Insights into thermal sensitivity: Effects of elevated temperature on growth, metabolic rate, and stress responses in Atlantic wolffish (Anarhichas lupus).

The Atlantic wolffish (Anarhichas lupus) is a cold-water fish with potential for aquaculture diversification. To unveil the mechanisms underlying the compromised growth in Atlantic wolffish when reared at higher temperatures, we investigated the relationship between temperature, growth rate, aerobic capacity, stress biomarkers, and gut barrier function. Juveniles acclimated to 10°C were maintained at 10°C (control) or exposed to 15°C for either 24 h (acute exposure) or 50 days (chronic exposure). Fish exposed to 15°C exhibited reduced growth, higher standard, and maximum metabolic rates compared to those at 10°C. In the chronically exposed group at 15°C, metabolic rates were lower than those of acutely exposed fish. The absolute aerobic scope exhibited no significant variation in temperatures; however, the factorial scope showed a notable reduction at 15°C in both acute and chronic exposed groups, aligning with a correlated decrease in individual growth rates. Chronic warming led to increased plasma glucose levels, indicating energy mobilization, but cortisol levels were unaffected. Furthermore, chronic warming resulted in reduced intestinal barrier function, as evidenced by increased ion permeability and a negative potential in the serosa layer. We conclude that warming elevates metabolic rates while reducing intestinal barrier function, thus increasing energy expenditure, collectively, limiting energy available for growth at this temperature from increased allostatic load. Thus, juvenile wolffish maintaining their aerobic scope under thermal stress experience slower growth. This research provides insights for improving the welfare and resilience of wolffish in aquaculture at elevated temperatures and understanding their response to increased environmental temperatures.

Acute Hyperglycemia-Induced Injury in Myocardial Infarction.

Acute hyperglycemia is a transient increase in plasma glucose level (PGL) frequently observed in patients with ST-elevation myocardial infarction (STEMI). The aim of this review is to clarify the molecular mechanisms whereby acute hyperglycemia impacts coronary flow and myocardial perfusion in patients with acute myocardial infarction (AMI) and to discuss the consequent clinical and prognostic implications. We conducted a comprehensive literature review on the molecular causes of myocardial damage driven by acute hyperglycemia in the context of AMI. The negative impact of high PGL on admission recognizes a multifactorial etiology involving endothelial function, oxidative stress, production of leukocyte adhesion molecules, platelet aggregation, and activation of the coagulation cascade. The current evidence suggests that all these pathophysiological mechanisms compromise myocardial perfusion as a whole and not only in the culprit coronary artery. Acute hyperglycemia on admission, regardless of whether or not in the context of a diabetes mellitus history, could be, thus, identified as a predictor of worse myocardial reperfusion and poorer prognosis in patients with AMI. In order to reduce hyperglycemia-related complications, it seems rational to pursue in these patients an adequate and quick control of PGL, despite the best pharmacological treatment for acute hyperglycemia still remaining a matter of debate.

Insulin and glucose tolerance assessment in guanylyl cyclase/natriuretic peptide receptor-A gene-targeted mutant mice

Atrial natriuretic peptide (ANP), acting through guanylyl cyclase/natriuretic peptide receptor-A (GC-A/NPRA) regulates blood pressure and cardiac homeostasis. The disruption of Npr1 (encoding NPRA) in mice exhibits hypertension and provokes renal and cardiac disorder. However, the underlying mechanisms are not yet precisely determined. The objective of this study was to determine whether Npr1 plays a role in regulating glucose homeostasis in global Npr1 gene-disrupted mice. The adult male (14-18 wks) Npr1 haplotype gene-knockout ( Npr1+/-, 1-copy), wild-type ( Npr1+/+, 2-copy), and gene-duplicated ( Npr1+ +/+ +, 4-copy) mice were fasted overnight (16 h) and given free access to water. The mice were administered with glucose both orally and intraperitoneally (2 g/kg body weight) to determine the oral glucose tolerance test (OGTT) and intraperitoneal glucose tolerance test (IPGTT). For the insulin tolerance test (ITT), mice were also fasted for 16 h and then received an ip-injection of insulin (1.0 UI/kg body weight). Blood glucose levels were determined by performing tail bleeds at 0, 15, 30, 60, 90, and 120 min using the AlphaTRAK blood glucose monitoring system. The results showed that administration of glucose resulted in a greater increases in blood glucose levels at 120 mins in 1-copy mice (OGTT: 237 ± 5 mg/dL, IPGT: 246 ± 6 mg/dL, ITT; 239 ± 7 mg/dL) than 2-copy male (OGTT: 131 ± 3 mg/dL, IPGT: 126 ± 6 mg/dL, ITT; 127 ± 6 mg/dL), respectively. The blood glucose was also significantly lower in 4-copy mice (OGTT: 113 ± 5 mg/dL, IPGT: 108 ± 7 mg/dL, ITT; 107 ± 3 mg/dL) than in 2-copy mice. Systolic blood pressure (SBP) was determined by non-invasive tail-cuff method (Visitech 2000). SBP was significantly greater in 1-copy mice (130 ± 4 mmHg) than 2-copy mice (100 ± 3 mmHg). Similarly, SBP was also significantly lower in 4-copy mice (90 ± 2) than 2-copy mice. The increase in plasma glucose levels was significantly lower in OGTT than in IPGTT. The present results suggest that decreased Npr1 gene copy number predisposes to hyperglycemia, insulin resistance, and hypertension. These results indicate that the Npr1 gene might regulate blood glucose homeostasis. This work was supported by the NIH grant (DK133833). Adaptation of Chinese perch (Siniperca chuatsi) to different levels of dietary carbohydrates

