Effects of Perfluorooctane Sulfonate on Cerebellar Cells via Inhibition of Type 2 Iodothyronine Deiodinase Activity.

Perfluorooctane sulfonate (PFOS) has been used in a wide variety of industrial and commercial products. The adverse effects of PFOS on the developing brain are becoming of a great concern. However, the molecular mechanisms of PFOS on brain development have not yet been clarified. We investigated the effect of early-life exposure to PFOS on brain development and the mechanism involved. We investigated the change in thyroid hormone (TH)-induced dendrite arborization of Purkinje cells in the primary culture of newborn rat cerebellum. We further examined the mechanism of PFOS on TH signaling by reporter gene assay, quantitative RT-PCR, and type 2 iodothyronine deiodinase (D2) assay. As low as 10-7 M PFOS suppressed thyroxine (T4)-, but not triiodothyronine (T3)-induced dendrite arborization of Purkinje cells. Reporter gene assay showed that PFOS did not affect TRα1- and TRβ1-mediated transcription in CV-1 cells. RT-PCR showed that PFOS suppressed D2 mRNA expression in the absence of T4 in primary cerebellar cells. D2 activity was also suppressed by PFOS in C6 glioma-derived cells. These results indicate that early-life exposure of PFOS disrupts TH-mediated cerebellar development possibly through the disruption of D2 activity and/or mRNA expression, which may cause cerebellar dysfunction.

Acute nitrite exposure interferes with intestinal thyroid hormone homeostasis in grass carp (Ctenopharyngodon idellus)

Nitrite in the aquatic environment potentially disturbs thyroid hormone (TH) homeostasis in peripheral tissues, but little is known about TH metabolism in the intestine. This study investigated the serum concentrations of THs and thyroid-stimulating hormone (TSH) as well as the activity of intestinal iodothyronine deiodinases (IDs) of grass carp (Ctenopharyngodon idellus) exposed to various concentrations of nitrite (0, 8, 25, or 50 mg/L) for 96 h. Acute nitrite exposure significantly altered the triiodothyronine (T3) levels and the morphology of thyroid follicles at 96 h. Thyroxine (T4), free T4 levels and intestinal IDs activities showed an increase trend under nitrite stress. After 96 h exposure, nitrite down-regulated the expressions levels of intestinal Akt1 protein, sugar transporter genes, and thyroid hormone receptor (TR) signaling pathway genes except for tr ɑ1 and tr ɑ2. Moreover, the expressions levels of pparγ, cpt1α, cd36, fabp2 and fatp4 were down-regulated, whereas fabp6 and lpl were up-regulated in the 50 mg/L exposure group at 96 h. The results indicate that acute nitrite exposure has the potential to disturb the homeostasis of intestinal TH metabolism, which in turn alters TRs genes transcription, down-regulates sugar transporter activities, and promotes the energy expenditure in gut of grass carp.

Deiodinases and the Three Types of Thyroid Hormone Deiodination Reactions.

Thyroid hormone (TH) signaling is strictly regulated by iodothyronine deiodinase activity, which both preserves the circulating levels of the biologically active triiodothyronine (T3) and regulates TH homeostasis at the local level, in a cell- and time-dependent manner. Three deiodinases have been identified—namely iodothyronine deiodinase 1 (DIO1), DIO2, and DIO3—that differ in their catalytic properties and tissue distribution. The deiodinases represent a dynamic system that changes in the different stages of life according to their functions and roles in various cell types and tissues. Deiodinase activity at the tissue level permits cell-targeted fine regulation of TH homeostasis, mediating the activation (DIO1 and DIO2) and inactivation (DIO3) of THs. Deiodinase homeostasis is the driving force that leads T3-target cells towards customized TH signaling, which takes into account both the hormonal circulating levels and the tissue-specific response. This review analyzes the complex role of deiodinases in physiological and pathological contexts, exploring new challenges and opportunities deriving from a deeper knowledge of the dynamics underlying their roles and functions.

Adaptive Thermogenesis Driving Catch-Up Fat Is Associated With Increased Muscle Type 3 and Decreased Hepatic Type 1 Iodothyronine Deiodinase Activities: A Functional and Proteomic Study.

