Synthesis, Crystal Structure, Hirshfeld Surface Analysis, and Computational Approach of a New Pyrazolo[3,4-g]isoquinoline Derivative as Potent against Leucine-Rich Repeat Kinase 2 (LRRK2).

Ethyl-2-((8-cyano-3,5,9a-trimethyl-1-(4-oxo-4,5-dihydrothiazol-2-yl)-4-phenyl-3a,4,9,9a-tetrahydro-1H-pyrazolo[3,4-g]isoquinolin-7-yl)thio)acetate (5) was synthesized, and its structure was characterized by IR, MS, and NMR (1H and 13C) and verified by a single-crystal X-ray structure determination. Compound 5 adopts a "pincer" conformation. In the crystal, the hydrogen bonds of -H···O, C-H···O, and O-H···S form thick layers of molecules that are parallel to (101). The layers are linked by C-H···π(ring) interactions. The Hirshfeld surface analysis shows that intermolecular hydrogen bonding plays a more important role than both intramolecular hydrogen bonding and π···π stacking in the crystal. The intramolecular noncovalent interactions in 5 were studied by QTAIM, NCI, and DFT-NBO calculations. Based on structural activity relationship studies, leucine-rich repeat kinase 2 (LRRK2) was found to bind 5 and was further subjected to molecular docking studies, molecular dynamics, and ADMET analysis to probe potential drug candidacy.

Elucidating the potential of nitrogen-containing heterocyclic core-based non-fullerene acceptors for organic photovoltaic applications

Intramolecular noncovalent interactions are recognized as an efficient route to construct non-fused-core-based non-fullerene acceptors (NFAs) for efficient and cost-effective organic solar cells (OSCs). Herein, we engineered ten distinct structured NFAs molecules based on nitrogen-containing six-membered heterocycle cores. We incorporated various efficient functional groups into the synthetic reference molecule (RM) to design a new efficient series of NFAs (RM1-RM10). These materials are intensively investigated by performing advanced quantum chemical simulations, and the results are compared with those of synthetic RM molecules. We revealed the potential of these materials by investigating their optical, electronic, optoelectronic, and photovoltaic characteristics. By implementing various theoretical models, we explore the inner hidden potential of this designed series (RM1-RM10), such as density of state, transition density matrix, and electrostatic potential studies have been performed. Our applied theoretical modeling approach enables these photovoltaic materials (RM1-RM10) to have customized attributes by tuning their energy levels, binding energy, optical features, and also the photovoltaic characteristics of these materials. Newly developed molecules (RM1-RM10) show promising optoelectronic properties, including a lower energy gap (ranging from 1.81 eV to 1.63 eV) and an absorbed maximum absorption wavelength (760.36 nm), compared to the reference values, which have an energy gap of 1.81 eV and a wavelength of 684.16 nm. Moreover, to demonstrate the charge transfer behavior of these materials, a donor: acceptor (PTB7-Th:RM9) complex study is also performed to reveal the charge transfer phenomenon at the donor: acceptor interface. Therefore, our presented molecular modeling strategy holds significant potential for manufacturing materials with tailored qualities, which is crucial in advancing the organic photovoltaics field.

Non-Fused π-Extension of Endcaps of Small Molecular Acceptors Enabling High-Performance Organic Solar Cells.

The modular structure of small molecular acceptors (SMAs) allows for versatile modifications of the materials and boosts the photovoltaic efficiencies of organic solar cells (OSCs) in recent years. As a critical component, the endcaps of SMAs have been intensively investigated and modified to control the molecular aggregation and photo-electronic conversion. However, most of the studies focus on halogenation or π-fusion extension of the endcap moieties, but overlook the non-fused π-extension approach, which could be a promising strategy to balance the self-aggregation and compatibility behaviors. Herein, we reported two new acceptors namely BTP-Th and BTP-FTh based on non-fused π-extension of the endcap by chlorinated-thiophene, of which the latter molecule has better co-planarity and crystallinity because of the intramolecular noncovalent interactions. Paired with donor PBDB-T, the optimal device of BTP-FTh reveals a greater efficiency of 14.81% that that of BTP-Th (13.91%). Nevertheless, the BTP-Th based device realizes a lower energy loss, enabling BTP-Th as a good candidate to serve as guest acceptor. As a result, the ternary solar cells of PM6:BTP-eC9:BTP-Th output a champion efficiency up to 18.71% with enhanced open-circuit voltage. This study highlights the significance of rational decoration of endcaps for the design of high-performance SMAs and photovoltaic cells.