The objective of the study was to investigate the effects of dietary carbohydrate on growth performance, feed utilization, proximate compositions, and hepatic glucolipid metabolism in Chinese perch. Triplicate groups of Chinese perch (29.51 ± 0.24 g) were fed five isonitrogenous and isolipidic diets containing graded levels of carbohydrate (2.4%, 9.7%, 16.5%, 23.9%, and 30.1%) for 8 weeks. The results based on second-order polynomial regression analysis of specific growth rate (SGR) proved that Chinese perch obtained the best growth performance at the 10.93% dietary carbohydrate level. Compared to 2.4%-carbohydrate group, 9.7%-carbohydrate group exhibited the increase in daily weight gain (DWG) and protein retention ratio (PER) and the decrease in feed conversion efficiency (FCR). Moreover, 9.7%-carbohydrate group showed the increase in the mRNA levels of pfk and cs involved in aerobic oxidation pathway, the content of liver glycogen increased significantly, and no change in glucose level. Above results suggested that a part of carbohydrates may be converted into energy for growth by aerobic oxidation pathway, or converted into glycogen for storage to maintain glucose homeostasis in Chinese perch fed with moderate-carbohydrate diet. Differently, 23.9%-carbohydrate and 30.1%-carbohydrate groups showed the decrease in DWG and PER, and the increase in FCR, compared to 2.4%-carbohydrate group. Additionally, 23.9%-carbohydrate and 30.1%-carbohydrate group presented the increase in the mRNA levels of g6pca and pc involved in gluconeogenesis, which caused the increase in plasma glucose level. However, other plasma indices related to TG-metabolite and its transport (total cholesterol, and low-density lipoprotein) did not show difference, which combined with elevated fatty acid synthesis-related gene accα mRNA level resulted in excessive liver lipid deposition. Above results indicated that unregulated gluconeogenesis and invalid outward transportation of liver TG-rich metabolites may account for the glucose intolerance of Chinese perch fed with carbohydrate-rich diet. Furthermore, compared to the results from the 4th week, the expression of gk, pfk, pc, cs, and accα genes significantly increased in the 30.1%-carbohydrate group at the 8th week. Both liver and muscle exhibited a significant elevation in crude fat content, while Daily DWG experienced a noticeable decrease. These results indicated that the glucose metabolism and growth of Chinese perch in the high-carbohydrate group are influenced by the interaction of feeding duration and carbohydrate levels. And within a certain time frame, Chinese perch can tolerate a diet rich in carbohydrates.