Refeeding after caloric restriction induces weight regain and a disproportionate recovering of fat mass rather than lean mass (catch-up fat) that, in humans, associates with higher risks to develop chronic dysmetabolism. Studies in a well-established rat model of semistarvation-refeeding have reported that catch-up fat associates with hyperinsulinemia, glucose redistribution from skeletal muscle to white adipose tissue and suppressed adaptive thermogenesis sustaining a high efficiency for fat deposition. The skeletal muscle of catch-up fat animals exhibits reduced insulin-stimulated glucose utilization, mitochondrial dysfunction, delayed in vivo contraction-relaxation kinetics, increased proportion of slow fibers and altered local thyroid hormone metabolism, with suggestions of a role for iodothyronine deiodinases. To obtain novel insights into the skeletal muscle response during catch-up fat in this rat model, the functional proteomes of tibialis anterior and soleus muscles, harvested after 2 weeks of caloric restriction and 1 week of refeeding, were studied. Furthermore, to assess the implication of thyroid hormone metabolism in catch-up fat, circulatory thyroid hormones as well as liver type 1 (D1) and liver and skeletal muscle type 3 (D3) iodothyronine deiodinase activities were evaluated. The proteomic profiling of both skeletal muscles indicated catch-up fat-induced alterations, reflecting metabolic and contractile adjustments in soleus muscle and changes in glucose utilization and oxidative stress in tibialis anterior muscle. In response to caloric restriction, D3 activity increased in both liver and skeletal muscle, and persisted only in skeletal muscle upon refeeding. In parallel, liver D1 activity decreased during caloric restriction, and persisted during catch-up fat at a time-point when circulating levels of T4, T3 and rT3 were all restored to those of controls. Thus, during catch-up fat, a local hypothyroidism may occur in liver and skeletal muscle despite systemic euthyroidism. The resulting reduced tissue thyroid hormone bioavailability, likely D1- and D3-dependent in liver and skeletal muscle, respectively, may be part of the adaptive thermogenesis sustaining catch-up fat. These results open new perspectives in understanding the metabolic processes associated with the high efficiency of body fat recovery after caloric restriction, revealing new implications for iodothyronine deiodinases as putative biological brakes contributing in suppressed thermogenesis driving catch-up fat during weight regain.

Iodothyronine deiodinases and reduced sensitivity to thyroid hormones.

Iodothyronine deiodinases are selenoproteins that regulate thyroid hormone metabolism. Of the three types of deiodinases, type 2 is the major regulator of intracellular triiodothyronine concentration in both the hypothalamus and pituitary, and therefore the major regulator of thyrotropin secretion. A defect in iodothyronine deiodinase activity can lead to a reduced sensitivity to thyroid hormones action and the most recent literature includes these defects in the so-called "syndromes of reduced sensitivity to thyroid hormones". To date, the pathogenic variants of the selenocysteine insertion sequence-binding protein 2 (SECISBP2) gene are the first and only inherited disorder of iodothyronine metabolism described. Moreover, there is a growing interest in understanding the possible role of polymorphisms of DIO1 and DIO2 genes in some pathological conditions and in determining the requirement of levothyroxine replacement and the role of combined levothyroxine-liothyronine therapy in carrying subjects affected by hypothyroidism and who need replacement therapy. Results on this topic are still conflicting and more studies are needed to assess the efficacy of combined levothyroxine-liothyronine replacement therapy in this subset of patients.

Acute microcystin-LR exposure interfere thyroid hormones homeostasis in adult zebrafish (Danio rerio).

Microcystin-LR (MC-LR) in the aquatic environment may disturb thyroid hormone (TH) homeostasis. It is not clear how MC-LR affects downstream biological processes after TH disturbance. After exposure to 50, 100, 200 and 400 μg/L MC-LR for 24, 48, 72, or 96 h, alterations of the TH metabolism of adult zebrafish at thyroxine (T4), triiodothyronine (T3) levels, and iodothyronine deiodinase (Dio) activity, were observed. After exposure to MC-LR at 400 μg/L, T3 and T4 levels decreased significantly in females (p < 0.05) and returned to normal levels at 96 h. In males, T4 levels were not significantly different between groups. The expression of corticotropin releasing hormone, thyroid-stimulating hormone beta subunit, transthyretin, sodium/iodide cotransporter, thrombopoietin, thyroid hormone receptor alpha and beta changed, but not in a dose-dependent manner. Acute MC-LR exposure induced a negative feedback regulation of the hypothalamic-pituitary-thyroid axis in adult zebrafish, and females were more sensitive than males. In conclusion, acute MC-LR exposure disrupted the TH metabolism by altering Dio activity and gene expression of the HPT axis; these changes may affect the complement system through regulation of c9 mRNA synthesis.

