Development of Insecticide Resistance in Field Populations of Onion Thrips, Thrips tabaci (Thysanoptera: Thripidae).

The present study evaluated insecticide resistance in field populations of onion thrips, Thrips tabaci Lindeman (Thysanoptera: Thripidae), collected from eight different onion-growing regions of Punjab, Pakistan. These field-collected populations were assessed for resistance development against eight commonly used active ingredients including deltamethrin, lambda-cyhalothrin, imidacloprid, acetamiprid, spinosad, spinetoram, cypermethrin, and abamectin. In leaf dip bioassays, T. tabaci adults showed varied levels of resistance towards different insecticides. Moderate or high levels of resistance to deltamethrin (58-86 fold), lambda-cyhalothrin (20-63 fold), and cypermethrin (22-54 fold) were observed in T. tabaci field populations. There were very low to moderate resistance levels to imidacloprid (10-38 fold), acetamiprid (5-29 fold), and abamectin (10-30 fold). The lowest levels of resistance were detected in thrips exposed to spinosad (3-13 fold) and spinetoram (3-8 fold). Insecticide resistance levels varied among populations collected from various geographic locations, but all populations exhibited elevated levels of resistance to deltamethrin. Thrips tabaci populations with higher resistance levels were most commonly found from the southern part of Punjab, Pakistan. Our findings revealed that spinosyns could be used as alternatives to conventional insecticides for the successful management of T. tabaci in onion fields.

CRISPR/Cas9-mediated knockout of NlCYP6CS1 gene reveals its role in detoxification of insecticides in Nilaparvata lugens (Hemiptera: Delphacidae).

The brown planthopper (Nilaparvata lugens) is one of the major rice insect pests in Asia. Recently, high levels of insecticide resistance have been frequently reported and cytochrome P450 monooxygenase (P450)-mediated metabolic detoxification is a common resistance mechanism in N. lugens. However, there has been no persuasive genetic method to prove the role of P450s in insecticide resistance in N. lugens. Here, CRISPR/Cas9 system was used to disrupt the P450 gene NlCYP6CS1 to elucidate its role in insecticide resistance in field populations of N. lugens. We successfully constructed a homozygous strain (Nl6CS1-KO) with a 5-bp deletion and 1-bp insertion mutation of NlCYP6CS1. Compared with a background resistant strain (Nl-R), the susceptibility of knockout strain Nl6CS1-KO to imidacloprid, nitenpyram, thiamethoxam, dinotefuran, and pymetrozine was increased by 2.3-, 3.4-, 7.0-, 4.2- and 3.9-fold, respectively, but not significantly changed to triflumezopyrim, chlorpyrifos and buprofezin. Life table analysis demonstrated that the Nl6CS1-KO strain resembled the Nl-R strain in terms of egg and nymph developmental duration and adult lifespan, but differed from the Nl-R strain in the survival rate of eggs and nymphs, reproduction, and body weight. Our study demonstrates the effect of functional deletion of NlCYP6CS1 on multiple insecticide resistance in N. lugens. For the first time, we applied CRISPR/Cas9 system to reveal the mechanism of insecticide resistance in N. lugens, which may shed light on similar studies in other hemipteran insects. © 2023 Society of Chemical Industry.

First Report on the Mechanisms of Insecticide Resistance in Field Populations of the Small Hive Beetle in Florida

