Evaluation of antioxidant and anti-inflammatory activity and identification of bioactive compound from the marine diatom, Odontella aurita extract

Increased production of reactive oxygen species (ROS) leads to oxidative stress, with its damaging effect extending to the mitochondria and plasma membrane. Further, prolonged inflammation can result in chronic disease development. The marine microdiatom Odontella aurita is recognized for its potential in food and pharmaceutical development. Moreover, it contains antioxidant and anti-inflammatory properties. However, studies regarding the efficacy of their varying extract forms and their underlying mechanisms remain scarce. Therefore, this study aims to explore the antioxidant and anti-inflammatory effects of Odontella aurita extracts obtained using various extraction methods (hot water, 70% ethanol, and chloroform:methanol (CM)). Among the three Odontella aurita extracts, the CM extract demonstrated superior efficacy in protecting RAW 264.7 cells from H2O2-induced cytotoxicity. It significantly lowered the levels of ROS and enhanced the expression of superoxide dismutase and glutathione peroxidase. Furthermore, the CM extract outperformed other extracts in inhibiting LPS-induced nitric oxide production, reducing mRNA levels in nitric oxide synthase, cyclooxygenase, and the proinflammatory cytokines interleukin IL-1β, IL-6, TNFα. Additionally, CM extract effectively suppressed the activation of NF-κB/IκBα and JAK2-STAT3 in LPS-induced RAW 264.7 cells. HPLC–UV analysis revealed a remarkable 33-fold higher fucoxanthin content in CM compared to the ethanol extract. GC–MS analysis identified elevated levels of cholest-5-en-3-ol, phytol, eicosapentaenoic acid methyl ester, methyl palmitate, palmitoleic acid methyl ester, and neophytadiene in the CM extract. These findings suggest that Odontella aurita CM extract is a promising antioxidant candidate for preventing or treating inflammatory diseases, consequently emphasizing its potential for further development.Graphical

Computational identification of potential bioactive compounds from Triphala against alcoholic liver injury by targeting alcohol dehydrogenase.

Alcoholic liver injury resulting from excessive alcohol consumption is a significant social concern. Alcohol dehydrogenase (ADH) plays a critical role in the conversion of alcohol to acetaldehyde, leading to tissue damage. The management of alcoholic liver injury encompasses nutritional support and, in severe cases liver transplantation, but potential adverse effects exist, and effective medications are currently unavailable. Natural products with their potential benefits and historical use in traditional medicine emerge as promising alternatives. Triphala, a traditional polyherbal formula demonstrates beneficial effects in addressing diverse health concerns, with a notable impact on treating alcoholic liver damage through enhanced liver metabolism. The present study aims to identify potential active phytocompounds in Triphala targeting ADH to prevent alcoholic liver injury. Screening 119 phytocompounds from the Triphala formulation revealed 62 of them showing binding affinity to the active site of the ADH1B protein. Promising lipid-like molecule from Terminalia bellirica, (4aS, 6aR, 6aR, 6bR, 7R, 8aR, 9R, 10R, 11R, 12aR, 14bS)-7, 10, 11-trihydroxy-9-(hydroxymethyl)-2, 2, 6a, 6b, 9, 12a-hexamethyl-1, 3, 4, 5, 6, 6a, 7, 8, 8a, 10, 11, 12, 13, 14b-tetradecahydropicene-4a-carboxylic acid showed high binding efficiency to a competitive ADH inhibitor, 4-Methylpyrazole. Pharmacokinetic analysis further confirmed the drug-likeness and non-hepatotoxicity of the top-ranked compound. Molecular dynamics simulation and MM-PBSA studies revealed the stability of the docked complexes with minimal fluctuation and consistency of the hydrogen bonds throughout the simulation. Together, computational investigations suggest that (4aS, 6aR, 6aR, 6bR, 7R, 8aR, 9R, 10R, 11R, 12aR, 14bS)-7, 10, 11-trihydroxy-9-(hydroxymethyl)-2, 2, 6a, 6b, 9, 12a-hexamethyl-1, 3, 4, 5, 6, 6a, 7, 8, 8a, 10, 11, 12, 13, 14b-tetradecahydropicene-4a-carboxylic acid from the Triphala formulation holds promise as an ADH inhibitor, suggesting an alternative therapy for alcoholic liver injury.

