Effects of Exogenous Oral Infusion of Volatile Fatty Acids on Ileal Microbiome Profiling and Epithelial Health in Goats

The role of volatile fatty acids (VFAs) in ruminal fermentation is well studied, but their effects on the ileal microbiome and epithelial health remain less explored. In this study, we investigated the impact of the exogenous oral infusion of three VFAs, sodium acetate (NaAc), propionate (NaPr), and butyrate (NaBu), on the VFA concentrations in ileal contents, as well as ileal microbiome profiling and epithelial health parameters (inflammatory cytokine and tight junctions) in goats. The data demonstrated that the oral infusion of three VFAs can enhance VFA production by increasing the proportions of each individual VFA and the total VFAs. Then, the microbiome revealed distinct microbial succession patterns and altered microbial diversities in response to the oral infusion of VFA treatments. Moreover, the oral infusion of each VFA had unique effects on the ileal bacterial community, with potential implications for epithelial health. Notably, the oral infusion of VFAs demonstrated potential anti-inflammatory effects, as evidenced by reduced IL-6 levels in the NaPr group and increased IL-10 levels in the NaAc group. Notably, the oral infusion of VFAs did not directly impact the tight junction concentrations, such as Claudin1, Occludin, and ZO-1. Lastly, the correlation analyses identified specific relationships between the ileal bacteria and epithelial health parameters, and Prevotella was positively correlated with IL-6 and IL-1β, while Bifidobacterium was positively correlated with IL-10. These results highlighted the crosstalk between VFAs, the ileal microbiome, and the health of the small intestine. Our findings had significant implications for optimizing ruminant nutrition, enhancing epithelial health, and promoting sustainable livestock production practices.

Rumen and milk odd- and branched-chain fatty acid proportions are minimally influenced by ruminal volatile fatty acid infusions

The objective of this study was to determine if ruminally infusing volatile fatty acid (VFA) increased concentration of their homologous odd- and branched-chain fatty acid (OBCFA) in rumen contents and milk. The influence of VFA on dry matter intake (DMI), blood metabolites, and blood insulin was also evaluated. Four mid-lactation cows were assigned to a 4×4 Latin square design with 48-h periods. Infusion treatments were acetate (AC), propionate (PR), isovalerate (IV), and anteisovalerate (AIV). Infusions began (time = 0) 5.5h before feeding at 17.4mmol of VFA/min and were terminated at 18h. Infusions rates were well above physiological levels for IV and AIV. Surprisingly, the greatest differences in rumen OBCFA were increases in rumen liquid iso C15:0 and nonbranched C17:0 for AIV. In addition, infusing AIV increased anteiso C15:0 and anteiso C17:0 in rumen solid contents. Infusing IV increased iso C15:0 in both rumen solids and milk. Propionate increased milk C15:0 and C17:0. Both gluconeogenic compounds, PR and AIV, had similar proportions of milk C15:0, which was greater than that obtained with AC and IV. Rumen and blood VFA were as expected, with increased concentrations of the VFA present in the infusate. At 23h, and consistently throughout infusions, DMI was similar for AC compared with PR and for AIV compared with IV. Both IV and AIV decreased DMI and energy balance; however, only IV increased plasma nonesterified fatty acids (121, 78, 172, and 102mM for AC, AIV, IV, and PR), increased β-hydroxybutyrate (10.8, 5.9, 51.9, 5.4mg/dL for AC, AIV, IV, and PR), and reduced plasma glucose (56.3, 59.1, 31.9, and 64.3mg/dL for AC, AIV, IV, and PR). Rumen and milk OBCFA responses were minimal following infusion of large amounts of IV and AIV, suggesting limited use of IV, and AIV for de novo OBCFA synthesis, either pre- or postabsorption. Minor increases in milk odd-chain fatty acids following large doses of ruminal PR support the presence of postabsorptive synthesis of these milk odd-chain fatty acids.

