Effects of Medium Additives on the Mycelial Growth and Polysaccharide Biosynthesis in Submerged Culture of Bjerkandera fumosa.

Medium additives have been shown to affect the synthesis of active products in fungi. This study investigated the effects of corn stalk, poplar sawdust, Tween-80, and oleic acid on mycelial biomass and physicochemical properties, as well as the bioactivity of polysaccharides, including exopolysaccharides (EPS) and intracellular polysaccharides (IPS), in the submerged culture of Bjerkandera fumosa. Results showed that the addition of corn stalk or poplar sawdust increased the production of EPS but decreased the production of IPS; Tween-80 had less effect on the production of EPS and IPS; and oleic acid stimulated polysaccharide production significantly. Polysaccharide property analysis showed that the addition of corn stalk or poplar sawdust promoted the production of high-molecular-weight components in polysaccharides and changed the monosaccharide composition of polysaccharides, as well as increased the mannose, glucuronic acid, and xylose contents of IPS. Tween-80 and oleic acid also changed the molecular weight distribution of polysaccharides but only slightly affected the composition of monosaccharides. The bioactivity assay indicated that the polysaccharides obtained by adding corn stalk possessed high hydroxyl radical scavenging and antitumor activities. The effect of poplar sawdust was slightly weaker than that of corn stalk. EPS and IPS obtained from a culture with Tween-80 and oleic acid possessed low antioxidant activity. Moreover, their antitumor activity was improved and lost, respectively. The results obtained in this work are useful for improving the understanding of the optimization and regulation of bioactive polysaccharide production in the submerged culture of B. fumosa.

Mycelial biomass and intracellular polysaccharides production, characterization, and activities in Auricularia auricula-judae cultured with different carbon sources

The carbon source, an essential factor for submerged culture, affects fungal polysaccharides production, structures, and activities. This study investigated the impact of carbon sources, including glucose, fructose, sucrose, and mannose, on mycelial biomass and the production, structural characterization, and bioactivities of intracellular polysaccharides (IPS) produced by submerged culture of Auricularia auricula-judae. Results showed that mycelial biomass and IPS production varied with different carbon sources, where using glucose as the carbon source produced the highest mycelial biomass (17.22 ± 0.29 g/L) and IPS (1.62 ± 0.04 g/L). Additionally, carbon sources were found to affect the molecular weight (Mw) distributions, monosaccharide compositions, structural characterization, and activities of IPSs. IPS produced with glucose as the carbon source exhibited the best in vitro antioxidant activities and had the strongest protection against alloxan-damaged islet cells. Correlation analysis revealed that Mw correlated positively with mycelial biomass (r = 0.97) and IPS yield (r = 1.00), while IPS antioxidant activities correlated positively with Mw and negatively with mannose content; the protective activity of IPS was positively related to its reducing power. These findings indicate a critical structure-function relationship for IPS and lay the foundation for utilizing liquid-fermented A. aruicula-judae mycelia and the IPS in functional food production.

High-Molecular-Weight Exopolysaccharides Production from Tuber borchii Cultivated by Submerged Fermentation

Truffles are known worldwide for their peculiar taste, aroma, and nutritious properties, which increase their economic value. However, due to the challenges associated with the natural cultivation of truffles, including cost and time, submerged fermentation has turned out to be a potential alternative. Therefore, in the current study, the cultivation of Tuber borchii in submerged fermentation was executed to enhance the production of mycelial biomass, exopolysaccharides (EPSs), and intracellular polysaccharides (IPSs). The mycelial growth and EPS and IPS production was greatly impacted by the choice and concentration of the screened carbon and nitrogen sources. The results showed that sucrose (80 g/L) and yeast extract (20 g/L) yielded maximum mycelial biomass (5.38 ± 0.01 g/L), EPS (0.70 ± 0.02 g/L), and IPS (1.76 ± 0.01 g/L). The time course analysis of truffle growth revealed that the highest growth and EPS and IPS production was observed on the 28th day of the submerged fermentation. Molecular weight analysis performed by the gel permeation chromatography method revealed a high proportion of high-molecular-weight EPS when 20 g/L yeast extract was used as media and the NaOH extraction step was carried out. Moreover, structural analysis of the EPS using Fourier-transform infrared spectroscopy (FTIR) confirmed that the EPS was β-(1-3)-glucan, which is known for its biomedical properties, including anti-cancer and anti-microbial activities. To the best of our knowledge, this study represents the first FTIR analysis for the structural characterization of β-(1-3)-glucan (EPS) produced from Tuber borchii grown in submerged fermentation.

