The Developmental Toxicity of Thymus schimperi Essential Oil in Rat Embryos and Fetuses.

Background In Ethiopian traditional medicine, the aerial parts of Thymus schimperi are widely used to treat diseases such as gonorrhea, cough, liver disease, kidney disease, hypertension, stomach pain, and fungal skin infections. In addition, they have been used as vegetables to flavor a broad variety of food products. However, there is an insufficient investigation of the toxic effect of Thymus schimperi essential oil. The aim of this study was, therefore, to evaluate the developmental toxicity of the essential oil of Thymus schimperi leaves on developing rat embryos and fetuses. Methods Essential oil of the aerial parts of Thymus schimperi was extracted by hydrodistillation. Pregnant Wistar albino rats were randomly divided into five groups. The doses 65 mg/kg, 130 mg/kg, and 260 mg/kg of the essential of Thymus schimperi were administered by force feeding to the III–V groups, respectively. Groups I and II were negative and ad libitum control groups. The embryos and fetuses were revealed on days 12 and 20 of gestations, respectively. The embryos were examined for developmental delays or growth retardation. Gross external, skeletal, and visceral anomalies in the fetuses were examined. Results In this study, the developmental scores of the number of implantation sites, crown-rump length, the number of somites, and morphological scores were significantly lower while the score of fetal resorptions was increased in a 12-day-old rat embryos treated with 260 mg/kg of the Thymus schimperi essential oil. There was also a significant delay in the development of the otic system, olfactory system, and a reduction in the number of branchial bars in 12-day-old embryos treated with 130 mg/kg and 260 mg/kg of the essential oil. However, external morphological examinations of rat fetuses revealed no detectable structural abnormalities. The fetal skull, vertebrae, hyoid, forelimb, and hindlimb ossification centers did not differ significantly across all the groups. Furthermore, there were no skeletal or soft-tissue malformations as a result of the essential oil treatment. Although the difference was not statistically significant, fetuses of the high-dose treatment group had a reduced number of ossification centers in the caudal vertebrae and hind limp phalanges. Conclusion The essential oil of Thymus schimperi at high doses has a detrimental effect on the development of rat embryos and fetuses. Its developmental toxicity is evidenced by significant delays in fetal and embryonic development, a decrease in the number of implantation sites, and an increase in fetal resorption. Furthermore, administration of the essential oil in higher doses resulted in a significant decrease in placenta weight and litter weight. In addition, the present study provided evidence that using the Thymus schimperi essential oil in a high dose could affect the developing embryo and fetus. Thus, it is recommended to discourage the use of Thymus schimperi essential oil in high doses.

Toxicity of Methanolic Extracts of Seeds of Moringa stenopetala, Moringaceae in Rat Embryos and Fetuses

Moringa stenopetala is a medicinal plant that has been used in Ethiopian traditional medicine as a remedy for the treatment of hypertension, diabetes, and stomach pain. The study is aimed at assessing the toxicity of the methanol extracts of the seeds of Moringa stenopetala on the developing embryo and fetuses of rats. The seeds of Moringa were extracted by maceration using 80% methanol. The extract (250–1000 mg/kg) was orally administered to pregnant Swiss albino rats from days 6 to12 of gestation. Embryos and fetuses were recovered by laparotomy on gestational day 12 and day 20, respectively, and were assessed for developmental anomalies. On day 20, significant prenatal growth retardation such as reduced litter weight and crown-rump length were observed in near term fetuses of 1000 mg/kg treated rats. Litter weight in 1000 mg/kg and pair-fed control groups was 2.41 g ± 0.108 and 3.08 g ± 0.093, respectively. Delay in the development of an otic, optic, and olfactory system, as well as a reduction in a number of branchial bars, occurred on day 12 embryos of 1000 mg/kg treated rats. The rate of fetal resorption in 1000 mg/kg and pair-fed control groups was 1.6 ± 0.55 and 0.42 ± 0.52, respectively. There was also a high incidence of fetal death in the 1000 mg/kg treated group but it was not statistically significant. The offspring's of Moringa-treated rats did not show gross external malformations at all doses. These findings suggest that the methanol seed extract of Moringa stenopetala is not safe to rat embryos and fetuses. Its toxic effects were evidenced by a significant delay in embryonic and fetal development and an increase in fetal resorptions and fetal death.