Psychosocial Well-being among Diabetic Individuals

Background: Quality of life (QoL) assessment is considered an important outcome measure in chronic disease management. With an increase in the prevalence of diabetes in India, it becomes imperative to assess QoL in diabetic patients for their better care and control. The present study aimed to assess the QoL among diabetic patients attending the diabetic outpatient clinic and probe its association with glycemic status and management. Methodology: A cross-sectional study was conducted for 4 months among 185 type 2 diabetes mellitus (T2DM) patients. Information on sociodemographic profile, diabetic history, and current medications was obtained. QoL was assessed using the General Health Questionnaire-28 (GHQ-28). The results were statistically analyzed. Results: Using GHQ-28, the psychosocial components of the patients in the fields related to somatic symptoms, anxiety and insomnia, social dysfunction, and severe depression were assessed. Significantly higher GHQ scores (P < 0.05) were noted in T2DM patients with higher glycated hemoglobin and increased plasma glucose levels, suggesting stronger associations of poorer QoL with compromised glycemic status. Insulin users exhibited higher GHQ scores (higher scores for depression subdomains) compared to noninsulin users. QoL was also found significantly dependent on the duration of diabetes and other comorbidities. Conclusion: The psychosocial well-being of the patients is significantly impacted in cases of chronic ailments like diabetes which adds to the total disease burden eventually. The present study warrants periodic assessment of QoL in diabetic individuals as a part of routine care along with advise of psychiatric counseling for those in need.

Optimization of a glucagon-like peptide 1 receptor antagonist antibody for treatment of hyperinsulinism.

Congenital hyperinsulinism (HI) is a genetic disorder in which pancreatic beta cell insulin secretion is excessive and results in hypoglycemia that can cause brain damage or death without treatment. Most patients with loss-of-function mutations in ABCC8 and KCNJ11, the genes encoding the beta cell ATP sensitive potassium channel (KATP), are unresponsive to diazoxide, the only FDA approved medical therapy, and require a pancreatectomy. The glucagon-like peptide 1 receptor (GLP-1R) antagonist exendin-(9-39) is an effective therapeutic agent to inhibit insulin secretion in both congenital and acquired hyperinsulinism. Previously, we identified a highly potent antagonist antibody, TB-001-003, which was derived from our synthetic antibody libraries that were designed to target G protein-coupled receptors. Here, we designed a combinatorial variant antibody library to optimize the activity of TB-001-003 against GLP-1R and performed phage-display on cells overexpressing GLP-1R. One antagonist, TB-222-023, is more potent than exendin-(9-39), also known as Avexitide. TB-222-023 effectively decreased insulin secretion in primary isolated pancreatic islets from a mouse model of hyperinsulinism, Sur1-/- mice, and in islets from an infant with HI as well as increased plasma glucose and decreased the insulin to glucose ratio in Sur1-/- mice. These findings demonstrate that targeting GLP-1R with an antibody antagonist is an effective and innovative strategy for treatment of hyperinsulinism.

Discovery of anthraquinones as DPP-IV inhibitors: Structure-activity relationships and inhibitory mechanism

Dipeptidyl peptidase IV (DPP-IV) is an integrated type II transmembrane protein that reduces endogenous insulin contents and increases plasma glucose levels by hydrolyzing glucagon-like peptide-1 (GLP-1). Inhibition of DPP-IV regulates and maintains glucose homeostasis, making it an attractive drug target for the treatment of diabetes II. Natural compounds have tremendous potential to regulate glucose metabolism. In this study, we examined the DPP-IV inhibitory activity of a series of natural anthraquinones and synthetic structural analogues on DPP-IV using fluorescence-based biochemical assays. The inhibitory efficiency differed among anthraquinone compounds with different structures. Alizarin (7), aloe emodin (11), emodin (13) emerged the outstanding inhibitory potential for DPP-IV with IC50 values lower than 5 μM. To clarifying the inhibitory mechanism, inhibitory kinetics were performed, which showed that alizarin red S (8) and 13 were effective non-competitive inhibitors of DPP-IV, while alizarin complexone (9), rhein (12), and anthraquinone-2-carboxylic acid (23) were mixed inhibitors. Emodin was determined as inhibitor with the strongest DPP-IV-binding affinity determined via molecular docking. Structure-activity relationship (SAR) demonstrated that hydroxyl group at C-1 and C-8 sites and hydroxyl, hydroxymethyl or carboxyl group at the C-2 or C-3 site were very essential for DPP-IV inhibition, replacement of hydroxyl group with amino group at C-1 could led to an increase of the inhibitory potential. Further fluorescence imaging showed that both compounds 7 and 13 significantly inhibited DPP-IV activity in RTPEC cells. Overall, the results indicated that anthraquinones would be a natural functional ingredient for inhibiting DPP-IV and provided new ideas for searching and developing potential antidiabetic compounds.

Induction of a single dose of streptozotocin (50 mg) in rat model causes insulin resistance with type 2 diabetes mellitus.

