Change in inorganic phosphate physical state can regulate transcription

ABSTRACTInorganic phosphate (Pi), a ubiquitous metabolite, is involved in all major biochemical pathways. We demonstrate that, in vitro, MgHPO4 (the intracellular Pi form) at physiological concentrations can exist in a metastable supersaturated dissolved state or as a precipitate. We have shown that in solution, MgHPO4 strongly stimulates exonuclease nascent transcript cleavage by RNA polymerase. We report here that MgHPO4 precipitate selectively and efficiently inhibits transcription initiation in vitro. In view of the MgHPO4 solubility and in vitro sensitivity of RNA synthesis to MgHPO4 precipitate, at physiological concentrations, MgHPO4 should cause a 50–98% inhibition of cellular RNA synthesis, thus exerting a strong regulatory action. The effects of Pi on transcription in vivo will, therefore, reflect the physical state of intracellular Pi.

Effect of actinomycin D on simian rotavirus (SA11) replication in cell culture.

Rotaviruses are the major cause of viral diarrhea in humans and animals. Actinomycin D (Act D) is an antibiotic that intercalates DNA and therefore inhibits DNA-dependent transcription. The current study was carried out to assess the influence of Act D on the replication of simian rotavirus (SA11) in cell culture. Virus-infected MA-104 cell cultures were studied in the presence of Act D at concentrations of 1.25 and 2.5 microg/ml. Treatment of rotavirus-infected cells with 2.5 microg/ml Act D 48 h post-infection reduced the cytoplasmic metachromasia after staining with acridine orange by 25%. Viral RNA labeled with 3H-uridine in the presence of the drug was separated by polyacrylamide gel electrophoresis. Viral RNA replication was not affected by Act D, but increased 3H-uridine uptake was demonstrable by infected cells in the presence of the drug. This possibly was due to the inhibition of cellular RNA synthesis by Act D, which thus enhances incorporation of the radionuclide into the viral RNA. Act D reduced the number of infected cells presenting virus-specific fluorescence 48 h post-infection by more than 50%. These data suggest that Act D may have complexed with viral RNA and prevented newly synthesized mRNA from being translated, but may not have prevented early replication.

Nitric oxide enhancement of fludarabine cytotoxicity for B-CLL lymphocytes.

Fludarabine is active but not curative in the treatment of chronic lymphocytic leukemia (B-CLL). Nitric oxide (NO) supplied from exogenous, NO-donating pro-drugs can also induce apoptosis and death of acute leukemia cells. This study investigated combinations of fludarabine with NO-donating pro-drugs for their cytotoxicity against freshly isolated B-CLL lymphocytes following a 72 h exposure in vitro. The median IC(50)for fludarabine was 2.2 microM (n = 85). The nitric oxide donors DETA-NO, PAPA-NO, and MAHMA-NO were also cytotoxic, and their effects were inversely related to rates of NO release. Neither DETA-NO depleted of NO nor DETA itself was effective, indicating that NO was required for cytotoxicity. Drug interactions were evaluated by a modified combination index method. Synergy was observed in combinations of fludarabine or nelarabine (506U78) with DETA-NO in 52% and 88% of samples, respectively. Interestingly, the combination of fludarabine and DETA-NO was more cytotoxic in B-CLL cells less sensitive to fludarabine. DETA-NO did not enhance the activity of other DNA anti-metabolites, topoisomerase I and II inhibitors, or alkylating agents. Finally, the anti-leukemic activity of fludarabine alone or in combination with DETA-NO was found to correlate with inhibition of cellular RNA synthesis. These results indicate that NO donors could enhance fludarabine therapy for B-CLL.

Specific interaction of heterogeneous nuclear ribonucleoprotein particle U with the leader RNA sequence of vesicular stomatitis virus.

The 3' ends of the genome and antigenome RNA of vesicular stomatitis virus (VSV) serve as the promoter sites for the RNA-dependent RNA polymerase in the initiation of transcription and replication, respectively. The leader RNA, the first transcript synthesized during the RNA synthetic step, contains sequences to initiate encapsidation with the nucleocapsid protein, which is a prerequisite for replication. It also plays a role in the inhibition of cellular RNA synthesis. To search for a specific cellular factor(s) which may interact with the leader RNA sequences and regulate these processes, we used a gel mobility shift assay to identify such a protein(s). By using nuclear extract, it was found that in addition to the previously reported La protein, a 120-kDa nuclear protein specifically interacts with the leader RNA. Biochemical and immunological studies identified the 120-kDa protein as heterogeneous nuclear ribonucleoprotein particle U (hnRNP U), which is involved in pre-mRNA processing. We also demonstrate that hnRNP U is associated with the leader RNA in the nuclei of VSV-infected cells and also packaged within the purified virions. By double immunofluorescence labeling and confocal microscopy, hnRNP U appears to colocalize with the virus in the cytoplasm of infected cells. These results strongly suggest that hnRNP U plays an important role in the life cycle of VSV.