Effects of Volatile Fatty Acids on IGF-I, IGFBP-3, GH, Insulin and Glucagon in Plasma, and IGF-I and IGFBP-3 in Different Tissues of Growing Sheep Nourished by Total Intragastric Infusions

Twelve SuffolkxSmall-tail-Han male sheep (body weight 21-26 kg), aged four months, were used to study the effects of volatile fatty acids (VFA) on IGF-I (insulin-like growth factor-I), IGFBP-3 (insulin-like growth factor binding protein-3), GH (growth hormone), insulin and glucagon in plasma, and IGF-I and IGFBP-3 in different tissues. The sheep were randomly divided into four groups with 3 sheep in each group. The sheep were sustained by total intragastric infusions and four levels of mixed VFA (the molar proportion of acetic acid, propionic acid and butyric acid was 65:25:10), which supplied 333, 378, 423 and 468 KJ energy/kg W 0.75 /d, were infused into the rumen as experimental Treatments I, II, III and IV, respectively. The experiment lasted 12 days, of which the first 8 days were for pretreatment and the last 4 days for collection of samples. At the end of the experiment, blood samples were taken and then the sheep were slaughtered and tissue samples from the rumen ventral sac, rumen dorsal sac, liver, duodenum and Longissimus dorsi muscle were obtained. IGF-I, IGFBP-3, GH, insulin and glucagon in plasma and IGF-I and IGFBP-3 in different tissues were analysed. Results showed that the concentration of IGF-I, IGFBP-3, GH, insulin or glucagon in plasma and the content of IGF-I and IGFBP-3 in the rumen dorsal sac, rumen ventral sac, liver or Longissimus dorsi muscle were increased with VFA infusion level (p 0.05). It was concluded that IGF-I, IGFBP-3, GH, insulin and glucagon in plasma and IGF-I and IGFBP-3 in rumen dorsal sac, rumen ventral sac, liver and Longissimus dorsi muscle were increased significantly with increasing level of ruminal infusion of mixed VFA.

Effects of monensin on volatile fatty acid metabolism in periparturient dairy cows using compartmental analysis

Eight multiparous periparturient Holstein cows fitted with ruminal cannulas were used in a split plot design to evaluate effects of monensin on ruminal volatile fatty acid (VFA) metabolism. Diets were supplemented with 300 mg/day of monensin, or no monensin, both prepartum and postpartum. Isotopic tracers, Na-1- 13C-acetate (Ac), Na-1- 13C-propionate (Pr), or Na-1- 13C-butyrate (Bu) were used as markers to describe VFA kinetics in the rumen. The Windows version of SAAM software (WinSAAM) was used to develop a steady state VFA model. A 9-compartment model was adequate to comprehensively describe ruminal VFA metabolism. The main VFA compartments consisted of Ac, Pr and Bu. The model estimated lower Bu and Ac interconversions with monensin, postpartum (Bu to Ac; 0.14 versus 0.12; P=0.04, and Ac to Bu; 0.32 versus 0.25; P=0.11) compared to when measured prepartum. Results demonstrate that dilution studies employing stable isotopes of VFA can be used to provide information on VFA metabolism of the periparturient dairy cow. A time frame of 320 min of labeled VFA infusion employing a single injection allows accurate quantification of VFA metabolism in the rumen. Compartmental kinetic analysis of major VFA in the rumen indicate that monensin reduced about 0.125 the portion of the Ac that contributes to Bu by reducing movements of Bu originated carbons to the Ac pool. Monensin may affect certain biochemical pathways of interconversion of Bu and Ac in the rumen. Propionate kinetic data suggests that Pr behaves as a single pool in the rumen. Monensin did not affect Pr production in the rumen suggesting that monensin improves the metabolic status of the transition cow in a way other than increasing Pr production in the rumen.

The relationship between the volatile fatty acids supply and the nitrogen retention in growing sheep nourished by total intragastric infusions

Twelve four-month old Suffolk × Small-tail- Han male sheep (live weight 21–26 kg), fitted with rumen and abomasum fistulas and nourished by total intragastric infusions, were used to study the relationship between the volatile fatty acids (VFA) supply and the nitrogen (N) retention in sheep. The animals were randomly divided into four groups and four levels of mixed VFA energy (the molar proportion of acetic acid, propionic acid and butyric acid was 65:25:10), i.e. 333, 378, 423 and 468 kJ kg −1 W 0.75 d −1, were infused into the rumen, as treatments I, II, III and IV, respectively. The results showed that the N retention was significantly increased ( P < 0.05) with the VFA infusion level. Significant regression relationship was found between the VFA supply ( x, g d −1) and the N retention ( y, mg d −1): y = 2.75 x − 403, r 2 = 0.86, n = 12, P < 0.01. It was concluded that to get efficient utilization of dietary N and high N retention in sheep, it is necessary to supply enough dietary energy.