Growth Potential of Selected Yeast Strains Cultivated on Xylose-Based Media Mimicking Lignocellulosic Wastewater Streams: High Production of Microbial Lipids by Rhodosporidium toruloides

The potential of Rhodosporidium toruloides, Candida oleophila, Metschnikowia pulcherima, and Cryptococcus curvatus species to produce single-cell-oil (SCO) and other valuable metabolites on low-cost media, based on commercial-type xylose, was investigated. Rhodosporidium strains were further evaluated in shake-flasks using different lignosulphonate (LS) concentrations, in media mimicking waste streams derived from the paper and pulp industry. Increasing the LS concentration up to 40 g/L resulted in enhanced dry cell weight (DCW) while SCO production increased up to ~5.0 g/L when R. toruloides NRRL Y-27012 and DSM 4444 were employed. The intra-cellular polysaccharide production ranged from 0.9 to 2.3 g/L in all fermentations. Subsequent fed-batch bioreactor experiments with R. toruloides NRRL Y-27012 using 20 g/L of LS and xylose, led to SCO production of 17.0 g/L with maximum lipids in DCW (YL/X) = 57.0% w/w. The fatty acid (FA) profile in cellular lipids showed that oleic (50.3–63.4% w/w) and palmitic acid (23.9–31.0%) were the major FAs. Only SCO from batch trials of R. toruloides strains contained α-linolenic acid. Media that was supplemented with various LS concentrations enhanced the unsaturation profile of SCO from R. toruloides NRRL Y-27012. SCO from R. toruloides strains could replace plant-based commodity oils in oleochemical-operations and/or it could be micro- and nano-encapsulated into novel food-based formulas offering healthier food-products.

Enhanced Effects of Iron on Mycelial Growth, Metabolism and In Vitro Antioxidant Activity of Polysaccharides from Lentinula edodes.

The polysaccharides found in Lentinula edodes have a variety of medicinal properties, such as anti-tumor and anti-viral effects, but their content in L. edodes sporophores is very low. In this study, Fe2+ was added to the liquid fermentation medium of L. edodes to analyze its effects on mycelial growth, polysaccharide and enzyme production, gene expression, and the activities of enzymes involved in polysaccharide biosynthesis, and in vitro antioxidation of polysaccharides. The results showed that when 200 mg/L of Fe2+ was added, with 7 days of shaking at 150 rpm and 3 days of static culture, the biomass reached its highest value (0.28 mg/50 mL) 50 days after the addition of Fe2+. Besides, Fe2+ addition also enhanced intracellular polysaccharide (IPS) and exopolysaccharide (EPS) productions, the levels of which were 2.98- and 1.79-fold higher than the control. The activities of the enzymes involved in polysaccharides biosynthesis, including phosphoglucomutase (PGM), phosphoglucose isomerase (PGI), and UDPG-pyrophosphorylase (UGP) were also increased under Fe2+ addition. Maximum PGI activity reached 1525.20 U/mg 30 days after Fe2+ addition, whereas PGM and UGP activities reached 3607.05 U/mg and 3823.27 U/mg 60 days after Fe2+ addition, respectively. The Pearson correlation coefficient showed a strong correlation (p < 0.01) between IPS production and PGM and UGP activities. The corresponding coding genes of the three enzymes were also upregulated. When evaluating the in vitro antioxidant activities of polysaccharides, EPS from all Fe2+-treated cultures exhibited significantly better capacity (p < 0.05) for scavenging -OH radicals. The results of the two-way ANOVA indicated that the abilities of polysaccharides to scavenge O2− radicals were significantly (p < 0.01) affected by Fe2+ concentration and incubation time. These results indicated that the addition of iron provided a good way to achieve desirable biomass, polysaccharide production, and the in vitro antioxidation of polysaccharides from L. edodes.

