Associations between polybrominated diphenyl ethers (PBDEs) levels in adipose tissues and female menstrual cycle and menstrual bleeding duration in Shantou, China

The endocrine-disrupting effects of human exposure to polybrominated diphenyl ethers (PBDEs) have been studied, but their associations with menstrual status were not clearly clarified. This study was to evaluate the associations between PBDE levels in adipose tissues and the menstrual cycle and menstrual bleeding duration alteration. A total of 298 female cases undergoing surgery were recruited from two hospitals in Shantou, China. Demographic, clinical, and pathological information were collected, and adipose tissues were obtained during mammary or abdominal surgery. Gas chromatography with mass spectrometry was used to analyze 14 PBDE congeners in the adipose samples. The associations between PBDE levels and menstrual cycle (MC) and menstrual duration (MD) were analyzed by logistic regression models, estimating odds ratios (ORs) and 95% confidence intervals (CIs). BDE-153 had the highest level in the adipose samples, followed by BDE-209, BDE-183 and BDE-47. Compared with referents, median levels of BDE-47, -71, −153, −183 were higher in women with MC > 30 days (all P < 0.05); BDE-47, -153, −183, −209 were also higher in women with MD > 5.5 days (all P < 0.05). After adjusted for age and parity, BDE-47, -71, −153, −183 were associated with prolonged MC (OR = 1.20, 1.15, 1.12, 1.11, respectively, all P < 0.05) in the logistic regression models; and BDE-47, -153, −183, −209 were associated with the prolonged MD (OR = 1.13, 1.09, 1.10, 1.11, respectively, all P < 0.05). Several individual PBDE congeners in female adipose were found associated with prolonged menstrual cycle and menstrual duration. PBDEs may influence reproductive health of women by altering menstrual status.

Assessment of binding potencies of polychlorinated biphenyls and polybrominated diphenyl ethers with Baikal seal and mouse constitutive androstane receptors: Comparisons across species and congeners

The present study evaluated the binding potencies (equilibrium dissociation constant: KD) of polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) with the constitutive androstane receptor (CAR)_ligand binding domain (LBD) of the Baikal seal (bsCAR_LBD) and mouse (mCAR_LBD) using a surface plasmon resonance (SPR) biosensor. The binding affinities of individual congeners with mCAR_LBD tended to be higher than those with bsCAR_LBD but the differences were within the same order of magnitude. Notably, PBDE congeners showed higher binding affinities for both CAR_LBDs than PCB congeners. In silico docking simulations demonstrated that PBDEs had more non-covalent interactions with specific amino acid residues in both CAR_LBDs than PCBs, supporting the results of their binding affinities. Binding affinity comparisons among congeners revealed the structural requirements for higher binding; mono or di ortho-, tri meta-, and di para‑chlorine substitutions for PCBs, and di or tri ortho-, mono meta-, and di para‑bromine substitutions for PBDEs. The binding potencies of these congeners unlikely accounted for their previously reported CAR-mediated transactivation potencies, implying that their transactivation is regulated in a ligand-dependent, but a distinct manner from ligand binding. Risk assessment analysis showed that the KD values of individual PCB and PBDE congeners were 1-4 orders of magnitude higher than their respective hepatic concentrations in wild Baikal seal population.

Ensuring the reliability of brominated flame retardant data on food and feed occurrence through harmonised analytical criteria and proficiency testing

The volume of occurrence data on food and animal feed contaminants such as polybrominated diphenyl ethers (PBDEs) and hexabromocyclododecanes (HBCDDs) is slowly increasing as more laboratories develop analytical capability. This data allows an evaluation of current background levels in different countries and regions and is also useful for estimating the health risk through dietary exposure and as evidence for the formulation of future control strategies. Existing data varies in the number of analytes reported and the quality measures applied. In order to ensure reliability and comparability, guidance on analytical criteria such as precision and trueness, limits of quantitation, recovery, positive identification, etc. is provided. These parameters are based on several years of collective experience and allow validation and regular quality control of analysis of individual PBDE congeners and HBCDD stereoisomers. The criteria-based approach also allows laboratories the flexibility to use different analytical methodologies and techniques for generating data. The effectiveness of this approach has been demonstrated by a successful proficiency testing scheme that has been used for a number of years and has attracted an increasing number of participants. The majority of participating laboratories (>80%) have been able to demonstrate performance within the 95% confidence interval (│z-score│≤ 2) and a further 10% of laboratories demonstrated performance with a z-score of (2 <│z-score│< 3). The combined support of these guidance criteria backed by successful proficiency testing will ensure the reliability and comparability of results, in particular, to refine risk assessments and to help the formulation of regulatory policy.

