Microbial- and Plant-Derived Bioactive Peptides and Their Applications against Foodborne Pathogens: Current Status and Future Prospects.

Bioactive peptides (BAPs) obtained from plants and microbes have been thoroughly explored and studied due to their prophylactic properties. The use of BAPs seems to be a promising substitute for several currently available antibiotics because of their antimicrobial properties against foodborne pathogens. BAPs have several other useful properties including antitumor, antihypertensive, antioxidant, antiobesity, and antidiabetic activities. Nowadays, scientists have attempted to recombinantly synthesize bioactive peptides to study their characteristics and potential uses, since BAPs are not found in large quantities in nature. Many pathogenic microorganisms including foodborne pathogens are becoming resistant to various antibiotics. To combat these pathogens, scientists are working to find novel, innovative, and safe antimicrobial agents. Plant- and microbe-based BAPs have demonstrated noteworthy antimicrobial activity against a wide range of pathogenic microorganisms, including foodborne pathogens. BAPs can kill pathogenic microorganisms by disrupting membrane integrity, inhibiting DNA and RNA synthesis, preventing protein synthesis, blocking protein activity, or interacting with certain intracellular targets. In addition, the positive effect of BAP consumption extends to gut microbiota modulation and affects the equilibrium of reactive oxygen species in the gut. This article discusses recombinant BAPs, BAPs generated from plants and microbes, and their antimicrobial applications and modes of action for controlling foodborne pathogens.

Line image sensor-based colony fingerprinting system for rapid pathogenic bacteria identification

The rapid identification of pathogenic bacteria is crucial across various industries, including food or beverage manufacturing. Bacterial microcolony image-based classification has emerged as a promising approach to expedite identification, automate inspections, and reduce costs. However, conventional imaging methods have significant practical limitations, namely low throughput caused by the limited imaging range and slow imaging speed. To address these challenges, we developed an imaging system based on a line image sensor for rapid and wide-field imaging compared to existing colony imaging methods. This system can image a standard Petri dish (92 mm in diameter) completely within 22 s, successfully acquiring bacterial microcolony images. This process yielded a set of discrimination parameters termed as colony fingerprints, which were employed for machine learning. We demonstrated the performance of our system by identifying Staphylococcus aureus in food products using a machine learning model trained on a colony fingerprint dataset of 15 species from 9 genera, including foodborne pathogens. While conventional mass spectrometry-based methods require 24 h of incubation, our colony fingerprinting approach achieved 96% accuracy in just 10 h of incubation. Line image sensor offer high imaging speeds and scalability, allowing for swift and straightforward microbiological testing, eliminating the need for specialized expertise and overcoming the limitations of conventional methods. This innovation marks a transformative shift in industrial applications.

Host-Microbe Interactions Regulate Intestinal Stem Cells and Tissue Turnover in Drosophila.

With the activity of intestinal stem cells and continuous turnover, the gut epithelium is one of the most dynamic tissues in animals. Due to its simple yet conserved tissue structure and enteric cell composition as well as advanced genetic and histologic techniques, Drosophila serves as a valuable model system for investigating the regulation of intestinal stem cells. The Drosophila gut epithelium is in constant contact with indigenous microbiota and encounters externally introduced "non-self" substances, including foodborne pathogens. Therefore, in addition to its role in digestion and nutrient absorption, another essential function of the gut epithelium is to control the expansion of microbes while maintaining its structural integrity, necessitating a tissue turnover process involving intestinal stem cell activity. As a result, the microbiome and pathogens serve as important factors in regulating intestinal tissue turnover. In this manuscript, I discuss crucial discoveries revealing the interaction between gut microbes and the host's innate immune system, closely associated with the regulation of intestinal stem cell proliferation and differentiation, ultimately contributing to epithelial homeostasis.

