The effect of serum triglyceride levels and different lipid-lowering methods on the prognosis of hypertriglyceridemic acute pancreatitis: a single-center 12-year retrospective study by propensity score matching

Aim To investigate the impact of triglyceride on hypertriglyceridemic acute pancreatitis (HTG-AP) and different lipid-lowering methods on triglyceride-lowering efficiency and HTG-AP. Methods The patients with HTG-AP from January 2012 to December 2023 in Civil Aviation General Hospital were analyzed, retrospectively. Patients were divided and compared according to whether their triglycerides were below 5.56 mmol/L at 48 and 72 h of admission. The patients were divided into control group, insulin group, and low molecular weight heparin (LMWH)+bezafibrate group based on the different methods of lipid-lowering. Propensity score matching (PSM) was employed to balance the baseline characteristics. Results There was no correlation between the severity of HTG-AP and the triglyceride at admission. The incidence of severity, local complications, and persistent organ failure (POF) were significantly decreased in patients with 48-h and 72-h triglyceride attainment. Following PSM, the incidence of infectious pancreatic necrosis (IPN) (3.3% vs. 13.3%) was significantly reduced in insulin group compared with control group (p < .05). Compared with control group, LMWH + bezafibrate group had higher lipid reduction efficiency, and the incidence of IPN (0.9% vs. 10.1%) and POF (8.3% vs. 19.3%) was significantly decreased (p < .05). There was no significant difference in the efficiency of lipid-lowering, complications, and POF between LMWH + bezafibrate group and insulin group (p > .05). Conclusion The severity of HTG-AP is not associated with the triglyceride levels at admission. However, rapid reduction of triglyceride levels can lower the incidence of local complications and respiratory failure. Compared with conservative treatment, insulin and LMWH + bezafibrate can both reduce the incidence of IPN in patients with HTG-AP.

Most of the escaped farmed salmon entering a river during a 5-year period were infected with one or more viruses.

Disease interactions between farmed and wild populations have been poorly documented for most aquaculture species, in part due to the complexities to study this. Here, we tested 567 farmed Atlantic salmon escapees, captured in a Norwegian river during 2014-2018, for five viral infections that are prevalent in global salmonid aquaculture. Over 90% of the escapees were infected with one or more viruses. Overall prevalences were: 75.7% for piscine orthoreovirus (PRV-1), 43.6% for salmonid alphavirus (SAV), 31.2% for piscine myocarditis virus (PMCV), 1.2% for infectious pancreatic necrosis virus (IPNV) and 0.4% for salmon anaemia virus (ISAV). A significantly higher prevalence of PMCV infection was observed in immature compared to mature individuals. The prevalence of both SAV and PMCV infections was higher in fish determined by fatty acid profiling to be 'recent' as opposed to 'early' escapees that had been in the wild for a longer period of time. This is the first study to establish a time-series of viral infection status of escapees entering a river with a native salmon population. Our results demonstrate that farmed escapees represent a continuous source of infectious agents which could potentially be transmitted to wild fish populations.

Farm level bio-economic modelling of aquatic animal disease and health interventions

A farm level bio-economic model, for aquatic animal production, of the relationships between inputs (e.g. purchased animals), outputs (e.g. harvested animals) and gross margin (GM) was developed to assess ex-ante the economics of disease and animal health interventions. Feed costs were calculated from estimates of food conversion ratio (FCR), animals harvested and mortality. The model was applied to a typical grow-out rainbow trout (Oncorhynchus mykiss) farm on Lake Titicaca, Peru and a typical shrimp (Paenus vannamei) farm in the Mekong Delta, Vietnam. The model was used in two analyses. Firstly, an approach to assess the burden of disease developed by the Global Burden of Animal Diseases (GBADs) project was adopted. Output under conditions of ‘ideal health’ was estimated by reducing mortality to zero and removing health costs. GM in both systems increased by approximately 25% when production was kept constant (and stocking rates reduced) and more than doubled if production was allowed to rise (and initial stocking increased). The increase in GM under conditions of ideal compared with current production provided an estimate of the maximum possible benefit from improved health management. Secondly, break-even analysis was used to assess the economics of vaccination against infectious pancreatic necrosis (IPN) vaccine (rainbow trout - RBT) and probiotics (shrimp). If initial stocking was kept constant, and production allowed to rise, break-even points for the intervention (when GM was the same with and without the intervention) were achieved when mortality was reduced by 16% in RBT fry and juvenile and 28% in shrimp. If production was kept constant and benefit realised by reduced initial stocking, the break-even point was achieved for i) vaccination of RBT when mortality in fry and juveniles was reduced by 39%, and ii) probiotics in shrimp production when there was a 15% reduction in mortality (nursery and grow-out), 10% increase in shrimp weight at harvest and 10% improvement in FCR. The results demonstrate how relatively simple models, parameterised with basic farm production data, can assess the burden of disease and quantify ex-ante the potential benefit of interventions. In the absence of trial data, these analyses support decision-making by farmers. The models can be adapted for many aquaculture systems. Farm level results can be extrapolated to estimate disease burden, and benefits of interventions, at regional or national level and thus support informed decision-making and allocation of resources to health management.

