The Influence of Maternal Hypoxia and Buspirone during Pregnancy on Cognitive Abilities and a Stress Response in Adult Male Offspring.

Spatial learning, memory, and reactivity of the hypothalamic-pituitary-adrenocortical system (HPA axis) were studied in adult male rats, whose mothers during pregnancy were subjected to acute moderate normobaric hypoxia, or repeated injections of buspirone, an agonist of type 1A serotonergic receptors (5HT1A), or their combination. Prenatal treatment with buspirone in rats with prenatal hypoxia impaired learning ability during the first day of 5-day training. A decrease in the effectiveness of long-term memory in comparison with short-term memory was revealed in two groups of rats: prenatal treatment with buspirone in combination with hypoxia and injection of physiological saline without hypoxia. The effectiveness of long-term memory and the level of corticosterone in response to stress did not differ between the groups, which can indicate adaptation of the 5HT1A receptor and the HPA axis to the prenatal buspirone and normobaric hypoxia during ontogeny.

Clinical efficacy of multiple intra-articular injection for hip osteoarthritis.

Intra-articular (IA) injection may be used when treating hip osteoarthritis (OA). Common injections include steroids, hyaluronic acid (HA), local anaesthetic, and platelet-rich plasma (PRP). Network meta-analysis allows for comparisons between two or more treatment groups and uses direct and indirect comparisons between interventions. This network meta-analysis aims to compare the efficacy of various IA injections used in the management of hip OA with a follow-up of up to six months. This systematic review and network meta-analysis used a Bayesian random-effects model to evaluate the direct and indirect comparisons among all treatment options. PubMed, Web of Science, Clinicaltrial.gov, EMBASE, MEDLINE, and the Cochrane Library were searched from inception to February 2023. Randomized controlled trials (RCTs) which evaluate the efficacy of HA, PRP, local anaesthetic, steroid, steroid+anaesthetic, HA+PRP, and physiological saline injection as a placebo, for patients with hip OA were included. In this meta-analysis of 16 RCTs with a total of 1,735 participants, steroid injection was found to be significantly more effective than placebo injection on reported pain at three months, but no significant difference was observed at six months. Furthermore, steroid injection was considerably more effective than placebo injection for functional outcomes at three months, while the combination of HA+PRP injection was substantially more effective at six months. Evidence suggests that steroid injection is more effective than saline injection for the treatment of hip joint pain, and restoration of functional outcomes.

Efficacy of Dexamethasone Injection at Different Sites on Postoperative Sequelae After Extracting Mandibular Impacted Third Molars: A Randomized Controlled Trial

BackgroundDexamethasone, an efficacious anti-inflammatory agent, is widely used after tooth extraction. However, its optimal injection site has yet to be investigated. PurposeWe compare the efficacy of dexamethasone injection at different sites on postoperative sequelae after extracting mandibular impacted third molars (MITMs). Study design, setting, and sampleA prospective randomized controlled trial was conducted. Healthy adults with fully MITMs scheduled for extraction were included. Exclusion criteria were 1) Patients with the systemic alteration that prevented the surgical procedure; 2) pregnancy, breastfeeding, premenstrual period; 3) hypersensitivity to the drug under test; and 4) those who did not return for postoperative follow-up at one, three, and seven days. Exposure variableThe subjects were randomized to three groups. An online randomization plan generator assigned each subject to a single treatment by randomly permuting blocks. Different sites for postoperative dexamethasone injections included the buccal side of the adjacent second molar and extraction sockets. Dexamethasone injection (4 mg) on the buccal side of the adjacent second molar (group 1), an injection on the buccal side of extraction sockets (group 2), an injection of physiological saline (0.8 mL) on the buccal side of the adjacent second molar (control). Outcome variablesThe outcome variables were postoperative facial swelling, limitation of the mouth opening, postoperative pain, and postoperative quality of life evaluation. The pain was assessed using a visual analog scale (VAS) at one, three and seven days, postoperatively. The quality of life was recorded throughout the Posse scale at seven days. CovariatesThe covariates are age, sex, length of operation, and type of impacted teeth and surgery. AnalysesThe statistical analysis was performed using ANOVA, repeated measures ANOVA, Chi-square, or Fisher’s exact tests with P-values < 0.05 considered statistically significant. ResultsOur study included 58 participants with a mean age of 19.48 ± 3.31 years; group 1 (n = 24), group 2 (n = 20), and control group (n = 14). On day three postoperative, the swelling and trismus were significantly less in group one than in the other two groups (P < 0.05), and group one had an overall postoperative quality of life compared to other groups (P < 0.05). Unaffected speech function was present in 73.7% of patients in group 1, while 50% of patients in group 2 had affected speech function three days after the operation (p < 0.05). The "unable to open mouth" of the “Eating subscale” and “felt tingling” had statistical significance (p < 0.05). ConclusionsDexamethasone injections on the buccal side of the adjacent second molar can be a viable option for treating facial swelling and limitation of mouth opening after total MITMs extraction.