Improved chromatography separation for polybrominated diphenyl ether congeners quantification in the food of animal origin

The improvement of polymers flames resistance for over fifty years using polybrominated diphenyl ethers (PBDEs) resulted in the environment and food of animal origin pollution. The number of PBDE congeners, their low concentration in food, and potentially co-eluting isobaric compounds require careful selection of chromatographic separation parameters. The aim of this study was the separation and determination of ten individual PBDE congeners in the fish muscle, oil, eggs, and milk required by European law (2014/118/EU). The analytical procedure of ten PBDE congeners quantification was based on the isotope (13C12) dilution method and the gas chromatography with high-resolution mass spectrometry detection. The analytes were extracted, then clean up using self-packed columns. The congeners separation using six capillary gas chromatography columns was checked. Two columns were selected for further study. The first one allowed for the separation of BDE-28 and BDE-154 from disturbing isobaric compounds, while the second one gave the appropriate sensitivity for highly brominated congeners. The method was successfully validated and positively evaluated in international proficiency tests. This work shows how critical is proper chromatographic separation for PBDE determination in food samples. Despite the use of sophisticated analytical techniques, false-positive results can still be obtained.

PBDE serum concentration and preschool maturity of children from Slovakia

Polybrominated diphenylethers (PBDEs) are persistent organic pollutants (POPs), they are considered endocrine disruptors and can bioaccumulate in nature, and in living tissue. Human exposure to and the presence of PBDEs in human samples is of concern due to their potential health risks. Young children are one of the most vulnerable populations to PBDE's potential health effects.Ninety-one serum samples of 6-year-old children, residing in a contaminated location, due to former production of polychlorinated biphenyls (PCBs), were analysed to examine children's exposure to PBDEs in Slovakia. Median serum concentrations found for individual PBDE congeners BDE-28+33, −47, −99, −100, −153, −154 and −183 were 0.015, 0.184, 0.079, 0.046, 0.176, 0.014, and 0.097 ng g−1 lipid weight, respectively.Children's preschool maturity was measured using the Wechsler Preschool and Primary Scale of Intelligence (WPPSI-III) test. In multivariate analyses BDE-153 serum concentrations were significantly inversely associated with WPPSI-III composite score (p = 0.011, β = −23.6), while adjusting for PCB-153 and sex. Significant negative associations were observed for BDE-153 serum concentrations (p = 0.002, β = −29.8) and WPPSI-III composite score, after controlling for PCB-118 and sex. Negative associations were also observed for BDE-47, BDE-100 and BDE-153, with different individual WPPSI subtest scores, after adjustment with PCB-153 and/or PCB-118 and sex. Serum concentrations of PCB-153 and PCB-118 were not statistically significantly associated with WPPSI-III composite score and individual subtest scores. These findings demonstrate adverse effects of PBDE serum exposure on preschool maturity of children, and PBDEs potentially negative impact on child neuropsychological development.

Levels of polybrominated diphenyl ethers in the Korean metropolitan population are declining: A trend from 2001 to 2013.