Ethnobotanical survey of medicinal plants used by various ethnic tribes of Mizoram, India.

Mizoram has a diverse range of ethnic and cultural groups, including the Lushai, Mara, Lai, Bawm, Hmar, Chakma, and Bru. Through practice and experience in their protracted battles with disease and the harsh natural environment, they have amassed rich folk medicine knowledge. The preservation of indigenous knowledge, identification of bioactive compounds, and the eventual discovery of novel medicinal plant species all depend on the documentation of the traditional uses of medicinal plants. Therefore, the present study was designed to document the ethnomedicinal knowledge related to the usage of medicinal plants in Mizoram, India. An ethnobotanical study of medicinal plants was carried out in 17 villages of 5 districts in Mizoram between September 2022 and January 2023. A total of 128 informants (77 males and 51 females) were interviewed. Information was gathered through field observations, group discussions, and semistructured interviews. The data were presented using descriptive statistics. To assess the importance of medicinal plant species, quantitative indices such as the informant consensus factor (ICF), fidelity level (FL), use value (UV), and cultural index (CI) was used. In our ethnobotanical investigation, 102 medicinal plant species from 95 genera and 58 families were recorded and documented. The majority of them (90 species) were obtained from wild habitats. The majority of the documented species were trees (48), followed by herbs (23 species) and shrubs (21 species). The most common method of preparation was decoction (67 species). Diabetes had the highest ICF value (0.81), followed by cancer, liver problems, and hypertension (0.8). The fidelity level (FL) of indigenous communities in the study area was evaluated to determine the dependability and consistency of herbal drug use. Indigenous knowledge and the variety of medicinal plant species that are used are of great values. The therapeutic applications of documented plants provide fundamental information for additional studies centered on pharmacological investigations and the preservation of the most significant species.

Phytochemical screening and antimicrobial activities of Guizotia abyssinica L. leaf and flower extracts

Sustainable agricultural systems demand less or no use of synthetic chemicals, which have various adverse effects on the environment and human health. In this context, studies on phytochemicals with antimicrobial and antioxidant properties hold great promise to suppress plant diseases and, thereby, reducing the application of synthetic chemical pesticides. Such an alternative approach requires screening and identification of bioactive compounds from various plant species. In the present study, Guizotia abyssinica L., an oilseed crop plant that usually requires less water with no pesticide inputs to grow, was screened for its potential antioxidant and antimicrobial phytochemicals. The methanolic and aqueous extracts of leaves and flowers of G. abyssinica were tested for their polyphenol content (total phenols and flavonoids) and antioxidant potential. The antioxidant and free radical scavenging activities of both methanolic and aqueous extracts were higher in comparison to many previously reported plant extracts. Furthermore, leaf extracts of G. abyssinica have shown slightly higher antioxidant activity than flower extracts. In vitro antimicrobial studies confirmed the inhibitory effect of methanolic extracts of leaves and flowers against phytopathogens like Xanthomonas oryzae RR24, Fusarium sp. and Alternaria sp. The chemical nature and composition of these crude extracts were analyzed by Fourier Transform Infrared Spectroscopy (FTIR) and Gas Chromatography Mass Spectrometry (GC-MS). Approximately, 28 bioactive compounds detected in GC-MS were predicted to have antimicrobial activity. Based on our results, we conclude that the extracts derived from G. abyssinica have demonstrated remarkable efficacy in inhibiting the proliferation of phytopathogens, thus exhibiting immense potential in promoting sustainable agricultural practices.

Extraction of bioactive compounds from Vitex megapotamica: A study and selection of variables