Abstract TMP110: A Novel Model of Ischemia in Rats With Middle Cerebral Artery Occlusion Using a Radiopaque Blood Clot and a Microcatheter Under Fluoroscopy

Introduction: Conventional thromboembolic stroke model requires ligation of the ECA, which is problematic for research of thrombolytic agents because cerebral blood flow is different from human stroke. We aimed to develop a novel thromboembolic stroke model that does not require arterial ligation, using a radiopaque blood clot and microcatheter. Methods: Using male Sprague Dawley rats (n=14) and two sham operated rats were included, a microcatheter (inner diameter, 0.42mm; outer diameter, 0.55mm) was navigated from the caudal ventral artery to the left ICA using fluoroscopy. A blood clot (0.86 mm in diameter, 3 mm in length) containing zirconium dioxide was advanced in the catheter by injection of physiological saline to establish local occlusion. Both fluoroscopy at 1, 3, 6, 24 h and TTC staining at 24 h after stroke model creation were performed. Neurological deficit score was measured. Results: The success rate in selectively embolized of ACA and MCA bifurcation was 93% (13 of 14 cases). The median operating time was 7 min (interquartile range; 4-15 min). The median infarct volume (mm 3 ) was 248 (interquartile range; 239-280) 24 h after occlusion. Neurological deficit scores before and at 1, 3, 6, and 24 h after model creation differed significantly ( P &lt; 0.001). Fluoroscopy showed the position of the radiopaque blood clot remains the same for 24 h after creating the model in 12 of 13 cases (92%). Sham-operated rats revealed no infarction and no neurological deficit. Conclusions: We present a novel rat model for focal infarct only in the MCA territory using a radiopaque blood clot. Fluoroscopy was able to identify whether the blood clot could migrate and recanalize.

A study on the treatment of Parkinson's disease and cognitive impairment with multiple system atrophy by intranasal injection of insulin

Parkinson's disease (PD) and multiple system atrophy (MSA) are both common neurodegenerative diseases. To investigate the effects of intranasal insulin injection on motor function and cognitive impairment in patients with Parkinson's disease (PD) and multiple system atrophy (MSA).Thirteen PD patients and one MSA patient participated in the experiment . Using a random number table method, the participants were assigned to an experimental group (n=8) and a control group (n=6). The experimental group received insulin injection, while the control group received physiological saline injection. Both groups received nasal injection for 4 consecutive weeks. Compare the general information, language fluency (FAS), Montreal Cognitive Assessment Scale (MoCA), Beck Depression Inventory (BDI), and Parkinson's Unified Rating Scale (UPDRS) results between the two groups. Compared to the control group, the experimental group had a significantly higher FAS score, but there was no significant difference in the FAS score before and after treatment in the experimental group, and the prognosis score improved compared to before. In the experimental group, a MSA patient showed improvement in FAS scores during the four week treatment period, with no significant progression of the disease. There was no significant improvement in UPDRS score, BDI score, and MoCA score before and after treatment, the difference was not significant statistically. The use of insulin therapy for cognitive and motor disorders related to PD is safe and feasible. Intranasal insulin may improve functional motor function and cognitive performance in Parkinson's disease.

Effects of fecal microbiota transplantation and fecal virome transplantation on LPS-induced intestinal injury in broilers