Polybrominated diphenyl ethers (PBDEs) have been banned or voluntarily withdrawn from commerce worldwide. Declining levels of PBDEs in humans have been reported elsewhere, but not in Korea. We monitored 7 individual PBDE congeners (BDE-28, BDE-47, BDE-99, BDE-100, BDE-153, BDE-154, and BDE-183) in 103 human serum samples collected in 2006. The arithmetic mean and median values for the sum of the 7 PBDEs (∑PBDEs) were 7.13 and 6.70 ng/g lipid, respectively. In addition, 6 pooled human serum samples collected in 2006, 2007, 2009, 2011, 2012, and 2013 were analyzed. A decrease in PBDE levels was observed, as reflected by the median value of ∑PBDEs from 5.98 in 2006 to 2.98 ng/g lipid in 2013 as well as in the pooled samples. The levels of individual congeners also decreased but at different declining rates. Based on these results and previously reported data, a definite decline in PBDE levels in the Korean metropolitan population could be observed from 2001 to 2013, providing the first evidence in Korea of the same decline observed elsewhere in the world. Dietary exposure to a binary mixture of polybrominated diphenyl ethers alters innate immunity and disease susceptibility in juvenile Chinook salmon (Oncorhynchus tshawytscha)

Polybrominated diphenyl ethers (PBDEs) have been used as flame retardants in consumer products and are now found in the aquatic environment. The presence of PBDEs puts the health and survival of aquatic species at risk due to the various toxic effects associated with exposure to these compounds. The effects of a binary dietary mixture of PBDEs on innate immunity and disease susceptibility of juvenile Chinook salmon (Oncorhynchus tshawytscha) were examined in the present study. Salmon were fed roughly 1:1 mixtures of two environmentally predominant PBDE congeners, BDE-47 and BDE-99. The six resulting whole body total PBDE concentrations ranged from less than the limit of quantification to 184 ng/g, wet weight (ww). The innate immune system was assessed by using two in vitro macrophage function assays. Specifically, assays that examined the ability of head kidney macrophages to: (1) engulf sheep red blood cells (SRBCs); and (2) produce a respiratory burst, as determined by the production of a reactive oxygen species, superoxide anion. Macrophages from salmon fed the BDE-47/99 mixture diets engulfed more SRBCs and produced greater superoxide anion than salmon fed the control diet. An increase in macrophage function was observed in fish with whole body total PBDE concentrations ranging from 2.81 ng/g, ww to 184 ng/g, ww. The mechanism for this increase in macrophage function due to PBDE exposure is currently unknown, but may be due to the ability of PBDEs to act as an endocrine receptor agonist and/or antagonist. Salmon exposed to the BDE-47/99 mixture diets were also challenged with the pathogenic bacteria, Vibrio (Listonella) anguillarum to determine disease susceptibility. Kaplan-Meier survival curves of fish exposed to the BDE-47/99 mixture and control diets were significantly different. The Cox proportional hazard risk ratios of disease-induced mortality in juvenile Chinook salmon with whole body concentrations of total PBDEs of 10.9, 36.8, and 184 ng/g, ww were significantly greater than the fish fed the control diet by 1.56, 1.83 and 1.50 times, respectively. Not all concentrations of the binary mixture diets had significant hazard ratios relative to the control diet, due to a non-monotonic concentration response curve. The mixture of PBDE congeners resulted in interactive effects that were generally non-additive and dependent upon the congener concentrations and metric examined. Consequently, predicting the interactive effects in juvenile Chinook salmon exposed to mixtures of PBDE congeners on innate immunity and disease susceptibility cannot be readily determined from the adverse effects of individual PBDE congeners.

Adipose tissue levels of polybrominated diphenyl ethers and breast cancer risk in Chinese women: A case–control study

