Halophilic bacteria are bacteria that can survive in environments with high salt levels, one of which is in salt ponds. Therefore, it is necessary to research to determine the morphology of halophilic bacteria for further use in the identification of bacterial species. This study aimed to determine the colony morphology and types of gram halophilic bacteria. Isolation of halophilic bacteria was carried out using the scatter method. Bacterial purification was carried out by streak method, then morphology identification and bacterial staining were carried out to determine the cell shape and gram of bacteria. The isolation results obtained 4 isolates from raw water (B.AB.1) and reservoir (B.B.1, B.B.2, and B.B.3). The results of the identification of the colony morphology of each bacterial isolate had different colors, shapes, colony edges, elevations, and surfaces. The results of the gram staining test showed that 2 isolates (B.B.1 and B.B.2) were gram-positive and 2 bacterial isolates (B.AB.1 and B.B.3) were gram-negative. The isolates of B.AB.1, B.B.2 and B.B.3 were in the form of bacilli, while the isolates of B.B1 were in the form of cocci.

Isolation of Halophilic Bacteria and Their Screening for Extracellular Enzyme Production

Halophiles are those microorganisms which are found at high salt concentration. These microorganisms have the capability to form a wide array of bioactive substances that have different applications in various industries. This work focussed at the isolation and screening of various halophilic bacterial strains from different places in Rajasthan and Tamil Nadu. Using a complete medium with different salt concentrations, 70 halophilic bacteria were isolated from 4 different sites. These isolated bacteria were then examined and screened out for the synthesis of different extracellular enzymes, such as cellulase, lipase and amylase. Out of the total, 24 isolates were found positive for amylase, 9 for lipase and 16 for cellulase. These enzymes can perform the hydrolytic activity at high salt concentrations. The various halophilic bacterial strains isolated from various places show a prospective for use in a range of biotechnological and molecular biology experiments and the extremozymes obtained from these bacteria have great industrial importance.

Isolation and Characterization of Halophilic Bacteria from Salinity Soil of Shatkhira, Bangladesh

Aim: The aim of this study was undertaken for the isolation and molecular characterization of the halophilic bacteria from salt affected soils.
 Place and Duration of Study: This study was conducted on the salinity affected area in Ramzan Nagor under Shamnagor Upazilla of Shatkhira District, Bangladesh. The collection of soil sample, isolation of halophilic bacteria and subsequent experiments were done from May to December 2018.
 Methodology: Salt affected soil could be good source of halophilic bacteria as it contain high amount of salt. Hence, one gram of salt affected soil sample which was containing high amount of salt was suspended to 100 ml sterile distilled water and one ml of sample from the top of the suspension were taken in to 250 ml Erlenmeyer flaks containing 100 ml of mineral salts (MS) medium. The primary enrichment was incubated for several days at 37°C with shaking at 120 rpm (revolution per minute) on an orbital shaker. Cultures which were found to be turbid after a period of up to 4 days were used as inocula in subsequent experiments.
 Results: Four halophilic bacterial strains viz. Bacillus sp. strain 8-15, Enterobacter sp. strain LCR75, Acinetobacter sp. strain 407 and Acinetobacter junii strain F27 were isolated from the saline soil and screened for their growth at a range of salt (NaCl) concentrations viz., 0%, 2%, 2.5%, 3%, 3.5%, 4%, 4.5% and 5%. Out of four isolates, Bacillus sp. showed the highest capacity to tolerate 5% salt concentration whereas Enterobacter sp. and other two isolates (Acinetobacter sp. and Acinetobacter junii) showed the capacity to tolerate 4% and 2.5% salt concentration respectively. Based on their morphological and biochemical characteristics the isolates were found to be Gram-positive and Gram-negative, rod shaped, motile bacteria which were later identified on the basis of 16S rRNA gene sequencing. The optimum culture conditions of the isolates were pH 7.0 and temperature 37°C. Results on the antibiogram profiles of the isolates revealed that the strains were sensitive to most of the antibiotics used in this study except Kenamycin, Ceftazidime and Ampicillin.
 Conclusion: The isolated halophilic bacteria may play an important role to optimize the salt affected soil; hence they could be important candidates for further research on slat bioremediation.