Predisposition in Dynamic Covalent Chemistry: The Role of Non-Covalent Interactions in the Assembly of Tetrahedral Boronate Cages.

Directional bonding strategies guide the design of complex molecular architectures, yet challenges arise due to emergent behavior. Rigid structures face geometric constraints and sensitivity to mismatches, hindering the efficient assembly of molecular organic cages (MOCs). Harnessing intramolecular non-covalent interactions offers a promising solution, broadening geometrical possibilities and enhancing adaptability to boost assembly yields. However, identifying these interactions remains challenging, with their full potential sometimes latent until final assembly. This study explores these challenges by synthesizing boronic acid tripods with varied oxygen positions at the tripodal feet and investigating their role in assembling tetrahedral boronate MOCs. Our results reveal substantial differences in the assembly efficiency among tripods. While the building blocks with oxygen in the benzylic position relative to the central aromatic ring form the MOCs in high yields, those with the oxygen atom directly bound to the central aromatic ring, only yield traces. Through X-ray crystallography and DFT analyses, we elucidate how intramolecular interactions profoundly influence the geometry of the building blocks and cages in a relay-like fashion, highlighting the importance of considering intramolecular interactions in the rational design of (supra)molecular architectures.

A highly crystalline face-on π-conjugated polymer based on alkoxythiophene-flanked benzobisthiazole for organic photovoltaics

AbstractThe use of noncovalent intramolecular interactions constitutes a powerful design strategy for preparing π-conjugated polymers featuring high backbone coplanarities and thereby high crystallinities. Herein, we report the design and synthesis of an alkoxythiophene-flanked benzobisthiazole (BBTz) as a new building unit for π-conjugated polymers, which was subsequently copolymerized to give a simple BBTz-bithiophene copolymer with alkyl and alkoxy groups (PDBTz2). Owing to the S···O noncovalent intramolecular interactions between the alkoxy oxygens and thiazole sulfurs in BBTz, PDBTz2 showed greater coplanarity and crystallinity than its alkyl counterpart, PDBTz1. Interestingly, the backbone orientation was completely altered from the edge-on orientation observed for PDBTz1 to a face-on orientation for PDBTz2, which is preferable for organic photovoltaics (OPVs). In addition, the electron-donating nature of the alkoxy group increased the HOMO energy level of PDBTz2 compared to that of PDBTz1, which enabled photoinduced hole transfer from a nonfullerene acceptor, Y6, to the polymer. As a result, the short-circuit current density of an organic photovoltaic cell based on PDBTz2 and Y6 was significantly greater than that of a cell based on PDBTz1 and Y6. This study confirmed that alkoxythiophene-flanked BBTz is a promising building unit for high-performance π-conjugated polymers.

Recent Advances in π-Stacking Interaction-Controlled Asymmetric Synthesis.

The π-stacking interaction is one of the most important intramolecular and intermolecular noncovalent interactions in organic chemistry. It plays an important role in stabilizing some structures and transition states in certain reactions via both intramolecular and intermolecular interactions, facilitating different selectivities, such as chemo-, regio-, and stereoselectivities. This minireview focuses on the recent examples of the π-stacking interaction-controlled asymmetric synthesis, including auxiliary-induced asymmetric synthesis, kinetic resolution, asymmetric synthesis of helicenes and heterohelicenes, and multilayer 3D chiral molecules.

