Phytohormonal balance and differential gene expression in chronically irradiated Scots pine populations from the chernobyl affected zone.

The impact of chronic radiation exposure on phytohormone content and expression of phytohormone- and stress-related genes of Scots pine in the zone affected by the Chernobyl accident was studied. Needle samples were collected from three plots with contrasting levels of radioactive contamination in the Polesye State Radiation-Ecological Reserve, Republic of Belarus, and two reference plots in the Kozeluzhsky forest in June 2022. The experimental plots were located within the artificial plantations of Scots pine established in 1982, before the accident in 1986. The activity of radionuclides 137Cs, 90Sr, 241Am, 238Pu, and 239+240Pu in soil and needles ensured dose rates ranging from 3.3 to 87 mGy × year-1, while at the reference plots, the range was 0.7‒0.8 mGy × year-1. Concentrations of plant hormones, including indole-3-acetic acid (IAA), indole-3-butyric acid (IBA), zeatin, and abscisic acid (ABA) in needles were evaluated using high-performance liquid chromatography (HPLC). We demonstrate that chronic radiation exposure is a significant stress factor that affects both phytohormonal balance and the expression of some important phytohormone- and stress-related genes. We found a tendency toward decreased ABA and auxin concentrations in trees from plots contaminated with radionuclides. The ratio (IAA + IBA + zeatin)/ABA was drastically raised at the most contaminated plots Masany and Kulazhin, reflecting the functional rearrangements of cellular metabolism that ensure plant adaptation under chronic radiation exposure. Changes in gene expression indicated modulation of ABA and Ca2+ signalling pathways, decreased potential of zeatin biosynthesis, and activation of heat shock proteins biosynthesis.

The correlation between embryo rescue and hormonal changes in seedless grapes.

After normal pollination and fertilization of pseudoparthenocarpic seedless grapes, their embryos often stop developing due to certain developmental factors, resulting in embryo abortion. Hybrid breeding using seedless grapes as the maternal parent requires embryo rescue breeding technology. This technology plays a crucial role in seedless grape breeding. Although previous studies have extensively explored this technology, knowledge regarding its impact on embryo abortion and the effectiveness of rescue techniques remains limited. This study aimed to investigate the correlation between embryo rescue and hormonal changes in seedless grapes. Four Eurasian seedless grape cultivars, "Thompson Seedless," "Flame Seedless," "Heshi Seedless," and "Ruby Seedless," were selected for examination. We investigated endogenous hormone levels, including indole-3-acetic acid (IAA), gibberellic acid (GA3), and abscisic acid (ABA), in both berries and in vitro ovules during the most suitable embryo rescue time for these cultivars. Based on the observed fluctuations in endogenous hormone levels and previous research findings, appropriate doses of exogenous hormones, such as IAA, GA3, and ABA, were applied during seedless grape embryo rescue. The results indicated significant differences in endogenous hormone levels between berries with varying ovule counts of the same cultivar and ovules cultured in vitro, suggesting a hormonal influence on ovule abortion and embryo development in seedless grapes. Further research has identified effective ratios of exogenous hormones: 30 mg·L-1 IAA + 30 mg·L-1 ABA for berry ovule development, 1.0 mg·L-1 IAA + 2.0mg·L-1 6-benzylaminopurine (6-BA) + 1.0 mg·L-1 GA3 + 1.0 mg·L-1 ABA for in vitro ovule development, and 1.0 mg·L-1 IAA + 2.0 mg·L-1 6-BA + 1.0 mg·L-1 GA3 for embryo germination and seedling formation. In summary, hormonal changes significantly influence ovule and embryo development and are closely related to seedless grape embryo rescue breeding. This study deepened our understanding of the correlation between seedless grape embryo rescue and hormonal changes. It also resulted in the successful production of a batch of embryo rescue seedlings, further improving embryo rescue breeding technology and providing new germplasm materials for developing new seedless grape cultivars.

In Vitro Morphogenesis of Tobacco: Modulation of Endogenous Growth Regulators by Tulsi (Holy Basil).

