A new synthesis of derivatives of imidazo[2,1-b]thiazole from 2-halogenoimidazoles has been effected. The reaction of 2-bromo-4,5-diphenylimidazoles with phenacyl bromide and its p-substituted derivatives has given the corresponding 2-bromo-1-phenylimidazoles, by the heating of which with thiourea and subsequent cyclization of the resulting 2-mercapto-1-phenacylimidazoles under the action of phosphorus oxychloride a number of 2,5,6-triarylimidazo[2,1-b]thiazoles has been synthesized.

The nature of the interaction of the phenyl and imidazole rings in the N-arylimidazoles

The ionization constants in 50% aqueous ethanol at 20‡ C and the rates of interaction with ethyl iodide in acetone at 50‡ C of the N-methyl, N-benzyl, and N-phenyl derivatives of imidazole and benzimidazole have been measured. It has been shown that in N-phenylbenzimidazole, the phenyl group departs from the plane of the molecule by an angle of about 40‡. Because of this, the influence of the N-phenyl group on the reactivity of the imidazole ring is greater in the imidazole series than in the benzimidazole series.

Investigations in the imidazole series. Synthesis of thiazolino-(3,2-α) benzimidazole and several of its derivatives

The reaction of 2-mercapto- and 5,6-dimethyl-2-mercaptobenzimidazoles with ethylene chloro (bro o)hydrins and 1,2-dichloro(dibromo)ethanes was studied. With dihaloethanes, the process proceeds in two directions, formation of thiazolino(3,2-a)benzimidazoles and 1,2-di-(2′-mercaptobenzimidazolyl) ethanes. Thiazolino(3,2-α)benzimidazole and several of its derivatives were synthesized by cyclization of 2-(β-chloroethyl)mercapto- and 1-(β-chloromethyl)-2-mercaptobenzimidazoles.

Studies of the imidazole series.

The bromination of 2-alkyl-4(5)-nitro-, 1,2-dialkyl-4-nitro-, and 1,2-dialkyl-5-nitroimidazoles was studied and at the same time a simple method was devised for obtaining 2-alkyl-4(5)-nitro-5(4)-bromo- and 1,2-dialkyl-4-nitro-5-bromoimidazoles. 1,2-Dialkyl-5-nitroimidazoles cannot be brominated. 1,2-Dialkyl-4-bromo-5-nitroimidazoles can be obtained by alkylation of 2-alkyl-4(5)-nitro-5(4)-bromoimidazoles by dialkyl sulfates.

Benz- and naphthazole research. XI. Formazanes of the naphth[1,2-d]imidazole series

Like 1-alkyl-2-hydrazinobenzimidazoles, 2-hydrazinonaphth[1,2-d]imidazole and 1-methyl- or 3-methyl-2-hydrazinonaphth[1, 2-d]imidazoles, when dissolved in dry alcohols, undergo autoxidation to 1, 5-dinaphth[1, 2-d]imidazolylformazanes.

KINETICS OF THE CATALYZED HYDROLYSIS OF p-NITROPHENYL ACETATE

Catalytic constants have been determined for the hydrolysis at 20 °C of p-nitrophenyl acetate in 9.56% (w/w) dioxane–water mixtures; catalysts used were hydroxide ions, imidazole and 2-methylimidazole, various substituted pyridines, serine, histidine and histidylhistidine. In the case of hydroxide ion catalysis, the rate constant falls off at high catalyst concentrations, and this is attributed to the establishment of an equilibrium involving the anion of the substrate. The results with the various catalysts indicate that there is no simple correlation between catalytic efficiency and pK or nucleophilicity. In the imidazole series there is evidence of steric hindrance when a group is present in a position next to the functional group. There is an indication of bifunctional catalysis in the hydrolysis by 2-(β-hydroxyethyl)pyridine and 4-(γ-hydroxypropyl)pyridine, both of which are much more effective than pyridine.

1) The strength of antispasmodic action on smooth muscles and toxicity of 1-β-diethylaminoethoxyphenyl-3-methyl-6, 7-methylenedioxyisoquinoline (I), derived from safrole, and its 3, 4-dihydro compound (II) were compared from which (II) was found to have weaker antispasmodic action but there was no significant difference between the toxicity of the two compounds. Analgesic action, thought to be the secondary symptoms of antispasmodic action, was examined in (II) by the modified Hardy method and an effect somewhat stronger than Pethidine Hydrochloride, by the same dose, was observed.2) 2-Methoxy-6-allyl-1-phenoxyethyldiallylamine, similar to (I) in chemical structure, possessed a strong action of uterine contraction and increased the tonus of smooth muscles. Its methiodide possesses the action of blocking the autonomous nerves but lost the uterine contracting action of the original compound, while it showed antispasmodic action similar to papaverine against the smooth muscles, i.e. actions similar to the methobromide of (I).3) Relationship between the chemical structure and pharmacological activities of safrole derivatives of isoquinoline, thiazole, and imidazole series was examined.