The small hive beetle (Aethina tumida Murray) is a serious threat to beekeeping and crops that rely on honeybees for pollination. The small hive beetle not only causes significant damage to honeybees by feeding on pollen and honey, attacking bee brood and causing stored honey to ferment, but also might serve as a vector of diseases. In addition, the small hive beetle has developed resistance to the pyrethroid and organophosphate insecticides registered for control of honeybee pests in the United States. The development of resistance in small hive beetle populations is a great concern to the beekeeping industry; thus, there is an urgent need for strategies to manage that resistance. Therefore, we used synergist probes to determine the mechanisms of resistance in a small hive beetle population to these insecticides. Our studies on the toxicity of insecticides alone or with the synergists piperonyl butoxide (PBO) and S,S,S,-tributyl phosphorotrithionate (DEF) suggested that mixed-function oxidases and esterases were the major resistance factors to these insecticides in a studied population of the small hive beetle. In contrast, there was no synergism with diethyl maleate (DEM), triphenyl phosphate (TPP) and formamidine. Therefore, glutathione-S-transferase, carboxylesterase and target site were not involved in insecticide resistance in the small hive beetle. Rotation of classes of insecticides (with different modes of action) and metabolic synergists were suggested for the development of successful resistance management programs. To the best of our knowledge, this is the first study of the mechanisms of resistance in small hive beetle populations in Florida and suggests an urgent need for alternative control strategies for these serious pests of honeybee colonies.

The detoxification enzymes activity profile in susceptible Aedes and Culex mosquitoes

The status of susceptible mosquito is important for monitoring and managing insecticide resistance in field population. The purpose of this study is to profile the differential expression level of enzymatic activities of metabolic enzymes exhibited in the mosquito species of Aedes aegypti, Aedes albopictus, and Culex quinquefasciatus from the susceptible strain. The fourth instar larvae of each strain were subjected to biochemical assay. The total protein content and enzymatic activities of Glutathione S-transferase (GST), α-esterase (α-est), β-esterase (β-est) Cytochrome P450 (Cyt P450) and Acetylcholinesterase (AChE) from each mosquito strains were elucidated. Significant difference (P<0.05) was detected between the total protein content between all species of susceptible strain mosquitoes. One-way ANOVA showed that the specific enzymatic activities of GST, α-est and Cyt P450 of all test mosquitoes were significantly different upon comparison with each species (P<0.05). The mean of enzymatic activities of insensitive AChE showed no significant difference upon comparison with each other (P>0.05). The enzymatic activities of β-est shows no significant difference between Ae. aegypti and Cx. quinquefasciatus mosquitoes (P>0.05) but the enzymatic activity of β-est in Ae. albopictus manifested significant difference upon comparison with the enzymatic activities of the other two test species (P<0.05). The results obtained may provide more information about the enzymatic activities of metabolic enzymes in Ae. aegypti, Ae. albopictus and Cx. quinquefasciatus mosquitoes which might be beneficial for public sector for the application of proper vector control measures.

Overexpression of UDP-glycosyltransferase potentially involved in insecticide resistance in Aphis gossypii Glover collected from Bt cotton fields in China.

The cotton aphid Aphis gossypii Glover is one of the most destructive insect pests. It has evolved resistance to numerous insecticides around the world due to the application of insecticides. Uridine diphosphate (UDP)-glycosyltransferases (UGTs) have been reported to potentially facilitate the detoxification process of imidacloprid and thiamethoxam in A. gossypii. In this study, the field populations of A. gossypii developed different levels of resistance to multiple insecticides. A UGT inhibitor, 5-nitrouracil, dramatically increased the toxicity of acetamiprid in resistant populations, moderately increased the toxicity of sulfoxaflor in the imidacloprid susceptible (IMI_S) population, and populations from Yuncheng in Shanxi Province (SXYC) and Jingzhou in Hubei Province (HBJZ), and increased the toxicity of bifenthrin in the IMI_S and HBJZ populations, but there was no synergism on omethoate or carbosulfan. Quantitative real-time PCR analysis revealed that UGT344B4 and UGT344C7 were overexpressed in all field populations, and UGT344N4 was overexpressed in the SDBZ and HBZJ populations. Furthermore, the suppression of UGT344B4 or UGT344C7 by RNA interference significantly increased the susceptibility to bifenthrin in the IMI_S population and the susceptibility to sulfoxaflor in the SXYC population. These results suggested that UGTs are potentially involved in the detoxification of neonicotinoid, sulfoximine, and pyrethroid insecticides in A. gossypii. Furthermore, the overexpression of UGTs could be associated with insecticide resistance in field populations of A. gossypii. The results might be helpful for the management of insecticide resistance in field populations of A. gossypii. © 2019 Society of Chemical Industry.