Transcription factor GlbHLH regulates hyphal growth, stress resistance, and polysaccharide biosynthesis in Ganoderma lucidum.

Basic helix-loop-helix (bHLH) transcription factors (TFs) participate in many physiological and cellular processes in eukaryotes. However, their functions remain unclear in the macro basidiomycete Ganoderma lucidum (G. lucidum). In this study, a gene encoding bHLH TF, GlbHLH, was identified in G. lucidum. The knockdown of GlbHLH by RNA interference reduced hyphal growth, hyphal branching, and resistant to osmotic, oxidative, and cell wall stress. The content of cell wall components β-1,3 glucan and chitin and the expression of their synthesis genes were decreased in the GlbHLH knockdown strains. The knockdown of GlbHLH led to an increase of intracellular reactive oxygen species by decreasing the enzyme activity and gene expression of antioxidant enzymes. Furthermore, the production of intracellular polysaccharides and extracellular polysaccharides was greatly decreased in the GlbHLH mutants. These results suggested that GlbHLH is involved in hyphal growth, stress response, and polysaccharide biosynthesis in G. lucidum.

Impact of different fermentation characteristics on the production of mycelial biomass, extra-cellular polysaccharides, intra-cellular polysaccharides, and on the antioxidant activities of Cordyceps militaris (L.) Fr. (strains AG-1, PSJ-1)

The mycelial biomass, antioxidant activity and production of extra- and intra-cellular polysaccharides production [EPS, IPS] of Cordyceps militaris strains AG-1, PSJ-1 were evaluated under different submerged liquid culture (SLC) conditions. At 24 0C mycelial biomass and polysaccharide production of AG-1, PSJ-1 was optimal using PVC media and static culture conditions; (AG-1: 21.85 ± 1.00; PSJ-1: 18.20 ± 1.84 g l-1 ), and oven drying at 40 oC (AG-1: 25.95 ± 0.84, PSJ-1: 23.55 ± 0.69 mg g-1) compared with hot water extraction (AG-1: 7.07 ± 0.15, PSJ-1: 7.39 ± 0.61 mg g-1). Maximum biomass, EPS and IPS production were observed when the initial pH was 6.7: AG-1: 12.92 ± 0.33, 209.70 ± 1.56, 32.62 ± 0.87; PSJ-1: 9.03 ± 0.24 g l-1 , 198.16 ± 0.85 mg g-1, 30.63 ± 1.96 mg g-1, respectively. The use 3.5 % coconut oil improved biomass, EPS, IPS production, which were 8.27 ± 0.09, 8.01 ± 0.01 g l-1 ; 1208.00 ± 8.60, 1110.40 ± 7.20 mg g-1; 32.43 ± 0.49, 29.74 ± 0.44, for AG-1 and PSJ-1, respectively. Both culture condition and oven drying methods had significant effects on H2O2 and radical scavenging activity, ABTS.+ radical activity, lipid peroxidation, and also had effects on total flavonoid and, total phenolic contents. The use of crude submerged liquid culture and oven drying on strains AG-1, PSJ-1 led to extracts with potent antioxidant activity, suggesting the therapeutic use of polysaccharides from strains AG-1, PSJ-1.

Optimisation of biomass, exopolysaccharide and intracellular polysaccharide production from the mycelium of an identified Ganoderma lucidum strain QRS 5120 using response surface methodology.