Studies on the mechanism of the selenite-induced decrease in cell attachment: effect of selenite on the levels of fibronectin receptor (alpha5beta1 integrin) mRNAs.

We previously reported that exposure of HeLa cells to selenite for 2 h results in a decrease in their ability to attach to fibronectin (Yan and Frenkel, Cancer Res. 52, 5803-5807 [1992]), as well as a decrease in the level of fibronectin receptor (alpha5beta1 integrin) at the cell surface (Yan and Frenkel, Biol. Trace Element Res. 46, 79-89 [1994]). We have now found that after exposure to selenite, there was a decrease in the total cellular content of the receptor protein, as well as in the level of the mRNAs for both of the subunits. Exposure of cells to actinomycin D (an inhibitor of RNA synthesis) also resulted in a decrease in the level of these mRNAs, suggesting that the effect of selenite is the result of its known inhibitory effect on RNA synthesis (Frenkel, Toxicol. Lett. 25, 219-223 [1985]). Exposure of cells to actinomycin D for 2 h also resulted in a decrease in the ability of cells to attach to fibronectin. Furthermore, both selenite and actinomycin D caused a decrease in integrin mRNA levels and in cell attachment to fibronectin only when high-density cells were exposed to the agents. In contrast, when low-density cells were exposed,neither agent had any detectable effect on mRNA levels or on cell attachment. These results have suggested the following scheme for the mechanism of the inhibition of cell attachment by selenite: After exposure to selenite for 2 h, there is a significant inhibition of cellular RNA synthesis, which results in a general decrease in the cellular level of those mRNAs with relatively short half-lives, including in particular those of the fibronectin receptor. This leads to a decrease in the intracellular level of the receptor protein and, consequently, in its level at the cell surface, which in turn causes a decrease in the rate of cell attachment to fibronectin.

Involvement of cellular sulfhydryl compounds in the inhibition of rna synthesis by selenite

Selenite has been shown previously to inhibit cellular RNA synthesis. Based upon our previous observation that selenite inhibits purified RNA polymerase only in the presence of a sulfhydryl compound (Frenkel et al., Mol Pharmacol 31: 112–116, 1987), we hypothesized that the inhibition of cellular RNA synthesis by selenite involves endogenous sulfhydryl compounds. We found that depletion of cells of endogenous sulfhydryl compounds, by exposure to diethylmaleate (DEM), virtually eliminated the inhibitory effect of a 1-hr exposure of cells to selenite. This inhibition was restored to normal or higher levels when the selenite was reacted with glutathione or cysteamine prior to addition to the DEM-treated cells. RNA synthesis in DEM-treated cells was inhibited after a 4-hr exposure to higher concentrations of selenite. In contrast to the effect of DEM, specific depletion of the cells of glutathione, by exposure to buthionine sulfoximine, had no effect on the inhibition of RNA synthesis by selenite. These results demonstrate the involvement of endogenous cellular sulfhydryl compounds in the inhibition of RNA synthesis by selenite, but indicate that glutathione, in particular, is not involved in this inhibition.

Rapid Communication: Mithramycin Selectively Inhibits Transcription Of G-C Containing DNA

Mithramycin induces a reversible inhibition of cellular RNA synthesis without affecting DNA synthesis. The authors have shown this drug induces myeloid differentiation of HL-60 promyelocytic leukemia cells and is an effective agent in certain patients with chronic granulocytic leukemia. In order to investigate the mechanism by which this drug inhibits RNA synthesis we have compared the effect of mithramycin on RNA synthesis by whole cells, isolated nuclei, and RNA synthesis by isolated E. coli RNA polymerase and eukaryotic RNA polymerase II. Exposure of HL-60 cells to mithramycin at concentrations of 4.6 X 10(-7) m or higher for 48 hours causes an almost immediate inhibition of RNA synthesis (up to 85% at 4 hours) with only modest cytotoxicity at these concentrations. Endogenous RNA synthesis by isolated nuclei can be inhibited by mithramycin only at high concentrations (greater than 10(-5) m), suggesting that mithramycin primarily may inhibit initiation, rather than elongation. Mithramycin inhibits in vitro transcription of salmon sperm DNA by E. coli RNA polymerase at DNA:drug ratios similar to those required for RNA synthesis inhibition in whole cells. Similar DNA binding studies with synthetic oligonucleotides demonstrate that mithramycin is a potent inhibitor of transcription of Poly dG.dC by E. coli RNA polymerase but has no effect on transcription of Poly dA.dT. The rapid inhibition of whole cell and isolated RNA polymerase transcription, and the relative insensitivity of isolated nuclei, suggest mithramycin may interact with specific DNA sequences in order to inhibit the initiation of RNA synthesis in intact cells.