Characterization of the Novel Leaderless Bacteriocin, Bawcin, from Bacillus wiedmannii

The rise of drug-resistant bacteria is a major threat to public health, highlighting the urgent need for new antimicrobial compounds and treatments. Bacteriocins, which are ribosomally synthesized antimicrobial peptides produced by bacteria, hold promise as alternatives to conventional antibiotics. In this study, we identified and characterized a novel leaderless bacteriocin, bawcin, the first bacteriocin to be characterized from a Bacillus wiedmannii species. Chemically synthesized and purified bawcin was shown to be active against a broad range of Gram-positive bacteria, including foodborne pathogens Staphylococcus aureus, Bacillus cereus, and Listeria monocytogenes. Stability screening revealed that bawcin is stable over a wide range of pH (2.0–10.0), temperature conditions (25–100 °C), and against the proteases, papain and pepsin. Lastly, three-dimensional structure homology modeling suggests that bawcin contains a saposin-fold with amphipathic helices and a highly cationic surface that may be critical for membrane interaction and the subsequent cell death of its targets. This study provides the foundational understanding of the activity and properties of bawcin, offering valuable insights into its applications across different antimicrobial uses, including as a natural preservative in food and livestock industries.

The Application of Hybridization Chain Reaction in the Detection of Foodborne Pathogens.

Today, with the globalization of the food trade progressing, food safety continues to warrant widespread attention. Foodborne diseases caused by contaminated food, including foodborne pathogens, seriously threaten public health and the economy. This has led to the development of more sensitive and accurate methods for detecting pathogenic bacteria. Many signal amplification techniques have been used to improve the sensitivity of foodborne pathogen detection. Among them, hybridization chain reaction (HCR), an isothermal nucleic acid hybridization signal amplification technique, has received increasing attention due to its enzyme-free and isothermal characteristics, and pathogenic bacteria detection methods using HCR for signal amplification have experienced rapid development in the last five years. In this review, we first describe the development of detection technologies for food contaminants represented by pathogens and introduce the fundamental principles, classifications, and characteristics of HCR. Furthermore, we highlight the application of various biosensors based on HCR nucleic acid amplification technology in detecting foodborne pathogens. Lastly, we summarize and offer insights into the prospects of HCR technology and its application in pathogen detection.

Impact of Acidulants on Salmonella and Escherichia coli O157:H7 in Water Microcosms Containing Organic Matter.

As demands for fresh water become more competitive between the processing plant and other consumers of water such as municipalities, interest has grown in recycling or reusing water for food processing. However, recycling the processing water from a poultry plant, for example, represents challenges due to increased organic loads and the presence of bacterial contaminants including foodborne pathogens. The objective in the current study was to evaluate the inactivation of Salmonella and E. coli O157:H7 using combinations (0.5% and 1%) of sodium bisulfate (SBS) and 1% lactic acid (LA) in water and water with organic matter in the form of horse blood serum (0.3%) with exposure times of 1 min and 5 min. Pathogen reductions after a 5 min exposure time were greater than corresponding reductions after a 1 min exposure time for all acid solutions. The Salmonella counts were significantly reduced (i.e., ≥1 log-unit) in all acid solutions after a 5 min exposure time with the combination of LA + SBS acid solutions being more effective than the corresponding 2% LA solutions. None of the acid solutions were effective in reducing the E. coli O157:H7 after a 1 min exposure time. The 1% LA + 1% SBS solution was the most effective acid solution against both pathogens and was the only acid solution effective in reducing E. coli O157:H7 by at least one log unit after 5 min of exposure.

Foodborne Clostridioides Species: Pathogenicity, Virulence and Biocontrol Options.

