First records and extension of the geographic distribution of Cynomops kuizha (Chiroptera: Molossidae) in the Colombian Caribbean

Abstract The insectivorous bat genus Cynomops belongs to the Neotropical fraction of the family Molossidae, which includes Free-tailed Bats or Mastiff Bats. Currently, nine species are recognized within Cynomops. In this study, we present the first records of Cynomops kuizha for the Colombian Caribbean Region. The two female specimens were recovered in the campus of the Puerta Roja of the University of Sucre, Sucre and represent a significant extension in the geographical and ecological distribution of the species. With these new records, the known distribution of this species is extended approximately 520 km northeast of the nearest previous record in the department of Valle del Cauca, Colombia. Identification of both specimens was carried out through morphological comparisons and analysis of the mitochondrial gene cytochrome-b (Cyt-b). Molecular analyses revealed that the Cyt-b sequences from the Sucre specimens are >99 % similar to those documented for C. kuizha in southwestern Colombia. With these new records, the known distribution of this species is extended approximately 520 km northeast of the nearest previous record in the department of Valle del Cauca, Colombia. Additionally, we discuss some morphological peculiarities observed in our Sucre specimens and their relationship with closely related species present in the area: Cynomops abrasus, Cynomops greenhalli, and Cynomops mastivus.

Identifikasi Jenis Anggang-anggang (Gerris marginatus) di Aliran Sungai Taman Wisata Alam Kerandangan Kabupaten Lombok Barat

Flywheels are a type of insect belonging to the Family Gerridae. Flywheels are insects that are usually found in still waters, such as ponds, rivers and lakes. The aim of this research is to identify the types of flywheels that exist in the river flow of Kerandangan Nature Tourism Park, West Lombok Regency. The type of research used is exploratory descriptive research. Identification of flywheels specimens is based on the identification book from Pennak (1978). The results of the research showed that one type of flywheels was found, namely Gerris sp. in the river flow of the Kerandangan Nature Tourism Park with a total of 529 individuals.

On the polysemic nature of traces and co-occurring patterns in anthropized material — contribution of a “Retoucher” with a flaked bone aspect from Roc-en-Pail (France)

This article presents a new archaeological material to discuss methodological issues encountered by scientists working both on minimally-modified bones from Mousterian assemblages, to those dealing with the identification of “retouchers” having a flaked bone aspect. The technological approach integrates complementary analytical study-frames in order to assess archaeological information. On the one hand, analogies with similar experimentally produced patterns reduce the functional identification of the archaeological specimen. They do not deal with a single artefact-type in the category of “retouchers”, which thus remains generic. On the other hand, the recording of the patterns in their chronological order, combined together with that of modifications relative to the diagenesis of the bone as an artefact, suggests the “retoucher” was reduced in a relatively fresh state by a carnivorous predator also. The evidence of this co-occurrence, if characterizing the successive anthropic-originated bone beds ultimately degraded by predators in situ, would suggest a relatively short period of human occupation generated by the use of the site in a singular cyclical conception “prey-hunter-predator” at regional scale.

Assessment of nine markers for phylogeny, species and haplotype identification of Kappaphycus species and Eucheuma denticulatum (Solieriaceae, Rhodophyta)

ABSTRACT Molecular studies have contributed to the taxonomy of carrageenan-producing Kappaphycus spp. and Eucheuma denticulatum. However, unresolved species complexes and the lack of standardization in the use of genetic markers impede the identification of specimens and the delineation of a robust taxonomic framework. Here, nine molecular markers (cox1, cox2–3 spacer, cox2, cox3, COB, ITS, psbA, UPA and rbcL) were used to generate a multilocus phylogeny for 113 fresh eucheumatoid samples and four herbarium specimens. Analyses of species delineation and genetic distances confirmed the monophyly of currently accepted taxa. These analyses suggest that clades previously reported as K. striatus KS1 and KS2 are conspecific, and that E. denticulatum EDA ‘spinosum’ and EDB ‘endong/cacing’ are also conspecific. The results also unveiled possible new taxa from Hawaii and Indonesia. Each molecular marker and combinations thereof were assessed with regard to species identification, ease of amplification and sequencing, and haplotype characterization. All genetic markers recorded at least 94% success in the amplification and sequencing of fresh specimens, with cox1 being the most phylogenetically informative. Automatic partitioning, phylogenetic and tree-based assessments showed cox1, cox2–3 spacer, cox2 and rbcL were able to correctly identify species while cox1+ rbcL, COB+rbcL, cox2+ rbcL or cox1+ COB+rbcL trees best represented the phylogeny with consistently high nodal support. Among individual markers, cox1 identified the greatest number of haplotypes, while UPA, partial rbcL (750 bp), ITS, cox3 and cox2–3 spacer were able to retrieve information from herbarium specimens of 12–16 years of age. These molecular results provide a basis for a database essential for the taxonomic framework, cultivar development and germplasm conservation of eucheumatoids.