The interesting roles and efficiencies of fecal microbiota transplantation (FMT) have attracted considerable attention and have been gradually evidenced in specific animal models. While the growing evidence that bacteriophages play roles in FMT efficacy has attracted considerable interest. In this study, we aimed to explore the effects of FMT and fecal virome transplantation (FVT) in improving inflammatory damage and ileal microbiota disorder in broilers. A total of 224 Arbor Acres broilers were selected at 1-day-old and randomly divided into the following four groups, with 56 broilers in each group: the CON group (the negative control group, sterile physiological saline injection + sterile phosphate-buffered saline (PBS) solution gavage), LPS group (the positive control group, lipopolysaccharide (LPS) injection + sterile PBS solution gavage), LPS + FMT group (LPS injection + FMT solution gavage), LPS + FVT group (LPS injection + FVT solution gavage). The results showed that: LPS injection significantly upregulated the mRNA expression levels of IFN-γ (P < 0.05) and IL-8 (P < 0.001) in ileal mucosa of broiler chickens at 11th day of age (D11), while LPS + FMT and LPS + FVT did not; LPS injection significantly upregulated the mRNA expression of ZO-1 in ileal mucosa at D11 (P < 0.01), while LPS + FMT and LPS + FVT did not; at D11, compare to CON group, LPS injection and LPS + FMT significantly increased the relative abundance of virulence factor Rab2 interacting conserved protein A related genes in broiler ileum contents (P < 0.05), while LPS + FVT had no significant difference with CON group (P > 0.05); at D11, LPS injection significantly downregulated the biosynthesis of antibiotics pathway (P < 0.05) in the ileal contents, while LPS + FVT did not. In conclusion, both FMT and FVT could promote the recovery of inflammation caused by LPS. Furthermore, FVT had shown less disadvantage stimulation on the broiler chickens and could reduce the risk of transmission of pathogenic genes, compared to FMT.

A study of Alfa Feto Protein parameter in Serum and Histological Effects of Pups Liver by SEM in Pregnant Rats Exposed to Overdose of Aminoglycoside antibiotics

The study has been conducted in Animal House/Faculty of Sciences/University of Kufa between December 2022 and February 2022, fifteen female Albino Rats are used. Gentamicin is aminoglycoside antibiotics widely used during pregnancy for treatment the infections. The present study has been intended to show Alfa Feto Protein parameter in serum and the histological effects of gentamicin on pups liver by SEM in female Albino Rats. The females Rats are randomly divided into three main groups, comprising five rats for each group .The control group is given inta peritoneal injection of physiological normal saline and the second and third group are given inta peritoneal injection of gentamicin doses twenty, forty mg/kg/day respectively for 20 days from the first day to the end of experimental allocated for each female.The rats are sacrificed in 20 th (dpc) to study the Alfa Feto Protein parameter in serum and histological effects of gentamicin on pups liver. The physiological study show negative results in all study groups due to absent of malformations in pups and present histological effects in external architecture structure of pups liver compare with control groups. In conclusion; aminoglycoside gentamicin caused histological effects on pupa liver and not absent of malformations in pups due to negative results of Alfa Feto Protein parameter.

Limosilactobacillus johnsoni and Limosilactobacillus mucosae and Their Extracellular Vesicles Alleviate Gut Inflammatory Injury by Mediating Macrophage Polarization in a Lipopolysaccharide-Challenged Piglet Model

BackgroundLimosilactobacillus johnsoni (L. j) and Limosilactobacillus mucosae (L. m) can alleviate the inflammatory response. ObjectivesThis study aimed to elucidate the underlying mechanisms by which L. j- and L. m-derived extracellular vesicles (EVs) mitigate lipopolysaccharide (LPS)-induced intestinal injury. MethodsPiglets were assigned to 4 groups: oral phosphate-buffered saline inoculation for 2 wk prior to intraperitoneal injection of physiological saline or LPS, and oral L. j/L. m inoculation for 2 wk prior to intraperitoneal injection of LPS. The intestinal integrity, macrophage markers, cytokine levels, and microbiota were determined. The cytokine levels and macrophage phenotype were detected after L. j/L. m and their EVs were coincubated with macrophages. The levels of cytokines, tight junction proteins, and apoptosis were measured after intestinal epithelial cells were cocultured with macrophages. ResultsLPS challenge decreased jejunal villus length; expression levels of zonula occludens-1 (ZO-1), occludin, arginase-1 (Arg1), and interleukin (IL)-10; and number of CD163+ cells and increased the expression levels of inducible nitric oxide synthase (iNOS), IL-1β, IL-6, and tumor necrosis factor (TNF)-α compared with that in the control. L. j and L. m pretreatment rescued the aforementioned indicators compared with LPS challenge. Pretreatment of L. j and L. m and their EVs reversed the levels of IL-1β, IL-6, TNF-α, and IL-10 and the gene expression of iNOS and Arg1 in the LPS group in macrophages. Pretreatment with L. j and L. m-derived EVs increased ZO-1 and occludin mRNA expression and reduced IL-1β, caspase-3, and bax gene expression in intestinal epithelial cells of the coculture system. Enzyme-treated EVs were less effective than native EVs. ConclusionsThis study suggests that EVs secreted by L. j and L. m control inflammation by modulating macrophage polarization, thereby improving intestinal barrier function.