Clostridioides species possess many virulence factors and alarming levels of muti-drug resistance which make them a significant risk to public health safety and a causative agent of livestock disease. Clostridioides result in serious systemic and gastrointestinal diseases such as myonecrosis, colitis, food poisoning and gastroenteritis. As foodborne pathogens, Clostridioides species are associated with significant incidences of morbidity and mortality where the application of broad-spectrum antibiotics predisposes patients to virulent Clostridioides colonisation. As part of the One Health approach, there is an urgent need to eliminate the use of antibiotics in food production to safeguard animals, humans and the environment. Alternative options are warranted to control foodborne pathogens at all stages of food production. Antimicrobial peptides and bacteriophages have demonstrated efficacy against Clostridioides species and may offer antimicrobial biocontrol options. The bacteriocin nisin, for example, has been implemented as a biopreservative for the control of Listeria, Staphylococcus and Clostridia species in food. Bacteriophage preparations have also gained recognition for the antibacterial action against highly virulent bacterial species including foodborne pathogens. Studies are warranted to mitigate the formulation and administration limitations associated with the application of such antimicrobials as biocontrol strategies. This review outlines foodborne Clostridioides species, their virulence factors, and potential biocontrol options for application in food production.

Knowledge about foodborne pathogens and PCR‐based microbial detection methods among Brazilian students and professionals: A survey

AbstractPolymerase chain reaction (PCR) is widely used in several research areas and routine laboratory analyses, including foodborne pathogens detection. We aimed to investigate the knowledge about PCR‐based methods and foodborne pathogens among undergraduate and graduate Brazilian students and professionals. A cross‐sectional survey was carried out using an online questionnaire as data collection method. The questionnaire was validated and distributed through email and social networks. Data from 1246 respondents was collected. The knowledge scores were verified by correspondence analysis and discussed, 75.8% of the participants answered that they knew a foodborne pathogen and 71.4% of the participants answered that they did not study molecular biology techniques during undergraduate course. The highest level of knowledge was found among professionals with Masters' and PhD degrees. In conclusion, participants are not knowledgeable about PCR‐based methods and the level of academic training influences the knowledge of analytical foundations. Most participants did not study PCR and its application in detecting foodborne pathogens during undergraduate course. We suggested that undergraduate courses in Food Engineering and in Food Science and Technology include mandatory molecular biology classes in academic programs.

Antibiotic resistance in the pathogenic bacteria isolated from environmental samples of the food production facilities

Foodborne pathogens, such as Salmonella spp. and Listeria monocytogenes, may be present at various stages of production. The overuse of antibiotics over the past few decades has led to the emergence of many antibiotic-resistant bacteria, including foodborne pathogens. We investigated the sensitivity of Salmonella spp and Listeria monocytogenes to various antibiotics. Studied strains of L. monocytogenes and Salmonella spp. they showed resistance to tetracycline, which totaled 66.7%. High resistance of strains to ampicillin was also noted (57.14%). All strains were sensitive to amikacin, azithromycin, Meropenem, ciprofloxacin, moxifloxacin, norfloxacin. All strains of Salmonella spp. were resistant to ampicillin and penicillin G. Resistance to streptomycin and trimethoprim/sulfamethoxazole was 91.7%. All strains of L. monocytogenes were resistant to nalidixic acid. 22.2% of the strains showed resistance to tetracycline and cefepime. 11.1% of the strains were resistant to chloramphenicol and trimethoprim/sulfamethoxazole.

A survey of enteric disease agents in UK deer populations

Wild deer products have been linked to outbreaks of Escherichia coli 0157 in humans and bovine tuberculosis (bTB) in domestic cats, raising concerns around foodborne diseases from venison. This study investigated the prevalence of several enteric disease agents in UK deer populations, including foodborne pathogens and other diseases of import to livestock health. Intestinal samples were collected from slaughtered farmed deer (N = 211, eight farms), shot wild deer (N = 136) and ground faecal samples were collected from two farms (N = 90), six parks (N = 228) and five zoos (N = 67). DNA was extracted and multiplex qPCR assays were run to amplify targets of, C. perfringens toxins, Campylobacter spp., E. coli toxins, Mycobacterium avium subsp. Paratuberculosis (MAP), Salmonella spp. and Yersinia spp. The most commonly amplified targets were E. coli astA (61.2%) and Campylobacter spp. (43.3%), but the prevalence of C. coli and C. jejuni were ≤ 3.0% and Salmonella spp., MAP and Yersinia pseudotuberculosis also had low overall prevalences of 1.6%, 3.3% and 2.6%, respectively. However, notable targets included C. perfringens toxins α (20.4%) and β2 (16.9%); E. coli stx1 (14.6% overall, 28.0% in abattoir samples) and stx2 (17.8% overall, 31.6% in wild samples); and Yersinia enterocolitica (10.8% overall, 25% in wild samples). The low prevalence of some foodborne pathogens is reassuring for food safety; further investigations into the commonly found targets are needed to quantify the risk to public health and the possibility of wild deer acting as reservoirs of disease.