Polybrominated diphenyl ethers (PBDEs) are suspected to be associated with breast cancer risk because of their estrogenic potencies. Epidemiological studies of PBDEs and breast cancer are scarce. Our study aimed to estimate the association between adipose-tissue PBDE concentrations and breast cancer risk. A total of 209 breast cancer cases and 165 controls were recruited from hospitals between January 2014 and May 2016 in Shantou, Chaoshan area, China. Concentrations of 14 PBDE congeners were measured in adipose tissues obtained from the breast for cases and the abdomen/breast for controls during surgery. Demographic and clinicopathologic characteristics were obtained from medical records. Breast cancer risk as well as clinicopathologic characteristics were evaluated by adipose-tissue PBDE level. Odds ratios (ORs) and 95% confidence intervals (95% CIs) for breast cancer risk associated with levels of PBDE congeners were estimated from logistic regression models for all cases and stratified by estrogen receptor (ER) status. Level of total PBDEs (∑PBDE) and most individual PBDE congeners were higher in breast cancer cases than controls (median ∑PBDE, 94.99 vs 73.72 ng/g lipid). In the adjusted univariate model for all cases, breast cancer risk was increased with both 2nd and 3rd tertiles versus the 1st tertile of BDE-47 level (OR 2.05 [95% CI 1.08–3.92]; 5.47 [2.96–10.11]) and BDE-209 level (2.48 [1.30–4.73]; 4.72 [2.52–8.83]) with trend (both P < 0.001) and with the 3rd tertile of BDE-28 level (2.83 [1.63–4.92]), BDE-99 (3.22 [1.85–5.60]), BDE-100 (5.45 [2.90–10.23]), BDE-138 (2.40 [1.37–4.20]), BDE-153 (1.74 [1.02–2.97]), BDE-154 (1.84 [1.05–3.22]), and ∑PBDE levels (1.83 [1.07–3.14]) but decreased with the 3rd tertile of BDE-71 level (0.38 [0.22–0.65]) with trend (all P < 0.01). After stratifying by ER-positive or -negative status, the adjusted results were similar for ER-positive patients except for BDE-153 and BDE-154, with no statistical significance. In the multivariate model for all cases, age, menarche age, BDE-47, 71, 99, 100, 183 and 209 were independent factors associated with breast-cancer risk. ∑PBDE and most individual PBDE congeners investigated were positively associated with breast cancer risk in women from the Chaoshan area, China. PBDE may play a role in the occurrence and development of breast cancer.

Polybrominated Diphenyl Ether (PBDE) Accumulation in Farmed Salmon Evaluated Using a Dynamic Sea-Cage Production Model.

Food is an important source of human exposure to hazardous chemicals. Chemical concentration in a food item depends on local environmental contamination, production conditions, and, for animal-derived foods, on feed. Here, we investigate these influences on the accumulation of individual polybrominated diphenyl ether congeners (PBDEs) in farmed Atlantic salmon ( Salmo salar). We develop a dynamic model over a full sea-cage salmon production cycle. To assess the influence of metabolic debromination on PBDE congener profiles, in vitro measurements of debromination rates in fish liver cells were extrapolated to whole-body metabolic rate constants. Model results indicate that the dominant factors governing PBDE concentration in Atlantic salmon fillet are uptake via contaminated feed and fish growth, whereas the influence of metabolic debromination is minor. PBDE concentrations in fish feed depend on several factors, including the geographic origin of fish feed ingredients, which are produced and traded globally. Human exposure to PBDE via salmon consumption is less influenced by environmental concentrations at the location of salmon farming than by environmental concentrations influencing feed components. This dependence of PBDE concentrations in salmon on the origin and composition of feed reveals the complexity of predicting contaminant concentrations in globally traded food.

From the Cover: BDE-47 and BDE-49 Inhibit Axonal Growth in Primary Rat Hippocampal Neuron-Glia Co-Cultures via Ryanodine Receptor-Dependent Mechanisms.

