Immunogenetic characteristics of the clinical course of allergic rhinitis in the UZBEK population depending on the polymorphism of the IL-17A RS2275913 gene

Summary Topicality. Over the past decade, according to WHO, there has been an increase in the incidence of allergic diseases. The pathogenesis of allergic diseases is being closely studied, in particular, the role of Th17 cells involved in the pathogenesis of allergic rhinitis and characterized by the predominant synthesis of cytokines IL-17A and IL-17F has been proven. Aim: to study the polymorphism of the IL-17A rs2275913 gene in the Uzbek population and the role in the clinical course, the effectiveness opharmacotherapy of allergic rhinitis with antihistamines. Material and methods. The study included 118 patients with allergic rhinitis. In 112 patients, seasonal allergic rhinitis was diagnosed with an exacerbation in summer (94.9%) and in the spring-summer period of pallination (82.2%). The control group included 200 practically healthy individuals. Genotyping of the IL17A rs2275913 polymorphism was performed using the real-time polymerase chain reaction (Real-Time) "SNP-EXPRESS"-RV method. Results. Patients with allergic rhinitis complained of seasonal and year-round symptoms, which included rhinorrhea, nasal obstruction, sneezing, itching, burning sensation in the nose, sniffling, snoring, apnea, voice change and nasality. Associations of IL-4, IL-4R, IL-17A, and IL-17F Polymorphisms with Colorectal Cancer Risk: A Meta-Analysis, Meta-Regression, and Trial Sequential Analysis.

Both interleukin (IL)-4 and IL-17 polymorphisms may be involved in the pathogenesis and progression of colorectal cancer (CRC). Herein, we designed a meta-analysis to assess the associations between IL-4, IL-4R, IL-17A, and IL-17F polymorphisms and CRC risk. Scopus, Web of Science, Cochrane Library, and PubMed databases were searched to retrieve articles published until October 21, 2021. We used crude odds ratio (OR) and 95% confidence interval assessing the association of the polymorphisms and CRC risk in 5 genetic models. Trial sequential analysis for the primary analyses was used to control random errors. Twenty-three studies (8: IL-4 rs2243250, 4: IL-4R rs1801275, 5: IL-17A rs2275913, and 6: IL-17F rs763780) were involved in the meta-analysis. The pooled OR (P-value) for the association between IL-4 rs2243250 polymorphism and the CRC risk was 1.11 (0.08), 1.27 (0.12), 1.07 (0.37), 1.09 (0.17), and 1.22 (0.12), for IL-4R rs1801275 polymorphism was 0.71 (0.18), 1.05 (0.76), 0.86 (0.37), 0.87 (0.41), and 0.69 (0.39), for IL-17A rs2275913 polymorphism was 1.83 (0.0003), 1.73 (0.06), 1.47 (<0.001), 1.61 (0.001), and 1.42 (0.15), and for IL-17F rs763780 polymorphism was 1.07 (0.48), 5.33 (0.02), 1.08 (0.49), 1.08 (0.47), and 8.42 (0.002) in allelic, homozygous, heterozygous, recessive, and dominant models, respectively. The G allele and GA genotype of IL-17A rs2275913 polymorphism and the CC genotype of IL-17F rs763780 polymorphism had an elevated risk in CRC cases. The ethnicity and genotyping method, sample size, control, and publication year were effective factors on the pooled results.

The associations between interleukin-17 single-nucleotide polymorphism and colorectal cancer susceptibility: a systematic review and meta-analysis

BackgroundNumerous case-control studies have reported associations between interleukin-17 (IL-17) polymorphisms and colorectal cancer; however, the results were inconsistent. The aim of this meta-analysis was to further clarify the effects of IL-17 polymorphisms on colorectal cancer susceptibility.Materials and methodRelevant studies were extracted from the electronic databases PubMed, Embase, Web of Science, China National Knowledge Infrastructure (CNKI), and the Chinese Biomedical Literature Database (CMB) up to April 2021. The odds ratio and 95% confidence interval were used to estimate the strength of the associations.ResultsTen articles including 2599 cases and 2845 controls were enrolled in our research after strict literature screening. Highly significant associations between the IL-17A rs2275913 polymorphism and increased colorectal cancer susceptibility were observed in all five gene models (allelic, dominant, recessive, homozygous, and heterozygous models), and subgroup analysis based on ethnicity revealed that these associations existed not only in the Asian population but also in the Caucasian population. However, the results showed no significantly elevated colorectal cancer risk correlated with the IL-17F rs763780 polymorphism, and a slightly lower colorectal cancer susceptibility for the Caucasian population was discovered in the recessive and homozygous models of this mutation.ConclusionThe IL-17A rs2275913 polymorphism may be an independent risk factor contributing to colorectal cancer susceptibility, while the IL-17F rs763780 polymorphism may decrease susceptibility to colorectal cancer. Future studies with large-scale samples are warranted to identify these associations.

