Effect of a pre-calving injectable trace mineral supplement on white blood cell function in seasonally calving pastoral dairy cows

ABSTRACT Aims To investigate the effect of injection of trace mineral supplement (TMS) 14–28 days before calving on white blood cell count (WBCC) and function, serum antioxidant capacity (SAC) and reactive oxygen species (ROS) in pasture-fed cattle after calving. Methods On each of two South Island, seasonally calving, pastoral dairy farms,1 month before dry-off, a random sample of 150 multiparous cows predicted to calve within 7 days of the herd’s planned start of calving (PSC) were stratified on individual somatic cell count, age, breed and expected calving date. On each farm, 14–24 days before PSC, 60 selected cows were randomly assigned for TMS (Zn, Mn, Se, Cu) injection, and 60 were controls. All 240 cows were contemporaneously injected with hydroxocobalamin, and controls with Se. Blood samples were collected pre-injection and 3, 12 and 40 days after calving. Phagocytic activity, count and proportion of neutrophils, lymphocytes and monocytes, WBCC, ROS, SAC were measured. Plasma concentrations of Se, Cu and glutathione peroxidase (GPx) were monitored from a random subset of animals. Differences attributable to TMS were estimated using mixed-multivariable Bayesian analysis, expressed as mean and highest density interval (HDI). Results Three and 40 days after calving, TMS-treated cows had 0.36 (90% HDI = 0.00–0.77) x 109 and 0.25 (90% HDI = 0.00–0.55) x 109 fewer neutrophils/L. Neutrophils comprised 6 (90% HDI = 0–11)% and 4 (90% HDI = 0–8)% less of the WBCC, and the neutrophil count was 14 (90% HDI = 0–27)% and 9 (90% HDI = 0–18)% less than controls. However, 3 days after calving, there were 7 (95% HDI = 2–12)% more cells phagocytosing and 2,900 (95% HDI = 2,600–3,200) more bacteria ingested/cell. Twelve and 40 days after calving, TMS-treated cows had 0.65 (95% HDI = 0.17–1.17) x 109 and 0.28 (95% HDI = 0.00–0.59) x 109 more lymphocytes/L. Lymphocytes comprised 10 (95% HDI = 3–18)% and 5 (95% HDI = 0–9)% more of the WBCC, and the lymphocyte count was 30 (95% HDI = 11–51)% and 9 (95% HDI = 0–9)% more than controls. There were no meaningful differences in ROS, SAC, ROS/SAC, other white blood cells, or WBCC. Plasma Cu, Se and GPx concentrations were above recommended thresholds. Conclusions Pre-calving TMS injection was associated with differences in white blood cell population and function that may reduce the risk of disease. Effect Of Injectable Trace Minerals on The Humoral Immune Response to Multivalent Viral Vaccine in Dairy Cows

Background: This study was done to investigate the effects of injectable trace minerals on the humeral immune response of dairy cows receiving multivalent viral vaccine. Materials and methods: Forty cows divided into 2 groups; average BW = 350 - 420 kg. All were seronegative for FMD virus, lumpy skin, BFV, BVD and IBRV. First group (Gp1) n= 20 Holstein cows vaccinated with FMD virus, lumpy skin, BFV, BVD and IBRV, and the 2 nd group (Gp2) vaccinated and treated with injectable trace mineral supplement 7 mL subcutaneous (MultiMin 90, USA) containing Cu, Zn, Mn (all as disodium EDTAsalts), and Se. Humoral immunity was detected by neutralizing antibody titers measured on day one, 7th week and 14th weeks experiment relative to vaccines administration. All cows were seronegative for each of the 5 viruses on day zero, serum mineral concentrations were evaluated on day one, 7 w and 14 w. Results: The higher average of milk production in 7 th and 14th week in the Gp2 compared to Gp1, there are clear increases in the concentration of minerals (Cu, Zn, Se, Mo, Co, Mn, Fe) in Gp2 compared to Gp1, which indicates better absorption of these minerals when they are injection. The mean antibodies titers were gradually increased from 7th weeks post-vaccines till reached higher level at 14th weeks post- vaccines for all viral vaccines as measured by SNT and ELISA test. The antibodies peak titer was observed higher at 14th w post-vaccination in Gp2 significantly greater than Gp1. Conclusion: Viral vaccination concurrent with ITM, induced a rapid protection against viral infection. Impact of an Injectable Trace Mineral Supplement on the Immune Response and Outcome of Mannheimia haemolytica Infection in Feedlot Cattle