Wild-cultivated medicinal mushroom Ganoderma lucidum was morphologically identified and sequenced using phylogenetic software. In submerged-liquid fermentation (SLF), biomass, exopolysaccharide (EPS) and intracellular polysaccharide (IPS) production of the identified G. lucidum was optimised based on initial pH, starting glucose concentration and agitation rate parameters using response surface methodology (RSM). Molecularly, the G. lucidum strain QRS 5120 generated 637 base pairs, which was commensurate with related Ganoderma species. In RSM, by applying central composite design (CCD), a polynomial model was fitted to the experimental data and was found to be significant in all parameters investigated. The strongest effect (p < 0.0001) was observed for initial pH for biomass, EPS and IPS production, while agitation showed a significant value (p < 0.005) for biomass. By applying the optimized conditions, the model was validated and generated 5.12 g/L of biomass (initial pH 4.01, 32.09 g/L of glucose and 102 rpm), 2.49 g/L EPS (initial pH 4, 24.25 g/L of glucose and 110 rpm) and 1.52 g/L of IPS (and initial pH 4, 40.43 g/L of glucose, 103 rpm) in 500 mL shake flask fermentation. The optimized parameters can be upscaled for efficient biomass, EPS and IPS production using G. lucidum.

Enhanced roles of biochar and organic fertilizer in microalgae for soil carbon sink.

Improved soil carbon sink capability is important for the mitigation of carbon dioxide emissions and the enhancement of soil productivity. Biochar and organic fertilizer (OF) showed a significant improving effect on microalgae in soil carbon sink capacity, and the ultimate soil total organic carbons with microalgae-OF, microalgae-biochar, microalgae-OF-biochar were about 16, 67 and 58% higher than that with microalgae alone, respectively, indicating that carbon fixation efficiency of microalgae applied in soil was improved with biochar and OF whilst the soil carbon capacity was promoted, the mechanism of which is illustrated through simulative experiments. Organic fertilizer could spur algal conversion of carbon into cell molecules by increasing intracellular polysaccharide production of microalgae. Biochar could change carbon metabolism pathway of microalgae through altering the yield of intracellular saccharides, and yield and type of extracellular saccharides. There was a superimposition effect on the soil carbon sink when biochar and OF were both present with microalgae.

Effects of cultural medium on the formation and antitumor activity of polysaccharides by Cordyceps gunnii

The effects of culture medium composition (i.e., carbon and nitrogen sources) on the growth of mycelia, molecular weight distribution and antitumor activity of intracellular polysaccharides (IPS) from Cordyceps gunnii were investigated. Sucrose and peptone were proved to be the best carbon and nitrogen sources for mycelia growth and remarkably improved IPS production. When the sucrose concentration was 2.0%, the mycelium yield reached up to 15.94±1.26g/L, but with lower IPS yield; whereas the sucrose concentration was 4.5%, IPS yield reached to a maximum of 138.78±3.89mg/100mL. The effects of different carbon/nitrogen (C/N) ratios with equal amounts of carbon source matter on the mycelia and IPS formation were optimized. It found that the yield of mycelia and IPS were both reached to the highest at a C/N ratio of 10:3. In addition, the IPS had the highest macro molecular polysaccharide content and antitumor activity when sucrose concentration was 3.5% and the C/N ratio was 10:1.5. Thus, there was a positive correlation between molecular weight distribution and antitumor activity of IPS by C.gunnii.

Sequestration and Distribution Characteristics of Cd(II) by Microcystis aeruginosa and Its Role in Colony Formation.

To investigate the sequestration and distribution characteristics of Cd(II) by Microcystis aeruginosa and its role in Microcystis colony formation, M. aeruginosa was exposed to six different Cd(II) concentrations for 10 days. Cd(II) exposure caused hormesis in the growth of M. aeruginosa. Low concentrations of Cd(II) significantly induced formation of small Microcystis colonies (P < 0.05) and increased the intracellular polysaccharide (IPS) and bound extracellular polysaccharide (bEPS) contents of M. aeruginosa significantly (P < 0.05). There was a linear relationship between the amount of Cd(II) sequestrated by algal cells and the amount added to cultures in the rapid adsorption process that occurred during the first 5 min of exposure. After 10 d, M. aeruginosa sequestrated nearly 80% of 0.2 mg L−1 added Cd(II), while >93% of Cd(II) was sequestrated in the groups with lower added concentrations of Cd(II). More than 80% of the sequestrated Cd(II) was bioadsorbed by bEPS. The Pearson correlation coefficients of exterior and interior factors related to colony formation of M. aeruginosa revealed that Cd(II) could stimulate the production of IPS and bEPS via increasing Cd(II) bioaccumulation and bioadsorption. Increased levels of cross-linking between Cd(II) and bEPS stimulated algal cell aggregation, which eventually promoted the formation of Microcystis colonies.