Polybrominated diphenyl ethers (PBDEs) are widespread environmental contaminants associated with adverse neurodevelopmental outcomes in children and preclinical models; however, the mechanisms by which PBDEs cause developmental neurotoxicity remain speculative. The structural similarity between PBDEs and nondioxin-like (NDL) polychlorinated biphenyls (PCBs) suggests shared toxicological properties. Consistent with this, both NDL PCBs and PBDEs have been shown to stabilize ryanodine receptors (RyRs) in the open configuration. NDL PCB effects on RyR activity are causally linked to increased dendritic arborization, but whether PBDEs similarly enhance dendritic growth is not known. In this study, we quantified the effects of individual PBDE congeners on not only dendritic but also axonal growth since both are regulated by RyR-dependent mechanisms, and both are critical determinants of neuronal connectivity. Neuronal-glial co-cultures dissociated from the neonatal rat hippocampus were exposed to BDE-47 or BDE-49 in the culture medium. At concentrations ranging from 20 pM to 2 µM, neither PBDE congener altered dendritic arborization. In contrast, at concentrations ≥ 200 pM, both congeners delayed neuronal polarization resulting in significant inhibition of axonal outgrowth during the first few days in vitro. The axon inhibitory effects of these PBDE congeners occurred independent of cytotoxicity, and were blocked by pharmacological antagonism of RyR or siRNA knockdown of RyR2. These results demonstrate that the molecular and cellular mechanisms by which PBDEs interfere with neurodevelopment overlap with but are distinct from those of NDL PCBs, and suggest that altered patterns of neuronal connectivity may contribute to the developmental neurotoxicity of PBDEs.

Mechanistic studies to evaluate the size dependent toxicity of titanium dioxide nanoparticles (TiO2 NPs) on the soil nematode, Caenorhabditis elegans

In recent years, polybrominated diphenyl ethers (PBDEs) are ubiquitous environmental contaminants, but its bioaccumulation and debromination in biota have remained largely unclear. In this study, we analyzed six PBDEs (BDE47, 99, 100, 153, 154, and 209) in various tissues (i.e., viscera, muscle, and gill) of 11 types of marine organisms including zooplankton, invertebrate and fish. The concentrations of six PBDE including BDE209 in marine organisms ranged from 0.75 to 7.29 ng/g dry weight, BDE209 from 0.46 to 6.78 ng/g dry weight, respectively. BDE209 was the dominant congener in all samples, followed by BDE47. The concentration ratio of BDE47, 99, 154 to ΣPBDEs in various tissues of organisms (i.e., Rapana venosa, shrimp, crab, cuttlefish, octopus, Synechogobius hasta, tonguefish and wolfish) increased, while the concentration ratio of BDE209 to ΣPBDEs decreased. Large differences of the concentration ratios between BDE99 and BDE100 in tissues of crab was found, ranging from 32:68 in crab viscera to 83:17 in crab leg muscle. Biomagnification factors for individual PBDE congeners ranged from 0.16 to 78.6. In general, the BMFs for BDE209 in muscle were higher than those in viscera within feeding relationships. The study results suggesting BDE209 can be biodegraded to BDE47 through BDE154 and BDE99 in marine organism, its metabolite importantly influenced by organism type not trophic level; higher percentage of BDE154 was found in viscera than that in other tissues in the analyzed marine organisms of Bohai Bay.

Development of an analytical method to quantify PBDEs, OH-BDEs, HBCDs, 2,4,6-TBP, EH-TBB, and BEH-TEBP in human serum.

Polybrominated diphenyl ethers (PBDEs) flame retardants (FRs) were phased-out in the mid-2000s (penta- and octaBDE) and 2013 (decaBDE); however, their hydroxylated metabolites (OH-BDEs) are still commonly detected in human serum. Today, novel FRs such as Firemaster® 550, a mixture that contains two brominated compounds, EH-TBB and BEH-TEBP are used as replacements for PBDEs in some applications, and there is a need to develop a comprehensive analytical method to assess exposure to both legacy PBDEs and novel FRs. This study developed a solid-phase extraction (SPE)-based method to analyze PBDEs, OH-BDEs, 2,4,6-tribromophenol (TBP), hexabromocylcododecane isomers (HBCDs), EH-TBB, and BEH-TEBP in human serum. Briefly, serum proteins were first denatured with formic acid, and then the target analytes were isolated using a SPE column. Finally, the extract was cleaned and fractioned using a silica SPE column. Method performance was assessed by spiking fetal bovine serum with 1-2ng of the target analytes, and method accuracy was quantified by comparison to a serum Standard Reference Material (SRM). The developed method showed good recovery and accuracy for all target analytes with the exception of the very low and very high molecular weight PBDE congeners. Using this method, 43 serum samples collected from the Healthy Pregnancy, Healthy Baby Study (HPHB) cohort in Durham, NC, USA were analyzed for FRs. A novel finding was the ubiquitous detection of 2,4,6-TBP, at levels greater than the individual PBDE congeners. Furthermore, 2,4,6-TBP was positively correlated with PBDEs, suggesting that they may have a similar source of exposure, or that 2,4,6-TBP may result from metabolism of PBDEs in vivo.