Streptozotocin (STZ) is a broad-spectrum antibiotic that is toxic to the insulin-producing beta cells of the pancreatic islets. STZ is currently used clinically for the treatment of metastatic islet cell carcinoma of the pancreas and the induction of diabetes mellitus (DM) in rodents. So far, there has been no previous research to show that STZ injection in rodents causes insulin resistance in type 2 diabetes mellitus (T2DM). The purpose of this study was to determine if rats (Sprague-Dawley) developed type 2 diabetes mellitus (insulin resistance) after 72 h of intraperitoneal administration of 50 mg/kg STZ. Rats with fasting blood glucose levels above 11.0 mM, 72 h post-STZ induction, were used. The body weight and plasma glucose levels were measured every week throughout the 60-day treatment period. The plasma, liver, kidney, pancreas, and smooth muscle cells were harvested for antioxidant, biochemical analysis, histology, and gene expression studies. The results revealed that STZ was able to destroy the pancreatic insulin-producing beta cell, as evidenced by an increase in plasma glucose level, insulin resistance, and oxidative stress. Biochemical investigation indicates that STZ can generate diabetes complications through hepatocellular damage, elevated HbA1c, kidney damage, hyperlipidemia, cardiovascular damage, and impairment of the insulin-signaling pathway.

Implications of the COVID-19 Pandemic and Social Isolation on the Cardiometabolic Profile of a Cohort of Individuals from the City of Buenos Aires

Background: The COVID-19 pandemic has shocked humanity. During the pandemic, the need for social isolation has encouraged low adherence to a healthy lifestyle in many individuals. However, evidence of the metabolic impact of COVID-19 pandemic on our field is scarce. Objective: To evaluate the impact of social isolation produced by the COVID-19 pandemic on the body weight and the cardiometabolic parameters of an adult population in the Autonomous City of Buenos Aires. Methods: Based on an observational design, we analyzed the data from patients who attended a prevention and health promotion program in the City of Buenos Aires. Data from participants who attended for testing in 2019 and repeated testing in 2021 were individualized. Medical records were used as source for collecting general data, anthropometric measurements, and laboratory values. The National Cholesterol Education Program (NCEP) criteria were used to define the presence of metabolic syndrome (MS). Results: A total of 558 patients with available evaluations in 2019 and 2021 were identified. The average age of the population was 52.2 ± 12.8 years, and 41% was female. An increase in body weight (82.1 ± 17.7 kg vs. 83.1 ± 18.5 kg; p<0.0001) and body mass index (29.4 ± 5.4 vs. 29.8 ± 5.7, p<0.0001) was observed. Increases in systolic (123.1 ± 15.1 mmHg vs. 126.6 ± 16.3 mmHg; p<0.0001) and diastolic (77.7 ± 9.3 mmHg vs. 79.6 mmHg± 9.4 mmHg, p<0.0001) blood pressure values were also observed. As regards the laboratory parameters, we noted an increase in plasma glucose levels with a median and an interquartile range (IQR) of 95 (89-103 mg/dL) vs. 99 (92-107 mg/dL), p<0.0001, and a decrease in HDL cholesterol (51, 8 ± 12.7 mg/dL vs. 49.3 ± 12.8 mg/dL, p<0.0001). No changes were observed in LDL cholesterol (116.4 ± 32.6 mg/dL vs. 116.1 ± 34 mg/dL; p=NS), total cholesterol (194.9 ± 37.4 vs. 193 ± 39.6 mg/dL; p=NS) or triglyceride levels, with a median (IQR) of 114.5 (83.2-162.7 mg/dL) vs. 118 (88-169 mg/dL; p=NS). This was accompanied by an increased prevalence of MS (21.5% vs. 34%; p<0.0001). The proportion of patients with carotid plaques also increased, without reaching statistical significance (36.4% vs. 40.7%; p=NS). Besides, it was observed that 18.8% of the patients increased their body weight by more than 5%. This population was represented by younger patients (47.6 ± 14 years vs. 53.3 ± 12 years; p< 0.0001) showing a reverse correlation between age and scope of weight increase (r=-0.1; p<0.02). Conclusion: Social isolation during the COVID-19 pandemic was shown to have important consequences on the risk factors of the population studied. The prospective implications of these findings might become apparent in the next few years if these metabolic alterations are not reversed.