Optimized extraction, composition, antioxidant and antimicrobial activities of exo and intracellular polysaccharides from submerged culture of Cordyceps cicadae.

BackgroundCordyceps cicadae is known as Jin Chan Hua in Traditional Chinese Medicine and known to possess different pharmacological activities. Presently, it was collected from the wild and isolated. Mycelial culture was optimized for extraction of polysaccharides under submerged culture conditions. Besides antioxidant, antibacterial activities of extracted polysaccharides were tested for first time.MethodsExo-polysaccharides (EPS) and intracellular polysaccharides (IPS) production was tested under different factors (medium capacity, rotation speed, pH, incubation time, temperature, carbon, nitrogen, minerals sources and carbon to nitrogen ratio) by orthogonal experiments using one-factor-at-a-time method. Monosaccharides composition of polysaccharides produced by C. cicadae was determined using high performance liquid chromatography. Antioxidant and antimicrobial activities on eight bacterial strains were checked by different standard procedures.ResultsFactors viz., medium capacity, rotation speed, incubation time, pH and temperature affected the EPS and IPS production under submerged culture conditions. EPS and IPS production was observed to vary with different carbon and nitrogen sources as well as C/N ratio. Glucose was the major component of polysaccharides (63.10 ± 4.15 %). Extracted EPS and IPS showed higher antioxidant potential with significant DPPH radical scavenging activity, ABTS radical scavenging activity, reducing power and iron chelating activity. Antimicrobial activities of EPS and IPS varied among the tested bacterial strains. IPS showed slightly higher inhibition rate to all the tested bacterial strains as compared to EPS. Maximum inhibition zones of IPS (12.9 ± 0.2 mm) and EPS (12.5 ± 0.3 mm) was observed against Pseudomonas aeruginosa at 10 % con. However, both EPS and IPS fractions showed broad spectrum for all the pathogenic microbial strains tested. The MIC of both the extracts ranged from 60–100 mg/mL.ConclusionsEPS and IPS production from submerged culture of C. cicadae with significant antioxidant and antibacterial potential can be enhanced with the combination of several factors which can be used for large scale industrial fermentation of C. cicadae.

Influence of fermentation conditions on polysaccharide production and the activities of enzymes involved in the polysaccharide synthesis of Cordyceps militaris.

The influence of different fermentation conditions on intracellular polysaccharide (IPS) production and activities of the phosphoglucomutase (PGM), UDPG-pyrophosphorylase (UGP), phosphoglucose isomerase (PGI), UDPG-dehydrogenase (UGD), and glucokinase (GK) implicated in metabolite synthesis in Cordyceps militaris was evaluated. The highest IPS production (327.57 ± 6.27mg/100mL) was obtained when the strain was grown in the optimal medium containing glucose (40g · L(-1)), beef extract (10g · L(-1)), and CaCO3 (0.5g · L(-1)), and the initial pH and temperature were 7 and 25°C, respectively. The activities of PGM, UGP, and PGI were proved to be influenced by the fermentation conditions. A strong correlation between the activities of these enzymes and the production of IPS was found. The transcription level of the pgm gene (encoding PGM) was 1.049 times and 1.467 times compared to the ugp gene and pgi gene (encoding UGP and PGI), respectively, in the optimal culture medium. This result indicated that PGM might be the highly key enzyme to regulate the biosynthesis of IPS of C. militaris in a liquid-submerged culture. Our study might be helpful for further research on the pathway of polysaccharide biosynthesis aimed to improve the IPS production of C. militaris.