Residual contact vial method for the rapid on-site detection of insecticide resistance in Thrips palmi

Abstract A residual contact vial plus water (RCVpW) bioassay method, in which water was supplemented to minimize control mortality, was established to monitor insecticide resistance in field populations of the melon thrips, Thrips palmi. In the RCVpW, median lethal doses (LD50) of six insecticides commonly used in T. palmi control, were determined at 8 h post-treatment, using a susceptible RDA strain according to the RCVpW protocol. Diagnostic doses for on-site resistance monitoring of the six insecticides, which were determined as doses two-fold higher than required to achieve LD90 in the RDA strain, were in the range of 0.299 to 164.3 μg−1 cm2. Insecticide resistance levels in five field populations of T. palmi were evaluated to test the applicability of RCVpW in monitoring the pest. Although the RDA strain exhibited 100% mortality to diagnostic doses, field populations showed a reduced mortality in response to all test insecticides, indicating different degrees of resistance. In particular, all test field populations exhibited a significantly low mortality in response to spinosad, suggesting a wide distribution of spinosad resistance. Synergistic bioassay revealed that cytochrome P450-mediated metabolic factor is involved in spinosad resistance in the Korean population. Interestingly, an apparently reduced mortality to emamectin benzoate and chlofenapyr was observed in some field populations, perhaps suggesting uneven distribution of resistance to these insecticides in field populations. Our study showed that the RCVpW protocol can be employed both as an on-site resistance monitoring method for major thrip species, and in the selection of appropriate insecticides for their control.

To plan and improve the efficiency of disinsection measures at livestock and poultry facilities, the information on the susceptibility of houseflies to modern insecticides is needed. The goal of this study was to determine the susceptibility level to fipronil, acetamiprid, ivermectin, and chlorfenapyr in Musca domestica L. field populations inhabiting agricultural facilities. The study was conducted in one poultry and in two livestock farms. The susceptibility of M. domestica adults of the first generation to insecticides was assessed by the method of dosed contacting, after which the median lethal doses and the resistance ratio were calculated relative to the laboratory culture. According to the results, the fly population from a cattlebreeding farm, in which neonicotinoid fly baits were used for ten years, was highly resistant to acetamiprid (resistance ratio 57) and tolerant to ivermectin (resistance ratio 10). The fly population from poultry farms, in which neonicotinoids were used as baits against flies for four years, was tolerant to acetamiprid (resistance ratio 5). The fly population from a livestock farm, in which insecticides have not been used for more than five years, was susceptible to all insecticides tested. The obtained results and literature data confirmed that the rate of the development of insecticide resistance in field populations of M. domestica depends on the form and duration of the application of insecticide formulations.

Detection and biochemical characterization of insecticide resistance in field populations of Asian citrus psyllid in Guangdong of China

The Asian citrus psyllid, Diaphorina citri Kuwayama, is one of the most damaging pests of citrus-producing regions throughout the world. The use of insecticides is the main strategy for controlling psyllid and has increased year by year. In this study, four field populations of D. citri were evaluated for resistance to nine different insecticides using the leaf-dip method. The results showed that the highest level of resistance for D. citri was found in imidacloprid with a resistance ratio of 15.12 in the Zengcheng population compared with the laboratory susceptible population. This was followed by chlorpyriphos (6.47), dinotefuran (6.16), thiamethoxam (6.04), lambda-cyhalothrin (4.78), and bifenthrin (4.16). Piperonyl butoxide (PBO) and triphenyl phosphate (TPP) showed significant synergism on imidacloprid effects in the Zengcheng population (3.84- and 2.46-fold, respectively). Nevertheless, diethyl maleate (DEM) had no synergism on imidacloprid. Biochemical enzyme assays suggested that general esterase, glutathione S-transferase and cytochrome P450 monooxygenase activities were higher in the field-collected populations than in the laboratory susceptible population. However, glutathione S-transferase may play a minor role in the resistance of adult D. citri to insecticides. At the molecular level, resistance of D. citri to imidacloprid is mainly related to the increased expression of CYP4C68 and CYP4G70 (>5-fold).

