Dihydrocaffeic acid grafted chitosan self-assembled nanomicelles with enhanced intestinal transport and antioxidant properties of chicoric acid.

Chicoric acid (CA) plays a crucial role as a functional factor within the realm of foods, showcasing a wide array of bioactivities. Nevertheless, its oral bioavailability is significantly limited. To optimize the intestinal absorption and bolster the antioxidant capacity of CA, a water-soluble dihydrocaffeic acid grafted chitosan copolymer (DA-g-CS) was synthesized using a conventional free radicals system, and subsequently utilized for the encapsulation of CA within self-assembled nanomicelles (DA-g-CS/CA). The average particle size of DA-g-CS/CA was 203.3nm, while the critical micelle concentration was 3.98×10-4 mg/mL. Intestinal transport studies revealed that DA-g-CS/CA penetrated cells via the macropinocytosis pathway, exhibiting the cellular uptake rate 1.64 times higher than that of CA. This substantial enhancement in the intestinal transport of CA underscores the significant improvements achieved through DA-g-CS/CA delivery. The pharmacokinetic results demonstrated that DA-g-CS/CA exhibited a remarkable bioavailability 2.24 times that of CA. Furthermore, the antioxidant assessment demonstrated that DA-g-CS/CA exhibited exceptional antioxidant properties in comparison to CA. It demonstrated enhanced protective and mitigating effects in the H2O2-induced oxidative damage model, while also displaying a stronger emphasis on protective effects rather than attenuating effects. These findings aim to establish a solid theoretical foundation for the advancement of CA in terms of its oral absorption and the development of functional food products.

Combining Human Organoids and Organ-on-a-Chip Technology to Model Intestinal Region-Specific Functionality

The intestinal mucosa is a complex physical and biochemical barrier that fulfills a myriad of important functions. It enables the transport, absorption, and metabolism of nutrients and xenobiotics while facilitating a symbiotic relationship with microbiota and restricting the invasion of microorganisms. Functional interaction between various cell types and their physical and biochemical environment is vital to establish and maintain intestinal tissue homeostasis. Modeling these complex interactions and integrated intestinal physiology in vitro is a formidable goal with the potential to transform the way new therapeutic targets and drug candidates are discovered and developed. Organoids and Organ-on-a-Chip technologies have recently been combined to generate human-relevant intestine chips suitable for studying the functional aspects of intestinal physiology and pathophysiology in vitro. Organoids derived from the biopsies of the small (duodenum) and large intestine are seeded into the top compartment of an organ chip and then successfully expandas monolayers while preserving the distinct cellular, molecular, and functional features of each intestinal region. Human intestine tissue-specific microvascular endothelial cells are incorporated in the bottom compartment of the organ chip to recreate the epithelial-endothelial interface. This novel platform facilitates luminal exposure to nutrients, drugs, and microorganisms, enabling studies of intestinal transport, permeability, and host-microbe interactions. Here, a detailed protocol is provided for the establishment of intestine chips representing the human duodenum (duodenum chip) and colon (colon chip), and their subsequent culture under continuous flow and peristalsis-like deformations. We demonstrate methods for assessing drug metabolism and CYP3A4 induction in duodenum chip using prototypical inducers and substrates. Lastly, we provide a step-by-step procedurefor the in vitro modeling of interferon gamma (IFNγ)-mediated barrier disruption (leaky gut syndrome) in a colon chip, including methods for evaluating the alteration of paracellular permeability, changes in cytokine secretion, and transcriptomic profiling of the cells within the chip.

β-Lactoglobulin Heptapeptide Reduces Oxidative Stress in Intestinal Epithelial Cells and Angiotensin II-Induced Vasoconstriction on Mouse Mesenteric Arteries by Induction of Nuclear Factor Erythroid 2-Related Factor 2 (Nrf2) Translocation.