Plant growth regulators (PGRs) play a vital role in the induction of morphogenesis in vitro. Synthetic PGRs are commonly used to induce organogenesis and somatic embryogenesis from various explants, while natural substances are rarely utilized. This study aimed to enhance the regenerative response in Nicotiana tabacum leaf explants using Tulsi (Ocimum sanctum) leaf extract and to elucidate the biochemical interactions during modulation of endogenous plant growth regulators, including indole-3-acetic acid (IAA), abscisic acid (ABA), zeatin, and 6-(γ, γ-dimethylallylamino) purine (2iP). Tulsi leaf extract significantly improved shoot production through interactions between endogenous hormones and those present in the extract, which enhanced stress mitigation. The 20% Tulsi leaf extract treatment produced significantly more shoots than the control, coinciding with increased endogenous IAA and zeatin levels starting on day 10 in culture. Furthermore, ABA and zeatin concentrations increased on days 15 and 25, respectively, in the 20% Tulsi extract treatment, suggesting their role in the induction of somatic embryo-like structures. ABA likely acts as an activator of stress responses, encouraging the development of these structures. Additionally, 2iP was involved in the induction of both forms of regeneration in the 10% and 20% extract treatments, especially in combination with ABA. These results suggest that Tulsi leaf extract holds promising potential as a natural supplement for increasing plant regeneration in vitro and advancing our understanding of how natural extracts of plant origin can be harnessed to optimize plant regeneration processes in vitro.

Transcriptome and Metabolome Reveal Key Genes from the Plant Hormone Signal Transduction Pathway Regulating Plant Height and Leaf Size in Capsicum baccatum.

Plant structure-related agronomic traits like plant height and leaf size are critical for growth, development, and crop yield. Defining the types of genes involved in regulating plant structure size is essential for the molecular-assisted breeding of peppers. This research conducted comparative transcriptome analyses using Capsicum baccatum germplasm HNUCB0112 and HNUCB0222 and their F2 generation as materials. A total of 6574 differentially expressed genes (DEGs) were detected, which contain 379 differentially expressed transcription factors, mainly including transcription factor families such as TCP, WRKY, AUX/IAA, and MYB. Seven classes of DEGs were annotated in the plant hormone signal transduction pathway, including indole acetic acid (IAA), gibberellin (GA), cytokinin (CK), abscisic acid (ABA), jasmonic acid (JA), ethylene (ET), and salicylic acid (SA). The 26 modules were obtained by WGCNA analysis, and the MEpink module was positively correlated with plant height and leaf size, and hub genes associated with plant height and leaf size were anticipated. Differential genes were verified by qRT-PCR, which was consistent with the RNA-Seq results, demonstrating the accuracy of the sequencing results. These results enhance our understanding of the developmental regulatory networks governing pepper key traits like plant height and leaf size and offer new information for future research on the pepper plant architecture system.

Panchagavya-Derived Brevibacillus brevis S1-3: Insights from the Draft Genome on its Antimicrobial and Plant Growth-Promoting Ability

Panchagavya has traditionally been used in Indian Ayurvedic practices because of its pro-agricultural and medicinal properties. This study presents the draft genome of a new Brevibacillus brevis S1-3 strain isolated from the fermented product Panchagavya. Through whole-genome sequencing, we determined that the genome of B. brevis S1-3 was 6,348,716 base pairs with a GC content of 54.3%. Genome assembly revealed the presence of 6107 protein-coding genes, 186 tRNA genes, and 13 rRNA genes. Genome annotation and analysis identified the genes involved in metabolism and other cellular processes. We also predicted the presence of several gene clusters associated with plant growth promotion, including indole acetic acid (IAA), gibberellic acid, ammonia, and nitrogen. Our study also revealed the genes responsible for the production of secondary metabolites that displayed a significant correlation with antimicrobial activity. Our results provide new insights into the genomic basis of the plant growth-promoting abilities of B. brevis and pave the way for further research in this field.

Plant growth regulators improve the growth and physiology of transplanted Thalassia Hemprichii fragments

IntroductionThe transplantation of seagrass fragments with shoots and rhizomes is the most common method for the ecological restoration of damaged seagrass meadows.MethodsThe aim of this study was to explore the effects of exogenous addition (10 mg per month for 3 months) of three commonly used plant growth regulators (PGRs), including indoleacetic acid (IAA), gibberellin (GA3), and paclobutrazol (PP333) on the growth and physiology of transplanted Thalassia hemprichii fragments (TTFs), with and without the rhizome apex (RA), using a simulation test. [Results] IAA and GA3 elevated the photosynthetic capacity and growth rate of TTF leaves but did not significantly alter leaf length and width. In contrast, PP333 reduced the leaf photosynthetic capacity and growth rate, while greatly increasing the leaf width and root viability. Additionally, PP333 treatment led to an increase in antioxidant enzyme activities (peroxidase [POD], superoxide dismutase [SOD], and catalase [CAT]), as well as malondialdehyde (MDA) and total phenol contents in TTFs, indicating some stress effects on the seagrass. Furthermore, IAA and GA3 decreased soluble sugar and protein contents and increased starch content in TTF tissues, whereas PP333 treatment elevated both nonstructural carbohydrate and soluble protein contents. The presence of RA positively affected the growth and physiology of T. hemprichii compared with TTFs without RA. There was a significant interaction between the PGRs and RA on leaf growth rate, chlorophyll fluorescence variables, and photosynthetic pigment content of the TTFs (p < 0.05).DiscussionThis study provides guidelines for the application of PGRs during the transplantation of T. hemprichii fragments for the restoration of seagrass meadows.