The overview of current evidence on the reproductive toxicity of dibutyl phthalate.

Over the past years, many legitimate concerns have been raised about the effects of dibutyl phthalate (DBP) as an endocrine disruptor, especially on reproduction. The aim of this publication is to critically review the literature related to the developmental and reproductive toxicity of DBP in animals. Several electronic databases were systematically searched until 2019. Studies were qualified for the review if they: linked exposure to DPB with reproduction, were published in English after 1990, and were conducted on animals. In the studies of the testicular effects of DBP on experimental animals, the most common effects of exposure included reduced fertility, atrophic changes in male gonads, degenerative changes in the epididymis, as well as a reduction in sperm count and motility, cryptorchidism, hypospadias, poor sperm quality and other genital defects (decreased testicular weight, delayed spermatogenesis, Leydig cell aggregation, impaired Sertoli cell maturation, and significant inhibitions of testicular enzymes). The embryotoxic effects of DBP on laboratory animals included mainly an increase in fetal resorption and a decrease in live births. The teratogenic effects of DBP also manifest as skeletal malformations in fetuses, malformations of male gonads and other genital effects. On the basis of the literature data, it is clearly demonstrated that DBP shows anti-androgenic effects; however, there are also reports confirming its weak estrogenic effect. Additionally, lower doses cause more adverse effects than the highest dose, which is an important fact because of the widespread environmental exposure to DBP. Identification of a Monoclonal Antibody That Attenuates Antiphospholipid Syndrome-Related Pregnancy Complications and Thrombosis.

In the antiphospholipid syndrome (APS), patients produce antiphospholipid antibodies (aPL) that promote thrombosis and adverse pregnancy outcomes. Current therapy with anticoagulation is only partially effective and associated with multiple complications. We previously discovered that aPL recognition of cell surface β2-glycoprotein I (β2-GPI) initiates apolipoprotein E receptor 2 (apoER2)-dependent signaling in endothelial cells and in placental trophoblasts that ultimately promotes thrombosis and fetal loss, respectively. Here we sought to identify a monoclonal antibody (mAb) to β2-GPI that negates aPL-induced processes in cell culture and APS disease endpoints in mice. In a screen measuring endothelial NO synthase (eNOS) activity in cultured endothelial cells, we found that whereas aPL inhibit eNOS, the mAb 1N11 does not, and instead 1N11 prevents aPL action. Coimmunoprecipitation studies revealed that 1N11 decreases pathogenic antibody binding to β2-GPI, and it blocks aPL-induced complex formation between β2-GPI and apoER2. 1N11 also prevents aPL antagonism of endothelial cell migration, and in mice it reverses the impairment in reendothelialization caused by aPL, which underlies the non-thrombotic vascular occlusion provoked by disease-causing antibodies. In addition, aPL inhibition of trophoblast proliferation and migration is negated by 1N11, and the more than 6-fold increase in fetal resorption caused by aPL in pregnant mice is prevented by 1N11. Furthermore, the promotion of thrombosis by aPL is negated by 1N11. Thus, 1N11 has been identified as an mAb that attenuates APS-related pregnancy complications and thrombosis in mice. 1N11 may provide an efficacious, mechanism-based therapy to combat the often devastating conditions suffered by APS patients.

Neutrophil Depletion Attenuates Placental Ischemia-Induced Hypertension in the Rat.