Sympathetic blockade on diet induced thermogenesis in the congenic LA/Ntul//-cp rat

The effects of overfeeding highly palatable diets via the cafeteria feeding diet method (Café) has been shown to be a reliable experimental model to induce diet induced thermogenesis (DIT) in young adult normally lean rats of several strains. Groups of young adult lean LA/Ntul//-cp rats were offered a Purina chow diet (CHOW) or the same diet plus a daily café supplement both ad libitum from 10 until 24 weeks of age. The drug α-methylparatyrosine (α-MPT) was administered to groups (250 mg α-MPT/kg BW, i.p.) to ablate sympathetic (SNS) activity or a sham injection of physiologic saline given, and measures of fasting resting thermogenesis obtained at thermal neutrality (30°C) before and after the α-MPT or sham administration. The Café diet resulted in a 67% increase in body weight (BW) and a ~25% increase in resting oxygen consumption (VO2) following café overfeeding in both groups, while only a 40% increase in BW and no additional increase in VO2 occurred in normally CHOW fed rats while the sham injection was without any variations from normal physiologic responses as predicted. Sympathetic blockade with α-MPT was associated with modest decreases in body temperature and an average ~12-15% decrease in VO2 in the café treated group, but when the VO2 data were arithmetically corrected to isothermal conditions only a net 3% decrease occurred in CHOW fed rats and was without effect on the SHAM group. Serum T3 concentrations increased by 92% and urinary catecholamine excretion of VMA &gt;250% following the café diet, but the urinary vanilmandelic acid (VMA) excretion was virtually nil following the sympathetic blockade. These observations indicate that the SNS-mediated contribution component to DIT following prolonged café overfeeding under conditions of thermal neutrality contribute approximately 50% of the thermic response. while under normal long term CHOW feeding the SNS component may be as little as 3 % of the thermic response, and thus, the total thermic response of DIT likely represents a combination of short acting SNS and longer acting non-SNS mechanisms, including a likely significant thyroidal component in normally lean animals.

Immunological effect of Lactic acid bacteria adjuvant on in ovo injection of Newcastle disease vaccine

In ovo immunization of chicken embryos with live vaccines is an effective strategy to protect chickens against various viral pathogens. The immunogenic efficacies of in ovo administration of lactic acid bacteria (LAB) in combination with live Newcastle disease (ND) vaccine were investigated in this study. Four hundred healthy 1-day-old fertilized specific pathogen-free (SPF) eggs of similar weights were randomly assigned to one of four treatments, with five replicates of each treatment and a total of 20 for each replicate. On day 18.5 of incubation, in ovo injections were given. The treatment groups are as follows: (I) no injection, (II) 0.9% physiological saline injection, (III) ND vaccine injection, and (IV) LAB as an adjuvant for ND vaccine injection. The ND vaccine adjuvanted with LAB significantly increased the daily weight gain, immune organ index, and small intestine histomorphological development in layer chicks while decreasing the feed conversion ratio (FCR). The results suggested that the LAB-adjuvant group significantly affected the relative expression of mucosal mucin protein (mucin-1) and zoccluding small circle protein-1 (ZO-1) (P < 0.05), whereas the relative expression of occludin mRNA was not significantly affected (P > 0.05) compared with the non-injected group. Meanwhile, we indicated that intra-amniotic synbiotic injection significantly maintained the balance of flora (P < 0.05). Compared with the non-injected group, the ND vaccine adjuvanted with the LAB group exhibited significant promotion of the HI and SIgA antibody titers in serum on day 21 (P < 0.05), induction of higher production of cytokines (IL-2, IL-4, IL-6, IFN-γ) in serum. In summary, in ovo injection of ND vaccine adjuvanted with LAB has a positive impact on the growth performance, immune function, and microbiome of growing chicks.

The study aims to analyze furocoumarin extracts isolated from poison hemlock cell culture influencing the correction of cisplatin-caused cytostatic myelosuppression. The experiments were carried out on 160 CD1 female mice weighing 18–25 g. Cytostatic myelosuppression was simulated by administering cisplatin intraperitoneally once at a maximum tolerated dose of 10 mg/kg. The control group received physiological saline injections. Poison hemlock (Conium maculatum L.) cell culture extract was administered at a dose of 30 mg/kg to prevent the development of disorders. 4 mg/kg of Warfarin Nycomed was used for comparison examination. The correctors were administered intragastrically on the sixth day after cisplatin injected for four days. The indicators were examined on the 1st, 2nd, 5th, 7th, 10th, 15th, 20th and 30th days following the administration of cisplatin. The bone marrow and peripheral blood values were examined using the standard method. Statistical analysis was carried out using Stat Plus Pro (build 7.3.0.0). Under conditions of cytostatic myelosuppression caused by the administration of 10 mg/kg (maximum tolerated dose) of cisplatin, the use of the poison hemlock cell culture extract contributes to the restoration of myeloid and erythroid hematopoietic germs, as well as the normalization of bone marrow and peripheral blood values. With increase in the number of mature neutrophils and lymphocytes, erythroblasts, and normoblasts, the total number of myelokaryocytes increased. The content of erythrocytes and leukocytes increased in the peripheral blood as the number of segmented neutrophils and lymphocytes increased. The dynamics of hematopoietic sprout recovery with poison hemlock cell culture extract are similar to those of Warfarin.