The transcription inhibitor α-amanitin which does not easily pass through the membrane of normal cells, shows a higher inhibition of cellular RNA synthesis in encephalomyocarditis (EMC) virus-infected HeLa cells as compared with uninfected cells. This selective inhibition is observed within minutes of virus addition to the culture medium. It depends on the multiplicity of infection used and the concentration of α-amanitin present. These results indicate that, very early during infection, α-amanitin is able to pass the cell membrane and get into the nucleus, where it blocks RNA synthesis. No effect is observed in the transport of [3H]uridine into the cells, indicating that the increased inhibition by α-amanitin affects RNA synthesis itself. This phenomenon occurs with different animal viruses irrespective of whether they possess a lipid envelope or not. Finally, no inhibition by NH4Cl or chloroquine is observed in the permeabilization induced by encephalomyocarditis virus, indicating that it is not a low-pH requiring process. However, it does require a metabolically active cell, since NaN3 plus 2′-deoxy-glucose blocked this process.

Persistent infection with mouse hepatitis virus 3 in mouse lymphoid cell lines

The sensitivity of mice to mouse hepatitis virus 3 (MHV3) varies according to strain, age, and immune status of the animals. In semisusceptible strains, mice surviving the acute phase of infection develop a chronic disease characterized by the occurrence of paralysis, virus persistence, and immunodeficiency. Persistent MHV3 infections established in vitro in YAC and RDM -4 mouse lymphoid cell lines were characterized by virus production, presence of cytoplasmic viral antigens, and cell lysis. The occurrence of cell "crisis" in YAC cells was manifested by a sharp increase in cell lysis and in the number of fluorescent cells and, concomitantly, by a marked decrease in virus titers. A relationship was observed among the percentage of fluorescent cells, cell lysis, and virus yield and was modulated by renewal of culture media, change in temperature, or inhibition of cellular RNA synthesis. Cell cloning and antibody treatment experiments indicated that viral transmission was performed by viral infection of newly permissive cells produced by the division of uninfected cells in the culture and not by transmission of viral information by infected dividing cells. The biological and biochemical properties of MHV3 variants derived from persistently infected YAC lymphoid cells were characterized. Thermosensitivity and thermolability of cloned viruses originating from persistently infected YAC cells, as well as parent virus suspensions, were studied. A similar heterogeneity was observed when YAC-derived cloned substrains (YAC-MHV3) were compared with parent-derived cloned viruses, indicating that no selection of temperature-sensitive mutants was induced in persistently infected YAC cells. However, the capacity of MHV3 to induce a lethal acute disease when injected into susceptible mice was lost very rapidly. The absence of pathogenicity was related to the induction of a subclinical infection which elicited defense mechanisms. These data suggest, therefore, that MHV3 replication in lymphoid cell lines leads to induction or selection of variants which maintain pathogenicity in vitro but display reduced pathogenic effects in vivo.

Inhibition of host cellular ribonucleic acid synthesis by glycoprotein of mumps virus

After infection with mumps virus, cellular ribonucleic acid synthesis of a murine lymphoma cell line, EL4, was appreciably depressed. The inactivation of viral infectivity by ultraviolet irradiation or the treatment of cells with mouse interferon did not abolish the inhibiting effect, suggesting that virus replication is not required for the depressed RNA synthesis. Envelope glycoproteins isolated from disrupted mumps virus caused inhibition of cellular RNA synthesis. The addition of low concentrations of specific antibody enhanced the inhibitory effect, probably through the formation of aggregates of glyco-proteins. On the contrary, the glycoproteins showed no effect on RNA synthesis in the presence of cytochalasin D.