Foliar application of plant growth regulators for enhancing heavy metal phytoextraction efficiency by Sedum alfredii Hance in contaminated soils: Lab to field experiments

The phytoremediation efficiency of plants in removing the heavy metals (HMs) might be influenced by their growth status and accumulation capacity of plants. Herein, we conducted a lab-scale experiment and a field try out to assess the optimal plant growth regulators (PGRs) including indole-3-acetic acid (IAA)/brassinolide (BR)/abscisic acid (ABA) in improving the phytoextraction potential of Sedum alfredii Hance (S. alfredii). The results of pot experiment revealed that application of IAA at 0.2 mg/L, BR at 0.4 mg/L, and ABA at 0.2 mg/L demonstrated notable potential as optimal dosage for Cd/Pb/Zn phytoextraction in S. alfredii. The findings of subcellular level of Cd/Pb/Zn in leaves showed that IAA (0.2 mg/L), BR (0.4 mg/L) or ABA (0.2 mg/L) promoted the HMs storage in the soluble and cell wall fraction, therefore contributing HMs subcellular compartmentation. In addition, application of PGRs notably enhanced the antioxidant system (SOD, CAT, POD, APX activities) while reducing lipid peroxidation (MDA content) in S. alfredii, consequently improving HMs tolerance and growth of S. alfredii. Moreover, the results of field trial showed that application of BR, IAA, or ABA+BR substantially improved the growth of S. alfredii by inducing plants biomass and augmenting the levels of photosynthetic pigment contents. Notably, ABA+BR noticed the highest theoretical biomass by 42.9 %, followed by IAA (41.6 %), and BR (36.4 %), as compared with CK. Additionally, ABA+BR treatment showed effectiveness in removing the Cd by 103.4 %, while BR and IAA led to a significant increase of Pb and Zn removal by 239 % and 116 %, respectively, when compared with CK. Overall, the results of this study highlights that the foliar application of IAA, BR, or ABA+BR can serve as viable strategy to boosting phytoremediation efficiency of S. alfredii in contaminated soil by improving the biomass and metal accumulation in harvestable parts.

Isolation and identification of Rhizospheric and Endophytic Bacteria from Cucumber plants irrigated with wastewater: Exploring their roles in plant growth promotion and disease suppression

Wastewater contains various emerging contaminants, including heavy metals, residues of pesticides, and pharmaceuticals. Therefore, irrigation with wastewater can enhance heavy metal contamination in soil and adversely affect plant growth. To mitigate this problem, plant growth-promoting bacteria (PGPR) can improve plant growth under heavy metal stress. This study aimed to isolate and characterize rhizospheric and endophytic bacteria from the rhizosphere soil and roots of a cucumber plant irrigated with municipal wastewater. A total of 121 morphologically distinct bacterial isolates from the rhizosphere and 90 bacterial isolates from the endophytic region were isolated and tested for heavy metal resistance and in vitro plant growth-promoting characteristics, including indole-3-acetic acid (IAA) production, phosphate solubilization, Hydrogen Cyanide (HCN) production, and siderophore production. Most of the bacteria analyzed from the rhizospheric and endophytic regions showed various plant growth-promoting characteristics and were tolerant to different heavy metals at various concentrations. Bacterial strains R1 (Proteus sp.) and E2 (Bacillus sp.) were antagonistic to Fusarium oxysporum f. sp. Lycopersici. Wastewater irrigation increases heavy metal-resistant bacteria in cucumber plants, which can alleviate heavy metal stress. Additionally, Proteus sp. and Bacillus sp. isolates are potential candidates for removing heavy metal-contaminated soil and could be potential biofertilizer candidates for selected plants and biocontrol agents.

Plant Growth Promotion Using Bacillus cereus.