Preeclampsia is characterized by reduced placental perfusion with placental ischemia and hypertension during pregnancy. Preeclamptic women also exhibit a heightened inflammatory state and greater number of neutrophils in the vasculature compared to normal pregnancy. Since neutrophils are associated with tissue injury and inflammation, we hypothesized that neutrophils are critical to placental ischemia-induced hypertension and fetal demise. Using the reduced uteroplacental perfusion pressure (RUPP) model of placental ischemia-induced hypertension in the rat, we determined the effect of neutrophil depletion on blood pressure and fetal resorptions. Neutrophils were depleted with repeated injections of polyclonal rabbit anti-rat polymorphonuclear leukocyte (PMN) antibody (antiPMN). Rats received either antiPMN or normal rabbit serum (Control) on 13.5, 15.5, 17.5, and 18.5 days post conception (dpc). On 14.5 dpc, rats underwent either Sham surgery or clip placement on ovarian arteries and abdominal aorta to reduce uterine perfusion pressure (RUPP). On 18.5 dpc, carotid arterial catheters were placed and mean arterial pressure (MAP) was measured on 19.5 dpc. Neutrophil-depleted rats had reduced circulating neutrophils from 14.5 to 19.5 dpc compared to Control, as well as decreased neutrophils in lung and placenta on 19.5 dpc. MAP increased in RUPP Control vs Sham Control rats, and neutrophil depletion attenuated this increase in MAP in RUPP rats without any effect on Sham rats. The RUPP-induced increase in fetal resorptions and complement activation product C3a were not affected by neutrophil depletion. Thus, these data are the first to indicate that neutrophils play an important role in RUPP hypertension and that cells of the innate immune system may significantly contribute to pregnancy-induced hypertension.

Expression of Apoptosis in Placenta of Experimental Antiphospholipid Syndrome Mouse

To study the histopathology and expression of apoptosis in placenta of pregnancy-complicated antiphospholipid syndrome (APS)-positive mouse models. ICR mice were immunized with IgG isotype of human anticardiolipin (aCL) and/or lupus anticoagulant (LA). The pathological and apoptotic expression was studied in the placenta of positive APS mice and compared with respective control samples. Mice with passive immunization of human aCL and/or LA produced an increase in fetal resorption along with markedly induced thrombosis in the placenta associated with increased placental apoptosis. In addition, fewer mitotic cells, less trophoblast giant cell invasion, and more shrunken cells in the deciduas were seen. Our study showed the pathogenic role of aCL and LA in pregnancy complications.

A Unique Preliminary Study on Placental Apoptosis in Mice with Passive Immunization of Anti-Phosphatidylethanolamine Antibodies and Anti-Factor XII Antibodies

Antiphospholipid antibodies have been investigated both in humans and in animal models. In contrast, there are fewer reports describing anti-phosphatidylethanolamine (aPE) antibodies in humans, and there are no reports of animal studies with aPE till date. Clinically, FXII deficiency or anti-FXII antibodies are sometimes associated with aPE in patients with recurrent pregnancy loss. Therefore, we asked whether aPE and/or anti-FXII in mice could cause fetal resorption, placental thrombosis and apoptosis. Moreover, antibodies to respective target antigens (LDC27 or IPP30) could cause pregnancy failure as well. Animal models were used to carry out these objectives. All the animals were immunized with different antibodies by passive immunization. Placental samples were used for various observations. Mice with passive immunization of aPE (or anti-LDC27) and aFXII (or anti-IPP30) produced a slight increase in fetal resorption, but markedly induced thrombosis and hemorrhage in the placenta associated with lower platelet counts and increased placental apoptosis. In addition, fewer mitotic cells, less trophoblast giant cell invasion, and more shrunken cells in the deciduas were seen. Our study supports the pathogenic role of aPE and aFXII in pregnancy complications and also suggests a beneficial role of LDC27 and IPP30 antigens on pregnancy failures.

