Imaging of Stroke in Rodents Using a Clinical Scanner and Inductively Coupled Specially Designed Receiver Coils.

Imaging of small laboratory animals in clinical MRI scanners is feasible but challenging. Compared with dedicated preclinical systems, clinical scanners have relatively low B0 field (1.5-3.0 T) and gradient strength (40-60 mT/m). This work explored the use of wireless inductively coupled coils (ICCs) combined with appropriate pulse sequence parameters to overcome these two drawbacks, with a special emphasis on the optimization of the coil passive detuning circuit for this application. A Bengal rose photothrombotic stroke model was used to induce cortical infarction in rats and mice. Animals were imaged in a 3T scanner using T2 and T1-weighted sequences. In all animals, the ICCs allowed acquisition of high-quality images of the infarcted brain at acute and chronic stages. Images obtained with the ICCs showed a substantial increase in SNR compared to clinical coils (by factors of 6 in the rat brain and 16-17 in the mouse brain), and the absence of wires made the animal preparation workflow straightforward.

Small Animal PET as Non-Invasive Tool for Preclinical Imaging

Increasingly, in vivo imaging of small laboratory animals has become critical for preclinical biomedical research allowing the non-invasive assessment of biological and biochemical processes in living subjects. Many di erent positron labeled compounds have been synthesized as tracer that target a range of speci c markers or pathways yielding quantitative, spatially and temporally indexed information on normal and diseased tissues such as tumors. Because of its non-invasive nature, nuclear techniques also enable to repeat studies in a single subject, facilitating longitudinal assay designs of rodent models of human cancer, cardiovascular, neurological and other diseases over the entire process, from the inception to progression of the disease, and monitoring the e ectiveness of treatment or other interventions (with each animal serving as its own control and thereby reducing biological variability). This also serves to reduce the number of experimental animals required for a particular study. In contrast to cell or tissue culture-based experiments, studies in intact animals incorporate all the interacting physiological factors present in vivo.

Performance Evaluation of the Small-Animal nanoScan PET/MRI System

nanoScan is a high-resolution integrated system for consecutive PET and MR imaging of small laboratory animals. We evaluated the performance of the system, using the NEMA NU 4-2008 protocol for the PET component and the NEMA MS 1-2007, MS 2-2008, and MS 3-2007 standards for the MR imaging component. The imaging system uses magnetically shielded position-sensitive photomultiplier tubes and a compact 1-T permanent-magnet MR imaging platform. Spatial resolution, sensitivity, counting rate capabilities, and image quality parameters were evaluated in accordance with the aforementioned NEMA standards. Further in vivo evaluation experiments complement the physical validation results. The spatial resolution of the PET system enabled the 0.8-mm rods of a Derenzo phantom to be resolved. With point source and 2-dimensional filtered backprojection reconstruction, the resolution varied from 1.50 to 2.01 mm in full width at half maximum in the radial direction and from 1.32 to 1.65 mm in the tangential direction within the radius of 25 mm. Peak absolute sensitivity was 8.41%. Scatter fraction was 17.3% and 34.0%, and maximum noise-equivalent counting rate was 406 and 119 kcps in the mouselike and ratlike phantom, respectively. The image quality test found a nonuniformity of 3.52% and a spillover ratio of 6.2% and 5.8% in water and air, respectively. In testing of the MR imaging component, artifact-free images with high signal-to-noise ratio were recorded. Geometric distortion was below 5%, and image uniformity was at least 94.5% and 96.6% for the 60- and 35-mm radiofrequency coils, respectively. The nanoScan integrated small-animal PET/MR imaging system has excellent spatial resolution and sensitivity. The performance characteristics of the PET and the MR imaging components are not compromised as a result of their integration onto a single platform. Because of its combination of features and performance parameters, the system provides crucial advantages for preclinical imaging studies over existing PET/CT systems, especially in neurologic and oncologic research.

An Event-Driven Motion Correction Method for Neurological PET Studies of Awake Laboratory Animals

The purpose of the study is to investigate the feasibility of an event driven motion correction method for neurological microPET imaging of small laboratory animals in the fully awake state. A motion tracking technique was developed using an optical motion tracking system and light (<1g) printed targets. This was interfaced to a microPET scanner. Recorded spatial transformations were applied in software to list mode events to create a motion-corrected sinogram. Motion correction was evaluated in microPET studies, in which a conscious animal was simulated by a phantom that was moved during data acquisition. The motion-affected scan was severely distorted compared with a reference scan of the stationary phantom. Motion correction yielded a nearly distortion-free reconstruction and a marked reduction in mean squared error. This work is an important step towards motion tracking and motion correction in neurological studies of awake animals in the small animal PET imaging environment.