AbstractThe study aimed to assess the impact of injectable trace mineral (“ITM”; Multimin90; Fort Collins, CO) supplementation on bacterial infection in cattle. Angus-crossbred steers (n = 32) were organized into two blocks by initial body weight. Steers were maintained on a ryelage and dry-rolled corn-based growing diet without supplementation of Zn, Cu, Mn, and Se for the duration of the study. The steers were transported 6 h, then randomized into three treatment groups: control received sterile saline (“CON”), ITM administered 1 day after transport (6 days before infection, “ITMPRE”), and ITM administered 2 days post infection (dpi) concurrent with antibiotic treatment (“ITMPOST”). Steers were infected with Mannheimia haemolytica on day 0, and all were treated with tulathromycin at 2 dpi. Plasma levels of Zn, Cu, and Se did not differ among treatments (P ≥ 0.74). Liver Se was higher in ITMPRE at 2 dpi (P < 0.05), and both ITM groups had higher liver Se at 5 dpi (P < 0.05) compared to CON. A time × treatment interaction was detected for liver Cu (P = 0.02). Clinical scores were lower (P < 0.05) in ITMPRE on 1 and 8 dpi and ITMPOST on 8 dpi compared to CON. Thoracic ultrasonography scores were lower in ITMPRE at 2 dpi compared to CON (P < 0.05) and ITMPOST (P < 0.1). No treatment effects (P > 0.10) were observed for bacterial detection from bronchoalveolar lavage (BAL) or nasopharyngeal swabs. At 5 dpi, both ITMPRE and ITMPOST showed higher frequencies of γδ T cells and NK cells in BAL compared to CON (P < 0.05). Before infection, leukocytes from ITMPRE steers produced more IL-6 (P < 0.01) in response to stimulation with the TLR agonist, Pam3CSK4. Use of ITM may be an effective strategy for improving disease resistance in feedlot cattle facing health challenges.

PSVIII-A-1 Effect of Source of Trace Mineral and Bismuth Subsalicylate on Growth, Ruminal H2s, and Trace Mineral Status of Growing Heifers

Abstract This study evaluated the effects of providing inorganic, chelated, and inorganic plus injectable trace mineral supplementation strategies with or without the inclusion of bismuth subsalicylate (BSS) on dry matter intake (DMI), water intake, ruminal hydrogen sulfide (H2S) concentration, and trace mineral status of growing beef heifers provided high sulfate (5,055 ± 228 mg/L) water. This study consisted of an 84-d feeding period with a 2×2 + 1 factorial treatment arrangement conducted using 2 blocks. Beef heifers (n = 15/block; 367 ± 33 kg BW) in each block were stratified according to their initial liver copper concentration. Mineral treatments included inorganic [added CuSO4 (7.76 mg/kg), ZnSO4 (22.92 mg/kg), MnSO4 (15.28 mg/kg)], chelated (100% of the Cu-methionine (7.68 mg/kg ), Zn-methionine (22.89 mg/kg), and Mn-methionine (15.27 mg/kg), and injectable trace mineral (15 mg/mL Cu, 10 mg/mL Mn, 60 mg/mL Zn) provided in combination with the inorganic mineral treatment. The BSS treatments were 0.0 (CON) or 0.2% (BSS) on a DM basis. Feed and water intake (weekly), ruminal H2S concentration (d 42 and 84), and liver (pre-study, d 42 and 84) and serum trace mineral concentrations (d 1, 28, 56, and 84) were evaluated. Initial liver trace mineral concentration was used as a covariate. Dry matter intake tended to be greater (mineral × BSS, P = 0.05) for heifers provided chelated minerals and fed CON, compared with BSS, and tended to be less for heifers fed inorganic and CON than BSS. Heifers fed chelated minerals drank 6.1 L/d more (P < 0.05) water than those provided inorganic minerals. Injectable trace-mineral provision did not affect DMI or water intake. Ruminal H2S was not affected by mineral type or BSS (P ≥ 0.33). The inclusion of BSS reduced (P < 0.01) liver Cu concentration from 60.5 to 31.3 ppm. Heifers provided the injectable minerals had greater (P < 0.01) liver Cu concentration than the inorganic without BSS treatment. Serum copper was not affected by BSS or mineral treatment (P ≥ 0.44). The liver concentration of Se was reduced (P < 0.01) by the inclusion of BSS. The serum Se concentration was not affected by mineral type, BSS, time, or any two- or three-way interactions (P ≥ 0.24). Bismuth subsalicylate did not affect ruminal H2S, and negatively affected liver Cu concentration. The use of organic trace mineral supplementation strategies did not affect the trace mineral status of beef heifers drinking high sulfate water, but use of an injectable trace mineral supplement increased liver Cu.