Efficient identification and distinction of musgravite and taaffeite with the utilization of ATR-FTIR spectroscopy and Raman spectroscopy

Taaffeite (BeMg3Al8O16) and musgravite (Be(Mg,Fe,Zn)2Al6O12) are two of the rarest gem kinds worldwide, and their scarcity greatly enhances their extraordinary worth. Due to their nearly matched physical properties, discriminating between the two gems using basic gemological equipment will be exceedingly difficult, considering that they both belong to the same mineral family. Distinguishing between these two categories is crucial due to the substantial variation in their rarity levels, which greatly impacts on their market pricing. Nevertheless, there is a lack of published data in the scientific literature about the spectroscopic characterization of musgravite and taaffeite.In this article, ATR-FTIR spectroscopy successfully distinguished Tanzanian musgravite from taaffeite for the first time. In addition, Raman spectroscopy and EPMA are employed for the identification of musgravite and taaffeite specimens. The EPMA results confirm that the Tanzanian gems under investigation have similar elemental compositions to those of the same kinds of stones discovered from other sources. The peaks observed in the ATR and Raman spectra serve as indicators for distinguishing between musgravite and taaffeite gemstones, with the goal of simplifying the identification process. The ATR and Raman spectra of musgravite and taaffeite are comprehensively analyzed and found to be achievable. The main Raman bands used to identify Tanzanian musgravite are situated at 412 and 713 cm−1, whereas for taaffeite, the significant bands were detected at 416 and 761 cm−1. The distinct ATR bands observed at 773 cm−1, corresponding to the vibration of Al–O, can be efficiently utilized as indications to differentiate Tanzanian musgravite from taaffeite. The results prove that ATR-FTIR spectroscopy, like Raman spectroscopy, is a very effective non-invasive method for rapidly distinguishing these precious gemstones.

A Handheld Tool for the Rapid Morphological Identification of Mosquito Species (VectorCam) for Community-Based Malaria Vector Surveillance: Summative Usability Study.

Malaria impacts nearly 250 million individuals annually. Specifically, Uganda has one of the highest burdens, with 13 million cases and nearly 20,000 deaths. Controlling the spread of malaria relies on vector surveillance, a system where collected mosquitos are analyzed for vector species' density in rural areas to plan interventions accordingly. However, this relies on trained entomologists known as vector control officers (VCOs) who identify species via microscopy. The global shortage of entomologists and this time-intensive process cause significant reporting delays. VectorCam is a low-cost artificial intelligence-based tool that identifies a mosquito's species, sex, and abdomen status with a picture and sends these results electronically from surveillance sites to decision makers, thereby deskilling the process to village health teams (VHTs). This study evaluates the usability of the VectorCam system among VHTs by assessing its efficiency, effectiveness, and satisfaction. The VectorCam system has imaging hardware and a phone app designed to identify mosquito species. Two users are needed: (1) an imager to capture images of mosquitos using the app and (2) a loader to load and unload mosquitos from the hardware. Critical success tasks for both roles were identified, which VCOs used to train and certify VHTs. In the first testing phase (phase 1), a VCO and a VHT were paired to assume the role of an imager or a loader. Afterward, they swapped. In phase 2, two VHTs were paired, mimicking real use. The time taken to image each mosquito, critical errors, and System Usability Scale (SUS) scores were recorded for each participant. Overall, 14 male and 6 female VHT members aged 20 to 70 years were recruited, of which 12 (60%) participants had smartphone use experience. The average throughput values for phases 1 and 2 for the imager were 70 (SD 30.3) seconds and 56.1 (SD 22.9) seconds per mosquito, respectively, indicating a decrease in the length of time for imaging a tray of mosquitos. The loader's average throughput values for phases 1 and 2 were 50.0 and 55.7 seconds per mosquito, respectively, indicating a slight increase in time. In terms of effectiveness, the imager had 8% (6/80) critical errors and the loader had 13% (10/80) critical errors in phase 1. In phase 2, the imager (for VHT pairs) had 14% (11/80) critical errors and the loader (for VHT pairs) had 12% (19/160) critical errors. The average SUS score of the system was 70.25, indicating positive usability. A Kruskal-Wallis analysis demonstrated no significant difference in SUS (H value) scores between genders or users with and without smartphone use experience. VectorCam is a usable system for deskilling the in-field identification of mosquito specimens in rural Uganda. Upcoming design updates will address the concerns of users and observers.