Phosphatidylinositol 3-Kinase Activity Is Critical for Glucose Metabolism and Embryo Survival in Murine Blastocysts

The phosphatidylinositol 3-kinase (PI3K) signal transduction pathway is a well known mediator of cell growth, proliferation, and survival signals. Whereas the expression and function of this pathway has been documented during mammalian development, evidence demonstrating the physiologic importance of this pathway in murine preimplantation embryos is beginning to emerge. This study demonstrates that inhibition of the PI3K pathway leads to the induction of apoptosis in both murine blastocysts and trophoblast stem cells. The apoptosis induced in both model systems correlates with a decrease in the expression of the glucose transporter GLUT1 at the plasma membrane. In addition, blastocysts cultured in the presence of the PI3K inhibitor LY-294002 display a decrease in both 2-deoxyglucose uptake and hexokinase activity as compared with control blastocysts. To determine the impact of PI3K inhibition on pregnancy outcome, embryo transfer experiments were performed. Blastocysts cultured in the presence of LY-294002 demonstrate a dramatic increase in fetal resorptions as compared with control embryos. Finally, we demonstrate that impairment of glucose metabolism via iodoacetate, a glyceraldehyde-3-phosphate dehydrogenase inhibitor, is sufficient to induce apoptosis in both blastocysts and trophoblast stem cells. Moreover, blastocysts treated with iodoacetate result in poor pregnancy outcome as determined by embryo transfer experiments. Taken together these data demonstrate the critical importance of the PI3K pathway in preimplantation embryo survival and pregnancy outcome and further emphasize the importance of glucose utilization and metabolism in cell survival pathways.

Mutagenic and Teratogenic Effects of Indomethacin and Cyclosporine-A on Dams and Fetuses of Mice.

Eighty pregnant female mice were alloted among 16 groups. The animals were given intraperitoneal injection equalized to the therapeutic doses used for human. Indomethacin was given (25, 75 mg/kg b.wt.) and cyclosporine-A was given (5, 10, 15 mg/kg b.wt.). They induced significant increase in fetal resorption and significant decrease in fetal body weight. Also, reduction in the sizes of the skeletons of embryos was observed. Dosage of 150 mg/kg b.wt. of indomethacin was the lethal dose to dams. Various chromosomal aberrations in maternal bone marrow cells and embryos cells and mitotic activity were recorded, quantitated and statistically analyzed. Indomethacin (75 mg/kg) induced more chromosomal aberrations in both pregnant females as well as fetuses than the lower dose (25 mg/kg). Cyclosporine-A (15 mg/kg b.wt.) induced an increase in chromosomal aberrations in bone marrow cells of female mice than lower dose (5, 10 mg/kg b.wt.), while no significant differences were observed between the (5, 10 mg/kg) doses. Also, cyclosporine-A (15 mg/kg) showed highly significant increase in chromosomal aberrations in embryos than medium (10 mg/kg) or lower doses (5 mg/kg).

Monoclonal antibodies against a dopamine-releasing protein (DARP) arrest fetal development, decrease brain catecholamines, and increase adrenal weight of neonatal rats

Monoclonal antibodies (MAb) were produced against a partially purified dopamine-releasing protein (DARP) to examine the feasible physiological role of this factor during the development of the rat. These IgM isotype MAbs, and not other IgM antibodies (controls), induced a remarkable increase in fetal resorption and stillbirth rate and impaired the developmental increase of catecholamine concentration in the corpus striatum and hypothalamus of newborn rats. Neonatal injections of the anti-DARP MAb (a single injection of 200 μg, 24 h after birth or 40 μg on alternate days during the first 10 days) decreased the dopamine (DA) concentration of the corpus striatum by 30% on Day 10 and 15% on Day 25 and drastically impaired (by 43% on Day 25) the developmental increase in hypothalamic DA. Furthermore, in the hypothalamus, there was a marked decrease in epinephrine and an increase in norepinephrine (NE) concentration, suggesting an impairment in PNMT function. Neonatal anti-DARP injections also resulted in increased adrenal weight (45 and 44% on Days 10 and 25, respectively) and elevated NE content (anti-DARP, 315 ± 12 ng, vs control, 223 ± 16 ng, n = 6). Intrafetal injection of anti-DARP MAb (40 μg) on E 17 resulted in increased resorption and stillbirth accounting for 83% fetal loss. A 10-μg dose of the antibody produced 33% of fetal resorption or stillborn fetuses, whereas control injections resulted in only 4.4% of fetal loss. These data strongly suggest that DARP may be a neurotrophic factor involved in the growth and differentiation of central catecholaminergic neurons and probably necessary for the maturation of PNMT during the early development of rat brain.