Evaluation of the stress‐reducing effect of trace mineral injection in beef calves

Little is known about the effects of trace mineral supplementation on the stress response in beef calves. To investigate the effect of injectable trace mineral supplementation (ITM) on the stress response in beef calves exposed to different types of stress. Thirty weaned Angus and Angus crossbred calves. The enrolled calves were randomly assigned to 2 groups: ITM, 15 calves received modified-live virus vaccine (MLV) and ITM SC and 15 calves received MLV and saline SC (CONT). The calves were exposed to 3 types of stress: the stress of MLV vaccination (d0), nasal aerosol with bovine viral diarrhea virus-2 (BVDV-2) challenge (d5), and liver biopsy (d26). The calves' body weights and health status were monitored. Leukocyte counts, serum cortisol concentration ([cort]), BVDV-2 serum neutralizing antibodies (SNA), and percentages of CD4+ , CD8+ , WC1+ , and CD25+ T-lymphocytes were measured. Serum cortisol concentration ([cort]) showed strong associations with the percentage of CD8+ (rs = .50), BVDV2-SNA (rs = -.43), and WC1CD25+ (rs = .41) cells, and rectal temperature (rs = .40). The highest [cort] was reported 3 days after aerosol BVDV-2 challenge. Serum [cort] was decreased in ITM-treated calves 3 days post-BVDV-2 challenge, compared with CONT calves, with an average decrease of 18.5 ng/μL (95% confidence interval [CI], -6.07 to -31.3). The ITM-treated calves were heavier and healthier (P < .01)than the CONT calves. Trace mineral supplementation appears to have stress mitigation effects in beef cattle that may reflect positively on growth and health performance. Viral exposure is associated with a high degree of stress, which is considered a major welfare concern.

Effect of injectable trace mineral supplementation on peripheral polymorphonuclear leukocyte function, antioxidant enzymes, health, and performance in dairy cows in semi-arid conditions

The objective of this study was to evaluate the effect of subcutaneous injections of 15 mg/mL Cu, 5 mg/mL Se, 60 mg/mL Zn, and 10 mg/mL Mn on health, performance, polymorphonuclear leukocyte (PMNL) function, circulating glutathione peroxidase (GPx) and superoxide dismutase (SOD) concentrations, and inflammation of dairy cows undergoing the transition period in high temperature-humidity index. A total of 923 multiparous cows from 2 commercial dairy farms were randomly allocated into 1 of 2 treatment groups as follows: control and injectable trace mineral supplementation (ITMS). Cows in the ITMS group received 7 mL of subcutaneous injections at dry-off (208 ± 3 d of gestation), 260 ± 3 d of gestation, and at 35 ± 3 d in milk (DIM). Data regarding health traits, reproductive performance, milk yield, and survivability were extracted from farm database software, and animals were followed-up until 300 DIM. For a subset of 142 cows from one herd, blood samples were collected at enrollment, and at 3 ± 1, 7 ± 1, 10 ± 1, and 35 ± 3 DIM to evaluate hematology, PMNL function, GPx and SOD concentrations, and circulating haptoglobin. Logistic regression was used to assess health and pregnancy per artificial insemination at first service. Cox proportional hazards models were used to evaluate hazard of pregnancy and culling. Mixed linear regression models accounting for repeated measures were used to assess all continuous variables collected over time. Parity, twinning, and previous gestation length were considered as potential confounders. Farm was included as a random effect. The ITMS cows tended to have lower incidence of metritis and stillbirth compared with control group. However, ITMS treatment did not influence the incidence of other diseases (e.g., mastitis, retained placenta), milk yield, reproductive performance, culling, and leukocyte count. Neutrophil-to-lymphocyte ratio, PMNL phagocytosis, and oxidative burst as well as intensity of the oxidative burst were greater for ITMS-treated cows in comparison to control cows. The ITMS cows had decreased expression of the adhesion molecule L-selectin on PMNL surface. The serum concentration of GPx and SOD were not affected by ITMS treatment. In conclusion, ITMS tended to reduce the incidence of metritis and stillbirth parturition, improved PMNL function, and improved the inflammatory status of dairy cows undergoing the transition period in high temperature-humidity index conditions. However, these findings did not translate into improved milk yield, reproductive performance, and survivability.