Alterações morfológicas induzidas por butirato, propionato e lactato sobre a mucosa ruminal e epiderme de bezerros: II. Aspectos ultra-estruturais

Avaliou-se o efeito de ácidos graxos voláteis (AGV) sobre a integridade do epitélio no rúmen, no plano nasolabial, na epicera e no perioplum traseiro e dianteiro de bezerros e validou-se a feitura de biópsias tegumentares como indicadores de alterações morfológicas da mucosa ruminal. Dezessete bezerros, com sonda no rúmen, receberam infusões intra-ruminais de AGV ou salina, durante 37 dias. Aos 89 dias de vida, após o abate, foram colhidas amostras dos tecidos. Os AGV aumentaram a área de epitélio total e a área de células metabolicamente ativas no epitélio ruminal, embora o butirato não tenha induzido ao desenvolvimento papilar. A área de epitélio não queratinizado no plano nasolabial foi reduzida pela infusão de AGV. Butirato e lactato foram mais indutores de alterações patológicas no epitélio ruminal. Não foram observadas lesões histológicas nos epitélios do plano nasolabial, da epicera e do perioplum, mostrando que essas são conseqüências do efeito direto dos AGV sobre o epitélio ruminal. Os efeitos indireto e direto dos AGV sobre a morfologia dos tecidos epiteliais queratinizados não foram iguais. Biópsias tegumentares podem ter utilidade como indicadores de alterações morfológicas da mucosa ruminal.

Alterações morfológicas induzidas por butirato, propionato e lactato sobre a mucosa ruminal e a epiderme de bezerros: I Aspectos histológicos

Dezessete bezerros foram utilizados para avaliar o efeito de ácidos graxos voláteis (AGV) sobre a morfologia ruminal, a epiderme do plano nasolabial, a epicera e o perioplum, e para validar a execução de biópsias tegumentares como indicadores de alterações da mucosa ruminal. Os animais receberam infusões intra-ruminal de butirato, propionato, lactato ou salina (controle) durante 37 dias. A insulina sorológica foi dosada no 22º dia experimental nos tempos de 0, 90, 180 e 360 minutos em relação à infusão diária da manhã. No 89º dia de vida, após o abate, foram coletados fragmentos ruminais e epidérmicos. Todos os AGV induziram aumento proporcionalmente maior no peso do ruminorretículo que no peso do omaso, sendo o butirato aparentemente mais estimulador da massa do estômago aglandular. Embora o butirato tenha sido mais estimulador da secreção de insulina, os AGV foram incapazes de induzir ganho nas dimensões papilares. Os AGV aumentaram a proliferação celular nos epitélios do rúmen e do perioplum traseiro, contrariamente ao efeito sobre o plano nasolabial e a epicera. Os efeitos dos AGV sobre a morfologia da mucosa ruminal e de outros tecidos queratinizados sugerem que danos morfológicos no rúmen e cascos podem ter causa comum. Biópsias tegumentares podem ter utilidade como indicadores de alterações morfológicas da mucosa ruminal.

Effects of ammonia load on amino acid utilization by growing steers (2004)

Ruminally cannulated steers were used in two experiments to study effects of rumen ammonia load on methionine and leucine utilization. All steers were limit-fed a diet based on soybean hulls, received ruminal infusions of volatile fatty acids and abomasal infusions of glucose to provide energy, and received an abomasal infusion containing a mixture of all essential amino acids except methionine in Exp. 1 or leucine in Exp. 2. Treatments were arranged as 3 × 2 factorials and included urea (0, 40, or 80 g/day) infused ruminally and methionine (2 or 5 g/day) infused abomasally in Exp. 1 and leucine (0, 4, or 8 g/day) infused abomasally and urea (0 or 80 g/day) infused ruminally in Exp. 2. In Exp. 1, supplementation with the greater amount of methionine improved retained nitrogen, but urea infusions did not alter nitrogen retention. In Exp. 2, leucine linearly increased retained nitrogen, and urea infusions also increased nitrogen retention. The efficiency of deposition of supplemental methionine ranged between 18 and 27%, whereas that for leucine ranged from 24 to 43%. Increasing ammonia load did not negatively impact whole-body protein deposition in growing steers when either methionine or leucine was limiting.

Methionine as a methyl group donor in growing cattle.