Plant growth-promoting bacteria (PGPB) appear to be a sensible competitor to conventional fertilization, including mineral fertilizers and chemical plant protection products. Undoubtedly, one of the most interesting bacteria exhibiting plant-stimulating traits is, more widely known as a pathogen, Bacillus cereus. To date, several environmentally safe strains of B. cereus have been isolated and described, including B. cereus WSE01, MEN8, YL6, SA1, ALT1, ERBP, GGBSTD1, AK1, AR156, C1L, and T4S. These strains have been studied under growth chamber, greenhouse, and field conditions and have shown many significant traits, including indole-3-acetic acid (IAA) and aminocyclopropane-1-carboxylic acid (ACC) deaminase production or phosphate solubilization, which allows direct plant growth promotion. It includes an increase in biometrics traits, chemical element content (e.g., N, P, and K), and biologically active substances content or activity, e.g., antioxidant enzymes and total soluble sugar. Hence, B. cereus has supported the growth of plant species such as soybean, maize, rice, and wheat. Importantly, some B. cereus strains can also promote plant growth under abiotic stresses, including drought, salinity, and heavy metal pollution. In addition, B. cereus strains produced extracellular enzymes and antibiotic lipopeptides or triggered induced systemic resistance, which allows indirect stimulation of plant growth. As far as biocontrol is concerned, these PGPB can suppress the development of agriculturally important phytopathogens, including bacterial phytopathogens (e.g., Pseudomonas syringae, Pectobacterium carotovorum, and Ralstonia solanacearum), fungal phytopathogens (e.g., Fusarium oxysporum, Botrytis cinerea, and Rhizoctonia solani), and other phytopathogenic organisms (e.g., Meloidogyne incognita (Nematoda) and Plasmodiophora brassicae (Protozoa)). In conclusion, it should be noted that there are still few studies on the effectiveness of B. cereus under field conditions, particularly, there is a lack of comprehensive analyses comparing the PGP effects of B. cereus and mineral fertilizers, which should be reduced in favor of decreasing the use of mineral fertilizers. It is also worth mentioning that there are still very few studies on the impact of B. cereus on the indigenous microbiota and its persistence after application to soil. Further studies would help to understand the interactions between B. cereus and indigenous microbiota, subsequently contributing to increasing its effectiveness in promoting plant growth.

Draft genome of Raoultella planticola, a high lead resistance bacterium from industrial wastewater

Isolation of heavy metals-resistant bacteria from their original habitat is a crucial step in bioremediation. Six lead (Pb) resistant bacterial strains were isolated and identified utilizing 16S rRNA to be Enterobacter ludwigii FACU 4, Shigella flexneri FACU, Microbacterium paraoxydans FACU, Klebsiella pneumoniae subsp. pneumonia FACU, Raoultella planticola FACU 3 and Staphylococcus xylosus FACU. It was determined that all these strains had their Minimum inhibitory concentration (MIC) to be 2500 ppm except R. planticola FACU 3 has a higher maximum tolerance concentration (MTC) up to 2700 ppm. We evaluated the survival of all six strains on lead stress, the efficiency of biosorption and lead uptake. It was found that R. planticola FACU 3 is the highest MTC and S. xylosus FACU was the lowest MTC in this evaluation. Therefore, transmission electron microscopy (TEM) confirmed the difference between the morphological responses of these two strains to lead stress. These findings led to explore more about the genome of R. planticola FACU 3 using illumine Miseq technology. Draft genome sequence analysis revealed the genome size of 5,648,460 bp and G + C content 55.8% and identified 5526 CDS, 75 tRNA and 4 rRNA. Sequencing technology facilitated the identification of about 47 genes related to resistance to many heavy metals including lead, arsenic, zinc, mercury, nickel, silver and chromium of R. planticola FACU 3 strain. Moreover, genome sequencing identified plant growth-promoting genes (PGPGs) including indole acetic acid (IAA) production, phosphate solubilization, phenazine production, trehalose metabolism and 4-hydroxybenzoate production genes and a lot of antibiotic-resistant genes.

Plant Growth Promotion and Biocontrol of Leaf Blight Caused by Nigrospora sphaerica on Passion Fruit by Endophytic Bacillus subtilis Strain GUCC4.