Impact of Heat Treatment on the Microbiological Quality of Frass Originating from Black Soldier Fly Larvae (Hermetia illucens).

Simple SummaryIndustrially produced insects can convert organic waste streams into proteins and other valuable products. The black soldier fly (Hermetia illucens) belongs to the most promising insect species. The residue, called frass, which remains after the production of the larvae, shows great potential as a soil improver and plant fertilizer, but it also contains a large amount of microorganisms, potentially including pathogenic species for humans. To minimize safety risks upon consumption of crops fertilized by this frass, a treatment that reduces pathogenic organisms, such as heating, is required. In this study, the impact of a heat treatment of 70 °C for 60 min (as prescribed by legislation) on frass of black soldier fly larvae was evaluated. The treatment resulted in a small reduction of the total microbial counts, and bacterial endospores were not at all reduced. However, when the foodborne pathogens Salmonella and Clostridium perfringens were added to the frass, the heat treatment led to undetectable amounts for both pathogens, as well as for Enterobacteriaceae. Consequently, a heat treatment of 70 °C for 60 min is likely appropriate to meet the microbiological criteria for the application of insect frass as biofertilizer or soil improver.Since black soldier fly larvae (BSFL, Hermetia illucens) are being produced at substantial volumes, concomitantly large amounts of the resulting by-product, called frass, are generated. This frass can potentially be applied as valuable plant fertilizer or soil improver. Since frass carries high microbial counts, potentially including foodborne pathogens, safety problems for consumers should be prevented. A heat treatment of 70 °C for 60 min is proposed to reduce harmful organisms in insect frass, based on EU regulations ((EU) No. 2021/1925). This study evaluated for the first time the impact of the proposed heat treatment on BSFL frass. This was done by applying the treatment on uninoculated frass as well as on frass inoculated with Salmonella or Clostridium perfringens at 5.0 log cfu/g. The heat treatment resulted in a reduction (maximum one log-cycle) of total viable counts and did not noticeably reduce bacterial endospores. In contrast, Enterobacteriaceae counts were reduced to below the detection limit (10 cfu/g). Heat treatment of inoculated frass resulted in absence of Salmonella in 25 g of frass and reduction of vegetative C. perfringens to below the detection limit (1 cfu/g). The proposed heat treatment appears to be appropriate to meet the microbiological regulations for insect frass.

Probiotic potential of commercial dairy-associated protective cultures: In vitro and in vivo protection against Listeria monocytogenes infection