Consideraciones anestésicas en la diabetes mellitus: revisión narrativa

La diabetes mellitus (DM) desencadena complicaciones agudas y crónicas. Los individuos con DM que requieren una intervención quirúrgica se consideran pacientes de riesgo elevado. En efecto, entre los pacientes diabéticos gran parte de las intervenciones quirúrgicas se consideran consecuencia de las complicaciones de la DM. Cabe destacar, que la cirugía y la anestesia general constituyen un estrés que incrementa los niveles plasmáticos de glucosa lo que determina una significativa tendencia a la hiperglicemia y la cetoacidosis. La cirugía, su tipo y duración incrementan la secreción de glucagón, catecolaminas e inhibe la secreción de insulina por las células beta pancreáticas. En casos de cirugía electiva se recomiendan sostener niveles de hemoglobina glucosilada ≤ 8.5%. En consecuencia, se demanda contar con los recursos necesarios para realizar el diagnóstico y tratamiento de la hiperglucemia perioperatoria, con el fin de disminuir sus complicaciones.

Both osmolality-dependent and independent mechanisms are associated with acute hyperglycemia-induced cardiovascular adverse reactions: Analysis of the mutual interactions leading to cardiovascular phenotypes in dogs.

Acute hyperglycemia causes various cardiovascular responses; however, the underlying pathophysiology in vivo is myriad and complex, of which mutual interactions remain poorly understood. We analyzed the cardiovascular effects of acute hyperglycemia in comparison with those of hyperosmolality alone. Three g/kg of D-glucose (n = 4) or D-mannitol (n = 4) was intravenously infused to isoflurane-anesthetized intact dogs. Glucose infusion increased plasma glucose level and osmolality, whereas mannitol infusion similarly changed osmolality to glucose infusion but decreased glucose level. Glucose infusion decreased total peripheral vascular resistance, but increased heart rate, left ventricular contraction, left ventricular preload and cardiac output without altering mean blood pressure. Mannitol infusion likewise changed them, but its positive chronotropic and inotropic effects were less potent than those of glucose infusion. Glucose infusion prolonged PR interval, QRS width and QTcV. Mannitol infusion similarly changed them, but its QTcV prolongation was smaller than that of glucose infusion. Glucose infusion-induced cardiovascular responses would be basically attributed to osmolality-dependent mechanisms, whereas its positive chronotropic and inotropic effects along with repolarization delay may be enhanced by osmolality-independent mechanisms, including hyperglycemia by itself and insulin release.

Assessment of Plasma Glucose and Lipid Profile Dysregulation among Patients on Antipsychotics in Sokoto

Background: Antipsychotic drugs used in the treatment of psychotic disorders have been implicated in negative effects on the blood glucose and lipid profile levels of patients. Methods: We studied the anthropometric parameters and then the plasma glucose and lipid profile levels of one hundred and seventy-two (172) patients between the ages of 18 and 45 years; who are on various oral antipsychotics. Also, forty (40) treatment-naïve psychiatric patients and forty (40) healthy controls were included in the study. The results were analysed using standard methods. Results: The mean plasma glucose level was significantly higher (p<0.05) in psychiatric patients on oral antipsychotic drugs compared to controls. The mean plasma glucose level of treatment-naïve psychiatric patients was also significantly higher (p<0.05) compared to that of healthy controls. Furthermore, there was a significant increase in plasma glucose levels of patients on second-generation antipsychotic drugs compared to those on first-generation antipsychotic drugs. The lipid profile of patients on antipsychotic drugs showed a similar trend. There were significant increases in the plasma levels of Total Cholesterol (TC), Low-Density Lipoprotein (LDL), Triglycerides (TG) and a significant decrease in the plasma level of High-Density. However, the plasma TG levels were significantly higher in the typical antipsychotic drugs group. Lipoprotein (HDL) levels of individuals that were on antipsychotics were significantly decreased when compared to healthy controls Conclusion: The lipid profile and plasma glucose concentration were altered by oral antipsychotic drugs in Sokoto, Nigeria. Therefore, it may be clinically expedient to vigorously assess the plasma glucose and lipid profile levels of all patients that are on oral antipsychotic drugs and interventions planned as appropriate. Nanoherbal Ocimum Basilicium L Reduce Mda And Increase Sod Level In Wistar Rats Induced Stz

