Effects of alkylating agents on spermatogenesis in the rabbit.

Serial sperm counts have been made in rabbits treated with alkylating agents. After single injections of Busulphan, Tretamine and isopropyl methanesulphonate, aspermia developed during the 10th and 11th weeks. Other simple monofunctional esters of methane and ethane sulphonic acids, which induced sterility in rats as a result of their action on spermatozoa and late spermatids, did not depress the sperm count or interfere with sperm motility in the rabbit. The results suggest that the duration of spermatogenesis in the rabbit is about 10 weeks and indicate that in this species, as in rats and mice, a steady turnover of spermatozoa occurs. Fertility tests in male rabbits after administration of Tretamine showed that small doses readily induced sterility which could be maintained by continued administration.

SummaryCurative tests of Vit. E activity of ubiquinone, its hydroquinone and acid reduction product in relation to muscular dystrophy in the rabbit were negative. Similarly, solanachromene exhibited no biological activity with respect to dystrophy in rabbits or sterility in rats, Vit. E-deficient dystrophic rabbits underwent a remission of gross symptoms within approximately 5 days following oral treatment with N,N'-diphenyl-p-phenylenediamine. The elevated plasma concentration of unesterified cholesterol declined to near-normal levels within 3 weeks, while skeletal muscle concentration responded more slowly. The results confirm previous findings indicating that the Vit. E requirement of rabbits and rats represents a need for suitable antioxygenic compounds.

Some observations on the genital function of vitamin E-deficient female non-pregnant guinea-pigs.

Vitamin E has been considered to be specially associated with fertility since the first biological effect observed in vitamin E deficiency was sterility in rats, resulting from internal haemorrhage, with death of the fetuses, and their subsequent resorption (Evans and Burr, 1927). Further research has, however, shown that vitamin E is involved in the general metabolism. There has been much discussion about its r6le in the body. Its action seems to lie important in protein metabolism as well as in carbohydrate and lipoid metabolism as recently reviewed by Harris and hf a son (1956). In the early stages of vitamin E-deficiency endocrine disorders have ken observed in animals consisting either in a decrease of gonadotropin production or a decreased reactivity to progesterone. For details of these observations reference is made to the recent survey of Ve r z A r (1956). The following investigation has been carried out in order to study the genital function in vitamin E-deficient female guineapigs.

Induction of translocations in mice by triethylenemelamine.

THE findings of Jackson and Bock1 that triethylenemelamine causes male sterility in rats instigated an investigation into the possibility that this effect is at least partially due to the production of dominant lethals through chromosome breakage. If this were true, one might expect to find that a proportion of the survivors carry translocations. In support of this idea is the work of Fahmy and Fahmy2,3 on Drosophila, where it was shown that triethylenemelamine induces a high frequency of translocations as well as dominant lethals. A search for translocations in the offspring of treated male mice was therefore started.