A multi-center evaluation of a novel IVF cryostorage device in an active clinical setting

The objective of this study was to evaluate the function, and usability of a novel automated software-guided cryostorage system in an active IVF laboratory setting. The investigational device (ID) was installed at 3 IVF laboratories (sites: α, β, and γ). A total of 15 embryologists were trained to use the ID. Mock patient specimens containing mirrored live patient data were handled using the ID. Temperature readings were recorded every minute. Successful identification, storage, and retrieval of mock patient specimens by the ID were evaluated. To assess an LN2 pressure builder, the frequency of use and events of workflow interruption were logged. Student’s t-test was used to determine statistical significance. The ID was in active use for 164 days total. During this time, 329 mock patient egg and embryo cohorts were handled by the ID. The mean ± SD temperatures during active use were: α, − 176.57 ± 1.83 °C; β, − 178.21 ± 2.75 °C; γ, − 178.98 ± 1.74 and did not differ significantly. The highest recorded temperatures were: α, − 165.14 °C; β, − 157.41 °C; γ, − 164.45 °C. A total of 1064 automation transactions on 409 specimen vessels were performed. Data was managed on 1501 eggs and embryos. The ID did not lose or misplace any specimen data or vessels, and no mock specimen was exposed to a detrimental (> − 150 °C) temperature excursion. Over the 25 LN2 pressure builder usages during 99 total days, there was 1 occurrence where usage interrupted workflow due to a lack of LN2 pressure. The ID has advantages over the current manual-based cryostorage systems, including radio frequency identification (RFID) tracking, automation of manual tasks, and software guidance to ensure accurate specimen storage and retrieval. The results of this study indicate that the ID can be integrated into active IVF laboratories.

Pseudopus pannonicus (Squamata), the largest known anguid lizard-Redescription of the type material and new specimens from the Neogene and Quaternary of Hungary and Poland.

Herein, we revise the material of the extinct taxon Pseudopus pannonicus from Central Europe, the largest known anguid lizard and iconic member of herpetofaunas from the Upper Cenozoic of Europe. The geographical position of Polgárdi 2, the type locality of P. pannonicus, as well as several other closely located important localities make Central Europe a valuable area of high interest for studies regarding European Cenozoic palaeoherpetological assemblages. We clarified the nature of the type material of Pseudopus pannonicus, showing that it consisted not only of the five specimens originally figured. Instead, the syntype series also includes a considerable number of specimens from Polgárdi 2 that were only described or figured 12 years after the original description of P. pannonicus. Detailed osteological descriptions are provided for specimens from the type series and Polish specimens, with the aid of high-resolution imaging (SEM and μCT scanning), and intraspecific variability is discussed. The articular surface with the lappet of the parietal overlapping the frontal is discussed as a character potentially relevant for the diagnosis of P. pannonicus. We updated the identification of several specimens of P. pannonicus and discussed the biogeographic implications of such revisions. In Poland, P. pannonicus is an abundant component of Neogene and early Quaternary herpetofaunas, known with certainty from the Middle Miocene of Przeworno (the oldest known occurrence of the species globally), the Early-Late Pliocene of Węże I, the Late Pliocene of Rębielice Królewskie I, the Late Pliocene of Węże II, and the Early Pleistocene of Kadzielnia (one of the youngest occurrences documented globally). An indeterminate anguine with Anguis affinities is newly reported from Rębielice Królewskie II. The taxonomic status of other large anguids from the Neogene of Europe is discussed and we conclude that most are junior synonyms of P. pannonicus. We also show that another purported synonym, that is, Ophisaurus intermedius from the Early Pleistocene of Romania, is instead a nomen nudum. We conducted phylogenetic analyses (18 taxa, 65 characters) to understand the relationship of P. pannonicus relative to other anguid representatives and anguid-related group (i.e., glyptosaurids). A single most parsimonious tree (length: 134 steps) was recovered. The clade Pseudopus is stable, comprising the two distinct sister subclades [Pseudopus laurillardi + Pseudopus ahnikoviensis] and [Pseudopus pannonicus + Pseudopus apodus]. These phylogenetic results are in accordance with previously published works.