In-vitro immunomodulatory responses and antiviral activities of antimicrobial peptide octominin against fish pathogenic viruses

Antimicrobial peptides (AMPs) are considered a novel approach to stimulate fish antiviral mechanisms for defense against a broad range of viral infections by enhancing immunomodulatory activities. Octominin is an AMP derived from the defense proteins of Octopus minor. In this study, preliminary screening of octominin against viral hemorrhagic septicemia virus (VHSV), infectious hematopoietic necrosis virus (IHNV), and infectious pancreatic necrosis virus (IPNV) was carried out. Moreover, immune responses upon octominin treatment and IHNV challenge were investigated using fathead minnow (FHM) cells. The CC50s of octominin for FHM and Chinook salmon embryo-214 (CHSE-214) cells were 2146.2 and 1865.2 μg/mL, respectively. With octominin treatment, EC50 resulted in 732.8, 435.1, and 925.9 μg/mL for VHSV, IHNV, and IPNV, respectively. The selectivity indices were 2.9, 4.9, and 2.0, respectively. The transcriptional analysis results demonstrated the induced transcription factors (Irf3; 143-fold, Irf7; 105-fold, and NF-κB; 8-fold), stress response gene (HspB8; 2-fold), and apoptosis functional gene (p53; 3-fold) in octominin treated (500 μg/mL) FHM cells for 48 h. Moreover, IHNV viral copy number was slightly decreased with the octominin treatment (500 μg/mL) in FHM cells. Overall results suggest that octominin could be a potential antiviral agent, although further studies are necessary to understand its mode of action and the mechanism of its antiviral activity.

Study of in vitro reproduction of infectious pancreatic necrosis virus (IPNV)

The aim of the work was to study the in vitro reproduction of new emergent isolates of infectious pancreatic necrosis virus (IPNV) in purpose to determine virulence properties. The new strains of IPNV isolated from trout farms in Ukraine during 2021-2022 were investigated. For this purpose, RTG-2 cell line was used, on which the infectious titer of the virus and the manifestation of CPE signs were determined. The infectious titer of selected isolates in RTG-2 cell line ranged from 1.1x103 to 1.0x107 TCID50/ml Isolated IPNV strains caused low (≤25 %), moderate (≤50 %) and high (≥70 %) mortality rates in trout fry. Based on biological properties and indicators of infectious titer, the investigated isolates VN11, VN18 and VN29 were determined as with low virulence, and isolates VN20, VN30, VN32 and VN39 were classified as highly virulent. Among the highly virulent isolates, VN32 had the highest titer of 1.0x107 TCID50/ml and caused mortality in rainbow trout with an average weight of 100 g. The IPNV strain isolated from the brook trout had an infectious titer of 1.58x106 TCID50/ml and caused fry mortality ranged from 15 to 25 %. The number of detected isolates and their cultural properties testify to the biodiversity of strains for the Infectious pancreatic necrosis virus in Ukraine. How many newly discovered isolates of the IPNV belong to one or another genotype remains to be determined. For this, a study of the nucleotide sequence of the gene encoding the capsid protein VP2 will be conducted, on the basis of which it will also be possible to determine virulence by changes in amino acids sequences