Occurrence and bioaccumulation of polybrominated diphenyl ethers in sediments and paddy ecosystems of Liaohe River Basin, northeast China

Concentrations of 16 polybrominated diphenyl ether (PBDE) congeners were measured in river sediments, paddy soils and three species of paddy-field organisms (crab, loach and carp) collected from the Liaohe River Basin, northeastern China. The total contents of PBDEs (∑16PBDEs) in sediments and paddy soils were in the ranges of 273.4–3246.3pg/g dry weight (dw), and 192.1–1783.8pg/gdw, respectively. BDE 209 was the dominant congener both in sediments and paddy soils. The concentrations of ∑16PBDEs in sediments were significantly higher than those in the adjacent paddy soils, indicating a potential transport of PBDEs from river to paddy ecosystems via river water irrigation. The biota–soil accumulation factor (BSAF) was calculated as the ratio between the lipid-normalized concentration in paddy-field organisms and the total organic carbon-normalized concentration in paddy soil. The average BSAF values of ∑15PBDEs followed the sequence of crab (3.6)>loach (3.3)>carp (2.1). BDE 154 had the highest BSAF value, and a parabolic trend between BSAF values of individual PBDE congeners and their logKOW values was observed. In view of the fact that crab had the larger BSAF value and higher lipid content, the ecological risk and health risk for crab cultivation in paddy fields should be of particular concern.

Dietary Exposure to Individual Polybrominated Diphenyl Ether Congeners BDE-47 and BDE-99 Alters Innate Immunity and Disease Susceptibility in Juvenile Chinook Salmon.

Polybrominated diphenyl ethers (PBDEs), used as commercial flame-retardants, are bioaccumulating in threatened Pacific salmon. However, little is known of PBDE effects on critical physiological functions required for optimal health and survival. BDE-47 and BDE-99 are the predominant PBDE congeners found in Chinook salmon collected from the Pacific Northwest. In the present study, both innate immunity (phagocytosis and production of superoxide anion) and pathogen challenge were used to evaluate health and survival in groups of juvenile Chinook salmon exposed orally to either BDE-47 or BDE-99 at environmentally relevant concentrations. Head kidney macrophages from Chinook salmon exposed to BDE-99, but not those exposed to BDE-47, were found to have a reduced ability in vitro to engulf foreign particles. However, both congeners increased the in vitro production of superoxide anion in head kidney macrophages. Salmon exposed to either congener had reduced survival during challenge with the pathogenic marine bacteria Listonella anguillarum. The concentration response curves generated for these end points were nonmonotonic and demonstrated a requirement for using multiple environmentally relevant PBDE concentrations for effect studies. Consequently, predicting risk from toxicity reference values traditionally generated with monotonic concentration responses may underestimate PBDE effect on critical physiological functions required for optimal health and survival in salmon.

Neurobehavioral function and low-level exposure to brominated flame retardants in adolescents: A cross-sectional study