Challenges of molecular barcode-based identification of earthworm specimens for biodiversity assessment

Specimen identification at the species level is a critical challenge for understanding community structure and conserving biodiversity. The use of mitochondrial DNA barcodes, in addition to morphology, has proven to be a useful tool for earthworm identification, but it has also raised difficulties. Thus, approaches to delineating molecular operational taxonomic units (MOTUs) from cytochrome c oxidase subunit 1 (COI) or 16S rDNA sequence data often reveal more mitochondrial lineages than morphospecies, raising the question of whether these MOTUs should be used as taxonomic entities in community structure studies. Here, we used a newly acquired dataset of 576 COI barcodes of earthworms from Metropolitan France that were clustered in 36 MOTUS, corresponding to 21 morphospecies. We also incorporated data gathered from the literature to investigate this question. In order to match our MOTUs with already-described mitochondrial lineages, we downloaded reference sequences from the GenBank and BOLD platforms. In light of the difficulties encountered in recovering these sequences, we recommend that any new mitochondrial lineage described in a study be named consistently with previous works. Next, we analyzed the biological, ecological, and molecular data available in the literature on the different mitochondrial lineages that matched our MOTUs in order to determine if there was a consensus for species delimitation. Although the study specimens mainly belong to the Lumbricidae, which is one of the most studied families of earthworms, the data are often missing to determine if the MOTUs correspond to different species. Pending revision of the taxonomy, MOTUs for which mitochondrial divergence has been confirmed by morphological differences should be considered distinct taxonomic entities in community structure studies. In the absence of morphological differences and pending more data, we propose to distinguish for these analyses, within sexually reproducing morphospecies, the MOTUs for which mitochondrial divergence has been confirmed by multilocus nuclear data, while in the case of reproduction by parthenogenesis, ecological differences between MOTUs are necessary to consider them as different taxonomic entities.

The specimens of green anaconda, Boa murina Linnaeus, 1758 (Squamata, Serpentes, Boidae, Eunectes), from the collection of Albertus Seba, and recommendations for tracing historical specimens

In a recent publication, Dubois et al. (2024) reviewed in detail the type material of Boa murina Linnaeus, 1758, now known as Eunectes murinus, and designated a specimen figured in Seba (1735), that is considered lost, as its lectotype. Here we explore the known details of the relevant dispersal routes for Seba’s Eunectes specimens and discuss the probabilities of their existence in several European museums. Based on this example, we provide recommendations for the identification of specimens used by historical authors like Seba as the basis for illustrations in their works.

Comparative analysis of the mitochondrial genomes of Smoothhound sharks provide insight into the phylogenetic relationships within the family Triakidae

The genus Mustelus comprises demersal smoothhound sharks, including M. canis and M. norrisi. Despite their economic value and known conservation concerns, only a few genetic and no genomic resources exist for these two species. In this study, we assembled and described the complete mitochondrial genomes of Mustelus canis and M. norrisi. The mitochondrial genomes of M. canis and M. norrisi are 16,758 bp and 16,769 bp in length, respectively. Both mitogenomes are A + T rich and contain 13 protein-coding genes (PCGs), 2 ribosomal RNA genes, 22 transfer RNA genes, and a control region (CR) 1122 bp and 1121 bp in length, respectively. Mitochondrial synteny observed in M. canis and M. norrisi is identical to that previously reported for congeneric species. In the studied mitochondrial genomes, all PCGs experienced purifying selection. In both species, 21 of the 22 tRNA genes exhibited a typical ‘cloverleaf’ secondary structure, except trnS1 that lacked a complete D-arm. The CR of the studied species lacked tandem repeats, but abundant A + T rich dinucleotide microsatellites were detected. Maximum likelihood and Bayesian inference phylogenetic analyses based on translated PCGs supported the monophyly of the family Triakidae. The same analyses indicated that the genus Mustelus was not monophyletic considering that all representatives of the genus Mustelus clustered together with Triakis megalopterus in a fully supported clade. The assembled mitochondrial genomes will aid with the accurate identification of specimens belonging to the genus Mustelus and support biomonitoring programs based on environmental DNA (eDNA).