Characterization of the activity of 2′-C- methylcytidine against infectious pancreatic necrosis virus replication

Infectious pancreatic necrosis virus (IPNV) is the pathogen of infectious pancreatic necrosis (IPN), which can cause high mortality in salmonids, harm the healthy development of salmon-trout aquaculture, and lead to huge economic losses. However, in China, there is currently neither a commercially available vaccine to prevent IPNV infection nor antiviral drugs to treat IPNV infection. The genome of IPNV consists of two segments of dsRNA named A and B. Segment B encodes the RNA-dependent RNA-polymerase (RdRp) VP1 which is essential for viral RNA replication and is therefore considered an important target for the development of antiviral drugs. In this study, we investigate whether 2′-C-methylcytidine (2CMC), a nucleoside analog which target viral polymerases, has an inhibitory effect on IPNV both in vitro and in vivo. The results show that 2CMC inhibits IPNV infection by inhibiting viral RNA replication rather than viral internalization or attachment. In vivo experiment results showed that 2CMC could inhibit viral RNA replication and reduce viral load in rainbow trout (Oncorhynchus mykiss). In our study, we have revealed that 2CMC has a potent inhibitory effect against IPNV infection. Our data suggest that 2CMC is an attractive anti-IPNV drug candidate which will be highly valuable for the development of potential therapeutics for IPNV.

Epigenetic reprogramming around IFN1 and IFNy2 promoters in rainbow trout cells inoculated with infectious pancreatic necrosis virus (IPNV).

Infectious pancreatic necrosis virus (IPNV) has proven to effectively evade the host antiviral responses. This study clarifies whether the modulation of the antiviral immune response exerted by IPNV involves epigenetic mechanisms. An in-silico characterization of the rainbow trout IFN1 and IFNγ2 promoters was performed, identifying the islands or sequences rich in CpG dinucleotides and the putative transcription factor binding sites (TBS) for both gene promoters. RTS11 cells (rainbow trout monocyte/macrophage) were infected with IPNV, and the course of viral infection was followed up to 48h post infection (hpi). Infected cells showed increased IFN1 and IFNγ2 transcriptional expression at 6 and 24 hpi, respectively. IPNV infection caused increases and decreases in global IFNγ2 promoter methylation at 6 and 24 hpi, respectively. The CpG dinucleotides at positions -392 and+38 of this promoter were the most sensitive to methylation changes. The IFN1 promoter remained fully unmethylated during the course of the infection, similar to the control. The changes in the methylation pattern observed for the IFNγ2 promoter were coincident with the changes in DNA methyltransferase (DNMT) expression levels, increasing at 6 hpi and decreasing below basal level at 24 hpi. Similarly, the H4 histones associated with the IFN1 and IFNγ2 promoters were hyperacetylated at 6 hpi, subsequently decreasing their acetylation below basal levels at 24 hpi, in both promoters. Coincidentally with the above, overexpression of histone acetyltransferase (HAT) was observed at 6 hpi and of histone deacetylase (HDAC) at 24 hpi, with return to baseline of HAT. These results suggest that IPNV would epigenetically modulate the expression of IFN1 by changing acetylation levels of the histones H4 associated with its promoter. Also, the modulation of the expression of IFNy2 would be by switching methylation/demethylation levels of its promoter, in addition to changes in acetylation levels of histones H4 associated with this promoter. This study is the first to demonstrate the effect of epigenetic reprogramming after IPNV infection in salmonid cells, demonstrating that promoter methylation/demethylation level and changes in the histone code associated with promoters may play a role in the modulation of the immune response induced by the virus.

CRISPR-Cas- induced IRF3 and MAVS knockouts in a salmonid cell line disrupt PRR signaling and affect viral replication.

