Imaging of micro-steps on as-grown surface of sapphire with X-ray phase contrast technique

Basal-faceted sapphire ribbons grown using the Stepanov–LaBelle technology have a low density of surface steps. We present our results on a study of steps on the surface of a ribbon, misoriented relative to the singular face (0001) by several arc minutes. The ribbon has been characterized by means of in-line phase contrast imaging technique at Pohang Light Source, South Korea. It was shown for the first time that a step height of about 1 μm can be determined directly from an image. The step height obtained using the phase contrast method was confirmed by atomic force microscopy measurements. We have found that the experimental contrast matches the theoretical simulations only if the calculated intensity profile has been convolved with a Gaussian function. The full width at half maximum of the Gaussian was independently got from previous measurements. We have obtained an analytical solution in the case of theoretical fully coherent phase contrast image. The inverse problem is easy to solve, since there is a direct proportionality between the contrast and the step height.

Direct Imaging of Radiation-Sensitive Organic Polymer-Based Nanocrystals at Sub-Ångström Resolution.

Seeing the atomic configuration of single organic nanoparticles at a sub-Å spatial resolution by transmission electron microscopy has been so far prevented by the high sensitivity of soft matter to radiation damage. This difficulty is related to the need to irradiate the particle with a total dose of a few electrons/Å2, not compatible with the electron beam density necessary to search the low-contrast nanoparticle, to control its drift, finely adjust the electron-optical conditions and particle orientation, and finally acquire an effective atomic-resolution image. On the other hand, the capability to study individual pristine nanoparticles, such as proteins, active pharmaceutical ingredients, and polymers, with peculiar sensitivity to the variation in the local structure, defects, and strain, would provide advancements in many fields, including materials science, medicine, biology, and pharmacology. Here, we report the direct sub-ångström-resolution imaging at room temperature of pristine unstained crystalline polymer-based nanoparticles. This result is obtained by combining low-dose in-line electron holography and phase-contrast imaging on state-of-the-art equipment, providing an effective tool for the quantitative sub-ångström imaging of soft matter.

A closer look at high-energy X-ray-induced bubble formation during soft tissue imaging.

Improving the scalability of tissue imaging throughput with bright, coherent X-rays requires identifying and mitigating artifacts resulting from the interactions between X-rays and matter. At synchrotron sources, long-term imaging of soft tissues in solution can result in gas bubble formation or cavitation, which dramatically compromises image quality and integrity of the samples. By combining in-line phase-contrast imaging with gas chromatography in real time, we were able to track the onset and evolution of high-energy X-ray-induced gas bubbles in ethanol-embedded soft tissue samples for tens of minutes (two to three times the typical scan times). We demonstrate quantitatively that vacuum degassing of the sample during preparation can significantly delay bubble formation, offering up to a twofold improvement in dose tolerance, depending on the tissue type. However, once nucleated, bubble growth is faster in degassed than undegassed samples, indicating their distinct metastable states at bubble onset. Gas chromatography analysis shows increased solvent vaporization concurrent with bubble formation, yet the quantities of dissolved gasses remain unchanged. By coupling features extracted from the radiographs with computational analysis of bubble characteristics, we uncover dose-controlled kinetics and nucleation site-specific growth. These hallmark signatures provide quantitative constraints on the driving mechanisms of bubble formation and growth. Overall, the observations highlight bubble formation as a critical yet often overlooked hurdle in upscaling X-ray imaging for biological tissues and soft materials and we offer an empirical foundation for their understanding and imaging protocol optimization. More importantly, our approaches establish a top-down scheme to decipher the complex, multiscale radiation-matter interactions in these applications.

Reduced volume of diabetic pancreatic islets in rodents detected by synchrotron X-ray phase-contrast microtomography and deep learning network

The pancreatic islet is a highly structured micro-organ that produces insulin in response to rising blood glucose. Here we develop a label-free and automatic imaging approach to visualize the islets in situ in diabetic rodents by the synchrotron radiation X-ray phase-contrast microtomography (SRμCT) at the ID17 station of the European Synchrotron Radiation Facility. The large-size images (3.2mm×15.97mm) were acquired in the pancreas in STZ-treated mice and diabetic GK rats. Each pancreas was dissected by 3000 reconstructed images. The image datasets were further analysed by a self-developed deep learning method, AA-Net. All islets in the pancreas were segmented and visualized by the three-dimension (3D) reconstruction. After quantifying the volumes of the islets, we found that the number of larger islets (=>1500 μm3) was reduced by 2-fold (wt 1004±94 vs GK 419±122, P<0.001) in chronically developed diabetic GK rat, while in STZ-treated diabetic mouse the large islets were decreased by half (189±33 vs 90±29, P<0.001) compared to the untreated mice. Our study provides a label-free tool for detecting and quantifying pancreatic islets in situ. It implies the possibility of monitoring the state of pancreatic islets in vivo diabetes without labelling.