Background: Animal and in vitro studies demonstrated a neurotoxic potential of brominated flame retardants, a group of chemicals used in many household and commercial products to prevent fire. Although the first reports of detrimental neurobehavioral effects in rodents appeared more than ten years ago, human data are sparse. Methods: As a part of a biomonitoring program for environmental health surveillance in Flanders, Belgium, we assessed the neurobehavioral function with the Neurobehavioral Evaluation System (NES-3), and collected blood samples in a group of high school students. Cross-sectional data on 515 adolescents (13.6-17 years of age) was available for the analysis. Multiple regression models accounting for potential confounders were used to investigate the associations between biomarkers of internal exposure to brominated flame retardants [serum levels of polybrominated diphenyl ether (PBDE) congeners 47, 99, 100, 153, 209, hexabromocyclododecane (HBCD), and tetrabromobisphenol A (TBBPA)] and cognitive performance. In addition, we investigated the association between brominated flame retardants and serum levels of FT3, FT4, and TSH. Results: A two-fold increase of the sum of serum PBDE’s was associated with a decrease of the number of taps with the preferred-hand in the Finger Tapping test by 5.31 (95% CI: 0.56 to 10.05, p = 0.029). The effects of the individual PBDE congeners on the motor speed were consistent. Serum levels above the level of quantification were associated with an average decrease of FT3 level by 0.18 pg/mL (95% CI: 0.03 to 0.34, p = 0.020) for PBDE-99 and by 0.15 pg/mL (95% CI: 0.004 to 0.29, p = 0.045) for PBDE-100, compared with concentrations below the level of quantification. PBDE-47 level above the level of quantification was associated with an average increase of TSH levels by 10.1% (95% CI: 0.8% to 20.2%, p = 0.033), compared with concentrations below the level of quantification. We did not observe effects of PBDE’s on neurobehavioral domains other than the motor function. HBCD and TBBPA did not show consistent associations with performance in the neurobehavioral tests. Conclusions: This study is one of few studies and so far the largest one investigating the neurobehavioral effects of brominated flame retardants in humans. Consistently with experimental animal data, PBDE exposure was associated with changes in the motor function and the serum levels of the thyroid hormones.

Tree bark as a passive air sampler to indicate atmospheric polybrominated diphenyl ethers (PBDEs) in southeastern China

The different barks were sampled to discuss the influence of the tree species, trunk circumference, and bark thickness on the accumulation processes of polybrominated diphenyl ethers (PBDEs) from air into the bark. The results of different PBDE concentrations indicated that barks with a thickness of 0-3mm collected from weeping willow, Camphor tree, and Masson pine, the trunk circumferences of which were 100 to 150cm, were better PBDEs passive samplers. Furthermore, tree bark and the corresponding air samples were collected at Anji (AJ), Hangzhou (HZ), Shanghai (SH), and Wenling (WL) to investigate the relationship between the PBDE concentrations in bark and those in air. In addition, the significant correlation (r (2) = 0.906; P < 0.05) indicated that atmospheric PBDEs were the principle source for the accumulation of PBDEs in the barks. In this study, the log K BA (bark-air partition coefficient) of individual PBDE congeners at the four sites were in the range from 5.69 to 6.79. Finally, the total PBDE concentration in WL was 5 to 20 times higher than in the other three cities. The result indicated that crude household workshops contributed a heavy amount of PBDEs pollution to the environment, which had been verified by the spatial distribution of PBDEs levels in barks collected at Wenling (range, 26.53-1317.68ng/g dw). The good correlation between the PBDE concentrations in the barks and the air samples and the variations of the PBDE concentrations in tree barks collected from different sites reflected that the bark could be used as a passive sampler to indicate the atmospheric PBDEs.

A national probabilistic study of polybrominated diphenyl ethers in fish from US lakes and reservoirs

Polybrominated diphenyl ethers (PBDEs) are persistent, bioaccumulative, and toxic chemicals that are present in air, water, soil, sediment, and biota (including fish). Most previous studies of PBDEs in fish were spatially focused on targeted waterbodies. National estimates were developed for PBDEs in fish from lakes and reservoirs of the conterminous US (excluding the Laurentian Great Lakes) using an unequal probability design. Predator (fillet) and bottom-dweller (whole-body) composites were collected during 2003 from 166 lakes selected randomly from the target population of 147,343 lakes. Both composite types comprised nationally representative samples that were extrapolated to the sampled population of 76,559 and 46,190 lakes for predators and bottom dwellers, respectively. Fish were analyzed for 34 individual PBDE congeners and six co-eluting congener pairs representing a total of 46 PBDEs. All samples contained detectable levels of PBDEs, and BDE-47 predominated. The maximum aggregated sums of congeners ranged from 38.3 ng/g (predators) to 125 ng/g (bottom dwellers). Maximum concentrations in fish from this national probabilistic study exceeded those reported from recent targeted studies of US inland lakes, but were lower than those from Great Lakes studies. The probabilistic design allowed the development of cumulative distribution functions to quantify PBDE concentrations versus the cumulative number of US lakes from the sampled population.

