Study on the Influencing Mechanism of Human Chorionic Gonadotropin (hCG) on Oocyte Maturation in Patients with Polycystic Ovary Syndrome.

The study was aimed at investigating the influence of human chorionic gonadotropin (hCG) hormone on oocyte maturation in the patients with polycystic ovary syndrome (PCOS). A total of 54 patients with PCOS who received in vitro maturation (IVM) treatment in the Cheeloo College of Medicine, Shandong University, were divided into two groups: one group who underwent hCG injections was the observation group (OG; n = 27) and other was the control group (CG; n = 27) with no hCG injection. The oocyte development and the expression of steroid hormone synthesis-related genes including gonadotropin-releasing hormone receptor (GnRHR), Conexin43, epidermal growth factor-related genes, luteinizing hormone/choriogonadotropin receptor (LHCGR), epiregulin (EREG), and vascular endothelial growth factor (VEGF) were examined. The human ovarian granulosa cell line (SVOG cells) and ovarian epithelial cell line (HOSEpiC cells) were employed to analyze the effect of hCG on the biological behaviour of cells. As a result, OG showed higher normal fertilization, cleavage, and high-qualified embryo rate than CG. Expression levels of GnRHR, Cx43, LHCGR, EREG, and VEGF were significantly elevated in granulosa cells in the OG group. Western blot revealed that phosphoinositide 3-kinase (PI3K), protein kinase B (AKT), and rapamycin (mTOR) proteins were decreased in granulosa cells under hCG intervention. A biological behaviour test indicated that the multiplication capacity of hCG-intervened SVOG and HOSEpiC was increased, while the apoptosis was decreased. In conclusion, hCG could accelerate follicular development and oocyte maturation by activating oocyte maturation genes in PCOS patients, which could significantly improve and popularize the application of IVM technology.

The influence of human chorionic gonadotropin on hormonal and haematological profile of postpubertal male albino rats exposed to chronic oral administration of alcohol

This study evaluated the influence of human chorionic gonadotropin on hormonal and haematological profile of postpubertal male albino rats exposed to chronic oral administration of alcohol. Twenty-four mature male albino rats were assigned to four groups (n=6). Group A was the control, given distilled water, Group B was given 30% ethanol (8 ml/kg) orally 3 times a week, Group C was given human chorionic gonadotropin (HcG) (50 IU/kg) subcutaneously 3 times a week and Group D was given HcG (50 IU/kg) subcutaneously + 30% ethanol (8 ml/kg) orally 3 times a week. The study was for 10 weeks, and hormonal profile and haematology were determined. The follicle stimulating hormone of Group B decreased significantly (P<0.05) when compared to Groups A, C and D. The luteinizing hormone was significantly lower (P<0.05) in Group B when compared to Groups A, C and D. The testosterone level was significantly higher (P<0.05) in Group D when compared to Groups A, B and C. The results obtained from haematology indicated that haemoglobin concentration of Group B was significantly lower (P<0.05) when compared to Groups A, C and D. Ethanol significantly reduced (P<0.05) platelets when compared to Groups A, C and D. There was a significant reduction (P<0.05) in the neutrophils and lymphocytes in Group B when compared to Groups A, C and D. The study showed that HcG improves the level of male hormones and haematology in postpubertal albino rats exposed to chronic alcohol administration.

Influence of Human Chorionic Gonadotropin on Maturation in Striped Murrel, Channa striatus

Gonadal development, gonadosomatic index (GSI), oocyte diameter, fecundity, histology and level of serum steroid hormones (testosterone and estradiol-17β) were observed in captive striped murrel (Channa striatus Bloch) implanted with human chorionic gonadotropin (HCG) capsules and cholesterol capsules as control for a period of five months after implantation. HCG implants induced a significant increase in the GSI of male and female fish. The GSI values were highest in the fourth month (December) and lowest in the first month (September) in HCG implanted fishes. Variation in ova diameter of control and HCG implanted fishes revealed the presence of large size ova in HCG implanted fishes throughout the study period. Similarly, a significant increase in the fecundity of HCG implanted fishes was also observed. Fecundity was highest in December (15415 eggs) in HCG implanted fish, whereas only 2245 eggs were observed from control fish in the above sampling period. In control fish, only perinucleolar and primary oocytes were observed throughout the study period. Similarly, in testis, spermatogonia and few spermatocytes were observed till the end of the study period. Histological examination of the gonads of HCG implanted fish showed that numerous oogonia were present in the ovary after one month of HCG implantation which continued to develop into vitellogenic oocytes in the third and fourth months. The ovary in the fifth month of sampling of HCG implanted fish consisted of oocytes having completed the process of vitellogenesis. The testicular histology of HCG implanted fish showed primary spermatocytes in the first month and testis contained a large reserve of primary spermatocytes and spermatids in the second and third months. A large number of spermatids were observed in the testis in the fourth month of sampling and the testis in the fifth month of sampling predominantly contained spermatozoa. Serum testosterone level was highest (18ng•ml-1) during the fourth month (December) in HCG implanted male fish and serum estradiol-17β level was highest (61.5pg•ml-1) in the third month in HCG implanted female fish. The results of the present work can be used as a reference study for controlled breeding and reproduction of striped murrel, C. striatus.