Interleukin 17A rs2275913 polymorphism is associated with susceptibility to systemic lupus erythematosus: A meta and trial sequential analysis.

The role of cytokines in the development of systemic lupus erythematosus (SLE) has received much attention. Interleukin-17A upregulates several inflammation-related genes and is thought to have a crucial role in SLE development. The susceptibility to SLE development has been linked to functional genetic variations of the IL-17A gene; nevertheless, the findings have been conflicting. We conducted a meta-analysis that included previously published reports to establish a definitive conclusion on the role of the IL-17A rs2275913 polymorphism in SLE propensity. The PubMed, Google Scholar, and Scopus databases were used to find eligible published articles. All analyses were conducted using Comprehensive Meta-analysis V3.1. Funnel plots and Egger's regression analysis were used to assess publication bias. Q statistics and I2 test explored the heterogeneity among the included studies. Combined odds ratio, 95% confidence interval were calculated for each comparison model. Based on the inclusion and exclusion criteria, a total of four reports, comprising of 608 SLE patients and 815 healthy controls, were considered for the present meta-analysis. The homozygous comparison (AA vs. GG: combined odds ratio= 2.046, p = 0.005) and recessive genetic model (AA vs. GG+GA: combined odds ratio=1.901, p = 0.010) analysis revealed a significant association of rs2275913 with susceptibility to the development of SLE. However, other genetic comparisons (A vs. G, GA vs. GG, AA+GA vs. GG) failed to demonstrate such association. Furthermore, trial sequential analysis revealed a sufficient number of studies, including enough cases and controls that have already been considered to conclude the role of IL17-A rs2275913 polymorphism in SLE. IL-17A rs2275913 polymorphism is associated with susceptibility to SLE development.

Association between the interleukin (IL)-17A rs2275913 polymorphism and rheumatoid arthritis susceptibility: a meta-analysis and trial sequential analysis.

ObjectiveThis meta-analysis was conducted to investigate the relationship between the interleukin (IL)-17A rs2275913 polymorphism and rheumatoid arthritis (RA) susceptibility.MethodsEligible studies were retrieved from PubMed, Embase, and Web of Science. The fixed- or random-effects model was used to calculate the pooled odds ratios (ORs) and 95% confidence intervals (95%CIs) on the basis of heterogeneity.ResultsOverall, 11 studies containing 4019 RA patients and 4137 controls were included in this meta-analysis. The results suggested a significant association between the IL-17A rs2275913 polymorphism and RA susceptibility in the overall population (allelic model A vs. G: OR = 0.89, 95%CI: 0.83–0.95; heterozygote model GA vs. GG: OR = 0.87, 95%CI: 0.78–0.96; homozygote model AA vs. GG: OR = 0.82, 95%CI: 0.71–0.96; dominant model GA + AA vs. GG: OR = 0.86, 95%CI: 0.78–0.94). In the subgroup analyses, the IL-17A rs2275913 polymorphism was significantly associated with RA risk in Europeans (allelic model A vs. G: OR = 0.87, 95%CI: 0.78–0.97; heterozygote model GA vs. GG: OR = 0.79, 95%CI: 0.68–0.93; dominant model GA + AA vs. GG: OR = 0.79, 95%CI: 0.68–0.92), but not in Africans or Americans.ConclusionThis study suggests that the IL-17A rs2275913 polymorphism is significantly associated with RA susceptibility in Europeans.INPLASY registration number: INPLASY202170056.

Association of IL-17A Polymorphism with Chronic Periodontitis in Type 1 Diabetic Patients.