Image quality and scan time optimisation for in situ phase contrast x-ray tomography of the intervertebral disc

In-line phase contrast synchrotron tomography combined with in situ mechanical loading enables the characterisation of soft tissue micromechanics via digital volume correlation (DVC) within whole organs. Optimising scan time is important for reducing radiation dose from multiple scans and to limit sample movement during acquisition. Also, although contrasted edges provided by in-line phase contrast tomography of soft tissues are useful for DVC, the effect of phase contrast imaging on its accuracy has yet to be investigated. Due to limited time at synchrotron facilities, scan parameters are often decided during imaging and their effect on DVC accuracy is not fully understood. Here, we used previously published data of intervertebral disc phase contrast tomography to evaluate the influence of i) fibrous image texture, ii) number of projections, iii) tomographic reconstruction method, and iv) phase contrast propagation distance on DVC results. A greater understanding of how image texture influences optimal DVC tracking was obtained by visualising objective function mapping, enabling tracking inaccuracies to be identified. When reducing the number of projections, DVC was minimally affected by image high frequency noise but with a compromise in accuracy. Iterative reconstruction methods improved image signal-to-noise and consequently significantly lowered DVC displacement uncertainty. Propagation distance was shown to affect DVC accuracy. Consistent DVC results were achieved within a propagation distance range which provided contrast to the smallest scale features, where; too short a distance provided insufficient features to track, whereas too long led to edge effect inconsistencies, particularly at greater deformations. Although limited to a single sample type and image setup, this study provides general guidelines for future investigations when optimising image quality and scan times for in situ phase contrast x-ray tomography of fibrous connective tissues.

In situ investigation on ultrasound (US)-generated bubbles' dynamics for membrane fouling control applications

The purpose of this research article is to gain a comprehensive understanding of the influence of ultrasound (US) power intensity on the stationary and moving microbubbles close to the membrane surface and its application on membrane fouling control. Compared to other imaging techniques, synchrotron in-line Phase Contrast Imaging technique, available at the biomedical imaging and therapy (BMIT) beamlines at the Canadian Light Source (CLS), allowed for in-situ quantitative analysis and real-time imaging of microbubbles in water inside a membrane filtration unit. An advanced non-invasive synchrotron propagation-based imaging (PBI) technique was employed to improve the contrast between the liquid and gas inside the bubble (interface of liquid and gas), which was crucial for the qualitative and quantitative analyses. ImageJ software was employed for the quantitative analyses of cavitational bubbles dynamics and their characteristics at different US power intensities. The bubble-related parameters, including the number, size, and fraction of total area occupied by bubbles, were determined for all captured images at three different US power intensities. Bubble characteristics at different times of US exposure and distances from the US transducer were also studied. Then, MATLAB software was used to generate the plots for the bubble size distribution and probability density for each image. The effect of US power intensity on bubble velocity, and its application in membrane fouling control was also investigated. The generated bubbles were mostly located in the middle of the images, close to the membrane surface, where the US waves were detected. It was also observed that the velocity of smaller bubbles was much larger than that of the larger bubbles; and they tended to move toward the US transducer. Bubble growth was much faster at 100 W than at lower US power intensities. The experimental results were in good agreement with the in-situ observation of microbubbles. The mass transfer across the membrane was improved as the US power increased from 25 W to 100 W. This was likely due to more frequent bubble explosions and energy releases into the water medium, causing turbulence flow, and resulting in more membrane surface cleaning.