Peptides derived from buffalo dairy products possess multiple healthy properties that cannot be exerted as long as they are encrypted in parent proteins. To evaluate the biological activities of encrypted peptide sequences from buffalo ricotta cheese, we performed a simulated gastrointestinal (GI) digestion. Chemical and pharmacological characterization of the digest led to the identification of a novel peptide endowed with antioxidant and antihypertensive action. The GI digest was fractionated by Semiprep-HPLC, and fractions were tested against reactive oxygen species (ROS) release in an H2O2-treated intestinal epithelial cell line. UHPLC-PDA-MS/MS analysis revealed the presence of an abundant β-lactoglobulin peptide (BRP2) in the most active fraction. Pharmacological characterization of BRP2 highlighted its antioxidant activity, involving ROS reduction, nuclear factor erythroid 2-related factor 2 (Nrf2) activation, and cytoprotective enzyme expression. The bioavailability of BRP2 was evaluated in intestinal transport studies through a Caco-2 cell monolayer. Equal bidirectional transport and linear permeability indicate that BRP2 was absorbed mainly through passive diffusion. In addition to its local effects, the BRP2 administration on mouse mesenteric arteries was able to reduce the angiotensin II-induced vasoconstriction by the Nrf2 nuclear translocation, the reduction of the active form of Ras-related C3 botulinum toxin substrate 1 (Rac1), and the NADPH oxidase activity. These data further highlight the role of buffalo ricotta cheese-derived peptides against oxidative stress-related diseases and suggest their health-promoting potential.

Validation of UHPLC–MS/MS methods for the determination of kaempferol and its metabolite 4-hydroxyphenyl acetic acid, and application to in vitro blood-brain barrier and intestinal drug permeability studies

Sedative and anxiolytic-like properties of flavonoids such as kaempferol and quercetin, and of some of their intestinal metabolites, have been demonstrated in pharmacological studies. However, routes of administration were shown to be critical for observing in vivo activity. Therefore, the ability to cross intestinal and blood-brain barriers was assessed in cell-based models for kaempferol (KMF), and for the major intestinal metabolite of KMF, 4-hydroxyphenylacetic acid (4-HPAA). Intestinal transport studies were performed with Caco-2 cells, and blood-brain barrier transport studies with an immortalized monoculture human model and a primary triple-co-culture rat model. UHPLC–MS/MS methods for KMF and 4-HPAA in Ringer-HEPES buffer and in Hank’s balanced salt solution were validated according to industry guidelines. For all methods, calibration curves were fitted by least-squares quadratic regression with 1/X2 as weighing factor, and mean coefficients of determination (R2) were >0.99. Data obtained with all barrier models showed high intestinal and blood-brain barrier permeation of KMF, and no permeability of 4-HPAA, when compared to barrier integrity markers.

The Appearance of a Leptin Effect on Glucose Absorption in Caco2 Cells Depends on Their Differentiation Level

Backdround/Aims: The aim of this work was to study the effect and mechanism of action of leptin added apically, on glucose absorption, using Caco-2 cells as a model. Methods: Cells were grown on inserts and treated with leptin, at different time points after confluence. Radiolabelled glucose was added to the upper chamber and samples from the lower chamber were collected and assayed for radioactivity. Results: Glucose absorption increased with an increase in the level of differentiation and was associated with an increase in the protein expression level of glucose transporters. Leptin reduced glucose absorption only by day 16 after confluence, the time at which apical leptin receptors started appearing. This inhibitory effect became higher the longer the post confluence period, and was prominent on day 23. The hormone effect was found to be mediated via a decrease in the number of glucose transporters (SGLT1 and GLUT2) and a decrease in the activity of the Na⁺/K⁺ ATPase which was assayed by measuring the amount of inorganic phosphate liberated in presence and absence of enzyme activators. Conclusion: It was concluded that by day 23 post confluence, Caco-2 cells are differentiated and are appropriate to use as a model for intestinal transport studies.

Effects of age and zinc supplementation on transport properties in the jejunum of piglets.

Zinc is effective in the prevention and treatment of post-weaning diarrhoea and in promoting piglet growth. Its effects on the absorption of nutrients and the secretory capacity of the intestinal epithelium are controversial. We investigated the effects of age, dietary pharmacological zinc supplementation and acute zinc exposure in vitro on small-intestinal transport properties of weaned piglets. We further examined whether the effect of zinc on secretory responses depended on the pathway by which chloride secretion is activated. A total of 96 piglets were weaned at 26 days of age and allocated to diets containing three different levels of zinc oxide (50, 150 and 2500 ppm). At the age of 32, 39, 46 and 53 days, piglets were killed, and isolated epithelia from the mid-jejunum were used for intestinal transport studies in conventional Ussing chambers, with 23 μm ZnSO4 being added to the serosal side for testing acute effects. Absorptive transport was stimulated by mucosal addition of d-glucose or l-glutamine. Secretion was activated by serosal addition of prostaglandin E2 , carbachol or by mucosal application of Escherichia coli heat-stable enterotoxin (Stp ). Jejunal transport properties showed significant age-dependent alterations (p < 0.03). Both absorptive and secretory responses were highest in the youngest piglets (32 d). The dietary zinc supplementation had no significant influence on jejunal absorptive and secretory responses. However, the pre-treatment of epithelia with ZnSO4 in vitro led to a small but significant decrease in both absorptive and secretory capacities (p < 0.05), with an exception for carbachol (p = 0.07). The results showed that, in piglets, chronic supplementation with zinc did not sustainably influence the jejunal transport properties in the post-weaning phase. Because transport properties are influenced by the addition of zinc in vitro, we suggest that possible epithelial effects of zinc depend on the acute presence of this ion.