Statement of the Problem: The association of genetic polymorphisms with periodontitis has been studied extensively. The interleukin -17 (IL-17) is a group of cytokines, which comprises six different molecules (IL-17A, B, C, D, E & F). Among these, IL-17A & F are the most commonly understood cytokine, which plays a critical role in inflammatory diseases and periodontal inflammation.Purpose: To evaluate whether IL-17A gene polymorphism is associated with increased risk of chronic periodontitis in type 1 diabetes patients.Materials and Method: This quantitative case- control study was carried out in 60 subjects in 4 groups. The study groups included group A: 15 type 1 diabetes patients (T1DM) with chronic periodontitis (CP), group B: 15 T1DM patients without CP, group C: 15 Non-diabetic patients with CP, group D: 15 Non-diabetic patients without CP. Blood samples were drawn from the subjects and analyzed for IL-17A polymorphism by using the polymerase chain reaction-restriction fragment length polymorphism method. Results: There was no statistical significant difference seen in the genotype distribution among CP patients with or without T1DM and healthy controls. Odds ratio and p value indicated that increased risks for CP were associated with IL-17A allele (G) in patients with T1DM. This allele was correlated with worse clinical parameters of CP in T1DM patients.Conclusion: The present study revealed that IL-17A (rs2275913) polymorphism was not associated with increased risk for CP in T1DM patients.

IL-23/Th17 pathway and IL-17A gene polymorphism in Egyptian children with immune thrombocytopenic purpura

BackgroundImmune thrombocytopenic purpura (ITP) is an acquired complex autoimmune thrombocytopenia. Uncontrolled cellular immune response is one of the key triggers for the loss of immune tolerance in ITP patients. The purpose of this study was to investigate the association of IL-23/Th17, IL-17A and IL-17A rs2275913 gene polymorphism with ITP in Egyptian children.Methods60 patients with ITP and 50 healthy control children from Minia city- Egypt were involved. Serum levels of IL-23 and IL-17A were determined by enzyme-linked immunosorbent assay. The frequency of Th17 cells was measured using flow cytometer. Genotyping for IL-17A was performed via polymerase chain reaction-restriction fragment length polymorphism.ResultsComparing children with ITP to controls, serum levels of IL-23 and IL-17A as well as Th17 cells percentage were significantly increased (p < 0.001). Also, higher levels of these ILs and Th17 cells percentage were associated with decreased platelet count within ITP patients (p < 0.001). Analysis of genotype frequencies for IL-17A rs2275913 polymorphism and its alleles (A, G) showed no significant difference between cases and controls. Likewise, no significant differences were demonstrated between acute and chronic ITP regarding both IL-17A rs2275913 polymorphism prevalence and levels of IL-23, IL-17A plus Th17 cells percentage. The frequency of A alleles was 85 and 86% within patients and controls, respectively.ConclusionsElevated levels of IL-23, IL-17A and Th17 cells may be involved in ITP pathogenesis while IL-17A polymorphism rs2275913 is not prevalent in Egyptian children with ITP.

Determination of High-Resolution HLA-DQB1 Suballeles and IL-17 Polymorphisms in Turkish Pediatric Patients.

Celiac disease (CD) is an autoimmune enteropathy in the small intestine caused by gluten intolerance of the patients. The most important genetic disease-related factor is human leukocyte antigen (HLA)-DQ polymorphism. Association between interleukin (IL)-17A expression of CD4 + T cells and various autoimmune diseases has been reported. The aim of this study was to investigate the relationship between single nucleotide polymorphism (rs2275913) IL-17A and HLA-DQ polymorphisms in Turkish pediatric celiac patients. Study group included 125 pediatric celiac patients with CD and 100 healthy pediatric controls. Deoxyribonucleic acid was isolated from peripheral blood samples. IL-17A polymorphism (rs2275913) was analyzed by polymerase chain reaction-restriction fragment polymorphism method. IL-17A polymorphism and low-/high-resolution HLA-DQ results of patients were evaluated. GG and GA genotype frequencies of IL-17A (rs2275913) polymorphism were significantly higher ( p < 0.05) in the CD patients than the control group. HLA-DQB1*02 and HLA-DQA1*05 alleles were detected in patients, while HLA-DQB1*03 and HLA-DQA1*01 alleles in the control group. Also, when we compared the patient and control groups in terms of HLA-DQ-DR haplotypes, HLA-DQB1*02-DQA1*05-DRB1*03 was found with the relative risk of 42.5 ( p < 0.05). As a result of high-resolution HLA-DQB1 typing, DQB1*02:01 and DQB1*03:02 were at high frequency ( p < 0.05; in 25 patient group). IL-17A (rs2275913) polymorphism genotype frequency was found to be significant in the patient group compared with the control group. The most common HLA-DQB1 suballele was observed as DQB1*02:01.