PSV-1 Injectable trace mineral supplementation on grazing beef cows: Effect on subsequent offspring birth weight and weaning weight

Abstract This study evaluated the effect of an injectable multi-mineral complex supplementation on grazing beef cows on subsequent offspring birth weight (BW) and adjusted weaning weight (AWW). Experiment 1 included 1,128 Angus crossbred multiparous suckled cows, from 9 locations in Virginia, that were randomly assigned to one of two treatments: 1) One dose of an injectable trace mineral (6 mL; Multimin® 90, Multimin USA, Ft. Collins, CO) at 71.4±16.8 d of lactation (n = 560); or 2) a negative control with no trace mineral injection (n = 568). In experiment 2, 986 Angus crossbred multiparous suckled cows, at 7 locations in Virginia, were randomly assigned to one of two treatments: 1) Two doses of an injectable trace mineral (6 mL; Multimin® 90) at 118.3±17 days before calving and at 67.7±17.4 d of lactation (n = 494); or 2) negative control with no trace mineral injection (n = 492). All cows had ad libitum access to forage and water, and received additional mineral supplementation in form of mineral blocks. Calves were weaned at approximately 7 months of age, and 205-day AWW was calculated based on birth day, age of weaning and age of dam. No differences between treatments were detected for BW (33.7±1.01 kg; P = 0.50) and AWW (212.2±5.0 kg; P = 0.25) when only one dose of injectable trace mineral was administered to the dam at early lactation. Similarly, when two doses of injectable trace mineral were administrated BW (35.6±1.1 kg; P = 0.70) and AWW (259.4±6.1 kg; P = 0.83) did not differ between treatments. In conclusion, one or two doses of injectable trace mineral administrated at late gestation and/or during lactation had no effect on offspring BW and AWW.

156 Injectable trace mineral supplementation on grazing beef cows: effect on overall mineral status and fertility

Abstract The objective of this study was to evaluate the effect of two doses of an injectable multi-mineral complex supplementation on beef cow fertility and overall mineral status. A total of 986 Angus crossbred multiparous cows at 8 locations in Virginia were enrolled in a 7-day CO-Synch+CIDR fixed-time artificial insemination (FTAI) protocol, followed by natural service for an approximately 70 d breeding season. All cows had ad libitum access to water, mineral blocks, and diet composed of hay, grass, sudangrass, sorghum grain and commodity pellets, depending on location. Approximately 30 days before calving (d -120, day of AI is d 0) cows were randomly assigned to one of two treatments: 1) Two doses (6 mL) of Multimin® 90 (Multimin USA, Ft. Collins, CO) at d -120 and d -10 (MIN; n = 494); or 2) a negative control with no trace mineral injection (CTRL; n = 492). Body condition score was recorded on d -10, 60 and 120. Estrus detection was recorded at d 0 using estrus detection patches. Pregnancy was diagnosed on d 60 and 120 by ultrasonography. Liver biopsies were collected on d -10 and 60 in a subgroup of 60 cows equally distributed between treatments and among all locations. No differences between treatments were detected in days post-partum (P = 0.66), BCS (P = 0.24) and estrus expression (P = 0.11). Mineral status of copper, manganese, selenium or zinc was similar between treatments before (P = 0.20) and after treatment (P = 0.17). No effect of treatment was detected in FTAI pregnancy rate (50.8%, P = 0.52) or overall pregnancy rate (89.6%, P = 0.64). Two doses of injectable trace mineral administrated approximately 30 days before calving and 10 days before FTAI did not improve pregnancy rate or overall mineral status in multiparous beef cows in Virginia.

42 Effect of injectable trace mineral supplementation on beef cows fertility and overall mineral status

Abstract The objective of this study was to evaluate the effect of an injectable multi-mineral complex supplementation on beef cows overall mineral status and fertility. This study involved 9 different locations in the state of Virginia and a total of 1,128 Angus crossbred multiparous cows. All cows had ad libitum access to forage and water, and received mineral supplementation in form of mineral blocks. All cows were enrolled in a 7-d CO-Synch + CIDR fixed-time AI (FTAI) protocol, followed by natural service for an approximately 70-d breeding season. At the initiation of the FTAI protocol (d 0) cows were stratified by days post-partum (82 ± 18 d) and randomly assigned to one of two treatments: 1) One dose of an injectable trace mineral containing zinc (60mg/ml), copper (15mg/ml), selenium (5mg/ml) and manganese (10mg/ml) on d 0 (n = 560; 1 ml/90 kg BW of Multimin® 90, Multimin USA, Ft. Collins, CO); or 2) a negative control with no trace mineral injection (n = 568). BCS was recorded at d 0, 61 and 121. Estrus detection was recorded at d 10 using an estrus detection patch. Pregnancy was diagnosed on days 71 and 131 by ultrasonography. Liver biopsies were collected on days 0 and 45 in a subgroup of 140 cows from both treatments and distributed among all locations. No differences between treatments were detected in days post-partum (P = 0.83), BCS (P ≥ 0.42) and estrus detection (P= 0.12), neither in mineral status of copper, manganese, selenium or zinc (P ≥ 0.28). No effect of treatment was detected (P ≥ 0.20) in AI pregnancy rate (57%) or overall pregnancy rate (93%). In conclusion, one dose of injectable trace mineral administrated 10 days before AI did not improve pregnancy rate or overall mineral status in multiparous beef cows enrolled in FTAI.