Intestinal Transportations of Main Chemical Compositions of Polygoni Multiflori Radix in Caco‐2 Cell Model

Context. Polygoni Multiflori Radix (PMR) is originated from the root of Polygonum multiflorum Thunb. and used in oriental countries for centuries. However, little researches pay close attention to the absorption of its major constituents. Objective. Transepithelial transport of TSG, RL, PL, and four anthraquinones is carried out. Materials and Methods. Caco-2 cell monolayer, which represented a well-established model for the study of intestinal transport of nutrients and xenobiotics, was used in this paper. Results. The apparent permeability coefficients (P app) in the Caco-2 cell monolayers were TSG (2.372 × 10−9) < EG (2.391 × 10−9) < EN (2.483 × 10−9) < PL (4.917 × 10−9) < RN (1.707 × 10−8) < RL (1.778 × 10−8) < AE (1.952 × 10−8). Thus, RN, RL, and AE were considered partly absorbed, while other constituents were hardly absorbed. Discussion and Conclusion. Glycosides showed poor permeabilities than aglycones. In the meantime, TSG and EN gave out poor recovery rates in this assay, which indicated that TSG and EN may accumulate or metabolise in the Caco-2 cells. In silico prediction indicated that Gibbs energy (r = 0.751, p < 0.05) and heat of form (r = 0.701, p < 0.05) were strongly positively correlated with P app.

In vitro study of intestinal transport of fluoride using the Caco-2 cell line

Water and food are the main sources of fluoride exposure and therefore it is necessary to study intestinal absorption in order to make a correct evaluation of the risk/benefit associated with exposure to fluoride. The present study characterizes intestinal transport of fluoride, using the Caco-2 cell line as a model of the intestinal epithelium, and evaluates the coefficients of apparent permeability and intracellular accumulation in various conditions (pH, temperature, opening of cell junctions, presence of anions). The results indicate that fluoride is an element with moderate absorption (<70%) in both directions (absorptive and secretory). Both in absorption (apical-basolateral) and in secretion (basolateral-apical) there is transport by the paracellular pathway, which may be considered predominant. Absorption and secretion of fluoride increase at acid pH levels, possibly because of its non-ionized state at these pHs and/or because of participation of a F(-)/H(+) cotransporter or a F(-)/OH(-) antiporter. The results also suggest transcellular participation of mechanisms involved in transport of Cl(-) and of an active transport in the secretory direction. The present study extend the knowledge on the cellular transport of fluoride and provide the basis for future studies aimed at identifying potential transporters involved in human fluoride absorption.

In vitro study of intestinal transport of inorganic and methylated arsenic species by Caco-2/HT29-MTX cocultures.

This study characterizes intestinal absorption of arsenic species using in vitro system Caco-2/HT29-MTX cocultures in various proportions (100/0 to 30/70). The species assayed were As(V), As(III), monomethylarsonic acid [MMA(V)], monomethylarsonous acid [MMA(III)], dimethylarsinic acid [DMA(V)], and dimethylarsinous acid [DMA(III)]. The results show that the apparent permeability (P(app)) values of pentavalent species increase significantly in the Caco-2/HT29-MTX cocultures in comparison with the Caco-2 monoculture, probably because of enhancement of paracellular transport. For MMA(III) and DMA(III), P(app) decreases in the Caco-2/HT29-MTX cell model, and for As(III), there is no change in P(app) between the two culture models. Transport studies of arsenic solubilized from cooked foods (rice, garlic, and seaweed) after applying an in vitro gastrointestinal digestion showed that arsenic absorption also varies with the model used, increasing with the incorporation of HT29-MTX in the culture. These results show the importance of choosing a suitable in vitro model when evaluating intestinal arsenic absorption processes.