One of the public health problems that is very serious and requires proper treatment for sufferers is diabetes mellitus (DM). An increase in plasma glucose levels that exceeds normal limits is one of the basics for diagnosing diabetes. This study aim to determine the antidiabetic activity of Nanoherbal Extract of Ocimum Basilicum L. This is an experimental study in which the research group was divided into seven groups, each including five rats weighing 200-250 grams. Which are (normal group, negative group, metformin, nanoherbal 25, 50, 100, and 200 mg/kgBW). Rats were induced by STZ 75 mg/kgBW + NCT 120 mg/kgBW until the blood glucose is above 200 mg/dL. The highest level of SOD is the control group 63.984 ± 2,065 ng/mL and the lowest is negative group is 22,41 ± 2,06 ng/mL. For group that given treatment show that the highest SOD level is in dose 200 mg/kgBW 71,72 ± 0,90 ng/mL and the lowest is in dose 25 mg/kgBW 31,814 ± 1,11 ng/mL and the group given metformin is 68,52 ± 32,39 ng/mL. The highest level of MDA is in group control which given only STZ and NCT 37,522 ± 1,64 ng/mL and the lowest MDA level is in normal group 7.199 ± 1,415 ng/mL. however, after given extract show that MDA level decreases which show in the dosen 200 mg/kgBW 7,76 ± 1,64 ng/mL that nearly to the value of normal MDA level while the highest in the treatment group is 30,83 ± 0,961 ng/ml. In summary, Nanoherbal Extract of Ocimum Basilicum L has antidiabetic activity by increasing the level of insulin and antioxidant SOD level while reducing the MDA level

Effects of Persimmon Fruit Polyphenols on Postprandial Plasma Glucose Elevation in Rats and Humans.

Persimmon is a fruit rich in polyphenols (proanthocyanidins or condensed tannins). Using rats and humans, the effects of Kaki-tannin (Nara-type), persimmon polyphenols prepared using a new method, on postprandial plasma glucose levels were investigated in this study. Kaki-tannin (Nara-type) comprised mainly proanthocyanidins, composed of epicatechin : epicatechin gallate : epigallocatechin : epigallocatechin gallate in a ratio of 1 : 1 : 2 : 2 with a molecular weight of approximately 8,000 Da, with epicatechin gallate as a terminal unit. These polyphenols inhibited amylolytic enzymes, such as α-amylase, maltase, sucrase, and α-glucosidase in vitro, and sodium-dependent glucose transporter 1 in Caco-2 cells. These results suggested that the polyphenols suppressed digestion and absorption in the intestinal tract. The ingestion of 250 mg/kg body weight of the polyphenols significantly suppressed increased blood glucose levels after carbohydrate (2 g/kg body weight of glucose or maltose) loading in rats. In a human trial, 1.88 g of Kaki-tannin (Nara-type) significantly delayed increased plasma glucose levels after carbohydrate (150 kcal of maltooligosaccharides) loading. Thus, Kaki-tannin (Nara-type) holds promise to be developed as a food material that potentially improve blood glucose elevation after meals.

Role of Insulin Resistance in Gestational Diabetes Mellitus: A Literature Review

Several physiologic, hormonal, and molecular processes contribute to the emergence of hyperglycaemia during pregnancy. Increased insulin resistance is seen during the course of a healthy pregnancy. During the early stages of normal pregnancies, the pancreatic β-cells secrete more insulin, which slows the rise in plasma glucose levels. This regulation explains the abnormally modest increases in plasma glucose levels brought on by elevated insulin resistance. The aim of this review was to evaluate the role of insulin resistance in gestational diabetes mellitus (GDM). The authors extensively searched various electronic databases like PubMed, Scopus, and Google Scholar for the collection of material regarding the role of insulin resistance in GDM. It was seen that hyperglycaemia results from the combination of the pregnancy’s increased insulin tolerance and pancreatic beta-cell insufficiency. Scientific studies have revealed that individuals who present with GDM are more likely to acquire chronic insulin resistance because of the superimposition of lower insulin production by the cells in that condition (GDM). Due to their significance in the development of postpartum diabetes mellitus, inflammation markers in GDM have been widely researched. Inflammation during GDM induces adaptive reactions in the placenta, which can have a substantial effect on the programming of foetal development.