AbstractFruits are widely used to obtain and study bioactive compounds. Among them, the tarumã (Vitex megapotamica) is still understudied. The extraction of bioactive compounds from the powdered pulp of the ripe fruit of the tarumã was studied from the selection of several variables of the extraction process through the experimental design Plackett–Burman: the pH, the solid:liquid ratio, agitation, and temperature in an extraction time of 3 h. The extracts obtained were evaluated for antioxidant activity by 2,2′‐azino‐bis‐(3‐ethylbenzothiazoline‐6‐sulfonic acid) (ABTS), 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH), and for total phenolic compounds, as well as the presence of flavonoids, anthocyanins, carotenoids, and identification of bioactive compounds was assessed by high‐performance liquid chromatography (HPLC). Using the experimental design, the best extraction condition was defined with a pH of 5, solid:liquid ratio of 1:30, stirring at 100 rpm, with a temperature of 80°C, obtaining 104.21 mg Gallic Acid Equivalent (GAE) 100 g−1 for phenolic compounds total, 742.71 μmol Trolox g−1 for ABTS, and EC50 of 2884.98 mg L−1 for DPPH. From HPLC, the presence of fumaric acid, gallic acid, p‐coumaric acid, and caffeic acid in the extracts was detected. Regarding flavonoids, anthocyanins, and carotenoids, the values obtained were 6.38 mg ECG g−1, 1.67 mg ECG g−1, and 2.89 mg 100 g−1, respectively. The selection of extraction conditions allowed obtaining better levels of bioactive compounds in the powdered pulp of tarumã extract, even with the use of water as a solvent, to avoid the presence of chemical residues in the extract, with antioxidant potential and viability for application in food products.Practical applicationsRecent research suggests a change in perspective regarding the consumption of healthier foods from plants and fruits that have important health properties. In this regard, tarumã (Vitex megapotamica) fruit has been standing out as a potential source of bioactive compounds with significant medicinal properties. The extracts obtained have a significant amount of phenolic compounds, as well as the presence of flavonoids, anthocyanins, and carotenoids, in which all of these have great potential bioactive activities. Thus, a good methodology of extraction and characterization of tarumã extract becomes a crucial part of any food, chemical, or pharmaceutical industry sector. In this regard, the present study with experimental design Plackett–Burman extraction is the way ahead due to high bioactive retention and the cost implications of the process.

Optimizing Ultrasound-Assisted Extraction of Bioactive Compounds from Canthium horridum Blume Leaves Utilizing Polyols: A Study on Skin-Related Activities

The focus on reducing organic solvent usage, owing to their negative environmental and health impacts, is driving a search for innovative green alternative solvents in academia and industry. Canthium horridum Blume (CH) exhibits many therapeutic activities, including antioxidant and anti-inflammatory efficacy. The objective of this study is to evaluate the optimal solvent concentration using a simplex-lattice design with an aqueous-polyols mixture and to optimize the parameters for extracting bioactive compounds and antioxidant activities from ultrasound-assisted extraction (UAE) of CH leaves through central composite design (CCD) in response surface methodology (RSM). This study examines the total phenolic content (TPC), antioxidant activities, comparison of different extraction conditions, identification of bioactive compounds, cell cytotoxicity, cellular antioxidant activity, and melanin content reduction efficacy of the extracts. According to the findings from the simplex-lattice model, the ideal solvent composition consisted of 32.57%w/w butylene glycol, 32.92%w/w glycerine, and 34.51%w/w water. Furthermore, based on the response model, optimal extraction conditions were identified as a 15-minute extraction time and a solvent-to-sample ratio of 32.94:1. In comparison to alternative extraction methods, ultrasonic-assisted extraction using the aqueous-glycerine-butylene glycol (GB-UAE) extract resulted in notably elevated TPC and antioxidant responses (p<0.05). Major antioxidant bioactive compounds included 4-(Butoxymethyl) phenol, 3-O-Caffeoyl-4-O-methylquinic acid, Quercetin 3-(2G-glucosylrutinoside), 2,4-Dihydroxybenzoic acid and other bioactive compounds. The GB-UAE extract revealed greater cell viability than UAE using ethanol (EtOH-UAE) extract in both cytotoxicity and cellular antioxidant assays at the same concentration. Additionally, it exhibited comparable melanin content reduction efficacy at a higher concentration compared to that of EtOH-UAE extract. The researcher anticipates that the current study will advance the utilization of an aqueous-polyols system for extracting bioactive compounds extending beyond CH leaves. Although the potential applications of CH leaves in cosmetics and pharmaceutical formulations have been identified, further comprehensive mechanistic and clinical studies are required to fully understand their effects.