Polybrominated Diphenyl Ethers in Air across China: Levels, Compositions, and Gas-Particle Partitioning

Air samples were concurrently collected using high volume air samplers for 24 h every week from September 2008 to August 2009 at 15 sites (11 urban, 1 suburban, and 3 background/rural) across China. Twelve polybrominated diphenyl ether (PBDE) congeners (BDE-17, -28, -47, -66, -85, -99, -100, -138, -153, -154, -183, and -209) were measured. Total PBDE concentrations (∑12PBDEs) in air (gas + particle phases) were in the range of 11.0-838 pg m(-3) with a mean of 232 ± 72 (mean ± SE) pg m(-3). The site with the highest concentration was Guangzhou (838 ± 126 pg m(-3)), followed by Beijing (781 ± 107 pg m(-3)). Significant positive correlations were found between PBDEs levels and urban population (R = 0.69, P < 0.05) and gross industrial output values (R = 0.87, P < 0.001) as well. BDE-209 was the dominating congener with the contribution of 64 ± 23% to ∑12PBDEs, followed by BDE-47(8 ± 8%) and -99(6 ± 5%) at all urban and suburban sites. At background/rural sites, however, BDE-47 was the dominating congener, followed by BDE-99, together accounting for 52 ± 21% of ∑12PBDEs, while BDE-209 was only 11 ± 2%. It was found that PBDEs at the 15 sites showed a primary distribution and fractionation pattern. This study produced more than 700 pairs of air samples in gaseous and particulate phases with a wide temperature range of ∼60 °C, providing a good opportunity to investigate gas-particle partitioning for individual PBDE congeners. The results of gas-particle partitioning analysis for PBDEs using both subcooled-liquid-vapor pressure (PL)-based and octanol-air partition coefficient (KOA)-based models indicated that PBDEs in air at all sampling sites had not reached equilibrium because the slope values (mO) in the KOA-based equation and the opposite slope values (mP) in the PL-based equation at all 15 sampling sites were less than 1. It also found that both mO and -mP were significantly and positively correlated with the annual average temperatures of sampling sites and also significantly and negatively correlated with the mole masses of PBDE congeners, indicating a general trend that the higher the temperature at the sampling site and the lower the mole mass of the PBDE congeners are, the closer to the equilibrium the congeners approach and vice versa. To our knowledge, this is the first study to report the correlations of the slope values for both the KOA-based and PL-based equations with temperatures at sampling sites and mole masses for individual PBDE congeners.

Polybrominated diphenyl ethers in humans - sources and exposure

Polybrominated diphenyl ethers (PBDEs) are a class of widely used flame retardants that are incorporated into a wide range of consumer products such as household appliances, plastics, textiles and computers, to prevent fire. They were first introduced in the environment in the 1970s and their concentrations have been increasing ever since. Their persistence in the environment, widespread distribution and bioaccumulation in humans and wildlife has rendered them chemicals of concern. The main route of entry of these chemicals into the human body is via the food web, but occupational exposure may also occur in the workplace during handling, repair and dismantling of flame retarded goods. Inhalation of indoor air and dermal uptake may also be another important route of entry of PBDEs into humans. PBDEs structural similarities to better known and studied chemicals like PCBs, are causes for concern. Individual PBDE congeners have been associated with neurotoxic effects following neonatal exposure in animals and effects on thyroid hormone function. PBDEs have also been associated with non-Hodgkin’s lymphoma in humans, teratogenicity and fetal toxicity. There are still many toxicity gaps, including their carcinogenic potential and human health effects. This paper reviews the sources and exposure of humans to PBDEs, highlighting recent scientific data.