Waterborne exposure to microcystin-LR alters thyroid hormone levels, iodothyronine deiodinase activities, and gene transcriptions in juvenile zebrafish (Danio rerio).

This study investigated the effects of microcystin (MC) on the regulation of thyroid hormone (TH) metabolism in juvenile zebrafish exposed to MC-LR. The results showed that acute MC-LR exposure at concentrations ranging from 50 μg/L to 400 μg/L led to significant reductions in thyroxine (T4) and triiodothyronine (T3) levels in juvenile zebrafish. The transcription levels of genes involved in TH synthesis, such as corticotropin-releasing hormone (crh), thyroid-stimulating hormone (tsh), thyroid peroxidase (tpo) and transthyretin (ttr), were significantly decreased followed by an increase after MC-LR exposure. Transcription of the TH nuclear receptors (tr-α and tr-β) was significantly reduced during the exposure period. Moreover, the activities of iodothyronine deiodinase type Ⅰ (ID1) and iodothyronine deiodinase type Ⅱ (ID2) showed initially decreased and then increased trend, while the activity of iodothyronine deiodinase type Ⅲ (ID3) significantly decreased during MC-LR exposure. In addition, the effect of MC-LR on deiodinase activities and T4 contents were important causes of the decreased T3 at the early exposure stage. These results indicated that acute MC-LR exposure significantly interfered with the transcription of genes related to TH synthesis, transport and metabolism, and affected normal function of the thyroid which leads to decrease of T4 and T3 in juvenile zebrafish. Therefore, the thyroid function is susceptible to interference by MC-LR, and it may cause adverse effects on the growth and development of juvenile zebrafish.

Short-term exercise training improves cardiac function associated to a better antioxidant response and lower type 3 iodothyronine deiodinase activity after myocardial infarction.

AimsWe assessed the effects of a short-term exercise training on cardiac function, oxidative stress markers, and type 3 iodothyronine deiodinase (D3) activity in cardiac tissue of spontaneously hypertensive rats (SHR) following experimental myocardial infarction (MI).MethodsTwenty-four SHR (aged 3 months) were allocated to 4 groups: sham+sedentary, sham+trained, MI+sedentary and MI+trained. MI was performed by permanent ligation of the coronary artery. Exercise training (treadmill) started 96 hours after MI and lasted for 4 weeks (~60% maximum effort, 4x/week and 40 min/day). Cardiac function (echocardiography), thioredoxin reductase (TRx), total carbonyl levels, among other oxidative stress markers and D3 activity were measured. A Generalized Estimating Equation was used, followed by Bonferroni’s test (p<0.05).ResultsMI resulted in an increase in left ventricular mass (p = 0.002) with decreased cardiac output (~22.0%, p = 0.047) and decreased ejection fraction (~41%, p = 0.008) as well as an increase in the carbonyl levels (p = 0.001) and D3 activity (~33%, p<0.001). Exercise training resulted in a decrease in left ventricular mass, restored cardiac output (~34%, p = 0.048) and ejection fraction (~20%, p = 0.040), increased TRx (~85%, p = 0.007) and reduced carbonyl levels (p<0.001) and D3 activity (p<0.001).ConclusionsOur short-term exercise training helped reverse the effects of MI on cardiac function. These benefits seem to derive from a more efficient antioxidant response and lower D3 activity in cardiac tissue.