The growth performance and metabolic responses of juvenile spotted rose snapper (Lutjanus guttatus) fed diets with different precooked cornstarch to protein ratio

The effect of diets with varying crude protein-to-precooked cornstarch (P:S) ratios on growth performance, feed efficiency, carbohydrase activity, and blood chemistry of juvenile Lutjanus guttatus was evaluated. Four isoenergetic (19.0 kJ g-1) diets containing different P:S ratios (46:18, 38:30, 34:35, and 31:40) were formulated to feed fish for ten weeks in triplicate. Each diet was tested in triplicate in groups of 35 fish per tank (average body weight 19.4 ± 0.3 g) to apparent satiation three times a day. The weight gain, specific growth rate, and feed conversion ratio obtained with P38S30 and P34S35 were not significantly different vs. P46S18. Survival does not present significant differences among treatments. Biological indices (condition factor, hepatosomatic index, viscerosomatic index, and intraperitoneal fat rate) showed no significant differences among treatments. Body protein content decreased significantly with increasing precooked cornstarch concentration, while body lipid levels increased. Blood chemistry values were within those of healthy juveniles, except for glucose values. An increase in pancreatic α-amylase and intestinal α-glucosidase blood chemistry values were within those of healthy juveniles, except for glucose values. An increase in pancreatic α-amylase and intestinal α-glucosidase levels concomitant with increasing precooked cornstarch concentration showed the capacity of juvenile L. gutattus to digest high precooked cornstarch levels. Growth performance, biological indices, and hematological parameters showed that L. guttatus could digest and metabolize up to 35% of precooked cornstarch through increased plasma glucose levels and the α-amylase and α-glucosidase activities. Without negative effect on growth performance.

The Effect of Glucose Administration on Yes‐associated Protein (YAP) Activity in Healthy and Cachectic Mouse Muscles

Yes‐associated protein (YAP) is a downstream effector of Hippo signaling and regulates skeletal muscle size and homeostasis. The change of YAP activity has been reported in aging and dystrophic muscles. Our preliminary data showed that tumor burden alters the YAP activity in skeletal muscle. Recent studies showed that glucose metabolism also regulates YAP activity in cancer cells; however, it is unknown whether glucose administration stimulates YAP activity in skeletal muscle. The purpose of this study was to determine whether the YAP activity would change after acute glucose administration in healthy and cachectic mouse muscles. We hypothesized that glucose could stimulate YAP activity, at least in healthy muscle. Approximately 4‐month‐old wild‐type (WT, n=10) and ApcMin/+ mice (Min, n=10) were used in this study. Each group of mice was assigned to either control (Con, n=5 per group) or glucose administration (Glu n=5 per group) treatment. At first, all the mice fasted for four hours. Glucose dissolved in phosphate‐buffered saline (PBS, 2 mg/kg) was administered intraperitoneally to Glu groups. Con groups received PBS injections. Thirty minutes after glucose administration, the mice were euthanized, and plantaris muscles (PLAN) were removed. The time to sacrifice was based on the peak insulin secretion during a glucose tolerance test. To ensure injection accuracy, the glucose levels were measured using a handheld glucometer before glucose injection and at death. After total protein was extracted, routine Western blotting was performed using 50~100 µg of the total protein. Each phosphorylated protein was normalized to its respective total protein. Unpaired t‐test (WT vs. Min) and a two‐way ANOVA (genotype x glucose treatment) were used for statistical analysis. The levels of significance were set at 0.05. The body weight of Min mice was smaller than that of WT mice (19.8±1.2 mg vs. 17.3±0.5 mg for WT and Min, respectively, p=0.05). Likewise, the PLAN muscle mass of Min mice was smaller than that of WT mice (19.8±1.2 mg vs. 17.3±0.5 mg for WT and Min, respectively, p=0.05). There was no significant difference in fasting glucose levels between WT and Min mice, and glucose administration increased plasma glucose levels regardless of genotype (main effect of glucose administration, p<0.05). The activity levels of glycogen synthase kinase 3 beta (GSK3β) were comparable at the baseline (1.1±0.1 vs. 1.1±0.2 for WT and Min, respectively) and after glucose administration (1.1±0.2 vs. 1.1±0.4 for WT and Min, respectively). Min mice showed a reduction in total ribosomal protein S6 level after fasting (1.0±0.2 vs. 0.4±0.2 for WT and Min, respectively, p<0.05) and in response to glucose administration (2.7±1.5 vs. 0.6±0.4 for WT and Min, respectively, p<0.05). Interestingly, the basal YAP levels were comparable between WT and Min mice (1.0±0.1 vs. 1.1±0.1 for WT and Min, respectively), and the glucose administration did not alter the YAP levels (1.1±0.1 vs. 1.1±0.2 for WT and Min, respectively). These data suggest that YAP activity is independent of acute glucose administration in healthy and cachectic mouse skeletal muscles.

