Effect of Yarrowia lipolytica yeast biomass with increased kynurenic acid content on selected metabolic indicators in mice.

The unconventional yeast species Yarrowia lipolytica is a valuable source of protein and many other nutrients. It can be used to produce hydrolytic enzymes and metabolites, including kynurenic acid (KYNA), an endogenous metabolite of tryptophan with a multidirectional effect on the body. The administration of Y. lipolytica with an increased content of KYNA in the diet may have a beneficial effect on metabolism, which was evaluated in a nutritional experiment on mice. In the dry biomass of Y. lipolytica S12 enriched in KYNA (high-KYNA yeast) and low-KYNA (control) yeast, the content of KYNA was determined by high-performance liquid chromatography. Then, proximate and amino acid composition and selected indicators of antioxidant status were compared. The effect of 5% high-KYNA yeast content in the diet on the growth, hematological and biochemical indices of blood and the redox status of the liver was determined in a 7-week experiment on adult male mice from an outbred colony derived from A/St, BALB/c, BN/a and C57BL/6J inbred strains. High-KYNA yeast was characterized by a greater concentration of KYNA than low-KYNA yeast (0.80 ± 0.08 vs. 0.29 ± 0.01 g/kg dry matter), lower content of crude protein with a less favorable amino acid composition and minerals, higher level of crude fiber and fat and lower ferric-reducing antioxidant power, concentration of phenols and glutathione. Consumption of the high-KYNA yeast diet did not affect the cumulative body weight gain per cage, cumulative food intake per cage and protein efficiency ratio compared to the control diet. A trend towards lower mean corpuscular volume and hematocrit, higher mean corpuscular hemoglobin concentration and lower serum total protein and globulins was observed, increased serum total cholesterol and urea were noted. Its ingestion resulted in a trend towards greater ferric-reducing antioxidant power in the liver and did not affect the degree of liver lipid and protein oxidation. The improvement of the quality of Y. lipolytica yeast biomass with increased content of KYNA, including its antioxidant potential, would be affected by the preserved level of protein and unchanged amino acid profile. It will be worth investigating the effect of such optimized yeast on model animals, including animals with metabolic diseases.

Pro-and Antioxidant Status in Newborn with COVID-19.

There is practically no information on the state of oxidative stress reactions in newborns with coronavirus infections. At the same time, such studies are extremely important and can contribute to better understanding of the process of reactivity in patients of different ages. The content of pro- and antioxidant status indicators was assessed in 44 newborns with confirmed COVID-19. It was found that the content of compounds with unsaturated double bonds, primary, secondary, and final LPO products were elevated in newborns with COVID-19. These changes were accompanied by higher SOD activity and retinol level and reduced activity of glutathione peroxidase. Contrary to popular opinion, newborns can be a COVID-19-susceptible age group and require more close monitoring of metabolic reactions during the period of neonatal adaptation that is an aggravating background during infection.

Evaluation of the preventive potential of graded dietary inclusion of Hyphaene thebaica (Linn) fruit in rat model of colon carcinogenesis.

The preventive effect of Hyphaene thebaica fruit in colon carcinogenesis was evaluated in Wistar rats at 0, 2.5, 5 and 10% inclusion rates for twelve weeks with concomitant 72-h intra-rectal N-methyl-N-nitrosourea (MNU) instillations. Indices of antioxidant status and carcinogenesis were analyzed using spectrophotometric, ELISA, histological and immunohistochemical techniques. The fruit protected against lipid peroxidation and level of early biomarkers of colon carcinogenesis, accompanied by decrease in some endogenous antioxidant enzymes functionality. It also prevented colon tissues against MNU-induced severe inflammations and damage to the mutL-homolog 1 (MLH1) gene. There was significant negative correlation between endogenous antioxidant enzyme activities and carcinoembryonic antigen (CEA) as well as lipid peroxidation, but relationship between total polyphenols and percentage expression of MLH1 proteins as well as endogenous antioxidant enzyme activities was positive. These results validate the folkloric use of H. thebaica fruit in the management of colorectal disorders. PRACTICAL APPLICATIONS: Hyphaene thebaica fruit which is widely consumed in northern Nigeria and other countries of sub-Saharan Africa is rich in fiber and antioxidant polyphenols. These two classes of compounds have demonstrated capacity to prevent colorectal cancer and cancer of other sites. Therefore, the validated protective Hyphaene thebaica fruit suggests that it can be processed for inclusion in beverages/diets as functional foods for prevention and management of colorectal disorders.