Passion fruit (Passiflora edulis Sims) is widely cultivated in tropic and sub-tropic regions for the production of fruit, flowers, cosmetics, and for pharmacological applications. Its high economic, nutritional, and medical values elicit the market demand, and the growing areas are rapidly increasing. Leaf blight caused by Nigrospora sphaerica is a new and emerging disease of passion fruit in Guizhou, in southwest China, where the unique karst mountainous landscape and climate conditions are considered potential areas of expansion for passion fruit production. Bacillus species are the most common biocontrol and plant-growth-promotion bacteria (PGPB) resources in agricultural systems. However, little is known about the endophytic existence of Bacillus spp. in the passion fruit phyllosphere as well as their potential as biocontrol agents and PGPB. In this study, 44 endophytic strains were isolated from 15 healthy passion fruit leaves, obtained from Guangxi province, China. Through purification and molecular identification, 42 of the isolates were ascribed to Bacillus species. Their inhibitory activity against N. sphaerica was tested in vitro. Eleven endophytic Bacillus spp. strains inhibited the pathogen by >65%. All of them produced biocontrol- and plant-growth-promotion-related metabolites, including indole-3-acetic acid (IAA), protease, cellulase, phosphatase, and solubilized phosphate. Furthermore, the plant growth promotion traits of the above 11 endophytic Bacillus strains were tested on passion fruit seedlings. One isolate, coded B. subtilis GUCC4, significantly increased passion fruit stem diameter, plant height, leaf length, leaf surface, fresh weight, and dry weight. In addition, B. subtilis GUCC4 reduced the proline content, which indicated its potential to positively regulate passion fruit biochemical properties and resulted in plant growth promotion effects. Finally, the biocontrol efficiencies of B. subtilis GUCC4 against N. sphaerica were determined in vivo under greenhouse conditions. Similarly to the fungicide mancozeb and to a commercial B. subtilis-based biofungicide, B. subtilis GUCC4 significantly reduced disease severity. These results suggest that B. subtilis GUCC4 has great potential as a biological control agent and as PGPB on passion fruit.

Dominance of Bacillus species in the wheat (Triticum aestivum L.) rhizosphere and their plant growth promoting potential under salt stress conditions.

Wheat (Triticum aestivum L.) is a major source of calorific intake in its various forms and is considered one of the most important staple foods. Improved wheat productivity can contribute substantially to addressing food security in the coming decades. Soil salinity is the most serious limiting factor in crop production and fertilizer use efficiency. In this study, 11 bacteria were isolated from wheat rhizosphere and examined for salt tolerance ability. WGT1, WGT2, WGT3, WGT6, WGT8, and WGT11 were able to tolerate NaCl salinity up to 4%. Bacterial isolates were characterized in vitro for plant growth-promoting properties including indole-3-acetic acid (IAA) production, phosphate solubilization, nitrogen fixation, zinc solubilization, biofilm formation, and cellulase-pectinase production. Six isolates, WGT1, WGT3, WGT4, WGT6, WGT8, and WGT9 showed IAA production ability ranging from 0.7-6 µg m/L. WGT8 displayed the highest IAA production. Five isolates, WGT1, WGT2, WGT5, WGT10, and WGT11, demonstrated phosphate solubilization ranging from 1.4-12.3 µg m/L. WGT2 showed the highest phosphate solubilization. Nitrogen fixation was shown by only two isolates, WGT1 and WGT8. Zinc solubilization was shown by WGT1 and WGT11 on minimal media. All isolates showed biofilm formation ability, where WGT4 exhibited maximum potential. Cellulase production ability was noticed in WGT1, WGT2, WGT4, and WGT5, while pectinase production was observed in WGT2 and WGT3. Phylogenetic identification of potential bacteria isolates confirmed their close relationship with various species of the genus Bacillus. WGT1, WGT2, and WGT3 showed the highest similarity with B. cereus, WGT6 with B. tianshenii, WGT8 with B. subtilis, and WGT11 with B. thuringiensis. Biofertilizer characteristics of salt-tolerant potential rhizospheric bacteria were evaluated by inoculating wheat plants under controlled conditions and field experiments. B. cereus WGT1 and B. thuringiensis WGT11 displayed the maximum potential to increase plant growth parameters and enhance grain yield by 37% and 31%, respectively. Potential bacteria of this study can tolerate salt stress, have the ability to produce plant growth promoting substances under salt stress and contribute significantly to enhance wheat grain yield. These bacterial isolates have the potential to be used as biofertilizers for improved wheat production under salinity conditions and contribute to the sustainable agriculture.

Low red/far-red ratio can induce cytokinin degradation resulting in the inhibition of tillering in wheat (Triticum aestivum L.).