Vaccines for the common cold.

The common cold is a spontaneously remitting infection of the upper respiratory tract, characterised by a runny nose, nasal congestion, sneezing, cough, malaise, sore throat, and fever (usually < 37.8 ºC). Whilst the common cold is generally not harmful, it is a cause of economic burden due to school and work absenteeism. In the United States, economic loss due to the common cold is estimated at more than USD 40 billion per year, including an estimate of 70 million workdays missed by employees, 189 million school days missed by children, and 126 million workdays missed by parents caring for children with a cold. Additionally, data from Europe show that the total cost per episode may be up to EUR 1102. There is also a large expenditure due to inappropriate antimicrobial prescription. Vaccine development for the common cold has been difficult due to antigenic variability of the common cold viruses; even bacteria can act as infective agents. Uncertainty remains regarding the efficacy and safety of interventions for preventing the common cold in healthy people, thus we performed an update of this Cochrane Review, which was first published in 2011 and updated in 2013 and 2017. To assess the clinical effectiveness and safety of vaccines for preventing the common cold in healthy people. We searched the Cochrane Central Register of Controlled Trials (CENTRAL) (April 2022), MEDLINE (1948 to April 2022), Embase (1974 to April 2022), CINAHL (1981 to April 2022), and LILACS (1982 to April 2022). We also searched three trials registers for ongoing studies, and four websites for additional trials (April 2022). We did not impose any language or date restrictions. Randomised controlled trials (RCTs) of any virus vaccine compared with placebo to prevent the common cold in healthy people. We used Cochrane's Screen4Me workflow to assess the initial search results. Four review authors independently performed title and abstract screening to identify potentially relevant studies. We retrieved the full-text articles for those studies deemed potentially relevant, and the review authors independently screened the full-text reports for inclusion in the review, recording reasons for exclusion of the excluded studies. Any disagreements were resolved by discussion or by consulting a third review author when needed. Two review authors independently collected data on a data extraction form, resolving any disagreements by consensus or by involving a third review author. We double-checked data transferred into Review Manager 5 software. Three review authors independently assessed risk of bias using RoB 1 tool as outlined in the Cochrane Handbook for Systematic Reviews of Interventions. We carried out statistical analysis using Review Manager 5. We did not conduct a meta-analysis, and we did not assess publication bias. We used GRADEpro GDT software to assess the certainty of the evidence and to create a summary of findings table. MAIN RESULTS: We did not identify any new RCTs for inclusion in this update. This review includes one RCT conducted in 1965 with an overall high risk of bias. The RCT included 2307 healthy young men in a military facility, all of whom were included in the analyses, and compared the effect of three adenovirus vaccines (live, inactivated type 4, and inactivated type 4 and 7) against a placebo (injection of physiological saline or gelatin capsule). There were 13 (1.14%) events in 1139 participants in the vaccine group, and 14 (1.19%) events in 1168 participants in the placebo group. Overall, we do not know if there is a difference between the adenovirus vaccine and placebo in reducing the incidence of the common cold (risk ratio 0.95, 95% confidence interval 0.45 to 2.02; very low-certainty evidence). Furthermore, no difference in adverse events when comparing live vaccine preparation with placebo was reported. We downgraded the certainty of the evidence to very low due to unclear risk of bias, indirectness because the population of this study was only young men, and imprecision because confidence intervals were wide and the number of events was low. The included study did not assess vaccine-related or all-cause mortality. AUTHORS' CONCLUSIONS: This Cochrane Review was based on one study with very low-certainty evidence, which showed that there may be no difference between the adenovirus vaccine and placebo in reducing the incidence of the common cold. We identified a need for well-designed, adequately powered RCTs to investigate vaccines for the common cold in healthy people. Future trials on interventions for preventing the common cold should assess a variety of virus vaccines for this condition, and should measure such outcomes as common cold incidence, vaccine safety, and mortality (all-cause and related to the vaccine).