Resistance to commonly used insecticides and phosphine fumigant in red palm weevil, Rhynchophorus ferrugineus (Olivier) in Pakistan.

The red palm weevil Rhynchophorus ferrugineus (Olivier) is an important pest of date palms in many regions of the world. This paper reports the first survey of insecticide resistance in field populations of R. ferrugineus in Pakistan which were collected from seven date palm growing areas across Punjab and Khyber Pakhtunkhwa (KPK) provinces, Pakistan. The resistance was assessed by the diet incorporation method against the formulated commonly used chemical insecticides profenophos, imidacloprid, chlorpyrifos, cypermethrin, deltamethrin, spinosad, lambda-cyhalothrin and a fumigant phosphine. Elevated levels of resistance were recorded for cypermethrin, deltamethrin and phosphine after a long history of insecticide use in Pakistan. Resistance Ratios (RRs) were 63- to 79-fold for phosphine, 16- to 74-fold for cypermethrin, 13- to 58-fold for deltamethrin, 2.6- to 44-fold for profenophos, 3- to 24-fold for chlorpyrifos, 2- to 12-fold for lambda-cyhalothrin and 1- to 10-fold for spinosad compared to a susceptible control line. Resistant R. ferrugineus populations were mainly found in southern Punjab and to some extent in KPK. The populations from Bahawalpur, Vehari, Layyah and Dera Ghazi Khan were most resistant to chemical insecticides, while all populations exhibited high levels of resistance to phosphine. Of the eight agents tested, lower LC50 and LC90 values were recorded for spinosad and lambda-cyhalothrin. These results suggest that spinosad and lambda-cyhalothrin exhibit unique modes of action and given their better environmental profile, these two insecticides could be used in insecticide rotation or assist in phasing out the use of older insecticides. A changed pattern of both insecticides can be used sensibly be recommended without evidence of dose rates and frequencies used.

Associated patterns of insecticide resistance in field populations of malaria vectors across Africa

The development of insecticide resistance in African malaria vectors threatens the continued efficacy of important vector control methods that rely on a limited set of insecticides. To understand the operational significance of resistance we require quantitative information about levels of resistance in field populations to the suite of vector control insecticides. Estimation of resistance is complicated by the sparsity of observations in field populations, variation in resistance over time and space at local and regional scales, and cross-resistance between different insecticide types. Using observations of the prevalence of resistance in mosquito species from the Anopheles gambiae complex sampled from 1,183 locations throughout Africa, we applied Bayesian geostatistical models to quantify patterns of covariation in resistance phenotypes across different insecticides. For resistance to the three pyrethroids tested, deltamethrin, permethrin, and λ-cyhalothrin, we found consistent forms of covariation across sub-Saharan Africa and covariation between resistance to these pyrethroids and resistance to DDT. We found no evidence of resistance interactions between carbamate and organophosphate insecticides or between these insecticides and those from other classes. For pyrethroids and DDT we found significant associations between predicted mean resistance and the observed frequency of kdr mutations in the Vgsc gene in field mosquito samples, with DDT showing the strongest association. These results improve our capacity to understand and predict resistance patterns throughout Africa and can guide the development of monitoring strategies.

Neonicotinoid insecticide resistance in the field populations of Sogatella furcifera (Horváth) in Central China from 2011 to 2015

The whitebacked planthopper Sogatella furcifera (Horváth) is an important pest of rice throughout Asia. Application of chemical insecticide is the main approach to suppress the field populations of S. furcifera. In this study, neonicotinoid insecticide resistance in field populations of S. furcifera were evaluated. The results showed that some field populations of S. furcifera had developed moderate level of resistance to imidacloprid (RR=1.1–16.4), thiamethoxam (RR=0.8–14.9), dinotefuran (RR=1.2–16.6) and acetamiprid (RR=3.3–12.2), low level of resistance to nitenpyram (RR=1.1–9.5) and clothianidin (RR=1.3–8.7) in Central China. Moreover, there were an increasing trend in neonicotinoid insecticide resistance in the period 2011–2015. The results of current study may form the basis to identify and evaluate the resistance tendency of S. furcifera against neonicotinoid insecticides, which could make effective management recommendations to avoid further development of insecticide resistance in S. furcifera.