Exposure to nitrite alters thyroid hormone levels and morphology in Takifugu rubripes

Nitrite (NO2−) can act as a toxic nitrogenous compound with the potential to disrupt endocrine systems in fish. The aim of the present study was to investigate the effects of nitrite on the thyroid endocrine system of Takifugu rubripes. Fish were exposed to 0, 0.5, 1, 3, and 6 mM nitrite concentrations. Blood was collected to assay the concentrations of thyroid-stimulating hormone (TSH), thyroxine (T4), triiodothyronine (T3), free thyroxine (FT4), free triiodothyronine (FT3), and 3,3,5′-triiodothyronine (rT3), as well as the activity of iodothyronine deiodinases (Dio1, Dio2, and Dio3,) after 0, 12, 24, 48, and 96 h of exposure to nitrite. The first branchial arch to the third branchial arch of T. rubripes were sampled and fixed, and thyroid morphology was observed. The results showed that exposure to nitrite significantly increased the concentrations of TSH, T3, FT3, and reduced the concentrations of T4, FT4, and rT3. The activity of Dio1 and Dio2 increased significantly, whereas Dio3 activity decreased significantly. Additionally, thyroid follicles degenerated and became blurred and most colloid material disappeared 96 h after exposure to high nitrite concentrations. Based on these results, high nitrite concentration exposure can disturb thyroid hormone homeostasis, alter thyroid follicle morphology, and result in thyroid endocrine toxicity.

N-Acetylcysteine Prevents Low T3 Syndrome and Attenuates Cardiac Dysfunction in a Male Rat Model of Myocardial Infarction.

Nonthyroidal illness syndrome (NTIS) affects patients with myocardial infarction (MI). Oxidative stress has been implicated as a causative factor of NTIS, and reversed via N-acetylcysteine (NAC). Male Wistar rats submitted to left anterior coronary artery occlusion received NAC or placebo. Decreases in triiodothyronine (T3) levels were noted in MI-placebo at 10 and 28 days post-MI, but not in MI-NAC. Groups exhibited similar infarct areas whereas MI-NAC exhibited higher ejection fraction than did MI-placebo. Left ventricular systolic and diastolic diameters were also preserved in MI-NAC, but not in MI-placebo. Ejection fraction was positively correlated with T3 levels. Oxidative balance was deranged only in MI-placebo animals. Increased type 3 iodothyronine deiodinase expression was detected in the cardiomyocytes of MI-placebo compared with normal heart tissue. NAC was shown to diminish type 3 iodothyronine deiodinase expression and activity in MI-NAC. These results show that restoring redox balance by NAC treatment prevents NTIS- related thyroid hormone derangement and preserves heart function in rats subjected to MI.

A Nonradioactive DEHAL Assay for Testing Substrates, Inhibitors, and Monitoring Endogenous Activity.

Iodotyrosine deiodinase (DEHAL1) is a crucial enzyme in iodine homeostasis. Unbound mono- and diiodotyrosines are indispensable byproducts of thyroid hormone biosynthesis. Their iodine needs to be recovered to avoid iodine deficiency, as observed in genetic defects in DEHAL1. Despite its importance, the enzyme is rarely studied. The deiodination process can be monitored by radioactive tracers or via techniques involving mass spectrometry. However, isotope-labeled molecules are expensive, not always commercially available, and their use is legally restricted, whereas mass spectrometry requires sophisticated, costly, and sensitive instrumentation. To circumvent these difficulties, we adapted the nonradioactive iodothyronine deiodinase assay to determine DEHAL1 activity by a colorimetric readout, based on the Sandell-Kolthoff reaction. DEHAL1 was recombinantly expressed and used to optimize the assay in microtiter format. We applied the setup to scenarios of alternative substrate screening or search for compounds potentially acting as endocrine disrupting compounds, without identifying novel readily accepted substrates or inhibitors yet. Next, the assay was adapted to ex vivo material, and activity was reliably determined from rodent kidney and other tissues. Analyzing two mouse models of hyperthyroidism, we observed a decreased renal Dehal1 activity and mRNA expression. Our results show that this nonradioactive DEHAL1 assay is suited to screen for potential endocrine disrupters and to monitor endogenous Dehal1 expression. We harmonized the assay protocols to enable iodothyronine deiodinase and DEHAL1 activity measurements from the same samples. Hereby, a more complete view on iodine metabolism by these predominant deiodinating activities can be obtained from a given sample by a similar process flow.

