Identification of Chlorogenic Acid, Caffeine, Melanoidin, Sucrose, and Protein Content of Local Indonesia Arabica Coffee Base on Its Cupping and Variety Variation

The increased demand for high-quality coffee led to the need to evaluate coffee quality for market acceptance. Indonesia has Arabica, which has the potential for high selling value. Arabica coffee varieties, including Ateng, Lini S, and Sigararutang, affect the content of compounds in coffee beans, such as chlorogenic acid, caffeine, melanoidin, sucrose, and protein. The objective of this study was to analyze the impact of Arabica coffee varieties on the compound composition and flavor characteristics of coffee. The UV-Vis spectrophotometric methods were used to determine the quantity of chlorogenic acid, caffeine, melanoidin, sucrose, and protein in different Arabica coffee varietals. The study revealed that the content of these compounds differed among the varieties. The Lini S variety had the highest chlorogenic acid and caffeine content, while the Ateng variety had the highest melanoidin, sucrose, and protein content. The varying levels of chlorogenic acid and caffeine in coffee significantly impact its flavor, resulting in a unique sour and bitter taste in coffee brew. Melanoidin, a high-molecular-weight brown compound, contributes to the malty flavor of coffee, along with sweet, nutty, caramel, and spice notes. Sucrose adds a sweet hint to coffee brew, while protein has a relatively minimal influence on coffee flavor.

Identification of chlorogenic acid in cotyledonous leaves and husks of common sunflower (Helianthus annuus L.)

The relevance of the study is conditioned by the need to optimise the production process of the sunflower farming ecosystem by increasing environmental sustainability. Among the by-products of processing, the use of which is still not effective, the release of the powerful antioxidant chlorogenic acid has not been sufficiently studied. In this regard, this paper is aimed at revealing the patterns of accumulation and identification of phenolic compounds in cotyledons and sunflower husks. The purpose of the study is to identify phenolic compounds in cotyledonous leaves and husks of various genotypes of common sunflower (Helianthus annuus L.) by thin-layer chromatography. In the course of research on this problem, the authors used field, biochemical, and statistical research methods. To identify chlorogenic acid, a solvent system was used: chloroform – glacial acetic acid – methanol – water (60:32:12:8). Chromatography analysis was performed in the SORBFLIL TLC software suite. The object of research is the Auris common sunflower hybrid. It is established that the qualitative composition of the phenolic complex and the number of individual compounds has a pronounced variety specificity. On the chromatogram, this substance has an Rf of ~ 0.60 (for sunflower husks) and an Rf of ~0.74 (for cotyledonous leaves). It was found that the most effective solvent for chlorogenic acid is aqueous ethanol. Extraction of chlorogenic acid with 40% ethanol provides up to 10 mg/g of chlorogenic acid. In addition to chlorogenic acid, a small amount of caffeic acid was found in the alcohol extract. The materials of the study are of practical value for improving the processes related to the development of the productivity of sunflower crops and the rational use of processing by-products

Identification of Chlorogenic Acid from Morus alba Leaves by UV-Vis Spectroscopy, FTIR, UPLC-QTOF-MS and Quantification by HPTLC

ABSTRACT The present study aims to characterize chlorogenic acid extracted from Morus alba leaves by using suitable methods such as UV-Vis spectroscopy, Fourier transform infrared spectroscopy (FTIR), ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOF-MS) and further quantification by high-performance thin layer chromatography (HPTLC). Sonication method and methanol solvent (50:50%, v/v) is found more effective for the extraction of chlorogenic acid from leaf extract. UV-Vis spectra has shown maximum absorbance at λmax 330 nm, and found overlapping with standard chlorogenic acid. The functional groups of chlorogenic acid were identified by FTIR which indicated the presence of hydroxyl, carboxyl and carbonyl groups. In UPLC-QTOF-MS study, the mass spectra and total ion chromatogram analysis revealed the presence of structural isomers for chlorogenic acid at same m/z of parent ion 353. Mass fragments of daughter ions indicated base peak at m/z 191, m/z 173 and 179 were assigned as 5-O-caffeoylquinic acid, 4-O-caffeoylquinic acid and 3-O-caffeoylquinic acid. Quantitative analysis of chlorogenic acid was performed by HPLTC, overlapping absorption spectra of plant extract revealed great consistency with standard, confirms the specificity of HPTLC method. Similar retention factor (Rf) at 0.14 confirms the presence of chlorogenic acid in leaf extract. The standard plot shows good linear relationship from 200 to 1000 ng/spot (R2 = 0.9986). The detection limit was 15.45 ng/spot while limit of quantification was 46.84 ng/spot, with good average recovery rate between 97.96–99.03%. The amount of chlorogenic acid was quantified 10.8 mg/g dried weight extract.