Holstein steers were used in two 5 x 5 Latin square experiments to evaluate the sparing of methionine by alternative sources of methyl groups (betaine and choline). Steers were housed in metabolism crates and limit-fed a soybean hull-based diet high in rumen degradable protein. To increase energy supply, ruminal infusions of volatile fatty acids and abomasal infusions of glucose were provided. An amino acid mixture, limiting in methionine, was infused abomasally to ensure that nonsulfur amino acids did not limit protein synthesis. Treatments for Exp. 1 were abomasal infusion of 1) water, 2) 2 g/d L-methionine, 3) 1.7 g/d L-cysteine, 4) 1.6 g/d betaine, and 5) 1.7 g/d L-cysteine + 1.6 g/d betaine. Treatments for Exp. 2 were abomasal infusion of 1) water, 2) 2 g/d L-methionine, 3) 8 g/d betaine, 4) 16 g/d betaine, and 5) 8 g/d choline. In both experiments, nitrogen retention increased in response to methionine (P < 0.05), demonstrating a deficiency of sulfur amino acids. Responses to cysteine, betaine, and choline were all small and not significant. The lack of response to cysteine indicates that the response to methionine was not due to transsulfuration to cysteine or that cysteine supply did not alter the flux of methionine through transsulfuration. The lack of response to betaine suggests that the steers' needs for methyl groups were met by the dietary conditions or that betaine was relatively inefficient in increasing the remethylation of homocysteine to methionine and, thereby, reducing the synthesis of cysteine from homocysteine. Under our experimental conditions, responses to methionine were likely due to a correction of a deficiency of methionine per se rather than of methyl group donors.

Comparison of non-tracer and tracer methods for determination of volatile fatty acid production rate in the rumen of sheep fed on two levels of intake

The aim of the present work was to estimate volatile fatty acid (VFA) production rate in the rumen of sheep fed two levels of intake using both a tracer (TM; by isotope dilution) and a non-tracer method (NTM; by supplementary infusion) in steady-state conditions. Six wethers received a diet containing 700 g lucerne hay and 300 g ground maize/kg in eight equal meals at 3 h intervals per d. The diet (9.8 MJ metabolizable energy (ME)/kg DM) was offered at 90 % ad libitum consumption (high intake, HI) or 45 % ad libitum consumption (low intake, LI) in a crossover design. Each sheep received five intrarumen VFA solutions infused continuously for 24 h at rates of 250 ml and 165 ml/h for the HI and LI respectively. The first infusion, considered as a control treatment (Con), consisted of a solution of [1-(13)C]propionate (7 mmol/d). The four other solutions were isoenergetic (1.9 MJ ME/kg DM intake) mixtures of unlabelled propionate (C(3)) and butyrate (C(4)) at different levels: 0.90 mol C(4)/kg DM intake; 0.60 mol C(3)/kg DM intake; 0.30 mol C(3)/kg DM intake; 1.35 mol C(3)/kg DM intake. The VFA infusions did not affect rumen fermentation of the basal diet (pH, osmotic pressure, protozoa numbers), and comparable DM digestibility of the diet among the different treatments was observed. Both estimation methods demonstrated a similar increase (1.7-fold) in the rumen VFA production rate of sheep fed at intakes varying between 0.9 to 1.7 times maintenance. Irrespective of the intake level, the rumen production rate of individual VFA was on average 1.5-fold higher when estimated by the TM compared with the NTM. Rumen VFA production rates estimated by the NTM and TM represented 80 % and 120 % ME intake respectively. The difference between NTM and TM estimates seems likely to be caused mainly by overestimation of the VFA production rates by the TM.

Response of one-humped camel (Camelus dromedarius) to intravenous glucagon injection and to infusion of glucose and volatile fatty acids, and the kinetics of glucagon disappearance from the blood.