Interferon (IFN) responses are critical in the resolution of viral infections and are actively targeted by many viruses. They also play a role in inducing protective responses after vaccination and have been successfully tested as vaccine adjuvants. IFN responses are well conserved and function very similar in teleosts and mammals. Like in mammals, IFN responses in piscine cells are initiated by intracellular detection of the viral infection by different pattern recognition receptors. Upon the recognition of viral components, IFN responses are rapidly induced to combat the infection. However, many viruses may still replicate and be able to inhibit or circumvent the IFN response by different means. By employing CRISPR Cas9 technology, we have disrupted proteins that are central for IFN signaling in the salmonid cell line CHSE-214. We successfully generated KO clones for the mitochondrial antiviral signaling protein MAVS, the transcription factors IRF3 and IRF7-1, as well as a double KO for IRF7-1/3 using an optimized protocol for delivery of CRISPR-Cas ribonucleoproteins through nucleofection. We found that MAVS and IRF3 KOs inhibited IFN and IFN-stimulated gene induction after intracellular poly I:C stimulation as determined through gene expression and promoter activation assays. In contrast, the IRF7-1 KO had no clear effect. This shows that MAVS and IRF3 are essential for initiation of intracellular RNA-induced IFN responses in CHSE-214 cells. To elucidate viral interference with IFN induction pathways, the KOs were infected with Salmon alphavirus 3 (SAV3) and infectious pancreatic necrosis virus (IPNV). SAV3 infection in control and IRF7-1 KO cells yielded similar titers and no cytopathic effect, while IRF3 and MAVS KOs presented with severe cytopathic effect and increased titers 6 days after SAV 3 infection. In contrast, IPNV yields were reduced in IRF3 and MAVS KOs, suggesting a dependency on interactions between viral proteins and pattern recognition receptor signaling components during viral replication. Aside from more insight in this signaling in salmonids, our results indicate a possible method to increase viral titers in salmonid cells.

Traditional Chinese medicine bufalin inhibits infectious pancreatic necrosis virus genogroups I and V infection in vitro and in vivo

Infectious pancreatic necrosis virus (IPNV) is the causative pathogen of infectious pancreatic necrosis (IPN), which causes high mortality and great economic losses in salmon and trout aquaculture. IPNVs are divided into seven genotypes, and all IPNV isolates that have been reported in China belong to genogroups I and V. Because of restrictions on imported vaccines, China has no commercial vaccine against IPNV; Therefore, more effective antiviral drugs and vaccines are needed. Traditional Chinese medicine (TCM) has been used to treat endemic and pandemic diseases and is reported effective against viral infections. We screened a TCM library containing 1483 compounds and found that bufalin exhibited anti-IPNV activity. Bufalin inhibits infections from IPNV genogroups I and V by inhibiting viral internalization and RNA replication rather than viral attachment. In vivo results showed that bufalin significantly inhibited viral loads in IPNV-targeted tissues in rainbow trout. Mechanistic research showed that bufalin's antiviral effects occurred via Na+/K+-ATPase. Our results indicate that bufalin inhibits IPNV infections both in vitro and in vivo and may be a promising drug for treating IPNV infections.

Assessment of listing and categorisation of animal diseases within the framework of the Animal Health Law (Regulation (EU) No 2016/429): infectious pancreatic necrosis (IPN).

Infectious pancreatic necrosis (IPN) was assessed according to the criteria of the Animal Health Law (AHL), in particular, the criteria of Article 7 on disease profile and impacts, Article 5 on its eligibility to be listed, Annex IV for its categorisation according to disease prevention and control rules as in Article 9, and Article 8 for listing animal species related to IPN. The assessment was performed following a methodology previously published. The outcome reported is the median of the probability ranges provided by the experts, which indicates whether each criterion is fulfilled (lower bound ≥ 66%) or not (upper bound ≤ 33%), or whether there is uncertainty about fulfilment. Reasoning points are reported for criteria with an uncertain outcome. According to the assessment here performed, it is uncertain whether IPN can be considered eligible to be listed for Union intervention according to Article 5 of the AHL (50-90% probability). According to the criteria in Annex IV, for the purpose of categorisation related to the level of prevention and control as in Article 9 of the AHL, the AHAW Panel concluded that IPN does not meet the criteria in Section 1 (Category A; 0-1% probability of meeting the criteria) and it is uncertain whether it meets the criteria in Sections 2, 3, 4 and 5 (Categories B, C, D and E; 33-66%, 33-66%, 50-90% and 50-99% probability of meeting the criteria, respectively). The animal species to be listed for IPN according to Article 8 criteria are provided.