The Effect of Maternal Dietary Selenium Supplementation on Blood Antioxidant and Metabolic Status of Ewes and Their Lambs.

This study investigated the effect of dietary selenium supplementation (organic and inorganic) of late-gestation ewes on blood selenium concentrations and metabolic and antioxidant status indicators in ewes and their lambs. In addition, the efficacy of selenium transfer from ewes to lambs during the suckling period was determined. The study was conducted on 30 Merinolandschaf ewes and their lambs and lasted four months. The feed mixture of the control group (group I) contained no added selenium, while the feed mixture of group II was enriched with 0.3 mg/kg of organic selenium sources and the third group with 0.3 mg/kg of inorganic selenium sources. In ewes and their lambs, selenium supplementation significantly (p < 0.01; p < 0.05) increased selenium concentration, glutathione peroxidase, and superoxide dismutase activity and decreased malondialdehyde concentration compared to the control group. Selenium supplementation had a positive effect on metabolism and hematological indicators in lambs. A positive correlation was found between antioxidant indicators in the whole blood of ewes and lambs. The good transfer of selenium from ewes to lambs was complemented by higher correlation coefficients when the feed mixture was supplemented with organic compared to inorganic selenium.

Metabolic Profile of Sow Blood Serum after Weaning.

The aim of our research was to determine the content of protein, carbohydrate, lipid, and mineral metabolites, as well as an antioxidant status of the sow's blood after weaning and to calculate the correlation between these parameters. The experiment was carried out on twenty clinically healthy crossbred sows (Yorkshire × Landrace). Twenty sows were allocated to one of two groups: (1) 1 day after weaning (group 1, n = 10) and (2) 8 days after weaning (group 2, n = 10). The basis of the sow diet was SK-1 compound feed, balanced in terms of nutrients and energy in accordance with modern standards and the recommended feeding regimen. Sows blood samples were taken and analyzed for the metabolites of nitrogenous, carbohydrate-lipid, and mineral metabolism and indicators of antioxidant status. The results showed that, in group 2, the total protein content was 89.07 g/l, which is 10.2% higher than that in group 1 (p < 0.05); it was mainly achieved due to the globulin fraction. The urea increased by 19.1% (p < 0.05), but the concentrations of magnesium and chlorides decreased by 20.2% (p < 0.01) and 5.43% (p < 0.05), as well as the alkaline phosphatase and ALT activities decreased by 42.5% (p < 0.05). Strong positive correlations of the ceruloplasmin with total protein (0.672) and very strong with globulins (0.780) were observed. There was a strong negative correlation between the AST activity and the TBA-AP content, as well as the values of phospholipids and TAWSA. There are moderate negative correlations of the TBA-AP with magnesium, TAWSA and ALT activity, and moderate positive correlations of the TBA-AP with total protein, albumin, triglycerides, and cholesterol. The revealed tendencies and dependencies will serve as the theoretical basis for the development of practical methods for regulating the level of free-radical reactions.

Alleviation of Dyslipidemia via a Traditional Balanced Korean Diet Represented by a Low Glycemic and Low Cholesterol Diet in Obese Women in a Randomized Controlled Trial.

A traditional balanced Korean diet (K-diet) may improve energy, glucose, and lipid metabolism. To evaluate this, we conducted a randomized crossover clinical trial, involving participants aged 30–40 years, who were randomly assigned to two groups—a K-diet or westernized Korean control diet daily, with an estimated energy requirement (EER) of 1900 kcal. After a 4-week washout period, they switched the diet and followed it for 4 weeks. The carbohydrate, protein, and fat ratios based on energy intake were close to the target values for the K-diet (65:15:20) and control diet (60:15:25). The glycemic index of the control diet and the K-diet was 50.3 ± 3.6 and 68.1 ± 2.9, respectively, and daily cholesterol contents in the control diet and K-diet were 280 and 150 mg, respectively. Anthropometric and biochemical parameters involved in energy, glucose, and lipid metabolism were measured while plasma metabolites were determined using UPLC-QTOF-MS before and after the 4-week intervention. After the four-week intervention, both diets improved anthropometric and biochemical variables, but the K-diet significantly reduced them compared to the control diet. Serum total cholesterol, non-high-density lipoprotein cholesterol, and triglyceride concentrations were significantly lower in the K-diet group than in the control diet group. The waist circumference (p = 0.108) and insulin resistance index (QUICKI, p = 0.089) tended to be lower in the K-diet group than in the control diet group. Plasma metabolites indicated that participants in the K-diet group tended to reduce insulin resistance compared to those in the control diet group. Amino acids, especially branched-chain amino acids, tyrosine, tryptophan, and glutamate, and L-homocysteine concentrations were considerably lower in the K-diet group than in the control diet group (p < 0.05). Plasma glutathione concentrations, an index of antioxidant status, and 3-hydroxybutyric acid concentrations, were higher in the K-diet group than in the control diet group. In conclusion, a K-diet with adequate calories to meet EER alleviated dyslipidemia by decreasing insulin resistance-related amino acids and increasing ketones in the circulation of obese women.