Taxonomic reassessment of Scabies (Bivalvia: Unionidae) species in China based on multilocus and mitogenomic phylogenetic analyses

Effective species conservation necessitates the ability to accurately differentiate among species, a challenge compounded by taxonomic uncertainties in freshwater mussels due to substantial intraspecific variation and pronounced phenotypic plasticity in shell morphology. The taxonomic status and species validity of Scabies longata and S. chinensis, two species endemic in China, have been under continuous debate since establishment. The lack of essential molecular data required for a comprehensive systematic study has resulted in the unresolved taxonomic status of these two species. This study presents molecular data, including COI barcoding, COI + 28S rRNA, and mitogenomic data combined with morphological characteristics to assess the validity of S. longata and S. chinensis. Both morphological and COI barcoding data support the conclusion that S. longata and S. chinensis are junior synonyms of Nodularia douglasiae and N. nuxpersicae respectively. Our findings suggest the absence of Scabies species in China. Mitochondrial phylogenetic analyses were used to further elucidate intrageneric relationships within the genus Nodularia, revealing the following relationships: (N. breviconcha (Nodularia sp. 1 (N. douglasiae (N. nuxpersicae, N. nipponensis)))). We underscore the significance of employing an integrated taxonomic approach for species identification, especially given the considerable morphological disparities between larvae and adult freshwater mussels. Proper morphological identification of adult specimens is essential for extracting meaningful taxonomic characters. Furthermore, our findings suggest a notable resemblance between the freshwater bivalve fauna in southern China and those east of the Mekong River. Occurrence of Four Freshwater Stingrays (Chondrichthyes: Potamotrygoninae) in the Uatumã River Basin, Amazon Region: A Field Study

The Amazon region has the largest diversity of freshwater stingrays; however, there are still places where information about this group is scarce. The present work aims to record the occurrence of freshwater stingrays in the Uatumã river basin, Amazonas, Brazil. For this, collections were carried out in three municipalities bathed by the Uatumã River, Presidente Figueiredo, Itapiranga, and São Sebastião do Uatumã. The rays were collected through fishing using a hand net and, after capture, were identified through the pattern of the dorsal and ventral color of the disc, the absence or presence of the labial groove, and the organization of the row of spines on the tail, and were also measured, weighed, and sexed. In total, 69 specimens of Potamotrygonines were captured, 59 of which were from the genus Potamotrygon and ten individuals from the genus Paratrygon. The present study provides preliminary information on the characteristics of freshwater stingray species that may occur in the Uatumã River. Furthermore, new research is necessary to improve the identification of specimens to determine the diversity and elucidate aspects related to the biology of elasmobranchs in the Uatumã River Basin, Brazil.

Molecular taxonomy of fish larvae in the Northwestern Arabian gulf: A baseline study from Kuwait's first marine protected area

Sulaibikhat Bay, a shallow embayment dominated by tides and extensive mud flats along the northern coast of Kuwait Bay, serves as a crucial nursery ground for various commercially important species of finfish and shellfish in the Northwestern Arabian Gulf (NWAG). As the location of Kuwait's inaugural marine protected area (MPA), Sulaibikhat Bay plays a vital role in fostering numerous fish and marine invertebrate species of commercial importance in the Arabian/Persian Gulf. Accurate identification of larval specimens from this habitat is essential for understanding their spawning dynamics and recruitment process. However, limitations in taxonomic identification hinder scientific understanding of fish larval recruitment. To address this, our baseline study employed an integrated taxonomic approach, combining conventional morphology with molecular techniques. Fish larvae collected from the MPA and surrounding waters in Spring (2022) were identified up to the genus and species levels using partial gene sequencing of 16 S rRNA and mitochondrial cytochrome oxidase (COI) genes. Morphological analysis indicated the prevalence of Sparidae, Sillaginidae, and Dorosomatidae families. Molecular analysis confirmed the presence of these families and identified two species, Crenidens crenidens (Sparidae) and Nematalosa nasus (Dorosomatidae), along with two genera, Diplodus sp. (Sparidae) and Sillago sp. (Sillaginidae), which exhibited maximum similarity to available COI sequences of Diplodus sargus, and Sillago maculata, respectively. This represents the first baseline molecular study on fish larvae from NWAG, laying the groundwork for future in-depth analyses with more larval specimens and spatiotemporal samplings from the entire exclusive economic zone (EEZ) of Kuwait in the Arabian/Persian Gulf.