Establishment of an In Vitro Model to Study Viral Infections of the Fish Intestinal Epithelium.

Viral infections are still a major concern for the aquaculture industry. For salmonid fish, even though breeding strategies and vaccine development have reduced disease outbreaks, viral diseases remain among the main challenges having a negative impact on the welfare of fish and causing massive economic losses for the industry. The main entry port for viruses into the fish is through mucosal surfaces including that of the gastrointestinal tract. The contradictory functions of this surface, both creating a barrier towards the external environment and at the same time being responsible for the uptake of nutrients and ion/water regulation make it particularly vulnerable. The connection between dietary components and viral infections in fish has been poorly investigated and until now, a fish intestinal in vitro model to investigate virus-host interactions has been lacking. Here, we established the permissiveness of the rainbow trout intestinal cell line RTgutGC towards the important salmonid viruses-infectious pancreatic necrosis virus (IPNV), salmonid alphavirus (subtype 3, SAV3) and infectious salmon anemia virus (ISAV)-and explored the infection mechanisms of the three different viruses in these cells at different virus to cell ratios. Cytopathic effect (CPE), virus replication in the RTgutGC cells, antiviral cell responses and viral effects on the barrier permeability of polarized cells were investigated. We found that all virus species infected and replicated in RTgutGC cells, although with different replication kinetics and ability to induce CPE and host responses. The onset and progression of CPE was more rapid at high multiplicity of infection (MOI) for IPNV and SAV3 while the opposite was true of ISAV. A positive correlation between the MOI used and the induction of antiviral responses was observed for IPNV while a negative correlation was detected for SAV3. Viral infections compromised barrier integrity at early time points prior to observations of CPE microscopically. Further, the replication of IPNV and ISAV had a more pronounced effect on barrier function than SAV3. The in vitro infection model established herein can thus provide a novel tool to generate knowledge about the infection pathways and mechanisms used to surpass the intestinal epithelium in salmonid fish, and to study how a virus can potentially compromise gut epithelial barrier functions.

Effects of artificial photoperiods on antigen-dependent immune responses in rainbow trout (Oncorhynchus mykiss).

In this study, we investigated the effects of the artificial photoperiods that mimic summer (16L:8D; 16 h Light: 8 h Dark) and winter (8L:16D) solstices, equinoxes (12L:12D), and the artificial 24-h light regimen (24L:0D) on the leukocyte populations and the T helper and regulatory type responses on rainbow trout (Oncorhynchus mykiss). Using flow cytometry analysis, we found that photoperiod induces changes in head kidney leukocyte subsets. The lymphoid subset increased in the 16L:8D summer solstice regime. The analysis using antibodies against B and T cells showed the increase of CD4-1+ T lymphocytes and other unidentified lymphoid cells, with no changes in the B cells. To investigate the modulatory influence of the photoperiod on the fish T cell response, we quantified in the head kidney the transcript levels of genes involved in the Th1 type response (t-bet, ifn-ƴ, il-12p35, il-12p40c), Th2 type response (gata3, il-4/13a), Th17 response (ror-ƴt, il-17a/f), T regulatory response (foxp3α, il-10a, tgf-β1), and the T cell growth factor il-2. The results showed that the seasonal photoperiod alone has a limited influence on the expression of these genes, as the only difference was observed in il-14/13a and il-10a transcripts of fish kept on the 16L:8D regimen. In addition, the 24L:0D treatment used in aquaculture produces a reduction of il-14/13a and il-17a/f. We also evaluated the effect of photoperiod in the presence of an antigenic stimulus. Thus, in fish immunized with the recombinant viral protein 1 (rVP1) of infectious pancreatic necrosis virus (IPNV), the photoperiod had a striking influence on the type of adaptive immune response. Each photoperiod fosters a unique immune signature of antigenic response. A classical type 1 response is observed in fish subjected to the 16L:8D photoperiod. In contrast, fish in the 12L:12D photoperiod showed only the upregulation of il-12p40c. Furthermore, none of the cytokines were increased in fish maintained on the artificial 24L:0D regimen, and a decrease in the master transcription factors (t-bet, ror-ƴt, and foxp3α) was observed. Thus, fish on the 12L:12D and 24L:0D photoperiod appear hyporesponsive regarding the T cell response. Altogether, this study showed that photoperiods modify the magnitude and quality of the T-helper response in rainbow trout and thus impact essential mechanisms for the generation of immune memory and protection against microorganisms.