Mixed scale dense convolutional networks for x-ray phase contrast imaging

X-ray in-line phase contrast imaging relies on the measurement of Fresnel diffraction intensity patterns due to the phase shift and the attenuation induced by the object. The recovery of phase and attenuation from one or several diffraction patterns is a nonlinear ill-posed inverse problem. In this work, we propose supervised learning approaches using mixed scale dense (MS-D) convolutional neural networks to simultaneously retrieve the phase and the attenuation from x-ray phase contrast images. This network architecture uses dilated convolutions to capture features at different image scales and densely connects all feature maps. The long range information in images becomes quickly available, and greater receptive field size can be obtained without losing resolution. This network architecture seems to account for the effect of the Fresnel operator very efficiently. We train the networks using simulated data of objects consisting of either homogeneous components, characterized by a fixed ratio of the induced refractive phase shifts and attenuation, or heterogeneous components, consisting of various materials. We also train the networks in the image domain by applying a simple initial reconstruction using the adjoint of the Fréchet derivative. We compare the results obtained with the MS-D network to reconstructions using U-Net, another popular network architecture, as well as to reconstructions using the contrast transfer function method, a direct phase and attenuation retrieval method based on linearization of the direct problem. The networks are evaluated using simulated noisy data as well as images acquired at NanoMAX (MAX IV, Lund, Sweden). In all cases, large improvements of the reconstruction errors are obtained on simulated data compared to the linearized method. Moreover, on experimental data, the networks improve the reconstruction quantitatively, improving the low-frequency behavior and the resolution.

Enhanced phase retrieval via deep concatenation networks for in-line X-ray phase contrast imaging

PurposeIn-line X-ray phase contrast imaging offers considerable additional information beyond that acquired from conventional absorption contrast X-ray imaging, showing promising potentials in clinical diagnosis, materials characterization and so on. Given the physically intractable factors tangled inside, conventional phase retrieval methods typically suffer from limited feasibility. A deep-learning-augmented reconstruction strategy is proposed to improve the phase retrieval in spatial resolution and noise compression. MethodsThe deep network is composed of a phase contrast refinement module and a phase retrieval module to stabilize and generalize the phase retrieval. The two modules are aggregated in a plug-and-play fashion with the final assembly finetuned using limited training data, essentially encouraging a semi-supervised training. Verification experiments were performed on simulated phase contrast images of histopathological images. The results were compared to those from conventional phase-attenuation duality method. ResultsThe deep-learning-augmented reconstruction strategy increases structural similarity and peak signal-to-noise ratio of phase retrieval result by more than 8% and 30%, and reduces root mean squared error by 46% compared with conventional phase-attenuation duality method. Conclusions: The pilot study of deep learning deployment in in-line X-ray phase-contrast imaging exhibit advantages against conventional methods in terms of spatial resolution and noise robustness.

Problems with Evaluation of Micro-Pore Size in Silicon Carbide Using Synchrotron X-ray Phase Contrast Imaging.

We report near- and far-field computer simulations of synchrotron X-ray phase-contrast images using a micropipe in a SiC crystal as a model system. Experimental images illustrate the theoretical results. The properties of nearly perfect single crystals of silicon carbide are strongly affected by μm-sized pores even if their distribution in a crystal bulk is sparse. A non-destructive technique to reveal the pores is in-line phase-contrast imaging with synchrotron radiation. A quantitative approach to evaluating pore sizes is the use of computer simulations of phase-contrast images. It was found that near-field phase-contrast images are formed at very short distances behind a sample. We estimated these distances for tiny pores. The Fresnel zones did not provide any information on the pore size in the far-field, but a contrast value within the first Fresnel zone could be used for simulations. Finally, general problems in evaluating a micro-pore size via image analysis are discussed.

In-situ synchrotron X-ray imaging of ultrasound (US)-generated bubbles: Influence of US frequency on microbubble cavitation for membrane fouling remediation

Gaining an in-depth understanding of the characteristics and dynamics of ultrasound (US)--generated bubbles is crucial to effectively remediate membrane fouling. The goal of present study is to conduct in-situ visualization of US-generated microbubbles in water to examine the influence of US frequency on the dynamics of microbubbles. This study utilized synchrotron in-line phase contrast imaging (In-line PCI) available at the biomedical imaging and therapy (BMIT) beamlines at the Canadian Light Source (CLS) to enhance the contrast of liquid/air interfaces at different US frequencies of 20, 28 and 40 KHz at 60 Watts. A high-speed camera was used to capture 2,000 frames per second of the bubble cavitation generated in water under the ultrasound influence. Key parameters at the polychromatic beamlines were optimized to maximize the phase contrast of gas/liquid of the microbubbles with a minimum size of 5.5 µm. ImageJ software was used to analyze the bubble characteristics and their behavior under the US exposure including the microbubble number, size, and fraction of the total area occupied by the bubbles at each US frequency. Furthermore, the bubble characteristics over the US exposure time and at different distances from the transducer were studied. The qualitative and quantitative data analyses showed that the microbubble number or size did not change over time; however, it was observed that most bubbles were created at the middle of the frames and close to the US field. The number of bubbles created under the US exposure increased with the frequency from 20 kHz to 40 kHz (about 4.6 times). However, larger bubbles were generated at 20 kHz such that the average bubble radius at 20 kHz was about 6.8 times of that at 40 kHz. Microbubble movement/traveling through water was monitored, and it was observed that the bubble velocity increased as the frequency was increased from 20 kHz to 40 kHz. The small bubbles moved faster, and the majority of them traveled upward towards the US transducer location. The growth pattern (a correlation between the mean growth ratio and the exposure time) of bubbles at 20 kHz and 60 W was obtained by tracking the oscillation of 22 representative microbubbles over the 700 ms of imaging. The mean growth ratio model was also obtained.