Shoot branching is inhibited by a low red/far-red ratio (R/FR). Prior studies have shown that the R/FR suppressed Arabidopsis thaliana branching by promotes bud abscisic acid (ABA) accumulation directly. Given that wheat tiller buds are wrapped in leaf sheaths and may not respond rapidly to a R/FR, systemic cytokinin (CTK) may be more critical. Here, systemic hormonal signals including indole-3-acetic acid (IAA), gibberellins (GA) and CTK and bud ABA signals in wheat were tested under a low R/FR. The results showed that a low R/FR reduced the percentage of tiller occurrence of tiller IV and the tiller number per plant. The low R/FR did not rapidly induced ABA accumulation in the tiller IV because of the protection of the leaf sheath and had little effect on IAA content and signaling in the tiller nodes. The significant change in the CTK levels was observed earlier than those of other hormone (ABA, IAA and GA) and exogenous cytokinin restored the CTK levels and tiller number per plant under low R/FR conditions. Further analysis revealed that the decrease in cytokinin levels was mainly associated with upregulation of cytokinin degradation genes (TaCKX5, TaCKX11) in tiller nodes. In addition, exposure to a decreased R/FR upregulated the expression of GA biosynthesis genes (TaGA20ox1, TaGA3ox2), resulting in elevated GA levels, which might further promote CTK degradation in tiller nodes and inhibit tillering. Therefore, our results provide evidence that the enhancement of cytokinin degradation is a novel mechanism underlying the wheat tillering response to a low R/FR.

Whole-genome analysis revealed the growth-promoting mechanism of endophytic bacterial strain Q2H1 in potato plants.

Endophytes are non-pathogenic inhabitants of healthy plant tissues and have been found to promote plant growth and health. The endophytic bacterial strain Q2H1 was isolated from the roots of the potato and was identified to exhibit growth-promoting effects in potato plants. Whole-genome sequencing was performed to reveal the mechanism underlying its growth-promoting effect. The obtained sequencing data of approximately 5.65 MB encompassed 5,533 coding sequences. Of note, nine secondary metabolite gene clusters, including siderophore gene clusters, closely associated with plant growth promotion (PGP) were predicted by antiSMASH software. Comparative genomic analysis revealed that Q2H1 belongs to the genus Peribacillus. By gene function annotation, those genes related to plant growth-promoting activities, including indole-3-acetic acid (IAA) synthesis in tryptophan metabolism, siderophore biosynthetic activity, phosphate solubilization, nitrogen fixation, and related genes, were summarized. IAA (14.4 μg/ml) was presumptively produced by Q2H1 using the Salkowski colorimetric method. A total of five genes, namely, phoU, pstB, pstA1, pstC, and pstS, were annotated for phosphate solubilization, which is associated with the ability of the Q2H1 strain to solubilize phosphate under in vitro conditions. It is revealed that genes in the Q2H1 genome associated with nitrogen fixation belonged to three groups, namely, nitrogen fixation (nifU, sufU, salA, and nifS), nitrogen metabolism (nirA, nrtB, and nasA), and glutamate synthesis (glnA, gltB, gltD, and gudB), supported by evidence that Q2H1 grew on medium without nitrogen. We have also identified a siderophore gene cluster located on the chromosome of Q2H1, including seven genes (viz., rbsR, rhbf, rhbE, rhbD, rhbC, rhbA, ddc, and an unknown gene). In the in vitro assay, a prominent brown circle around the colony was produced on the chrome azurol S medium at 48 and 72 h post-inoculation, indicating that the siderophore gene cluster in Q2H1 harbored the ability to produce siderophores. In summary, these findings implied that identifying strain-specific genes for their metabolic pathways in bacterial endophytes may reveal a variety of significant functions of plant growth-promoting mechanisms.

Comprehensive Analysis of NAC Genes Reveals Differential Expression Patterns in Response to Pst DC3000 and Their Overlapping Expression Pattern during PTI and ETI in Tomato.