Efficient all-small-molecule organic solar cells processed with non-halogen solvent

All-small-molecule organic solar cells with good batch-to-batch reproducibility combined with non-halogen solvent processing show great potential for commercialization. However, non-halogen solvent processing of all-small-molecule organic solar cells are rarely reported and its power conversion efficiencies are very difficult to improve. Herein, we designed and synthesized a small molecule donor BM-ClEH that can take advantage of strong aggregation property induced by intramolecular chlorine-sulfur non-covalent interaction to improve molecular pre-aggregation in tetrahydrofuran and corresponding micromorphology after film formation. Tetrahydrofuran-fabricated all-small-molecule organic solar cells based on BM-ClEH:BO-4Cl achieved high power conversion efficiencies of 15.0% in binary device and 16.1% in ternary device under thermal annealing treatment. In contrast, weakly aggregated BM-HEH without chlorine-sulfur non-covalent bond is almost inefficient under same processing conditions due to poor pre-aggregation induced disordered π-π stacking, indistinct phase separation and exciton dissociation. This work promotes the development of non-halogen solvent processing of all-small-molecule organic solar cells and provides further guidance.

Conformational polymorphs of the homoleptic scorpionate sandwich complex [(TpPh,Me)2Ni], TpPh,Me=hydrotris(3-phenyl-5-methylpyrazol-1-yl)borate: Ligand substituent effects include intramolecular non-covalent interactions

The homoleptic scorpionate sandwich complex [(TpPh,Me)2Ni] (1), where TpPh,Me = hydrotris(3-phenyl-5-methylpyrazol-1-yl)borate, crystallizes as a mixture of two polymorphs. A major monoclinic fraction crystallizes as well-formed parallelepipeds and a minor triclinic fraction forms irregularly shaped, anhedral crystals. These exhibit distinct melting points (288 and 294 °C, respectively) as observed by differential scanning calorimetry. We obtained X-ray crystal structures of both polymorphs at 100 K. The complex adopts different conformations between the two lattices. These are supported by distinctive ensembles of intramolecular non-covalent interactions, specifically π-stacking and perpendicular C–H···π contacts between opposing 3-phenylpyrazolyl arms on the co-ligands. The conformation found in the major monoclinic polymorph consists of a counter-rotation of the ligands that reduces the ideal complex symmetry from D3d to D3 and results in three-fold pair-wise association of 3-phenylpyrazolyl arms. In contrast, an alternate conformation of 1 in the minor triclinic polymorph consists of two triads, with an ideal complex symmetry of C2h. Also affected are the tetragonality of the ligand fields and interligand N–Ni–N′ bond angles. A review of sandwich complex structures [(TpR)2M], where R is an aromatic substituent, shows these conformations are adopted broadly. Hence, substituent effects in second-generation scorpionate ligands are not merely steric in origin.

Biginelli dihydropyrimidines carrying azole rings: Synthesis, computational studies, and evaluation of alpha‐glucosidase inhibitory and antimicrobial activities

In the current study, we designed three novel compounds by merging two valuable nitrogen-containing pharmacophores, namely dihydropyrimidine (DHPM) and azole, within a single molecule. To obtain the target molecules, azole-linked benzaldehydes were initially synthesized via nucleophilic aromatic substitution reaction between 4-fluorobenzaldehyde and pyrazole, imidazole, or 1,2,4-triazole. Afterward, Biginelli reaction was applied to yield azole-carrying DHPMs. Following structural confirmation, the obtained compounds were tested for their alpha (α)-glucosidase inhibitory and antimicrobial activities. According to the biological data, DHPM-P and DHPM-T showed strong inhibition values whereas DHPM-I failed to effectively inhibit the α-glucosidase enzyme. This situation was explained by molecular docking studies of all compounds into the binding site of alpha-glucosidase. The title compounds also presented moderate antibacterial and antifungal activities. Computational methods based on density functional tight binding (DFTB) and density functional theory (DFT) calculations, along with molecular dynamics (MD) simulations were used to analyze the reactive properties of the compounds. A combination of DFTB and DFT calculations was applied to obtain the lowest energy conformations of the molecules. Further computational analysis included calculations of local reactivity descriptors, such as molecular electrostatic potential and average local ionization energy, by applying DFT calculations. DFT calculations were also used to study intramolecular non-covalent interactions. MD simulations were employed to understand how the title molecules behave in water and to obtain their solubility parameters.

Noncovalent Interaction Boosts Performance and Stability of Organic Solar Cells Based on Giant-Molecule Acceptors.