Teratological assessment of the antiprotozoal, diminazene diaceturate, in rats

Diminazene diaceturate was dissolved in deionized water and administered to pregnant rats by oral gavage once daily on days 8–15 of pregnancy at dose levels of 0, 100, 250, 500 and 1000 mg/kg. On day 21 of pregnancy, the dams were killed and the number of implants, resorptions and live fetuses counted. All fetuses were examined by routine teratological method. A significant increase in fetal resorptions and decrease in fetal body weights were found at the 1000 mg/kg dose. No significant increase in the incidence of anomalous fetuses was observed in external, skeletal and internal examinations even at the maternally toxic dose of 1000 mg/kg. Thus, these data indicate that diminazene diaceturate is not teratogenic in rats.

Embryolethal and teratogenic effects of bromofenofos in rats.

Bromofenofos, an organophosphorus anthelmintic, was administered by gavage to rats as a single dose (50 mg/kg) on one of days 6 through 14 of pregnancy. The dams were killed on day 21, and the fetuses were removed, weighed and examined by routine teratological methods. A significant increase in fetal resorptions occurred after administration on days 9 through 13, with a maximum on day 10. Approximately 72% of the implants were resorbed after administration on day 10. Fetal body weights were significantly decreased when dams were treated on day 8 or later. The greatest decrease in fetal body weights was observed on day 10, when the fetuses weighed less than the controls by about 44% on the average. The incidence of fetuses with gross, skeletal and internal malformations was significantly increased on days 8 through 10, on days 8 through 11 and on days 8 and 9, respectively. Although various types of malformations were observed, most of them occurred on day 8, when no significant increase in fetal resorptions did occur. Cleft lip, short tail, brachygnathia, anal atresia, absence of genital tubercle, fused pelvic legs and perineal testicles were seen on day 8 as gross malformations. Skeletal malformations mainly affected the vertebrae and ribs. Major internal malformations on day 8 were hydronephrosis, hydroureter, anophthalmia, cleft palate, agenesis of the bladder and renal agenesis. Anophthalmia and/or microphthalmia were observed on days 8 through 10, with the highest incidence on day 9.(ABSTRACT TRUNCATED AT 250 WORDS)

A total of 305 timed-pregnant Sprague-Dawley rats were exposed for 6 h a day on each of three consecutive days in one of three periods, i.e., pregnancy days 14-16, 11-13, or 8-10, either to 0.55 times the minimum alveolar concentration (MAC) of nitrous oxide (75%) or to 0.75 MAC of halothane (0.8%), isoflurane (1.05%) or enflurane (1.65%); an additional 232 positive-control (retinoic acid) and air control rats were studied. Reproductive indices were determined, and the 5178 offspring delivered at cesarean section were examined for external, internal, and skeletal abnormalities. There were no major or minor teratologic effects in anesthetic treated groups, although several developmental variants were observed in halothane- and enflurane-treated groups. Nitrous oxide exposure on days 14-16 resulted in a three-fold increase in fetal resorptions. The results suggest that the volatile anesthetics are not teratogenic and confirm that nitrous oxide may be associated with increased reproductive loss.

Teratology evaluation of methyl tertiary butyl ether in rats and mice.

Mated CD Sprague-Dawley rats and CD-1 mice were exposed during the period of organogenesis to target concentrations of 0, 250, 1000, and 2500 ppm methyl t-butyl ether (MTBE). None of the control or test-group animals died during the treatment or posttreatment periods. Females were sacrificed on d 20 (rats) or d 18 (mice). No adverse effects of treatment were reflected in maternal parameters of body weight, water consumption, or liver weight or in physical examination data for either species. Food consumption fell in the groups of treated rats during d 9-12; similar but nonsignificant effects were observed for mice during d 12-15. In rats, no treatment-related changes were recorded in the uterine implantation data, fetal size parameters, or fetal sex distribution data. Examination of fetuses for external abnormalities, skeletal malformations, or ossification variations did not reveal any changes caused by MTBE exposure. A slight increase in fetal resorptions was observed in the groups of mice exposed to low and high concentrations; this increase was attributed to two females in each group that had an unusually high number of resorptions, rather than to the treatment itself. No significant effects were observed in any groups of treated mice on external and soft-tissue examination or evaluation of skeletal abnormalities or ossification variations. The incidence of fused sternebrae in the high-concentration group increased slightly, which might be attributed to fetotoxicity.