NAC (NAM/ATAF/CUC) transcription factors belong to a unique gene family in plants, which play vital roles in regulating diverse biological processes, including growth, development, senescence, and in response to biotic and abiotic stresses. Tomato (Solanum lycopersicum), as the most highly valued vegetable and fruit crop worldwide, is constantly attacked by Pseudomonas syringae pv. tomato DC3000 (Pst DC3000), causing huge losses in production. Thus, it is essential to conduct a comprehensive identification of the SlNAC genes involved in response to Pst DC3000 in tomato. In this study, a complete overview of this gene family in tomato is presented, including genome localization, protein domain architectures, physical and chemical features, and nuclear location score. Phylogenetic analysis identified 20 SlNAC genes as putative stress-responsive genes, named SSlNAC 1-20. Expression profiles analysis revealed that 18 of these 20 SSlNAC genes were significantly induced in defense response to Pst DC3000 stress. Furthermore, the RNA-seq data were mined and analyzed, and the results revealed the expression pattern of the 20 SSlNAC genes in response to Pst DC3000 during the PTI and ETI. Among them, SSlNAC3, SSlNAC4, SSlNAC7, SSlNAC8, SSlNAC12, SSlNAC17, and SSlNAC19 were up-regulated against Pst DC3000 during PTI and ETI, which suggested that these genes may participate in both the PTI and ETI pathway during the interaction between tomato and Pst DC3000. In addition, SSlNAC genes induced by exogenous hormones, including indole-3-acetic acid (IAA), abscisic acid (ABA), salicylic acid (SA), and methyl jasmonic acid (MeJA), were also recovered. These results implied that SSlNAC genes may participate in the Pst DC3000 stress response by multiple regulatory pathways of the phytohormones. In all, this study provides important clues for further functional analysis and of the regulatory mechanism of SSlNAC genes under Pst DC3000 stress.

Influence of Plant Growth Regulators and Artificial Light on the Growth and Accumulation of Inulin of Dedifferentiated Chicory (Cichorium intybus L.) Callus Cells.

Chicory (Chicorium intybus L.) is a perennial herb of the family Asteraceae, widely distributed in Asia and Europe, commonly used industrially as a raw material for extracting inulin because of a high content of inulin and biologically active compounds. Light conditions and plant growth regulators (PGRs) are two of many factors that affect the growth and inulin content of chicory callus. The aim of this work is to study the effect of PGRs and light conditions on proliferation and accumulation of inulin of chicory callus in vitro. In this study, we used semi-solid MS medium supplemented with different auxins (including Indole-3-acetic acid (IAA), naphthylacetic acid (NAA), and 2,4-dichlorophenoxyacetic acid (2,4-D)) at a concentration of 5.5–9.5 mg/L in combination with 2.0 mg/L 6 benzylaminopurine (BA) to determine induction and proliferation of callus. The increasing value of callus fresh weight was used to assess the growth of the callus in treatments. The results showed that a steady increase in callus fresh weight and inulin content in callus cells was obtained when they were cultured on MS medium supplemented with a combination of 2.0 mg/L BA with 7.5 mg/L IAA in lighting conditions with radiation equalized by the flux density of photosynthetic photons and ratios of radiation levels in the region of FR—far red > R—red. Increasing demand for organic inulin sources in production practice can be met by our finding.

Plant growth promoting activities of P solubilizing bacteria and their impact on disease resistance in groundnut, <em>Arachis hypogaea</em> L. against soil borne fungal pathogens

Plant growth promoting (PGP) activities of soil bacteria directly help plants in taking up the nutrients, attuning the growth hormones and indirectly safeguard by inhibiting diverse groups of fungal pathogens. In this study, we explored the native P solubilizing bacteria (PSB) isolated from the acid soils (pH < 5.5) of Odisha for selection of efficient PGPR with antifungal potential. Five PSB strainswere checked for their P solubilization efficiencies with Ca3(PO4)2,AlPO4,FePO4 and Fe3(PO4)2. The bioconversion of P by all the five strains in the broth medium followed the order Ca-P > Fe(III)-P > Fe(II)-P >Al-P. The strains interestingly showed potential plant growthpromoting properties including indole acetic acid (IAA) andsiderophore production in in vitro tests. These five strains also exhibited antifungal activities against fungal pathogens( Pythium aphanidermatum, Fusarium oxysporum, Pythium debaryanum, Thanatephorus cucumeris and Aspergillus niger )of groundnut. A field study was carried out with two of the above PSB strains [identified as Bacillus amyloliquefaciens (KT633845) and Burkholderia cepacia (KT717633)] with groundnut. Both the stains significantly influenced the plantgrowth (plant height, nodule no. and nodule dry weight) and pod yield. However, these two strains inoculated along withdoses of inorganic phosphate (SSP, single super phosphate) resulted in significantly higher pod yield as well as residual soil P.Additionally; the prevalence of both seedling mortality and plant mortality due to collar rot and stem rot were found to bereduced significantly in the inoculated plots. The findings substantiate the growth promoting ability of the two P solubilizingstrains, and thus qualifies to be used as biofertilizers either alone or as components of INM practices