Reassortant viruses threatening fish aquaculture

AbstractAquaculture provides more than half of fish destined for human consumption worldwide. In aquaculture, infectious diseases triggered by viruses are amongst the major cause of mortality of farmed fish. The cohabitation of different virus strains in the same geographical area opens the possibility to natural reassortment. Virus reassortment is a characteristic recombination process of segmented viruses in which different viruses merge their genome by the shuffling of complete segments. This is possible when two or more viral strains are cohabitating into a single host, boosting their probability to concatenate their viral cycles, getting to pack segments from distinct parental strains. Cohabitation of different viruses in a geographical area, and therefore the possibility of natural reassortment, has been aided by the increment in the transport of eggs and larvae between facilities around the world. Reassortant viruses have been described to show special tropisms for fish species opposed from those affected by parental virus strains, entailing an emerging threat for aquaculture but also for wild populations. To date, outbreaks of reassortant strains of infectious pancreatic necrosis virus, nodavirus (NNV), infectious salmon anaemia virus and tilapia lake virus have been described. Unfortunately, only the characterization of reassortant NNV has been explored. The aim of this review is compiling the information available concerning reassortant viruses affecting fish species of relevance for aquaculture highlighting their potential impact for the sector. Additionally, recent research advances in host‐NNV interaction are also included.

Characterization of a Novel Infectious Pancreatic Necrosis Virus (IPNV) from Genogroup 6 Identified in Sea Trout (Salmo trutta) from Lake Vänern, Sweden.

In November 2016, infectious pancreatic necrosis virus (IPNV) was isolated from a broodstock female of landlocked sea trout (Salmo trutta) in Lake Vänern in Sweden. VP2 gene sequencing placed the IPNV isolate in genogroup 6, for which pathogenicity is largely unknown. Lake Vänern hosts landlocked sea trout and salmon populations that are endangered, and thus the introduction of new pathogens poses a major threat. In this study we characterized the novel isolate by conducting an infection trial on three salmonid species present in Lake Vänern, whole genome sequencing of the isolate, and prevalence studies in the wild sea trout and salmon in Lake Vänern. During the infection trial, the pathogenicity of the Swedish isolate was compared to that of a pathogenic genogroup 5 isolate. Dead or moribund fish were collected, pooled, and analyzed by cell culture to identify infected individuals. In the trial, the Swedish isolate was detected in fewer sample pools in all three species compared to the genogroup 5 isolate. In addition, the prevalence studies showed a low prevalence (0.2-0.5%) of the virus in the feral salmonids in Lake Vänern. Together the data suggest that the novel Swedish IPNV genogroup 6 isolate is only mildly pathogenic to salmonids.

Differential response of RTGUTGC and RTGILL-W1 rainbow trout epithelial cell lines to viral stimulation.

Mucosal surfaces constitute the main route of entry of pathogens into the host. In fish, these mucosal tissues include, among others, the gastrointestinal tract, the gills and the skin. However, knowledge about the mechanisms of regulation of immunity in these tissues is still scarce, being essential to generate a solid base that allows the development of prevention strategies against these infectious agents. In this work, we have used the RTgutGC and RTgill-W1 epithelial-like cell lines, derived from the gastrointestinal tract and the gill of rainbow trout (Oncorhynchus mykiss), respectively, to investigate the transcriptional response of mucosal epithelial cells to a viral mimic, the dsRNA poly I:C, as well as to two important viral rainbow trout pathogens, namely viral haemorrhagic septicaemia virus (VHSV) and infectious pancreatic necrosis virus (IPNV). Additionally, we have established how the exposure to poly I:C affected the susceptibility of RTgutGC and RTgill-W1 cells to both viruses. Our results reveal important differences in the way these two cell lines respond to viral stimuli, providing interesting information on these cell lines that have emerged in the past years as useful tools to study mucosal responses in fish.