Changes in blood antioxidant status in American football players and soccer players over a training macrocycle

BackgroundThe effectiveness of sport training programs should be assessed regularly against biochemical indices. This study assesses changes in the antioxidant status indices in American football players (AF) and soccer players (SP) over a training macrocycle. MethodsThe study was carried out with Poland's American Football League players (AF, n = 11, age 24.0 ± 3.7 years) and first-league soccer players (SP, n = 11, age 26.5 ± 3.8 years). Resting venous blood samples were collected from the players at the beginning of the three periods (preparatory, competition, and transition) making up the training macrocycle to determine the activity levels of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT), glutathione reductase (GR), creatine kinase (CK), and lactate dehydrogenase (LDH), as well as the concentrations of non-enzymatic antioxidants (uric acid-UA and glutathione-GSH) and the levels of malondialdehyde (MDA). ResultsThe period effect on SOD (p < 0.001), CAT (p < 0.05), GPx (p < 0.05), GSH (p < 0.0001) and UA (p < 0.0001), and the group × period interaction effect on SOD, CAT and GPx (p < 0.05), GSH (p < 0.001), and UA (p < 0.01) proved to be significant. Also significant were the group effect on MDA (p < 0.001) and LDH (p < 0.0001) and the period effect on MDA (p < 0.01) and LDH (p < 0.001). The activity of SOD and CAT and the concentration of GSH were higher in both AF (12%, 2%, and 15%, respectively) and SP (33%, 10%, and 42%) at the start of the competition period than in the preparatory period, but the concentration of MDA and the activity of CK and LDH was lower (0.8%, 29%, 5% (AF) and 2%, 11%, 5% (SP). The highest activity of GPx and LDH and the greatest concentrations of UA and MDA occurred in the early transition period. ConclusionThe study revealed an association between American footballers' and soccer players’ training loads in the preparatory period and moderate improvements in their blood antioxidant status at the beginning of the competition period.

Efficacy of using a complex of minerals and vitamins for prevention of complications in bone tissue and the digestive tract in rats with hypothyroidism

Significant consequences of hypothyroidism are impairments in all types of metabolism, including bone metabolism, pathological changes in the digestive tract and the development of deficiency of vitamins, macro-and micronutrients.The relevance of the study is due to the widespread occurrence of hypothyroidism and the feasibility of developing effective methods for the prevention of the pathology and its complications. The study was aimed at the condition of bone tissue and digestive tract in rats with hypothyroidism, as well as the effectiveness of preventive addition of a complex of vitamins and minerals. The studies were performed on rats with hypothyroidism that was caused using thyrostatic mercazolyl for 50 days, administered orally. Prophylaxis was performed by adding a prophylactic complex of vitamins P, C, D and minerals Ca, Mg, Cu, Se, Mn to the daily diet. In the bone tissue of rats, the indicators of destruction (elastase and acid phosphatase activity) and mineralization (alkaline phosphatase activity, calcium content) as well as the antioxidant state (activities of catalase, glutathione reductase, malonic dialdehyde content) were studied. In the gums and mucous membranes of the digestive tract of rats, we determined indicators of antioxidant status (catalase activity, malonic dialdehydecontent), inflammation (activities of elastase, acid phosphatase) and dysbiosis (activities of urease activity, lysozyme). In the bone tissue of rats with hypothyroidism, activation of destruction processes, reduction of calcium levels, activation of lipid peroxidation and antioxidant enzymes were found. Hypothyroidism also led to pathological disorders in the gums of rats and gastric mucosa, small and large intestine, namely – the development of inflammation, increased membrane permeability, activation of lipid peroxidation, increased contamination with opportunistic bacteria against the background of reduced antioxidant protection. In general, preventive use of the complex contributed to the normalization of the studied parameters in the gums, mucous membranes of the stomach, intestines, as well as remodeling processes and antioxidant-prooxidative state in the bone tissue of animals with hypothyroidism. In the conditions of hypothyroidism, the proposed complex of vitamins and minerals provided notable antioxidant, anti-inflammatory and antidysbiotic effects in the tissues of the gastrointestinal tract, thus overcoming the calcium deficiency in the blood and stopping the destructive and oxidative processes in the bone tissue of animals with hypothyroidism.