Dual-energy phase retrieval algorithm for inline phase sensitive x-ray imaging system.

Phase retrieval is vital for quantitative x-ray phase contrast imaging. This work presents an iterative method to simultaneously retrieve the x-ray absorption and phase images from a single x-ray exposure. The proposed approach uses the photon-counting detectors' energy-resolving capability in providing multiple spectrally resolved phase contrast images from a single x-ray exposure. The retrieval method is derived, presented, and experimentally tested with a multi-material phantom in an inline phase contrast imaging setup. By separating the contributions of photoelectric absorption and Compton scattering to the attenuation, the authors divide the phase contrast image into two portions, the attenuation map arises from photoelectric absorption and a pseudo phase contrast image generated by electron density. This way one can apply the Phase Attenuation Dualiby (PAD) algorithm and Fresnel propagation for the iteration. The retrieval results from the experimental images show that this iterative method is fast, accurate, robust against noise, and thus yields noticeable enhancement in contrast to noise ratios.

Iterative reconstruction algorithm based on discriminant adaptive-weighted TV regularization for fibrous biological tissues using in-line X-ray phase-contrast imaging.

In-line X-ray phase-contrast computed tomography (IL-PCCT) can produce high-contrast and high-resolution images of biological samples, and it has a great advantage with regard to imaging the microstructures and morphologies of fibrous biological tissues (FBTs). Filtered back projection (FBP) is widely used in ILPCCT. However, it requires long scanning times and high radiation doses to produce high-quality CT images, and this restricts its applicability in biomedical and preclinical studies on FBTs. To solve this problem, a novel IL-PCCT reconstruction algorithm is proposed to decrease the radiation dose by reducing the number of projections and reconstruct high-quality CT images of FBTs. The proposed algorithm incorporates the FBP method into the iterative reconstruction framework. Considering the area types and anisotropic edge properties of FBTs, a discriminant adaptive-weighted total variation model is introduced to optimize the intermediate reconstructed images. A fibrous phantom simulation and real experiment were performed to assess the performance of the proposed algorithm. Simulation and experimental results demonstrated that the proposed algorithm is an effective IL-PCCT reconstruction method for FBTs with incomplete projection data, and it has a great ability to suppress artifacts and preserve the edges of fibrous structures.

Three-dimensional construction of micrometer level in rat stomach by synchrotron radiation

BackgroundThe structural changes of gastric mucosa are considered as an important window of early gastric lesions. This article shows an imaging method of the stomach that does not use imaging agents. X-ray phase-contrast images of different stages of gastric development were taken using micrometer level X-ray in-line phase-contrast imaging (XILPCI) technique on synchrotron radiation facility. The aim of the study was to demonstrate that the imaging technique is an appropriate method for micron imaging of the gastric structures.MethodsThe stomachs of 4-, 6- and 12-week-old rats were removed and cleaned. XILPCI has 1000 times greater soft tissue contrast than that of X-ray traditional absorption radiography. The projection images of the rats stomachs were recorded by an XILPCI charge coupled device (CCD) at 9-μm image resolution.ResultsThe X-ray in-line phase-contrast images of the different stages of rats’ gastric specimens clearly showed the gastric architectures and the details of the gastro-duodenal region. 3-dimensional (3D) stomach anatomical structure images were reconstruction.ConclusionThe reconstructed gastric 3D images can clearly display the internal structure of the stomach. XILPCI may be a useful method for medical research in the future.