High biodiversity in the tropics is good for ecosystem services; however, challenges in taxonomy and identification usually come from such high biodiversity. Spiders are no exception to the challenges. Identifying spiders in tropical places like Thailand is difficult and time consuming. To reduce the difficulty of identifying Thai spiders, a data retrieval system for geographical occurrence and photographic identification was conducted to deploy on an online platform, Spiders in Thailand (SIT) via the website "spiderthailand.info". This allows professional arachnologists and amateur spider lovers to visit and check the geographical distribution of Thai spiders and to quickly access pictures for comparative photographic identification. To facilitate Thai spider identification, there were two parts, the database and the website, which are connected to each other. Data of Thai spiders were extracted from the World Spider Catalog to build a database comprising geographical occurrence and pictures of spider species in Thailand. The database was then linked with the website to display data. The dataset of pictures and illustrations extracted from taxonomic literature of the World Spider Catalog were included in the database for connecting with the online platform, Spiders in Thailand (SIT) via the website "spiderthailand.info" which facilitated access to pictures and illustrations, expediting the identification of Thai spider specimens. Geographical occurrences of Thai spiders consisted of 1419 records belonging to 670 species of 228 genera and 50 families. Amongst those, 461 species from 133 genera of 41 families were distributed only in Thailand. Around Thailand, 756 geographical localities were reported for spider occurrences. From 76 provinces and one additional special administrative area (Bangkok), 58 provinces showed occurrence records of spiders and 18 provinces showed non-occurrence records. Those provinces of non-occurrence records of spiders were Amnat Charoen, Ang Thong, Bueng Kan, Chai Nat, Maha Sarakham, Mukdahan, Nakhon Phanom, Nong Bua Lam Phu, Nonthaburi, Phayao, Phichit, Phra Nakhon Si Ayutthaya, Samut Prakan, Samut Sakhon, Si Sa Ket, Sing Buri, Uthai Thani and Yasothon. Most spiders were reported from Chiang Mai Province.

Identification of long-preserved specimens reveals the historical geographic range of the Patagonian lamprey Geotria macrostoma (Burmeister, 1868) in southern South America.

The lamprey genus Geotria Gray, 1851 currently includes only two species: G. australis and G. macrostoma. However, taxonomic relationships within the genus have traditionally been ambiguous and difficult to establish due to the extreme changes in morphology, dentition, and coloration that lampreys undergo during their life cycles, particularly during upstream migration and sexual maturation. Consequently, several lamprey specimens held in museum collections have remained unidentified, especially those from Argentina. In this study, a series of morphometric characters were subjected to discriminant function analysis (DFA) to identify the lamprey species collected during 1867-2004 from the de la Plata River and Patagonia. These specimens are housed at the Museo Argentino de Ciencias Naturales "Bernardino Rivadavia" in Buenos Aires, the Museo de Historia Natural de Montevideo, and the Naturhistoriska riksmuseet in Stockholm. Based on the proportions of the length of the oral disc, prebranchial, and pre-caudal body regions, and the depth of the trunk, DFA provided conclusive evidence that the specimens corresponded to the recently revalidated G. macrostoma (Burmeister, 1868), which was originally incorrectly named as Petromyzon macrostomus Burmeister, 1868, Exomegas macrostomus (Berg, 1899), Geotria chilensis (Berg, 1895), and Geotria macrostoma f. gallegensis Smitt, 1901, as well as other nontype museum individuals of uncertain taxonomic status. The identifications of these long-preserved museum specimens provided key information on the historical geographic range of Argentinian lampreys and suggest that the disappearance of the species reported from northern localities (the Pampean Region) can be attributed to the degradation of their critical habitats, primarily caused by anthropogenic impact and climate change.