Designing giant-molecule acceptors is deemed as an up-and-coming strategy to construct stable organic solar cells (OSCs) with high performance. Herein, two giant dimeric acceptors, namely, DYV and DYFV, have been designed and synthesized by linking two Y-series derivatives with a vinyl unit. DYFV exhibits more red-shifted absorption, down-shifted energy levels, and enhanced intermolecular packing than DYV because the intramolecular noncovalent interaction (H···F) of DYFV leads to better coplanarity of the backbone. The D18:DYFV film owns a distinct nanofibrous nanophase separation structure, a more dominant face-on orientation, and more balanced carrier mobilities. Therefore, the D18:DYFV OSC achieves a higher photoelectron conversion efficiency of 17.88% and a longer-term stability with a t80 over 45,000 h compared with the D18:DYV device. The study demonstrates that the intramolecular noncovalent interaction is a superior strategy to design giant-molecule acceptors and boost the photovoltaic performance and stability of the OSCs.

Manipulating the functionality and structures of π-conjugated polymers utilizing intramolecular noncovalent interactions towards efficient organic photovoltaics.

Careful control of electronic properties, structural order, and solubility of π-conjugated polymers is central to the improvement of organic photovoltaic (OPV) performance. In this work, we designed and synthesized a series of naphthobisthiadiazole-quaterthiophene copolymers by systematically replacing the alkyl groups with ester groups and changing the position of the fluorine groups in the quaterthiophene moiety. These alterations lowered the HOMO and LUMO energy levels and systematically varied the combination of intramolecular noncovalent interactions such as O⋯S and F⋯S interactions in the backbone. More importantly, although the introduction of such noncovalent interactions often lowers the solubility owing to the interlocking of backbone linkages, we found that careful design of the noncovalent interactions afforded polymers with relatively high solubility and high crystallinity at the same time. As a result, the power conversion efficiency of OPV cells that used fullerene (PC61BM) and nonfullerene (Y12) as the acceptor was improved. Our work offers important information for the development of high-performance π-conjugated polymers for OPVs.

Investigating the diastereoselective synthesis of a macrocycle under Curtin-Hammett control.

This work sheds new light on the stereoselective synthesis of chiral macrocycles containing twisted aromatic units, valuable π-conjugated materials for recognition, sensing, and optoelectronics. For the first time, we use the Curtin-Hammett principle to investigate a chiral macrocyclisation reaction, revealing the potential for supramolecular π-π interactions to direct the outcome of a dynamic kinetic resolution, favouring the opposite macrocyclic product to that expected under reversible, thermodynamically controlled conditions. Specifically, a dynamic, racemic perylene diimide dye (1 : 1 P : M) is strapped with an enantiopure (S)-1,1'-bi-2-naphthol group (P-BINOL) to form two diastereomeric macrocyclic products, the homochiral macrocycle (PP) and the heterochiral species (PM). We find there is notable selectivity for the PM macrocycle (dr = 4 : 1), which is rationalised by kinetic templation from intramolecular aromatic non-covalent interactions between the P-BINOL π-donor and the M-PDI π-acceptor during the macrocyclisation reaction.

Amphiphilic Polyurethane with Cluster-Induced Emission for Multichannel Bioimaging in Living Cell Systems.

The development of single-component materials with low cytotoxicity and multichannel fluorescence imaging capability is a research hotspot. In the present work, highly electron-deficient pyrazine monomers were covalently connected into a polyurethane backbone using addition polymerization with terminal poly(ethylene glycol) monomethyl ether units containing a high density of electron pairs. Thereby, an amphiphilic polyurethane-pyrazine (PUP) derivative has been synthesized. The polymer displays cluster-induced emission through compact inter- and/or intramolecular noncovalent interactions and extensive through-space electron coupling and delocalization. Molecular rigidity facilitates red-shifted emission. Based on hydrophilic/hydrophobic interactions and excitation dependence emission at low concentrations, PUP has been self-assembled into fluorescent nanoparticles (PUP NPs) without additional surfactant. PUP NPs have been used for cellular multicolor imaging to provide a variety of switchable colors on demand. This work provides a simple molecular design for environmentally sustainable, luminescent materials with excellent photophysical properties, biocompatibility, low cytotoxicity, and color modulation.

Discovery of Unusual Ajmaline-Macroline Type Bisindole Alkaloids from Alstonia macrophylla by Building Blocks-Based Molecular Networking.