The Analysis of Intestinal Lavage Method Efficiency in Patients with Chronic Pancreatitis Exacerbation

The clinical picture in patients with chronic pancreatitis (CP) which develops due to acute pancreatitis (AP) (in month after attack of AP) is characterized by intestinal insufficiency syndrome (IIS). The multifactorial mechanism of IIS in CP as a result of AP requires direct attention of the physician on the elimination of the pathological symptoms, which are the main cause of malabsorption, trophological insufficiency, abdominal discomfort and pain, and as consequence, reduced quality of life and social adaptation.The objective: to investigate the dynamic of the clinical signs, anthropometric parameters and pro- and antioxidant status indicators after treatment by proposed treatment with the intestinal lavage course in patients with chronic pancreatitis which is as result of acute pancreatitis.Materials and methods. The results of investigation and outpatient treatment of 34 patients with CP (complicated by IIS) as a result of AP were analyzed.Results. It was established the effectiveness of the optimized treatment complex at the level of MA – 39,55 %, CM254 – 8,85 %, CM280 – 7,09 %, SOD – 7,75 %, SH-groups – 5,18 %, catalase – 17,78 %. The effectiveness of the optimized treatment complex by the level of PSA was 41,15 %, ferritin – 18,61 %, albumin – 15,13 %. The implementation of the proposed treatment course in the standart treatment of patients with chronic pancreatitis which developed due to acute pancreatitis has led to a significant more effective reduction of pain, improved coprogram results and normalization of endogenous intoxication indicators and positive changes antioxidant system.Conclusion. The conducted analysis of results showed that the parameters of C-reactive protein, albumin and ferritin can be used as markers of the intoxication expresion, severity of inflammation and nutrient deficiency, as well as determining the number of intestinal lavage procedures: when the content of CRP 3,0–7,0 mg/l, ferritin – 100,0–150,0 ng/ml, albumin – 35,0–32,0 g/l one lavage procedure was performed; with content of CRP – 7,0–30,0 mg/l, ferritin – 50,0–100,0 ng/ml, albumin – 32,0–29,0 g/l – 3 procedures were perfomed, with a CRP content of 30,0–50,0 mg/l, ferritin – 20,0–50,0 ng/ml, albumin ≤29,0 g/l 5 procedures of intestinal lavage were performed.

Association between genotoxic properties of inhalation anesthetics and oxidative stress biomarkers.

Exposure to inhalation anesthetics (IAs) has been associated with DNA damage as reflected in the increased frequency of micronuclei (MN) and chromosomal aberrations (CAs). The present study was undertaken to ascertain whether there was any correlation between increased MN and CA and the extent of oxidative stress as well as the antioxidant status of a group of operating room personnel exposed to a mixture of IAs, including nitrous oxide, isoflurane, and sevoflurane. In this cross-sectional study, 60 operating room personnel (exposed group) in whom the frequencies of MN and CA had already been shown to be significantly higher than those of a referent group, as well as 60 unexposed nurses, were studied. Venous blood samples were taken from all participants, and malondialdehyde (MDA) levels as an index of oxidative stress (OS) and the activity of superoxide dismutase (SOD) and levels of total antioxidant capacity (TAC) as indices of antioxidant status were measured. The level of TAC (1.76 ± 0.59 mM vs. 2.13 ± 0.64 mM, p = 0.001) and the activity of SOD (11.22 ± 5.11 U/ml vs. 13.36 ± 4.12 U/ml, p = 0.01) were significantly lower, while the mean value of MDA was significantly higher (2.46 ± 0.66 µM vs. 2.19 ± 0.68 µM, p = 0.03) in the exposed group than in the nonexposed group. After adjusting for potential confounders, there were statistically significant associations between exposure to IAs, gender, SOD, and TAC with MN frequency and between exposure to IAs and SOD with numbers of CA. The findings of the present study indicated that exposure to IAs was associated with OS, and this, in turn, may be causally linked with DNA damage.

Ethanolic Extract of Moringa oleifera Leaves Influences NF-κB Signaling Pathway to Restore Kidney Tissue from Cobalt-Mediated Oxidative Injury and Inflammation in Rats.