The IL-17A rs2275913 polymorphism is associated with colorectal cancer risk.

ObjectiveThe association of the IL-17A rs2275913 polymorphism with the risk of colorectal cancer (CRC) has been previously reported. However, the results are inconsistent. In this study, we comprehensively assessed the effect of the rs2275913 polymorphism on CRC risk.MethodsThe rs2275913 polymorphism of 208 CRC patients and 312 age- and gender-matched healthy controls was genotyped by the polymerase chain reaction-restriction fragment length polymorphism method, and then analyzed by logistic regression. In addition, a pooled analysis based on five single-center studies was performed using Stata 12.0 software.ResultsLogistic regression analysis indicated that the IL-17A rs2275913 polymorphism was associated with CRC risk (GA vs. GG: OR = 1.53, 95% CI = 1.02–2.28; AA vs. GG: OR = 1.89, 95% CI = 1.11–3.20; GA+AA vs. GG: OR = 1.62, 95% CI = 1.11–2.37; A vs. G: OR = 1.38, 95% CI = 1.07–1.77). Further pooled analysis also indicated a statistically significant association between the rs2275913 polymorphism and CRC risk in Asians and Northern Africans.ConclusionThis study suggested that the IL-17A rs2275913 polymorphism may act as a biomarker for predicting CRC risk. However, further functional research should be performed to clarify the role of the rs2275913 polymorphism in the etiology of CRC.

IL17A rs2275913 polymorphism and features of immunological parameters in patients with persistent allergic rhinitis during allergen-specific immunotherapy

Allergic rhinitis (AR) represents a global healthcare challenge. Epidemiology data demonstrate that around 20% of all-age group subjects suffer from allergic rhinitis. Over the last decades, AR incidence and morbidity have been markedly increased due to poorly understood causes. For instance, in the last decade AR prevalence has been elevated by 2-fold in Uzbekistan. However, medical records related to AR prevalence based on patient visit rate infer that it is dozens of times lower than actual data and reflects in no way severity of the problem, but sufficient enough to outline its large-scale spread. Allergen-specific immunotherapy (ASIT) requiring further development and adjustments represents one of the most promising approaches to treat allergic diseases. Some researchers note rise in respiratory tract allergic disease (AD) prevalence including caused by pollen allergens. Therapeutic interventions in this type of pathology emerging due to chronic inflammatory process mainly in airway mucosa are aimed at achieving good control over disease symptoms, lowering risk of subsequent exacerbations and preventing AD aggravation. IL-17 belongs to the Th17 cell-derived cytokines that was described relatively recently. IL17 genes encode six proteins (molecular weight 20-30 kDa), among which IL17A and IL17F display peak sequence homology and were studied in numerous cell types. IL-17 family proteins take part in various reactions of immune response being mainly secreted by Th17 cells. It was shown that immunological mechanisms particularly mediated by cytokines such as IL-17A involved in inflammation, regeneration and fibrogenesis are crucial in progression of diverse infectious diseases.

IL17A rs2275913 gene polymorphism in Uzbek ethnic group and its linkage with development of allergic rhinitis

Currently, new insights into the general mechanisms of developing allergic diseases suggest that Th17 cells are involved in the pathogenesis of allergic rhinitis characterized by dominant production of cytokines IL-17A and IL-17F. Objective: to assess IL17A rs2275913 gene polymorphism in the Uzbek ethnic group and its linkage with development of allergic rhinitis. There were examined 83 patients with allergic rhinitis, among which 38 (46%) had intermittent form and 45 (54%) – with persistent disease form. In control group there were included 123 apparently healthy subjects. Genotyping of IL17A rs2275913 polymorphism was carried out by using real-time polymerase chain reaction (Real-Time) “SNP-EXPRESS”-RV. While analyzing frequency distribution of the G and A alleles within the IL17A gene, it was found that the A allele dominated (38.7% vs 19.5%, respectively, χ2 = 15.9; p 0.05). The G/G genotype in the A/G IL17A gene polymorphism was much less abundant in AR patients compared to apparently healthy patients in control group. It was detected increased frequency of heterozygous G/A allele within the IL17A gene in AR patients vs. control group (56.9% vs 30.2%, respectively, χ2 = 11.9; p 0.05; OR = 3.1). While comparing the A/A genotype of the IL17A gene it was shown that its prevalence was significantly higher in AR patients compared to healthy subjects (14.1% vs 5.9%, respectively, χ2 = 4.6; p 0.05). Our findings demonstrated that the AA genotype in the IL17A rs2275913 gene is associated with developing AR in the Uzbek ethnic group. Such trait may be used as AR predictor, provide valuable information for design and implementation of pathogenetically justified methodological approaches to therapy and prevention of allergic rhinitis.