Diagnostic Doses of Insecticides for Adult Aedes aegypti to Assess Insecticide Resistance in Cuba.

The objective of this study was to determine diagnostic doses (DDs) of 5 insecticides for the Rockefeller susceptible strain of Aedes aegypti , using the Centers for Disease Control and Prevention (CDC) bottle bioassay as a tool for monitoring insecticide resistance in the Cuban vector control program. The 30-min DD values determined in this study were 13.5 μg/ml, 6.5 μg/ml, 6 μg/ml, 90.0 μg/ml, and 15.0 μg/ml for cypermethrin, deltamethrin, lambda-cyhalothrin, chlorpyrifos, and propoxur, respectively. To compare the reliability of CDC bottle bioassay with the World Health Organization susceptible test, 3 insecticide-resistant strains were evaluated for deltamethrin and lambda-cyhalothrin. Results showed that the bottles can be used effectively from 21 to 25 days after treatment and reused up to 4 times, depending on the storage time. The CDC bottle bioassay is an effective tool to assess insecticide resistance in field populations of Ae. aegypti in Cuba and can be incorporated into vector management programs using the diagnostic doses determined in this study.

Insecticide resistance in field populations of Leucinodes orbonalis (Lepidoptera: Crambidae) in India

AbstractThe status of insecticide resistance in field populations of eggplant fruit and shoot borer, Leucinodes orbonalis (Guenée) (Lepidoptera: Crambidae) from the major vegetable growing regions of India was determined during the cropping seasons of 2009–2010 and 2010–2011. Six commonly used insecticides: carbaryl, chlorpyriphos, deltamethrin, endosulfan, fenvalerate, and profenofos were tested against L. orbonalis larvae. The resistance ratios (RR) at the lethal dosage (LD)50 levels were estimated as RR=LD50 field strain/LD50 susceptible strain. The L. orbonalis populations exhibited widespread resistance to tested insecticides. The highest average RR in the two-year study was observed in the assays of populations with deltamethrin (21.50–82.42-fold) followed by assays conducted with endosulfan (24.47–68.26-fold), chlorpyriphos (22.17–63.14-fold), carbaryl (39.18–49.09-fold), and fenvalerate (14.00–44.66-fold); and the lowest average RRs were observed in the assays with profenofos (16.65–39.43-fold). The high levels of LD50 values can be attributed to the long-term indiscriminate use of these insecticides in eggplant (Solanum melongena Linnaeus; Solanaceae) growing regions.

Diagnostic doses and times for Phlebotomus papatasi and Lutzomyia longipalpis sand flies (Diptera: Psychodidae: Phlebotominae) using the CDC bottle bioassay to assess insecticide resistance.

BackgroundInsecticide resistance to synthetic chemical insecticides is a worldwide concern in phlebotomine sand flies (Diptera: Psychodidae), the vectors of Leishmania spp. parasites. The CDC bottle bioassay assesses resistance by testing populations against verified diagnostic doses and diagnostic times for an insecticide, but the assay has been used limitedly with sand flies. The objective of this study was to determine diagnostic doses and diagnostic times for laboratory Lutzomyia longipalpis (Lutz & Nieva) and Phlebotomus papatasi (Scopoli) to ten insecticides, including pyrethroids, organophosphates, carbamates, and DDT, that are used worldwide to control vectors.MethodsBioassays were conducted in 1,000-ml glass bottles each containing 10–25 sand flies from laboratory colonies of L. longipalpis or P. papatasi. Four pyrethroids, three organophosphates, two carbamates and one organochlorine, were evaluated. A series of concentrations were tested for each insecticide, and four replicates were conducted for each concentration. Diagnostic doses were determined only during the exposure bioassay for the organophosphates and carbamates. For the pyrethroids and DDT, diagnostic doses were determined for both the exposure bioassay and after a 24-hour recovery period.ResultsBoth species are highly susceptible to the carbamates as their diagnostic doses are under 7.0 μg/ml. Both species are also highly susceptible to DDT during the exposure assay as their diagnostic doses are 7.5 μg/ml, yet their diagnostic doses for the 24-h recovery period are 650.0 μg/ml for Lu. longipalpis and 470.0 μg/ml for P. papatasi.ConclusionsDiagnostic doses and diagnostic times can now be incorporated into vector management programs that use the CDC bottle bioassay to assess insecticide resistance in field populations of Lu. longipalpis and P. papatasi. These findings provide initial starting points for determining diagnostic doses and diagnostic times for other sand fly vector species and wild populations using the CDC bottle bioassay.