Effect of chlorine and monochloramine in drinking water on the developing rat fetus

AbstractChlorine dioxide (ClO2) or monochloramine (NH2Cl) may be considered as an alternative to chlorine (HOCl) as a disinfectant in public water supplies, since chlorination produces trihalomethanes. This study was conducted to determine the effect of chlorine and monochloramine on rat fetuses. Female rats were administered 0, 1, 10 or 100 mg l−1 NH2Cl or HOCl daily in the drinking water for 2 1/2 months prior to and throughout gestation. Rats were sacrificed on Day 20 of gestation and fetuses were preserved for soft‐tissue and skeletal examination. No significant increase in fetal resorptions was found in any treated group. A slight increase in skeletal variants (such as incompletely ossified or missing sternebrae, or rudimentary ribs) was seen in 10 and 100 mg l−1 HOCl groups. Soft‐tissue defects, mainly improper orientation of the heart or adrenal agenesis, were observed in the 100 mg l−1 HOCl group. The percentage of total defects (skeletal and soft‐tissue) was increased significantly over control in the 100 mg l−1 HOCl group; however, monochloramine did not produce any significant changes in rat fetuses at any dose level.

Pregnancy and fetal development in hamsters treated with prostaglandin F 2α

The influence of PGF 2α on pregnancy and fetal outcome was investigated in the hamster. Following subcutaneous treatment with 50, 100, 200, 400, and 800 μg PGF 2α prenatal loss was significantly increased only at the highest dose level. The offspring of the treated animals were all alive and normal. Fetal weight was not affected. However, following intravenous injection of 100 and 200 μg PGF 2α there was a significant reduction in fetal weight, and at the 400 μg dose level an increase in fetal resorptions. All fetuses recovered were alive and normal. These results indicate that PGF 2α is not teratogenic in hamsters despite the apparent greater sensitivity of the hamster embryo to prostaglandin.

Pregnancy failure in hypophysectomized rats following LH-RH administration

Daily LH-RH administration induced a dose related increase in fetal resorption in rats following hypophysectomy on Day 11 or Day 12 of pregnancy. Ovarian and adrenal weights, as well as serum progesterone levels, were significantly decreased by Day 18. Serum progesterone was also significantly lower than control in those animals that remained pregnant after receiving LH-RH post-hypophysectomy. These observations suggest that LH-RH exerts an anti-pregnancy effect via a placental:ovarian axis in this species.

Synergistic teratogenic effects of arsenic and hyperthermia in hamsters

Similar malformations of the central nervous system including encephaloceles and exencephaly are induced by treatment of pregnant hamsters with sodium arsenate or hyperthermia on the eight day of gestation. These two treatments are synergistic in that combinations of minimal teratogenic levels of each cause a marked increase in fetal resorptions and the frequency and severity of developmental malformations as compared to separate treatments. The possible relationship of maternal hyperpyrexia and environmental teratogens to congenital malformations is discussed.

Oxonic acid and fetal development: II. Teratogenicity in rats

Feeding the uricase inhibitor potassium oxonate (KOx) as 3% of the diet to pregnant rats beginning on the 8th day of gestation for either 2 or 3 days caused a time-related increase in fetal resorptions, with the incidence in the experimental group treated for the additional day being 72% higher than the 2-day group. The shorter treatment period permitted more rat fetuses to survive to term; however 9.3% of these showed gross malformations, exencephaly and/or visceral herniation. Thus, KOx is demonstrated to be both embryotoxic and teratologic in the rat.