Three unusual ajmaline-macroline type bisindole alkaloids, alsmaphylines A-C, together with their postulated biogenetic precursors, were isolated from the stem barks and leaves of Alstonia macrophylla via the building blocks-based molecular network (BBMN) strategy. Alsmaphyline A represents a rare ajmaline-macroline type bisindole alkaloid with an S-shape polycyclic ring system. Alsmaphylines B and C are two novel ajmaline-macroline type bisindole alkaloids with N-1-C-21' linkages, and the former possesses an unconventional stacked conformation due to the presence of intramolecular noncovalent interactions. The chemical structures including absolute configurations of alsmaphylines A-C were established by comprehensive spectroscopic analyses, electronic circular dichroism (ECD) calculations, and single-crystal X-ray crystallography. In addition, a plausible biosynthetic pathway of these bisindole alkaloids as well as their ability to promote the protein synthesis on HT22 cells were discussed.

Over 18% Efficiency Ternary Organic Solar Cells with 300nm Thick Active Layer Enabled by an Oligomeric Acceptor.

The development of high-efficiency thickness-insensitive organic solar cells (OSCs) is crucially important for the mass production of solar panels. However, increasing the active layer thickness usually induces a substantial loss in efficiency. Herein, a ternary strategy in which an oligomer DY-TF is incorporated into PM6:L8-BO system as a guest component is adopted to break this dilemma. The S···F intramolecular noncovalent interactions in the backbone endow DY-TF with a high planarity. Upon the addition of DY-TF, the crystallinity of the blend is effectively improved, leading to increased charge carrier mobility, which is highly desirable in the fabrication of thick-film devices. As a result, thin-film PM6:L8-BO:DY-TF-based device (110nm) shows a power conversion efficiency (PCE) of 19.13%. Impressively, when the active layer thickness increases to 300nm, an efficiency of 18.23% (certified as 17.8%) is achieved, representing the highest efficiency reported for 300 nm thick OSCs thus far. Additionally, blade-coated thick device (300nm) delivers a promising PCE of 17.38%. This work brings new insights into the construction of efficient OSCs with high thickness tolerance, showing great potential for roll-to-roll printing of large-area solar cells.

Synthesis and characterization of supramolecular assembly probenecid cocrystal

To improve the bioavailability and provide a lead compound for developing formulations of the highly water-insoluble prodrug probenecid (PRO), enhancing its solubility is a crucial challenge that needs to be addressed. Herein, we report the synthesis and solid-state characterization of a new pharmaceutical cocrystal, PRO-BPE, synthesized using trans-1,2-bis(4-pyridyl)ethylene (BPE) to improve the solubility of PRO. The PRO-BPE cocrystals were characterized using various analytical techniques, including differential scanning calorimetry, thermogravimetric analysis, scanning electron microscopy, Fourier transform-infrared spectroscopy, and X-ray powder diffraction. Single-crystal X-ray diffraction was performed to determine the crystal structure of PRO-BPE. PRO-BPE crystallized in the triclinic crystal system P-1 space group and its asymmetric unit consists of half a BPE molecule and one PRO molecule. The N1 atoms located at both ends of the BPE molecule form stable hydrogen bonds with two PRO carboxyl hydrogens through N1···H4—O4 (1.777 Å, 166.78°) interactions, whereas the O3 atoms of the carboxylic groups form C1—H1···O3 (2.479 Å, 132.10°) hydrogen bonds with two pyridyl hydrogens, resulting in a stable seven-membered ring structure. Furthermore, these fragments are further assembled via C—H···O intermolecular interactions to form a two-dimensional layered structure. Interestingly, the two pyridyl rings of each BPE molecule engage in π···π interactions with the benzene rings of two other non-coplanar PRO molecules. Finally, a three-dimensional framework structure is formed through intramolecular non-covalent interactions. Comprehensive characterization analyses provided further information regarding PRO-BPE cocrystal formation. Based on the equilibrium solubility results, the water solubility of the PRO-BPE cocrystal was superior to that of pure PRO, with an increase of 2.43 times. In summary, this study provides the first evidence that cocrystals could enhance the solubility of PRO and offers valuable insights into the solid-state properties of PRO-BPE cocrystals.