Use of UV irradiation to identify the genetic information of vesicular stomatitis virus responsible for shutting off cellular RNA synthesis.

UV irradiation of infectious vesicular stomatitis virus was employed to study the relationship between the expression of certain viral gene functions and viral inhibition of RNA synthesis in mouse myeloma (MPC-11) cells. Viral infectivity, protein synthesis, and viral mRNA synthesis were all highly susceptible to inactivation by UV radiation; however, low levels of viral transcriptase activity were detected in vitro in virus preparations subjected to large doses of UV radiation. In sharp contrast, the capacity of vesicular stomatitis virus to shut off cellular transcription was quite resistant to UV radiation. The data presented here indicate that viral transcription is essential to inhibit host RNA metabolism, even though synthesis of viral polypeptides in the inhibited cells could not be detected. At those levels of UV radiation that inactivated all viral gene functions, except viral inhibition of cellular RNA synthesis, the only viral product detected was non-adenylated, low-molecular-weight RNA species.

Transcription of vesicular stomatitis virus is required to shut off cellular RNA synthesis

RNA synthesis by mouse myeloma (MPC-11) cells was rapidly and progressively shut off by infection with vesicular stomatitis virus temperature-sensitive (ts) mutants permissive for transcription. In sharp contrast, mutants or defective vesicular stomatitis virions restricted in transcription were incapable of causing progressive inhibition of cellular RNA synthesis even at massive multiplicities of infection. A viral product synthesized 30 to 60 min after permissive infection with tsG114(I) appeared to be essential for prolonged inhibition of RNA synthesis in cells switched up to restrictive temperature.

Inhibition of RNA synthesis in mouse myeloma cells infected with vesicular stomatitis virus

Infection of mouse myeloma cells (MPC-11) with vesicular stomatitis (VS) virus resulted in rapid and marked reduction in cellular RNA synthesis considerably before cell viability was compromised. Mouse myeloma cells responded maximally to viral infection at a multiplicity of 1 and were considerably more se;sitive to shut-off of RNA synthesis than were mouse L cells or BHK-21 cells. This inhibition of cellular RNA synthesis was shown not to be caused by differential membrane permeability of infected and uninfected MPC-11 cells to [3H]uridine, nor was it due to greater degradation of previously synthesized RNA. VS viral infection appeared not to impede transport of newly synthesized nuclear RNA to the cytoplasm; moreover, infected cells accumulated polyadenylated mRNA at the same rate as did uninfected cells. Polyacrylamide gel electrophoresis of newly synthesized nuclear RNA demonstrated that the polydisperse nature and size distribution were not affected by VS viral infection. Isolated nuclei of infected MPC-11 cells also inhibited greatly impaired capacity to synthesize RNA despite the absence of cytoplasmic factors. Infected-cell cytosol did not inhibit transcription by uninfected-cell nuclei, nor did uninfected-cell cytosol reverse viral inhibition of nuclear transcription. Studies with alpha-amanitin revealed that VS viral infection inhibited the activity of polymerases I, II, and III, but only polymerase II was affected progressively throughout infection and to a much greater extent. These data suggest that, even at low multiplicities of infection, VS virus rapidly shuts off cellular RNA synthesis at the level of nuclear transcription.

Drug-containing lipid vesicles render drug-resistant cell sensitive to actinomycin D.

DEVELOPMENT of cellular resistance to actinomycin D (AD) has been reported in tumour cell populations in vivo1,2 and in vitro3–6. Such cells have a lower capacity than normal to incorporate AD3,6–8, prompting suggestions that the AD-resistant phenotype is determined by changes in plasma membrane function that reduce cellular permeability to this drug. Support for this proposed mechanism of drug resistance has been provided by experiments showing that treatment of AD-resistant cells with membrane-active detergents, to increase plasma membrane permeability, enhances AD uptake by resistant cells and reduces the concentration of AD required to inhibit cell growth8,9. This approach, while instructive, has the disadvantage that the detergent causes significant alteration in cell viability and also impairs cell growth8. We now report a new method for enhancing drug uptake by permeability restriction, drug-resistant cells which uses lipid vesicles as carrier agents to introduce AD directly into cells. As reported elsewhere10,11, negatively-charged lipid vesicles composed of lipids or lipid mixtures that are “fluid” at 37 °C are incorporated into cultured cells by fusion with the cellular plasma membrane. Entrapment of drugs inside such vesicles and subsequent fusion of the vesicles with cells offers a potential method for bypassing the plasma membrane transport barrier to introduce drugs directly into the intracellular compartment. Since vesicles are not cytotoxic10–12, their use as drug carriers allows direct evaluation of the cellular response to the introduced drug, free from the complications8,9,13 associated with the use of detergents. The results presented here indicate that uptake of vesicles containing AD by AD-resistant cells produces inhibition of cellular RNA synthesis and cell growth at AD concentrations that have no effect on cells when added as free drug to the culture medium.