The effects of glucagon injection and infusion of glucose and volatile fatty acids were studied in one-humped camels. Twenty adult male camels were divided into four equal groups. The first group was infused with physiological saline and served as a control. The second group was injected with a single dose of glucagon, the third group was infused with glucose (50%) in sterile saline, and the fourth group was infused with a volatile fatty acid (VFA) mixture. In the first, third and fourth groups, sampling was performed before the beginning of infusions (control time), and at 15, 30, 60 and 120 min post-infusion. Plasma glucagon concentrations were monitored in the second group at 5, 10, 15, 20, 30, 45, 90, 105 and 120 min after injection. For glucagon injection, glucose concentration peaked at 15 min post-injection, and tended to decrease thereafter. Plasma glucose concentrations showed significant rises above the basal value at all times after glucose infusion. VFA infusion had no apparent effect on plasma glucose concentration. After injection of glucagon, the plasma lactate concentration dropped significantly at 15 and 30 min, then increased gradually until it reached the original concentration of lactate at 120 min. However, glucose infusion elevated the plasma lactate concentration only at the end of the infusion period. A decrease in plasma lactate was observed at 60 min after VFA infusion. The present investigation provides evidence that the glucagon level in camels is higher than that in other ruminants and in man, and suggests that this is a probable species specificity, which would explain the higher level of glucose in the blood of camels than in that of other ruminants. The disappearance curve of injected glucagon had, as in other ruminants, an exponential two-compartment function. The hormone was rapidly distributed and was eliminated with a high rate of clearance.

The effect of nutrients on feed intake in ruminants.

The purpose of the present review is to examine the role played by nutrients in controlling feed intake in ruminants, in light of their particular anatomical, physiological, nutritional and behavioural characteristics. The ration is first digested in the rumen for several hours by microbial fermentation. Volatile fatty acids, which constitute 50-75% of a ruminant's energy supply, considerably depress feed intake when administered by short-term infusion into the rumen. However, this effect seems to be largely due to osmolarity problems. Only propionate seems to have a specific action, unrelated to osmolarity, in the mesenteric or portal veins. Nitrogenous nutrients have little short-term effect on feed intake, except when there is excess NH3 in the rumen. Metabolic cues from intestinal digestion, particularly of glucose and starch, have very little short- or long-term influence in controlling feed intake, in comparison with rumen digestion cues. However, the short-term responses in feeding behaviour do not always reflect longer-term effects on feed intake control. The effects of volatile fatty acid infusion on feed intake are much less significant over the long term, except in the case of propionate. The nutrients required for good microbial activity (proteins in the rumen) generally promote feed intake, whereas nutrients that disrupt rumen functioning (lipids) reduce feed intake. After a learning period, preferences are always governed by a tendency toward optimum rumen functioning, rather than by animal nutritional requirements, although the two factors are not independent. Ruminants, due to their particular anatomical and nutritional characteristics, have, in the course of their evolution, developed specific feed intake control mechanisms based on nutritional cues.

Effects of ruminal infusion of volatile fatty acids on plasma concentration of growth hormone and insulin in sheep

In an initial experiment we observed postprandial changes in plasma concentrations of growth hormone (GH), insulin, glucagon, and somatostatin (SRIF) in sheep. We then examined whether increasing the rumen concentration of volatile fatty acids (VFA) by infusing a VFA mixture at three rates (53.5, 107, and 214 μmol/kg/min for 4 hr) mimicked the postprandial changes in hormone secretion. Feeding significantly (P < 0.05) suppressed the plasma GH concentration for 6 hr, whereas it significantly (P < 0.05) increased plasma concentrations of insulin, glucagon, and SRIF. Plasma glucose levels tended to decrease after feeding but then gradually increased over the prefeeding level (P < 0.05). Intraruminal infusion of the VFA mixture at 107 μmol/kg/min caused similar changes in ruminal VFA concentrations to those seen after feeding. The infusion significantly (P < 0.05) suppressed GH secretion in a dose-dependent manner, whereas it caused a significant (P < 0.05) increase in insulin and glucose concentrations without changing glucagon concentrations. From these results, we conclude that the postprandial change in ruminal VFA concentration may be a physiological signal which modifies GH and insulin secretion in sheep.

Effect of glucose supply on fasting nitrogen excretion and effect of level and type of volatile fatty acid infusion on response to protein infusion in cattle