Analysis of microbiota-host communication mediated by butyrate in Atlantic salmon

Butyrate is a microbiota-produced metabolite, sensed by host short-chain fatty acid receptors FFAR2 (Gpr43), FFAR3 (Gpr41), HCAR2 (Gpr109A), and Histone deacetylase (HDAC) that promotes microbiota-host crosstalk. Butyrate influences energy uptake, developmental and immune response in mammals. This microbial metabolite is produced by around 79 anaerobic genera present in the mammalian gut, yet little is known about the role of butyrate in the host-microbiota interaction in salmonid fish. To further our knowledge of this interaction, we analyzed the intestinal microbiota and genome of Atlantic salmon (Salmo salar), searching for butyrate-producing genera and host butyrate receptors. We identified Firmicutes, Proteobacteria, and Actinobacteria as the main butyrate-producing bacteria in the salmon gut microbiota. In the Atlantic salmon genome, we identified an expansion of genes orthologous to FFAR2 and HCAR2 receptors, and class I and IIa HDACs that are sensitive to butyrate. In addition, we determined the expression levels of orthologous of HCAR2 in the gut, spleen, and head-kidney, and FFAR2 in RTgutGC cells. The effect of butyrate on the Atlantic salmon immune response was evaluated by analyzing the pro and anti-inflammatory cytokines response in vitro in SHK-1 cells by RT-qPCR. Butyrate decreased the expression of the pro-inflammatory cytokine IL-1β and increased anti-inflammatory IL-10 and TGF-β cytokines. Butyrate also reduced the expression of interferon-alpha, Mx, and PKR, and decreased the viral load at a higher concentration (4 mM) in cells treated with this molecule before the infection with Infectious Pancreatic Necrosis Virus (IPNV) by mechanisms independent of FFAR2, FFAR3 and HCAR2 expression that probably inhibit HDAC. Moreover, butyrate modified phosphorylation of cytoplasmic proteins in RTgutGC cells. Our data allow us to infer that Atlantic salmon have the ability to sense butyrate produced by their gut microbiota via different specific targets, through which butyrate modulates the immune response of pro and anti-inflammatory cytokines and the antiviral response.

Correlation of Uric Acid with Disease Severity, Prognosis, and Complications in Patients with Severe Acute Pancreatitis

This study aimed to investigate the role of serum uric acid (UA) in early identification of the severity, prognosis and complications in patients with severe acute pancreatitis (SAP). The clinical data of SAP patients were collected retrospectively (n=268), and patients were divided into high UA group and low UA group based on the median UA level. Propensity score matching was used to make the baseline data comparable, and 85 patients were selected in each group. Spearman correlation analysis was performed to assess the relationship between UA level and severity and prognosis. The ROC curve was designed to evaluate the ability of UA to predict complications. In the high UA group, disease severity scores were higher, ICU stay and total hospital days were longer. UA levels were found to be positively associated to disease severity scores, ICU stay and total hospital days. The ROC curve revealed that the UA level had an useful predictive value for infectious pancreatic necrosis (IFN). UA level can be used as a convenient indicator of disease severity and IFN risk in SAP patients.

Evaluation of an ultrafiltration membrane for the removal of fish viruses and bacteria in aquaculture water

Ultrafiltration (UF) membranes are used to successfully remove waterborne virus and bacteria from wastewater and drinking water. However, UF membrane application in aquaculture water treatment is limited. In this study we evaluate the performance of a capillary polyethersulfone UF membrane to remove two benchmark waterborne fish pathogens: i) the infectious pancreatic necrosis virus - IPNV, which is an unenveloped icosahedral virus, and ii) the bacterium Aeromonas salmonicida, which is a Gram-negative, facultative anaerobic bacilli. Moreover, the UF membrane bench-scale unit was tested at two temperatures according to salmonid aquaculture: low (4 - 7°C) and high (16 - 19°C). Sterilised natural seawater was spiked with laboratory cultured pathogens, the water was filtrated, and the membrane permeate collected. Both pathogen solution and permeate were evaluated using a cell culture method to estimate the colony-forming units (CFU/ml) for bacteria presence, a median tissue culture infectious dose (TCID50/ml) assay for virus presence, and real-time quantitative polymerase chain reaction (RT-qPCR) for both bacteria and virus presence. The membrane permeate was negative for both virus and bacteria for all analysis and for both low and high temperatures. The results from this bench-scale study are encouraging for the application of UF membrane technology in aquaculture water treatment to prevent virus and bacteria outbreaks. Further studies should validate this UF membrane technology results in commercial aquaculture conditions.