Type 2 Iodothyronine Deiodinase Activity Is Required for Rapid Stimulation of PI3K by Thyroxine in Human Umbilical Vein Endothelial Cells.

Thyroid hormones (THs) exert a number of physiological effects on the cardiovascular system. Some of the nongenomic actions of T3 are achieved by cross coupling the TH receptor (TR) with the phosphatidylinositol 3-kinase (PI3K)/protein kinase Akt (Akt) pathway. We observed that both T3 and T4 rapidly stimulated Akt phosphorylation and Ras-related C3 botulinum toxin substrate 1 (Rac1) activation, which resulted in cell migration, in a PI3K-dependent manner in human umbilical vein endothelial cells (HUVECs). We identified the expression of type 2 iodothyronine deiodinase (D2), which converts T4 to T3, and TRα1 in HUVECs. D2 activity was significantly stimulated by (Bu)2cAMP in HUVECs. The blockade of D2 activity through transfection of small interfering RNA (siRNA) specific to D2 as well as by addition of iopanoic acid, a potent D2 inhibitor, abolished Akt phosphorylation, Rac activation, and cell migration induced by T4 but not by T3. The inhibition of TRα1 expression by the transfection of siRNA for TRα1 canceled Akt phosphorylation, Rac activation, and cell migration induced by T3 and T4. These findings suggest that conversion of T4 to T3 by D2 is required for TRα1/PI3K-mediated nongenomic actions of T4 in HUVECs, including stimulation of Akt phosphorylation and Rac activation, which result in cell migration.

Postnatal overnutrition programs the thyroid hormone metabolism and function in adulthood.

Early overnutrition (EO) during lactation leads to obesity, leptin resistance and lower thyroid hormone (TH) levels during adulthood. To better understand the biological significance of this thyroid hypofunction, we studied the long-term effects of postnatal EO on both the function of hypothalamic-pituitary-thyroid (HPT) axis and the metabolism and action of TH. To induce EO, the litter size was reduced to three pups per litter (small litter (SL) group) on the third day of lactation. In the controls (normal litter group), litter size was adjusted to 10 pups per litter. Rats were killed at PN180. TRH content and in vitro TSH were evaluated. Iodothyronine deiodinase (D1 and D2) activities were measured in different tissues. Mitochondrial α-glycerol-3-phosphate dehydrogenase (mGPD), uncoupling protein 1 (UCP1) and TH receptor (TRβ1) were evaluated to assess TH action. The SL group presented lower TRH, intra-pituitary and released TSH levels, despite unchanged plasma TSH. They presented lower D1 activity in thyroid, muscle and white adipose tissue (WAT) and higher D2 activity in the hypothalamus, pituitary, brown adipose tissue (BAT) and WAT, which confirmed the hypothyroidism. UCP1 in BAT and TRβ1 in WAT were decreased, which can contribute to a lower catabolic status. Despite the lower TH, the D2 activity in the thyroid, heart and testes was unchanged. Hepatic D1, mGPD and TRβ1 were also unchanged in SL rats, suggesting that the TH conversion and action were preserved in the liver, even with lower TH. Thus, this model indicates that postnatal EO changes thyroid function in adult life in a tissue-specific way, which can help in the understanding of obesogenesis.

Disruption of type 2 iodothyronine deiodinase activity in cultured human glial cells by polybrominated diphenyl ethers.