Effect of an injectable trace mineral supplement on the immune response of dairy calves

On a spring calving, pastoral dairy farm, the first 40 heifer calves born after calving mid-point (50% of the herd calved) were blood sampled within 24 h. Thirty were selected, using stratified randomisation to form two equal groups (treatment and control) with the same distribution of serum total protein, copper, selenium, zinc, and manganese concentrations, age and breed. From the remaining 10 calves, five were randomly selected into a sentinel group to assess field exposure to Salmonella spp.All calves received two injections of a killed vaccine containing Salmonella spp. antigens at 2 and 6 weeks of age. Concurrently, the treatment group were injected with 1 mL/50 kg trace mineral supplement (TMS) containing 40 mg zinc, 10 mg manganese, 5 mg selenium, 15 mg copper per mL. Sentinel animals received no injections. All animals were bled from 2 to 9 weeks for assay of immune function. At three and four weeks, white blood cells from TMS calves had an increased percentage of cells phagocytosing (effect size = 9.36 and 4.35) and increased number of bacteria ingested per cell (effect size = 0.93 and 1.52). No differences were detected in gamma interferon response (effect size <0.15) or Salmonella sp. antibody titres (effect size <0.20).

Effects of maternal supplementation with an injectable trace mineral on subsequent calf performance and inflammatory response1.

Newly weaned, commercial Angus steers [body weight (BW) = 204 ± 19 kg; n = 24; 12 steers from dams administered an injectable trace mineral (MM; Mulimin90) and 12 steers from control (CON) dams] were utilized to determine the effects of maternal supplementation with an injectable trace mineral on the inflammatory response of subsequent steers subjected to a lipopolysaccharide (LPS) challenge at the initiation of a 42-d receiving period. On day -2 steers were weaned, and the following day, shipped 354 km to the Beef Cattle and Sheep Field Laboratory in Urbana, IL. On day 0, steers were administered an intravenous LPS challenge. Body temperature and blood samples were collected from steers prior to LPS administration (0 h) and again at 0.5, 1, 2, 3, 4, 5, and 6 h. Blood samples were analyzed for trace mineral and cortisol at 0 and 2 h and glucose, insulin, LPS-binding protein (LBP), interleukin-1β (IL-1β), interleukin-6 (IL-6), haptoglobin, ceruloplasmin, and fibrinogen at 0, 0.5, 1, 2, 3, 4, 5, and 6 h. Calf BW was collected at trial initiation and subsequently every 14 d. Dry matter intake was collected daily and average daily gain (ADG) and feed efficiency were assessed. Initial plasma Zn tended (P = 0.06) to be greater for MM steers. However, there was no difference (P ≥ 0.31) in trace mineral status or serum cortisol at any other time. Total area under the curve (TAUC) for body temperature was lesser (P > 0.01) for MM steers. Basal LBP concentrations and TAUC for LBP tended (P ≤ 0.10) to be greater for MM steers. Peak concentration of IL-6 tended (P = 0.09) to be reached earlier for CON steers. However, there was no difference (P ≥ 0.11) in glucose, insulin, IL-6, ceruloplasmin, haptoglobin, and fibrinogen concentrations between treatments. Calf performance and feed efficiency did not differ (P ≥ 0.17) between treatments except ADG from day 28 to 42, which was greater (P = 0.03) for CON steers. Maternal supplementation with an injectable trace mineral tended to improve steer plasma Zn status at 0 h and tended to increase basal concentrations of LBP and overall LBP production when steers were administered an LPS challenge. Additionally, MM steers exhibited a more favorable change in body temperature following LPS administration. However, injectable trace mineral supplementation of dams during gestation had minimal to no effect on cytokine and acute-phase protein concentrations, as well as overall calf performance and efficiency during a 42-d receiving period.

