use. To retrieve most of the cellular metabolites, four extraction solvent systems, including methyl tert-butyl ether/methanol/water, methanol/water, acetonitrile/water and acetonitrile/methnol/water, were evaluated. And acetonitrile/methnol/water was found to be the most efficient one as, among other solvents, it yielded the highest metabolite abundances for most of the metabolites. Furthermore, the protocol was subjected to analytical validation using a series of selected representative metabolites and yielded a linear range of 0.00128–100 μg mL−1 with coefficients of 0.9953–0.9999 and recovery levels of 90%–110%. These results suggest the reliability of the proposed method for L. plantarum ZS2058.

Characterization of the triple-component linoleic acid isomerase in Lactobacillus plantarum ZS2058 by genetic manipulation.

To assess the mechanism for conjugated linoleic acid (CLA) production in Lactobacillus plantarum ZS2058. CLA has attracted great interests for decades due to its health-associated benefits including anticancer, anti-atherogenic, anti-obesity and modulation of the immune system. A number of microbial CLA producers were widely reported including lactic acid bacteria. Lactobacillus plantarum ZS2058, an isolate from Chinese traditional fermented food, could convert LA to CLA with various intermediates. To characterize the genetic determinants for generating CLA, a cre-lox-based system was utilized to delete the genes encoding myosin cross-reactive antigen (MCRA), short-chain dehydrogenase/oxidoreductase (DH) and acetoacetate decarboxylase (DC) in Lact. plantarum ZS2058, respectively. Neither intermediate was detected in the corresponding gene deletion mutant. Meanwhile all those mutants could recover the ability to convert linoleic acid to CLA when the corresponding gene was completed. The results indicated that CLA production was a multiple-step reaction catalysed by triple-component linoleate isomerase system encoded by mcra, dh and dc. Multicomponent linoleic acid isomerase provided important results for illustration unique mechanism for CLA production in Lact. plantarum ZS2058. Lactobacilli with CLA production ability offer novel opportunities for functional food development.

Complete genome sequence of Lactobacillus plantarum ZS2058, a probiotic strain with high conjugated linoleic acid production ability

Lactobacillus plantarum ZS2058 was isolated from sauerkraut and identified to synthesize the beneficial metabolite conjugated linoleic acid. The genome contains a 319,7363-bp chromosome and three plasmids. The sequence will facilitate identification and characterization of the genetic determinants for its putative biological benefits.

Synthesis of conjugated linoleic acid by the linoleate isomerase complex in food-derived lactobacilli.

To assess strains of lactobacilli for their capacity to produce functional fatty acid-conjugated linoleic acid. To assess the linoleate isomerase for CLA production in the most efficient CLA producer. In this study, strains of food-derived lactobacilli were cultured in media with linoleic acid and CLA production was assessed. Most of the selected strains produced CLA at different levels, with Lactobacillus plantarum ZS2058 being the most efficient CLA producer converting over 50% of linoleic acid to c9, t11-CLA and t9, t11-CLA. Some intermediates 10-hydroxy-cis-12-octadecenoic acid, 10-oxo-cis-12-octadecenoic acid and 10-oxo-trans-11-octadecenoic acid were determined via GC-MS. The genes coding the multicomponent linoleate isomerase containing myosin-cross-reactive antigen, short-chain dehydrogenase/oxidoreductase and acetoacetate decarboxylase for CLA production in Lact.plantarum ZS2058 were cloned and expressed in Escherichia coli. With the mixture of recombinant E.coli, c9, t11-CLA and three kinds of intermediates were produced from linoleic acid, which were in line with those in the lactobacilli. The ability for CLA production by lactobacilli exhibited variation. Lactobacillus plantarum and Lact.bulgaricus were the most efficient producers in the selected strains. Lact.plantarum ZS2058 converted linoleic acid to CLAs with 10-hydroxy-cis-12-octadecenoic acid, 10-oxo-cis-12-octadecenoic acid and 10-oxo-trans-11-octadecenoic acid as intermediates. The multiple-step reactions for CLA production catalysed by multicomponent linoleate isomerase in Lact.plantarum ZS2058 were confirmed successfully. Multicomponent linoleate isomerase provides important results for the illustration of the mechanism for CLA production in lactic acid bacteria. Food-derived lactobacilli with CLA production ability offers novel opportunities for functional foods development.