Polybrominated diphenyl ether (PBDE) flame retardants are endocrine disruptors and suspected neurodevelopmental toxicants. While the direct mechanisms of neurodevelopmental toxicity have not been fully elucidated, it is conceivable that alterations in thyroid hormone levels in the developing brain may contribute to these effects. Cells within the brain locally convert thyroxine (T4) to the biologically active triiodothyronine (T3) through the action of the selenodeiodinase type 2 iodothyronine deiodinase (DIO2). Previous studies have demonstrated that PBDEs can alter hepatic deiodinase activity both in vitro and in vivo; however, the effects of PBDEs on the deiodinase isoforms expressed in the brain are not well understood. Here, we studied the effects of several individual PBDEs and hydroxylated metabolites (OH-BDEs) on DIO2 activity in astrocytes, a specialized glial cell responsible for production of more than 50% of the T3 required by the brain. Primary human astrocytes and H4 glioma cells were exposed to individual PBDEs or OH-BDEs at concentrations up to 5 μM. BDE-99 decreased DIO2 activity by 50% in primary astrocyte cells and by up to 80% in the H4 cells at doses of ≥500 nM. 3-OH-BDE-47, 6-OH-BDE-47, and 5'-OH-BDE-99 also decreased DIO2 activity in cultured H4 glioma cells by 45-80% at doses of approximately 1-5 μM. Multiple mechanisms appear to contribute to the decreased DIO2 activity, including weakened expression of DIO2 mRNA, competitive inhibition of DIO2, and enhanced post-translational degradation of DIO2. We conclude that decreases in DIO2 activity caused by exposure to PBDEs may play a role in the neurodevelopmental deficits caused by these toxicants.

The Role of Thyroid Hormone on Functional Organization in the Cerebellum

The thyroid hormone (L-triiodothyronine (T3), thyroxine (T4)) plays a critical role in cerebellar development. Circulating T4 preferentially crosses the blood-brain barrier through several amino acid transporters. Then, it is taken up by astrocyte to convert into T3, which is a bioactive ligand for nuclear thyroid hormone receptor (TR). Liganded TR regulates the expression of target genes that may play an important role in cerebellar development and function. Thus, thyroid hormone deficiency results in the change in neuronal excitability and aberrant neurotransmitter transport, which induces abnormal motor coordination, decreased locomotor activity, and increased anxiety. In addition to genomic action of the thyroid hormone, T4 alters actin polymerization and iodothyronine deiodinase activity in astrocyte through non-genomic pathway, which may also contribute to the normal brain development. Taken together, thyroid hormone regulates cerebellar development and plasticity through multiple signal transduction pathways.

Molecular mechanism of thyroid hormone action in carcinogenesis

Thyroid hormones, 3,5,3'5'-tetraiodothyronine (thyroxine) and 3,5,3'-triiodothyronine (T3) contribute to the regulation of key cellular processes, including proliferation, differentiation, apoptosis and metabolism. Thus, deregulation of thyroid hormone pathway can result in serious disturbances of cellular physiology such as those observed in tumoral transformation. The intra- and extracellular concentrations of thyroid hormones (TH) are regulated by three types of iodothyronine deiodinases (DIO1, DIO2, and DIO3). These enzymes catalyze two types of reactions: deiodination in position 5' of iodothyronines (catalyzed by DIO1 and DIO2), resulting in activation of thyroid hormones, and deiodination in position 5 of iodothyronines (catalyzed by DIO1 and DIO3), which results in inactivation of thyroid hormones. Changes in expression and activity of iodothyronine deiodinases result in modulations of thyroid hormone concentrations and thus influence cellular processes. The actions of thyroid hormone are mediated by thyroid hormone receptors (TRs) which influence intracellular processes via modulation of expression of target genes. In this process, known as genomic mechanism of TH action, thyroid hormone receptors act as transcription factors whose activity is modulated by the presence of ligand, T3. TH can also influence cellular processes via non-genomic pathways in which TH modulate activity of membrane or intracellular receptors and activate different intracellular signaling pathways, involving protein kinases, for instance PI3K, and Akt/PKB. In the process of carcinogenesis the signaling pathway of thyroid hormones is disturbed. This is illustrated by observations of mutations and deregulated expression of thyroid hormone receptors, altered expression and activity of iodothyronine deiodinases as well as decreased intracellular concentrations of T3 in tumours. Whether these changes play a causative role in the process of carcinogenesis is currently a matter of discussion. In vitro studies in cancer cell lines and mouse models showed that increased expression of DIO3 together with enhanced proteasomal degradation of DIO2 in basal cell carcinoma stimulates tumoral proliferation. Moreover, thyroid hormone receptor beta-1 was shown to act as a powerful suppressor of tumour invasiveness and metastasis in mouse models of breast cancer and hepatocarcinoma. These studies suggest that local tissue hypothyroidism may rather support tumoral transformation and even initiate enhanced cancerous proliferation. In contrast, studies on the effects of systemic hypothyroidism and cancer risk show opposite or conflicting results. For instance, hypothyroidism was associated with increased risk of hepatocellular carcinoma, and cancers of lung and prostate. In case of breast cancer, different studies reported that hypothyroidism was linked to reduced risk of the disease in humans while when studied in mice, hypothyroidism reduced tumour growth but enhanced metastasis. Finally, studies performed on cell line and mouse xenografts of thyroid and kidney cancers showed that inhibition of integrin-mediated thyroid hormone signaling can effectively result in inhibition of tumour growth. All these studies show that the role of thyroid hormones in carcinogenic process is complex and presumably depends on type of cancer and extra- or intracellular effects of TH signaling. The results showing benefits from the use of TH pathway genes in cancer treatment and diagnosis suggest that further and more in depth studies are needed.