This study aimed to describe the protective efficacy of Moringa oleifera ethanolic extract (MOEE) against the impact of cobalt chloride (CoCl2) exposure on the rat’s kidney. Fifty male rats were assigned to five equal groups: a control group, a MOEE-administered group (400 mg/kg body weight (bw), daily via gastric tube), a CoCl2-intoxicated group (300 mg/L, daily in drinking water), a protective group, and a therapeutic co-administered group that received MOEE prior to or following and concurrently with CoCl2, respectively. The antioxidant status indices (superoxide dismutase (SOD), catalase (CAT), and reduced glutathione (GSH)), oxidative stress markers (hydrogen peroxide (H2O2), 8-hydroxy-2-deoxyguanosine (8-OHdG), and malondialdehyde (MDA)), and inflammatory response markers (nitric oxide (NO), tumor necrosis factor (TNF-α), myeloperoxidase (MPO), and C-reactive protein (CRP)) were evaluated. The expression profiles of pro-inflammatory cytokines (nuclear factor-kappa B (NF-kB) and interleukin-6 (IL-6)) were also measured by real-time quantitative polymerase chain reaction (qRT-PCR). The results showed that CoCl2 exposure was associated with significant elevations of oxidative stress and inflammatory indices with reductions in the endogenous tissue antioxidants’ concentrations. Moreover, CoCl2 enhanced the activity of the NF-κB inflammatory-signaling pathway that plays a role in the associated inflammation of the kidney. MOEE ameliorated CoCl2-induced renal oxidative damage and inflammatory injury with the suppression of the mRNA expression pattern of pro-inflammatory cytokine-encoding genes. MOEE is more effective when it is administered with CoCl2 exposure as a prophylactic regimen. In conclusion, MOEE administration exhibited protective effects in counteracting CoCl2-induced renal injury in rats.

Eucalyptus viminalis leaf extract alters the productivity and blood parameters of healthy broiler chickens

Background and Aim:As an alternative to natural and chemically synthesized direct-acting bactericides, there has been an increase in the use of plant extracts, which possess a set of phytochemicals with potential for microbial disease control; this is due to the spectrum of secondary metabolites present in extracts, which include phenolic compounds, quinones, flavonoids, alkaloids, terpenoids, and polyacetylenes. The biologically active substances within plant extracts, which perform protective functions for plant tissues, can have ambiguous effects on the animal body. Therefore, the aim of this study was to assess the ability of gamma-octalactone, isolated from Eucalyptus viminalis extract, to inhibit various LuxI/LuxR quorum-sensing (QS) systems in bacteria, and to evaluate its effect on broiler chickens.Materials and Methods:Phytochemical analysis of E. viminalis extract was performed. The ability of gamma-octalactone to inhibit QS was evaluated using four different LuxI/LuxR bacterial test systems. In vivo assessments were performed on one hundred and twenty 7-day-old broiler chickens (Arbor Acres cross), split into four groups of 30 chickens: 1. Control group: Basic diet (BD); 2. experimental Group I: BD + gamma-octalactone at a dosage of 0.05 ml/kg live weight/day; 3. experimental Group II: BD + gamma-octalactone at a dosage of 0.1 ml/kg live weight/day; and 4. experimental Group III: BD + gamma-octalactone at a dosage of 0.2 ml/kg live weight/day. Hematological blood parameters were assessed using an automatic hematological analyzer (URIT-2900 Vet Plus, URIT Medial Electronic Co., China) and an automatic biochemical analyzer (CS-T240, Dirui Industrial Co., Ltd., China). Statistical analyses were performed using SPSS Statistics Version 20 (IBM); averages (M), standard deviations (σ), and standard deviation errors (m) were calculated. Results with p≤0.05 were considered significant.Results:Based on the phytochemical analysis results, libraries of compounds with putative QS inhibitory properties were compiled. Gamma-octalactone exhibited a pronounced inhibitory effect on the LuxI/LuxR QS systems, characterized by EC50 values of 0.15-0.4 mM. In the in vivo portion of this study, broiler chicken live weights increased in all experimental groups, with the most significant increase in Group III (14.0%), in relation to the control group. Blood serum from the experimental group chickens had significantly higher levels of triglycerides and uric acid (p≤0.05), in comparison to the control group chickens. With respect to blood serum enzyme activity and antioxidant status indicators, the experimental group chickens had a higher level of gamma-glutamyl transferase, an enzyme associated with amino acid metabolism, than those in the control group; this increase was especially pronounced in Group III, with 37.0% increase (p≤0.05). Superoxide dismutase and catalase levels were higher in the experimental groups than the control group, corresponding to increases of 30.4-56.2% (p≤0.05), 33.3-83.3%, and 27.9-45.5% (p≤0.05) in Groups I, II, and III (p≤0.05), respectively. Morphological blood parameters did not display significant changes due to gamma-octalactone.Conclusion:According to the results of this in vivo study in broiler chickens, gamma-octalactone, isolated from E. viminalis leaf extract and supplied at a dosage of 0.2 ml/kg live weight/day, led to an increase in the activity of blood plasma digestive enzymes, increased live weight, and had a positive effect on lipid metabolism and antioxidant status.