Identification of Orissa frog Fejervarya orissaensis from Bangladesh based on 16s and 12s rRNA genes

In this study, we attempted species-level identification of frog specimen collected from Faridpur district of Bangladesh beyond it’s location outside Orissa, India. Specimen was identified morphologically at genus level as Fejervarya sp. belonging to the family Dicroglossidae using finger formula F3>F4>F1=F1 where F denote as toe finger. From the study two nucleotide sequences of 16S and 12S rRNA genes were obtained which contained 508bp and 408bp respectively. The Sequences were submitted to Gene Bank database with the accession number OQ231604 and OQ240197 for 16S and 12S rRNA gene sequences. Furthermore, the 16S rRNA gene sequence was used as molecular bar-code for the identified Orissa frog F. orissaensis species from Bangladesh. GC content of partial 12S and 16S rRNA genes have been calculated as 44% and 45% respectively. For 16S rRNA gene sequence there was no intra specific divergence. Whereas the inter specific polymorphic divergence were calculated 4.13% and 6.3% when the collected Orissa frog F. orissaensis was compared with that of F. iskandari and F. kupitzi, respectively. In case of 12S rRNA gene intra specific divergence was found 2.45% where the inter specific divergence were 4.41% and 6.86% when the collected Orissa frog F. orissaensis was compared with that of F. iskandari and F. limnocaris, respectively. Maximum likelihood tree also indicates that our sample Orissa frog formed a monophyletic group with F. orissaensis in both the cases of 16S and 12S rRNA genes and thus can be concluded as closely related. Therefore, the collected specimen was identified to be belonging to Fejervarya orissaensis which would be first report from Bangladesh outside Orissa, India. Bangladesh J. Zool. 51(3): 301-313, 2023

Identification of fish specimens of the Tocantins River, Brazil, using DNA barcoding.

The fish fauna of the Tocantins River possesses many endemic species; however, it is little studied in molecular terms and is quite threatened by the construction of several hydroelectric dams. Therefore, the objective of this study was to identify the ichthyofauna of the Tocantins River using DNA barcoding. For this, collections were carried out in five points of this river, which resulted in the capture of 725 individuals from which partial sequences of the cytochrome oxidase subunit I (COI) gene were obtained for genetic analysis. A total of 443 haplotypes were recovered with the mean intraspecific K2P genetic distance of 1.82%. Altogether, 138 species were identified based on morphological criteria, which was a quantity that was much lower than that indicated by the four molecular methods (assemble species by automatic partitioning [ASAP], barcode index number [BIN], generalized mixed Yule coalescent (GMYC), and Bayesian Poisson tree processes [bPTP]) through which 152-157 molecular entities were identified. In all, 41 unique BINs were obtained based on the data generated in the BOLDSystems platform. According to the result indicated by ASAP (species delimitation approach considered the most appropriate in the present study), there was an increase of 17 molecular entities (12.32%), when compared to the number of species identified through their morphological criteria, as it can show cryptic diversity, candidates for new species, and misidentifications. There were 21 incongruities indicated between the different identification approaches for species. Therefore, it is suggested that these taxonomic problems be cautiously evaluated by experts to solve such taxonomic issues.

Molecular Evidence Reveals Taxonomic Uncertainties and Cryptic Diversity in the Neotropical Catfish of the Genus Pimelodus (Siluriformes: Pimelodidae).

Pimelodus is the most speciose genus of the family Pimelodidae, and is amply distributed in the Neotropical region. The species-level taxonomy and phylogenetic relationships within this genus are still poorly resolved, however. These taxonomic problems and the general lack of data have generated major uncertainties with regard to the identification of specimens from different localities. In the present study, we applied a single-locus species delimitation approach to identify the MOTUs found within the genus Pimelodus and provide sound evidence for the evaluation of the species richness of this genus in the different river basins of the Neotropical region. The study was based on the analysis of sequences of the mitochondrial COI gene of 13 nominal species, which resulted in the identification of 24 consensus MOTUs. Only six nominal species were recovered as well-defined molecular entities by both the traditional barcoding analysis and the molecular delimitation methods, while the other seven presented cryptic diversity or persistent taxonomic uncertainties. The lineages identified from the Parnaíba ecoregions, Amazonas Estuary and Coastal Drainages may represent a much greater diversity of Pimelodus species than that recognized currently, although a more detailed study of this diversity will be necessary to provide a more definitive classification of the genus.
