Bajgelman1, 2* 1 National Center For Research in Energy and Materials – CNPEM, Brazilian Biosciences National Laboratory– LNBio , Brazil 2 University of Campinas, Medical School, Brazil GM-CSF-expressing tumor cell-based vaccine (GVAX) induces a potent antitumor response and is considered as a gold standard to compare efficacy of new strategies that are being developed to produce specific anti-tumor immunity. Although GVAX has surprising results in animal models, there is no comparable success in the clinic. This observation can be associated to the complexity of the immune system and low immunogenicity of tumor cells that may be protected by immunosuppressive mechanisms which mediate activation and proliferation of effectors lymphocytes. The costimulatory TNFSF receptors can modulate phenotypic properties of T cell, reducing the activation threshold, inducing a greater level of proliferation and preventing cell death after activation. In this work, we evaluate the anti tumor effect of modified B16F10 irradiated tumor cells expressing TNFRSF4, also know as OX40 ligand (CD134), which receptor is expressed on the surface of activated T cells and play a role in the maintenance of the immune response.We have performed in vivo assays in C57BL/6 mice that were challenged with wild type B16 F10 tumor cells and treated with modified B16F10 irradiated cells. We compare tumor progression in 4 groups of mice divided according to the modified B16 F10 they received (A) PBS, (B) GVAX, (C) OX40L and (D) combination of GVAX and OX40L. Our data suggests that OX40L induces the same reduction in tumor mass as the gold standard GVAX, and the combination of the two approaches provides a synergistic effect, enhancing the elimination of tumor cells.