X-Ray Phase-Contrast Image Reconstruction Based on the Chambolle-Pock Algorithm

The X-ray in-line phase-contrast imaging technique requires that the light source provides a spatially coherent X-ray beam typically generated by a microfocus X-ray tube in most laboratories. However, its small focal spot limits the lightness. An arrayed X-ray point source can overcome this drawback and provide an adequately bright spatial coherent light. However, the arrayed source will introduce artefacts to the phase-contrast image due to the point source spatial distribution. To eliminate these artefacts, an optimization-based algorithm is used to reconstruct the image as an optimization problem. An iterative method was developed to numerically solve the optimization problem by using the first-order primal-dual algorithm. The simulations demonstrate its convergence and reconstruction accuracy.

Developing a Microbubble-Based Contrast Agent for Synchrotron In-Line Phase Contrast Imaging.

X-ray phase contrast imaging generates contrast from refraction of X-rays, enhancing soft tissue contrast compared to conventional absorption-based imaging. Our goal is to develop a contrast agent for X-ray in-line phase contrast imaging (PCI) based on ultrasound microbubbles (MBs), by assessing size, shell material, and concentration. Polydisperse perfluorobutane-core lipid-shelled MBs were synthesized and size separated into five groups between 1 and 10μm. We generated two size populations of polyvinyl-alcohol (PVA)-MBs, 2-3μm and 3-4μm, whose shells were either coated or integrated with iron oxide nanoparticles (SPIONs). Microbubbles were then embedded in agar at three concentrations: 5×107, 5×106 and 5×105 MBs/ml. In-line phase contrast imaging was performed at the Canadian Light Source with filtered white beam micro-computed tomography. Phase contrast intensity was measured by both counting detectable MBs, and comparing mean pixel values (MPV) in minimum and maximum intensity projections of the overall samples. Individual lipid-MBs 6-10μm, lipid-MBs 4-6μm and PVA-MBs coated with SPIONs were detectable at each concentration. At the highest concentration, lipid-MBs 6-10μm and 4-6μm showed an overall increase in positive contrast, whereas at a moderate concentration, only lipid-MBs 6-10μm displayed an increase. Negative contrast was also observed from two largest lipid-MBs at high concentration. These data indicate that lipid-MBs larger than 4μm are candidates for PCI, and 5×106 MBs/ml may be the lowest concentration suitable for generating visible phase contrast in vivo. Identifying a suitable MB for PCI may facilitate future clinical translation.

Methodology development and application of X-ray imaging beamline at SSRF

This paper introduces some latest developments regarding the X-ray imaging methodology and applications of the X-ray imaging and biomedical application beamline (BL13W1) at Shanghai Synchrotron Radiation Facility in the past 5 years. The photon energy range of the beamline is 8–72.5 keV. Several sets of X-ray imaging detectors with different pixel sizes (0.19–24 μm) are used to realize X-ray microcomputed tomography (X-ray micro-CT) and X-ray in-line phase-contrast imaging. To satisfy the requirements of user experiments, new X-ray imaging methods and image processing techniques are developed. In vivo dynamic micro-CT experiments with living insects are performed in 0.5 s (sampling rate of 2 Hz, 2 tomograms/s) with a monochromatic beam from a wiggler source and in 40 ms (sampling rate of 25 Hz, 25 tomograms/s) with a white beam from a bending magnet source. A new X-ray imaging method known as move contrast X-ray imaging is proposed, with which blood flow and moving tissues in raw images can be distinguished according to their moving frequencies in the time domain. Furthermore, X-ray speckle-tracking imaging with twice exposures to eliminate the edge enhancement effect is developed. A high-precision quantification method is realized to measure complex three-dimensional blood vessels obtained via X-ray micro-CT. X-ray imaging methods such as three-dimensional X-ray diffraction microscopy, small-angle X-ray scattering CT, and X-ray fluorescence CT are developed, in which the X-ray micro-CT imaging method is combined with other contrast mechanisms such as diffraction, scattering, and fluorescence contrasts respectively. Moreover, an X-ray nano-CT experiment is performed with a 100 nm spatial resolution. Typical user experimental results from the fields of material science, biomedicine, paleontology, physics, chemistry, and environmental science obtained on the beamline are provided.