During pregnancy, the trophoblast develops from the fusion of cytotrophoblastic cells into a syncytiotrophoblast. As the exchange of molecules through gap junctions is considered to play a role in the control of cell and tissue differentiation, the cell to cell diffusion of a fluorescent dye was investigated in human trophoblastic cells differentiating in culture. The fluorescence recovery after photobleaching technique was used to estimate the transfer of 6-carboxyfluorescein from contiguous cellular elements into photobleached cells. Fluorescence recovery follows a slow exponential time course when the cell to cell exchange process is rate limited by the presence of gap junctional channels between contiguous cells, contrasting with a much faster step-like course in the case of fusion of the plasma membranes. In the presence of 10% fetal calf serum, Percoll-purified cytotrophoblastic cells develop into cellular aggregates, then into a syncytium, within 24-48 h after plating. During this in vitro differentiation, fluorescence recoveries after photobleaching with a time course typical for gap junctions were observed between aggregated cytotrophoblastic cells, between cytotrophoblastic cells and syncytiotrophoblasts, and between contiguous syncytiotrophoblasts. The maximum percentage of gap junctional coupling occurs on the fourth day. This fluorescence recovery is attributed to the diffusion of dye through gap junctions, because it can be interrupted by exposure to a known junctional uncoupler (3 mM heptanol). The effects of hCG on this gap junctional communication during trophoblast differentiation were investigated. In the presence of 500 mIU/ml hCG in the culture medium, the percentage of coupled cells was increased at all stages of culture, and the highest proportion of coupled cells was observed after 2 days of culture vs. 4 days in control medium. Moreover, the diffusion rate constant k (the inverse value of the time constant measured on recovery curves) was also significantly increased in the presence of chorionic hormone. It is concluded that during trophoblast differentiation, the development of a cell to cell communication through gap junctions precedes the formation of a morphological syncytium by cell fusion. This gap junctional communication is promoted by hCG. Furthermore, our study confirms the differentiating role and the autocrine action of hCG in the physiology of the trophoblast.

Influence of human chorionic gonadotropin (hCG) and hCG internalization by granulosa cells on the rate of in vitro fertilization and embryonic development of human oocytes

Follicular fluids and granulosa cells were obtained from 28 aspirated follicles of nine women undergoing laparoscopy in an in vitro fertilization program. Follicular growth was stimulated by a human menopausal gonadotropin regimen and laparoscopy was performed 32 hours after human chorionic gonadotropin (hCG) administration. Follicular fluid 17 beta-estradiol (E2) levels were higher and hCG levels were lower in follicles with oocytes that fertilized and cleaved beyond two blastomeres (greater than two-cell group) than in those with nonfertilizable oocytes (NF group) (P less than 0.05). Compared to those from the NF group, granulosa cells from the greater than two-cell group secreted less progesterone (P) in vitro and had a fourfold increase in percentage of cells with internalized hCG. These results demonstrate that the steroidogenic capacity of granulosa cells from follicles whose oocytes fertilize and undergo accelerated embryonic development in vitro differs from the capacity of granulosa cells from NF follicles. This difference may be due to their enhanced ability to bind and subsequently internalize hCG.

Influence of human chorionic gonadotropin in vivo on steroid formation and gonadotropin responsiveness of isolated human preovulatory follicular cells

Granulosa and thecal cells of preovulatory follicles taken from 12 women were isolated and incubated separately for 2 hours in the presence and absence of human chorionic gonadotropin (hCG). To six of these women, an ovulatory dose of hCG (9000 IU) had been given 24 to 30 hours before excision of the follicle. Following incubation, cellular cyclic adenosine 3':5' monophosphate (cAMP) levels and the medium content of progesterone (P), androstenedione (A), and 17 beta-estradiol (E2) were determined. All follicles appeared healthy and well developed, and the oocytes recovered were morphologically normal and mature. Exposure to hCG in vivo caused a shift in steroidogenesis from A toward P formation in isolated thecal cells and a marked increase in the P production by the granulosa cells of the preovulatory follicles. Furthermore, the thecal cells, but not the granulosa cells, developed refractoriness to further stimulation with hCG in vitro.

Influence of Human Chorionic Gonadotropin on Pregnancy Rates in Lactating Dairy and Beef Cows

Single subcutaneous injection of 1500 IU human chorionic gonadotropin or placebo injections were given at the time of insemination in lactating dairy and beef cows to determine their effects on rates of pregnancy. Pregnancy rates at first service in 161 control and 145 treated dairy cows were 52.8% and 44.8%, respectively. Similar rates for 136 control and 145 treated beef cows were 54.4% and 54.5%. Injections of human chorionic gonadotropin were not effective in stimulating conception rates in lactating dairy or beer cows under field conditions.