Effects of betaine on growth performance, intestinal health, and immune response of goslings challenged with lipopolysaccharide

The objective of this experiment was to investigate the effects of betaine on growth performance, serum parameters, intestinal health, and immune performance of goslings in response to lipopolysaccharide (LPS) challenge. A total of 168 healthy male 15-day-old Jiangnan White Goslings were randomly divided into 4 groups, with 6 replicates per treatment and seven goslings per replicate. A 2 × 2 factorial arrangement included 2 factors, that is, LPS challenge (injection of LPS or physiological saline) and betaine (added 0 or 0.06% betaine in diet). The results indicated that LPS challenge significantly reduced the average daily feed intake (ADFI), average daily gain (ADG), and body weight (BW) at 21 D of the goslings, while dietary betaine supplementation tended to increase the ADFI during the LPS stress period (P = 0.08) and BW at 21 D of the goslings (P = 0.09). The LPS-challenged goslings showed higher pro-inflammatory cytokines (interleukin-1 [IL-1β], interleukin-6 [IL-6], tumor necrosis factor-α (TNF-α), and Interferon-gamma [IFN-γ]) and lower anti-inflammatory cytokine (Interleukin-10 [IL-10]) (P < 0.05) at 21 D of age. Dietary betaine supplementation alleviated LPS-induced increase in pro-inflammatory cytokines. The LPS challenge significantly decreased duodenal and jejunal villus height (VH) and villus height and crypt depth ratio (VCR), while the addition of betaine significantly increased duodenal VH and VCR (P < 0.05). On the other hand, addition of betaine significantly alleviated decline of enzyme activity on lipase, amylase, trypsin, and chymotrypsin in the intestinal of goslings. The LPS challenge significantly increased the content of serum D-lactic acid (D-LA) and the activity of diamine oxidase (DAO) at 21 D of the goslings. The LPS challenge and betaine addition significantly increased the mRNA expression of Occcludin (OCLN) in jejunal mucosa at 28 D of the goslings (P < 0.05). In conclusion, our research demonstrated that betaine can alleviate the decline of growth performance and immune performance in goslings caused by LPS. The results also indicate betaine possesses anti-inflammation properties and improves intestinal barrier functions. We recommend that 0.06% betaine be added into the diet to improve the intestinal health and immune performance of goslings.

Histopathological effects of retinol on blastocyst implantation in pregnant rats

Retinol is lipid soluble vitamin widely used during pregnancy as a food supplement.The study has been conducted in Animal House/Faculty of Sciences/University of Kufa between December 2021 and February 2022, fifteen female Albino Rats are used. The present study has been intended to show the histolopathologiacl effects of retinol on blastocyst implantation in female Albino Rats. The females Rats are randomly divided into three main groups, comprising five rats for each group. The control group is given orally injection of physiological normal saline and the second and third group are given orally injection of retinol doses ten, twenty μcg/kg/day respectively for seven days from the first day to the end of experimental allocated for each female.The rats are sacrificed in 7 th (dpc) to study the implantation of embryos in decidual tissues. Retinol doses ten, twenty μcg/kg/day caused macroscopic malformations in embryos of female rats compare to control group. The histopathlogical study of female rats embryos treated with ten, twenty μcg/kg/day of retinol have shown deformations of decidual tissues zonation.In conclusion; retinol caused malformations of embryos in 7 th (dpc) and histopathlogical effects in decidual tissue zonations of blastocyst.

Teratogenic effects of vitamin A on embryos of pregnant rats

Vitamin A is lipid soluble vitamin widely used during pregnancy as a food supplement. The study has been conducted in Animal House/Faculty of Sciences/University of Kufa between December 2021 and February 2022, thirty female Albino Rats are used. The present study has been intended to show the teratogenic effects of Vitamin A on embryos in female Albino Rats. The females Rats are randomly divided into four main groups,The first and second control groups are given orally injection of physiological normal saline and the third group 15th (dpc) are given orally injection of vitamin A doses ten, twenty μcg/kg/day respectively for fifteen days from the first day to the end of experimental allocated for each female. The fouth group 20 th (dpc) are given orally injection of vitamin A doses ten, twenty μcg/kg/day respectively for tewnty days from the first day to the end of experimental allocated for each female. The rats are sacrificed in 15,20 th (dpc) to study the teratogenic effects on embryonic bodies.Retinol doses ten, twenty μcg/kg/day caused macroscopic malformations in embryos of female rats compare to control group.In conclusion; Vitamin A caused malformations of embryos in 15 th and 20 th (dpc).

Acute cholangitis due to haemobilia complicating percutaneous cholecystostomy: First literature case report.