Two experiments were carried out on cattle nourished entirely by intragastric infusion, to determine the extent to which glucose or a glucose precursor determines the response to protein infusion in energy-undernourished animals. In order to determine the requirement for glucose in 1-year-old fasting cattle, glucose was infused at increments to supply 0, 1.5, 2.5, 3.5, 4.5, 5.5 and 6.5 g/kg metabolic body weight (W0.75) and the effects on plasma beta-hydroxybutyrate and N excretion were measured. At 5.5 g glucose/kg W0.75 plasma beta-hydroxybutyrate was reduced to a basal level of 1.65 mmol/l and fasting N excretion reduced from 529 to 280 mg N/kg W0.75. No further reduction was observed with the higher level of 6.5 g glucose/kg W0.75. In the second trial, three steers were used in a 3 x 3 Latin square design and infused with a volatile fatty acid mixture of 65, 27 and 8 mol acetic, propionic and butyric acids respectively/100 mol, either at an estimated maintenance energy level of 450 kJ/kg W0.75 and supplying a calculated glucose equivalent level of 13.0 g/kg W0.75 (M1A), or at 1.5 x maintenance supplying a glucose equivalent of 20 g/kg W0.75 (M1.5A). Another mixture infused at the maintenance energy level contained 49, 43 and 8 mol acetic, propionic and butyric acids respectively/100 mol but with a glucose equivalent of 20 g/kg W0.75 (M1P). Casein was infused at each of these energy treatments to supply 0, 200, 400, 800, 1600 and 2500 mg N/kg W0.75 daily, and N balance and blood metabolites were measured. N retention increased linearly (r 0.98) with casein infusion. The coefficients for N retention were 0.55, 0.57 and 0.64 for M1A, M1.5A and M1P respectively. The mean efficiency of N utilization was 0.58. The results suggest that provided the glucose need is met there is no relationship between energy supply and efficiency and level of protein retention. However, the results also indicate that glucose requirement in cattle may be higher than that previously observed in sheep.

Effects of intraruminal infusions of propionate and butyrate with two different protein supplements on milk production and blood metabolites in dairy cows receiving grass silage‐based diet

Four cows were used in a balanced 4×4 Latin square with 2 week experimental periods to investigate the effects of intraruminal infusions of volatile fatty acids and protein source on milk production and blood metabolites. The four treatments in a 2×2 factorial arrangement were isoenergetic intraruminal infusions of propionate (500 g day−1) or butyrate (417 g day−1) each given with isonitrogenous protein supplementation of fish meal (FM) or barley protein (BP). The cows were fed restrictively with 9 kg dry matter day−1 of formic acid treated grass silage and 8 kg day−1 of concentrate. Propionate infusion increased milk yield (24·9 vs 23·4 kg day−1; P<0·05), milk protein yield (832 vs 778 g day−1; P=0·05) and milk lactose content (44·7 vs 43·5 g kg−1; P<0·05) and yield (1113 vs 1023 g day−1; P<0·01), whereas butyrate infusion was associated with a higher milk fat content (44·7 vs 39·4 g kg−1; P<0·01) and yield (1033 vs 974 g day−1; P<0·01). FM tended (P<0·10) to increase milk yield, but had no significant effects on milk composition or milk component yields compared with BP. Butyrate infusion increased blood ketones, plasma non-esterified fatty acids and glycine relative to propionate infusion. The concentrations of ammonia N in rumen fluid and urea in plasma and milk were similar for both protein supplements. The profile of amino acids in plasma was similar for both protein supplements except for the higher concentrations of phenylalanine, proline and tyrosine with BP. The results show that protein utilisation can be improved by increasing the supply of propionate from rumen fermentation in cows given a grass silage-based diet. © 1998 SCI.

The four treatments in a 2 x 2 factorial arrangement were isoenergetic intraruminal infusions of propionate (500 g day -1 ) or butyrate (417 g day -1 ) each given with isonitrogenous protein supplementation of fish meal (FM) or barley protein (BP). The cows were fed restrictively with 9 kg dry matter day -1 of formic acid treated grass silage and 8 kg day -1 of concentrate. Propionate infusion increased milk yield (24.9 vs 23.4 kg day -1 ; P < 0.05), milk protein yield (832 vs 778 g day -1 ; P = 0-05) and milk lactose content (44.7 vs 43.5 g kg -1 ; P < 0.05) and yield (1113 vs 1023 g day -1 ; P < 0.01), whereas butyrate infusion was associated with a higher milk fat content (44.7 vs 39.4 g kg -1 ; P < 0.01) and yield (1033 vs 974 g day -1 ; P < 0.01). FM tended (P < 0.10) to increase milk yield, but had no significant effects on milk composition or milk component yields compared with BP. Butyrate infusion increased blood ketones, plasma non-esterified fatty acids and glycine relative to propionate infusion. The concentrations of ammonia N in rumen fluid and urea in plasma and milk were similar for both protein supplements. The profile of amino acids in plasma was similar for both protein supplements except for the higher concentrations of phenylalanine, proline and tyrosine with BP. The results show that protein utilisation can be improved by increasing the supply of propionate from rumen fermentation in cows given a grass silage-based diet.