Effects of chlorpyrifos on enzymatic systems of Cydia pomonella (Lepidoptera: Tortricidae) adults.

The control program of codling moth (Cydia pomonella L.) in the Río Negro and Neuquén Valley is intended to neonate larvae. However, adults may be subjected to sublethal pesticide concentrations generating stress which might enhance both mutation rates and activity of the detoxification system. This study assessed the exposure effects of chlorpyrifos on target enzyme and, both detoxifying and antioxidant systems of surviving adults from both a laboratory susceptible strain (LSS) and a field population (FP). The results showed that the FP was as susceptible to chlorpyrifos as the LSS and, both exhibited a similar chlorpyrifos-inhibitory concentration 50 (IC50 ) of acetylcholinesterase (AChE). The FP displayed higher carboxylesterase (CarE) and 7-ethoxycoumarine O-deethylase (ECOD) activities than LSS. Both LSS and FP showed an increase on CarE activity after the exposure to low-chlorpyrifos concentrations, followed by enzyme inhibition at higher concentrations. There were no significant differences neither in the activities of glutathione S-transferases (GST), catalase (CAT) and superoxide dismutase (SOD) nor in the reduced glutathione (GSH) content between LSS and FP. Moreover, these enzymes were unaffected by chlorpyrifos. In conclusion, control adults from the FP exhibited higher CarE and ECOD activities than control adults from the LSS. AChE and CarE activities were the most affected by chlorpyrifos. Control strategies used for C. pomonella, such as rotations of insecticides with different modes of action, will probably delay the evolution of insecticide resistance in FPs from the study area.

Susceptibility of field populations of the diamondback moth, Plutella xylostella, to a selection of insecticides in Central China

The diamondback moth (DBM), Plutella xylostella (L.) (Lepidoptera: Plutellidae), is a globally distributed and important economic pest. Chemical control is the primary approach to regulate populations of this pest. However, resistance to insecticides evolves following heavy and frequent use. Therefore, the insecticide resistance in field populations of P. xylostella collected from Central China from 2013 to 2014 was determined with a leaf-dipping method. Based on the results of the monitoring, P. xylostella has developed high levels of resistance to beta-cypermethrin (resistance ratio=69.76–335.76-fold), Bt (WG-001) (RR=35.43–167.36), and chlorfluazuron (RR=13.60–104.95) and medium levels of resistance to chlorantraniliprole (RR=1.19–14.26), chlorfenapyr (RR=4.22–13.44), spinosad (RR=5.89–21.45), indoxacarb (RR=4.01–34.45), and abamectin (RR=23.88–95.15). By contrast, the field populations of P. xylostella remained susceptible to or developed low levels of resistance to diafenthiuron (RR=1.61–8.05), spinetoram (RR=0.88–2.35), and cyantraniliprole (RR=0.4–2.15). Moreover, the LC50 values of field populations of P. xylostella were highly positively correlated between chlorantraniliprole and cyantraniliprole (r=0.88, P=0.045), chlorantraniliprole and spinosad (r=0.66, P=0.039), spinosad and diafenthiuron (r=0.57, P=0.0060), and chlorfenapyr and diafenthiuron (r=0.51, P=0.016). Additionally, the activities of detoxification enzymes in field populations of P. xylostella were significantly positively correlated with the log LC50 values of chlorantraniliprole and spinosad. The results of this study provide an important base for developing effective and successful strategies to manage insecticide resistance in P. xylostella.