BackgroundLaparoscopic cholecystectomy is the standard treatment for acute cholecystitis. Cholecystostomy is a good option in patients with significant comorbidities. We report a case of a patient having had a percutaneous cholecystostomy for acute cholecystitis complicated with haemobilia and acute cholangitis.Presentation of a caseA woman aged 64 years old, with a history of diabetes, arterial hypertension, and chronic obstructive pulmonary disease was admitted to our institution with acute cholecystitis.We opted for transhepatic percutaneous cholecystostomy (PC) and antibiotics.On the fourth day, the patient had acute cholangitis due to haemobilia.We injected physiologic saline serum through the drain of cholecystostomy to dissolve the blood clot. There was a clinical improvement.We performed laparoscopic cholecystectomy two months later. The patient had an uneventful recovery with a follow-up of five months.DiscussionWe report the first literature report of acute cholangitis due to haemobilia complicating percutaneous cholecystostomy in a patient admitted for cholecystitis.We highlight the importance of the injection of saline physiologic serum from the catheter. Medical treatment with antibiotics may be enough knowing that blood clots can disappear spontaneously. In case of failure, ERCP with sphincterotomy should be performed.ConclusionHaemobilia causing acute cholangitis is a rare complication of percutaneous cholecystostomy. Conservative treatment with antibiotics and injection of saline physiologic serum from the catheter is a good treatment option. In case of failure, ERCP should not be delayed.

Sympathetic blockade on diet induced thermogenesis in the congenic LA/Ntul//-cp rat

The effects of overfeeding highly palatable diets via the cafeteria feeding diet method (Café) has been shown to be a reliable experimental model to induce diet induced thermogenesis (DIT) in young adult normally lean rats of several strains. Groups of young adult lean LA/ Ntul//-cp rats were offered a Purina chow diet (CHOW) or the same diet plus a daily café supplement both ad libitum from 10 until 24 weeks of age. The drug α-methylparatyrosine (α-MPT) was administered to groups (250 mg α-MPT/kg BW, i.p.) to ablate sympathetic (SNS) activity or a sham injection of physiologic saline given, and measures of fasting resting thermogenesis obtained at thermal neutrality (30°C) before and after the α-MPT or sham administration. The Café diet resulted in a 67% increase in body weight (BW) and a ~25% increase in resting oxygen consumption (VO2) following café overfeeding in both groups, while only a 40% increase in BW and no additional increase in VO2 occurred in normally CHOW fed rats while the sham injection was without any variations from normal physiologic responses as predicted. Sympathetic blockade with α-MPT was associated with modest decreases in body temperature and an average ~12-15% decrease in VO2 in the café treated group, but when the VO2 data were arithmetically corrected to isothermal conditions only a net 3% decrease occurred in CHOW fed rats and was without effect on the SHAM group. Serum T3 concentrations increased by 92% and urinary catecholamine excretion of VMA &gt;250% following the café diet, but the urinary vanilmandelic acid (VMA) excretion was virtually nil following the sympathetic blockade. These observations indicate that the SNS-mediated contribution component to DIT following prolonged café overfeeding under conditions of thermal neutrality contribute approximately 50% of the thermic response. While under normal long term CHOW feeding the SNS component may be as little as 3% of the thermic response, and thus, the total thermic response of DIT likely represents a combination of short acting SNS and longer acting non-SNS mechanisms, including a likely significant thyroidal component in normally lean animals.

The influence of epidural volume extension on spinal block with hyperbaric bupivacaine for elective knee arthroplasty

BackgroundEpidural volume extension (EVE) via a combined spinal–epidural is the enhancement of a small-dose intrathecal block by an epidural injection of physiological saline solution. The aim of this study was to investigate the sensory and motor block characteristics of spinal anesthesia after EVE. Eighty patients enrolled in this prospective, randomized, double-blind study. Group I (n=40) received 10 mg hyperbaric bupivacaine and group II (n=40) first received 10 mg hyperbaric bupivacaine intrathecally and subsequently 10 mL saline via epidural catheter.ResultsIn the first 30 min after combined spinal–epidural anesthesia, the maximum sensory block level was significantly higher in group II than in group I. The Bromage score was significantly higher in group II than in group I at 3–6 and 9 min of the intraoperative period. EVE increased the mean Smax significantly in group 2 than group 1 (p<0.05). Tmax was statistically similar between group 1 and group 2 (p>0.05). EVE significantly altered Time10 and Tadeq in group 2 (p<0.05).ConclusionsEVE with saline (10 mL) offer early onset of sensory and motor block and a high level of sensory block. And also delay supplemental epidural dose requirement intraoperatively.