Fatty Acid Profile and Antioxidant Status Fingerprint in Sarcopenic Elderly Patients: Role of Diet and Exercise.

Plasma fatty acids (FAs) and oxidant status contribute to the etiology of sarcopenia in the elderly concurring to age-related muscle loss and elderly frailty through several mechanisms including changes in FA composition within the sarcolemma, promotion of chronic low-grade inflammation, and insulin resistance. The aim of this study was to determine the FA profile and pro-antioxidant status in sarcopenic frail elderly patients enrolled in a nutritional and physical activity program and to evaluate their correlation with clinical markers. Moreover, the possible changes, produced after a short-term clinical protocol, were evaluated. Plasma and erythrocyte FA composition and pro-antioxidant status were analyzed in sarcopenic elderly subjects recruited for the randomized clinical study and treated with a placebo or dietary supplement, a personalized diet, and standardized physical activity. Subjects were tested before and after 30 days of treatment. Pearson correlations between biochemical parameters and patients’ characteristics at recruitment indicate interesting features of sarcopenic status such as negative correlation among the plasma FA profile, age, and physical characteristics. Physical activity and dietetic program alone for 30 days induced a decrease of saturated FA concentration with a significant increase of dihomo-gamma-linolenic acid. Supplementation plus physical activity induced a significant decrease of linoleic acid, omega-6 polyunsaturated FAs, and an increase of stearic and oleic acid concentration. Moreover, glutathione reductase activity, which is an indicator of antioxidant status, significantly increased in erythrocytes. Changes over time between groups indicate significant differences for saturated FAs, which suggest that the amino acid supplementation restores FA levels that are consumed during physical activity. A relationship between FA and clinical/metabolic status revealed unique correlations and a specific metabolic and lipidomic fingerprint in sarcopenic elderly. The results indicate the positive beneficial role of supplementation and physical activity on plasma FA status and the antioxidant system as a co-adjuvant approach in sarcopenic, frail, elderly patients.

Decreased Level of Nitric Oxide in Preeclamptic Pregnancy: A Relationship with PON1 Arylesterase Activity

Aims: The aim of the study was to evaluate the status and diagnostic utility of PON1. (Paraoxonase-1) Arylesterase and nitric oxide as indicator of antioxidant status in preeclampsia.&#x0D; Study Design: Analytical case control study.&#x0D; Place and Duration of Study: Sample: Department of obstretics and gynecology Department, G. M. C. Ambajogai, between July 2010 and July 2012.&#x0D; Methodology: We conducted a case-control study of 57 women with preeclampsia and 57 women with uncomplicated deliveries. We measured PON1 Arylesterase activity, Nitric oxide and lipid profile.&#x0D; Results: Serum levels of LDLc (low density lipoprotein cholesterol) are higher in cases than in controls and are statistically significant (p=0.023). However serum HDLc (high density lipoprotein cholesterol) levels are decreased significantly (p = 0.017). Serum PON1 Arylesterase showed significant decrease in cases152.68 KU/L versus controls 180.89 KU/L, p value=0.002. Serum nitric oxide also showed significant decrease in cases 22.77 ± 4.792 umol/L versus controls 25.127 umol/L, p=0.010. PON1 Arylesterase activity is found to be positively correlated with serum HDL cholesterol (r = 0.449, p value&lt; 0.001). Multivariate logistic regression analysis was done.&#x0D; Conclusion: Our observed results show decrease in the antioxidant PON1 Arylesterase activity point towards their role in the pathogenesis of Preeclampsia.

The effect of the dietary inclusion levels and sources of zinc on the performance, metabolism, redox and immune status of turkeys