Phosphate-solubilising bacteria promote horticultural plant growth through phosphate solubilisation and phytohormone regulation

ABSTRACT Microorganisms in the plant's rhizosphere are capable of solubilising insoluble soil phosphates and can promote plant growth. In this study, a total of 30 bacteria were isolated from soils in Changji city, and one highly efficient strain (B5) was identified as a member of Pseudomonadaceae. B5 exhibited phosphorus (P) solubilisation of 519.66 mg/L in a culture medium containing tricalcium phosphate (Ca3 (PO4)2). Results showed that strain B5 had a high P utilisation rate in a medium containing glucose (monosaccharide), ammonium sulphate and tricalcium phosphate. B5 treatment significantly promoted plant growth and increased the dry weight, the chlorophyll content as well as P content of tomato, marigold and cucumber. Moreover, B5 produced plant growth-promoting hormones, including indole-3-acetic acid (IAA) (4.18 g/L) and gibberellin (GA) (19.8 mg/L). In addition, organic acids including oxalic acid, succinic acid, gluconic acid, arabic-hexanoic acid and glutraic acid were produced by B5 under different phosphorus sources. Our results indicated that B5 is a potential candidate for development as a bioinoculant for horticultural application.

Effect of ion source polarity and dopants on the detection of auxin plant hormones by ion mobility-mass spectrometry.

Ion mobility spectrometry (IMS) equipped with a corona discharge (CD) ion source was used for measurement of three auxin plant hormones including indole-3-acetic acid (IAA), indole-3-propionic acid (IPA), and indole-3-butyric acid (IBA). The measurements were performed in both positive and negative polarities of the CD ion source. Dopant gases NH3, CCl4, and CHBr3 were used to modify the ionization mechanism. A time-of-flight mass spectrometer (TOFMS) orthogonal to the IMS cell was used for identification of the product ions. Density functional theory was used to rationalize formation of the ions, theoretically. The mixtures of the auxins were analyzed by CD-IMS. The separation performance depended on the ion polarity and the dopants. In the positive polarity without dopants, auxins were ionized via protonation and three distinguished peaks were observed. Application of NH3 dopant resulted in two ionization channels, protonation, and NH4+ attachment leading to peak overlapping. In the negative polarity, two ionization reactions were operative, via deprotonation and O2- attachment. The separation of the monomer peaks was not achieved while the peaks of anionic dimers [2M-H]- were separated well. The best LOD (4ng) was obtained in negative polarity with CCl4 dopant. Methylation (esterification) of IAA improved LODs by about one order.

Biostimulant Activity of <i>Sargassum</i> sp. Extracts on Early Growth of <i>Zea mays</i> L. and the Phytohormones Content Analysis

Seaweed has been gaining global interest in agriculture for the development of marine-based plant biostimulants. This research aimed to study the effect of three different liquid extracts of Sargassum sp., acidic, alkaline, and water extract, on the germination and early growth of maize and to evaluate the phytohormones content responsible for the growth. Phytohormones content including Indole-3-acetic acid (IAA), gibberellins (GA), kinetin and zeatin were analyzed using high-performance liquid chromatography (HPLC) and bioassay was performed twice on maize. Parameters observed on the bioassay were germination percentage, number of roots, shoot length, shoot weight and root weight under 4 different concentrations with 0.5; 1.5; 3.5; and 5% in the first bioassay and 3.5% concentration in the second bioassay. Both bioassays following randomized complete design and the data were analyzed using one-way ANOVA using post hoc test of Duncan Multiple Range Test (DMRT) at error probability of 5% in Genestat software. Phytohormones content in the seaweed extract indicated that alkaline extract was rich in IAA, gibberellin, and zeatin content, while water extract showed the highest kinetin content. The first bioassay indicated that lower concentration of the seaweed extracts gave better growth in all extracts, therefore a 3.5% concentration was chosen for the second bioassay with higher replication for each treatment. The second bioassay confirmed alkaline extract resulted in the highest germination while the highest seedling height, number of roots, shoot and root weight were resulted from acidic extract treatment. In conclusion, Sargassum sp. extracts obtained from acidic, alkaline, and water-based extraction methods, were able to improve the shoot and root growth of maize plants. The acidic extract showed the highest growth promotion among other extracts with the lowest phytohormones content.