In-Line Phase Contrast Mammography, Phase Contrast Digital Breast Tomosynthesis, and Phase Contrast Breast Computed Tomography With a Dedicated CT Scanner and a Microfocus X-Ray Tube: Experimental Phantom Study

We performed laboratory investigations of the image quality for 2-D [digital mammography (DM)] and 3-D [digital breast tomosynthesis (DBT) and computed tomography (CT)] in-line phase contrast imaging dedicated to the breast. We employed the cone-beam, microfocus CT scanner developed in-house. Planar imaging tests were carried out with BR50/50 and polymethylmethacrylate test objects, at tube voltages of 60 kV, 80 kV, and 100 kV. CT volume images, acquired at 80 kV at dose levels comparable to two-view mammography (mean glandular dose <inline-formula xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink"> <tex-math notation="LaTeX">$\cong ~4$ </tex-math></inline-formula> mGy), as well as at 120 kV, were reconstructed from phase contrast projection images as well as from the corresponding phase retrieved projection images (Paganin’s algorithm). We assumed 3-D spatial uniform distribution of the ratio <inline-formula xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink"> <tex-math notation="LaTeX">$\delta /\beta $ </tex-math></inline-formula> of the X-ray optical coefficients of the sample material, at the effective energy of the X-ray beam. In 2-D imaging, all phantom details showed higher contrast after phase retrieval. In particular, contrast-to-noise ratio between a simulated mass and phantom background in phase retrieved images resulted up to 6.5 times higher than for absorption-based images. CT and DBT phase retrieval images of a homogeneous 50/50 test object at 120 kV produced higher signal-to-noise ratio of microcalcifications than in phase contrast DBT and CT of the same object, at the same glandular dose. In CT reconstructed volume images, microcalcifications down to 0.230–0.290-mm size presented an increased visibility after phase retrieval, in a two-components 50/50 breast phantom. For the first time, the advantages of in-line phase imaging in microfocus breast imaging have been shown with one setup for all techniques (DM, DBT, and breast CT). These results motivate further research on in-line phase contrast 2-D and 3-D imaging of the breast with a microfocus X-ray tube for potential application in the clinic of in-line phase contrast DM, DBT, and breast CT at dose levels comparable with corresponding attenuation-based exams.

Tomographic reconstruction with a generative adversarial network.

This paper presents a deep learning algorithm for tomographic reconstruction (GANrec). The algorithm uses a generative adversarial network (GAN) to solve the inverse of the Radon transform directly. It works for independent sinograms without additional training steps. The GAN has been developed to fit the input sinogram with the model sinogram generated from the predicted reconstruction. Good quality reconstructions can be obtained during the minimization of the fitting errors. The reconstruction is a self-training procedure based on the physics model, instead of on training data. The algorithm showed significant improvements in the reconstruction accuracy, especially for missing-wedge tomography acquired at less than 180° rotational range. It was also validated by reconstructing a missing-wedge X-ray ptychographic tomography (PXCT) data set of a macroporous zeolite particle, for which only 51 projections over 70° could be collected. The GANrec recovered the 3D pore structure with reasonable quality for further analysis. This reconstruction concept can work universally for most of the ill-posed inverse problems if the forward model is well defined, such as phase retrieval of in-line phase-contrast imaging.

Synchrotron Radiation-Based Three-Dimensional Visualization of Angioarchitectural Remodeling in Hippocampus of Epileptic Rats.

Characterizing the three-dimensional (3D) morphological alterations of microvessels under both normal and seizure conditions is crucial for a better understanding of epilepsy. However, conventional imaging techniques cannot detect microvessels on micron/sub-micron scales without angiography. In this study, synchrotron radiation (SR)-based X-ray in-line phase-contrast imaging (ILPCI) and quantitative 3D characterization were used to acquire high-resolution, high-contrast images of rat brain tissue under both normal and seizure conditions. The number of blood microvessels was markedly increased on days 1 and 14, but decreased on day 60 after seizures. The surface area, diameter distribution, mean tortuosity, and number of bifurcations and network segments also showed similar trends. These pathological changes were confirmed by histological tests. Thus, SR-based ILPCI provides systematic and detailed views of cerebrovascular anatomy at the micron level without using contrast-enhancing agents. This holds considerable promise for better diagnosis and understanding of the pathogenesis and development of epilepsy.

Study of micropores in single crystals by in-line phase contrast imaging with synchrotron radiation

Some single crystals that are of importance for engineering contain micropores of various shapes, including micropipes, empty cylinders whose section sizes vary from fractions of a micrometer to several micrometers. We describe the properties of such objects in a silicon carbide single crystal and a method of observing and exploring these objects, namely, in-line phase contrast imaging with synchrotron radiation. We also review the history of the development and current status of this method.