Protective bacterial cultures (PCs) are commercially available to producers to control undesirable microbes in foods, including foodborne pathogens such as Listeria monocytogenes. They are generally recognized as safe for consumption and many are capable of producing bacteriocins. Yet their potential to act as probiotics and confer a health benefit on the host is not known. This study investigated the ability of three commercial PCs to survive human gastrointestinal conditions and exert anti-infective properties against L. monocytogenes. Counts of two PCs of Lactiplantibacillus plantarum remained unchanged after exposure to simulated gastrointestinal conditions, whereas counts of the PC Lactococcus lactis subsp. lactis were reduced by 5.3 log CFU/mL. Cultures of Lactiplantibacillus plantarum and Lactococcus lactis subsp. lactis adhered to human Caco-2 epithelial cells at ∼ 6 log CFU/mL. This pretreatment reduced subsequent L. monocytogenes adhesion and invasion by 1–1.6 log CFU/mL and 3.8–4.9 log CFU/mL, respectively, compared to control. L. monocytogenes-induced cytotoxicity was also reduced from 29.1% in untreated monolayers to ∼ 8% in those treated with PCs. Pretreatment of Caco-2 monolayers with Lactococcus lactis subsp. lactis and one PC of Lactiplantibacillus plantarum reduced L. monocytogenes translocation by ≥ 1.2 log CFU/mL compared to control (≥ 94.5% inhibition). All PCs significantly reduced DextranFITC permeability through Caco-2 monolayers to approximately half that of control. Pretreatment with PCs also reduced L. monocytogenes-induced mortality in Caenorhabditis elegans. These findings demonstrate the potential for commercially produced PCs to exert probiotic effects in the host through protection against L. monocytogenes infection, thus providing an additional benefit to food safety beyond inhibiting pathogen growth, survival, and virulence in foods.

Low-energy electron beam has severe impact on seedling development compared to cold atmospheric pressure plasma

Sprouts are germinated seeds that are often consumed due to their high nutritional content and health benefits. However, the conditions for germination strongly support the proliferation of present bacteria, including foodborne pathogens. Since sprouts are consumed raw or minimally processed, they are frequently linked to cases of food poisoning. Therefore, a seed decontamination method that provides efficient inactivation of microbial pathogens, while maintaining the germination capacity and quality of the seeds is in high demand. This study aimed to investigate and compare seed decontamination by cold atmospheric-pressure plasma and low-energy electron beam with respect to their impact on seed and seedling quality. The results show that both technologies provide great potential for inactivation of microorganisms on seeds, while cold plasma yielded a higher efficiency with 5 log units compared to a maximum of 3 log units after electron beam treatment. Both techniques accelerated seed germination, defined by the percentage of hypocotyl and leaf emergence at 3 days, with short plasma treatment (< 120 s) and all applied doses of electron beam treatment (8–60 kGy). However, even the lowest dose of electron beam treatment at 8 kGy in this study caused root abnormalities in seedlings, suggesting a detrimental effect on the seed tissue. Seeds treated with cold plasma had an eroded seed coat and increased seed wettability compared to electron beam treated seeds. However, these effects cannot explain the increase in the germination capacity of seeds as this was observed for both techniques. Future studies should focus on the investigation of the mechanisms causing accelerated seed germination and root abnormalities by characterizing the molecular and physiological impact of cold plasma and electron beam on seed tissue.

Computer simulation of submicron fluid flows in microfluidic chips and their applications in food analysis.

In recent years, countries around the world have maintained a zero-tolerance attitude toward safety problems in the food industry. In order to ensure human health, a fast, sensitive, and high-throughput analysis of food contaminants is necessary to ensure safe food products on the market. Microfluidics, as a high-efficiency and sensitive detection technology, has many advantages in the detection of food contaminants, including foodborne pathogens, pesticides, heavy metal ions, toxic substances, and so forth, especially in conjunction with a variety of submicron fluid driving methods, making food detection and analysis more efficient and accurate. This review introduces the principle of submicron fluid driving modes and discusses the driving simulation of submicron fluid in microfluidic chips. In addition, the latest developments in the application of simulation in food analysis from 2006 to 2020 are discussed, and the computer simulation of submicron fluid flow in microfluidic chips and its application and development trend in food analysis are also highlighted. The review indicates that microfluidic technology, using numerical simulation as an auxiliary tool, combined with traditional methods has greatly improved the detection and analysis of food products. In addition, microfluidics combined with a variety of control methods embodies the ability of specific, multifunctional, and sensitive detection and analysis of food products. The development of high-sensitivity, high-throughput, portable, integrated microfluidic chips will enable the technology to be applied in practice.

Antimicrobials from Seaweeds for Food Applications.