Effects of injectable trace minerals on the immune response to Mannheimia haemolytica and Pasteurella multocida following vaccination of dairy calves with a commercial attenuated-live bacterin vaccine

ABSTRACT The objective was to evaluate the effects of an injectable trace mineral (ITM) supplement containing Zn, Mn, Se, and Cu on the humoral and cell mediated immune responses to vaccine antigens in dairy calves receiving an attenuated-live bacterin vaccine containing Mannheimia haemolytica and Pasteurella multocida. Thirty 3-mo-old dairy calves received 2 doses (21 d apart) of an attenuated-live M. haemolytica and P. multocida bacterin vaccine (Once PMH, Merck Animal Health, Summit, NJ), and a 5-way modified-live-virus vaccine (Express 5, Boehringer Ingelheim, Vetmedica, St. Joseph, MO). On the day of primary vaccination, animals were randomly assigned to 1 of the 2 treatment groups (n = 15 per group): ITM (ITM administration) or control (sterile saline injection). Treatments were administered concurrently with vaccinations. Blood samples were collected for determination of antibody titers against M. haemolytica and P. multocida and of antigen-induced proliferation and interferon-γ secretion by peripheral blood mononuclear cells. Injectable trace-mineral supplementation improves sperm motility and morphology of young beef bulls

ABSTRACT This experiment evaluated effects of supplemental s.c. trace-mineral injections on growth and breeding soundness of bull calves. Weaned bulls (n = 488; initial BW = 308 ± 45 kg, initial age = 203 ± 17 d) of 2 breeds (Angus and Charolais) and originating from 13 ranches in the Great Plains were transported to a common confinement facility and assigned randomly to 2 treatments: (1) s.c. injections of trace mineral (TM) containing 15 mg/mL Cu, 5 mg/mL Se, 10 mg/mL Mn, and 60 mg/mL Zn or (2) s.c. injections of physiological saline (control). Treatments were administered at arrival (d −2 or −1; 1 mL per 45 kg of BW) and on d 90 ± 1 (1 mL per 68 kg of BW). On d 0, bulls were stratified by treatment, breed, and ranch of origin and assigned randomly to 8 pens in which they were fed a growing diet for 225 d. The diet was formulated to promote a 1.5-kg ADG at a DMI of 2.6% of BW and to meet or exceed NRC (2000) requirements for Ca, Co, Cu, I, Mg, Mn, Na, P, K, Se, and Zn. Initial BW were measured and pretreatment blood plasma samples were collected on d −2 or −1. Breeding soundness examinations (BSE) were conducted and BW were measured at 10 and 12 mo of age (d 90 ± 1 and d 150 ± 1, respectively). Scrotal circumference was measured and semen samples were collected via electro-ejaculation. Motility and morphology of sperm were evaluated via light microscopy. Scrotal circumference, BW, and ADG did not differ (P ≥ 0.16) between treatments at any time. Proportions of control- and TM-treated bulls achieving minimal satisfactory BSE classifications did not differ at 10 mo of age (P = 0.98; 50 ± 3.8% for both TM and control) or at 12 mo of age (P = 0.43; 89 and 86 ± 2.2% for TM and control, respectively). Conversely, improved (P = 0.05) sperm motility was detected in TM-treated bulls compared with control-treated bulls at 12 mo of age; moreover, TM-treated bulls had greater (P ≤ 0.05) improvements in sperm morphology and motility between 10 and 12 mo of age than control-treated bulls. Among bulls that failed BSE at 10 mo of age, more TM-treated bulls tended (P = 0.10) to pass BSE at 12 mo of age than control-treated bulls (98 and 94 ± 1.6% for TM and control, respectively). Under the conditions of this experiment, sperm motility and morphology at 12 mo of age were improved in bulls treated with injectable TM at 7 and 10 mo of age compared with bulls treated with saline.

Effects of injectable trace minerals on humoral and cell-mediated immune responses to Bovine viral diarrhea virus, Bovine herpes virus 1 and Bovine respiratory syncytial virus following administration of a modified-live virus vaccine in dairy calves