Near-infrared non-fused electron acceptors for efficient organic photovoltaics

Developing narrow-bandgap organic semiconductors is important to facilitate the advancement of organic photovoltaics (OPVs). Herein, two near-infrared non-fused ring acceptors (NIR NFRAs), PTBFTT-F and PTBFTT-Cl have been developed with A-πA-πD-D-πD-πA-A non-fused structures. It is revealed that the introduction of electron deficient π-bridge (πA) and multiple intramolecular noncovalent interactions effectively retained the structural planarity and intramolecular charge transfer of NFRAs, extending strong NIR photon absorption up to 950 nm. Further, the chlorinated acceptor, with the enlarged π-surface compared to the fluorinated counterpart, promoted not only molecular stacking in solid, but also the desirable photochemical stability in ambient, which are helpful to thereby improve the exciton and charge dynamics for the corresponding OPVs. Overall, this work provides valuable insights into the design of NIR organic semiconductors.

Long but Strong C-C Single Bonds: Challenges for Theory.

Theoretical challenges in describing molecules with anomalously long single C-C bonds are analyzed in terms of the relative contributions of stabilizing and destabilizing intramolecular interactions. Diamondoid dimers that are stable despite the presence of C-C bonds up to 1.7 Å long, as well as other bulky molecules stabilized due to intramolecular noncovalent interactions (London dispersions) are discussed. The unexpected stability of highly crowded molecules, such as diamondoid dimers and tert-butyl-substituted hexaphenylethanes, calls for reconsideration of the "steric effect" traditionally thought to destabilize the molecule. Alternatively, "steric attraction" helps to understand bonding in sterically overloaded molecules, whose structural and energetic analysis requires a proper theoretical description of noncovalent interactions.

Highly efficient and stable thermally activated delayed fluorescent palladium(II) complexes for organic light-emitting diodes

Transition metal complexes exhibiting thermally activated delayed fluorescence (TADF) remain underdeveloped for organic light-emitting diodes (OLEDs). Here, we describe a design of TADF Pd(II) complexes featuring metal-perturbed intraligand charge-transfer excited states. Two orange- and red-emitting complexes with efficiencies of 82 and 89% and lifetimes of 2.19 and 0.97 μs have been developed. Combined transient spectroscopic and theoretical studies on one complex reveal a metal-perturbed fast intersystem crossing process. OLEDs using the Pd(II) complexes show maximum external quantum efficiencies of 27.5 to 31.4% and small roll-offs down to 1% at 1000 cd m −2 . Moreover, the Pd(II) complexes show exceptional operational stability with LT 95 values over 220 hours at 1000 cd m −2 , benefiting from the use of strong σ-donating ligands and the presence of multiple intramolecular noncovalent interactions beside their short emission lifetimes. This study demonstrates a promising approach for developing efficient and robust luminescent complexes without using the third-row transition metals.

Realizing High-Efficiency Orange-Red Thermally Activated Delayed Fluorescence Materials through the Construction of Intramolecular Noncovalent Interactions.

The development of highly efficient orange and red thermally activated delayed fluorescence (TADF) materials for constructing full-color and white organic light-emitting diodes (OLEDs) remains insufficient because of the formidable challenges in molecular design, such as the severe radiationless decay and the intrinsic trade-off between the efficiencies of radiative decay and reverse intersystem crossing (RISC). Herein, we design two high-efficiency orange and orange-red TADF molecules by constructing intermolecular noncovalent interactions. This strategy could not only ensure high emission efficiency via suppression of the nonradiative relaxation and enhancement of the radiative transition but also create intermediate triplet excited states to ensure the RISC process. Both emitters exhibit typical TADF characteristics, with a fast radiative rate and a low nonradiative rate. Photoluminescence quantum yields (PLQYs) of the orange (TPA-PT) and orange-red (DMAC-PT) materials reach up to 94 and 87%, respectively. Benefiting from the excellent photophysical properties and stability, OLEDs based on these TADF emitters realize orange to orange-red electroluminescence with high external quantum efficiencies reaching 26.2%. The current study demonstrates that the introduction of intermolecular noncovalent interactions is a feasible strategy for designing highly efficient orange to red TADF materials.