In vitro study of intestinal transport of arsenite, monomethylarsonous acid, and dimethylarsinous acid by Caco-2 cell line

Arsenic is a pollutant widely distributed in the environment. There are numerous studies on the toxicity of trivalent arsenic forms As(III), MMA(III), and DMA(III), but few data are available on the processes of digestion and absorption of these arsenic species and the mechanisms involved are unknown. The present study evaluated the processes involved in intestinal absorption of trivalent arsenic species, using the Caco-2 cell model as system. The apparent permeability values obtained for As(III) in apical-basolateral direction (4.6±0.3)×10−6cm/s, showing moderate intestinal absorption. Transport of MMA(III) [Papp=(7.0±0.9)×10−6cm/s] and DMA(III) [Papp=(10.6±1.4)×10−6cm/s] is greater than that of As(III). The cellular retention of As(III) (0.9–2.4%) was less than that observed for MMA(III) (30%) and DMA(III) (35%).A substantial paracellular component was observed in transport of As(III) and MMA(III), whereas DMA(III) does not use this pathway for its absorption. For all the trivalent species, transport depends on temperature, with an active transcellular component for MMA(III) and DMA(III). Variations in pH do not affect transport of these species. The presence of GSH and green tea extract significantly alters transport of As(III) across Caco-2 cells.

Studies of intestinal drug transport using an in silico epithelio-mimetic device

We report the development and use of a synthetic, discrete event, discrete space model that functions as an epithelio-mimetic device (EMD). It is intended to facilitate the study of intestinal transport of drug-like compounds. We represent passive paracellular and transcellular transport, carrier-mediated transport and active efflux using stand-alone components. Systematic verification of the EMD over a wide physiologically realistic range is essential before we can use it to address questions regarding the details of the interacting mechanisms that are believed to influence absorption. We report details of key verification experiments. We demonstrate that this device can generate behaviors similar to those observed in the in vitro Caco-2 transwell system. To do that we used a series of hypothetical drugs and we simulated behaviors for two clinically used drugs, alfentanil and digoxin. The results support the feasibility and practicability of the EMD as a tool to expand the experimental options for better understanding the biological processes involved in intestinal transport and absorption of compounds of interest.

Radiolabeling of methylphosphonate and phosphorothioate oligonucleotides and evaluation of their transport in everted rat jejunum sacs.

The therapeutic use of antisense oligonucleotides will likely involve their administration over protracted periods of time. The oral route of drug dosing offers many advantages over other possible routes when chronic drug administration is necessary. However, little is known about the potential for oligonucleotide uptake from the gastrointestinal tract. This issue is addressed in the current work. We have developed a simple procedure for radiolabeling oligonucleotides by reductive alkylation with 14C-formaldehyde. We have utilized this approach, as well as 5' addition of fluorophores, to prepare labeled methylphosphonate and phosphorothioate oligonucleotides for use in intestinal transport studies. An everted rat gut sac model was employed to compare the transport of oligonucleotides to that of model compounds whose permeation properties are better understood. We demonstrate that both methylphosphonate and phosphorothioate oligonucleotides are passively transported across the intestinal epithelium, probably by a paracellular route. The rates of transport for both types of oligonucleotides were similar, and were significantly greater than that of the very high MW polymer blue dextran, but were lower than the transport rate of valproic acid, a low MW compound known to have high oral availability. A significant degree of permeation of oligonucleotides across the gastrointestinal epithelium does occur, but it is still unclear whether this is sufficient to permit effective oral administration of oligonucleotides as drugs.

Characterization of small intestinal water, sodium, and potassium transport and morphology in the pig.

The suitability of the young pig for studies of intestinal transport was investigated by determination of net water, sodium, and potassium absorption, and morphometry in jejunum and ileum. Active net absorption was seen in both jejunum and ileum. There was no difference in absorption between segments and there was a good correlation between water and electrolyte transport. Heart rate, arterial pressure, and absorption were constant with time. Villus height and width were similar to those of human, but crypt depth was greater, and the ratio of villus height to crypt depth less. The mucosa was partially permeable to polyethylene glycol 4000 and this marker is not suitable for studies of transport in young pigs.

Impaired intestinal electrolyte transport in rats infested with the common parasiteSyphacia muris