The exponential growth of emerging multidrug-resistant microorganisms, including foodborne pathogens affecting the shelf-life and quality of foods, has recently increased the needs of the food industry to search for novel, natural and eco-friendly antimicrobial agents. Macroalgae are a bio-diverse group distributed worldwide, known to produce multiple compounds of diverse chemical nature, different to those produced by terrestrial plants. These novel compounds have shown promising health benefits when incorporated into foods, including antimicrobial properties. This review aims to provide an overview of the general methods and novel compounds with antimicrobial properties recently isolated and characterized from macroalgae, emphasizing the molecular pathways of their antimicrobial mechanisms of action. The current scientific evidence on the use of macroalgae or macroalgal extracts to increase the shelf-life of foods and prevent the development of foodborne pathogens in real food products and their influence on the sensory attributes of multiple foods (i.e., meat, dairy, beverages, fish and bakery products) will also be discussed, together with the main challenges and future trends of the use of marine natural products as antimicrobials.

Crystal structure and site-directed mutagenesis of circular bacteriocin plantacyclin B21AG reveals cationic and aromatic residues important for antimicrobial activity

Plantacyclin B21AG is a circular bacteriocin produced by Lactiplantibacillus plantarum B21 which displays antimicrobial activity against various Gram-positive bacteria including foodborne pathogens, Listeria monocytogenes and Clostridium perfringens. It is a 58-amino acid cyclised antimicrobial peptide, with the N and C termini covalently linked together. The circular peptide backbone contributes to remarkable stability, conferring partial proteolytic resistance and structural integrity under a wide temperature and pH range. Here, we report the first crystal structure of a circular bacteriocin from a food grade Lactobacillus. The protein was crystallised using the hanging drop vapour diffusion method and the structure solved to a resolution of 1.8 Å. Sequence alignment against 18 previously characterised circular bacteriocins revealed the presence of conserved charged and aromatic residues. Alanine substitution mutagenesis validated the importance of these residues. Minimum inhibitory concentration analysis of these Ala mutants showed that Phe8Ala and Trp45Ala mutants displayed a 48- and 32-fold reduction in activity, compared to wild type. The Lys19Ala mutant displayed the weakest activity, with a 128-fold reduction. These experiments demonstrate the relative importance of aromatic and cationic residues for the antimicrobial activity of plantacyclin B21AG and by extension, other circular bacteriocins sharing these evolutionarily conserved residues.

Discovery and characterisation of circular bacteriocin plantacyclin B21AG from Lactiplantibacillus plantarum B21

Lactiplantibacillus plantarum B21 isolated from Vietnamese sausage (nem chua) has previously displayed broad antimicrobial activity against Gram-positive bacteria including foodborne pathogens Listeria monocytogenes and Clostridium perfringens. This study successfully identified the antimicrobial agent as plantacyclin B21AG, a 5668 Da circular bacteriocin demonstrating high thermostability, resistance to a wide range of pH, proteolytic resistance and temporal stability. We report a reverse genetics approach to identify and characterise plantacyclin B21AG from first principles. The bacteriocin was purified from culture supernatant by a three-step process consisting of concentration, n-butanol extraction and cation exchange chromatography. A de novo peptide sequencing using LC-MS/MS techniques identified two putative peptide fragments which were mapped to the genome sequence of L. plantarum B21. This revealed an ORF corresponding to a putative circular bacteriocin with a 33-amino acid leader peptide and a 58-amino acid mature peptide encoded on a native plasmid pB21AG01. The bacteriocin is shown to be a small cationic predominantly α-helical protein (69%). The corresponding gene cluster, consisted of seven genes associated with post-translational circularisation, immunity and secretion. Whilst plantacyclin B21AG is 86% identical to the newly published plantaricyclin A it is more highly cationic having a net charge of +3 due to an additional basic residue in the putative membrane interaction region. This and other substitutions may well go some way to explaining functional differences. The robust nature of plantacyclin B21AG, its antimicrobial activity and associated machinery for cyclisation make it an interesting biotechnological target for development, both as a food-safe antimicrobial or potentially a platform technology for recombinant protein circularisation.