Effects of oral administration of polystyrene nanoplastics on plasma glucose metabolism in mice

Microplastic (MP) and nanoplastic (NP) induce neurotoxicity, cytotoxicity, and reproductive system toxicity in mammals. However, the impacts of NPs on the endocrine system are obscure. Here, monodisperse polystyrene nanoplastics (PS-NPs) were prepared by emulsion polymerization and the accumulation of fluorescent PS-NPs in various organs, including the liver, kidney, spleen, and pancreas, was examined. The oral administration of PS-NPs induced visceral organ injury, and the main toxicities were damage to hepatic function and the abnormity of lipid metabolism. Global transcriptome sequencing (RNA-Seq) revealed the impact of PS-NPs on the genes involved in reactive oxygen species (ROS) generation and the PI3K/Akt signaling pathway, which is associated with glucose metabolism in mice. Chronic exposure to PS-NPs significantly increased plasma glucose levels and ROS levels, but did not affect plasma insulin secretion. The phosphorylation of insulin receptor substrate (IRS)-1 at Ser307 was raised, which decreased the phosphorylation of Akt (at Ser473) in the PI3K/Akt pathway. Collectively, these findings suggested that the oral administration of PS-NPs significantly increased ROS, hepatic triglycerides, and cholesterol accumulation. The high levels of ROS disturbed the PI3K/Akt pathway, causing insulin resistance and increased plasma glucose in the mouse liver.

The Evaluation of Fasting Plasma Glucose (FPG) Levels in Hypertensive Patients Using Calcium Channel Blocker (CCB) Drugs Class in Undata Regional Hospital

Background: Hypertension plays a major role in the development of heart disease which is one of the most leading causes of death worldwide. Antihypertensives must be selected carefully to avoid affecting metabolic parameters. A risky case involving the increase of plasma glucose levels using Calcium Channel Blocker (CCB) antihypertensive, amlodipine, has been reported. Objectives: This study aims to examine the effects of CCB treatments on blood glucose levels in hypertensive patients at the Undata District General Hospital of Central Sulawesi Province. Material and Methods: This study used a cross-sectional design with a prospective approach. The instruments & materials used in this study are Accu-Chek Performa glucometer along with the patient's blood sample material. The research subjects were 31 patients who met the inclusion criteria; patients had been treated with CCB therapy for 3 months, patients were not diagnosed with diabetes mellitus, andpatients did not use drugs that might affect the blood glucose levels. Results: The results showed an average value of fasting plasma glucose (FPG) levels of hypertension patients using CCB class showed an increase from baseline of 124,619 mg/dL to 141,612 mg/dL with a difference of 16,993 mg/dL. Statistical analysis conducted also obtained a significance value of p 0,000 which indicates that the use of amlodipine can significantly increase fasting patients' plasma glucose levels. Conclusions: If observed clinically, the increasing in fasting plasma glucose values ​​falls into the category of mild to moderate hyperglycemic.

Pattern of Alpha Fetoprotein and Activities of Liver Enzymes in Type 2 Diabetic Patients Attending Nnamdi Azikiwe University Teaching Hospital Nnewi, Anambra State, Nigeria.

Diabetes mellitus (DM) is a metabolic disorder characterized by hyperglycemia, which has become health problem of global interest, linked to renal disease. The study is to assess the activities of some liver enzymes and alpha fetoprotein level in type 2 diabetic patients. 81 participants comprised 38 diabetics and 43 controls were randomly recruited. Informed, oral and written consent was obtained from the participants. Ethical approval was obtained. 6mls of fasting blood samples were collected. Plasma glucose, alpha feto-protein (AFP) levels; serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and gamma glutamyltransferase (GGT) activities were analyzed using standard laboratory methods. The results showed significantly increased plasma glucose level in the diabetics than in controls (p=0.000), as well as in male diabetics than in female diabetics (p=0.048). But, the mean age, AFP, ALT, AST, ALP and GGT activities were the same in the diabetics and the controls; and in male and female diabetics at p>0.05 in each case. Relatively strong positive significant correlation was observed between ALT Vs AST (r=0.566; p=0.000) and a weaker negative association exist between ALT Vs Age (r= -0.353; p=0.022) in diabetics but no associations were observed with other parameters studied. In conclusion the positive association between ALT and AST may reveal transaminase derangements in liver status of type 2 diabetic subjects studied.