Meta-Analysis of Risk Association between Interleukin-17A Gene Polymorphism and Chronic Periodontitis.

Among this, IL-17A is the most commonly understood cytokine, and its polymorphism plays a critical role in inflammatory diseases and periodontal inflammation. The present study was aimed at pooling the data available for meta-analysis and to evaluate whether IL-17A (rs2275913) polymorphism is associated with the susceptibility of chronic periodontitis.

The association between IL-17 gene variants and risk of colorectal cancer in a Chinese population: A case-control study.

Interleukin (IL)-17 have been reported to be associated with the pathogenesis of colorectal cancer (CRC). Few studies investigated the association between IL-17 gene polymorphisms and risk of CRC with inconsistent findings. Thus, we recruited 352 CRC cases and 433 controls in a Chinese population and their genotyping was done using polymerase chain reaction-restriction fragment length polymorphism method. Our data showed that IL-17A rs2275913 polymorphism was associated with the increased risk of CRC, while no association was observed for IL-17F rs763780 polymorphism. Stratified analyses revealed that the significant association was also obtained in the females, smokers, drinkers and age ≥ 60 years groups for rs2275913 polymorphism. Moreover, the CC and/or GC genotype of rs2275913 polymorphism were correlated with TNM stage and lymph node metastasis. No association was shown between IL-17F rs763780 polymorphism and clinical characteristics of CRC. In conclusion, our data indicate that IL-17A rs2275913 polymorphism but not IL-17F rs763780 polymorphism contributes to increased risk for CRC patients in this Chinese population.

Association between IL-17 gene polymorphisms and circulating IL-17 levels in osteoarthritis: ameta-analysis.

This study systemically reviewed the evidence regarding associations between polymorphisms in interleukin-17 (IL-17) genes and osteoarthritis (OA) susceptibility, and the relationship between circulating IL-17 levels and OA. We performed ameta-analysis of the associations between the IL-17A rs2275913 and IL-17F rs763780 polymorphisms and risk for OA and serum/plasma IL-17 levels in OA patients and controls. Eight studies including 2214 OA patients and 2474 controls were included. Our meta-analysis identified asignificant association between OA and the AA genotype of the IL-17A rs2275913 polymorphism in apooled cohort of affected individuals, compared to the case in apooled cohort of control participants (OR = 1.516, 95% CI = 1.260-1.825, P < 0.001), and asignificant association between OA and the CC genotype of the IL-17F rs763780 polymorphism (OR = 2.257, 95% CI = 1.376-3.704, p = 0.001). OA site-based stratification identified an association between the AA genotype of the IL-17A rs2275913 polymorphism and the CC genotype of the IL-17F rs763780 polymorphism and knee OA, but not hip OA. Furthermore, the same patterns of significant associations between OA and the IL-17A rs2275913 and IL-17F rs763780 polymorphisms were identified based on homozygote contrasts. The OA patients showed significantly higher IL-17 levels than the control subjects (SMD = 1.830, 95% CI = 1.184-2.477, P < 0.001). Our meta-analysis revealed associations between the IL-17A rs2275913 and IL-17F rs763780 polymorphisms and OA susceptibility, and the presence of significantly higher circulating IL-17 levels in OA patients.