Sucrose fed-batch strategy enhanced biomass, polysaccharide, and ganoderic acids production in fermentation of Ganoderma lucidum 5.26.

Ganoderma, as a Chinese traditional medicine, has multiple bioactivities. However, industrial production was limited due to low yield during Ganoderma fermentation. In this work, sucrose was found to greatly enhance intracellular polysaccharide (IPS) content and specific extracellular polysaccharide (EPS) production rate. The mechanism was studied by analyzing the activities of enzymes related to polysaccharide biosynthesis. The results revealed that sucrose regulated the activities of phosphoglucomutase and phosphoglucose isomerase. When glucose and sucrose mixture was used as carbon source, biomass, polysaccharide and ganoderic acids (GAs) production was greatly enhanced. A sucrose fed-batch strategy was developed in 10-L bioreactor, and was scaled up to 300-L bioreactor. The biomass, EPS and IPS production was 25.5, 2.9 and 4.8 g/L, respectively, which was the highest biomass and IPS production in pilot scale. This study provides information for further understanding the regulation mechanism of Ganoderma polysaccharide biosynthesis. It demonstrates that sucrose fed-batch is a useful strategy for enhancing Ganoderma biomass, polysaccharide and GAs production.

Optimised isolation of polysaccharides from Lentinula edodes strain NCBI JX915793 using response surface methodology and their antibacterial activities

Mycelial growth in a defined medium by submerged fermentation is a rapid and alternative method for obtaining fungal biomass of consistent quality. Biomass, exopolysaccharides (EPS) and intracellular polysaccharides (IPS) production were optimised by response surface methodology in Lentinula edodes strain LeS (NCBI JX915793). The optimised conditions were pH 5.0, temperature 26°C, incubation period of 25 days and agitation rate of 52 r/min for L. edodes strain LeS. Under the calculated optimal culture conditions, biomass production (5.88 mg mL− 1), EPS production (0.40 mg mL− 1) and IPS production (12.45 mg g− 1) were in agreement with the predicted values for biomass (5.93 mg mL− 1), EPS (0.55 mg mL− 1) and IPS production (12.64 mg g− 1). Crude lentinan exhibited highest antibacterial effects followed by alcoholic, crude and aqueous extracts. The results obtained may be useful for highly effective yield of biomass and bioactive metabolites.

Uniform design for optimizing biomass and intracellular polysaccharide production from self-flocculating Scenedesmus sp.-BH

Four microalgae species were isolated from freshwaters in Xinjiang, one of which was identified as Scenedesmus sp.-BH. Analysis of the specimens showed not only the characterization of self-flocculation with a flocculating rate of 92.29 % in 12 h, but relatively high biomass and intracellular polysaccharide (IPS) production (250.5 and 5.55 mg/L) under original BG11 medium. Uniform design was applied to optimize the nutritional conditions of Scenedesmus sp.-BH. The maximum biomass yield of 556.7 ± 16.97 mg/L appeared at NaNO3 0.77 g/L, K2HPO4·3H2O 0.24 g/L, MgSO4·7H2O 0.24 g/L, ferric ammonium citrate 0.021 g/L, and NaHCO3 3.85 g/L, while a maximum IPS yield of 14.3 ± 0.46 mg/L appeared at NaNO3 0.004 g/L, K2HPO4·3H2O 0.24 g/L, MgSO4·7H2O 0.21 g/L, ferric ammonium citrate 0.024 g/L, and NaHCO3 1.42 g/L. Both biomass and IPS production were over two-fold of those cultured under the original condition, which demonstrated that uniform design could successfully improve the production of biomass and IPS.