Protein utilization during energy undernutrition in sheep sustained by intragastric infusion. Effect of body fatness on the protein metabolism of energy-restricted sheep.

The effect of body fat content on the protein metabolism of energy-restricted sheep has been studied in two experiments. In the first experiment, six Suffolk-cross wether sheep, three weighing about 39 kg and three of about 61 kg, were given progressively increasing amounts of casein-N from 0 to 3000 mg N/kg metabolic body weight (W0.75) daily with constant energy, 91 kJ/kg W0.75 daily, from a high-propionic acid mixture of volatile fatty acids (VFA). In the second experiment, two lean and two fat sheep of similar body weights were given progressively increasing amounts of casein with the same VFA mixture. All the animals attained a positive N balance when they were in negative energy balance. N balance was not affected by body fatness of the magnitude studied, although lean animals utilized increasing levels of standard VFA (acetate-propionate-butyrate 65:25:10, molar proportions) infusion for N accretion more efficiently than fat animals. Endogenous energy was utilized for protein accretion with an efficiency of 0.56. Supply of glucogenic VFA equivalent to 28 mmol glucose/kg W0.75 reduced fasting N excretion by 39%. Fasting heat production decreased from 335 to 300 kJ/kg W0.75 with the infusion of casein and glucogenic VFA. It is argued that fasting induces additional heat losses due to raised protein metabolism and is unsuited as a baseline for dietary assessment.

Effect of Volatile Fatty Acid Infusion on Development of the Rumen Epithelium in Neonatal Sheep

The purpose of this study was to determine whether the continuous intraruminal infusion of calculated physiological concentrations of volatile fatty acids (VFA) stimulated the metabolic development of the neonatal rumen. Eight 1-wk-old lambs were assigned to one of three treatments: saline infusion (three lambs), VFA infusion (three lambs), or no infusion (two lambs). Rumen catheters were surgically implanted into lambs in the infusion groups. The amount of VFA infused, beginning at 2 wk of age, increased weekly in equal increments of 12.5% of the estimated net energy requirement until, at 6 wk of age, lambs received 50% of their estimated net energy requirement from the infused VFA. All lambs consumed milk replacer for ad libitum intake and had free access to water. The lambs that were infused with VFA tended to have longer rumen papillae. There were no differences in width or number of papillae per square centimeter across treatments. Rumen epithelial cells isolated from lambs that were infused with VFA tended to oxidize less glucose and produce more acetoacetate than did cells from lambs that were infused with saline or from uninfused lambs. β-Hydroxybutyrate production by isolated rumen epithelial cells and concentrations of blood glucose, acetoacetate, and β-hydroxybutyrate were not different among the three treatments. Thus, infusion of physiological concentrations of VFA appears to stimulate some aspects of rumen metabolic development.

Plasma Levels of Energy Metabolites and Pancreatic Hormones in Relation to the Level of Intake and Intraruminal Infusions of Volatile Fatty Acids in Fed Wether Sheep

Abstract The effect of feeding level and intraruminal infusion of volatile fatty acids (VFA) on the jugular plasma concentrations of energy metabolites and insulin and glucagon was studied in wether sheep. Three animals (42 kg ± 0.5 of body weight) of the Swedish Landrace breed were fed a grass hay and barley diet (70:30 on a dry matter basis) at three levels of intake (L1, L2, L3) in a change-over design. In addition to the diet, 3 different VFA mixtures and water (control) were continuously infused into the rumen during 16 days (one mixture during 4 consecutive days). The energy content of the VFA mixtures infused was equivalent to 17% of the calculated metabolizable energy intake per day. Plasma levels of non-esterified fatty acids and citrate decreased, and plasma insulin increased with increasing energy intake. When propionate was absent from the VFA infusion mixtures, plasma citrate tended to increase with increasing energy intake. The plasma citrate levels were negatively correlated to the concentrations of glucose and insulin in plasma. The concentrations of glucose and insulin in plasma were increased by the infusion of propionate in animals fed at L3. There was a high correlation between plasma glucose and insulin. Plasma glucagon levels were not affected either by level of intake or by VFA infusions. No evidence was found to support the concept that propionate is a major regulator of insulin and glucagon secretion in fed sheep.