Our objective was to evaluate the effect of an injectable trace mineral (ITM) supplement containing zinc, manganese, selenium, and copper on the humoral and cell mediated immune (CMI) responses to vaccine antigens in dairy calves receiving a modified-live viral (MLV) vaccine containing BVDV, BHV1, PI3V and BRSV. A total of 30 dairy calves (3.5 months of age) were administered a priming dose of the MLV vaccine containing BHV1, BVDV1 & 2, BRSV, PI3V, and an attenuated-live Mannheimia-Pasteurella bacterin subcutaneously (SQ). Calves were randomly assigned to 1 of 2 groups: (1) administration of ITM SQ (ITM, n=15) or (2) injection of sterile saline SQ (Control; n=15). Three weeks later, calves received a booster of the same vaccine combination SQ, and a second administration of ITM, or sterile saline, according to the treatment group. Blood samples were collected on days 0, 7, 14, 21, 28, 42, 56, and 90 post-vaccination for determination of antibody titer, viral recall antigen-induced IFN-γ production, and viral antigen-induced proliferation by peripheral blood mononuclear cells (PBMC). Administration of ITM concurrently with MLV vaccination resulted in higher antibody titers to BVDV1 on day 28 after priming vaccination compared to the control group (P=0.03). Calves treated with ITM showed an earlier enhancement in PBMC proliferation to BVDV1 following vaccination compared to the control group. Proliferation of PBMC after BVDV stimulation tended to be higher on day 14 after priming vaccination in calves treated with ITM than in the control group (P=0.08). Calves that received ITM showed higher PBMC proliferation to BRSV stimulation on day 7 after priming vaccination compared to the control group (P=0.01). Moreover, calves in the ITM group also had an enhanced production IFN-γ by PBMC after stimulation with BRSV on day 21 after priming vaccination compared to day 0 (P<0.01). In conclusion, administration of ITM concurrently with MLV vaccination in dairy calves resulted in increased antibody titer to BVDV1, and greater PBMC proliferation to BVDV1 and BRSV recall stimulation compared to the control group, suggesting that ITM might represent a promising tool to enhance the humoral and CMI responses to MLV vaccines in cattle.

Effects of injectable trace mineral supplementation in lactating dairy cows with elevated somatic cell counts

Objectives of this clinical trial were to evaluate the effects of injectable trace mineral supplementation (ITMS) on somatic cell count (SCC), linear score (LS), milk yield, milk fat and protein contents, subclinical mastitis cure, and incidence of clinical mastitis in cows with elevated SCC. Holstein cows from a commercial dairy farm in New York were evaluated for subclinical mastitis, defined as SCC ≥200×103 cells/mL on the test day preceding enrollment. Cows with a history of treatment for clinical mastitis in the current lactation and those pregnant for more than 150d were not eligible for enrollment. Cows fitting inclusion criteria were randomly allocated to 1 of 2 treatment groups. Cows assigned to ITMS (n=306) received 1 subcutaneous injection containing zinc (300mg), manganese (50mg), selenium (25mg), and copper (75mg) at enrollment (d 0). Control cows (CTRL; n=314) received 1 subcutaneous injection of sterile saline solution. Following treatment, visual assessment of milk was performed daily, and cows with abnormal milk (i.e., presence of flakes, clots, or serous milk) were diagnosed with clinical mastitis (CM). Chronic clinical mastitis was defined as cows with 3 or more cases of CM. Milk yield, milk fat and protein contents, SCC, and LS were evaluated once monthly. Additionally, randomly selected animals were sampled to test serum concentrations of selected minerals on d0 and 30 (n=30 cows/treatment). Treatment did not affect serum concentrations of calcium, magnesium, phosphorus, potassium, copper, iron, manganese, selenium, and zinc on d30. Injectable supplementation with trace minerals did not improve overall cure of subclinical mastitis (CTRL=42.8 vs. ITMS=46.5%), although a tendency was observed in cows with 3 or more lactations (CTRL=27.1 vs. ITMS=40.0%). Supplementation did not reduce treatment incidence of CM (CTRL=48.2 vs. ITMS=41.7%); however, it tended to reduce the proportion of cows diagnosed with chronic CM (CTRL=16.9 vs. ITMS=12.0%), particularly among first-lactation cows (CTRL=18.4 vs. ITMS=7.6%). Cure of subclinical mastitis was associated with higher serum concentrations of phosphorus and selenium on d30. Supplementing trace minerals to cows with elevated SCC had no effect on milk yield, milk fat and protein contents, SCC, and LS.

Effect of trace mineral supplementation on selected minerals, energy metabolites, oxidative stress, and immune parameters and its association with uterine diseases in dairy cattle