1H-NMR Metabolomic Study of the Mushroom Pleurotus djamor for the Identification of Nematocidal Compounds

Due to the increasing populations of anthelmintic-resistant gastrointestinal nematodes and as a consequence of the adverse effects of synthetic drugs, this study focuses on the search for secondary metabolites with nematocidal activity from the edible mushroom Pleurotus djamor using The proton nuclear magnetic resonance (1H-NMR) metabolomics. The highest activity was shown by the ethyl acetate fractions of mycelium (EC50 290.8 µg/mL) and basidiomes (EC50 282.7 µg/mL). Principal component analysis (PCA) and hierarchical data analysis (HCA) of the 1H-NMR metabolic profiles data showed that the ethanolic extracts, the ethyl acetate, butanol, and water fractions from mycelium have different metabolic profiles than those from basidiomes, while low polarity (hexane) fractions from both stages of fungal development show similar profiles. Orthogonal partial least squares discriminant analysis (OPLS-DA) allowed the identification of signals in the 1H-NMR metabolic profile associated with nematocidal activity. The signals yielded via OPLS-DA and bidimensional NMR analysis allowed the identification of uracil as a component in the ethyl acetate fraction from basidiomes, with an EC50 of 237.7 µg/mL. The results obtained showed that chemometric analyses of the 1H-NMR metabolic profiles represent a viable strategy for the identification of bioactive compounds from samples with complex chemical profiles.

Phytochemical screening and GC-FID identification of bioactive compounds in n-hexane, ethylacetate and methanol fractions of methanolic leaves extract of Azanza garckeana

Azanza garckeana is a tropical African plant commonly used for food and medicine. This study was aimed at evaluating the antioxidant, phytochemicals and the bioactive compounds present in n-hexane, ethylacetate and methanol fractions of the methanolic extract of Azanza garckeana leaves. Phytochemical screening and antioxidant assayed were carried out using standard methods, while bioactive compounds were analyzed using gas chromatography-flame ionization detector (GC-FID). The phytochemical compositions revealed considerable level of tannins and saponins with moderate concentration of flavonoids and steroids in the methanol fraction; whereas n-hexane fraction also showed considerable amount of tannins followed by saponins and a low concentration of steroids. However, ethylacetate fraction had the highest yield of tannins, steroids, and terpenoids which were significantly (p < 0.05) higher than methanolic fraction and n-hexane fraction respectively. The antioxidant activity of the methanolic crude extract of A. garckeana leaves showed that the % DPPH radical scavenging activity of the crude extract increased with decreased in concentration when compared with the standard antioxidant (ascorbic acid). The result of GC-FID analysis revealed the presence of nineteen constituents in n-hexane fraction, while ethylacetate and methanol fractions each have twenty constituents. This result showed that Azanza garckeana leaves contain health-promoting bioactive compounds with potent antioxidants.

Cheminformatics and QSAR‐Based Identification of Natural Bioactive Compounds as Potent Inhibitors of SARS‐CoV‐2 N‐7 Methyltransferases

AbstractEver since the emergence of the COVID‐19 pandemic, to date, no specific and efficient therapeutic cure is available for the SARS‐CoV‐2 viral infection. Few reports have been observed with the late cardiovascular or neurodegenerative disorders that are associated with altered immunity. The nsp14 protein of SARS‐CoV‐2 is a key target for antiviral drug development as it controls replication of virus and evasion of host immune system. In this study, a consensus of cheminformatics approaches involving virtual screening, molecular docking, ADMET profiling, molecular dynamics simulations, free‐energy landscape, MM‐GBSA, DFT and 2D‐QSAR analysis is employed to discover the inhibitor of nsp14 protein. Our investigation reveals four natural bioactive compounds, i. e. ZINC2132169, ZINC8791872, ZINC8952607 and ZINC6624334 that possess enhanced binding affinity towards nsp14 protein with no predicted toxicity. Molecular docking, MD simulations and MM‐GBSA analysis established that all hit compounds strongly bind at the same pocket through hydrophobic interactions and hydrogen bonding. DFT analysis revealed greater reactivity of hit compounds and their stable complex formation with nsp14 protein, while the 2D‐QSAR (R2=0.92) predicted their pIC50 value in the range of 5.30–6.71 nM. These findings imply that hit compounds would be potent drug candidates in controlling the SARS‐CoV‐2 by inhibiting nsp14 protein.