Comparison of Monte Carlo simulated and measured performance parameters of miniPET scanner

In vivo imaging of small laboratory animals is a valuable tool in the development of new drugs. For this purpose, miniPET, an easy to scale modular small animal PET camera has been developed at our institutes. The system has four modules, which makes it possible to rotate the whole detector system around the axis of the field of view. Data collection and image reconstruction are performed using a data acquisition (DAQ) module with Ethernet communication facility and a computer cluster of commercial PCs. Performance tests were carried out to determine system parameters, such as energy resolution, sensitivity and noise equivalent count rate. A modified GEANT4-based GATE Monte Carlo software package was used to simulate PET data analogous to those of the performance measurements. GATE was run on a Linux cluster of 10 processors (64 bit, Xeon with 3.0 GHz) and controlled by a SUN grid engine. The application of this special computer cluster reduced the time necessary for the simulations by an order of magnitude. The simulated energy spectra, maximum rate of true coincidences and sensitivity of the camera were in good agreement with the measured parameters.

Bioluminescence Imaging

Bioluminescence refers to the process of visible light emission in living organisms. Bioluminescence imaging is a powerful methodology that has been developed over the last decade as a tool for molecular imaging of small laboratory animals, enabling the study of ongoing biological processes in vivo. This form of optical imaging is low cost and noninvasive and facilitates real-time analysis of disease processes at the molecular level in living organisms. In this article, we provide a brief introduction to bioluminescence imaging technology and discuss its applications in mouse models of lung inflammation/injury, bacterial pneumonia, and tumor growth and metastasis.

Development of position sensitive detectors for use in positron emission tomography of small laboratory animals

We report the development of high resolution detector modules for use in PET imaging of small laboratory animals. The detectors are based on thin blocks of NaI coupled to a light guide which serves to share the scintillation light between four photomultiplier tubes. Monte Carlo simulations of photon transport within the scintillator and light guide, together with initial experimental results, suggest that a detector resolution of approximately 2 mm is obtainable using 18F or 11C labeled tracers.

Design and engineering aspects of a high resolution positron tomograph for small animal imaging

Describes the Sherbrooke positron emission tomograph, a very high resolution device dedicated to dynamic imaging of small laboratory animals. Its distinctive features are: small discrete scintillation detectors based on avalanche photodiodes (APD) to achieve uniform, isotropic, very high spatial resolution; parallel processing for low deadtime and high count rate capability; multispectral data acquisition hardware to improve sensitivity and scatter correction; modularity to allow design flexibility and upgradability. The system implements the clam-shell sampling scheme and a rotating rod transmission source. All acquisition parameters can be adjusted under computer control. Temperature stability at the detector site is ensured by the use of thermoelectric modules. The initial system consists of one layer of 256 modules (two rings of detectors) defining 3 image slices in a 118 mm diameter by 10.5 mm thick field. The axial field can be extended to 50 mm using 4 layers of modules (8 rings of detectors). The design constraints and engineering aspects of an APD-based PET scanner are reviewed and preliminary results are reported. >

Analytical study of performance in a 3D PET scanner

The use of arrays of small discrete detectors and thin slices to achieve a high isotropic spatial resolution in positron emission tomography (PET) results in systems with a low ring sensitivity. In a multi-ring system, the overall sensitivity can be considerably improved by removing the interslice collimators to make full use of all cross slice coincidences, but this is achieved at the expense of increased scatter and accidental rates in the image. For imaging of small laboratory animals (diameter of 10 cm or less), the relieved burden of scatters, and to some extent accidentals, suggests that volumetric imaging may be of particular value. In order to evaluate the performance to be expected from a small animal PET scanner (10 cm diameter field) with and without the interplane collimators, the incident event rates for singles (unscattered and single scattered) and true, scatter and accidental coincidences were evaluated analytically. The performance was evaluated for various source sizes and activities. It is concluded that the volumetric configuration is more advantageous than the multislice configuration to image small animals.