The objective of this study was to evaluate the relationship between selected minerals’ serum levels, energy metabolites, oxidative stress indicators, IL-8 and haptoglobin levels, and the potential for uterine diseases. Additionally, we investigated the effect of injectable trace mineral supplementation (ITMS) on metabolism, immune function, and animal health under field conditions involving a dairy herd with high milk production. The study was conducted in 1 dairy farm located near Ithaca, New York, with 270 multiparous cows were enrolled from October 3, 2012 until January 10, 2013. Cows were randomly allocated into 1 of 2 treatments groups: ITMS or control. Cows randomly assigned to the ITMS group received 2 injections of trace minerals at 230 and 260 d of gestation; each injection contained 300mg of Zn, 50mg of Mn, 25mg of Se, and 75mg of Cu. Retained placenta (RP) and metritis were diagnosed and treated by trained farm personnel. Clinical endometritis evaluation was performed by the investigators. Blood mineral levels, plasma nonesterified fatty acids and serum β-hydroxybutyrate concentrations, plasma IL-8 concentrations, serum haptoglobin concentration, and serum superoxidase dismutase and plasma glutathione peroxidase activities were measured at various time points before and after calving. Four groups of mixed general linear models were fitted to the data using MIXED procedure of SAS. Injectable trace mineral-supplemented cows had increased serum concentration of Cu, Se, and Zn. Conversely, ITMS did not affect energy metabolites or immune and oxidative stress parameters. Serum concentration of Ca, Cu, K, Mg, Mo, Ps, Pt, Se, and Zn varied according to days relative to parturition. Cows with RP had reduced serum concentrations of Ca, Mg, Mo, and Zn when compared with cows without RP. Cows affected with metritis had significantly lower serum concentrations of Ca, Mo, soluble P, total P, Se, and Zn than nonaffected cows. Serum concentration of Ca, Cu, Mo, and Zn were reduced in cows diagnosed with endometritis in comparison to nonaffected ones.

Effect of an injectable trace mineral supplement containing selenium, copper, zinc, and manganese on immunity, health, and growth of dairy calves

The objective of this study was to evaluate the effect of 2 subcutaneous injections of a multimineral preparation, each containing 60mg of zinc, 10mg of manganese, 5mg of selenium, and 15mg of copper at 3 and 30d after birth on immunity, health, and growth of dairy calves during the preweaning period. The study was conducted in upstate New York in 2 commercial dairy farms. A total of 790 Holstein heifer calves were randomly allocated at birth into 1 of 2 treatments: trace mineral supplement (TMS) treated or control. Blood samples were collected at 3, 14, and 35d after birth to evaluate glutathione peroxidase (GPx) activity, superoxide dismutase (SOD) activity, haptoglobin, and neutrophil and monocyte function. Incidence of diseases and average daily gain was evaluated in the first 50d of life. At 14d of life, TMS-treated calves had increased neutrophil activity compared with control calves. Moreover, TMS-treated calves had greater GPx activity on d 14 after birth than control calves. The TMS treatment reduced the incidence of diarrhea (TMS=41.7% vs. control=49.7%) and combined incidence of pneumonia or otitis or both (TMS=41.7% vs. control=49.1%). Additionally, GPx was greater for calves diagnosed with otitis at d 35 after birth. However, calves diagnosed with pneumonia had decreased GPx activity at d 35 after birth. Serum SOD and haptoglobin concentrations were not affected by treatment or disease. Moreover, no effects were observed on average daily gain and survivability between TMS-treated and control calves during the preweaning period. Supplementation with trace minerals at 3 and 30d of life increased neutrophil function and GPx activity and reduced the incidence of health disorders.

Effect of an injectable trace mineral supplement containing selenium, copper, zinc, and manganese on the health and production of lactating Holstein cows

The objective of this study was to evaluate the effect of a subcutaneous injection of a multimineral preparation containing 300mg of zinc, 50mg of manganese, 25mg of selenium, and 75mg of copper at 230 and 260days of gestation and 35days postpartum, on the health, milk production and reproductive performance of lactating Holstein cows. A randomized field trial was conducted on three large commercial dairy farms located near Ithaca, New York, USA, with 1416 cows enrolled. All cows were housed and offered a total mixed ration consisting of approximately 55% forage and 45% concentrate on a dry matter basis of the diet, which supplied 2–6 times the NRC requirements for the supplemented elements. Dry cows and pregnant heifers were blocked by parity and randomly allocated to one of two treatments: Trace mineral supplemented (TMS) or control.For multiparous cows, subcutaneous TMS significantly decreased linear somatic cell count scores (normalized data) as compared to control cows. The incidence of subclinical mastitis for TMS and control cows was 10.4% and 8.0%, respectively (P=0.005). The main effect of treatment on clinical mastitis was not significant but the interaction of treatment and parity was significant. For primiparous cows, the incidence of clinical mastitis was 11.8% and 15.6% for control and TMS cows, respectively (P=0.33); for multiparous cows, the incidence of clinical mastitis for control and TMS cows was 25.4% and 19.7%, respectively (P=0.03). Additionally, control cows had increased odds of stillbirth and endometritis (odds ratios 1.69 and 1.30, respectively). The incidence of endometritis was 34.2% and 28.6% for control and TMS cows, respectively (P=0.039) but treatment had no effect on reproductive performance, milk production or other health traits. Further research is required to confirm these findings and to establish whether the response seen in this study was related to the supplementation of a particular mineral.
