Accessibility of gonococcal and meningococcal surface antigens: immunogold labeling for quantitative electron microscopy

The parallel application of two electron microscopic immunogold labeling procedures was used to assess the surface exposure and accessibility of gonococcal and meningococcal surface antigens. Monoclonal antibodies were used as markers for the surface antigens, i.e., outer membrane proteins and lipooligosaccharides. To evaluate the labeling densities obtained after incubation of whole bacteria in suspension or ultrathin cryosections of bacteria, a method of electron microscopic quantitation was developed. Incubation of whole bacterial suspensions with monoclonal antibodies and protein A-gold resulted in specific labeling of the bacterial surfaces. However, the labeling densities varied largely in each cell. By contrast, cryosections showed uniform heavy labeling densities at the surface of the outer membranes of all cells. Apparently, by sectioning the cells the antigen-masking barrier could be evaded, and steric hindrance was no longer restrictive. Thus, a better estimate of both the presence and the surface exposure, i.e., the accessibility of antigens, could be made. Such information is essential for us to better understand host-bacterial interactions and to develop new vaccines.

Growth cone advance mediated by fibronectin-associated filopodia is inhibited by a phorbol ester tumor promoter

In serum-supplemented medium, exposure to the tumor promoter 4β-phorbol 12β-myristate 13α-acetate (PMA) increases the proportion of SH-SY5Y neuroblastoma cells with neurites and increases the average neurite length. In the present study, under serum-free conditions, PMA treatment had the opposite effects, i.e., retarded neurite sprouting and partially inhibited neurite elongation. This inhibition in neurite outgrowth was partially antagonized by the addition of serum fibronectin (FN) to the medium or substratum. In the absence of PMA, SH-SY5Y cells grown under serum-free conditions showed extensive neurite outgrowth as well as the capacity to secrete FN into their microenvironment and form FN-containing substratum-attachment sites. Immunogold labeling and whole mount transmission electron microscopy (WMTEM) demonstrated FN-containing contact pads at sites where filopodia attached to the substratum and focal plaques on the underside of growth cone margins. The appearance and abundance of FN-containing contact pads and focal plaques were increased by the addition of exogenous FN to defined medium. Focal plaques appeared in close association with microfilament bundles, and nearly always with bundles that projected into filopodia attached to the substratum by contact pads. A method for immunolabeling FN in the filopodial contact pads of living cultures provided more direct evidence that filopodia and contact pads have a major role in FN-mediated attachment and are central in determining growth cone shape and the rate and direction of advance. In support of this view, we show that PMA treatment retards neurite sprouting, alters growth cone morphology and motility, and eliminates the appearance of microfilament bundles, filopodia, and FN-containing substratum-attachment plaques.

Class I major histocompatibility complex antigen expression on early murine trophoblast and its induction by lymphokines in vitro. II. The role of gamma interferon in the responses of primary and secondary giant cells

The induction of paternal Class I and II MHC antigens by crude lymphokine preparations or purified recombinant gamma interferon was investigated on (C57BL/6 J × CBA/ H)F 1 primary and secondary trophoblast giant cell outgrowths from 3.5-day post-coital (pc) blastocyst and 7.5-day pc ectoplacental cone preparations, respectively, using sensitive immunogold labelling techniques and electron microscopy. Class I MHC (but not Class II) antigens could readily be induced on secondary trophoblast giant cells, by incubation in vitro with gamma interferon for 40 h. However, repeated attempts to induce detectable MHC antigens on primary trophoblast giant cells failed. Mock-treated (C57BL/6 J × CBA/ H)F 1 secondary trophoblast giant cell control preparations failed to express detectable MHC antigens. These findings suggest that, at the time of implantation, there is a time window during which MHC antigens are neither expressed constitutively nor are inducible by soluble factors which normally modulate cell surface MHC antigen concentration.

Demonstration of calicivirus in human faeces by immunosorbent and immunogold-labelling electron microscopy methods

Immunosorbent electron microscopy (ISEM) and immuno-gold staining (IGS) electron microscopy methods have been applied to human faeces, shown by direct electron microscopy (EM) to contain calicivirus. Caliciviruses were successfully trapped on grids coated with positive rabbit or human anti-sera against calicivirus, but not with negative sera. Caliciviruses were specifically labelled with gold particles, when treated with positive rabbit or human antisera against calicivirus followed by protein A-gold and goat anti-rabbit or anti-human IgG gold conjugates in an indirect method in suspension. Goat anti-human IgM gold complexes did not react with the available antisera. Attempts to label caliciviruses trapped on the grids were unsuccessful. No cross-reactions were observed with Norwalk agent-like particles, hepatitis A virus or poliovirus type 3 by any of the methods. The results indicate that ISEM and IGS may be useful techniques for detection and identification of small viruses present in low concentrations in faeces.

Analysis of antigenic determinants on internal and external proteins of influenza virus and identification of antigenic subpopulations of virions in recent field isolates using monoclonal antibodies and immunogold labelling

An electron microscopic immunogold labelling technique employing monoclonal antibodies has been applied to the antigenic analysis of influenza A and B viruses. Reassortant influenza A H3N2 viruses containing haemagglutinin molecules from viruses isolated between 1968 and 1982 were analysed with a panel of monoclonal antibodies raised against viruses which appeared over the same period. The immunogold labelling technique clearly demonstrated the antigenic drift in the haemagglutinin molecule that occurred between 1968 and 1982. When the technique was applied to the examination of viruses from a more geographically restricted influenza epidemic in a semi-closed community, antigenic variants were found. Furthermore the technique enabled the identification of distinct antigenic variant subpopulations within a single clinical isolate. Analysis of the HA of MDCK cell or egg grown virus by this procedure provided data to support the hypothesis that the host cell exerts selective pressure on subpopulations of virus resulting in the emergence of antigenic variants.