Optimization of fermentation process for the production of intracellular polysaccharide from Paecilomyces cicadae and the immuno-stimulating activity of intracellular polysaccharide

Optimization of fermentation process for the production of intracellular polysaccharide (IPS) from the fungus Paecilomyces cicadae and the immuno-stimulating activity of IPS were carried out. The quantitative effects of initial pH, fermentation temperature and time on the yield of IPS content produced by P. cicadae in submerged fermentation were investigated separately using response surface methodology (RSM). The three factors chosen for the present investigation were based on the results of a previous Plackett-Burman (PB) design. The experimental data obtained were fitted to a second-order polynomial equation using multiple regression analysis and also analyzed by appropriate statistical methods. RSM analysis showed good correspondence between experimental and predicted values. It was found that three parameters represented significant effect. Probability value (p < 0.0001) demonstrated a very high significance for the regression model. By solving the regression equation and analyzing the response surface contour plots, the optimal process parameters were determined, i.e. fermentation temperature 24.53 °C, initial pH 7.46 and fermentation time 73.9 h. The maximum predicted yield of IPS was 356.02 μg/ml under the optimal conditions. Meanwhile, IPS from P. cicadae was found to have direct immuno-stimulating activity in vitro on murine macrophage RAW264.7 proliferative response and to stimulate nitric oxide generation in a dose-dependent manner.

Application of Response Surface Methodology for Optimization of Liquid Fermentation Medium for Polysaccharide Production by &lt;i&gt;Tricholoma Mongolicum&lt;/i&gt; Imai

The fermentation medium of Tricholoma Mongolicum Imai for producing intracellular polysaccharide and extracellular polysaccharide was optimized by response surface methodology. The effects of carbon source, nitrogen source and mineral salt on polysaccharide production were examined by single factor experiments, glucose and yeast was found to be the best carbon source and nitrogen source. Based on this, the optimization of the Box-Benhnken design combined with response surface analysis showed that the optimal medium contained 40g/L glucose; 20g/L yeast extract; 0.40% KH2PO4; 0.40% MgSO4.The intracellular polysaccharide production and extracellular polysaccharide in medium reached 2.0512g/L and 2.394 g/L , respectively.

Optimization for Production of Intracellular Polysaccharide from Cordyceps ophioglossoides L2 in Submerged Culture and Its Antioxidant Activities in vitro

Cordyceps ophioglossoides is a valuable traditional medicinal material. We have found that intracellular polysaccharide (IPS) is the major biologically active ingredient in Cordyceps ophioglossoides. This study is the first time to optimize the yield of IPS from Cordyceps ophioglossoides. The optimal medium for IPS production consists of glucose 54.50 g·L−1, yeast powder 25.50 g·L−1, NaH2PO4 0.4 g·L−1 and K2HPO4 0.4 g·L−1. The suggested culture conditions are 24 °C, initial pH 4.5 with a rotary speed of 120 r·min−1 for 168 h. The yield of IPS is 737.93 mg·L−1, which is 50% higher than the yield under the conditions prior to optimization. The anti-oxidative activities of IPS in Cordyceps ophioglossoides L2 are also characterized using various in vitro assay. The anti-oxidative activity may explain the reason why IPS from Cordyceps ophioglossoides can be used to fight against neurodegenerative diseases and menopausal symptoms.

Medium modification to enhance the formation of bioactive metabolites in shake flask cultures of Antrodia cinnamomea by adding citrus peel extract

Antrodia cinnamomea has recently become a well-known medicinal mushroom in Taiwan. Bioactive compounds found in A. cinnamomea include: polysaccharide, sesquiterpene lactone, steroids and triterpenoids. The aim of this study was to evaluate the feasibility of adding citrus peel extract to enhance the formation of bioactive metabolites in the submerged culture of A. cinnamomea. With the exception of grapefruit, citrus peel extracts tested were proved to be beneficial to mycelial growth and to the production of intracellular polysaccharide. Lemon was the most effective for enhancing bioactive metabolite production. With an addition of 2% (v/v), the mycelium biomass concentration and intracellular polysaccharide content rose from 11.96g DW/L of the control and 123.6mg/g DW to 21.96g DW/L and 230.8mg/g DW, respectively, on day 8. The production of triterpenoids also increased from 86.7 to 282.9mg/L. Moreover, this study also demonstrates that although the addition of peel extract could cause the lengthening of the exponential phase and reduce the specific growth rate, the production rate of biomass, intracellular polysaccharide and triterpenoids was still enhanced significantly.