Activation of Age-Related Nuclear Factor-κB Signaling Pathway Leads to Chronic Inflammation and Pituitary Fibrosis

The purpose of the present study was to investigate the mechanism of pituitary fibrosis in elderly people. First, 20 pituitary glands obtained from 11 elderly people and 9 young people were studied using Masson's trichrome staining for fibrosis detection. Second, pituitary glands from 12 male rats, including 6 aged rats (OM group) and 6 young rats (YM group), were also studied. Western blotting was performed to detect collagen 1 and phosphorylation of the nuclear factor (NF)-κB subunit p65 in the OM and YM groups. The levels of 8 proinflammatory cytokines (interleukin [IL]-1α, IL-1β, IL-2, IL-4, IL-6, IL-8, interferon-γ, and tumor necrosis factor-α) in the rat pituitary glands were detected using liquid suspension chip technology. Enzyme-linked immunosorbent assays were performed to detect the growth hormone (GH) levels in the venous blood samples from the rats. Next, 12 aged rats were randomly divided into 2 groups: the QNZ (Q)+OM and normal physiological saline (N)+OM groups. The Q+OM and N+OM groups had undergone intervention by intraperitoneally injection of QNZ and physiological saline (1 mg/kg) for 28 days, respectively. Finally, biochemical and histological examinations were performed, including Masson's trichrome staining for fibrosis, Western blotting for phosphorylation of p65, Millipore multiplex bead arrays (Millipore, Billerica, Massachusetts, USA) for proinflammatory cytokine levels, and enzyme-linked immunosorbent assays for GH secretion. Fibrosis was detected in the elderly patient group. Collagen 1, phosphorylation of the NF-κB signaling pathway, and the proinflammatory cytokine levels showed a significant increase in the OM group. Compared with the N+OM group, pituitary fibrosis was alleviated in the Q+OM group, with an increase in GH secretion and decreased proinflammatory cytokine levels and NF-κB. Pituitary fibrosis was found in the elderly group, and the pathological change was antagonized by decreasing the proinflammatory cytokine levels using QNZ and further increasing GH secretion.

L. reuteri ZJ617 inhibits inflammatory and autophagy signaling pathways in gut-liver axis in piglet induced by lipopolysaccharide

BackgroundThis study investigated the protective effects of L. reuteri ZJ617 on intestinal and liver injury and the underlying mechanisms in modulating inflammatory, autophagy, and apoptosis signaling pathways in a piglet challenged with lipopolysaccharide (LPS).MethodsDuroc × Landrace × Large White piglets were assigned to 3 groups (n = 6/group): control (CON) and LPS groups received oral phosphate-buffered saline for 2 weeks before intraperitoneal injection (i.p.) of physiological saline or LPS (25 μg/kg body weight), respectively, while the ZJ617 + LPS group was orally inoculated with ZJ617 for 2 weeks before i.p. of LPS. Piglets were sacrificed 4 h after LPS injection to determine intestinal integrity, serum biochemical parameters, inflammatory signaling involved in molecular and liver injury pathways.ResultsCompared with controls, LPS stimulation significantly increased intestinal phosphorylated-p38 MAPK, phosphorylated-ERK and JNK protein levels and decreased IκBα protein expression, while serum LPS, TNF-α, and IL-6 concentrations (P < 0.05) increased. ZJ617 pretreatment significantly countered the effects induced by LPS alone, with the exception of p-JNK protein levels. Compared with controls, LPS stimulation significantly increased LC3, Atg5, and Beclin-1 protein expression (P < 0.05) but decreased ZO-1, claudin-3, and occludin protein expression (P < 0.05) and increased serum DAO and D-xylose levels, effects that were all countered by ZJ617 pretreatment. LPS induced significantly higher hepatic LC3, Atg5, Beclin-1, SOD-2, and Bax protein expression (P < 0.05) and lower hepatic total bile acid (TBA) levels (P < 0.05) compared with controls. ZJ617 pretreatment significantly decreased hepatic Beclin-1, SOD2, and Bax protein expression (P < 0.05) and showed a tendency to decrease hepatic TBA (P = 0.0743) induced by LPS treatment. Pretreatment of ZJ617 before LPS injection induced the production of 5 significant metabolites in the intestinal contents: capric acid, isoleucine 1TMS, glycerol-1-phosphate byproduct, linoleic acid, alanine-alanine (P < 0.05).ConclusionsThese results demonstrated that ZJ617 pretreatment alleviated LPS-induced intestinal tight junction protein destruction, and intestinal and hepatic inflammatory and autophagy signal activation in the piglets.