First detection of Cyclopterus lumpus virus in England, following a mortality event in farmed cleaner fish

Lumpfish are used routinely as cleaner fish to delouse farmed Atlantic salmon (Salmo salar) and are often translocated globally. In September 2021, an aquaculture production business in Dorset, England experienced considerable mortalities in consignments of lumpfish (Cyclopterus lumpus) imported from Norway. Following suspicion of infection with Cyclopterus lumpus virus (CLuV) by a veterinarian, commercial testing provided presumptive confirmation of a CLuV infection using reverse-transcription real-time polymerase chain reaction (RT-rPCR). Affected fish were later resampled by the Centre of Environment, Fisheries, and Aquaculture Science, Fish Health Inspectorate for screening of CLuV by RT-rPCR, and confirmation by conventional RT-PCR and sequence analysis. Additional samples were tested using isolation of viruses on cell cultures, bacteriology, histopathology, and PCR/rPCR techniques to investigate whether other pathogens of concern to the salmon aquaculture industry were also present. All samples tested negative for the presence of viral haemorrhagic septicaemia virus, infectious haematopoietic necrosis virus , nervous necrosis virus, infectious pancreatic necrosis virus , infectious salmon anaemia virus , ranavirus, and salmonid alphavirus. Four pools of ten samples tested positive by RT-rPCR for CLuV (Ct values between 19 and 29) and was confirmed using conventional RT-PCR. Sequence analysis showed that amplicons shared a 99.63 % match with the published sequences for CLuV. The CLuV has only recently been reported in aquatic animals and its full impact is not yet known. To the best of our knowledge this is the first report of CLuV in England.

Development of a recombinant adenovirus-vectored vaccine against both infectious hematopoietic necrosis virus and infectious pancreatic necrosis virus in rainbow trout (Oncorhynchus mykiss)

Infectious hematopoietic necrosis virus (IHNV) and infectious pancreatic necrosis virus (IPNV) are typical pathogens of rainbow trout Oncorhynchus mykiss, and the concurrent infection of the two viruses is very common among modern trout hatcheries, which has caused huge economic losses to the rainbow trout farming industry. To prevent and control the spread of IHNV and IPNV in juvenile trout simultaneously, in this study a bivalent recombinant adenovirus vaccine with IHNV Glycoprotein (G) and IPNV VP2 genes was developed. After immunizing juvenile trout with this bivalent vaccine via the immersion route, the expression levels of IHNV G and IPNV VP2 and the representative immune genes in vaccinated and control rainbow trout were tested to evaluate the correlation of immune responses with the expression of viral genes. The neutralizing antibody level induced by this bivalent vaccine as well as the protection efficacy of the vaccine against IHNV and IPNV was also evaluated. The results showed that IHNV G and IPNV VP2 were successfully expressed in juvenile trout, and all the innate and adaptive immune genes were up-regulated. This indicated that the level of the innate and adaptive immune responses were significantly increased, which might be induced by the high expression of the two viral proteins. Compared with the controls, high levels of neutralizing antibodies against IHNV and IPNV were induced in the vaccinated trout. Besides, the bivalent recombinant adenovirus vaccine showed high protection rate against IHNV, with the relative percent survival (RPS) of 81.25%, as well as against IPNV, with the RPS of 78.95%. Taken together, our findings clearly demonstrated that replication-defective adenovirus can be developed as a qualified vector for fish vaccines and IHNV G and IPNV VP2 were two suitable antigenic genes that could induce effective immune protection against these two pathogens. This study provided new insights into developing bivalent vectored vaccines and controlling the spread of IHNV and IPNV simultaneously in juvenile trout.