Association of the IL-17A rs2275913 and MIF rs755622 polymorphisms with the risk of gastric and colorectal cancer

BackgroundInterleukin 17 (IL-17) and Macrophage migration inhibitory factor (MIF) are pro-inflammatory cytokines that play an important role in gastric and colorectal cancer pathogenesis. AimThe main objective of our study is to investigate the potential association of IL-17A (−197G > A) rs2275913 and MIF (−173 G > C) rs755622 polymorphisms with susceptibility to gastric and colorectal cancers in Moroccan population. MethodsWe conducted a case-control study that included 70 gastric cancer patients; 70 colorectal cancer patients; and 70 healthy controls. Genotyping was performed with real-time polymerase chain reaction (PCR), using a TaqMan single-nucleotide polymorphism (SNP) genotyping assay. ResultsThe mutant genotype AA of the IL-17 rs2275913 polymorphism was correlated with a higher risk of developing colorectal cancer than the GG and AG genotypes combined [odds ratio (OR) = 2.7284; 95% confidence interval (CI) = 1.6491–4.5141, p = .0001]. The wild type genotype GG of the MIF rs755622 polymorphism has a slight protective effect against colorectal [odds ratio (OR) = 0.1262; 95% confidence interval (CI) = 0.0148–1.0733, p = .0581] and gastric cancer [odds ratio (OR) = 0.0749; 95% confidence interval (CI) = 0.0090–0.6237, p = .0165]. ConclusionThe IL-17A rs2275913 (−197G > A) polymorphism could contribute to the risk of colorectal cancer and may be utilized as a genetic screening marker in assessing colorectal cancer risk in the Moroccan population.

Association between interleukin-17A/F single nucleotide polymorphisms and susceptibility to osteoarthritis in a Chinese population.

Interleukin-17A/F (IL-17A/F) might play a role in the pathophysiology of osteoarthritis (OA), but several studies exploring the association between IL-17A/F single nucleotide polymorphisms and OA in different populations present inconsistent results. Thus, this case–control study, involving 410 OA cases and 507 controls, was aimed to investigate such association in a Chinese population. Genotyping was performed using standard polymerase chain reaction and restriction fragment length polymorphism. It was found that AA genotype or A allele carriers of IL-17A gene rs2275913 polymorphism were associated with OA susceptibility. Stratified analyses showed IL-17A rs2275913 polymorphism was evidently associated with a significantly increased risk for OA among males, <60 years old patients, smokers, and drinkers. No significant association was observed between IL-17F gene rs763780 polymorphism and OA risk. In conclusion, IL-17A rs2275913 polymorphism is involved in the development of OA in Chinese Han population. This finding should be confirmed by a larger study from diverse ethnic populations.

GENETIC POLYMORPHISMS OF IL-17A IN ASSOCIATION WITH THE RISK OF PULMONARY TUBERCULOSIS IN IRAQI PATIENTS

Objectives: Tuberculosis (TB) is an infectious disease that usually affects the lungs caused by Mycobacterium tuberculosis, TB is the second biggest killer, globally. The aim of this study was to examine the association between IL-17A rs2275913 SNP and pulmonary TB susceptibility in Iraqi population.Methods: From January 2017 to April 2017, 80 pulmonary TB patients were selected as the case group, another 40 healthy control were enrolled as the control group. The genotype frequencies of IL-17A rs2275913 was detected using amplification refractory mutation system.Results: The results of IL-17A serum level demonstrated that there were significant differences (p&lt;0.05) between patients groups. The result showed that A allele have a higher frequency (55% vs. 50%) in TB patients than the control sample with OR value of 1.22 and EF value 0.1 and G have lower frequency (45% vs. 50%) in TB patients than control sample with OR value of 0.82 with PF value of 0.09, but this difference was not statistically significant (p&gt;0.05).Conclusion: There were no significant associations between IL-17A rs2275913 polymorphism and risk of TB in Iraqi population.

Association of IL17 and IL23R gene polymorphisms with rheumatic heart disease in South Indian population

ABSTRACTBackground: IL-23/Th17 signaling pathway plays a crucial role in the cell-mediated immune response against bacterial infections and also in the pathogenesis of inflammatory and autoimmune diseases. Recent studies indicate that Th17 cell-associated cytokines are involved in the progression and maintenance of valvular lesions in rheumatic heart disease (RHD). Variants in the genes of cytokines that are potentially involved in Th17 response may influence interindividual differences in their expression levels, thereby contributing to the pathogenesis of immune-mediated diseases such as RHD.Objective: The aim of the study is to investigate the association of IL17A, IL17F, and IL23R gene variants with the risk perception of RHD.Methods: A total of 225 individuals (99 RHD patients and 126 healthy siblings) were recruited for the study. The IL17A (rs2275913), IL17F (rs763780), and IL23R (rs10889677) polymorphisms were determined by polymerase chain reaction restriction fragment length polymorphisms and amplification-refractory mutation system-polymerase chain reaction methods, respectively.Results: The frequency of IL17A (rs2275913) A/A genotype was significantly high in pooled RHD patients (odds ratio [OR] = 2.76; pc = 0.021), rheumatic fever (RF) patients (OR = 14.5; pc = 0.0001), and mitral valvular lesions patients (OR = 2.74; pc = 0.039) when compared to healthy siblings. However, the IL17F (rs763780) and IL23R (rs10889677) polymorphisms did not show any association with RHD.Conclusions: The results suggest that IL17A (rs2275913) polymorphism is associated with the development of RF/RHD in South Indian population. Further studies are required to substantiate the association of these genes with the disease risk.