The aim of this experiment was to determine whether the inclusion levels of supplemental Zn in turkey diets could be reduced without compromising important metabolic functions, the immune status and growth parameters of birds, and whether the above goal can be achieved with the use of zinc (Zn) nanoparticles. The experiment was performed on 648 one-day-old Hybrid Converter turkey hens divided into 6 groups with 6 replicates (18 birds per pen), in a two-factorial design with three dietary supplemental levels of Zn (100, 50 and 10 mg/kg) and two dietary sources of Zn - conventional zinc oxide (ZnO) and Zn nanoparticles (NP-ZnO). The applied inclusion levels and sources of Zn had no effect on the growth performance of turkeys, including liveability. The replacement of ZnO with NP-ZnO in turkey diets led to an increase in the levels of albumin (ALB), triacylglycerols (TG) and alkaline phosphatase (ALP) activity in the blood serum as well as minor changes in redox indicators including a decrease in plasma malondialdehyde (MDA) levels, catalase (CAT) activity in the liver, and total glutathione (GSH + GSSG) and MDA levels in the bursa of Fabricius. The addition of 50 mg and 100 mg of Zn exerted similar effects on blood biochemical parameters and antioxidant status indicators in the liver. Only in the bursa of Fabricius, the moderate dietary inclusion level of Zn contributed to a decrease in GSH + GSSG levels. The results of this study indicate that the recommended level of ZnO supplementation in turkey diets can be reduced to 50 mg/kg. A decrease in the dose of supplemental Zn from 100 to 10 mg/kg did not compromise the growth performance of turkeys, but it had a negative effect on lipid metabolism by increasing plasma TG concentrations. The replacement of ZnO with NP-ZnO had no beneficial influence on performance, the immune and antioxidant status of turkeys.

Tissue Inflammatory Markers and Oxidative Stress in Swine Subjected to a 40% TBSA Burn Injury

Burn injury is well known to induce a robust inflammatory response and hypermetabolic state. Intravenous (IV) crystalloid fluid resuscitation of burn patients is the foundation of modern burn care to improve tissue perfusion and outcomes. However, under military scenarios of far‐forward burn care or under civilian mass casualty situations, the availability of large volumes of such fluids for IV burn resuscitation will be logistically challenging and definitive burn care may be delayed for days. More recently, enteral resuscitation of burns has received renewed attention and some evidence suggests that it may reduce IV fluid requirements to support urine output. The present study investigated the early response of tissue inflammatory markers and evidence for oxidative stress in swine subjected to a 40% burn and resuscitated with enteral fluids compared to no fluids or a standard of care IV fluid regimen. Anesthetized Yorkshire swine (n=6 per group) were subjected to a 40% full thickness total body surface area (TBSA) burn with brass probes heated to 100°C. They were then randomized to 4 groups: 1) no fluid (NF), 2) 15 ml/kg/d WHO oral rehydration solution (ORS) (15‐ORS), 3) 70 ml/kg ORS (70‐ORS) or 4) 2 ml/kg/% burn IV lactated Ringer's (Modified Brooke (MB) formula‐ a standard of care). After 48 h, animals were euthanized and heart, liver, kidney, lung and small intestine collected and analyzed for tissue cytokine levels and indices of oxidative stress. As expected, urine output was higher in the 70‐ORS and MB groups than the NF group (p&lt;0.01). Although plasma creatinine and BUN peaked 6 h after burn, levels returned to baseline by 48 h in all groups except 15‐ORS and NF. In the kidney, levels of IL‐1β, IL‐6 and TNF‐α tended to be higher in the 70‐ORS and MB groups, whereas IL‐10 levels were similar among groups. In liver, heart and jejunum, only IL‐1β levels tended to be higher in MB than the other groups, with a similar trend in IL‐10 concentrations. With regard to oxidant status in kidney, total antioxidant levels, glutathione reductase (GR) and glutathione peroxidase activities were higher in 70‐ORS than the other groups. Mn‐superoxide dismutase (SOD) activity was higher in both 70‐ORS and MB groups, with no differences in catalase activity or nitric oxide levels. In liver, the lowest levels of total antioxidants, SOD, GR and catalase activities were observed in the NF groups compared to the others. No significant differences in indices of antioxidant status were observed in jejunum, heart or lung among groups. Typically, these variables in 15‐ORS were similar to the NF group in all tissues. The results of this study indicate that enteral fluids may be as effective as a standard of care IV fluid regimen in maintaining antioxidant status in vital organs without major changes in pro‐inflammatory cytokines. Determination of antioxidant indices in dairy cows during the periparturient period