Identification of new endocrine disruptive transthyretin ligands in polluted waters using pull-down assay coupled to non-target mass spectrometry

Surface and treated wastewater are contaminated with highly complex mixtures of micropollutants, which may cause numerous adverse effects, often mediated by endocrine disruption. However, there is limited knowledge regarding some important modes of action, such as interference with thyroid hormone (TH) regulation, and the compounds driving these effects. This study describes an effective approach for the identification of compounds with the potential to bind to transthyretin (TTR; protein distributing TH to target tissues), based on their specific separation in a pull-down assay followed by non-target analysis (NTA). The method was optimized with known TTR ligands and applied to complex water samples. The specific separation of TTR ligands provided a substantial reduction of chromatographic features from the original samples. The applied NTA workflow resulted in the identification of 34 structures. Twelve compounds with available standards were quantified in the original extracts and their TH-displacement potency was confirmed. Eleven compounds were discovered as TTR binders for the first time and linear alkylbenzene sulfonates (LAS) were highlighted as contaminants of concern. Pull-down assay combined with NTA proved to be a well-functioning approach for the identification of unknown bioactive compounds in complex mixtures with great application potential across various biological targets and environmental compartments.

Metabolomic and chemometric analyses of St. John's wort and related Asian Hypericum species linked to bioactivity

Ethnopharmacological relevancePlants in the genus Hypericum (Hypericaceae), include more than 500 species worldwide, and many are valued for their medicinal properties, and are used as traditional herbal medicines. However, only H. perforatum is officially recognized as herbal drug in several pharmacopoeias, and used as an antidepressant clinically. Hypericum perforatum had been used as an herbal medicine since the Han Dynasty (206 B.C. -220 A.D.) in China. It taxonomically belongs to the section Hypericum in the genus Hypericum. There are about 42 species in the section Hypericum, with six species occurring in China. All six are recorded as traditional herbal medicines for treating aliments, including hepatitis, malaria, traumatic hemorrhage, irregular menstruation, wounds, and bruises. Aim of the studyThe study aimed to characterize the chemical profiles of five phylogenetically related Hypericum species, and compare their metabolites with three H. perforatum products. Informed by ethnobotanical use, the extracts prepared from the five species were further investigated into anticancer, anti-inflammatory and antiplasmodial activity. This study tested the hypothesis that systematic metabolomic and bioactivity characterization of species in section Hypericum will help to validate their phytotherapeutic use and reveal potential drug lead compounds. Materials and methodsTargeted and non-targeted metabolic analyses coupled with chemometrics were conducted on H. perforatum and four medicinal species, H. attenuatum, H. enshiense, H. erectum, and H. faberi, native to China from section Hypericum. UPLC-QTOF-MS/MS and UPLC-TQD-MS/MS were used for non-targeted and targeted metabolic analyses, respectively. Cytotoxicity bioassays on four cancer cell lines, anti-inflammation tests and anti-plasmodial activity on Plasmodium falciparum 3D7, selected based on traditional medicinal use, were evaluated on extracts from Hypericum species. Progenesis QI and EZinfo were used for chemometrics analysis to link the chemical profile and bioassay activity to aid in the identification of bioactive compounds. ResultsIn total, 58 compounds were identified from the five species, including compounds with well-characterized bioactivity. Hypericum attenuatum, H. erectum, and H. perforatum, displayed the highest cytotoxicity, and contain the cytotoxic compounds petiolin A, prolificin A, and hypercohin G, respectively. Hypericum faberi and H. perforatum showed the highest anti-inflammatory activity, with pseudohypericin, quercetin and chlorogenic acid being observed at higher concentrations. Hypericum perforatum and H. erectum showed anti-plasmodial activity, with higher hyperforin and xanthones in these species that may account for the anti-plasmodial activity. ConclusionsThis study characterized the chemical differences among five Hypericum species using metabolomics. These ethnomedically important species were tested for their biological activities in three distinct in vitro assays. The ethnobotanical data were useful for identifying bioactive Hypericum species. Hypericum attenuatum, H. erectum and H. faberi are promising phytotherapeutic species, although they are much less studied than H. perforatum, St. John's wort. Combining ethnobotanical surveys with chemometric analyses and bioactivity screening can greatly enhance the discovery of promising active constituents.