Genetic polymorphisms of IL-17A rs2275913, rs3748067 and IL-17F rs763780 in gastric cancer risk: evidence from 8124 cases and 9873 controls.

Interleukin-17 (IL-17) is a critical cytokine involved in inflammation-associated cancers. Single nucleotide polymorphisms (SNPs) might promote carcinogenesis. In this current meta-analysis, we investigated the association of IL-17A and IL-17F gene polymorphisms with gastric cancer (GC) risk. Eligible genetic association studies were retrieved from PubMed, Web of Science and Scopus database sources. Two reviewers independently assessed methodological quality and extracted data from eligible articles. Pooled odds ratios (ORs) and 95% confidence intervals (CIs) were calculated. Quantitative data synthesis was conducted using comprehensive meta-analysis v2. Subgroup analysis and heterogeneity analysis were performed. Begg's funnel plot and Egger's regression tests were used to judge publication bias. In silico data analysis was executed to analyze the functional and structural impact of the SNPs. A total of 21 case-control studies for rs2275913 c.-197G > A (7660 patients and 9409 controls), 9 studies for rs3748067 c.*1249C > T (3378 patients and 4120 controls), and 14 studies for rs763780 c.482A > G (4481 patients and 5354 controls) were included. The pooled estimate revealed an association between IL-17A rs2275913 polymorphism and the risk of GC under all genetic models (A vs. G, OR 1.187, 95% CI 1.086-1.297, P < 0.001; GA vs. GG, OR 1.108, 95% CI 1.008-1.218, P = 0.033; AA vs. GG, OR 1.484, 95% CI 1.236-1.781, P < 0.001), while no evidence of association was found with IL-17A rs3748067 or IL-17F rs763780 polymorphisms. Our results showed that IL-17A promoter rs2275913 variant might represent a potential risk factor for gastric cancer susceptibility.

IL-17A, MCP-1, CCR-2, and ABCA1 polymorphisms in children with non-alcoholic fatty liver disease

ObjectiveThe prevalence of non-alcoholic fatty liver disease in children has risen significantly, owing to the worldwide childhood obesity epidemic in the last two decades. Non-alcoholic fatty liver disease is closely linked to sedentary lifestyle, increased body mass index, and visceral adiposity. In addition, individual genetic variations also have a role in the development and progression of non-alcoholic fatty liver disease. The aim of this study was to investigate the gene polymorphisms of MCP-1 (-2518 A/G) (rs1024611), CCR-2 (190 G/A) (rs1799864), ABCA1 (883 G/A) (rs4149313), and IL-17A (-197 G/A) (rs2275913) in obese Turkish children with non-alcoholic fatty liver disease. MethodsThe study recruited 186 obese children aged 10–17 years, including 101 children with non-alcoholic fatty liver disease and 85 children without non-alcoholic fatty liver disease. Anthropometric measurements, insulin resistance, a liver panel, a lipid profile, liver ultrasound examination, and genotyping of the four variants were performed. ResultsNo difference was found between the groups in respect to age and gender, body mass index, waist/hip ratio, or body fat ratio. In addition to the elevated ALT levels, AST and GGT levels were found significantly higher in the non-alcoholic fatty liver disease group compared to the non non-alcoholic fatty liver disease group (p<0.05). The A-allele of IL-17A (-197 G/A) (rs2275913) was associated with non-alcoholic fatty liver disease (odds ratio [OR] 2.05, 95% confidence interval: 1.12–3.77, p=0.02). ConclusionsThe findings of this study suggest that there may be an association between IL-17A (-197 G/A) (rs2275913) polymorphism and non-alcoholic fatty liver disease development in obese Turkish children.