Novel mutations and mutation combinations of ryanodine receptor in a chlorantraniliprole resistant population of Plutella xylostella (L.).

A previous study documented a glycine to glutamic acid mutation (G4946E) in ryanodine receptor (RyR) was highly correlated to diamide insecticide resistance in field populations of Plutella xylostella (Lepidoptera: Plutellidae). In this study, a field population collected in Yunnan province, China, exhibited a 2128-fold resistance to chlorantraniliprole. Sequence comparison between resistant and susceptible P. xylostella revealed three novel mutations including a glutamic acid to valine substitution (E1338D), a glutamine to leucine substitution (Q4594L) and an isoleucine to methionine substitution (I4790M) in highly conserved regions of RyR. Frequency analysis of all four mutations in this field population showed that the three new mutations showed a high frequency of 100%, while the G4946E had a frequency of 20%. Furthermore, the florescent ligand binding assay revealed that the RyR containing multiple mutations displayed a significantly lower affinity to the chlorantraniliprole. The combined results suggested that the co-existence of different combinations of the four mutations was involved in the chlorantraniliprole resistance. An allele-specific PCR based method was developed for the diagnosis of the four mutations in the field populations of P. xylostella.

High level of metaflumizone resistance and multiple insecticide resistance in field populations of Spodoptera exigua (Lepidoptera: Noctuidae) in Guangdong Province, China

Spodoptera exigua is a phytophagous pest that causes critical economic losses in vegetable crops, and insecticides are commonly used against it in vegetable growing areas. However, excessive and frequent applications of insecticides cause resistance in S. exigua. The current resistance in field populations of S. exigua collected from Huizhou, Guangdong Province, China to 12 insecticides was investigated. S. exiguahad developed very high resistance to lambda cyhalothrin (2925- to 3449-fold), chlorpyrifos (>1786-fold), emamectin benzoate (174- to 867-fold), and metaflumizone (60.3- to 942-fold). High resistance to tebufenozide (51.5- to 75.4-fold) and chlorfluazuron (60.4- to 63.0-fold) was also found. Synergism assays revealed that the resistance to metaflumizone and lambda cyhalothrin was associated with esterase and microsomal oxidases, respectively. The resistance to emamectin benzoate was not affected by detoxification enzymes inhibitors and might be conferred by other mechanisms. The selection of the field population by metaflumizone for 10generations in the laboratory resulted in a 6.1-fold increase in metaflumizone resistance but did not lead to increases in resistance to other insecticides. After metaflumizone selection, susceptibilities to spinosad and endosulfan did not change, and the susceptibilities to indoxacarb, methomyl, pyridalyl, tebufenozide, chlorfluazuron, emamectin benzoate and lambda cyhalothrin decreased slightly. However, no statistically significant differences in the resistance levels were observed among the selected population, its starting strain and the unselected strain. The resistance to chlorantraniliprole noticeably decreased in unselected strain and the strain subjected to selection for 10 generations compared with their starting strains. Lack of cross-resistance to tested insecticides suggested the involvement of multiple mechanisms of resistance and the need for wise application of these insecticides for the management of S. exigua.

How many genetic options for evolving insecticide resistance in heliothine and spodopteran pests?

The widely accepted paradigm for the development of insecticide resistance in field populations of insects is of selection for one or a very few genes of major effect. Limited genetic mapping data for organophosphate and pyrethroid resistance in heliothine and spodopteran pests generally agrees with this paradigm. However, other biochemical and transcriptomic data suggest a more complex set of changes in multiple P450 and esterase gene/enzyme systems in resistant strains of these species. We discuss possible explanations for this paradox, including the likely embedding of these genes in regulatory cascades and emerging evidence for their arrangement in large clusters of closely related genes. We conclude that there could indeed be an unusually large number of genetic options for evolving resistance in these species. © 2013 Society of Chemical Industry