Fractionation and identification of bioactive compounds from a salt marsh plant Sesuvium potrucastrulam (L.) and its antioxidant activity

Sesuvium portulacastrum (L.) is a fast-growing herbaceous perennial and halophyte belonging to the family Aizoaceae. Bioactive compound identification from halophytes is more helpful for the drug development process. The present investigation was to fractionate and identify the bioactive compounds of leaf extracts from Sesuvium potrucastrulam and evaluate their antioxidant potential. The Soxhlet extraction method was used in this study, and column chromatography was done for the partial purification. The bactericidal activity of the fraction was determined using the agar-well diffusion technique, and the effective fraction was analysed by GC-MS. A hydrogen peroxide-reducing assay was carried out on the antioxidant activity of the elite fraction. Five active fractions were collected from the crude extract. Fraction (F3) exhibited promising antibacterial and antioxidant activity. GS-MS analysis suggested the active compounds of the elite fraction are n-Hexadecanoic acid (29.70%), oleic acid (8.08%), octadecatrienoic acid (8.01%), 2-methoxy-4-vinylphenol (6.65%), tertracosamethyl-cyclododecasiloxane (6.55%), and lineolic acid (4.68%).

Development of Anti-Inflammatory Drug from Crataeva Nurvala: In Silico and In Vitro Approach

ABSTRACT Background: Crataeva nurvala, a medicinal plant with potential therapeutic properties, offers a promising avenue for the development of novel anti-inflammatory drugs. This study adopted a combined in silico and in vitro approach to investigate the anti-inflammatory potential of compounds derived from Crataeva nurvala. Materials and Methods: In the in silico phase, virtual screening and molecular docking analyses were conducted to identify bioactive compounds from Crataeva nurvala that could interact with key inflammatory targets. Subsequently, selected compounds were synthesized and subjected to in vitro experimentation. Cellular models were employed to assess the anti-inflammatory effects of Crataeva nurvala-derived compounds, focusing on the modulation of pro-inflammatory cytokine levels and the underlying signaling pathways. Results: Virtual screening and molecular docking led to the identification of several bioactive compounds with favorable interactions with inflammatory targets. In the in vitro experiments, treatment with Crataeva nurvala-derived compounds resulted in a significant reduction in pro-inflammatory cytokine production. Moreover, the compounds exhibited the ability to modulate inflammatory signaling pathways, further substantiating their anti-inflammatory potential. Conclusions: This study not only contributes to the development of effective anti-inflammatory drugs but also underscores the value of harnessing natural sources such as Crataeva nurvala for therapeutic interventions in inflammatory disorders. The dual-phase strategy presented here provides a robust framework for anti-inflammatory drug discovery and validation.

NanoRAPIDS as an analytical pipeline for the discovery of novel bioactive metabolites in complex culture extracts at the nanoscale.

Microbial natural products form the basis of most of the antibiotics used in the clinic. The vast majority has not yet been discovered, among others because the hidden chemical space is obscured by previously identified (and typically abundant) antibiotics in culture extracts. Efficient dereplication is therefore key to the discovery of our future medicines. Here we present an analytical platform for the efficient identification and prioritization of low abundance bioactive compounds at nanoliter scale, called nanoRAPIDS. NanoRAPIDS encompasses analytical scale separation and nanofractionation of natural extracts, followed by the bioassay of interest, automated mass spectrometry identification, and Global Natural Products Social molecular networking (GNPS) for dereplication. As little as 10 μL crude extract is fractionated into 384 fractions. First, bioactive congeners of iturins and surfactins were identified in Bacillus, based on their bioactivity. Subsequently, bioactive molecules were identified in an extensive network of angucyclines elicited by catechol in cultures of Streptomyces sp. This allowed the discovery of a highly unusual N-acetylcysteine conjugate of saquayamycin, despite low production levels in an otherwise abundant molecular family. These data underline the utility and broad application of the technology for the prioritization of minor bioactive compounds in complex extracts.

The cytotoxicity effect and identification of bioactive compounds of Prismatomeris glabra crude leaf extracts against breast cancer cells