Comparative molecular studies of halophilic bacteria from saline water and soil in the Saudi environment

Halophilic bacteria are a microorganism that grows optimally in the presence of the very high concentration of sodium chloride. Halophiles are vital sources of various enzymes including hydrolases, which are very stable and catalytically highly efficient at high salt concentration and other extreme conditions such as high temperature, pH and presence of organic solvents. Several hydrolases such as amylases, proteases, and lipases have been obtained from halophilic bacteria and are commonly used for various industrial applications. We initiated a screening project to isolate and characterize the halophilic bacteria from the Red Sea, which is one of the saltiest bodies of water in the world. Water and soil samples, collected from the Red Sea coast, Jeddah, Saudi Arabia, were screened for isolation of halophilic bacteria. Ten bacterial isolates were obtained, which were characterized by biochemical tests and 16S rRNA gene sequencing. Hydrolase producing bacteria among the isolates were screened by plate assay on starch and gelatin agar plates for amylase and protease, respectively. Two bacterial isolates i.e Bacillus haynesii and Enterobacter cloacae subsp. were found to possess significant amylase and protease activity. Further characterization of both the strains is in progress.

Microflora microbiological characteristics of saline soils

The purpose of current work was isolation of halophilic bacteria from saline lands in some regions of the Krasnodar Territory, after mycological analysis, there followed the study and characterization of microorganisms with further creation on effective biocomplex basis, contributing to restoration of arid and saline irrigated areas. Soil samples were taken from saline lands of the Krasnodar Territory, the salt composition of which was studied in the work process. There were determined total titer of microorganisms and the titer of nitrogen-fixing bacteria on the Corresponding selective media in the samples. Twenty-five pure bacterial cultures were isolated from the samples and its morphophysiological and biochemical properties, as well as its nitrogenfixing activity, were studied. As a result, seven cultures with relatively high nitrogen-fixing activity were selected, as well as resistance to various environmental factors, particularly, to high concentrations of NaCl, рН alkaline values and to sharp temperature drops. After appropriate phylogenetic analysis, selected crops may become a potential target for creating a complex biological fertilizer, contributing to the restoration of dry and saline lands and increase their fertility.

Alleviation of Salinity Stress on Some Growth Parameters of Wheat by Exopolysaccharide-Producing Bacteria

After isolation of halophilic bacteria from saline soils, the effects of two superior isolates producing exopolysaccharide TP7 and TP5 was investigated on some wheat growth parameters such as shoot and root dry weight and concentration of K+, Ca2+, Na+ and K+/Na+, Ca2+/Na+ ratios under saline stress. Inoculation with bacteria (TP7, TP5 and both of strains) had a positive effect on shoot weight (74.71, 57.52 and 82.45%) at EC 8 dS m−1 and (45.26, 60.95 and 71.28%) at EC 16 dS m−1. Also, TP7 and TP5 isolates can increase plant resistance to salt and drought stresses by decreasing Na+ uptake and in 16 dS m−1 led to an increase in dry weight of root in comparison with control by 97.95, 128.29 and 193.53%, respectively. In higher-salinity treatments, all of treatments (TP5, TP7 and both of strains) had a significant effect on K+ concentration, especially in EC 16 dS m−1 by 29.58, 45.94 and 53.56% and 35.50, 52.37 and 58.65% in water stress and non-water stress, respectively. The determination of 16S rRNA gene sequences of isolates TP7 and TP5 showed that they are mostly close to Bacillus subtilis susp. inaquosorum (98.49% sequence similarity) and Marinobacter lipolyticus SM19 (T) (97.69% sequence similarity), respectively. Generally, results showed that these isolates can be used as a tool for reducing adverse effects of salinity and drought stresses in wheat plant.

Isolation of Halophilic Bacteria from Inland Petroleum- Producing Wells

The goals of this study were to isolate microorganisms from oil well-produced water, identify the microorganisms, and test the microorganisms’ salt tolerance. Saltwater collected from two well locations producing from different zones in Jones County, Texas, was spread onto Mannitol Salt Agar (MSA). Isolates showed a 16S rDNA gene sequence identity of 99% with Idiomarina baltica and Marinobacter persicus. Salt tolerance assays indicated an optimal growth concentration of 10-12.5% NaCl for the Idiomarina isolate and a decrease in growth beyond 5% NaCl for the Marinobacter isolate. In conclusion, organisms that are phylogenetically similar to marine microorganisms are present in oil well environments, and have variable salt tolerances, which may prove useful in microbialmediated hydrocarbon bioremediation of high salinity environments.