The aim of the present study was to evaluate indicators of antioxidant status – glutathione peroxidase (GPx); superoxide dismutase (SOD); total antioxidant status (TAS); vitamins A, E and beta carotene in 10 dairy cows of the Holstein breed from 15-20 days ante partum (a.p.) to 25–30 days post partum (p.p.). Blood samples were collected 5 × during this period: 15–20 days a.p., 1–3 days a.p., 2–3 days p.p., 10–15 days p.p. and 25–30 days p.p. The mean GPx activity was significantly (P = 0.013) lower in the 10–15 days p.p. compared to 15–20 days a.p. (P &lt; 0.05) and 1–3 days a.p. (P &lt; 0.01). The increase of SOD values was monitored throughout the whole experiment. The mean TAS concentration was significantly (P &lt; 0.01) decreased 2–3 days p.p. compared to 25–30 days p.p. A significantly lower mean concentration of vitamin A was also found 2–3 days p.p. compared to the concentration 15–20 days a.p. (P &lt; 0.05) and 25–30 days p.p. (P &lt; 0.001). A significant (P &lt; 0.001) decrease in vitamin E concentration was recorded in cows 2–3 days p.p. compared to cows 15–20 days a.p. and 25–30 days p.p. The mean concentration of beta carotene was also decreased immediately after calving. Significant changes in the concentration of antioxidant parameters during the periparturient period indicate the occurrence of oxidative stress in dairy cows which can contribute to increased incidence of metabolic diseases.

Genetic polymorphism and antioxidant activity in interventions of tobacco-related diseases of the abdominal aorta.

IntroductionThe aim of this study was to determine the genetic polymorphisms of some antioxidant enzymes together with oxidative stress and the response of some antioxidant enzymes against this situation in vascular and endovascular interventions performed for diseases of the infrarenal abdominal aorta.Material and methodsTwenty-four current or ex-smoker patients (eight aortoiliac occlusive disease (AOD), 16 abdominal aortic aneurysm (AAA)) who were operated were included in this pilot study. Malonyl dialdehyde (MDA) levels, as an indicator of oxidative stress, reduced glutathione, catalase and superoxide dismutase enzymes, which are indicators of antioxidant status, which were measured in aortofemoral bypass in AODs, and in endovascular abdominal aortic aneurysms repairs in the preoperative, operative, and postoperative periods. Genetic polymorphisms of these antioxidant enzymes developing a response to the damage in the preoperative blood samples were determined by using the PCR-RFLP method.ResultsThe lack of a significant increase of MDA (nmol/ml preoperative: 2.57 ±0.19, operative: 2.59 ±0.21, postoperative: 2.64 ±0.22, p = 0.63) in the oxidative damage in the operative and postoperative periods compared to the preoperative period prevented the damage and was thought to be associated with the elevation of some effective antioxidant parameters in the operative and postoperative periods.ConclusionsIt may be thought that both types of interventions are quite reliable in terms of oxidative damage and, accordingly, the effect of the procedure-associated oxidative damage in the postoperative complications is low or ineffective. Two genotypes were obtained in each of the three gene areas of the patients, and no statistical significance was determined between the genotypes (p > 0.05).

C60 Fullerene Prevents Restraint Stress-Induced Oxidative Disorders in Rat Tissues: Possible Involvement of the Nrf2/ARE-Antioxidant Pathway.

The effects of C60FAS (50 and 500 μg/kg) supplementation, in a normal physiological state and after restraint stress exposure, on prooxidant/antioxidant balance in rat tissues were explored and compared with the effects of the known exogenous antioxidant N-acetylcysteine. Oxidative stress biomarkers (ROS, O2·−, H2O2, and lipid peroxidation) and indices of antioxidant status (MnSOD, catalase, GPx, GST, γ-GCL, GR activities, and GSH level) were measured in the brain and the heart. In addition, protein expression of Nrf2 in the nuclear and cytosol fractions as well as the protein level of antiradical enzyme MnSOD and GSH-related enzymes γ-GCLC, GPx, and GSTP as downstream targets of Nrf2 was evaluated by western blot analysis. Under a stress condition, C60FAS attenuates ROS generation and O2·− and H2O2 releases and thus decreases lipid peroxidation as well as increases rat tissue antioxidant capacity. We have shown that C60FAS supplementation has dose-dependent and tissue-specific effects. C60FAS strengthened the antiradical defense through the upregulation of MnSOD in brain cells and maintained MnSOD protein content at the control level in the myocardium. Moreover, C60FAS enhanced the GSH level and the activity/protein expression of GSH-related enzymes. Correlation of these changes with Nrf2 protein content suggests that under stress exposure, along with other mechanisms, the Nrf2/ARE-antioxidant pathway may be involved in regulation of glutathione homeostasis. In our study, in an in vivo model, when C60FAS (50 and 500 μg/kg) was applied alone, no significant changes in Nrf2 protein expression as well as in activity/protein levels of MnSOD and GSH-related enzymes in both tissues types were observed. All these facts allow us to assume that in the in vivo model, C60FAS affects on the brain and heart endogenous antioxidative statuses only during the oxidative stress condition.