An incidentally discovered infestation with the nematode Syphacia muris of cecum and colon in spontaneously hypertensive (SHR) and normotensive control (WKY) rats was investigated over a two-year period. Infestation rates in WKY were higher than in SHR, while clinical signs as well as histological changes of colonic tissues were absent in both strains. In vivo net water absorption (microliter/hr/cm2) in control worm-free SHR turned into secretion in infested rats, ie, from 74.2 +/- 23.2 to -7.5 +/- 35.0 (P less than 0.001); this corresponded with a decrease in net absorption (mumol/hr/cm2) of Na from 18.5 +/- 2.4 to 9.3 +/- 4.3 (P less than 0.001) and of Cl from 14.0 +/- 3.2 to 3.2 +/- 5.7 (P less than 0.001). In WKY, net water absorption decreased from 112.2 +/- 23.2 to 48.0 +/- 25.1 (P less than 0.001) and Na and Cl absorption from 22.3 +/- 3.1 to 16.0 +/- 4.2 (P less than 0.005) and from 19.4 +/- 2.7 to 10.9 +/- 4.7 (P less than 0.005), respectively. Antihelminthic treatment with 0.007% pyrvinium pamoate in the ration (four weeks on, six months off) eradicated Syphacia muris in both rat strains. Body weight gain of young rats on normal and pyrvinium pamoate-substituted diet studied over 18 months was similar, indicating a good tolerance of the treatment. It is concluded that results obtained during comparative intestinal transport studies between SHR and WKY may not only be impaired but also significantly distorted by Syphacia muris infestation as SHR appear to be more susceptible to effects induced by this common parasite than WKY.

Fluxplus, a microcomputer program for interactive calculation of ‘in vitro’ solute fluxes, uptakes and accumulations in studies of intestinal transport

A new microcomputer program for interactive comparisons, analysis and calculation of in vitro solute transepithelial fluxes and nutrient uptakes and accumulations by intestinal tissues is described. The program is written in Microsoft GWBASIC for the widely distributed IBM-PC. Flux, uptake and accumulation values are obtained by computation from initial sequential data files and stored in sequential or random-organized files, depending on the specific process. Computed unidirectional fluxes are saved together with their associate electrical variables. The program calculates univariate statistics for unidirectional and net fluxes, and allows comparisons between selected groups of experiments by using the Student's t-test. An homocedasticity test is also provided. Active uptake or accumulations of solutes are calculated by substracting the predicted diffusive values at a determined concentration to total uptake or accumulation and from this, kinetic parameters are computed. The program was designed to be user-friendly and includes several functions to permit the user to manage and examine stored data. Examples of outputs illustrate applications of the program.

Food restriction and recovery of nonabsorbed indicators from the small intestine of the rat.

Indicators of net water movement are used in a variety of intestinal transport studies. Such indicators should be virtually unabsorbed under study conditions. The effects of stimuli that alter intestinal mass on indicator recovery have not been examined. We used the male albino rat to study the effects of food restriction (50%) and fasting on recovery of indicators perfused intraluminally through the entire small intestine. Recovery of PEG 4000 and phenol red was essentially complete (98-100%) from control and restricted groups, but was 96% from the fasted group. PEG 400, consisting of a homologous series of polymers of which seven were measured (MW 242-506), was also perfused. The lower-molecular-weight polymers were absorbed. Absorption decreased with increasing molecular weight, and the slope of the relation of absorption to molecular weight was the same for all groups.

Triple-lumen perfusion of the canine jejunum

The triple-lumen gut perfusion technique was validated in five dogs with established jejunal fistulas. Transit time through the 30-cm test segment was 6.2 min when perfused at 4 ml/min with a balanced electrolyte solution (Na 140, K 5, Cl 110 and HCO3 35 mM), and this remained constant during a 3-hr test period. Fasting intestinal content was very small in the dog. After 35 min equilibration, steady-state absorption was achieved during perfusion with electrolyte solution containing polyethylene glycol (5 g/ liter) as a nonabsorbable marker (10 studies). The triple-lumen gut perfusion technique is well suited to the study of intestinal transport in vivo in dogs with chronic intestinal fistulas.

Water and electrolyte movement and mucosal morphology in the jejunum of patients with portal hypertension

Intestinal secretion and intercellular space dilatation can be induced in animal models by acute elevation of intravascular volume or portal pressure. We examined whether patients with increased portal venous pressure might represent a clinical counterpart to these animal models. Portal venous pressure, determined by hepatic wedge pressure measurement, was elevated to 10–55 mmHg (mean 29 mmHg) in 8 patients with chronic liver disease without diarrhea. Intestinal transport studies utilizing a steady-state perfusion technique revealed normal absorption of a plasmalike electrolyte solution. A solution designed to unmask intestinal secretion demonstrated no difference from control subjects in the movement of water, electrolytes, or protein into the intestinal lumen. There was no correlation of absorption or secretion with hepatic wedge pressure. Jejunal biopsy revealed a significant increase in dilatation of intercellular spaces in patients compared to controls; this increase was not correlated with hepatic wedge pressure, but was significantly inversely correlated to plasma renin and aldosterone concentration. We conclude that patients with chronic liver disease and portal hypertension absorb water and electrolytes normally, but have mild morphologic alterations in the intestinal mucosa, possibly related to intravascular volume status.