The effect of low dose lipopolysaccharide on thyroid hormone-regulated actin cytoskeleton modulation and type 2 iodothyronine deiodinase activity in astrocytes

Systemic infection/inflammation can severely interfere with brain development. Lipopolysaccharide (LPS) is a major cell wall component of gram-negative bacteria and commonly used to model the response by infections. Since perinatal exposure to LPS shows neurodevelopmental defects partly similar to those seen in perinatal hypothyroidism, we examined the effect of LPS on thyroxin (T4)-mediated signalings in astrocytes. Initially, C6 rat glioma-derived clonal cells were used, whose biological nature is similar to that of astrocytes. To measure the effects of LPS and T4, actin polymerization and D2 activity assays were carried out. LPS treatment (10 ng/mL) markedly induced actin depolymerization, whereas 10 nM T4 promoted actin polymerization. Furthermore, T4 partly rescued LPS-induced actin depolymerization. LPS treatment (10 ng/mL) increased D2 activity, whereas T4 (10 nM) suppressed this activity. T4 restored LPS-increased D2 activity at 10 nM. LPS-induced actin depolymerization and D2 activity were blocked by p38 MAP kinase inhibitor. Such effects were not seen in T4-mediated changes. Furthermore, similar results were found in the cerebellar primary astrocyte. These results indicate that, although LPS affects T4-regulated cellular events such as actin polymerization and D2 activity, which may induce neurodevelopmental defects similar to those in perinatal hypothyroidism, LPS signaling pathways are independent of T4 signaling pathways.

Deiodinase Activities in Thyroids and Tissues of Iodine-Deficient Female Rats

Severe iodine deficiency is characterized by goiter, preferential synthesis, and secretion of T(3) in thyroids, hypothyroxinemia in plasma and tissues, normal or low plasma T(3), and slightly increased plasma TSH. We studied changes in deiodinase activities and mRNA in several tissues of rats maintained on low-iodine diets (LIDs) or LIDs supplemented with iodine (LID+I). T(4) and T(3) concentrations decreased in plasma, tissues, and thyroids of LID rats, and T(4) decreased more than T(3) (50%). The highest type 1 iodothyronine deiodinase (D1) activities were found in the thyroid, kidney, and the liver; pituitary, lung, and ovary had lower D1 activities; but the lowest levels were found in the heart and skeletal muscle. D1 activity decreased in all tissues of LID rats (10-40% of LID+I rats), except for ovary and thyroids, which D1 activity increased 2.5-fold. Maximal type 2 iodothyronine deiodinase (D2) activities were found in thyroid, brown adipose tissue, and pituitary, increasing 6.5-fold in thyroids of LID rats and about 20-fold in the whole gland. D2 always increased in response to LID, and maximal increases were found in the cerebral cortex (19-fold), thyroid, brown adipose tissue, and pituitary (6-fold). Lower D2 activities were found in the ovary, heart, and adrenal gland, which increased in LID. Type 3 iodothyronine deiodinase activity was undetectable. Thyroidal Dio1 and Dio2 mRNA increased in the LID rats, and Dio1 decreased in the lung, with no changes in mRNA expression in other tissues. Our data indicate that LID induces changes in deiodinase activities, especially in the thyroid, to counteract the low T(4) synthesis and secretion, contributing to maintain the local T(3) concentrations in the tissues with D2 activity.