Microorganisms Move a Short Distance into an Almond Orchard from an Adjacent Upwind Poultry Operation.

Over a 2-year period, drag swabs of orchard soil surface and air, soil, and almond leaf samples were collected in an almond orchard adjacent to (35 m from the first row of trees) and downwind from a poultry operation and in two almond orchards (controls) that were surrounded by other orchards. Samples were evaluated for aerobic plate count, generic Escherichia coli, other coliforms, the presence of Salmonella, bacterial community structure (analyzed through sequencing of the 16S rRNA gene), and amounts of dry solids (dust) on leaf surfaces on trees 0, 60, and 120 m into each orchard. E. coli was isolated from 41 of 206 (20%) and 1 of 207 (0.48%) air samples in the almond-poultry and control orchards, respectively. Salmonella was not isolated from any of the 529 samples evaluated. On average, the amount of dry solids on leaves collected from trees closest to the poultry operation was more than 2-fold greater than from trees 120 m into the orchard or from any of the trees in the control orchards. Members of the family Staphylococcaceae-often associated with poultry-were, on average, significantly (P < 0.001) more abundant in the phyllosphere of trees closest to the poultry operation (10% of relative abundance) than in trees 120 m into the orchard (1.7% relative abundance) or from any of the trees in control orchards (0.41% relative abundance). Poultry-associated microorganisms from a commercial operation transferred a short distance into an adjacent downwind almond orchard.IMPORTANCE The movement of microorganisms, including foodborne pathogens, from animal operations into adjacent plant crop-growing environments is not well characterized. This study provides evidence that dust and bioaerosols moved from a commercial poultry operation a short distance downwind into an almond orchard and altered the microbiome recovered from the leaves. These data provide growers with information they can use to assess food safety risks on their property.

Currently, leafy green salads are often consumed because they are considered as practical and healthy. However, during their preparation, both inadequate washing and contact with non-hygienic surfaces may increase their microbial load. This may cause several health problems for individuals consuming salads. The overuse of antibiotics has led to the emergence of multidrug resistant bacteria, including foodborne pathogens. Biofilm production ability of these pathogenic bacteria makes it difficult to treat infections caused by these pathogens. The aim of this study is to isolate and identify P. fluorescens from leafy green salads collected from different restaurants. A total of 72 isolates were isolated from leafy green salads, and 29 of these isolates were identified by PCR as Pseudomonas and 9 of them identified as P. fluorescens. All P. fluorescens isolates were resistant to ampicillin, amoxicillin, cefuroxime, ceftazidime and ceftriaxone antibiotics. The results of this study showed that additional attention for the hygiene conditions is needed during the preparation and storage stages of leafy green salads.

Emerging Technologies for Food Safety: High Pressure Processing (HPP) and Cold Plasma Technology (CPT) for Decontamination of Foods

Foods may become contaminated from a variety of sources, therefore it is imperative to understand and discover easy, cheap and effective means of decontaminating foods. Heat, although effective, economical and easily available, has been reported to produce undesirable effects on food such as loss of taste and nutrition. High Pressure Processing can inactivate the cells of the food borne pathogens and organisms responsible for food spoilage regardless of the temperature without making changes to the texture, color or flavor. Equipment involved in HPP includes a pressure vessel, pumps generating high hydrostatic pressure or intensifiers. Its success depends on certain factors such as pressure of water, temperature used during the treatment, and the properties and state of the food and categories of microorganisms found in food.Cold Plasma Technology (CPT) is a novel, non-thermal food processing technology that uses energetic and reactive gases to inactivate contaminating microorganisms in food products. CPT is environmentally friendly that uses natural gases such as nitrogen, argon, air, hydrogen, and oxygen. Depending on the plasma type, it can inactivate a wide range of microorganisms including food borne pathogens and spoilage organisms. This technology hasa low running cost (Cost of natural gases and electricity).Both HPP and CPT can spread and work on the entire food sample, regardless of shape and size.