Effects of rifampicin, streptolydigin and actinomycin D on the replication of Col E1 plasmid DNA in Escherichia coli

We recently reported (Clewell et al., 1972) on an inhibitory effect of rifampicin on Col E1 plasmid replication. The present study represents a further characterization of this phenomenon as well as a study of the effects of two other known inhibitors of RNA synthesis, Streptolydigin and actinomycin D. During treatment of cells with chloramphenicol the colicinogenic factor E 1 (Col E1) continues to replicate for more than ton hours. During this time 4 to 5 S RNA is also synthesized. When varying concentrations of rifampicin were included during chloramphenicol treatment, inhibition of plasmid DNA synthesis correlated very closely with inhibition of cellular RNA synthesis. Similar experiments testing the effects of Streptolydigin and actinomycin D (during chloramphenicol treatment) showed that cellular RNA synthesis was at least 100 times more sensitive to these drugs than was plasmid DNA synthesis. When actively growing cells (i.e. cells not treated with chloramphenicol) were treated with a high concentration of rifampicin (250 μg/ml), chromosomal DNA synthesis continued to an extent representing about a 50% increase in DNA, while plasmid DNA synthesis appeared to stop abruptly.

The synthesis of surface coat materials in normal and transformed cells.

The rate of synthesis and thickness of the surface coat material in a range of virus-transformed and chemically-transformed cell lines were measured by ellipsometry. Cell lines transformed by polyoma virus, SV 40 virus, Rous sarcoma virus and murine sarcoma virus had a significantly thicker coat than the normal parent cells. An increase in the thickness of the cell coat was not a consistent feature of the transformed cell state since this change was not detected in cell lines transformed by methylcholanthrene. The rate of synthesis of the surface coat was significantly faster in transformed cells than in normal cells. Coat synthesis in normal and transformed cells was inhibited rapidly by treatment with cycloheximide. Inhibition of cellular RNA synthesis by actinomycin D produced rapid inhibition of coat synthesis in normal and chemically-transformed cell, but in certain virus-transformed cell lines coat synthesis continued for up to 2 1 2 h. The significance of these changes in the pattern of coat synthesis in transformed cells in relation to their altered surface properties is discussed.

Light and electron microscopic autoradiography of coxsackievirus A 1 infected muscles: viral RNA synthesis and inhibition of host cell RNA synthesis.

Light and electron microscopic autoradiography using tritiated uridine as radioactive precursor, shows that the viral RNA synthesis in coxsackievirus infected muscles of newborn mice takes place to a very small amount in and around the nucleus during the first 8 to 10 hours p. i. The main part of viral RNA is synthesized in cytoplasmic vacuoles around the 10th hour p. i., weak synthetic activity is found until complete cell destruction at about 15 hours p. i. The time course of viral induced inhibition of cellular RNA synthesis is demonstrated by light microscopic autoradiography and shows good agreement with the known biochemical data obtained with picornavirus infected cultured cells.

Comparison of interferon induction by active and inactive Semliki Forest virus.

SUMMARY Semliki Forest virus, inactivated by incubation at 37°, induced considerably more interferon than did the live virus. Infection by live virus produced an earlier termination of interferon synthesis (at 8 hr) which correlated with a profound inhibition of cellular RNA synthesis. Heat-inactivated Semliki Forest virus did not inhibit cellular RNA synthesis until much later and permitted continuation of interferon synthesis for an additional 4 hr, thereby increasing the yield of interferon by about four- to sixfold.

Macromolecular Synthesis in Cells infected with Reovirus Type 2 and the Effect of Ara-C

WE have shown that it is possible to demonstrate in RA cells (an established human amnion culture) and in HeLa cells infected with reovirus type 2 the synthesis of double stranded viral RNA as early as 4–5 h after infection1,2. Although we did not observe any inhibition of cellular RNA synthesis, it was possible to distinguish viral RNA by its relative resistance to treatment with ribonuclease. This communication reports the macromolecular synthesis of DNA and protein in RA cells undergoing a single cycle infection with reovirus.