BackgroundDespite the fact that natives of Southeast Asia have been consuming Prismatomeris glabra for decades for a variety of health benefits, research on this species is not as extensive as that on other species due to its limited distribution. The purpose of this study was to determine the cytotoxicity and identify the bioactive compounds of P. glabra crude leaf extracts against the MCF-7 cell line.ResultsWe first examined the potential cytotoxic activity of P. glabra using the MTT assay against the MCF-7 cell line to determine the IC50 of the plant extracts at various concentrations at three time points (24 h, 48 h, and 72 h). Across all time points, the MTT assay revealed that the aqueous extract exhibited the lowest IC50 values (p < 0.05) compared to the ethanol and methanol extracts. All plant extracts exerted the ability to induce cell death in the MCF-7 cell line at all time points, and the optimal time for P. glabra to manifest its antiproliferative activities and promote cell death was 48 h. LC–MS analysis was conducted to reveal the components in plant extracts. Forty compounds were discovered in P. glabra's extracts, with the majority being flavonoids and triterpenoids. Five similar compounds were present in all three extracts. Further research should be conducted on these compounds to unveil a compound that fulfils the criteria as a promising anticancer agent. This research is of the utmost importance, as it provides a fundamental framework for the identification of alternative therapies for breast cancer and contributes implicitly to the development of new drugs.ConclusionsThis study discovered that P. glabra crude leaf extracts have the potential to inhibit the MCF-7 cell line by inducing cell death.

Identification of Bioactive Compounds in Chrysobalanus icaco Seed Kernel Using Gas Chromatography-Mass Spectrometry

Chrysobalanus icaco seed kernel (i.e Cocoplum) is good source of oil and is utilized as spice in soups or stews in some African cuisines. The work aimed to identify compounds in methanol and dichloromethane/methanol (1:1, v/v) flour extract from its seed kernels using gas chromatography-mass spectrometry. Eleven compounds were identified from the methanol extract while thirteen compounds were identified from the dichloromethane/methanol extract. The most abundant compounds from the methanol extract were cis-13-octadecenoic acid methyl ester, cis vaccenic acid and n-Hexadecanoic acid. They had relative abundance of 28.83, 17.14 and 15.40% respectively. 2,6-octadienal-3,7-dimethyl (E) (ie α-citral), citral, cis-3-hexenyl cis-3-hexenoate and trans-2,7-dimethyl-4,6-octadiene-2-ol were the most abundant in the dichloromethane/methanol extract. These had relative abundance of 12.86, 18.92, 10.56 and 20.59% respectively. Compounds which were common to both methanol and dichloromethane/methanol extracts of C. icaco seed kernel were 2,6-octadienal-3,7-dimethyl (E), hexadecanoic acid methyl ester, n-Hexadecanoic acid, heptadecanoic acid, 16 methyl, methyl ester, cis-vaccenic acid and oleic acid. Both solvents extracted varied concentrations of compounds which fall in different classes of either being an unsaturated long chain aldehyde, fatty acids, fatty acid methyl esters, indene derivatives, monoterpenoid alchohol. The compounds identified in both extracts of Chrysobalamus icaco seed kernels have various beneficial bioactivities and sensory attributessuch as being flavor compounds which confers C.icaco seed kernel a peculiar aroma. As such, Chrysobalamus icaco seed kernel is a functional food and can serve as a good raw material for edible oil from which bioactive compounds can be isolated and utilized in relevant food systems and in the preparation of neutraceuticals and pharmaceuticals. Therefore, its use in cuisines is greatly encouraged.

Identification of Characteristic Bioactive Compounds in Silkie Chickens, Their Effects on Meat Quality, and Their Gene Regulatory Network.

Silkie chicken, an important chicken breed with high medicinal and nutritional value, has a long history of being used as a dietary supplement in China. However, the compounds with health-promoting effects in Silkie chickens remain unclear. In the present study, we conducted a comprehensive analysis of metabolic and lipidomic profiles to identify the characteristic bioactive compounds in Silkie chickens, using a common chicken breed as control. The results showed that the levels of 13 metabolites including estradiol, four lipid subclasses including cardiolipin (CL), eight lipid molecules, and three fatty acids including docosahexaenoic acid (C22:6) were significantly increased in Silkie chickens, which have physiological activities such as resisting chronic diseases and improving cognition. These characteristic bioactive compounds have effects on meat quality characteristics, including improving its water-holding capacity and umami taste and increasing the content of aromatic compounds and phenols. The differentially expressed genes (DEGs) between the two chicken breeds revealed the regulatory network for these characteristic bioactive compounds. Fifteen DEGs, including HSD17B1, are involved in the synthesis of characteristic metabolites. Eleven DEGs, including ELOVL2, were involved in the synthesis and transport of characteristic lipids and fatty acids. In summary, we identified characteristic bioactive compounds in Silkie chickens, and analyzed their effects on meat quality characteristics. This study provided important insight into Silkie chicken meat as a functional food.