A robust platform based on ultra-high performance liquid chromatography Quadrupole time of flight tandem mass spectrometry with a two-step data mining strategy in the investigation, classification, and identification of chlorogenic acids in Ainsliaea fragrans Champ

The chlorogenic acids are the major bioactive constituents of the whole plant of Ainsliaea fragrans Champ. (Xingxiang Tuerfeng). These compounds are usually present as isomeric forms in Xingxiang Tuerfeng. Therefore, an efficient approach should be developed for the rapid discovery and identification of chlorogenic acids isomers through the fragmentation pathway and rules. In this study, the collision induced dissociation tandem mass spectrometry (CID-MS/MS) fragmentation routes of chlorogenic acids were systematically investigated by UHPLC–QTOF–MS/MS in the negative ion mode using eight chlorogenic acids standards. As a result, diagnostic product ions for rapid discovery and classification of chlorogenic acids isomers were determined according to their MS/MS fragmentation patterns and intensity analysis. Based on these findings, a novel two-step data mining strategy was established. The first key step was to screen different kinds of substitution and the skeleton of the quinic acid using the characteristic product ions and neutral losses. The second key step was to screen and classify different types of chlorogenic acids using their diagnostic product ions. It was apply to the rapid investigation, classification, and identification of chlorogenic acids. And the same carbon skeletons from a complex extract of Ainsliaea fragrans Champ. were effectively identified. 88 constituents, including 14 chlorogenic acids types, were rapidly discovered and identified, and in particular, 12 types of chlorogenic acids, including p-CoQC, FQA, BQC, CQA-Glu, CFQA, p-Co-CQC, di-p-CoQC, BCQA, di-CQA-Glu, PCQA, tri-QCA, and P-di-CQA, were first discovered in Ainsliaea fragrans Champ. In conclusion, UHPLC-QTOF-MS/MS method together with a systematic two-step data mining strategy was established as a feasible, effective, and rational technique for analyzing chlorogenic acids. Additionally, this study laid a foundation for the study of the active substances and quality control of Ainsliaea fragrans Champ.

Identification of Chlorogenic Acid as a Resistance Factor for Thrips in Chrysanthemum

Western flower thrips (Frankliniella occidentalis) has become a key insect pest of agricultural and horticultural crops worldwide. Little is known about host plant resistance to thrips. In this study, we investigated thrips resistance in chrysanthemum (Dendranthema grandiflora). We identified thrips-resistant chrysanthemums applying bioassays. Subsequently, nuclear magnetic resonance (NMR)-based metabolomics was applied to compare the metabolome of thrips-resistant and -susceptible chrysanthemums. NMR facilitates wide-range coverage of the metabolome. We show that thrips-resistant and -susceptible chrysanthemums can be discriminated on basis of their metabolomic profiles. Thrips-resistant chrysanthemums contained higher amounts of the phenylpropanoids chlorogenic acid and feruloyl quinic acid. Both phenylpropanoids are known for their inhibitory effect on herbivores as well as pathogens. Thus, chlorogenic and feruloyl quinic acid are the compounds of choice to improve host plants resistance to thrips in ornamentals and crops. The effect of chlorogenic acid on thrips was further studied in bioassays with artificial diets. These experiments confirmed the negative effects on thrips. Our results prove NMR to be an important tool to identify different metabolites involved in herbivore resistance. It constitutes a significant advance in the study of plant-insect relationships, providing key information on the implementation of herbivore resistance breeding strategies in plants.
